Eradication of MRSA osteomyelitis requires elimination of distinct biofilms. To overcome this, we developed bisphosphonate-conjugated sitafloxacin (BCS, BV600072) and hydroxybisphosphonate-conjugate sitafloxacin (HBCS, BV63072), which achieve "target-and-release" drug delivery proximal to the bone infection and have prophylactic efficacy against MRSA static biofilm in vitro and in vivo. Here we evaluated their therapeutic efficacy in a murine 1-stage exchange femoral plate model with bioluminescent MRSA (USA300LAC::lux). Osteomyelitis was confirmed by CFU on the explants and longitudinal bioluminescent imaging (BLI) after debridement and implant exchange surgery on day 7, and mice were randomized into seven groups: 1) Baseline (harvested at day 7, no treatment); 2) HPBP (bisphosphonate control for BCS) + vancomycin; 3) HPHBP (hydroxybisphosphonate control for HBCS) + vancomycin; 4) vancomycin; 5) sitafloxacin; 6) BCS + vancomycin; and 7) HBCS + vancomycin. BLI confirmed infection persisted in all groups except for mice treated with BCS or HBCS + vancomycin. Radiology revealed catastrophic femur fractures in all groups except mice treated with BCS or HBCS + vancomycin, which also displayed decreases in peri-implant bone loss, osteoclast numbers, and biofilm. To confirm this, we assessed the efficacy of vancomycin, sitafloxacin, and HBCS monotherapy in a transtibial implant model. The results showed complete lack of vancomycin efficacy while all mice treated with HBCS had evidence of infection control, and some had evidence of osseous integrated septic implants, suggestive of biofilm eradication. Taken together these studies demonstrate that HBCS adjuvant with standard of care debridement and vancomycin therapy has the potential to eradicate MRSA osteomyelitis. [ABSTRACT FROM AUTHOR]

Family language policy (FLP) is increasingly recognised as a distinct domain of language policy concerned with the family as an arena of language policy formulation and implementation. While FLP is a relatively new research area, its conceptualisation of family and language practice requires re-examination due to social changes and technological developments, including the expansion of digital communication within families and the rise of globally dispersed families a product of global migration and transnationalism. In this systematic review of migrant FLP research, we investigate how the notions of family and language practice are conceptualised in research. Following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines, we identified a total of 163 articles for analysis. Our analysis reveals that the majority of studies were conducted in nuclear families, i.e., those consisting of a father, a mother, and one or more children. Studies also tend to conceptualise the family as fixed and physically located in one place. Paradoxically, around half of the studies acknowledge the presence of geographically dispersed family relations, but this does not necessarily affect their conceptualisation of what comprises a family. Language practice was conceptualised as physical and face-to-face communication in 51% of instances, with only 11% incorporating an analysis of digital communications. Based on our review, we recommend that FLP researchers researching migrant families reconceptualise the family as geographically dispersed and language practice as digital and multimodal when necessary. Such a reconceptualisation will help researchers understand the hitherto underexamined contributions of dispersed family members and multimodal digital communications in migrant FLP. According to the WHO, the number of mental disorder patients, especially depression patients, has overgrown and become a leading contributor to the global burden of disease. With the rising of tools such as artificial intelligence, using physiological data to explore new possible physiological indicators of mental disorder and creating new applications for mental disorder diagnosis has become a new research hot topic. We present a multi-modal open dataset for mental-disorder analysis. The dataset includes EEG and recordings of spoken language data from clinically depressed patients and matching normal controls, who were carefully diagnosed and selected by professional psychiatrists in hospitals. The EEG dataset includes data collected using a traditional 128-electrodes mounted elastic cap and a wearable 3-electrode EEG collector for pervasive computing applications. The 128-electrodes EEG signals of 53 participants were recorded as both in resting state and while doing the Dot probe tasks; the 3-electrode EEG signals of 55 participants were recorded in resting-state; the audio data of 52 participants were recorded during interviewing, reading, and picture description. Electroencephalography (EEG)-based studies focus on depression recognition using data mining methods, while those on mild depression are yet in infancy, especially in effective monitoring and quantitative measure aspects. Aiming at mild depression recognition, this study proposed a computer-aided detection (CAD) system using convolutional neural network (ConvNet). However, the architecture of ConvNet derived by trial and error and the CAD system used in clinical practice should be built on the basis of the local database; we therefore applied transfer learning when constructing ConvNet architecture. We also focused on the role of different aspects of EEG, i.e., spectral, spatial, and temporal information, in the recognition of mild depression and found that the spectral information of EEG played a major role and the temporal information of EEG provided a statistically significant improvement to accuracy. The proposed system provided the accuracy of 85.62% for recognition of mild depression and normal controls with 24-fold cross-validation (the training and test sets are divided based on the subjects). Thus, the system can be clinically used for the objective, accurate, and rapid diagnosis of mild depression. First principles was carried out studying the properties of (Ti, Nb)C compounds based on density functional theory. The integration of mechanical behavior, electronic structures, and thermodynamic properties can be optimized by mediating the concentration of the titanium alloying element. The results revealed that these transition metal compounds were stable with the negative formation energy. Nb0.5Ti0.5C (29.15 GPa) demonstrated the largest hardness characterized by moduli (B, G) because of the stable shell configuration. NbC exhibited the strongest anisotropy from the universal anisotropic index (AU) and three-dimensional surface contours. TixNb1−xC compounds displayed relatively strong stress responses along the [001], [110], and [111] directions. Due to the weakening p–d bonding, the ideal tensile strength gradually decreased with the increasing titanium concentration. The electronic structures revealed that the bonding characteristics of the (Ti, Nb)C compounds were a mixture of metallic and covalent bonds. On the other hand, NbC and TiC exhibited a minimum (740.55 K) and maximum (919.29 K) Debye temperature, indicating the stronger metalic bonds of NbC and covalent bonds of TiC. [ABSTRACT FROM AUTHOR]

To improve the high-rate capacity and cycle ability, LiZrO was successfully coated on LiNiCoMnO materials via wet chemical method. The crystal structure and electrochemical properties of the bare and coated material are studied by X-ray diffractometry (XRD), scanning electron microscope (SEM), transmission electron microscopy (TEM), cyclic voltammetry, and electrochemical impedance spectroscopy (EIS). The XRD and SEM results indicated that the lattice structure of LiZrO-coated materials was the same as the pristine one. Transmission electron microscopy showed that there was a thin LiZrO coating layer on the surface. LiZrO-coating improves the rate performance and cycling stability. Within the cutoff voltage of 2.6-4.8 V, the 1 wt% LiZrO-coated samples exhibited an initial discharge capacity of 190 mAh g and with a capacity retention about 85 % after 50 cycles at 0.1 C. LiCoPO has been successfully synthesized by a simple rheological phase method. The effects of synthesis temperature on the properties of LiCoPO are also investigated. The results show that a well-crystallized olivine structure LiCoPO with no obvious impurity phase is obtained. The electrochemical properties of LiCoPO were examined by galvanostatic charge/discharge tests, cyclic voltammetry (CV), and electrochemical impedance spectroscopy (EIS). The results indicate that the sintering temperature has a significant effect on the electrochemical properties of LiCoPO. The sample fired at 600 °C shows the best electrochemical properties. The initial discharge capacity is 101.0, 96.8, and 91.6 mAh g at 0.1, 0.2, and 1 C, respectively. The significantly improved electrochemical properties of LiCoPO are attributed to the better crystallized rheological phase production with better dispersed and smaller particles, which can greatly facilitate the diffusion of Li. Differences in the binding affinities of bisphosphonates for bone mineral have been proposed to determine their localizations and duration of action within bone. The main objective of this study was to test the hypothesis that mineral binding affinity affects bisphosphonate distribution at the basic multicellular unit (BMU) level within both cortical and cancellous bone. To accomplish this objective, skeletally mature female rabbits ( n = 8) were injected simultaneously with both low- and high-affinity bisphosphonate analogs bound to different fluorophores. Skeletal distribution was assessed in the rib, tibia, and vertebra using confocal microscopy. The staining intensity ratio between osteocytes contained within the cement line of newly formed rib osteons or within the reversal line of hemiosteons in vertebral trabeculae compared to osteocytes outside the cement/reversal line was greater for the high-affinity compared to the low-affinity compound. This indicates that the low-affinity compound distributes more equally across the cement/reversal line compared to a high-affinity compound, which concentrates mostly near surfaces. These data, from an animal model that undergoes intracortical remodeling similar to humans, demonstrate that the affinity of bisphosphonates for the bone determines the reach of the drugs in both cortical and cancellous bone. Inter-recti distance (IRD) and linea alba or umbilical hernia were measured by ultrasound. Strength of abdominal muscle was measured by a manual muscle test. Low back pain (LBP), urinary incontinence (UI) and quality of life (QOL) were measured by questionnaires including the Oswestry Disability Index, the International Consultation on Incontinence Questionnaire-Urinary Incontinence Short Form, 36-Item Short Form Health Survey respectively.Women with DRA experienced more severe LBP, and poorer QOL only 10 years postpartum according to the diagnostic criterion of IRD > 2cm. However, when the diagnostic criterion was raised to IRD > 3cm, women with DRA reported weaker abdominal muscle strength, more severe LBP 3, 5, and 10 years postpartum, poorer QOL 3, 5, 10, and 20 years postpartum, and higher incidence of linea alba or umbilical hernia 5 and 20 years postpartum.When using IRD > 2cm as the diagnostic criterion, the impact of DRA is minimal. However, when utilizing IRD > 3cm as the diagnostic criterion, DRA is associated with increased linea alba or umbilical hernia, weakened abdominal muscle strength, increased LBP, and decreased QOL. Most of the effects are particularly evident within 3–10 years postpartum, but becomes insignificant 20 and 30 years postpartum.
