Comparative studies of aging are a promising approach to identifying general properties of and processes leading to aging. While to date, many comparative studies of aging in animals have focused on relatively narrow species groups, methodological innovations now allow for studies that include evolutionary distant species. However, comparative studies of aging across a wide range of species that have distinct life histories introduce additional challenges in experimental design. Here, we discuss these challenges, highlight the most pressing problems that need to be solved, and provide suggestions based on current approaches to successfully carry out comparative aging studies across the animal kingdom. Exceptionally long-lived species, including many bats, rarely show overt signs of aging, making it difficult to determine why species differ in lifespan. Here, we use DNA methylation (DNAm) profiles from 712 known-age bats, representing 26 species, to identify epigenetic changes associated with age and longevity. We demonstrate that DNAm accurately predicts chronological age. Across species, longevity is negatively associated with the rate of DNAm change at age-associated sites. Furthermore, analysis of several bat genomes reveals that hypermethylated age- and longevity-associated sites are disproportionately located in promoter regions of key transcription factors (TF) and enriched for histone and chromatin features associated with transcriptional regulation. Predicted TF binding site motifs and enrichment analyses indicate that age-related methylation change is influenced by developmental processes, while longevity-related DNAm change is associated with innate immunity or tumorigenesis genes, suggesting that bat longevity results from augmented immune response and cancer suppression. Extra-group paternity, in which offspring are sired by a male outside the breeding group, may alter the distribution of reproductive success in a population, thus affecting the opportunity for sexual selection. Both inter- and intraspecific studies have focused largely on mating systems in which females choose their social mates, and less is known about extra-group paternity in polygynous systems in which social mates are largely dictated by male-male competition. In this study, we examine the frequency and distribution of extra-group paternity in a harem-forming bat, Phyllostomus hastatus. We find that despite aggressive harem defense, males are unable to fully monopolize reproduction within their harem and over 70% of harems contain extra-group offspring. Harem males in better body condition suffered less paternity loss, but we found no effect of male age or body size. Even though the age and size of individual females did not predict offspring paternity, we found a significant effect of age heterogeneity within the group. Harems composed of differently aged females were more likely to contain extra-group offspring. Our results not only provide evidence for the role of male defense in preventing extra-group paternity but also suggest that social group composition has consequences for male reproductive success. Significance statement: In polygynous societies, the ability to monopolize mating is critical to reproductive success. As the group size increases, defense often becomes more difficult, resulting in increased rates of extra-group paternity. We find that among greater spear-nosed bats, extra-group offspring occur in most harems, but the overall rate of extra-group paternity is relatively low despite their large harems (15–30 females). Variation in the rate of extra-group paternity between harems is explained by male body condition (i.e., relative mass) but not harem size. Additionally, the rate of extra-group paternity is not explained entirely by male attributes, as we find that age heterogeneity within the female group is a significant predictor of the extra-group paternity rate. Our results support the hypothesis that not only the physical condition of a male influences his ability to monopolize reproduction but also the group composition influences the female mating behavior. Chemical signals are ubiquitous, but often overlooked as potentially important for conveying information relevant for sexual selection. The male greater spear-nosed bat, Phyllostomus hastatus, possesses a sexually dimorphic gland on the chest that produces an odoriferous secretion. Here, we investigate the potential for this glandular secretion to act as a sexually selected olfactory signal by examining gland activity in and out of the mating season and determining if variation in its chemical composition reflects variation in male mating status or attributes of the individual. Based on gas chromatography-mass spectrometry (GC-MS) measurements of samples collected from wild bats roosting in caves in Trinidad, West Indies, we find that males that defend and roost with groups of females (harem holders) have significantly different chemical profiles from males found roosting in all male groups (bachelors). Additionally, profiles differed significantly among individuals. Taken together, these results suggest that this chemical signal has the potential to communicate both mating status and individual identity and thus could be used to mediate interactions among individuals within this harem-based social mating system. Alloparental care poses an evolutionary dilemma because effort is expended on non-filial offspring. Thus, instances of alloparental care have been attributed to either mistaken identity, (i.e., recognition errors) or active cooperation. In greater spear-nosed bats ( Phyllostomus hastatus), reproductive females roost together in stable long-term social groups in cave ceilings. Non-volant pups frequently fall from roost sites to the cave floor where they can die unless retrieved by an adult. In this study, we examined the function of adult female visits to non-filial young and tested whether visits were attributable to recognition errors or to cooperation. We found that females visited non-filial pups from their own social group more than expected. Females from different social groups attacked and sometimes killed pups, and male pups were attacked more frequently than female pups. Visits by group mates benefited fallen pups by reducing the likelihood of attack by females from other groups. In contrast to the mistaken identity hypothesis, we found that some females leave their own pups to approach and remain with group mates’ pups. We used microsatellite markers to estimate relatedness and test whether kinship could explain this alloparental care and found that females were unrelated to the pups they guard. We conclude that females who reside in highly stable social groups exhibit cooperative behavior that cannot be explained by kinship and is unlikely to be due to direct or generalized reciprocity. The cytological basis of X chromosome meiotic drive or sex ratio (SR) has been reported for several species of Drosophila but not for other species. Here we describe how sperm development in the stalk-eyed fly, Cyrtodiopsis whitei, influences progeny sex proportion, in order to determine if a common developmental mechanism could cause meiotic drive in these distantly related taxa. Because age has been found to affect the degree of segregation distortion in some Drosophila, we tested flies from six to 26 weeks of age. We find that spermatocyst bundles in SR males frequently contain incompletely elongated spermatid nuclei independently of male age. Older males have, however, more spermatocyst bundles in their testes than younger males. Abnormal spermatid elongation affects male fertility since SR males produce 74% as many progeny per week as ST males. The proportion of spermatocyst bundles with improperly elongated spermatid nuclei explains 71% of the variation in progeny sex proportion. After reviewing the literature on sperm development and meiotic drive, we conclude that the cytological basis of meiotic drive in diopsids closely resembles Drosophila. Across species in both groups, the production of fertile males is associated with less than half of all spermatids not elongating normally in a spermatocyst bundle. Evolution of male weapons or status signals has been hypothesized to precede evolution of female mating preferences for those traits. We used staged male fights among three species of Malaysian stalk-eyed flies (Diptera: Diopsidae) to determine if elongated eye span, which is preferred by females in two sexually dimorphic species, influences contest outcome. Extreme sexual dimorphism, with large males possessing longer eye span than females, is shared by Cyrtodiopsis whitei and C. dalmanni. In contrast, C. quinqueguttata exhibits a more ancestral condition – short, sexually monomorphic eye stalks. Videotape analysis of 20-min paired contests revealed that males with larger eye span and body size won more fights in the dimorphic, but not monomorphic, species. To determine if males from the dimorphic species use eye span directly to resolve contests, we competed male C. dalmanni from lines that had undergone artificial selection for 30 generations to increase or decrease eye span. We found that eye span, independently of body size, determines contest outcome in selected-line males. Furthermore, in both dimorphic species, the average encounter duration declined as the eye span difference between contestants increased, as expected if males use eye span to assess opponent size. The number of encounters also increased with age in dimorphic, but not monomorphic, species. Selected-line males did not differ from outbred males in either fight duration or number of encounters. Variability at seven polymorphic allozyme loci and observations of dispersal and mating provide evidence for nonrandom genetic structure among adult female groups of the highly social bat, Desmodus rotundus. The average degree of relatedness, estimated by allelic correlations at each locus, within three and six groups of females is 0.018 (SE=0.013) and 0.032 (SE=0.023), respectively. Even though these estimates do not differ significantly from zero, a multivariate analysis of variance of individual allele frequencies reveals that three of six pairwise comparisons of groups reach significance. This genetic heterogeneity within a population does not lead to increased genetic subdivision between populations. Mean classificatory ability of the discriminant functions drops from 84% for assignment to group to 56% for assignment to population. This pattern of genetic variability is due to recruitment of female offspring into their natal groups and forced male dispersal. Occasional movements of unrelated females between groups lead to the formation of multiple matrilines within groups. Although males fight viciously for access to the top of preferred female roosting sites and top males mate preferentially with females in that roost, mean maximum paternity for top males is only 46%. Consequently, male mating success is sufficiently random to maintain gametic equilibria among all pairs of loci. Given an infant mortality of 54%, mean male tenure of 17 months, and a birth interval of 10 months, females are unlikely to be related through common male ancestors. In one group, the average degree of relatedness, derived from matrilineal pedigrees, is 0.11 (SD=0.17). Computer simulations of the growth of a group of female D. routundus show that the low level of relatedness within groups is expected even if the proportion of unrelated females allowed into a group decreases. Abstract: Among mammals, bats exhibit extreme variation in sociality, with some species living largely solitary lives while others form colonies of more than a million individuals. Some tropical species form groups during the day that persist throughout the year while many temperate species only gather into groups during hibernation or parturition. How groups form and then persist has now been described for a number of species, but the degree to which kinship explains patterns of association has never been quantified across species. Here, we use social network analysis and genetic data to determine the extent to which relatedness contributes to associations among individuals estimated from free-ranging animals across nine species from four families of bats. Network analysis reveals that all species show evidence of emergent social structure. Variation in the strength of the relationship between genetic relatedness and social association appears to be related to the degree of roost switching, i.e., species in which individuals change roosts frequently tend to exhibit higher levels of association among relatives. Sex-biased dispersal determines whether associations were between male or female relatives. The strength of associations among kin does not predict known occurrence of complex behaviors, such as dominance or various types of cooperation, indicating that kinship is not a prerequisite for social complexity in bats.Significance statement: The number of differentiated relationships has been proposed as a way to measure social complexity. Among primates, relationships can be differentiated on the basis of rank, age, kinship, or association. Application of this approach to other groups of mammals that vary in sociality could help reveal ecological, behavioral, or cognitive similarities and differences between species. As a first step toward this approach, we used social network analysis on long-term individual records and estimated relatedness using genetic markers for nine species of bats. We confirmed nonrandom emergent social structure in all species. Kinship was predictive of social association among individuals of the same sex in a few species, but largely independent of the occurrence of complex behaviors, such as dominance among males or cooperation among females.
