Strange metals possess highly unconventional electrical properties, such as a linear-in-temperature resistivity1–6, an inverse Hall angle that varies as temperature squared7–9 and a linear-in-field magnetoresistance10–13. Identifying the origin of these collective anomalies has proved fundamentally challenging, even in materials such as the hole-doped cuprates that possess a simple bandstructure. The prevailing consensus is that strange metallicity in the cuprates is tied to a quantum critical point at a doping p* inside the superconducting dome14,15. Here we study the high-field in-plane magnetoresistance of two superconducting cuprate families at doping levels beyond p*. At all dopings, the magnetoresistance exhibits quadrature scaling and becomes linear at high values of the ratio of the field and the temperature, indicating that the strange-metal regime extends well beyond p*. Moreover, the magnitude of the magnetoresistance is found to be much larger than predicted by conventional theory and is insensitive to both impurity scattering and magnetic field orientation. These observations, coupled with analysis of the zero-field and Hall resistivities, suggest that despite having a single band, the cuprate strange-metal region hosts two charge sectors, one containing coherent quasiparticles, the other scale-invariant ‘Planckian’ dissipators.Measurements of high-field magnetotransport in overdoped cuprates indicate that the strange-metal regime exists beyond the critical doping, and that it has both coherent and incoherent contributions. A visualization of thermal diffusion in a horizontal pipe flow is conducted to elucidate the axial diffusion characteristics of two fluids of differing temperatures, particularly those flows between the laminar and turbulent flow regimes. Tracer particles are used to detect the thermal diffusion interface as well as to measure streamwise velocity profiles. We found that the Hagen–Poiseuille parabolic velocity profile established a long axial diffusion length that drives thermal convection in the pipe cross-sectional plane. This situation results in a 3D interface at Reynolds number, Re = 600, and a vertically stratified interface at Re = 1900. In turbulent flows at Re = 5700, an anti-diffusion spot appears in the front section before undergoing rapid mixing under turbulence. From the visualized particle images, the apparent axial coefficients of diffusion were measured and compared with the friction properties of the pipe. Superconductivity is ubiquitous as evidenced by the observation in many crystals including carrier-doped oxides and diamond. Amorphous solids are no exception. However, it remains to be discovered in quasicrystals, in which atoms are ordered over long distances but not in a periodically repeating arrangement. Here we report electrical resistivity, magnetization, and specific-heat measurements of Al–Zn–Mg quasicrystal, presenting convincing evidence for the emergence of bulk superconductivity at a very low transition temperature of Tc ≅ 0:05 K. We also find superconductivity in its approximant crystals, structures that are periodic, but that are very similar to quasicrystals. These observations demonstrate that the effective interaction between electrons remains attractive under variation of the atomic arrangement from periodic to quasiperiodic one. The discovery of the superconducting quasicrystal, in which the fractal geometry interplays with superconductivity, opens the door to a new type of superconductivity, fractal superconductivity. Biological invasions have severe economic and ecological impacts. The yellow-legged hornet, Vespa velutina was originally distributed throughout most of the Oriental region, but it has invasive populations in South Korea, Europe, and most recently, Japan. This species is a predator of honeybees, raising concerns about costs to beekeeping. A previous study concluded that South Korean and European populations originated from China. In this study, we investigate the genetic structure of V. velutina to infer the origin of Japanese populations. A maximum likelihood tree based on three mitochondrial genes (COI, Cytb, and 16S rRNA) revealed that V. velutina consists of an Indonesian-Malaysian clade and an Asian Continent clade, although some Indonesian samples are included in the latter clade. Sequences from individuals in the Japanese and South Korean populations were identical, suggesting that V. velutina in Japan came from South Korea. The single haplotype found in these introduced populations suggests that they originated from a very small number of founders from the natural distributional area. Nevertheless, their distribution continues to expand in introduced areas. Special attention should be paid to diapausing queens of the hornet, as they are likely responsible for these introductions. Interband tunneling in semiconductors [1, 2] is one of the clearest manifestations of quantum mechanics in electronic devices. In the case of interband tunneling in a heavily doped PN junction, in reverse bias, electrons tunnel from the filled states in the valence band to the empty states in the conduction band. Under forward bias, tunneling current is observed when electrons tunnel from filled states in the conduction band to empty states in the valence band, as shown in Fig. 8.1. As the conduction band on the n-side becomes biased higher in energy than the valence band, states are not available for electrons to tunnel into. As a result, the current decreases, leading to a characteristic negative differential resistance regime. [ABSTRACT FROM AUTHOR]

We have performed electron spin resonance (ESR) measurements in the low temperature magnetic-ordered state of polycrystalline samples of CdYbS. This material is one of the rare-earth spinel compounds in which Yb ions form the pyrochlore lattice, and consequently, CdYbS possesses anomalous properties caused by geometrical frustration and anisotropic terms. Some ESR signals have been detected, and their resonance fields are well-fitted by linear lines in the frequency vs. magnetic-field plane. One of the ESR modes appears to intersect the origin of this plane, suggesting a nearly gapless feature in the magnetically ordered state of this material. Two additional ESR modes appear above H = 2.6 T, indicating that there is a field-induced phase transition at H. Given the nearly gapless feature of CdYbS, we expect that the ground state of CdYbS must be either the Palmer-Chalker state or the ψ state. [ABSTRACT FROM AUTHOR]

We have performed high-field multi-frequency electron spin resonance (ESR) measurements of a single crystal of the S = 1/2 quasi one-dimensional (1D) Ising-like antiferromagnet BaCoVO at 1.5 K in magnetic fields of up to 14 T applied along [110] and [100] directions. Two main ESR branches, which are ascending and descending with increasing magnetic fields, are observed from 400 GHz at zero magnetic field in both cases. The low ESR mode shows a softening near 10 T for H || [100], while no softening was observed below 14 T for H || [110]. This difference in the magnetic excitations may explain the distinct phase diagrams for these two directions. [ABSTRACT FROM AUTHOR]

Power generation performance of a thermoelectric module consisting of the Heusler FeVAl alloy was evaluated. For construction of the module, W-doped FeVAl alloys were prepared using powder metallurgy process. Power generation tests of the module consisting of 18 pairs of p- n junctions were conducted on a heat source of 373-673 K in vacuum. The reduction of thermal conductivity and improvement of thermoelectric figure of merit by W-doping enhanced the conversion efficiency and the output power. High output power density of 0.7 W/cm was obtained by virtue of the high thermoelectric power factor of the Heusler alloy. Tumour necrosis factor-alpha (TNF-α) is an important mediator in the pathogenesis of rheumatoid arthritis (RA) and hypertension. TNF-α inhibitors improve clinical symptoms and inhibit joint destruction in RA, but their effect on blood pressure (BP) has not been fully investigated. We measured 24-h BP using an ambulatory BP monitor in 16 RA patients treated with a TNF-α inhibitor, infliximab, to investigate its influence on BP and its association with the regulatory factors of BP and renin-angiotensin-aldosterone and sympathetic nervous systems. Infliximab significantly reduced the 24-h systolic BP (SBP) from 127.4±21.8 to 120.1±23.4 mm Hg (P<0.0001). Particularly, morning BP (0600-0800 h) decreased from 129.7±19.7 to 116.9±13.4 mm Hg (P<0.0001), and daytime BP decreased from 131.8±15.1 to 122.5±13.7 mm Hg (P<0.0001). Infliximab significantly reduced the plasma level of norepinephrine and plasma renin activity (PRA) (from 347.5±180.7 to 283.0±181.8 pg ml−1 and 2.6±2.7 to 2.1±2.9 ng ml−1 h−1, respectively) but did not significantly reduce the plasma levels of dopamine and epinephrine. The reduction in morning SBP correlated with the reduction in the norepinephrine level (P<0.05) but not with that in PRA and inflammatory parameters related to RA. This study shows the effect of infliximab on ambulatory BP, especially daytime BP, which may be partly accounted for by the reduction of sympathetic nerve activity after infliximab treatment. Effects of element substitutions on thermoelectric properties of Heusler FeVAl alloys were evaluated. By W substitution at the V site, the thermal conductivity is reduced effectively because of the enhancement of phonon scattering resulting from the introduction of W atoms, which have much greater atomic mass and volume than the constituent elements of FeVAl alloy. W substitution is also effective to obtain a large negative Seebeck coefficient and high electrical conductivity through an electron injection effect. To change the conduction type from n-type to p-type, additional Ti substitution at the V site, which reduces the valence electron density, was examined. A positive Seebeck coefficient as high as that of conventional p-type FeVAl alloy was obtained using a sufficient amount of Ti substitution. Electrical resistivity was reduced by the hole doping effect of the Ti substitution while maintaining low thermal conductivity. Compared with the conventional solo-Ti-substituted p-type FeVAl alloy, the ZT value was improved, reaching 0.13 at 450 K. [ABSTRACT FROM AUTHOR]

Functional single-nucleotide polymorphisms (SNPs) in inositol 1,4,5-trisphosphate 3-kinase C (ITPKC) (rs28493229) and caspase-3 (CASP3) (rs113420705; formerly rs72689236) are associated with susceptibility to Kawasaki's disease (KD). To evaluate the involvement of these 2 SNPs in the risk for intravenous immunoglobulin (IVIG) unresponsiveness, we investigated 204 Japanese KD patients who received a single IVIG dose of 2 g kg−1 (n=70) or 1 g kg−1 daily for 2 days (n=134). The susceptibility allele of both SNPs showed a trend of overrepresentation in IVIG non-responders and, in combined analysis of these SNPs, patients with at least 1 susceptible allele at both loci had a higher risk for IVIG unresponsiveness (P=0.0014). In 335 prospectively collected KD patients who were treated with IVIG (2 g kg−1), this 2-locus model showed a more significant association with resistance to initial and additional IVIG (P=0.011) compared with individual SNPs. We observed a significant association when all KD patients with coronary artery lesions were analyzed with the 2-locus model (P=0.0031). Our findings strongly suggest the existence of genetic factors affecting patients' responses to treatment and the risk for cardiac complications, and provide clues toward understanding the pathophysiology of KD inflammation. A complicating factor in unravelling the theory of high-temperature (high-Tc) superconductivity is the presence of a 'pseudogap' in the density of states, the origin of which has been debated since its discovery. Some believe the pseudogap is a broken symmetry state distinct from superconductivity, whereas others believe it arises from short-range correlations without symmetry breaking. A number of broken symmetries have been imaged and identified with the pseudogap state, but it remains crucial to disentangle any electronic symmetry breaking from the pre-existing structural symmetry of the crystal. We use scanning tunnelling microscopy to observe an orthorhombic structural distortion across the cuprate superconducting Bi2Sr2Can?1CunO2n+4+x (BSCCO) family tree, which breaks two-dimensional inversion symmetry in the surface BiO layer. Although this inversion-symmetry-breaking structure can impact electronic measurements, we show from its insensitivity to temperature, magnetic field and doping, that it cannot be the long-sought pseudogap state. To detect this picometre-scale variation in lattice structure, we have implemented a new algorithm that will serve as a powerful tool in the search for broken symmetry electronic states in cuprates, as well as in other materials. We previously reported that polyamidoamine STARBURST dendrimer (generation 3, G3) (dendrimer) conjugate with α-cyclodextrin (α-CyD) having an average degree of substitution of 2.4 of α-CyD (α-CDE) provided remarkable aspects as novel carriers for DNA and small-interfering RNA. To develop novel α-CDE derivatives with tumor cell specificity, we prepared folate-appended α-CDEs (Fol-α-CDEs) and folate-polyethylene glycol (PEG)-appended α-CDEs (Fol-PαCs) with the various degrees of substitution of folate (DSF), and evaluated in vitro and in vivo gene transfer activity, cytotoxicity, cellular association and physicochemical properties. In vitro gene transfer activity of Fol-α-CDEs (G3, DSF 2, 5 or 7) was lower than that of α-CDE (G3) in KB cells, folate receptor (FR)-overexpressing cancer cells. Of the three Fol-PαCs (G3, DSF 2, 5 or 7), Fol-PαC (G3, DSF 5) had the highest gene transfer activity in KB cells. The activity of Fol-PαC (G3, DSF 5) was significantly higher than that of α-CDE (G3) in KB cells, but not in A549 cells, FR-negative cells. Negligible cytotoxicity of the plasmid DNA (pDNA) complex with Fol-PαC (G3, DSF 5) was observed in KB cells or A549 cells up to a charge ratio of 100/1 (carrier/pDNA). The cellular association of the pDNA complex with Fol-PαC (G3, DSF 5) could be mediated by FR on KB cells, resulting in its efficient cellular uptake. Fol-PαC (G3, DSF 5) had a higher binding affinity with folate-binding protein than α-CDE (G3), although the physicochemical properties of pDNA complex with Fol-PαC (G3, DSF 5) were almost comparable to that with α-CDE (G3), although the onset charge ratio and the compaction ability of Fol-PαC (G3, DSF 5) were slightly different. Fol-PαC (G3, DSF 5) tended to show a higher gene transfer activity than α-CDE (G3) 12 h after intratumoral administration in mice. These results suggest that Fol-PαC (G3, DSF 5), not Fol-α-CDEs, could be potentially used as a FR-overexpressing cancer cell-selective DNA carrier. [ABSTRACT FROM AUTHOR]

The nucleation of ferrite precipitates at austenite grain faces, edges (triple lines), and corners (quadruple points) was studied in a Co-15Fe alloy in which the matrix phase was retained upon cooling to room temperature by serial sectioning coupled with electron backscatter diffraction analysis. Nearly half of the edges and corners were vacant at an undercooling of 60 K from the γ/( α + γ) boundary where the precipitation occurred significantly at grain faces. A significant proportion of precipitates had Kurdjumov-Sachs (K-S) and to a lesser extent Nishiyama-Wassermann (N-W) orientation relationships with more than one grain at all boundary sites. Vacant edges and corners were readily observed, of which the misorientations of matrix grain boundaries would permit a precipitate to have a specific orientation relationship with multiple grains. Small differences in the nucleation activation energy among the grain faces, edges, and corners may lend support to a view proposed from experiments of nucleation in Fe-C base alloys that ferrite nuclei are more or less surrounded by low-energy facets of α/ γ phase boundary. We conducted feeding experiments on threadsail filefish Stephanolepis cirrhifer juveniles for 16 days to evaluate the efficacy of moon jellyfish Aurelia aurita as a prey item. Four treatments, each with 40 individuals, were prepared to compare fish growth performance. The treatments consisted of filefish that were starved (control) (S), fed only jellyfish (J), fed only krill (K), and fed both jellyfish and krill (JK). Fish in the S treatment exhibited a 50% mortality rate and reduced body weight, whereas the J treatment exhibited a zero mortality rate and increased body weight. Fish in the JK treatments showed a significantly faster growth than those in the K treatment. Filefish consumed as much as 24 and 13 times their own body weight in jellyfish per day in the J and JK treatments, respectively. This is the first report showing that growth can be sustained by feeding a marine fish only jellyfish, and indicates the potential of jellyfish as a fish prey in both nature and captivity. Accessory breast is synonymous with polymastia or supernumerary breast tissue. An accessory breast without a nipple or areola is rare. We report a case of fibroadenoma of an accessory breast with no nipple or areola in a 41-year-old woman who presented with a right axillary mass associated with five small nodules in the normally situated breast. Magnetic resonance imaging (MRI) showed the accessory breast surrounding the tumor. We ignored the presence of the component surrounding the mass and made a preoperative diagnosis of an axillary mass of possible metastases from multiple breast cancers or breast cancer of unknown origin associated with multiple breast fibroadenomas. From a retrospective view, based on the histological results, MRI and dynamic MRI demonstrated a tiny component of breast-like tissue surrounding the axillary mass and an enhancement pattern typical of fibroadenoma for the axillary mass. For the later diagnosis of the axillary mass, the interpretation of whether the component of breast tissue surrounding the axillary mass was present is crucial. If the component exists, a tumor that originated from the accessory breast should be foremost in the differential diagnosis. Dynamic MRI appears to contribute to the diagnosis of fibroadenoma of an accessory breast before biopsy or surgical resection. [ABSTRACT FROM AUTHOR]

In order to develop practical thermoelectric materials consisting solely of environmentally friendly elements, we investigated the thermoelectric properties of the AlMn-type ( P6/ mmc, hp26) AlMnSi alloys and the AlMnSi-type ( Pm-3, cP138) AlMnFeSi alloys, both of which possess a pseudogap at the Fermi level. The formation range in which the single phase is obtained was determined for these two phases. The electrical resistivity, Seebeck coefficient, and thermal conductivity of the samples involving no secondary phase were measured over the temperature range of 2 K to 300 K. It is found that the thermoelectric properties of these phases are qualitatively accounted for in terms of the pseudogap at the Fermi level in the electronic density of states and the disordering in local atomic arrangements. Particle–wave duality suggests we think of electrons as waves stretched across a sample, with wavevector k proportional to their momentum. Their arrangement in ‘k-space’, and in particular the shape of the Fermi surface, where the highest-energy electrons of the system reside, determine many material properties. Here we use a novel extension of Fourier-transform scanning tunnelling microscopy to probe the Fermi surface of the strongly inhomogeneous Bi-based cuprate superconductors. Surprisingly, we find that, rather than being globally defined, the Fermi surface changes on nanometre length scales. Just as shifting tide lines expose variations of water height, changing Fermi surfaces indicate strong local doping variations. This discovery, unprecedented in any material, paves the way for an understanding of other inhomogeneous characteristics of the cuprates, such as the pseudogap magnitude, and highlights a new approach to the study of nanoscale inhomogeneity in general. We present a muon-spin rotation study of the optimally doped cuprate superconductor (BiPb)2(SrLa)2 CuO6+ δ . The measured magnetic field dependence of the in-plane magnetic penetration λ ab suggests superconductivity with a dominant d-wave order parameter. The comparison of the temperature dependence of λ ab with calculations, assuming the angular gap symmetry as obtained from photoemission measurements, is consistent with a partial suppression of the quasi-particle weight towards the anti-nodal region of the Fermi surface. This suggests that the superconducting and the pseudogap state are dominated by different parts of the Fermi surface. [ABSTRACT FROM AUTHOR]

One of the main challenges in understanding high-Tc superconductivity is to disentangle the rich variety of states of matter that may coexist, cooperate or compete with d-wave superconductivity. At centre stage is the pseudogap phase, which occupies a large portion of the cuprate phase diagram surrounding the superconducting dome. Using scanning tunnelling microscopy, we find that a static, non-dispersive, ‘checkerboard’-like electronic modulation exists in a broad regime of the cuprate phase diagram and exhibits strong doping dependence. The continuous increase of checkerboard periodicity with hole density strongly suggests that the checkerboard originates from charge-density-wave formation in the antinodal region of the cuprate Fermi surface. These results reveal a coherent picture for static electronic orderings in the cuprates and shed important new light on the nature of the pseudogap phase. [ABSTRACT FROM AUTHOR]

Authors investigated symptomatology of depressive state among workers in a 20-year study. A cohort of 167 men of a company in Japan, aged 18-40 years old at baseline in 1985 was researched as a study sample. Depressive symptoms were assessed by the Zung's self-rating depression scale (SDS) consisting of 6 somatic- and 14 psychological-symptom items. In a logistic regression analysis, sleep disturbance and fatigue, and total somatic-symptom scores were related to both future and long-term depressive state (all P < 0.05), after adjusting age and other variables. Our study and previous reports suggest that castration results in increased bone turnover and lowered BMD and that these changes might be attenuated by anti-androgens, such as BL and EMP. Recent studies have shown that castration for PC decreases bone mineral density (BMD), while estrogen therapy or bicalutamide (BL) monotherapy maintains BMD. However, the effect of combined androgen blockade (CAB) on bone turnover is not well studied. A total of 204 men were evaluated in the study (control group: n = 56, castration group: n = 102, ‘CAB with BL’ group: n = 22, ‘CAB with estramustine phosphate (EMP)’ group: n = 24). We measured steroid hormone levels, BMD (measured at one-third distal radius), bone turnover markers (levels of urinary N-telopeptide cross links of type 1 collagen (u-NTx) and deoxypyridinoline (u-DPD), serum concentrations of osteocalcin (OC)) in order to assess differences between groups. The BMD % Z score of the castration group was significantly lower than that of the control group or the ‘CAB with EMP’ group (90.6% vs. 95.5%, 98.6%; p < 0.042, p < 0.044, respectively). Levels of u-NTx, u-DPD, OC of the castration group were the highest followed by the control group, then the ‘CAB with BL’ group and the ‘CAB with EMP’ group. Our study and previous reports suggests that castration results in increased bone turnover and lowered BMD and that these changes might be attenuated by anti-androgens, such as BL and EMP. Adeno-associated virus (AAV) is used in gene-therapy studies, but its tissue distribution is unknown in natural infection. We examined cynomolgus AAVs (previously isolated AAV10 and AAV11 and novel AAVcy.7) for their tissue distribution in 14 cynomolgi by type-specific PCR. We found AAV10, AAV11, and AAVcy.7 in 6, 10, and 14 monkeys, respectively, and two or three types in 11 monkeys, showing that these AAVs are widespread in the monkeys. We detected AAV at a higher level mainly in the lymphatic tissues and ileum, which suggests that AAV may invade the host through Peyer’s patches in the ileum and infect immune cells. In conventional superconductors, the superconducting gap in the electronic excitation spectrum prevents scattering of low-energy electrons. In high-temperature superconductors (HTSs), an extra gap, the pseudogap, develops well above the superconducting transition temperature TC. Here, we present a new avenue of investigating the pseudogap state, using scanning tunnelling microscopy (STM) of resonances generated by single-atom scatterers. Previous studies on the superconducting state of HTSs have led to a fairly consistent picture in which potential scatterers, such as Zn, strongly suppress superconductivity in an atomic-scale region, while generating low-energy excitations with a spatial distribution—as imaged by STM—indicative of the d-wave nature of the superconducting gap. Surprisingly, we find that similar native impurity resonances coexist spatially with the superconducting gap at low temperatures and survive virtually unchanged on warming through TC. These findings demonstrate that properties of impurity resonances in HTSs are not determined by the nature of the superconducting state, as previously suggested, but instead provide new insights into the pseudogap state. Tight junctions (TJs) create a paracellular permeability barrier. Although reactive oxygen species have been implicated as mediators of inflammation in inflammatory bowel diseases, their influence on the function of colonic epithelial TJs remains unknown. Oxidative stress-mediated colonic epithelial permeability was significantly attenuated by a p38 mitogen-activated protein (MAP) kinase inhibitor, SB203580. Although the amount of TJ proteins was not altered, hydrogen peroxide (H2O2) changed the localization of claudin-4 protein from an NP-40 insoluble fraction to a soluble fraction and from an apical TJ to lateral membrane. The p38 MAP kinase inactivator Wip1 significantly attenuated phosphorylation of p38 MAP kinase, and oxidative stress mediated permeability. H2O2-induced changes in claudin-4 localization were abolished by SB203580 pretreatment as well as Wip1-expressing adenovirus infection. This is the first study to demonstrate that exogenous Wip1 functions to protect oxidative stress-mediated colonic mucosal permeability and that H2O2-induced claudin-4 dislocalization is abolished by Wip1. The nature and behaviour of electronic states in high-temperature superconductors are the centre of much debate. The pseudogap state, observed above the superconducting transition temperature, Tc, is seen by some as a precursor to the superconducting state. Others view it as a competing phase. Recently, this discussion has focused on the number of energy gaps in the system. Some experiments indicate a single energy gap, implying that the pseudogap is a precursor state. Others indicate two, suggesting that it is a competing or coexisting phase. Here, we use temperature-dependent scanning tunnelling spectroscopy of (Bi1−yPby)2Sr2CuO6+x to clarify the situation. We find a previously unobserved narrow and homogeneous gap that vanishes near Tc, superimposed on the typically observed inhomogeneous and broad gap, which is only weakly temperature dependent. Amplification and overexpression of the miR-17-92 microRNAs (miRNA) cluster at 13q31.3 has recently reported, with pointers to functional involvement in the development of B-cell lymphomas and lung cancers. In the present study, we show that inhibition of miR-17-5p and miR-20a with antisense oligonucleotides (ONs) can induce apoptosis selectively in lung cancer cells overexpressing miR-17-92, suggesting the possibility of ‘OncomiR addiction’ to expression of these miRNAs in a subset of lung cancers. In marked contrast, antisense ONs against miR-18a and miR-19a did not exhibit such inhibitory effects, whereas inhibition of miR-92-1 resulted in only modest reduction of cell growth, showing significant distinctions among miRNAs of the miR-17-92 cluster in terms of their roles in cancer cell growth. During the course of this study, we also found that enforced expression of a genomic region, termed C2, residing 3′ to miR-17-92 in the intron 3 of C13orf25 led to marked growth inhibition in association with double stranded RNA-dependent protein kinase activation. Finally, this study also revealed that the vast majority of C13orf25 transcripts are detected as Drosha-processed cleavage products on Northern blot analysis and that a novel polyadenylation site is present 3′ to the miR-17-92 cluster and 5′ to the C2 region. We analyzed leaf shape variations in Ainsliaea apiculata Sch. Bip. to evaluate the uniqueness of morphological characters in populations on Yakushima Island, Kagoshima Prefecture, Japan. Leaf size and shape from populations on Yakushima Island ( n = 300) were compared with those from populations in other areas of Japan ( n = 300). A considerable amount of variation occurred in leaf size in A. apiculata populations both on Yakushima Island and elsewhere, but clear discontinuities in leaf size were not detected. Some variants previously thought to be endemic to Yakushima Island, i.e., A. apiculata var. acerifolia and A. apiculata var. rotundifolia, were also found in other locations in Japan. Moreover, these leaf types were found to be continuous with the typical leaf shape of A. apiculata var. apiculata via various intermediate types, suggesting the need for future revision of these taxa. Based on these results, we reevaluated the uniqueness of the Yakushima populations of A. apiculata in terms of leaf variation. Elongin A is a transcription elongation factor that increases the overall rate of mRNA chain elongation by RNA polymerase II. To gain more insight into the physiological functions of Elongin A, we generated Elongin A-deficient mice. Elongin A homozygous mutant (Elongin A−/−) embryos demonstrated a severely retarded development and died at between days 10.5 and 12.5 of gestation, most likely due to extensive apoptosis. Moreover, mouse embryonic fibroblasts (MEFs) derived from Elongin A−/− embryos exhibited not only increased apoptosis but also senescence-like growth defects accompanied by the activation of p38 MAPK and p53. Knockdown of Elongin A in MEFs by RNA interference also dramatically induced the senescent phenotype. A study using inhibitors of p38 MAPK and p53 and the generation of Elongin A-deficient mice with p53-null background suggests that both the p38 MAPK and p53 pathways are responsible for the induction of senescence-like phenotypes, whereas additional signaling pathways appear to be involved in the mediation of apoptosis in Elongin A−/− cells. HIV-1 protease inhibitor (PI), nelfinavir (NFV) induced growth arrest and apoptosis of NCI-H460 and -H520, A549, EBC-1 and ABC-1 non-small-cell lung cancer (NSCLC) cells in association with upregulation of p21waf1, p27kip1 and p53, and downregulation of Bcl-2 and matrix metalloproteinase (MMP)-2 proteins. We found that NFV blocked Akt signalling in these cells as measured by Akt kinase assay with glycogen synthase kinase-3alpha/beta (GSK-3alpha/beta) as a substrate. To explore the role of Akt signalling in NFV-mediated growth inhibition of NSCLC cells, we blocked this signal pathway by transfection of Akt small interfering RNA (siRNA) in these cells; transient transfection of Akt siRNA in NCI-H460 cells decreased the level of Bcl-2 protein and slowed their proliferation compared to the nonspecific siRNA-transfected cells. Conversely, forced-expression of Akt partially reversed NFV-mediated growth inhibition of these cells, suggesting that Akt may be a molecular target of NFV in NSCLC cells. Also, we found that inhibition of Akt signalling by NFV enhanced the ability of docetaxel to inhibit the growth of NCI-H460 and -H520 cells, as measured by MTT assay. Importantly, NFV slowed the proliferation and induced apoptosis of NCI-H460 cells present as tumour xenografts in nude mice without adverse systemic effects. Skeletrophin (mindbomb homolog 2 (MIB2)) is a RING (Really Interesting New Gene) finger-dependent ubiquitin ligase, which targets the intracellular region of Notch ligands. A previous immunohistochemical study demonstrated that skeletrophin was downregulated in many melanomas. In the present study, we have identified a promoter region of skeletrophin on a CpG island and detected aberrant methylation of this region in six of 31 invasive melanomas, but in none of 25 benign nevi or five non-invasive superficial spreading melanomas. Subsequently, we found that a zinc-finger transcriptional factor Snail, which is overexpressed in many melanoma cells, repressed the skeletrophin promoter activity via an E-box-related element and was involved in downregulation of skeletrophin. An activator protein-2, which has a tumor suppressor-like role in melanoma, increased skeletrophin expression. Interestingly, exogenously expressed skeletrophin reduced melanoma cell invasion in vitro and in vivo. Colony formation in soft agar was also reduced in a RING motif-dependent manner, without affecting cell growth. We also found that skeletrophin downregulated transcription of the Met oncogene, which encodes the hepatocyte growth factor receptor and plays a role in the determination of the invasive phenotype of many malignant tumors. Finally, exogenously expressed skeletrophin, but not its RING mutant, increased transcription of Hes1 gene, a downstream effector of Notch pathway in melanoma cells. Novel, simple, rapid, highly sensitive, and direct determination of iodide and thiocyanate ions in seawater has been performed by liquid chromatography (LC) with UV detection at 220 nm. The separation was achieved on a C30 column of conventional size (150 mm × 4.6 mm i.d.) modified with poly(ethylene glycol); an aqueous solution of 300 m M sodium sulfate and 50 m M sodium chloride was used as mobile phase. Detection limits ( S/ N=3) obtained by injecting a 20-μL sample were 0.5 and 6 ng mL−1 for iodide and thiocyanate, respectively. The method was successfully used for rapid and direct determination of iodide and thiocyanate in seawater samples, collected from the coasts of Japan, without any extra pretreatment. Pure silica gel (Pia Seed 5S-60-SIL) has been investigated as a cation-exchange stationary phase for ion chromatography of common monovalent and divalent cations (Li+, Na+, NH4 +, K+, Mg2+, and Ca2+) with conductimetric detection; dilute oxalic acid (0.05 m m oxalic acid, pH 4.1, to 1 m m oxalic acid, pH 3.0) was used as mobile phase. The Pia Seed 5S-60-SIL silica gel acted as a cation-exchange stationary phase for these cations when 0.2 m m oxalic acid at pH 3.6 was used as the mobile phase. Excellent simultaneous separation and highly sensitive indirect conductimetric detection of these cations were achieved in 20 min on a 150 mm × 4.6 mm i.d. Pia Seed 5S-60-SIL silica gel column with 0.2 m m oxalic acid containing 4 m m 18-crown-6 (1,4,7,10,13,16-hexaoxacycloctadecane), pH 3.7, as mobile phase (detection limits (signal-to-noise ratio, 3, injection volume, 20 μL), were 0.15 μ m for Li+, 0.16 μ m for Na+, 0.21 μ m for NH4 +, 1.0 μ m for K+, 0.17 μ m for Mg2+, and 0.25 μ m for Ca2+). Ifin vitro-cultured cells could be converted into individual fish, they would have numerous applications in the field of genome biology and biotechnology. In spite of the benefits, however, no such system is presently available. Because primordial germ cells (PGCs) have the potential to be converted into individual fish via maturation and fertilization processes, we chose them to be the starting material for our system. As the first step, we visualized live PGCs in rainbow trout. Because thevasatranscript is restricted to the germ cell lineage, its regulatory regions are activated only in PGCs. Therefore, we produced transgenic strains carrying the green fluorescent protein (GFP) gene driven by thevasagene regulatory regions. The resulting transgenic embryos showed green fluorescence specifically in PGCs. As the second step, the GFP-labeled PGCs were purified by flow cytometry. The genital ridges isolated from the transgenic embryos were dissociated by trypsin and sorted into GFP-positive and GFP-negative cells. The GFP-positive cells possessed morphological characteristics typical of PGCs. In addition, thevasagene was expressed only in the GFP-positive cells, confirming that they were PGCs. To obtain functional gametes derived from isolated PGCs, a technique for converting PGCs into eggs and sperm is necessary. For this purpose, we developed a method for transplanting PGCs into developing embryos in order to incorporate them into the germ cell lineage of the recipient embryos. Approximately 10 PGCs isolated from newly hatched embryos were transplanted into the peritoneal cavity of recipient hatchlings. The transplanted PGCs actively migrated towards, and were finally incorporated into, the genital ridge. The PGCs that settled in the genital ridges of the recipient embryos proliferated, started meiosis, and differentiated into eggs and sperm in synchrony with the germ cells of allogenic recipient embryos. Further, the donor-derived gametes produced normal progenies through fertilization. These techniques, in combination within vitroculture, genetic modification, and cryopreservation of PGCs, are expected to have numerous applications in the field of fish bioengineering. Pure silica gels (Pia Seed 5S-60-SIL) calcined at 200, 400, 600, 800 and 1000°C for 5 h have been used as cation-exchange stationary phases in ion chromatography with indirect photometric detection for common monovalent and divalent cations (Li+, Na+, NH4 +, K+, Mg2+ and Ca2+); 0.75 mm tyramine (4-(2-aminoethyl)phenol)-0.25 mm oxalic acid, pH 5.0, containing crown ethers (18-crown-6 (1,4,7,10,13,15-hexaoxacyclooctadecane) or 15-crown-5 (1,4,7,10,13-pentaoxacyclopentadecane)) was used as mobile phase. With increasing calcination temperature, the amounts of the crown ethers adsorbed on the calcined silica gel column increased and, consequently, the effect of the crown ethers as retention modifiers for these cations increased. Excellent simultaneous separation and highly sensitive detection of these cations at 275 nm were achieved in 17 min by use of a 150 mm×4.6 mm i.d. column packed with silica gel calcined at 1000°C and use of 0.75 mm tyramine-0.25 mm oxalic acid, pH 5.0, containing either 0.5 mm 18-crown-6 or 5.0 mm 15-crown-5 as mobile phase. S-1 is an anticancer drug in which tegafur is combined with modulators, gimeracil and oteracil potassium. We encountered a patient with gastric cancer for whom oral administration of S-1 was effective, and we report this case. A 79-year-old woman visited our hospital with a major complaint of anorexia. Tests revealed severe anemia, with a hemoglobin (Hb) level of 6.5 g/dl, and the patient was admitted to the hospital for treatment. The primary lesion was a large type-1 gastric cancer (poorly differentiated adenocarcinoma) in the middle gastric body. In addition, a lesion with a diameter of 50 mm was observed in the left hepatic lobe and small metastatic lesions were also scattered in the right lobe. When a performance status (PS) of 0 was obtained after her systemic condition had been improved, S-1 was started, at a dose of 80 mg/day (with one course consisting of administration for 4 weeks, followed by 2 weeks' rest). No severe adverse drug reaction was observed. After one course of administration was completed, the patient received administrations at the outpatient clinic. Upon the completion of two courses, computed tomography (CT) showed disappearance of the metastatic lesions and marked regression of the primary lesion. A 65-year-old woman was admitted to our hospital for forgetfulness, depression and eccentric behavior that had been first noticed 2 years prior to admission. She showed memory impairment, perseveration and repeated violent actions, but no limb-kinetic apraxia. She died 12 years after the onset of symptoms. At autopsy, the unfixed brain weighed 820 g. Atrophy was circumscribed in the frontal lobe on both sides. The globus pallidus and the caudate nucleus were markedly atrophic and gold yellow in color, and the substantia nigra was strikingly pale. The cortical area showed neuronal loss and status spongiosus of the second and third cortical layers with ballooned neurons. Marked neuronal loss was observed in the dorsomedial nucleus of the thalamus, Meynert basal nucleus and substantia nigra. With Holzer stain, fibrillary gliosis was found to be severe in the frontal lobe, globus pallidus, subthalamic nucleus, hippocampus, dorsomedial nucleus of thalamus, substantia nigra, pontine tegmentum and inferior olivary nucleus. With Bielschowsky-Hirano stain, neurofibrillary tangles were observed in the cortex, hippocampus, substantia nigra, dentate nucleus, subthalamic nucleus, pontine nucleus, the inferior olivary nucleus, dorsomedial nucleus of the thalamus and, to a lesser extent, the neostriatum. Strikingly numerous argyrophilic and tau-positive threads were present in the cerebral white matter. These neuropathological findings corresponded to corticobasal degeneration, but lesions characteristic of progressive supranuclear palsy were also found. Moreover, widespread iron deposition throughout the central nervous system was the most striking finding of the present case. The effect of fluoride on the hydrolysis of tetracalcium phosphate (TTCP; Ca4(PO4)2O) in 0.1 mol/l KH2PO4 containing 62–83 mmol/l KF was studied with the help of X-ray fluorescence measurements. Fluorine X-ray emission and absorption spectra of the final product of hydrolysis and reference samples (CaF2 and Ca5(PO4)3F) were measured at Beamline BL-2C of Photon Factory (PF, Tsukuba). Based on these measurements we concluded that hydrolysis of TTCP in the presence of KF converts it into fluorapatite. Luteinizing hormone- (LH)-dependent ovarian follicle maturation has been recently described in two stages for teleost fishes. The oocyte's ability to respond to the steroidal maturation-inducing hormone (MIH), also known as oocyte maturational competence (OMC), is acquired during the first stage+ADs- whereas the MIH-dependent resumption of meiosis occurs during the second stage. However, studies directly addressing OMC have been performed with a limited number of species and therefore the general relevance of the two-stage model and its mechanisms remain uncertain. In this study, we examined the hormonal regulation of OMC and its basic transduction mechanisms in ovarian follicles of the sciaenid teleost, Nibe ( Nibea mitsukurii). Exposure to MIH +AFs-17,20+ACY-bgr+ADs--dihydroxy-4-pregnen-3-one or 17,20+ACY-bgr+ADs-,21-trihydroxy-4-pregnen-3-one] stimulated germinal vesicle breakdown (index of meiotic resumption) in full-grown follicles primed with human chorionic gonadotropin (HCG, an LH-like gonadotropin) but not in those pre-cultured in plain incubation medium. The induction of OMC by HCG was mimicked by protein kinase A (PKA) activators (forskolin and dibutyryl cyclic AMP), and blocked by specific inhibitors of PKA (H89 and H8) as well as inhibitors of RNA (actinomycin D) and protein (cycloheximide) synthesis. Forskolin-induced OMC was also inhibited by actinomycin D and cycloheximide. A strong activator of protein kinase C, PMA, inhibited HCG-dependent OMC. In conclusion, OMC in Nibe ovarian follicles is gonadotropin-dependent and requires activation of the PKA pathway followed by gene transcription and translation events. These observations are consistent with the two-stage model of ovarian follicle maturation proposed for other teleosts, and suggest that Nibe can be used as new model species for mechanistic studies of ovarian follicle differentiation and maturation in fishes. Loss of human mismatch repair (hMSH2) gene function has been linked to hereditary non-polyposis colorectal cancer (HNPCC), Muir-Torre syndrome (MTS), and sporadic cancers, excluding skin cancers unrelated to MTS. We immunohistochemically examined 125 squamous cell carcinomas (SCCs) using a monoclonal antibody to the hMSH2 protein and compared the results with those for 106 precursor lesions of SCC, consisting of actinic keratosis (AK), Bowenoid type of actinic keratosis (BAK), and Bowen's disease (BOD). In contrast to the homogeneous immunoreactivity of proliferating cells composed of AK, BAK, and BOD, heterogeneous and diminished immunostaining to hMSH2 was observed in tumor cells of SCCs examined. In addition, two SCCs (2 of 125; 1.6%) at multiple loci exhibited a complete lack of immunoreaction to hMSH2. Immunohistochemical staining of hMSH2 was semiquantitatively scored as 0 (0% of total cells examined), 1 (less than 10%), 2 (10-50%), or 3 (more than 50%). Percentage preservation of and average score for hMSH2 expression in normal, AK, BAK, BOD, and SCC were 56% and 2.06, 100% and 2.80, 94% and 2.88, 83% and 2.78, 63% and 2.36, respectively. The percentage preservation of and average scores for hMSH2 in AK, BAK, and BOD were significantly higher than those in presumably normal skin (P<0.01). There were no significant differences in the percentage preservation of and average scores for hMSH2 between presumably normal skin and SCC. The score for hMSH2 expression was significantly correlated with score for sun exposure in presumably normal skin of each lesion (R=0.70). These findings for hMSH2 expression in precursor lesions and SCC suggest that promotion or activation of hMSH2 expression may be induced by the increased DNA damage caused by sun exposure and that diminished expression of it might occur according to the transformation from precancerous lesions to SCC. To evaluate the efficacy of long-term administration of acyclovir as prophylaxis against varicella-zoster virus (VZV) reactivation, we analyzed the medical records of 86 consecutive adult patients who obtained engraftment after allogeneic hematopoietic stem cell transplantation from January 1996 to March 2000. We started long-term low-dose (400 mg/day) oral administration of acyclovir in June 1999, and this was continued until the end of immunosuppressive therapy after transplantation. There was no breakthrough reactivation of VZV in patients receiving acyclovir. Five patients who were receiving cyclosporine or prednisolone developed VZV reactivation after discontinuing acyclovir. With this prophylaxis, the cumulative incidence of VZV reactivation at 1 year after transplantation decreased from 33% to 10% (P = 0.025). On multivariate analysis, the use of long-term acyclovir was identified as a significant independent parameter for the development of VZV reactivation. These findings suggest the efficacy of long-term prophylaxis with low-dose acyclovir. Resumption of acyclovir upon restarting immunosuppressive therapy might be important for the further prevention of VZV reactivation. Primary lymphoid tumors of the lacrimal sac are quite rare, and all reported cases are of B-cell tumors with good prognosis. To our knowledge, this is the first case of primary natural killer (NK) cell lymphoma of the lacrimal sac. A 55-year-old woman presented with a lacrimal sac tumor, and histological diagnosis of NK cell lymphoma was made. Although disease was initially localized to the right lacrimal sac, it invaded into the adjacent ethmoidal sinus before chemotherapy was initiated (clinical stage IIE). Epstein-Barr virus (EBV)-encoded small RNA (EBER) was detected in lymphoma cells by in situ hybridization. Systemic chemotherapy combined with intrathecal chemotherapy followed by local radiotherapy was performed, and the patient achieved complete remission. However, shortly after completion of chemoradiotherapy, the lymphoma relapsed with rapid systemic dissemination. Although high-dose chemotherapy with autologous peripheral blood stem cell transplantation (autoPBSCT) has been shown or confirmed to be an effective treatment for high-risk and relapsed non-Hodgkin's lymphoma (NHL), relapse after autoPBSCT remains a serious problem. In a clinical trial to overcome relapse, we adopted a treatment plan in which PBSCs purified in vitro to CD34+ cells to deplete tumor cells (CD34+ autoPBSCT), total body irradiation (TBI) of 1200 cGy, and melphalan, 180 mg/m2, were used as a preconditioning regimen. Eighteen patients with relapsed or high-risk NHL participated in the study. This study compared the incidence of complications following CD34+ autoPBSCT preconditioned with the TBI regimen (n = 10): the TBI group; CD34+ autoPBSCT with the non-TBI regimen (n = 8): the non-TBI group; and unselected autoPBSCT with the non-TBI regimen (n = 19): the unselected autoPBSCT control group. After day 30 posttransplantation, 6 of 10 patients treated with the TBI regimen developed 11 infectious complications in total, compared with only 1 of 8 patients treated with the non-TBI regimen and 4 of 19 patients given unselected autoPBSCT. Two fatal complications occurred in the TBI group, but none occurred in the other 2 groups. The CD4+ lymphocyte count at 1 month posttransplantation was significantly lower in the TBI group than in the unselected autoPBSCT group. These findings suggest that the addition of TBI to the preconditioning regimen for CD34+ autoPBSCT is associated with an increased incidence of severe infectious complications after transplantation. Using an axenic encystation system in vitro, we examined the effect of wortmannin, a potent inhibitor of phosphatidylinositol 3-kinase (PI 3-kinase), which is a signaling molecule responsible for numerous cellular responses, on the encystation of Entamoeba invadens. Wortmannin inhibited both encystation and growth of E. invadens strain IP-1 in a dose-dependent manner, the former being more resistant to the drug than the latter. There was little decrease in the number of trophozoites after 3 days of culture in encystation medium containing wortmannin; and the cells remained motile, suggesting that the inhibitory effect of the drug on encystation was not due to its toxic effect on trophozoites. The addition of wortmannin after the induction of encystation was also inhibitory for encystation. Trophozoites incubated for 1 day in encystation medium with wortmannin did not encyst after removal of the drug, suggesting that the drug effect was not reversible in encystation medium. In contrast, trophozoites cultured in growth medium with wortmannin did encyst after their transfer to encystation medium without the drug. Encystation with wortmannin was more strongly inhibited among trophozoites grown in the presence of the drug than among those grown in the absence of the drug. The process of cyst maturation was slightly affected by wortmannin. These results suggest a possible role for PI 3-kinase in the signaling involved in the encystation of E. invadens. MUC1 mucin is known to serve as a target molecule in the killing of breast cancer cells by cytotoxic T-lymphocytes (CTLs). We searched for a possible mechanism allowing tumour cells to escape from autologous CTLs. When the killing of breast cancer cells by autologous lymphocytes was examined in 26 patients with breast cancer, significant tumour cell lysis was observed in 8 patients, whereas virtually no autologous tumour cell lysis was detected in as many as 18 patients. In the patients who showed negligible tumour cell lysis, the autologous tumour cells expressed MUC1-related antigenic epitopes much more weakly than the tumour cells in the patients who exhibited strong cytotoxicity (significant statistically at P < 0.0005-0.0045), suggesting that the unresponsiveness of cancer cells to CTLs observed in these patients was mainly due to loss of MUC1 expression or modulation of its antigenicity. A breast cancer cell line, NZK-1, established from one of the cytotoxicity-negative patients, did not express MUC1 and was resistant to killing by CTLs, while control breast cancer cell lines expressing MUC-1 were readily killed by CTLs. Transfection of NZK-1 cells with MUC1 cDNA induced significant lysis by autologous T-lymphocytes. These results supported the importance of MUC1 mucin in autologous anti-tumour immunity, but suggested that the major escape mechanism of tumour cells from autologous T-lymphocytes is the loss and/or modulation of MUC1 antigenicity on tumour cells, which would limit the effectiveness of possible immunotherapy designed to target the MUC1 mucin. T cadherin is a unique cadherin cell adhesion molecule that is anchored to the surface membrane through a glycosyl phosphatidyl inositol (GPI) moiety. In the present study, we postulated that T cadherin could regulate surfactant protein (SP)-D gene expression in human bronchioloalveolar type-II cells. We transfected A549 cells (human lung cancer cell line with alveolar type-II cell characteristics) with the T-cadherin expression vector. Both original and control plasmid-transfected A549 cells expressed SP-D; however, neither human nor murine T-cadherin-transfected A549 cells expressed SP-D mRNA. The downregulation of SP-D production in human T-cadherin-expressed A549 cells was also demonstrated using Western immunoblotting techniques. Control vector-transfected A549 cells showed a positive band of SP-D but not of T cadherin. In contrast, T-cadherin-transfected A549 cells, which expressed T-cadherin protein, did not produce SP-D. We further examined the relationship of T cadherin and SP-D expression in secondary pulmonary alveolar proteinosis associated with hematolymphoid malignancies. SP-D was detected in bronchioloalveolar type-II cells in alveolar proteinosis. However, little or no T-cadherin expression was detected in alveolar type-II cells in these patients.
