Electrospun nanofibrous mats represent a new generation of medical textiles with promising applications in heart valve tissue reconstruction. It is important for biomaterials to mimic the biological and mechanical microenvironment of native extracellular matrix (ECM). However, the major challenges are still remaining for current biomedical materials, including appropriate mechanical properties, biocompatibility, and hemocompatibility. In the present work, the novel composite nanofibrous mats of poly(p-dioxanone) (PDO) and poly(ester-urethane)ureas (PEUU) are fabricated by electrospinning system. The optimal combination ratio of PDO to PEUU may balance the mechanical properties and cellular compatibility to match the newly formed tissue. In PDO/PEUU composite nanofibrous mats, PEUU can provide the biomimetic elastomeric behavior, and PDO could endow the excellent biocompatibility. In comparison to nanofibrous mat of neat PDO, the composite showed significantly improved mechanical properties, with 5-fold higher initial elongation at break. Furthermore, human umbilical vein endothelial cells (HUVECs) were cultured on the composite to evaluate its ability to rapidly endothelialize as heart valve tissue engineering. The results revealed that PDO/PEUU composite nanofibrous mats could promote cell adhesion and proliferation, especially for the ratio of 60/40. Overall, PDO/PEUU composite nanofibrous mats (60/40) show the excellent mechanical properties, appropriate biocompatibility and hemocompatibility which meet the necessary norm for tissue engineering and may be suitable for potential heart valve tissue reconstruction. Solid-state phase transformation kinetics in the near-equilibrium regime behaves differently from that in extremely non-equilibrium regime since their thermodynamic states are different. Incorporating temperature- and transformed fraction-dependent thermodynamic terms, a thermo-kinetic model is derived to describe the transformation kinetics in the near-equilibrium regime. The model predicts a sluggish stage in isothermally conducted transformation and a temperature-dependent stage in non-isothermally conducted transformation. Then, the kinetics of γ/ α transformations in two binary substitutional Fe-based alloys (i.e. Fe-3.28 at.% Mn and Fe-1.73 at.% Co), measured by isothermal and non-isothermal dilatometry, is investigated by the newly proposed model. The model quantitatively describes the retarded isothermal kinetics in Fe-3.28 at.% Mn alloy and the abnormal non-isothermal kinetics in Fe-1.73 at.% Co alloy. [ABSTRACT FROM AUTHOR]

Poly(lactide- co-glycolide)-poly(ethylene glycol)-poly(lactide- co-glycolide) (PLGA-PEG-PLGA) triblock copolymer was synthesized through the ring-opening polymerization of LA and GA with PEG as macroinitiator and stannous octoate as catalyst. The amphiphilic copolymer self-assembled into micelles in aqueous solutions, and formed hydrogels as the increase of temperature at relatively high concentrations (> 15 wt%). The favorable degradability of the hydrogel was confirmed by in vitro and in vivo degradation experiments. The good cellular and tissular compatibilities of the thermogel were demonstrated. The excellent adhesion and proliferation of bone marrow mesenchymal stem cells endowed PLGA-PEG-PLGA thermogelling hydrogel with fascinating prospect for cartilage tissue engineering. A series of biodegradable hydrogels based on dextran and poly(L-glutamic acid) were fabricated for effective vancomycin loading and release. The preparation of hydrogels was simply achieved by photo cross-linking of methacrylated dextran and poly(L-glutamic acid)- g-hydroxyethyl methacrylate (PGH) in the presence of photoinitiator I2959. The structures of hydrogels were characterized by FTIR and SEM. The swelling and enzymatic degradation behaviors of hydrogels were examined to be dependent on the poly(L-glutamic acid) content in the hydrogels. The higher content of poly(L-glutamic acid) in the gel, the higher swelling ratio and quicker degradation were observed. More interestingly, the hydrogel with higher PGH ratio showed higher vancomycin (VCM) loading content, which might be due to the electrostatic interaction between carboxylate groups in hydrogel and ammonium group of VCM. In vitro drug release from the VCM-loaded hydrogels in aqueous solution exhibited sustained release of VCM up to 72 h, while the in vitro antibacterial test based on the VCM-loaded hydrogel showed an efficient Methicillin-Resistant S. aureus (MRSA) inhibition extending out to 7 days. These results demonstrated that the biodegradable hydrogels which formed by in situ photo-cross linking would be promising as scaffolds or coatings for local antibacterial drug release in tissue engineering. Summary. In this study, we compared the effects of pimobenda (PIM), a putative Ca2+-sensitizer and phosphodiesterase (PDE) inhibitor, on the L-type Ca2+ current (ICa) of guinea-pig ventricular myocytes and contractile tension of ventricular papillary muscles with those of a nonselective PDE inhibitor, isobutylmethylxanthine (IBMX), and selective PDE-III inhibitors, that is, milrinone (MIL) and cilostazol (CIL). The efficacy (maximum attainable effect) of these drugs for increasing ICa or developed tension (DT) ranged in the order of IBMX ≫ MIL > PIM > CIL. This finding suggests that the positive inotropic effect of each drug is roughly proportional to its increasing effect on ICa. The additional effect of PIM (a Ca2+-sensitizing effect) was not identified in “intact” preparations, and the potentiating effects of PIM on the DT and ICa were virtually the same as those observed for MIL. To isolate the Ca2+-sensitizing effect of PIM on the DT, we studied the effects of PIM in the presence of H89, an isoquinoline derivative possessing a selective inhibitory effect on cAMP-dependent protein kinase. In the absence of H89, 50 µM PIM increased the DT by 68 ± 11% (mean ± SE, n = 6). However, in the presence of 20 µM H89, which completely blocked the PIM-induced increase in ICa, PIM (50 µM) significantly increased the DT by 19 ± 6% (n = 6), thereby indicating the presence of a positive inotropic effect attributable to a mechanism other than increased intracellular cAMP, that is, a Ca2+-sensitizing effect. The latter notion was supported by the finding that in the presence of H89 (20 µM), the PIM-induced augmentation of DT was accompanied by a prolongation of the time to 50% relaxation of contractile tension. In contrast, MIL (50 µM) and forskolin, a direct activator of adenylate cyclase (1–10 nM), did not increase DT in the presence of 20 µM H89. These results suggest that the fraction of positive inotropic effect of PIM attributable to its Ca2+-sensitizing effect is masked by its potent PDE-III inhibitory effect in “intact” ventricular preparations.
