Biological nitrification inhibition (BNI) is a plant function where root systems release antibiotic compounds (BNIs) specifically aimed at suppressing nitrifiers to limit soil-nitrate formation in the root zone. Little is known about BNI-activity in maize (Zea mays L.), the most important food, feed, and energy crop. Two categories of BNIs are released from maize roots; hydrophobic and hydrophilic BNIs, that determine BNI-capacity in root systems. Zeanone is a recently discovered hydrophobic compound with BNI-activity, released from maize roots. The objectives of this study were to understand/quantify the relationship between zeanone activity and hydrophobic BNI-capacity. We assessed genetic variability among 250 CIMMYT maize lines (CMLs) characterized for hydrophobic BNI-capacity and zeanone activity, towards developing genetic markers linked to this trait in maize. CMLs with high BNI-capacity and ability to release zeanone from roots were identified. GWAS was performed using 27,085 SNPs (with unique positions on the B73v.4 reference genome, and false discovery rate = 10), and phenotypic information for BNI-capacity and zeanone production from root systems. Eighteen significant markers were identified; three associated with specific BNI-activity (SBNI), four with BNI-activity per plant (BNIPP), another ten were common between SBNI and BNIPP, and one with zeanone release. Further, 30 annotated genes were associated with the significant SNPs; most of these genes are involved in pathways of "biological process", and one (AMT5) in ammonium regulation in maize roots. Although the inbred lines in this study were not developed for BNI-traits, the identification of markers associated with BNI-capacity suggests the possibility of using these genomic tools in marker-assisted selection to improve hydrophobic BNI-capacity in maize. The capacity of several plant species or landraces to inhibit nitrification in soil (biological nitrification inhibition, BNI) has been assessed in certain tropical pastures. These assessments are commonly based on potential net nitrification rates, which do not differentiate between gross nitrification and other processes that may reduce the amount of nitrate in soil. In a greenhouse experiment using two genotypes of Urochloa humidicola with contrasting BNI capacity in vitro, we evaluated gross N transformation rates before and after (7 and 21 days) N fertilization, while periodically measuring N2O emissions. Gross nitrification rates (in fact gross nitrate production assessed by pool dilution technique) were comparable in both genotypes and were low in comparison to strong microbial NH4+ immobilization. The N2O emissions were higher in pots with low-BNI plants. The discrepancy between the potential net nitrification rates assessed in laboratory assays (higher in low-BNI plants) and gross nitrification in pot or field experiments (no differences between genotypes) can be attributed to the out-competition of ammonia oxidizers by plant N uptake and ammonia immobilizing heterotrophic microbes, resulting in low nitrification under conditions where growing plants are present. This study confirmed the capacity of certain U. humidicola genotypes to reduce N2O emissions but warrants further investigation of the underlying mechanisms. It also questions the relevance of BNI in the rhizosphere of this plant species as other mechanisms (rather than the inhibition of gross nitrification) seem to be more important in maintaining low-nitrate soil environments in soil–plant systems of U. humidicola. This position paper summarizes the current understanding of biological nitrification inhibition (BNI) to identify research needs for accelerating the development of BNI as a N2O mitigation strategy for grazed livestock systems. We propose that the initial research focus should be on the systematic screening of agronomically desirable plants for their BNI potency and N2O reduction potential. This requires the development of in situ screening methods that can be combined with reliable N2O emission measurements and microbial and metabolomic analyses to confirm the selective inhibition of nitrification. As BNI-induced reductions in N2O emissions can occur by directly inhibiting nitrification, or via indirect effects on other N transformations, it is also important to measure gross N transformation rates to disentangle these direct and indirect effects. However, an equally important challenge will be to discern the apparent influence of soil N fertility status on the release of BNIs, particularly for more intensively managed grazing systems.
