Male breast cancer (MBC) is a rare but aggressive malignancy with cellular and immunological characteristics that remain unclear. Here, we perform transcriptomic analysis for 111,038 single cells from tumor tissues of six MBC and thirteen female breast cancer (FBC) patients. We find that that MBC has significantly lower infiltration of T cells relative to FBC. Metastasis-related programs are more active in cancer cells from MBC. The activated fatty acid metabolism involved with FASN is related to cancer cell metastasis and low immune infiltration of MBC. T cells in MBC show activation of p38 MAPK and lipid oxidation pathways, indicating a dysfunctional state. In contrast, T cells in FBC exhibit higher expression of cytotoxic markers and immune activation pathways mediated by immune-modulatory cytokines. Moreover, we identify the inhibitory interactions between cancer cells and T cells in MBC. Our study provides important information for understanding the tumor immunology and metabolism of MBC. Male breast cancer may involve different populations of immune cells. Mixed-cation lead mixed-halide perovskites simultaneously possess structural stability and high power conversion efficiency. A thorough study of both carrier and exciton dynamics is needed to understand the photophysical properties that underpin its superior photovoltaic performance. By utilizing a broadband transient absorption spectroscopy, we observe the carrier and exciton dynamics in a FA0.85Cs0.15Pb(I0.97Br0.03)3 (FCPIB) perovskite by simultaneously resolving the carrier and exciton contribution to the transient change of the absorption spectra, from which the carrier density and exciton oscillator strength can be determined. Our data reveal a quick and significant conversion of the photogenerated carriers to excitons, on top of the usual carrier recombination process. Moreover, the decay of carrier density shows a change of kinetics from a second-order recombination at high pump fluence to a third-order recombination at low pump fluence. Our analysis utilizes band anharmonicity, presents an independent determination of electronic temperature and quasi-Fermi energy, and reveals an interesting interplay among the processes of carrier cooling, exciton formation/decay and carrier recombination, all as a function of time after photoexcitation. Our work demonstrates the use of pump fluence as a knob to tune the relative populations of carriers and excitons in halide perovskite materials. Lead halide perovskites have many suitable features as materials for solar cells and a better understanding of the underlying physics will help engineer improved performance values. Background: Circular RNAs (circRNAs), a special class of noncoding RNAs with the characteristic of covalent closed-loop structure, have been widely found in various organisms. Growing evidence has shown that circRNAs play a crucial role in regulating biological functions of cancers. However, the specific role of circRNAs in esophageal squamous cell carcinoma (ESCC) remains largely unknown.Aim: The present study aims to investigate the effects of circ-TTC17 in ESCC clinical samples as well as cells.Methods: Sanger sequencing and agarose gel electrophoresis were used to verify the specificity of circ-TTC17. Expression levels of circ-TTC17 in ESCC cells, plasma, and tissues were measured by quantitative real-time polymerase chain reaction. A colony formation experiment, CCK-8 assay, and wound-healing assay were applied to detect the functions of circ-TTC17 in KYSE30 and KYSE450 cells. A nucleus-cytoplasm fractionation experiment was used to probe the location of circ-TTC17 in ESCC cells. Finally, a network of circ-TTC17 with its targeted miRNAs interactions and corresponding mRNAs was analyzed and framed by bioinformatics.Results: The expression level of circ-TTC17 was found to be significantly higher in ESCC cells, plasma, and tissues compared with normal cases. In vitro experiments indicated that circ-TTC17 promoted proliferation and migration of ESCC cells. Bioinformatics predictions showed that circ-TTC17 might regulate progress of ESCC by acting as a sponge for microRNAs (miRNAs).Conclusions: The results of this study demonstrate that upregulated circ-TTC17 plays a key role in promoting proliferation and migration of ESCC cells and has potential to become a novel biomarker for diagnosis, treatment, and prognosis of ESCC in the future. Purpose: Esophageal squamous cell carcinoma (ESCC) is one of the most aggressive squamous cell carcinomas and is generally resistant to chemotherapy. In the present study, the cytotoxic activity of Rabdocoestin B (Rabd-B) against ESCC and the underlying mechanisms were investigated.Methods: The inhibitory effect of Rabd-B on KYSE30 and KYSE450 was evaluated by Cell Counting Kit-8 (CCK8) and colony formation assays in vitro. The cell cycle distribution and apoptosis of cells treated with Rabd-B were determined by flow cytometry. The mechanisms underlying the effects of Rabd-B were systematically examined by Western blot. The in vivo anti-tumor ability of Rabd-B was measured in mouse xenograft models and cisplatin (DDP) was used as positive control.Results: Rabd-B efficiently induced G2/M phase arrest in ESCC cells by upregulating the Chk1/Chk2-Cdc25C axis to inhibit the G2→M transition facilitated by Cdc2/Cyclin B1. Furthermore, Rabd-B suppressed ATM/ATR phosphorylation, thereby inhibiting BRCA1-mediated DNA repair, which resulted in mitotic catastrophe and induced cell apoptosis. Rabd-B also decreased the activity of the Akt and NF-κB survival signaling pathways and ultimately initiated the caspase-9-dependent intrinsic apoptotic pathway in ESCC cells. The apoptosis induced by Rabd-B could be partially reversed by a caspase-9-specific inhibitor (Z-LEHD-FMK) and a pan-caspase inhibitor (Z-VAD-FMK). Moreover, Rabd-B effectively suppressed tumor growth in mouse xenografts which was comparable to that of DDP without significant injuries to the mice.Conclusion: Taken together, these findings indicate that Rabd-B is a promising precursor compound that may be useful as a treatment for ESCC and thus warrants further investigation. Four new diterpenoids, phyllostachysins I-L ( 1- 4), along with a known one, hebeiabinin B (5), were isolated from the aerial parts of Isodon phyllostachys ( I. phyllostachys). Compounds 1 and 2 feature an ent-kaurane backbone, and 3, 4 and 5 bear an ent-abietane skeleton. Their structures were elucidated by means of spectroscopic analysis. Compounds 3 and 4 exhibited significant cytotoxic activity against human tumor cell lines (HL-60, SMMC-7721, A-549, MCF-7 and SW-480), and strongly inhibited NO production in LPS-stimulated RAW264.7 cells. Naturally occurring molecules derived from higher plants, animals, microorganisms and minerals play an important role in the discovery and development of novel therapeutic agents. The identification of molecular targets is of interest to elucidate the mode of action of these compounds, and it may be employed to set up target-based assays and allow structure-activity relationship studies to guide medicinal chemistry efforts toward lead optimization. In recent years, plant-derived natural compounds possessing potential anti-tumor activities have been garnering much interest and efforts are underway to identify their molecular targets. Here, we attempt to summarize the discoveries of several natural compounds with activities against hematological malignancies, such as adenanthin, oridonin, gambogic acid and wogonoside, the identification of their targets, and their modes of actions. Applications of ZnO nanomaterials in optoelectronics are still limited due to their insufficient photoluminescence efficiency. In order to optimize the photoluminescence properties of ZnO nanorods, the UV emission of vertically aligned ZnO nanorods grown on a Si substrate, in correlation with Ga ion irradiation at different ion energies (0.5 keV-16 keV), was investigated in the present study. We found that the UV intensity increased rapidly with increasing Ga ion energy, up to its maximum around 2 keV, at which point the intensity was approximately 50 times higher than that produced by as-grown ZnO nanorods. The gentle bombardment of low-energy Ga ions removes defects from ZnO nanorod surfaces. The Ga ions, on the other hand, implant into the nanorods, resulting in compressive strain. It is believed that the perfect arrangement of the crystal lattice upon removal of surface defects and the introduction of compressive strain are two factors that contribute to the significant enhancement of UV light generation. Waveguide design becomes more and more important with the increasing demand of efficient routing and interconnection of electromagnetic signals for advanced information technologies. Here we review the basic theory, special techniques, and potential applications of transformation optics in waveguide design. Three major design strategies of transformation optics and their limitations in waveguide applications are discussed. The recent new trends of transformation techniques by imposing specific restrictions from materials are emphasized for practical implementation. Plasmonic waveguide is used as an example to compare these transformation strategies. Applications beyond waveguides but with great practical value such as super-resolution hyperlens design are also discussed in the context of waveguiding phenomena from the perspective of transformation optics. Microlasers with controllable polarization of output emission are vital for on-chip optical communications, optical sensors and optical switches. In this work, we report a high quality (Q) factor, low-threshold polymer microfiber laser and the possibility of achieving laser emission with a desired polarization. The microfiber is fabricated by direct drawing from a dye-doped polymer solution and it can generate whispering gallery mode (WGM) lasing under optical pulse excitation. When the microfiber is pumped from the side with pumping direction perpendicular to the microfiber's axis, the polarization direction of the output laser is found to be the same as that of the pump laser. Lasing emission with either transverse electric (TE) or transverse magnetic (TM) modes can be obtained and these two polarization states can be switched over by tuning the pumping laser. Furthermore, emission with both TE and TM modes can also be observed by changing the orientation of the microfiber relatively to pumping direction. All-inorganic cesium lead halide perovskites have been emerging as the promising semiconductor materials for next-generation optoelectronics. However, the fundamental question of how the environmental atmosphere affects their photophysical properties, which is closely related to the practical applications, remains elusive. Here, we report the dynamic switching between radiative exciton recombination and non-radiative carrier trapping in CsPbBr3 by controlling the atmospheric conditions. Specifically, we show that the photoluminescence (PL) intensity from the CsPbBr3 crystals can be boosted by ~ 60 times by changing the surrounding from vacuum to air. Based on the comprehensive optical characterization, near-ambient pressure X-ray photoelectron spectroscopy (NAP-XPS) as well as density functional theory (DFT) calculations, we unravel that the physisorption of oxygen molecules, which repairs the trap states by passivating the PL-quenching bromine vacancies, is accountable for the enhanced PL in air. These results are helpful for better understanding the optical properties of all-inorganic perovskites. Perovskite-based optoelectronics are expected to become an important component of devices such as solar cells and light-emitting diodes. However, how they are affected by environmental conditions is still not fully understood. Background: Gynecomastia denotes the benign proliferation of glandular breast tissue and stands as a recognized risk factor for male breast cancer. Nonetheless, the underlying carcinogenic mechanisms orchestrating the progression from gynecomastia to cancer remain poorly understood.This study employed single-cell RNA sequencing (scRNA-seq) to meticulously dissect the cellular landscape of gynecomastia and unravel potential associations with male breast cancer at a single-cell resolution. Pseudotime and evolutionary analyses were executed to delineate the distinct features characterizing gynecomastia and male breast cancer. The TCGA database, along with cell-cell communication analysis and immunohistochemistry staining, was harnessed to validate differential gene expression, specifically focusing on CD13.From the copy number variation profiles and evolutionary tree, we inferred shared mutation characteristics (18p+ and 18q+) underpinning both conditions. The developmental trajectory unveiled an intriguing overlap between gynecomastia and malignant epithelial cells. Moreover, the differential gene CD13 emerged as a common denominator in both gynecomastia and male breast cancer when compared with normal mammary tissue. Cell-cell interaction analysis and communication dynamics within the tumor microenvironment spotlighted distinctions between CD13+ and CD13− subsets, with the former exhibiting elevated expression of FGFR1-FGF7.Our investigation provides novel insights into the evolutionary progression from gynecomastia to male breast cancer, shedding light on the pivotal role of CD13 in driving this transition.
