Background: Cancer-associated fibroblasts, as a major component of the tumor microenvironment, have been shown to exhibit protumorigenic effects in pancreatic ductal adenocarcinoma. Moreover, cancer-associated fibroblasts-derived exosomes have been reported to promote tumor development, but exact mechanisms have not been elucidated. The purpose of this study was to investigate the processes by which exosomes generated from cancer-associated fibroblasts promote tumor growth. Methods: twenty-one patients with pancreatic ductal adenocarcinoma who evaluated preoperatively as potentially surgically resectable without distant metastasis and pathologically examined postoperatively as pancreatic ductal cell carcinoma were included. We determined the expression of Leptin as well as downstream proteins at the clinical and cellular levels. Cancer-associated fibroblast-derived exosomes were characterised by nanoparticle transmission electron microscopy and tracking analysis. To ascertain the mechanism mediating the action of exosomal Leptin in pancreatic ductal adenocarcinoma, we performed CCK-8 assay, colony formation assays, transwell and wound healing assays in PSN1 cells to evaluate cell proliferation, migration and invasion. Western blotting was used to detect the level of Leptin, ABL2 and exosome markers. qRT-PCR was employed to evaluate miR-224-3p. Cancer-associated fibroblasts markers and exosome uptake were verified by immunofluorescence. Results: Western blotting assays show that Leptin is present inside tissues and cancer-associated fibroblasts in pancreatic ductal adenocarcinoma. Cancer-associated fibroblasts stimulated PSN1 cells growth, migration and invasion in vitro by secreting the exosomal Leptin. Exosomal Leptin could regulate miR-224-3p, which targets negative regulation of ABL2. Inhibiting Leptin significantly limited PSN1 cells growth, migration and invasion. In vitro analyses revealed that miR-224-3p mimics mitigate the inhibitory effect of cancer-associated fibroblasts knockdown of Leptin on PSN1 cells development, but overexpression of ABL2 partly abolished the tumor-promoting phenotype of miR-224-3p mimics. Conclusion: Our results revealed that cancer-associated fibroblasts mediate pancreatic ductal adenocarcinoma development by regulating the miR-224-3p/ABL2 molecular axis through the secretion of the exosomal Leptin. Human papillomavirus (HPV) infection is a major contributor to cervical cancer. Persistent HPV infection can trigger the expression of IL-32, yet the precise role of IL-32 in the occurrence and development of cervical cancer remains elusive. To investigate this, qRT‒PCR and western blotting were utilized to measure the mRNA and protein expression levels; bioinformatics analysis was used to screen differentially expressed miRNAs; wound healing and transwell assays were conducted to evaluate cell migration and invasion capabilities. Comparative analysis revealed significantly elevated IL-32 expression in cervical cancer tissues and cell lines compared to control groups. In SiHa and/or HeLa, overexpression of IL-32 and IL-32 exposure markedly upregulated miR-205, whereas its knockdown resulted in a substantial downregulation of miR-205. Furthermore, miR-205 also could significantly regulate the expression of IL-32 in HeLa and SiHa cells. Upregulation and downregulation of IL-32 led to a significant increase or decrease in NFκB expression, respectively. Treatment with BAY11-7082 (an NFκB inhibitor) notably decreased miR-205 expression but had no effect on IL-32 levels. qRT‒PCR and western blotting analyses demonstrated that both overexpression and underexpression of IL-32 and miR-205 significantly enhanced or reduced MMP2 and MMP9 expression in cervical cancer cells, respectively. Knockdown of IL-32 significantly inhibited the migration and invasion of HeLa and SiHa; conversely, treatment with rIL-32α and rIL-32γ notably promoted their migration and invasion. In brief, IL-32 is highly expressed via the formation of a positive regulatory loop with NFκB/miR-205, contributing to the persistence of inflammation and promoting the progression of cervical cancer. Objectives: We aimed to explore imaging features including tissue characterization and myocardial deformation in diabetic heart failure with preserved ejection fraction (HFpEF) patients by magnetic resonance imaging (MRI) and investigate its prognostic value for adverse outcomes. Materials and methods: Patients with HFpEF who underwent cardiac MRI between January 2010 and December 2016 were enrolled. Feature-tracking (FT) analysis and myocardial fibrosis were assessed by cardiac MRI. Cox proportional regression analysis was performed to determine the association between MRI variables and primary outcomes. Primary outcomes were all-cause death or heart failure hospitalization during the follow-up period. Results: Of the 335 enrolled patients with HFpEF, 191 had diabetes mellitus (DM) (mean age: 58.7 years ± 10.8; 137 men). During a median follow-up of 10.2 years, 91 diabetic HFpEF and 56 non-diabetic HFpEF patients experienced primary outcomes. DM was a significant predictor of worse prognosis in HFpEF. In diabetic HFpEF, the addition of conventional imaging variables (left ventricular ejection fraction, left atrial volume index, extent of late gadolinium enhancement (LGE)) and global longitudinal strain (GLS) resulted in a significant increase in the area under the receiver operating characteristic curve (from 0.693 to 0.760, p < 0.05). After adjustment for multiple clinical and imaging variables, each 1% worsening in GLS was associated with a 9.8% increased risk of adverse events (p = 0.004). Conclusions: Diabetic HFpEF is characterized by more severely impaired strains and myocardial fibrosis, which is identified as a high-risk HFpEF phenotype. In diabetic HFpEF, comprehensive cardiac MRI provides incremental value in predicting prognosis. Particularly, MRI-FT measurement of GLS is an independent predictor of adverse outcome in diabetic HFpEF. Clinical relevance statement: Our findings suggested that MRI-derived variables, especially global longitudinal strain, played a crucial role in risk stratification and predicting worse prognosis in diabetic heart failure with preserved ejection fraction, which could assist in identifying high-risk patients and guiding therapeutic decision-making. In this study, we described a novel process for separating Li and Fe from spent LiFePO4 power battery cathode materials. The phase evolution and the reaction mechanism of the LiFePO4-NaHSO4·H2O system during the roasting and water leaching process were studied by TG-DSC, XRD, SEM, thermodynamics, and ICP. At 25°C–600°C, the LiFePO4-NaHSO4·H2O system exhibited significant weight loss and endothermic phenomena. The results show that the phase transition of LiFePO4-NaHSO4·H2O system with the increase of NaHSO4·H2O added as follows: LiFePO4, NaHSO4·H2O → Li3Fe2(PO4)3, Fe2O3, LiNaSO4 → Li3Fe2(PO4)3, Fe2(SO4)3, LiNaSO4 → LiNaSO4, Fe2(SO4)3, NaFeP2O7. The surface of the roasted product particles became flat and dense, and plate-like crystal precipitation occurred. The reaction mechanism of the roasting process may be as follows: the production of SO3(g) promoted the transformation of Li3Fe2(PO4)3 and Fe2O3 into LiNaSO4 and Fe2(SO4)3, Li3PO4 is converted into metallic sulfates prior to FePO4, and P 2 O 7 4 - was more stable than PO 4 3 - at higher temperatures. Vascular dementia (VaD) is a cognitive disorder characterized by a decline in cognitive function resulting from cerebrovascular disease. The hippocampus is particularly susceptible to ischemic insults, leading to memory deficits in VaD. Astaxanthin (AST) has shown potential therapeutic effects in neurodegenerative diseases. However, the mechanisms underlying its protective effects in VaD and against hippocampal neuronal death remain unclear. In this study, We used the bilateral common carotid artery occlusion (BCCAO) method to establish a chronic cerebral hypoperfusion (CCH) rat model of VaD and administered a gastric infusion of AST at 25 mg/kg per day for 4 weeks to explore its therapeutic effects. Memory impairments were assessed using Y-maze and Morris water maze tests. We also performed biochemical analyses to evaluate levels of hippocampal neuronal death and apoptosis-related proteins, as well as the impact of astaxanthin on the PI3K/Akt/mTOR pathway and oxidative stress. Our results demonstrated that AST significantly rescued memory impairments in VaD rats. Furthermore, astaxanthin treatment protected against hippocampal neuronal death and attenuated apoptosis. We also observed that AST modulated the PI3K/Akt/mTOR pathway, suggesting its involvement in promoting neuronal survival and synaptic plasticity. Additionally, AST exhibited antioxidant properties, mitigating oxidative stress in the hippocampus. These findings provide valuable insights into the potential therapeutic effects of AST in VaD. By elucidating the mechanisms underlying the actions of AST, this study highlights the importance of protecting hippocampal neurons and suggests potential targets for intervention in VaD. There are still some unanswered questions include long-term effects and optimal dosage of the use in human. In this paper, a photoacoustic trace gas sensor based on a multi-pass enhanced open spherical photoacoustic cell (M-OPAC) was developed. A high-performance spherical photoacoustic (PA) cell was specially designed with high resonant frequency for suppressing various environmental noises, and integrated with a Herriot-type multi-pass optical system for enhancing PA signal. For evaluating the developed PA sensor system, details of theoretical simulation and experimental test were investigated for ambient water vapor (H2O) detection by utilizing a DFB diode laser with a central wavelength of 1391 nm, and wavelength modulation spectroscopy with second harmonic (WMS-2f) detection technique was employed. Compared to the conventional single-pass PA detection strategy, the developed multi-pass PA sensor shows a sensitivity enhancement of 4.3 times and demonstrates a good immunity to ambient noises. Moreover, the developed spherical open photoacoustic cell can be used for open monitoring of atmospheric adsorbent gases, which indicates a great potential for in-site and real-time gas monitoring without complex gas sampling procedures. The high activity of ethyl acetate (EA) combustion is studied over the CuO/CeO2 catalysts which are prepared by the ball milling method, and the relationships between catalytic activity and active sites of CuO and Cu–O–Ce structure are investigated. The finely dispersed CuO particles (around 1.0 nm) and Cu–O–Ce structure are prepared in the CuO/CeO2 catalysts when 5 wt% Cu loading is acquired with the calcination temperature at 500 °C. Complete conversion of ethyl acetate to CO2 is achieved at 210 °C. Via controlling calcination temperatures and treatments by alkaline/acid, it is demonstrated that the synergistic effect between highly dispersed CuO and Cu–O–Ce structure results in a better ability of oxygen activation and catalytic activity for the 5-CuO/CeO2(500) catalyst. Diabetic cardiomyopathy (DCM) is a common cardiovascular complication of diabetes, which may threaten the quality of life and shorten life expectancy in the diabetic population. However, the molecular mechanisms underlying the diabetes cardiomyopathy are not fully elucidated. We analyzed two datasets from Gene Expression Omnibus (GEO). Differentially expressed and weighted gene correlation network analysis (WGCNA) was used to screen key genes and molecules. Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis, and protein–protein interaction (PPI) network analysis were constructed to identify hub genes. The diagnostic value of the hub gene was evaluated using the receiver operating characteristic (ROC). Quantitative real-time PCR (RT-qPCR) was used to validate the hub genes. A total of 13 differentially co-expressed modules were selected by WGCNA and differential expression analysis. KEGG and GO analysis showed these DEGs were mainly enriched in lipid metabolism and myocardial hypertrophy pathway, cytomembrane, and mitochondrion. As a result, six genes were identified as hub genes. Finally, five genes (Pdk4, Lipe, Serpine1, Igf1r, and Bcl2l1) were found significantly changed in both the validation dataset and experimental mice with DCM. The Grey Wolf Optimization (GWO) is a highly effective meta-heuristic algorithm leveraging swarm intelligence to tackle real-world optimization problems. However, when confronted with large-scale problems, GWO encounters hurdles in convergence speed and problem-solving capabilities. To address this, we propose an Improved Adaptive Grey Wolf Optimization (IAGWO), which significantly enhances exploration of the search space through refined search mechanisms and adaptive strategy. Primarily, we introduce the incorporation of velocity and the Inverse Multiquadratic Function (IMF) into the search mechanism. This integration not only accelerates convergence speed but also maintains accuracy. Secondly, we implement an adaptive strategy for population updates, enhancing the algorithm's search and optimization capabilities dynamically. The efficacy of our proposed IAGWO is demonstrated through comparative experiments conducted on benchmark test sets, including CEC 2017, CEC 2020, CEC 2022, and CEC 2013 large-scale global optimization suites. At CEC2017, CEC 2020 (10/20 dimensions), CEC 2022 (10/20 dimensions), and CEC 2013, respectively, it outperformed other comparative algorithms by 88.2%, 91.5%, 85.4%, 96.2%, 97.4%, and 97.2%. Results affirm that our algorithm surpasses state-of-the-art approaches in addressing large-scale problems. Diabetic foot ulcer (DFU) is a leading cause of high-level amputation in DM patients, with a low wound healing rate and a high incidence of infection. Vascular endothelial growth factor (VEGF) plays an important role in diabetes mellitus (DM) related complications. This study aims to explore the VEGF expression and its predictive value for prognosis in DFU, in order to provide basis for the prevention of DFU related adverse events. We analyzed 502 patients, with 328 in healing group and 174 in non-healing/recurrent group. The general clinical data and laboratory indicators of patients were compared through Spearman correlation analysis, ROC analysis and logistic regression analysis. Finally, the independent risk factors for adverse prognosis in DFU patients were confirmed. Spearman analysis reveals a positive correlation between the DFU healing rate and ABI, VEGF in wound tissue, and positive rate of VEGF expression, and a negative correlation with DM duration, FPG, HbA1c, TC, Scr, BUN, and serum VEGF. Further logistic regression analysis finds that the DM duration, FPG, HbA1c, ABI, serum VEGF, VEGF in wound tissue, and positive rate of VEGF expression are the independent risk factors for adverse prognosis in DFU (p < 0.05). DM duration, FPG, HbA1c, ABI, serum VEGF, VEGF in wound tissue, and positive rate of VEGF expression are the independent risk factors for prognosis in DFU patients. Sunspots play a crucial role in both weather forecasting and the monitoring of solar storms. In this work, we propose a novel combined model for sunspot prediction using improved gated recurrent units (GRU) guided by pinball loss for probabilistic forecasts. Specifically, we optimize the GRU parameters using the slime mould algorithm and employ a seasonal-trend decomposition procedure based on loess to tackle challenges related to sequence prediction, such as self-correlations and non-stationarity. To address prediction uncertainty, we replace the traditional l 2 -norm loss with pinball loss. This modification extends the conventional GRU-based point forecasting to a probabilistic framework expressed as quantiles. We apply our proposed model to analyze a well-established historical sunspot dataset for both single- and multi-step ahead forecasting. The results demonstrate the effectiveness of our combined model in predicting sunspot values, surpassing the performance of other existing methods. [ABSTRACT FROM AUTHOR]

Background: Treatment of metastatic renal cell carcinoma (mRCC) remains a challenge worldwide. Here, we introduced a phase I trial of autologous RAK cell therapy in patients with mRCC whose cancers progressed after prior systemic therapy. Although RAK cells have been used in clinic for many years, there has been no dose-escalation study to demonstrate its safety and efficacy. Methods: We conducted a phase I trial with a 3 + 3 dose-escalation design to investigate the dose-related safety and efficacy of RAK cells in patients with mRCC whose cancers have failed to response to systemic therapy (ChiCTR1900021334). Results: Autologous RAK cells, primarily composed of CD8+ T and NKT cells, were infused intravenously to patients at a dose of 5 × 109, 1 × 1010 or 1.5 × 1010 cells every 28 days per cycle. Our study demonstrated general safety of RAK cells in a total of 12 patients. Four patients (33.3%) showed tumor shrinkage, two of them achieved durable partial responses. Peripheral blood analysis showed a significant increase in absolute counts of CD3+ and CD8+ T cells after infusion, with a greater fold change observed in naive CD8+ T cells (CD8+CD45RA+). Higher peak values of IL-2 and IFN-γ were observed in responders after RAK infusion. Conclusion: This study suggests that autologous RAK cell immunotherapy is safe and has clinical activity in previously treated mRCC patients. The improvement in peripheral blood immune profiling after RAK cell infusion highlights its potential as a cancer treatment. Further investigation is necessary to understand its clinical utility. High mobility group protein AT-hook 1 (HMGA1), an architectural transcription factor, has previously been reportedto play an essential role in architectural remodeling processes. However, its effects on cardiovascular diseases, particularly sepsis-induced cardiomyopathy, have remained unclear. The study aimed to investigate the role of HMGA1 in lipopolysaccharide-induced cardiomyopathy. Mice subjected to lipopolysaccharide for 12 h resulted in cardiac dysfunction. We used an adeno-associated virus 9 delivery system to achieve cardiac-specific expression of the HMGA1 gene in the mice. H9c2 cardiomyocytes were infected with Ad-HMGA1 to overexpress HMGA1 or transfected with si-HMGA1 to knock down HMGA1. Echocardiography was applied to measure cardiac function. RT-PCR was used to detect the transcriptional level of inflammatory cytokines. CD45 and CD68 immunohistochemical staining were used to detect inflammatory cell infiltration and TUNEL staining to evaluate the cardiomyocyte apoptosis, MitoSox was used to detect mitochondrial reactive oxygen species, JC-1 was used todetect Mitochondrial membrane potential. Our findings revealed that the overexpression of HMGA1 exacerbated myocardial inflammation and apoptosis in response to lipopolysaccharide treatment. Additionally, we also observed that H9c2 cardiomyocytes with HMGA1 overexpression exhibited enhanced inflammation and apoptosis upon stimulation with lipopolysaccharide for 12 h. Conversely, HMGA1 knockdown in H9c2 cardiomyocytes attenuated lipopolysaccharide-induced cardiomyocyte inflammation and apoptosis. Further investigations into the molecular mechanisms underlying these effects showed that HMGA1 promoted lipopolysaccharide-induced mitochondrial-dependent cardiomyocyte apoptosis. The study reveals that HMGA1 worsens myocardial inflammation and apoptosis in response to lipopolysaccharide treatment. In this paper, group consensus is investigated for a class of nonlinear multi-agent systems suffered from the DoS attacks. Firstly, a first-order nonlinear multi-agent system is constructed, which is divided into M subsystems and each subsystem has an unique leader. Then a protocol is proposed and a Lyapunov function candidate is chosen. By means of the stability theory, a sufficient criterion, which involves the duration of DoS attacks, coupling strength and control gain, is obtained for achieving group consensus in first-order system. That is, the nodes in each subsystem can track the leader of that group. Furthermore, the result is extended to nonlinear second-order multi-agent systems and the controller is also improved to obtain sufficient conditions for group consensus. Additionally, the lower bounds of the coupling strength and average interval of DoS attacks can be determined from the obtained sufficient conditions. Objectives: To explore individual weight of cardiac magnetic resonance (CMR) metrics to predict mid-term outcomes in patients with dilated cardiomyopathy (DCM), and develop a risk algorithm for mid-term outcome based on CMR biomarkers. Materials and methods: Patients with DCM who underwent CMR imaging were prospectively enrolled in this study. The primary endpoint was a composite of heart failure (HF) death, sudden cardiac death (SCD), aborted SCD, and heart transplantation. Results: A total of 407 patients (age 48.1 ± 13.8 years, 331 men) were included in the final analysis. During a median follow-up of 21.7 months, 63 patients reached the primary endpoint. NYHA class III/IV (HR = 2.347 [1.073–5.133], p = 0.033), left ventricular ejection fraction (HR = 0.940 [0.909–0.973], p < 0.001), late gadolinium enhancement (LGE) > 0.9% and ≤ 6.6% (HR = 3.559 [1.020–12.412], p = 0.046), LGE > 6.6% (HR = 6.028 [1.814–20.038], p = 0.003), and mean extracellular volume (ECV) fraction ≥ 32.8% (HR = 5.922 [2.566–13.665], p < 0.001) had a significant prognostic association with the primary endpoints (C-statistic: 0.853 [0.810–0.896]). Competing risk regression analyses showed that patients with mean ECV fraction ≥ 32.8%, LGE ≥ 5.9%, global circumferential strain ≥ − 5.6%, or global longitudinal strain ≥ − 7.3% had significantly shorter event-free survival due to HF death and heart transplantation. Patients with mean ECV fraction ≥ 32.8% and LGE ≥ 5.9% had significantly shorter event-free survival due to SCD or aborted SCD. Conclusion: ECV fraction may be the best independently risk factor for the mid-term outcomes in patients with DCM, surpassing LVEF and LGE. LGE has a better prognostic value than other CMR metrics for SCD and aborted SCD. The risk stratification model we developed may be a promising non-invasive tool for decision-making and prognosis. Clinical relevance statement: "One-stop" assessment of cardiac function and myocardial characterization using cardiac magnetic resonance might improve risk stratification of patients with DCM. In this prospective study, we propose a novel risk algorithm in DCM including NYHA functional class, LVEF, LGE, and ECV. Polyimide aerogel fibers (PAFs) have good thermal insulation due to their low density and ultra-high porosity. In this work, in order to further improve the temperature regulation performance of PAFs in extremely cold environments, a novel polyimide/CsxWO3 composite aerogel fiber (PCAF) was prepared by the freeze-spinning technology. The as-prepared PCAFs exhibit lightweight, high-strength and thermal insulation properties. Particularly, the outstanding near-infrared absorption of CsxWO3 could endow the PCAF textiles with excellent photothermal conversion and radiative heating under solar irradiation in cold environments. It was clarified that the photothermal conversion efficiency of the PCAF-8 can be increased by about 3 times compared with the pure PAF after exposure to infrared radiation for 100 s. This result indicates that the PCAFs possess excellent self-heating and photothermal temperature regulation performance, which are significantly different from PAF. The thermal insulation and self-heating of the PCAFs due to their porous structure and photothermal conversion can synergistically keep warm and inhibit human heat loss in the cold, reaching a kind of dual-channel thermal management mode. In addition, the CsxWO3 component with visible light transmission is helpful to maintaining the original color of the fiber, avoiding the dull and unappealing color of conventional photothermal coatings. Therefore, this work provides a new strategy for outdoor personal thermal management in cold weather, making PCAFs a promising solution for extreme weather applications beyond traditional PAFs. The widespread used organoarsenicals have drawn attention for decades due to their potential environment risks. In this study, a heterogeneous system of goethite/oxalate irradiated using UVA light (λ = 365 nm) was applied for the removal of ASA, a kind of organoarsenicals used in animal feeding operations as additives, from the aqueous phase through photodegradation. Results showed that the presence of 5 mM of oxalate significantly enhanced the photodegradation efficiency of ASA in the 0.1 g/L of goethite suspended system from 28 to ~100% within 180 min reaction at pH 5. Acid conditions favored the photoreaction rate, compared with neutral and basic conditions. This reaction process was also influenced by the initial concentration of oxalate and ASA. Furthermore, the mechanism study was conducted by quenching experiments and revealed the important roles of ·OH in the degradation of ASA in the goethite/oxalate/UVA system. By analyzing the reaction products, both inorganic arsenic (As(III) and As(V)) and ammonia were detected during the photodegradation of ASA. These findings help to gain a better understanding of the geochemical behavior of ASA in surface water and can also provide a potential treatment method for the organoarsenicals contaminated water. The manufacturing technique known as investment casting has found extensive application in producing metal components featuring intricate geometries. The production efficiency of the wax patterns is an essential issue in the investment casting industry, especially for the mass production of wax patterns. A conformal cooling channel (CCC) performs the rapid uniform cooling process for injection molding. However, the significant pressure drop along the cooling channels is a distinct disadvantage of CCC. In this study, an innovative waterfall cooling channel (WCC) was proposed and implemented. The WCC cools the injected products by surface contact, replacing the conventional line contact to cool the injected products. The WCC was optimized using Moldex3D simulation software. Rapid tools with two kinds of cooling channels were designed and implemented. The cooling time of the molded part was investigated using a low-pressure wax injection molding machine. Considering a water cup characterized by a mouth diameter of 70 mm, a height of 60 mm, and a thickness of 2 mm, the experimental results confirmed that the use of WCC can save the cooling time of the product by about 265 s compared with the CCC. This result shows that the WCC can increase cooling efficiency by approximately 17.47% compared with conventional CCC. [ABSTRACT FROM AUTHOR]

Neuroinflammation caused by microglia in the central nervous system (CNS) is observed after myocardial infarction (MI). However, the inflammatory response mechanism remains unclear. BuChang Naoxintong capsule (NXT) is a Chinese medicine for treating ischemic cardio-cerebrovascular diseases, requiring more studies to understand the pharmacodynamic mechanism. Permanent ligation of the left anterior descending coronary artery (LAD) was performed in rats. Additionally, histopathological staining in the left ventricular (LV) and immunofluorescence within the brain cortex after 1 d and 7 d of MI were performed to determine the NXT pharmacodynamic action and best administration dosage. Proteomics helped obtain the essential proteins related to neuroinflammation and MI in the heart and brain tissue after 7 d of MI. Based on TTC, HE, Masson, and immunofluorescence staining results of CD206 and IBA-1, NXT demonstrated a better pharmacodynamic action towards myocardial injury and neuroinflammation after 7 d of MI. Moreover, the human equivalent dosage of NXT (220 mg/kg) became the best administration dose. The proteome bioinformatics analysis in the LV and brain cortex was performed. Thus, the elongation of very long-chain fatty acids protein 5 (ELOVL5) and ATP-binding cassette subfamily G member 4 (ABCG4) became critical proteins related to MI and neuroinflammation. The western blotting results indicated that ABCG4 expression possessed the same trend as the proteomics results. The auto-dock results revealed that ABCG4 had a good binding ability with Ferulic acid, Paeoniflorin, and Tanshinone II A, the key ingredients of NXT. The cellular thermal shift assay results demonstrated that ABCG4 showed better thermal stability post-NXT treatment. NXT can improve myocardial injury, such as heart infarct size, pathological injury, myocardial fibrosis, and inflammatory cell infiltration. Additionally, brain neuroinflammation induced by microglia after MI affects the expression and structure of ABCG4. Thus, ABCG4 could be the key protein associated with MI and neuroinflammation. Background: Previous studies have observed elevated myeloid cells in the peripheral blood of patients with Parkinson's disease (PD), but the causal relationship between them remains to be elucidated. We investigated whether there is a causal relationship between different subtypes of peripheral blood myeloid cells and PD using Mendelian randomization (MR) combined with bioinformatics analysis. Exploring the etiology of PD from the perspective of genetics can remove confounding factors and provide a more reliable theoretical basis for elucidating the pathogenesis of PD. Methods: Comprehensive two-sample MR analysis and sensitivity analyses were conducted to explore the causal associations between 64 myeloid cell signatures and PD risk. The Venn diagram and protein-protein interaction network analysis of instrumental variables (IV) corresponding genes were used to further investigate the potential mechanism of myeloid cells influencing the pathogenesis of PD. Results: We investigated the impact of four immunophenotypes on the risk of PD, including Im MDSC% CD33dim HLA DR− CD66b− (relative count), CD33dim HLA DR+ CD11b+% CD33dim HLA DR+ (relative count), and CD11b on Mo MDSC (MFI) and CD11b on CD33br HLA DR+ CD14dim (MFI), while an immunophenotype's protective effect on PD was observed CD45 on Im MDSC (MFI). The results of bioinformatics analysis showed that CD33, NTRK2, PLD2, GRIK2 and RELN had protein interactions with the risk genes of PD. Conclusions: Our study has demonstrated a close genetic correlation between different subtypes of myeloid cells and PD, providing guidance for early identification and immunotherapeutic development in patients with PD. Variational inequalities (VIs) have become a general framework for efficient problems in various fields such as optimal control and nonlinear programming. Some real problems in application areas such as signal processing and network resource allocation can be transformed into VIs. VIs have been frequently solved using neurodynamic approaches, which have good performance and low computational cost. In this paper, two novel projection neural networks (PNNs) with impulsive effects are presented to deal with VIs. Based on the impulsive system theory and PNN, an impulsive projection neural network (IPNN) and its modified version are proposed. Then the stability properties of the presented IPNN and modified impulsive projection neural network (MIPNN) are analyzed by Lyapunov functions and impulsive system theory. VIs can be addressed with the proposed IPNN and MIPNN. Furthermore, two PNNs are applied to sparse signal reconstruction. It is also shown that the solutions of the proposed IPNN and MIPNN converge to the solution of the corresponding VIs. Meanwhile, sparse signals can also be accurately reconstructed by IPNN and MIPNN. Compared with the classical neural network, the newly designed IPNN and MIPNN have an improvement in the convergence rates because of the introduction of impulsive effects. This study systematically investigated the pollution levels and migration trends of PBDEs in soils and plants around engineering plastics factory, and identified the ecological risks of PBDEs in the environment around typical pollution sources.The results showed that 13 kinds of PBDEs were widely detected in the surrounding areas, and the concentration level was higher than the general environmental pollution level. The total PBDE concentrations (∑13PBDEs) in soils ranged from 14.6 to 278.4 ng/g dry weight (dw), and in plants ranged from 11.5 to 176 ng/g dw. Both soil and plant samples showed that BDE-209 was the most important congener, the pollution level in soil and plant was similar, and the composition of PBDEs congener was similar. In the soil column (50 cm), the radial migration of PBDEs was mainly concentrated in the 0–30 cm section. Except for BDE-66, which was mainly located in the 20–30 cm soil layer, the concentration of PBDEs was the highest in the 0–10 cm region. Furthermore, the environmental risks of PBDEs in soil and plants were evaluated by hazard quotient method, and the HQ values were all < 1, which did not exhibit any ecological risk. Corms serve as the primary mode of reproduction for saffron, which growth state determines the yield and quality of saffron under field cultivation conditions. In this study, fourteen rhizobacteria were isolated from the rhizosphere soil of C. sativus. The findings of this study indicate that all of the rhizobacterial isolates have the capacity to synthesize indole-3-acetic acid and gibberellic acid. Among the 14 rhizobacterial isolates, four isolates exhibited the capability to solubilize phosphate, six isolates showed nitrogen fixation ability, and four isolates were identified as effective producers of siderophore. Only one isolate exhibited the capability to degrade 1-aminocyclopropane-1-carboxylate. As indicated by the findings from the field trials, it was observed that SR144 (Paenibacillus silvae), SR163 (Bacillus idriensis), SR269 (Pseudomonas sp.), SR307 (Stenotrophomonas sp.), and SR383 (Stenotrophomonas maltophilia) had a positive impact on the fresh weight of corms and leaf length of C. sativus. Additionally, a reduction in the malondialdehyde content was observed, accompanied by an elevation in the antioxidant enzyme system in corms. Further research indicates that SR307 and SR383 cause an increase in the total bacterial count, resulting in elevated levels of microbial biomass carbon and nitrogen in rhizosphere soil, and the levels of soil sucrase, urease, dehydrogenase, catalase, and alkaline phosphatase activities were also increased. Only SR383 exhibits an increase in acid phosphatase activity. This study investigated the effect of rapamycin alone and in combination with chemotherapy (doxorubicin and cytarabine) on AML. Human acute monocytic leukemia cell line SHI-1 and NPG AML model mice created by intravenous injection of SHI-1 cell were treated with rapamycin, chemotherapy, or rapamycin plus chemotherapy. Analysis by cell counting kit-8, western blot, flow cytometry, and immunohistochemistry was performed, and results suggested that both rapamycin and chemotherapy inhibited proliferation of SHI-1 cells both in vitro and in vivo, suppressed neoplasm growth in vivo, and promoted survival of NPG AML mice. The antitumor effect of rapamycin plus chemotherapy was better than that of rapamycin alone and chemotherapy alone. In addition, western blot results demonstrated that rapamycin inhibited the phosphorylation of mTOR downstream targets 4EBP1 and S6K1 in SHI-1 cells, and increased the pro-apoptosis-related protein Bax and autophagy-associated proteins Beclin-1, LC3B-II, and ATG5 while reducing the anti-apoptosis-related protein Bcl-2. In conclusion, the results of this study indicate that rapamycin acts synergistically with doxorubicin and cytarabine in AML treatment, and its underlying mechanism might be associated with mTORC1 pathway-mediated apoptosis and autophagy. A core issue in the interdisciplinary study of human morality is its ontogeny in diverse cultures, but systematic, naturalistic data in specific cultural contexts are rare to find. This study conducts a novel analysis of 213 children's socio-moral behavior in a historical, non-Western, rural setting, based on a unique dataset of naturalistic observations from the first field research on Han Chinese children. Using multilevel multinomial modeling, we examined a range of proactive behaviors in 0-to-12-year-old children's peer cooperation and conflict in an entire community in postwar Taiwan. We modeled the effects of age, sex, kinship, and behavioral roles, and revealed complex interactions between these four variables in shaping children's moral development. We discovered linkages between coercive and non-coercive behaviors as children strategically negotiated leadership dynamics. We identified connections between prosocial and aggressive behaviors, illuminating the nuances of morality in real life. Our analysis also revealed gendered patterns and age-related trends that deviated from cultural norms and contradicted popular assumptions about Chinese family values. These findings highlight the importance of naturalistic observations in cultural contexts for understanding how we become moral persons. Nonlinear optical signal processing (NOSP) has the potential to significantly improve the throughput, flexibility, and cost-efficiency of optical communication networks by exploiting the intrinsically ultrafast optical nonlinear wave mixing. It can support digital signal processing speeds of up to terabits per second, far exceeding the line rate of the electronic counterpart. In NOSP, high-intensity light fields are used to generate nonlinear optical responses, which can be used to process optical signals. Great efforts have been devoted to developing new materials and structures for NOSP. However, one of the challenges in implementing NOSP is the requirement of high-intensity light fields, which is difficult to generate and maintain. This has been a major roadblock to realize practical NOSP systems for high-speed, high-capacity optical communications. Here, we propose using a parity-time (PT) symmetric microresonator system to significantly enhance the light intensity and support high-speed operation by relieving the bandwidth-efficiency limit imposed on conventional single resonator systems. The design concept is the co-existence of a PT symmetry broken regime for a narrow-linewidth pump wave and near-exceptional point operation for broadband signal and idler waves. This enables us to achieve a new NOSP system with two orders of magnitude improvement in efficiency compared to a single resonator. With a highly nonlinear AlGaAs-on-Insulator platform, we demonstrate an NOSP at a data rate approaching 40 gigabits per second with a record low pump power of one milliwatt. These findings pave the way for the development of fully chip-scale NOSP devices with pump light sources integrated together, potentially leading to a wide range of applications in optical communication networks and classical or quantum computation. The study investigates the association of coffee consumption and odds of osteoporosis/osteopenia among individuals older than 50 years in the United States. In NHANES 2005–2014, drinking ≤ 2 cups(16 oz) of coffee per day can reduce the risk of osteoporosis/osteopenia at the femoral neck and lumbar spine in US adults. Previous epidemiological studies revealed that daily coffee intake reduced the incidence of a cluster of metabolic diseases, however, the link between coffee consumption and prevalence of osteoporosis/osteopenia still remain inconclusive and awaits further confirmation. Based on data collection from 2005 to 2014 survey cycles, National Health and Nutrition Examination Survey (NHANES), a sample size of 8789 participants aged 50 and above completing two nonconsecutive 24-h dietary recalls were eventually enrolled for analysis. Associations between coffee intake and BMD were assessed. A lower odds of having femoral neck osteopenia/osteoporosis (FOO) was observed in participants with moderate intake of coffee (≤ 2 cups per day), rather than other beverages (OR 0.83; 95% CI, 0.72–0.95; p = 0.01). Moreover, significant associations existed between daily caffeine intake and both FOO and lumbar-spine osteopenia/osteoporosis (LOO). Even after adjusting for decaffeinated coffee, tea, sugar-sweetened beverages (SSBs), and coffee consumption, osteopenia and osteoporosis the odds remained lower at both femoral and neck levels. Our data suggest moderate habitual coffee intake (≤ 2 cups coffee/day) would have protective effects against osteoporosis/osteopenia of femoral neck and spine, among US adults over the age of 50. Background: Metadherin (MTDH) and ubiquitin specific protease 7 (USP7) have been identified to involve in the tumorigenesis of cervical cancer (CC). USP7 is one of the deubiquitinating enzymes. Here, this study aimed to explore whether USP7 affected CC progression via interacting with MTDH and regulating its stability via deubiquitination. Methods: qRT-PCR and western blotting assays detected the levels of genes and proteins. Functional analysis was conducted using 5-ethynyl-2’-deoxyuridine (EdU), flow cytometry, transwell, and tube formation assays, respectively. Proteins between USP7 and MTDH were identified by co-immunoprecipitation assay. A mouse xenograft model was established for in vivo analysis. Results: MTDH was highly expressed in CC tissues and cells, silencing of MTDH suppressed CC cell proliferation, migration, invasion, angiogenesis, and macrophage M2 polarization. Mechanistically, USP7 directly bound to MTDH, and maintained its stability by removing ubiquitination on MTDH. CC tissues and cells showed high USP7 expression, and USP7 knockdown also inhibited CC cell proliferation, migration, invasion, angiogenesis and macrophage M2 polarization, and these effects mediated by USP7 knockdown were reversed by MTDH overexpression. Moreover, USP7 knockdown impeded CC growth in vivo by regulating MTDH. Conclusion: Collectively, USP7 promoted CC cell proliferation, migration, invasion, angiogenesis, and macrophage M2 polarization in vitro, as well as tumor growth in vivo by regulating MTDH. Background: Previous studies have shown that visible light (VL), especially blue light (BL), could cause significant skin damage. With the emergence of VL protection products, a harmonization of light protection methods has been proposed, but it has not been widely applied in the Chinese population. Objective: Based on this framework, we propose an accurate and simplified method to evaluate the efficacy of BL photoprotection for the Chinese population. Methods: All subjects (n = 30) were irradiated daily using a blue LED light for four consecutive days. Each irradiation dose was 3/4 MPPD (minimum persistent pigmentation darkening). The skin pigmentation parameters, including L*, M, and ITA°, were recorded. We proposed the blue light protection factor (BPF) metric based on the skin pigmentation parameters to evaluate the anti-blue light efficacies of different products. Results: We found that the level of pigmentation rose progressively and linearly as blue light exposure increased. We proposed a metric, BPF, to reflect the anti-blue light efficacy of products based on the linear changes in skin pigment characteristics following daily BL exposure. Moreover, we discovered that the BPF metric could clearly distinguish the anti-blue light efficacies between two products and the control group, suggesting that BPF is an efficient and simple-to-use metric for anti-blue light evaluation. Conclusion: Our study proposed an accurate and simplified method with an easy-to-use metric, BPF, to accurately characterize the anti-blue light efficacies of cosmetic products, providing support for further development of anti-blue light cosmetics. The COVID-19 pandemic was a stressful event for all. It threatened people's physical and mental health. Previous studies have found that resilience can help people deal with stress and difficulties more effectively and prevent them from mental health problems. Coping style mediates the relationship between resilience and psychological well-being. Given the novelty and recency of the COVID-19, it is unknown whether resilience can also protect individuals in new difficulties, and whether the previous effective coping styles can be applied to new situations. The current study aimed to explore the influence mechanism of college students' resilience on negative stress response during the pandemic, and the chain mediation of coping style and positive adaptive response. A questionnaire survey was conducted among 910 college students (Mage = 20.58 years; 41.1% men, 58.9% women) studying in Shanghai by stratified random cluster sampling. At that time, they were in 27 different provinces and cities because of the COVID-19 pandemic lockdown. The research tools included Connor-Davidson Resilience Scale (CD-RISC), Simplified Coping Style Questionnaire (SCSQ), Positive Adaptive Response Questionnaire (PARQ), and Negative Stress Reaction Questionnaire (NSRQ). The results showed that: (1) College students' resilience, coping style, positive adaptive response and negative stress reaction were significantly correlated with each other; (2) Resilience could not directly affect negative stress reaction, but it could affect negative stress reaction through the chain mediation of coping style and positive adaptive response. These findings suggest that resilience enhancement intervention programs need to focus on the cultivation of individual positive coping style; In response to major emergencies such as the COVID-19 pandemic, officials can help people reduce negative stress reaction by disseminating positive coping strategies through the media. This article, published in Chemistry of Natural Compounds, discusses the production of isoflavone metabolites and their microbial transformation by a soil-derived Saccharopolyspora antimicrobica DSMZ45119. Actinomycetes, specifically the Saccharopolyspora genus, have been a focus of research due to their production of biologically active secondary metabolites. The study implemented the one strain many compounds (OSMAC) approach to cultivate DSMZ45119 on six different media and analyzed the resulting metabolites. The researchers isolated and characterized six known isoflavone aglycones and glycosides from the strain. The article also discusses the antimicrobial, cytotoxic, immunosuppressive, and antioxidant activities of these compounds. Background: Semi-enclosed bays are important links in the material cycle and energy flow between terrestrial and marine ecosystems. They are also areas of intense human activities and heavily influenced by such activities as aquaculture, industrial and agricultural wastewater discharge. The rate of biodiversity loss and changes in ecosystems have prompted research into the relationship between species diversity and ecosystem functioning. Studies have shown that application of functional diversity indices is useful for assessing the status of ecosystem functioning. We quantitatively sampled macrofauna in a semi-enclosed bay in four seasons and analyzed the relationship between species, functional diversity and secondary production, biomass and feeding functional groups of macrofauna. Results: The annual secondary production was 325.01 kJ m−2 year−1. Detritivorous, carnivorous and planktophagous feeders were the main functional groups of macrofauna. Differences in the spatial–temporal distribution of functional groups were influenced by Ruditapes philippinarum and Hemileucon bidentatus. Functional richness had significant negative correlations with macrofaunal biomass and secondary production. Functional divergence, functional dispersion and Rao's quadratic entropy had highly significant negative correlations with macrofaunal biomass and secondary production. Conclusion: The results showed that high overlap of ecological niches can increase competition for habitat resources, leading to a decline in biomass and secondary production. In addition, aquaculture could promote the use of habitat resources to some extent, while it could increase competition for ecosystem resources (including habitat resources, atmospheric resources, water resources, etc.). Functional diversity is a good indicator for the ecosystem functioning and the competition status for habitat resources, which can provide insights into the current state of ecosystem function. Myeloid cells are abundant and plastic immune cell subsets in the liver, to which pro-tumorigenic, inflammatory and immunosuppressive roles have been assigned in the course of tumorigenesis. Yet several aspects underlying their dynamic alterations in hepatocellular carcinoma (HCC) progression remain elusive, including the impact of distinct genetic mutations in shaping a cancer-permissive tumor microenvironment (TME). Here, in newly generated, clinically-relevant somatic female HCC mouse models, we identify cancer genetics' specific and stage-dependent alterations of the liver TME associated with distinct histopathological and malignant HCC features. Mitogen-activated protein kinase (MAPK)-activated, NrasG12D-driven tumors exhibit a mixed phenotype of prominent inflammation and immunosuppression in a T cell-excluded TME. Mechanistically, we report a NrasG12D cancer cell-driven, MEK-ERK1/2-SP1-dependent GM-CSF secretion enabling the accumulation of immunosuppressive and proinflammatory monocyte-derived Ly6Clow cells. GM-CSF blockade curbs the accumulation of these cells, reduces inflammation, induces cancer cell death and prolongs animal survival. Furthermore, GM-CSF neutralization synergizes with a vascular endothelial growth factor (VEGF) inhibitor to restrain HCC outgrowth. These findings underscore the profound alterations of the myeloid TME consequential to MAPK pathway activation intensity and the potential of GM-CSF inhibition as a myeloid-centric therapy tailored to subsets of HCC patients. Distinct genetic mutations can shape the tumor immune microenvironment. Myocardial infarction (MI) induces neuroinflammation indirectly, chronic neuroinflammation may cause neurodegenerative diseases. Changes in the proteomics of heart and brain tissue after MI may shed new light on the mechanisms involved in neuroinflammation. This study explored brain and heart protein changes after MI with a data-independent acquisition (DIA) mode proteomics approach. Permanent ligation of the left anterior descending coronary artery (LAD) was performed in the heart of rats, and the immunofluorescence of microglia in the brain cortex was performed at 1d, 3d, 5d, and 7d after MI to detect the neuroinflammation. Then proteomics was accomplished to obtain the vital proteins in the heart and brain post-MI. The results show that the number of microglia was significantly increased in the Model-1d group, the Model-3d group, the Model-5d group, and the Model-7d group compared to the Sham group. Various proteins were obtained through DIA proteomics. Linking to key targets of brain disease, 14 proteins were obtained in the brain cortex. In this study we aimed to investigate the prevalence of SARS-CoV-2 infection in psoriasis patients, and outcomes of SARS-CoV-2 infection and associated risk factors. A cross-sectional survey was conducted from February 2023 to March 2023. Information was obtained with online questionnaire about psoriasis patients on demographic characteristics, clinical characteristics, SARS-CoV-2 infection and outcomes, vaccination, and routine protection against COVID-19. Logistic regression analysis was used to explore risk factors with SARS-CoV-2 infection and exacerbation of psoriasis. A total of 613 participants were recruited. 516 (84.2%) were infected, and associated factors were sex, working status, routine protection against COVID-19, COVID-19 vaccination, impaired nail, infection exacerbate psoriasis, and severity of psoriasis. Among the patients infected with SARS-CoV-2, 30 (5.8%) required hospitalization, 122 (23.6%) had psoriasis exacerbation due to SARS-CoV-2 infection, and associated factors were subtype of psoriasis, discontinuation of psoriasis treatment during SARS-CoV-2 infection, response following COVID-19 vaccination, and severity of psoriasis. Booster dose vaccination contributed a low probability of COVID-19 sequelae. COVID-19 vaccine's effectiveness was unsatisfactory, while booster dose vaccination reduced the occurrence of COVID-19 sequelae in psoriasis patients of Southwest China. Patients treated with psoriasis shown to be safe, without a higher incidence of SARS-CoV-2 infection or COVID-19hospitalization compared to untreated patients. The p–n type Sm2WO6-Co9S8 heterojunction with two-dimensional coupling interface was successfully synthesized by electrostatic self-assembly. The unique rhombic morphology of Sm2WO6 and Co9S8 nanoparticles also effectively improve the light utilization efficiency. The intervention of Co9S8 enhances the visible light absorption capacity of m2WO6/Co9S8. The Sm2WO6/Co9S8 greatly increases the specific surface area and provides a rich reaction site. Recombination of photogenerated carriers in Sm2WO6/Co9S8 heterojunctions is effectively inhibited. The p–n type Sm2WO6-Co9S8 heterojunction results show that the optimized Sm2WO6/Co9S8 heterojunction exhibits significant hydrogen evolution activity, and the optimal Sm2WO6/Co9S8-25 composite has 117.5 μmol hydrogen evolution activity, it's close to four times Co9S8, close to 143 times Sm2WO6. The stability of the composite Sm2WO6/Co9S8 is the better. Under the built-in electric field induced by p–n heterojunction, using of p–n heterojunction interactions and electron transport modes, the Sm2WO6/Co9S8 can effectively achieve the remarkable effect of its internal carrier separation. With the increasing scarcity of resources, the search for green and available energy has become an urgent need. Among them, photocatalytic decomposition of water to hydrogen production has been widely studied. According to zeta potential and XRD characterization, NiCo2O4 and CuS were successfully reassembled by electrostatic self-assembly to construct S-scheme heterojunction induced by built-in electric site, which can accelerate the reaction. At the same time, under simulated visible light irradiation, the composite catalyst can rapidly decompose aqueous triethanolamine solution to produce hydrogen. The composite catalyst has high photocatalytic activity and shows a durable and efficient hydrogen generation effect. By studying the photogenerated carrier separation and transfer of the catalyst, the results show that the composite catalyst can accelerate the photogenerated carrier separation and transfer, so that it can participate in the photocatalytic reaction faster. Room temperature sodium-sulfur (Na-S) batteries, known for their high energy density and low cost, are one of the most promising next-generation energy storage systems. However, the polysulfide shuttling and uncontrollable Na dendrite growth as well as safety issues caused by the use of organic liquid electrolytes in Na-S cells, have severely hindered their commercialization. Solid-state electrolytes instead of liquid electrolytes are considered to be the most direct and effective solution to solve the above problems. However, its practical application is still greatly challenged due to the poor interfacial compatibility between the all-solid-state electrolytes and the anode/cathode, ionic conductivity, and the shuttle effect caused by the presence of liquid phase in the quasi-solid-state electrolytes. This paper presents a comprehensive review of solid-state Na-S batteries from the perspective of regulating interfacial compatibility and improving ionic conductivity as well as suppressing polysulfide shuttle. According to different components, solid-state electrolytes were divided into five categories: solid inorganic electrolytes, solid polymer electrolytes, polymer/inorganic solid hybrid electrolytes, gel polymer electrolytes, and liquid–solid inorganic hybrid electrolytes. Sodium metal is one of the ideal anodes for high-performance rechargeable batteries because of its high specific capacity (~ 1166 mAh·g−1), low reduction potential (−2.71 V compared to standard hydrogen electrodes), and low cost. However, the unstable solid electrolyte interphase, uncontrolled dendrite growth, and inevitable volume expansion hinder the practical application of sodium metal anodes. At present, many strategies have been developed to achieve stable sodium metal anodes. Here, we systematically summarize the latest strategies adopted in interface engineering, current collector design, and the emerging methods to improve the reaction kinetics of sodium deposition processes. First, the strategies of constructing protective layers are reviewed, including inorganic, organic, and mixed protective layers through electrolyte additives or pretreatments. Then, the classification of metal-based, carbon-based, and composite porous frames is discussed, including their function in reducing local deposition current density and the effect of introducing sodiophilic sites. Third, the recent progress of alloys, nanoparticles, and single atoms in improving Na deposition kinetics is systematically reviewed. Context: Adipose triglyceride lipase (ATGL), a key enzyme responsible for lipolysis, catalyzes the first step of lipolysis and converts triglycerides to diacylglycerols and free fatty acids (FFA). Our previous work suggested that phillyrin treatment improves insulin resistance in HFD-fed mice, which was associated with ATGL inhibition. In this study, using docking simulation, we explored the binding pose of phillyrin and atglistatin (a mouse ATGL inhibitor) to ATGL in mouse. From the docking results, the interactions with Ser47 and Asp166 were speculated to have caused phillyrin to inhibit ATGL in mice. Further, molecular dynamics simulation of 100 ns and MM-GBSA were conducted for the protein–ligand complex, which indicated that the system was stable and that phillyrin displayed a better affinity to ATGL than did atglistatin throughout the simulation period. Moreover, the results of pharmacological validation were consistent with those of the in silico simulations. In summary, our study illustrates the potential of molecular docking to accurately predict the binding protein produced by AlphaFold and suggests that phillyrin is a potential small molecule that targets and inhibits ATGL enzymatic activity. Methods: The ATGL-predicted protein structure, verified by PROCHECK, was determined using AlphaFold. Molecular docking, molecular dynamics simulation, and prime molecular mechanic–generalized born surface area were performed using LigPrep, Desmond, and prime MM-GBSA modules of Schrödinger software release 2021-2, respectively. Background: The incidence of comorbidity between type 2 diabetes mellitus (T2DM) and metabolic-associated fatty liver disease (MAFLD) is high, and patients tend to be younger. When people develop metabolic diseases such as T2DM and MAFLD, the original homeostasis of the gut microbiota in the body is disrupted, and gut flora drift occurs. This study investigated the relationship between the number of gut flora and MAFLD in young-onset T2DM. Methods: This retrospective study analyzed 44 adolescent T2DM patients who were divided into a non-MAFLD group and a MAFLD group. Anthropometric measurements, clinical and biochemical markers, inflammatory markers, thyroid function assessments, and stool specimens were collected. Real-time PCR was performed to quantify several important gut flora constituents at the genus level. Student's t-test and the chi-square test were applied for group comparisons, and binary regression models were used to explore the relationship between gut flora and MAFLD in young-onset T2DM. Results: Among the 44 subjects, 26 (59.1%) were diagnosed with MAFLD, and 18 (40.9%) were not. Compared with the non-MAFLD group, body mass index (BMI), abdominal circumference, and levels of blood uric acid and thyroid stimulating hormone (TSH) in the MAFLD group were significantly increased, and age level and high-density lipoprotein cholesterol (HDL-C) were significantly decreased (p < 0.05). Compared with the non-MAFLD group, the abundance of Faecalibacterium prausnitzii and Bifidobacterium in the MAFLD group was significantly reduced, and the abundance of Enterococcus and Lactobacillus was significantly increased (p < 0.05). In the multivariate regression analysis, Faecalibacterium prausnitzii and Bifidobacterium were independent protective factors for MAFLD in young-onset T2DM, after excluding confounding factors. Conclusion: In young-onset T2DM, there was a difference in gut flora between patients with MAFLD and those without MAFLD.
