The stock structure of small pelagic fishes is difficult to determine due to their patchy distribution and complex movement patterns. We integrate genetic, morphological, otolith, growth, reproductive and fishery data collected over 60 years using a Stock Differentiation Index (SDI). The absence of strong separation (SDI > 0.66) of most adjacent sub-groups supports the hypothesis that sardine ( Sardinops sagax) in Australian waters is a meta-population, but with effective isolation of at least four stocks: south western coast (off Western Australia); Great Australian Bight and Spencer Gulf; Bass Strait and Port Phillip Bay (off Victoria and Tasmania); and eastern Australia. There is also evidence for sub-division of the stocks off Western Australia and the east coast. We examine age-related and inter-annual patterns of stock structure off South Australia and the east coast through integrated analysis of otolith chemistry and shape data. For the east coast, there were significant differences between northern and southern sub-groups for all three age cohorts examined. Fish were correctly classified to sampling region with a high degree of success (>80%), supporting the sub-division of the east coast stock suggested by the SDI. For South Australia, there were significant differences among two sub-groups for most cohorts examined across two sampling years. However, spatial discriminatory power was poor, with allocation success ranging from 48 to 64%. Results suggest that movements between the two South Australian sub-groups may vary among years, which is consistent with inconclusive SDI (0.5). Integrating historical data using a SDI is suitable for identifying fishery management units. Integrated analysis of otoliths from archival collections is useful for examining temporal variability in stock structure, which is also important for fisheries management. Climate change driven alterations in the distribution and abundance of marine species, and the timing of their life history events (phenology), are being reported around the globe. However, we have limited capacity to detect and predict these responses, even for comparatively well studied commercial fishery species. Fisheries provide significant socio-economic benefits for many coastal communities, and early warning of potential changes to fish stocks will provide managers and other stakeholders with the best opportunity to adapt to these impacts. Rapid assessment methods that can estimate the sensitivity of species to climate change in a wide range of contexts are needed. This study establishes an objective, flexible and cost effective framework for prioritising future ecological research and subsequent investment in adaptation responses in the face of resource constraints. We build on an ecological risk assessment framework to assess relative sensitivities of commercial species to climate change drivers, specifically in relation to their distribution, abundance and phenology, and demonstrate our approach using key species within the fast warming region of south-eastern Australia. Our approach has enabled fisheries managers to understand likely changes to fisheries under a range of climate change scenarios, highlighted critical research gaps and priorities, and assisted marine industries to identify adaptation strategies that maximise positive outcomes. Background: 4-( N-( S-glutathionylacetyl)amino) phenylarsenoxide (GSAO) is a water-soluble mitochondrial toxin that binds to adenine nucleotide translocase in the inner mitochondrial membrane, thereby targeting cell proliferation. This phase 1 study investigated safety, dose-limiting toxicities (DLTs), maximum tolerated dose (MTD) and pharmacokinetics (PK) of GSAO as a daily 1-h infusion for 5 days a week for 2 weeks in every three. Pharmacodynamics of GSAO was evaluated by dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) and circulating markers of angiogenesis. Methods: Patients with advanced solid tumours received GSAO in a dose-escalation trial according to a standard ‘3 + 3’ design that was guided by toxicity and, for the final dose escalation, by arsenic PK data. Results: A total of 34 patients were treated with GSAO across 9 dose levels (1.3–44.0 mg/m 2). Treatment was well tolerated with few adverse events. An additional three patients were enrolled at the 12.4 mg/m 2 dose level following a DLT of derangement of liver function tests (grade 4). At the 44.0 mg/m 2 dose level, two out of three patients had DLTs (reversible encephalopathy; paroxysmal atrial fibrillation). Conclusions: The MTD of GSAO was 22.0 mg/m 2/day. We analysed a 25-year time series of fishery catch and effort data, and age/size information for four large-bodied, native fish species to investigate the hypotheses that under conditions of reduced freshwater inflows and high fishing pressure: (1) the structure of fish assemblages in the lower Murray River system have changed, (2) species diversity of fishes has declined and (3) population age structures of large-bodied, late-maturing, native fish ( Macquaria ambigua, Argyrosomus japonicus, Rhombosolea tapirina and Acanthopagrus butcheri) have been reduced. Annual catches and effort in the lower Murray River system were stable for 25 years, but proportional contribution to the total catch from each of freshwater, estuarine and adjacent marine habitats, and the species within them varied. Fish assemblages generally differed between subsequent 5-year periods, with the exception of 1989-1993 when floods occurred in 4 out of 5 years, and the following 5-year period (1994-1998). Species richness declined steeply over 25 years in freshwater and estuarine habitat and species diversity (Hill's H) also declined after 2001 in estuarine habitat. Species with rapid growth and early maturation (opportunistic strategists), increasingly dominated catches, whilst species with slow growth and late maturation (periodic strategists) declined. Truncated population age structures suggested longevity overfishing of three periodic strategist species: golden perch ( M. ambigua), black bream ( A. butcheri), mulloway ( A. japonicus) and a fourth species with an intermediate strategy, greenback flounder ( R. tapirina). This has implications for management because loss of older/larger individuals suggests reduced capacity to withstand or recover from deteriorated environmental conditions associated with a historically extreme drought in the lower Murray River system. Management of these species should seek to preserve the remnant population age structures and then to rebuild age structures by allowing recruits to become established in the adult population. We recommend that assessment of multi-species fisheries in changeable environments, such as occur in estuaries and other end-river environments, requires a suite of indicators that address changes in fish assemblages and populations. The effects of acute (24 h) exposure to the antidepressants amitriptyline, imipramine (both tricyclics), fluoxetine (a selective serotonin re-uptake inhibitor) and tranylcypromine (a monoamine oxidase inhibitor) on DNA damage in cultured C6 rat glioma cells were determined using an alkaline comet assay. The effects of manipulation of intracellular cyclic AMP by pretreatment with dibutyryl cyclic AMP (dBcAMP) and 3-isobutyl-1-methylxanthine (IBMX) were also studied. For fluoxetine, the effects of addition of exogenous glutathione (GSH) and pretreatment with L-buthionine sulfoximine (BSO) were also assessed. There were increases in DNA damage with increasing concentrations of antidepressants. IBMX pretreatment protected against antidepressant-induced DNA damage in C6 cells pretreated with dBcAMP. Addition of exogenous reduced GSH and BSO increased DNA damage after fluoxetine exposure. The data show that the antidepressants induce significant amounts DNA damage in C6 cells.
