Spinal cord injury (SCI) is a catastrophic accidence with little effective treatment, and inflammation played an important role in that. Previous studies showed photobiomodulation (PBM) could effectively downregulate the process of inflammation with modification of macrophage polarization after SCI; however, the potential mechanism behind that is still unclear. In the presented study, we aimed to investigate the effect of PBM on the expression level of versican, a matrix molecular believed to be associated with inflammation, and tried to find the mechanism on how that could regulate the inflammation process. Using immunofluorescence technique and western blot, we found the expression level of versican is increased after injury and markedly downregulated by irradiation treatment. Using virus intrathecal injection, we found the knock-down of versican could produce the effect similar to that of PBM and might have an effect on inflammation and macrophage polarization after SCI. To further verify the deduction, we peptide the supernatant of astrocytes to induce M0, M1, and M2 macrophages. We found that the versican produced by astrocytes might have a role on the promotion of M2 macrophages to inflammatory polarization. Finally, we investigated the potential pathway in the regulation of M2 polarization with the induction of versican. This study tried to give an interpretation on the mechanism of inflammation inhibition for PBM in the perspective of matrix regulation. Flexures are commonly used in various fields, including optics, microscopy, robotics, and precision engineering since they offer several advantages over traditional motion systems that employ mechanical bearings or sliding mechanisms such as high precision, minimal friction and hysteresis, lack of backlash, compact and lightweight, no lubrication and high stiffness. Flexure may experience cross-coupling effects, which occurs when motion along one axis affects motion along another axis due to mechanical interactions between the flexures. This can lead to unwanted motion errors of flexures or flexure-induced motion errors. This paper discusses identification and compensation of the flexure-induced motion error with a voice coil motor. Firstly, we built one DOF motion system supported by flexure and drove the motion system with a voice coil motor. Flexure-induced motion errors are then identified experimentally using single sine excitation. The single sine excitation can quantitively estimate the inclination angle of the flexure motion system, which makes cross-coupling error. Finally, a feedforward compensation method for the flexure-induced motion error with the voice coil motor is proposed and verified through experimentation. [ABSTRACT FROM AUTHOR]

Background: Diabetes, a chronic disease worldwide, may be associated with a poorer prognosis in patients with coronavirus disease 2019 (COVID-19). While some antihyperglycemic medications may be beneficial, others may increase the risk of adverse clinical outcomes of COVID-19. We aimed to analyze the effect of antihyperglycemic medications on COVID-19. Methods: We searched the Web of Science, Cochrane Library, EMBASE, PubMed, and Scopus databases from December 2019 to June 2022 to identify literature related to patients with COVID-19 and type 2 diabetes mellitus (T2DM) treated with antihyperglycemic medications. Results: 56 studies were included in the analysis. Metformin (OR 0.66; 95% CI 0.58–0.74; p < 0.05), Glucagon-like peptide-1 receptor agonist (GLP-1ra) (OR 0.73; 95% CI 0.59–0.91; p < 0.05), and sodium-dependent glucose transporters 2 inhibitor (SGLT 2i) (OR 0.77; 95% CI 0.69–0.87; p < 0.05) were associated with lower mortality risk, while insulin was associated with increased mortality risk (OR 1.40; 95% CI 1.26–1.55; p < 0.05). Meanwhile, metformin (OR 0.65; 95% CI 0.50–0.85; p < 0.05) and GLP-1ra (OR 0.84; 95% CI 0.76–0.94; p < 0.05) were significantly associated with decreased severe manifestation risk. What's more, metformin (OR 0.77; 95% CI 0.62–0.96; p < 0.05), GLP-1ra (OR 0.86; 95% CI 0.81–0.92; p < 0.05), and SGLT 2i (OR 0.87; 95% CI 0.79–0.97; p < 0.05) were also associated with a decreased risk of hospitalization, but insulin were associated with an increased risk of hospitalization (OR 1.31; 95% CI 1.12–1.52; p < 0.05). Nevertheless, the results of the subgroup analyses showed that the effects of different glucose-lowering agents on COVID-19 may be related to in-hospital use or out-hospital use, elderly or non-elderly patients use, and different geography. Conclusion: Metformin, GLP-1ra, and SGLT 2i have shown a positive effect on clinical outcomes in COVID-19, particularly in non-elderly individuals. However, insulin use may pose a higher risk, especially in elderly patients, so need with caution. Meanwhile, DPP-4i, TZD, α-GLUi, and sulfonylureas appeared to have a neutral effect. Objective: Teprotumumab plays an important role in thyroid eye disease pathogenesis and progression. We intend to mine the adverse event (AE) signals from a relevant database, thereby contributing to the safe use of teprotumumab. Methods: The data obtained from the ASCII data packages in the FAERS database from January 2020 to the second quarter of 2023 were imported into the SAS software (version 9.4) for data cleaning and analysis. Disproportionality analysis was performed using the reporting odds ratio (ROR) in conjunction with the United Kingdom Medicines and Healthcare Products Regulatory Agency (MHRA) omnibus standard method to detect positive signals. Participants: This retrospective observational study relied on adverse drug reactions reported to the FDA through FAERS, which is a standard public system for spontaneous reporting. Results: Collectively, 2171 AE reports for teprotumumab were collected, among which 108 significant signals were identified involving 17 system organ classes. The SOC of ear and labyrinth disorders included the most AE signals and reports. Muscle spasms, fatigue, headache, nausea, diarrhea, alopecia, blood glucose increased, hypoacusis, tinnitus, and diabetes mellitus were the top ten PTs ranked by the frequency of reporting, meanwhile, the two high-strength signals of thyroid-stimulating immunoglobulin increase (ROR 662.89, 95% CI 182.40–2409.19) and gingival recession (ROR 125.13, 95% CI 79.70–196.45) were not documented in the drug instruction. Meanwhile, we found a higher risk of increased blood glucose, deafness, and decreased appetite for male patients, and headache for female patients. Conclusions: Clinical application of teprotumumab should be closely monitored for ototoxicity, nail abnormalities, and menstrual changes, as well as for AEs not mentioned in the drug instruction, including gingival recession, thyroid-stimulating immunoglobulin increase, and so on. Because of its high toxicity and severe degradation, phenol has caused serious environmental pollution, and it is urgent to explore a stable and efficient degradation method of phenol. In this study, Ni/Fe bimetallic atomic cluster composite catalyst activated peroxymonosulfate (PMS) to degrade phenol efficiently. The experimental results show that the phenol degradation rate can reach 100% in a wide range of pH (3–11), and the system offers good stability in repeated experiments. The free radical quenching investigation and electrochemical characterization showed that the non-free radical pathway dominated the degradation system, and a large amount of singlet oxygen and rapid electron transfer promoted the efficient degradation of phenol in wastewater. Transitions between distinct obstructed atomic insulators (OAIs) protected by crystalline symmetries, where electrons form molecular orbitals centering away from the atom positions, must go through an intermediate metallic phase. In this work, we find that the intermediate metals will become a scale-invariant critical metal phase (CMP) under certain types of quenched disorder that respect the magnetic crystalline symmetries on average. We explicitly construct models respecting average C2zT, m, and C4zT and show their scale-invariance under chemical potential disorder by the finite-size scaling method. Conventional theories, such as weak anti-localization and topological phase transition, cannot explain the underlying mechanism. A quantitative mapping between lattice and network models shows that the CMP can be understood through a semi-classical percolation problem. Ultimately, we systematically classify all the OAI transitions protected by (magnetic) groups P m , P 2 ′ , P 4 ′ , and P 6 ′ with and without spin-orbit coupling, most of which can support CMP. The authors identify a novel delocalization mechanism for topologically trivial obstructed insulators. One potential approach for treating obesity is to increase energy expenditure in brown and white adipose tissue. Here we aimed to achieve this outcome by targeting mitochondrial uncoupler compounds selectively to adipose tissue, thus avoiding side effects from uncoupling in other tissues. Selective drug accumulation in adipose tissue has been observed with many lipophilic compounds and dyes. Hence, we explored the feasibility of conjugating uncoupler compounds with a lipophilic C8-hydrocarbon chain via an ether bond. We found that substituting the trifluoromethoxy group in the uncoupler FCCP with a C8-hydrocarbon chain resulted in potent uncoupling activity. Nonetheless, the compound did not elicit therapeutic effects in mice, likely as a consequence of metabolic instability resulting from rapid ether bond cleavage. A lipophilic analog of the uncoupler compound 2,6-dinitrophenol, in which a C8-hydrocarbon chain was conjugated via an ether bond in the para-position (2,6-dinitro-4-(octyloxy)phenol), exhibited increased uncoupling activity compared to the parent compound. However, in vivo pharmacokinetics studies suggested that 2,6-dinitro-4-(octyloxy)phenol was also metabolically unstable. In conclusion, conjugation of a hydrophobic hydrocarbon chain to uncoupler compounds resulted in sustained or improved uncoupling activity. The genus Acer L. (Sapindaceae Juss.) nowadays thrives in the temperate-subtropical areas of the Northern Hemisphere, especially in Yunnan and its adjacent areas in southwestern China. However, the fossil records of Acer from China, especially fruit fossils, are rather limited compared to its modern prosperity and high species diversity there. In this study, we described three taxa of Acer, viz. A. cf. caesium Wall. ex Brandi, A. cf. laurinum Hassk. ex Miq. and Acer sp., based on samara fossils from the Upper Pliocene of Tengchong, Yunnan, southwestern China. The samaras of Acer cf. caesium are characterised by large size, suborbicular to elliptical nutlet shape, and acute divergent angle; those of Acer cf. laurinum are characterised by large size, acute divergent angle, long elliptical nutlet shape and 5-7 veined nutlet surface; and those of Acer sp. are possibly underdeveloped and distinguished from all extant and fossil species of Acer by small size, distinct sulcus, and specific sub-triangular to semicircular nutlet shape. The occurrence of these fossils from Tengchong indicates that Acer was prosperous with high species diversity in western Yunnan during the Late Pliocene. The migration and differentiation of the genus Acer in southwestern China might have been accelerated by the rapid uplift of the Hengduan Mountains (HDM) and the post-Pliocene global cooling trend. In the last 40 years, Chinese cities have seen extensive development across a wide array of spheres. In this study, we applied a power-law and multifractal analysis to characterise 22 indices for 288 prefecture-level cities in mainland China. The data used to characterise the indices is representative of the year 2012. The results show that the Gross domestic product (GDP) is regular and exhibits multifractal spatial characteristics. Specifically, most developmental fields exhibit coupling in conjunction with being chaotic. Furthermore, eight indices with multifractal characteristics clearly reflect the spatial complexity of the corresponding fields and the volatility between prefecture-level cities. Overall, the tertiary industry has undergone nongeneralised development. On the other hand, the construction industry resembles a bubble economy, and the spatial layout of traditional industries has a homogenising effect in Middle and East China. This study examines China's overall spatial characteristics and states based on the above-mentioned analyses; additionally, the study focuses on the early 2010s. During spinal cord injury (SCI), the homeostasis of the cellular microenvironment in the injured area is seriously disrupted, which makes it extremely difficult for injured neurons with regenerative ability to repair, emphasizing the importance of restoring the cellular microenvironment at the injury site. Neurons interact closely with other nerve cells in the central nervous system (CNS) and regulate these cells. However, the specific mechanisms by which neurons modulate the cellular microenvironment remain unclear. Exosomes were isolated from the primary neurons, and their effects on astrocytes, microglia, oligodendrocyte progenitor cells (OPCs), neurons, and neural stem cells were investigated by quantifying the expression of related proteins and mRNA. A mouse SCI model was established, and neuron-derived exosomes were injected into the mice by the caudal vein to observe the recovery of motor function in mice and the changes in the nerve cells in the lesion area. Neuron-derived exosomes could reverse the activation of microglia and astrocytes and promote the maturation of OPCs in vivo and in vitro. In addition, neuron-derived exosomes promoted neurite outgrowth of neurons and the differentiation of neural stem cells into neurons. Moreover, our experiments showed that neuron-derived exosomes enhanced motor function recovery and nerve regeneration in mice with SCI. Our findings highlight that neuron-derived exosomes could promote the repair of the injured spinal cord by regulating the cellular microenvironment of neurons and could be a promising treatment for spinal cord injury. TMEM151A, located at 11q13.2 and encoding transmembrane protein 151A, was recently reported as causative for autosomal dominant paroxysmal kinesigenic dyskinesia (PKD). Here, through comprehensive analysis of sporadic and familial cases, we expand the clinical and mutation spectrum of PKD. In doing so, we clarify the clinical and genetic features of Chinese PKD patients harboring TMEM151A variants and further explore the relationship between TMEM151A mutations and PKD. Whole exome sequencing was performed on 26 sporadic PKD patients and nine familial PKD pedigrees without PRRT2 variants. Quantitative real-time PCR was used to assess the gene expression of frameshift mutant TMEM151A in a PKD patient. TMEM151A variants reported to date were reviewed. Four TMEM151A variants were detected in four unrelated families with 12 individuals, including a frameshift mutation [c.606_607insA (p.Val203fs)], two missense mutations [c.166G > A (p.Gly56Arg) and c.791T > C (p.Val264Ala)], and a non-pathogenic variant [c.994G > A (p.Gly332Arg)]. The monoallelic frameshift mutation [c.606_607insA (p.Val203fs)] may cause TMEM151A mRNA decay, suggesting a potential pathogenic mechanism of haploinsufficiency. Patients with TMEM151A variants had short-duration attacks and presented with dystonia. Our study provides a detailed clinical description of PKD patients with TMEM151A mutations and reports a new disease-causing mutation, expanding the known phenotypes caused by TMEM151A mutations and providing further detail about the pathoetiology of PKD. Diosgenin, a steroidal sapogenin, obtained from Trigonella foenum-graecum, Dioscorea, and Rhizoma polgonati, has shown high potential and interest in the treatment of various cancers such as oral squamous cell carcinoma, laryngeal cancer, esophageal cancer, liver cancer, gastric cancer, lung cancer, cervical cancer, prostate cancer, glioma, and leukemia. This article aims to provide an overview of the in vivo, in vitro, and clinical studies reporting the diosgenin's anticancer effects. Preclinical studies have shown promising effects of diosgenin on inhibiting tumor cell proliferation and growth, promoting apoptosis, inducing differentiation and autophagy, inhibiting tumor cell metastasis and invasion, blocking cell cycle, regulating immunity and improving gut microbiome. Clinical investigations have revealed clinical dosage and safety property of diosgenin. Furthermore, in order to improve the biological activity and bioavailability of diosgenin, this review focuses on the development of diosgenin nano drug carriers, combined drugs and the diosgenin derivatives. Skeleton-based action recognition (SBAR) is an important task in the field of computer vision. Learning effective action representations from skeleton sequences and improving the performance of action recognition models remain challenging problems. To capture effective features from skeleton sequences, a novel feature called a short-term path signature (STPS) is proposed in this work. Based on the STPS, a plug-and-play module is proposed to achieve improved SBAR. In this module, the STPS is applied as input, and a spatial-temporal graph convolutional network (ST-GCN) is used to learn action features. Finally, a multistream ST-GCN is built to achieve SBAR. The proposed method is verified on the NTU-RGB+D dataset. Several ablation experiments are conducted to verify the effectiveness of the proposed module. China ranks first in the world with respect to area and production of potato, an important food and vegetable crop. How to adjust the structure of fertilizer application, to become cost effective and improve quality and efficiency has become an important issue to be solved in potato cultivation. The potential of organic and microbial fertilizers to improve the quality and yield of potato under the condition of reduced chemical fertilizer was investigated for two years in field trials. The experiment was designed to provide a theoretical basis for the reduction of fertilizer application and efficiency model for potato. The results showed that compared with chemical fertilizer alone, the application of organic fertilizer or microbial fertilizer significantly increased potato yield by 23.5–34.5%, increased potato starch content by 4.20–17.5%, reduced potato nitrate content by 9.02–56.0%, and significantly improved soil physical and chemical properties. The microbial fertilizer could significantly increase soil alkaline nitrogen, available phosphorus, available potassium and organic matter content. This conclusion was verified in the 2 year of the trial. The results of two-way ANOVA showed that fertilizer application level had a significant effect on potato nutritional quality, while fertilizer type was the main effector of soil physicochemical properties. The results of redundancy analysis showed that soil alkaline nitrogen, available phosphorus content and pH value had significant effects on the nutritional quality of potato. [ABSTRACT FROM AUTHOR]

In this study, the nonlinear second harmonic generation(SHG) and third harmonic generation(THG) in a GaAlAs/GaAs Rosen–Morse quantum well(QW) are studied theoretically. The analytical expressions of the SHG and THG of the Rosen–Morse QW are given systematically by using the density matrix theory and the iterative method. Also, the energy levels and the wave function of the Rosen–Morse QW system are gained with finite difference method. The effects of the structure parameter and applied electric field on energy levels and nonlinear optical properties are discussed in detail. It is found that the nonlinear SHG and THG are closely related to the structure parameter and applied electric field. The result show that the red- and blue-shift of the resonance peaks of the nonlinear SHG and THG of the system can be achieved by adjusting the system parameters, and the optimal system parameter for obtaining the maximum peak intensity are given. In offshore engineering, the phenomenon of free span often occurs, and the pipeline may have multiple free spans adjacent to each other, forming a multi-span pipeline. The interaction of different spans makes the structural vibration characteristics more complex, which may change the fatigue characteristics of the pipeline and affect the safety of the structure. In this paper, model tests were designed to explore the vortex-induced vibration (VIV) characteristics of multi-span pipelines and investigate the multi-span interaction mechanism. The experimental study mainly focused on the dynamic response of double-span pipelines, and further extended to triple-span pipelines, hoping that the results can be applied to more complex environment. The effects of span-length ratio, buried depth and axial force on VIV of the pipeline were investigated and discussed. The dynamic response of the pipeline varied with the span length. There was obvious interaction between different spans of multi-span pipelines, and the pipe-sediment interaction obviously affected the vibration characteristics of pipeline. The differences of pipeline burial depth and axial force changed the structural stiffness. With the increase of buried depth, the response amplitude presented a downward trend. The spanwise evolutions were asymmetric caused by the pipe-sediment interaction and multi-span interaction. The damage to human body caused by laser and ultraviolet light from electronic products and sunlight has attracted attention. In this paper, on the basis of previous work, the near-infrared absorbing powder Er4Al2O9 was prepared by a more straightforward co-precipitation method, and intended to apply on laser protection. To achieve compatible absorption at 1064 and 1540 nm, the effects of doping Sm3+ and Dy3+ on the phase composition and reflectivity were inquired. The morphology, phase composition and reflectance of the fines were characterized by X-ray diffraction, Scanning Electron Microscopy and the ultraviolet-visible spectrophotometer (UV3600PC). No significant variation on the crystal structure was found of doped Sm3+ and Dy3+. While, the introduction of Sm3+/Dy3+ could greatly change the morphology of Er4Al2O9 powder, which will further affect the reflectivity performance. Through the co-doping of Sm3+ and Dy3+, it is expected to further broaden the characteristic absorption peak at 1100 nm. The results show that the absorption peak of Sm3+ at 1100 nm could be broadened with the increase of Dy3+ doping amount, and the reflectivity of Er0.8Dy2.0Sm1.6Al2O9 at 1064 and 1540 nm reaches the lowest value. Compared with the ordinary un-doped powder, the reflectivity of the co-doped powder at 1064 nm decreased by 26.41%, indicating a good application prospect. [ABSTRACT FROM AUTHOR]

The purpose of this preclinical study in a sheep model was to confirm the feasibility and safety of the LuX-Valve transjugular tricuspid valve (TV) replacement apparatus and to optimize the implantation procedure before beginning first-in-man study. The LuX-Valve was implanted in a sheep model (n = 8) via transjugular approach. Six of eight sheep underwent successful implantation procedure on beating heart. The first two sheep died during the prostheses deployment. In the remaining 6 sheep that survived, postoperative echocardiography results showed there was no paravalvular leakage (PVL) and central tricuspid regurgitation in 5 animals, whereas 1 animal had mild PVL. The mean transvalvular gradient was 1.1 ± 0.9 mm Hg at the 4-week follow-up. No right ventricular outflow tract (RVOT) obstruction, device malposition, pericardial effusion, coronary artery compression, or arrhythmias were observed. This technology may be a promising alternative for TR patients who are at high risk for open-heart surgery. With the development of the Internet and the formal arrival of the era of big data, people record, store and process data in electronic form, while the bulk of the data in the real life are asynchronous event sequence data. For modeling the asynchronous event sequence, neural point process is one of the most mainstream solutions. With the more and more in-depth study of neural point process, in order to boost the prediction accuracy of the model, the complexity of the model cannot be overestimated, or the selected model itself has more nonlinearity. For example, the neural point process based on attention mechanism will lead to great complexity of the model. Meanwhile, with the development of deep learning, people find that the traditional multi-layer perceptron has great potential. Now many model architectures built with pure multi-layer perceptron without attention have been proposed, and the effect is better than the attention mechanism. Therefore, the multi-layer perceptron has been "reborn" and has attracted extensive attention. Inspired by this, we propose the Linear Normalization Attention Hawkes Process (LNAHP), which substitutes the multi-head dot-product attention from the transformer for linear normalization attention, and learns the hidden representation through two linear transformation layers and normalization operation, which markedly reduces the complexity of the model. The performance for different evaluation metrics for the LNAHP is verified and compared to the current baselines on real datasets from different fields and synthetic datasets, which proves the effectiveness of the LNAHP. Objective: Outcome of pediatric acute liver failure (PALF) in countries with limited availability of LT is not well described. We evaluated the outcome and prognostic indicators of PALF in Malaysia where emergency LT for ALF is limited. Methods: In this retrospective review on children < 18 years with PALF, we compared clinical and laboratory parameters between survival after supportive treatment and after LT or succumbed without LT. The predictive values of Liver Injury Unit (LIU; peak laboratory values for international normalized ratio [INR], ammonia, total bilirubin) and upon admission (aLIU) on outcome of PALF was evaluated using receiver operator characteristic (ROC) curves. Results: Of 77 children (39 males [51%]; median age 2.8 years) with PALF, the overall survival was 55% (n = 42); 52% (n = 40) survived with supportive management, 2.6% (n = 2) after LT. As compared to children who survived without LT, children who had LT/died had lower hemoglobin, aspartate transferase, γ-glutamyl transpeptidase (GGT), and higher serum bilirubin, alkaline phosphatase, ammonia, and serum sodium (p < 0.05). On multivariate analysis, significant independent predictor for death or LT were peak bilirubin > 452 μmol/L and peak GGT < 96 IU/L. The C-index of LIU and aLIU score were 0.79 and 0.68, respectively, indicating that LIU score was a good model in predicting outcome of PALF. Conclusions: Overall survival of PALF remained poor. High peak bilirubin and low GGT predict poor outcome of PALF. LIU score is a good model in predicting outcome of PALF and maybe useful in selecting children for emergency LT. [ABSTRACT FROM AUTHOR]

This study conducts a systematic literature review and meta-analysis regarding the potential influence of serum uric acid levels on cerebral small vessel diseases and the cognitive status in the prodromal stages of dementia. We identified four different cerebral small vessel diseases and three specific domains of cognitive performance to be considered in the literature search. The analysis contained 14 studies (13 cross-sectional design and one longitudinal design) with 11,502 participants measuring the relationship between uric acid and cerebral small vessel disease. In both continuous and categorical analyses, significant associations were found between hyperuricemia and cerebral small vessel diseases (continuous data: pooled OR: 1.00, 95%CI: 1.00–1.01 and categorical data: pooled OR: 1.42, 95%CI: 1.15–1.75). For the relationship between uric acid and cognitive performance, 19 studies with 49,901 participants were considered, including eight cohort studies, and 11 cross-sectional studies. The cross-sectional data showed that a marginal relationship existed between uric acid and global cognition (β: 0.00, 95%CI: -0.01–0.00). The pooled analysis of cohort studies indicated that higher uric acid had a deleterious effect on attention and executive function (continuous data: β: -0.02, 95%CI: -0.04–0.00 and categorical data: β: -0.03, 95%CI: -0.07–0.00). Conclusion: Our study indicated that a higher level of uric acid had an adverse effect on brain health. Furthermore, a high level of uric acid is related to cognitive decline in attention and executive function, which may exist a long time before the diagnosis of dementia. [ABSTRACT FROM AUTHOR]

Ischaemic stroke is one of the most common causes of mortality and morbidity.circDlgap4 has been implicated in ischemia/reperfusion injury through an unknown mechanism. Here, we studied the function of circDlgap4/AUF1 in ischaemic stroke and its underlying molecular mechanism. N2a cells and primary mouse cortical neurons were subjected to OGD to mimic neuronal injury during ischemia. BV-2 cells were treated with LPS to mimic neuroinflammation. The MTT assay was used to assess cell viability, while flow cytometry was used to measure cell apoptosis. qRT–PCR, western blotting, immunohistochemistry, and immunostaining were employed to determine the levels of circDlgap4, AUF1, NRF2/HO-1, proinflammatory cytokines, NF-κB pathway–related proteins, and IBA-1. RIP and RNA pulldown assays were employed to validate the interactions of circDlgap4/AUF1, AUF1/NRF2, and AUF1/cytokine mRNAs. mRNA degradation was used to determine the effects on mRNA stability. The tMCAO model was used as an in vivo model of ischaemic stroke. TCC staining and neurological scoring were performed to evaluate ischaemic injury. circDlgap4 was decreased following OGD and during tMCAO. circDlgap4 overexpression inhibited OGD-induced cell death and oxidative stress and LPS-induced increases in proinflammatory cytokines by increasing NRF2/HO-1. Knockdown of AUF1 blocked the effects of circDlgap4 overexpression. Mechanistically, RIP, RNA pulldown, and mRNA degradation assay results showed circDlgap4/AUF1/NRF2 mRNA formed a complex to stabilize NRF2 mRNA. Furthermore, AUF1 directly interacted with TNF-α, IL-1β, and COX-2 mRNAs, and circDlgap4/AUF1 binding promoted the degradation of these mRNAs. Finally, circDlgap4 ameliorated ischaemic injury in vivo. circDlgap4 alleviates ischaemic stroke injury by suppressing oxidative stress and neuroinflammation by binding to AUF1. Topological electronic flattened bands near or at the Fermi level are a promising route towards unconventional superconductivity and correlated insulating states. However, the related experiments are mostly limited to engineered materials, such as moiré systems1–3. Here we present a catalogue of the naturally occuring three-dimensional stoichiometric materials with flat bands around the Fermi level. We consider 55,206 materials from the Inorganic Crystal Structure Database catalogued using the Topological Quantum Chemistry website4,5, which provides their structural parameters, space group, band structure, density of states and topological characterization. We combine several direct signatures and properties of band flatness with a high-throughput analysis of all crystal structures. In particular, we identify materials hosting line-graph or bipartite sublattices—in either two or three dimensions—that probably lead to flat bands. From this trove of information, we create the Materials Flatband Database website, a powerful search engine for future theoretical and experimental studies. We use the database to extract a curated list of 2,379 high-quality flat-band materials, from which we identify 345 promising candidates that potentially host flat bands with charge centres that are not strongly localized on the atomic sites. We showcase five representative materials and provide a theoretical explanation for the origin of their flat bands close to the Fermi energy using the S-matrix method introduced in a parallel work6.A catalogue of the naturally occurring three-dimensional stoichiometric materials with flat bands around the Fermi level provides a powerful search engine for future theoretical and experimental studies. In recent years, mining the knowledge from asynchronous sequences by Hawkes process is a subject worthy of continued attention, and Hawkes processes based on the neural network have gradually become the most hotly researched fields, especially based on the recurrence neural network (RNN). However, these models still contain some inherent shortcomings of RNN, such as vanishing and exploding gradient and long-term dependency problems. Meanwhile, transformer-based on self-attention has achieved great success in sequential modeling like text processing and speech recognition. Although the Transformer Hawkes process (THP) has gained huge performance improvement, THPs do not effectively utilize the temporal information in the asynchronous events, for these asynchronous sequences, the event occurrence instants are as important as the types of events, while conventional THPs simply convert temporal information into position encoding and add them as the input of transformer. With this in mind, we come up with a new kind of Transformer-based Hawkes process model, temporal attention augmented transformer Hawkes Process (TAA-THP), we modify the traditional dot-product attention structure and introduce the temporal encoding into attention structure. We conduct numerous experiments on a wide range of synthetic and real-life datasets to validate the performance of our proposed TAA-THP model, a significant improvement compared with existing baseline models on the different measurements is achieved, including log-likelihood on the test dataset, and prediction accuracies of event types and occurrence times. In addition, through the ablation studies, we vividly demonstrate the merit of introducing additional temporal attention by comparing the performance of the model with and without temporal attention. Neuropathic pain (NP) involves metabolic processes that are regulated by metabolic genes and their non-coding regulator genes such as microRNAs (miRNAs). Here, we aimed at exploring the key miRNA signatures regulating metabolic genes involved in NP pathogenesis. We downloaded NP-related data from public databases and identified differentially expressed microRNAs (miRNAs) and mRNAs through differential gene expression analysis. The miRNA target prediction was performed, and integration with the differentially expressed metabolic genes (DEMGs) was used for constructing the miRNA-DEMG network. Subsequently, functional enrichment analysis and protein–protein interaction (PPI) analysis were performed to explore the role of DEMGs in the regulatory network. The drug prediction was performed based on the DEMGs in the miRNA-DEMG network. A total of 8251 differentially expressed mRNAs (4193 upregulated and 4058 downregulated), and 959 differentially expressed miRNAs (455 upregulated and 504 downregulated) were identified. Moreover, after target gene prediction, a miRNA-DEMG network composed of 22 miRNAs and 113 mRNAs was constructed. The network was constituted of 135 nodes and 236 edges. We found that DEMGs in the network were mainly enriched in metabolic pathways and metabolic processes. A total of 1200 drugs were predicted as potential therapeutics for NP based on the differentially expressed genes, while 170 drugs were predicted for the DEMGs in the miRNA-DEMG network. Conclusively, our study predicted drugs that may be effective against the metabolic changes induced by miRNA dysregulation in NP. A novel ratiometric fluorescence nanoprobe based on long-wavelength emission carbon dots (CDs) was designed for high sensitive and selective detection of Zn2+. The CDs were conveniently prepared by a one-step solvothermal treatment of formamide and glutathione (GSH). Under single excitation wavelength (420 nm), the obtained CDs exhibit three emission peaks at 470, 650, and 685 nm, respectively. For the long-wavelength emission region of the CDs, the fluorescence at 685 nm can be quenched with different levels upon the addition of most metal ions. However, the presence of Zn2+ not only results in the fluorescence quenching at 685 nm effectively but also enhances at 650 nm remarkably, which may be due to the formation of CD-Zn2+ chelate complex inducing the dispersion of CDs aggregates and changes in the group distribution on the surface of CDs. Taking the advantage of the unique fluorescence response induced by Zn2+, the prepared CDs were successfully employed as nanoprobe for self-ratiometric fluorescence determination of Zn2+ with F650/F685 as signal output. A good linear relationship in the concentration range 0.01 to 2 μM, and a detection limit as low as 5.1 nM has been obtained. Purpose: There have been no definite conclusions about the biliopancreatic limb (BPL) and alimentary limb (AL) lengths in Roux-en-Y gastric bypass (RYGB) operations for different populations and BMIs. Western scholars have performed many studies on the lengths of the BPL and AL in patients with a BMI ≥ 35 kg/m2. However, for diabetic Chinese patients with BMI < 35 kg/m2, few people have compared the effects of different BPL and AL lengths on patient prognosis. Patients and Methods: Clinical data were collected prospectively and analyzed retrospectively for 87 patients with type 2 diabetes (T2DM) who underwent RYGB with a BPL of 50 cm and an AL of 50 cm (BPL50/AL50) or with a BPL of 100 cm and an AL of 100 cm (BPL100/AL100) and who were followed up for 5 years. Results: The cohort included 42 patients in the BPL50/AL50 group and 45 patients in the BPL100/AL100 group. At 5 years, there were significant differences in BMI, total weight loss (TWL%), glycosylated hemoglobin, and homeostasis model assessment insulin resistance between BPL50/AL50 and BPL100/AL100 (P < 0.05). Diabetes remission rate of the BPL100/AL100 group was significantly higher than that of the BPL50/AL50 group. Diabetes remission at 1 year after surgery correlated with the length limb (BPL + AL), duration of diabetes and TWL%. There was no difference in complications between BPL50/AL50 and BPL100/AL100. Conclusions: RYGB with BPL100/AL100 is a safe and effective treatment for diabetic patients with a BMI < 35 kg/m2 and offers significant improvement in weight loss and glycemic control. Various exotic topological phases of Floquet systems have been shown to arise from crystalline symmetries. Yet, a general theory for Floquet topology that is applicable to all crystalline symmetry groups is still in need. In this work, we propose such a theory for (effectively) non-interacting Floquet crystals. We first introduce quotient winding data to classify the dynamics of the Floquet crystals with equivalent symmetry data, and then construct dynamical symmetry indicators (DSIs) to sufficiently indicate the inherently dynamical Floquet crystals. The DSI and quotient winding data, as well as the symmetry data, are all computationally efficient since they only involve a small number of Bloch momenta. We demonstrate the high efficiency by computing all elementary DSI sets for all spinless and spinful plane groups using the mathematical theory of monoid, and find a large number of different nontrivial classifications, which contain both first-order and higher-order 2+1D anomalous Floquet topological phases. Using the framework, we further find a new 3+1D anomalous Floquet second-order topological insulator (AFSOTI) phase with anomalous chiral hinge modes. A general theory for Floquet topology applicable to all crystalline symmetry groups is lacking. Here, the authors propose such a theory for noninteracting Floquet crystals and predict an inversion-protected Floquet higher-order topological phase with anomalous chiral hinge modes. A novel ratiometric fluorescence nanoprobe based on carbon dots (CDs) and Cu nanoclusters (CuNCs) was designed for the label-free determination of uric acid (UA). The metal-organic framework (MOF) encapsulated CuNCs (ZIF-CuNC), and nitrogen-doped CDs can self-assemble into well-defined spherical nanocomposites (CD@ZIF-CuNC) due to physical adsorption. Under the excitation wavelength of 360 nm, the CD@ZIF-CuNC nanocomposites exhibit two evident intrinsic emissions peaked at 460 nm (CDs) and 620 nm (ZIF-CuNC), respectively. In the presence of H2O2, the fluorescence of CD@ZIF-CuNC at 620 nm is quenched remarkably within 1 min, while little effect on the emission at 460 nm is observed. Therefore, taking the fluorescence at 620 nm as the report signal and 460 nm as the reference signal, ratiometric quantitative determination of H2O2 was achieved with a linear range of 1–100 μM and a detection limit of 0.30 μM. The CD@ZIF-CuNC nanoprobe was successfully applied to the determination of UA that is catalyzed by uricase to produce H2O2, obtaining the linear range of 1–30 μM and the detection limit of 0.33 μM. Odorant-binding proteins (OBPs) are prevalent in the antennal transcriptomes of different orders of insects. Studies on OBPs have focused on their role in the insect chemosensory system, but knowledge of their functions in the insect testis is limited. We sequenced the transcriptomes of the Athetis dissimilis reproductive organs and analyzed the expression of AdisOBP genes in different tissues. We identified 23 OBPs in the testis and ovaries and 31 OBPs in antennal transcriptomes. The results of real-time quantitative PCR revealed that 23 of the 54 OBP genes were highly expressed in both female and male antennae, including three that exhibited male-biased expression and 15 that exhibited female-biased expression. A total of 24 OBPs were highly expressed in the testis of A. dissimilis, while expression of OBPs in the ovaries was very low. These findings highlight the functional diversity of OBPs in insects and can facilitate further studies on the OBPs in A. dissimilis and lepidopteran species. [ABSTRACT FROM AUTHOR]

Objective: To investigate whether ginsenoside Rb1 (Rb1) can protect human umbilical vein endothelial cells (HUVECs) against high glucose-induced apoptosis and examine the underlying mechanism. Methods: HUVECs were divided into 5 groups: control group (5.5 mmol/L glucose), high glucose (HG, 40 mmol/L) treatment group, Rb1 (50 µ mol/L) treatment group, Rb1 plus HG treatment group, and Rb1 and 3-(1H-1,2,3-triazol-4-yl) pyridine (3-TYP, 16 µ mol/L) plus HG treatment group. Cell viability was evaluated by cell counting kit-8 assay. Mitochondrial and intracellular reactive oxygen species were detected by MitoSox Red mitochondrial superoxide indicator and dichloro-dihydro-fluorescein diacetate assay, respectively. Annexin V/propidium iodide staining and fluorescent dye staining were used to measure the apoptosis and the mitochondrial membrane potential of HUVECs, respectively. The protein expressions of apoptosis-related proteins [Bcl-2, Bax, cleaved caspase-3 and cytochrome c (Cyt-c)], mitochondrial biogenesis-related proteins [proliferator-activated receptor gamma coactivator 1-alpha, nuclear respiratory factor-1 and mitochondrial transcription factor A)], acetylation levels of forkhead box O3a and SOD2, and sirtuin-3 (SIRT3) signalling pathway were measured by immunoblotting and immunoprecipitation. Results: Rb1 ameliorated survival in cells in which apoptosis was induced by high glucose (P<0.05 or P<0.01). Upon the addition of Rb1, mitochondrial and intracellular reactive oxygen species generation and malondialdehyde levels were decreased (P<0.01), while the activities of antioxidant enzymes were increased (P<0.05 or P<0.01). Rb1 preserved the mitochondrial membrane potential and reduced the release of Cyt-c from the mitochondria into the cytosol (P<0.01). In addition, Rb1 upregulated mitochondrial biogenesis-associated proteins (P<0.01). Notably, the cytoprotective effects of Rb1 were correlated with SIRT3 signalling pathway activation (P<0.01). The effect of Rb1 against high glucose-induced mitochondria-related apoptosis was restrained by 3-TYP (P<0.05 or P<0.01). Conclusion: Rb1 could protect HUVECs from high glucose-induced apoptosis by promoting mitochondrial function and suppressing oxidative stress through the SIRT3 signalling pathway. Artificial enzyme cascade systems with confinement effect are highly important in synthetic biology and biomedicine. Herein, a framework nucleic acid-based confined enzyme cascade (FNA-CEC) for synergistic cancer therapy in vivo was developed. The FNA-CEC consisted of glucose oxidase and horseradish peroxidase precisely assembled on an addressable DNA tetrahedron scaffold within few nanometers. Glucose oxidase (GOx) can trigger efficient glucose depletion for tumor starvation therapy, and increase the local concentration of H2O2in situ for enhanced downstream horseradish peroxidase (HRP)-activated prodrug therapy. Due to the spatial-confinement on DNA tetrahedron scaffold, the efficiency of intermediate metabolites transportation between the enzyme cascades was improved. Moreover, FNA-CEC was applied for efficient synergistic cancer therapy in vitro and in vivo. Chemosensory receptors in the dendritic membrane of olfactory cells are critical for the molecular recognition and discrimination of odorants. Tropidothorax elegans is a major pest of agricultural, ornamental, and medicinal plants. However, very little is known about olfactory genes in T. elegans. The purpose of this study was to obtain chemosensory receptor genes by sequencing the antennal transcriptome of T. elegans using Illumina sequencing technology. We identified 153 candidate chemosensory receptors, including 121 olfactory receptors (including one olfactory receptor co-receptor), 10 ionotropic receptors (including one IR8a and one IR25a), and 22 gustatory receptors (GRs). TeleOR76, 104 and 112 displayed more highly expression level than TeleOrco. Other TeleGR genes were expressed at very low levels except TeleGR1 and 20. TeleIR76b was the most highly expressed among TeleIR genes. Our results provide valuable biological information for studies of the olfactory communication system of T. elegans. As the main biological components of the air ecosystem, airborne microorganisms play an important role in ecosystem function, and most existing studies focus on their health and environmental effects, while ignoring ecological effects. In this study, air microbial samples from two sampling points in the coastal city of Qingdao were collected by a large volume air sampler. The Illumina MiSeq and PacBio high-throughput sequencing methods were used to analyze the microbial community structure and diversity of the two sampling points. The results showed that there were significant differences in the number and relative abundances of microorganisms identified by the two platforms, and with the refinement of the sample classification, the differences in the community became increasingly significant. Proteobacteria and Firmicutes were the dominant bacterial phyla among the airborne microorganisms. MiSeq sequencing showed that the relative abundances of unidentified bacteria were as high as 8.03–16.42% in MiSeq sequencing, while they were almost undetected in PacBio sequencing. Based on the different sequencing platforms, Bacilli, Gammaproteobacteria and Alphaproteobacteria could be seen as significant gaps in the two sampling points. Betaproteobacteria showed differences only in the coastal area, while the other 14 classes were differentially abundant in the urban area. Pseudomonas was the main genus in the aerosol, followed by Lactococcus. The relative abundances of bacterial taxa displayed statistically significant differences in the different functional areas. There were more diverse microbes in the urban area than in the coastal area; moreover, the relative abundances of dominant genera showed significant differences, such as Variovorax and Deinococcus. The discoveries of intrinsically magnetic topological materials, including semimetals with a large anomalous Hall effect and axion insulators1–3, have directed fundamental research in solid-state materials. Topological quantum chemistry4 has enabled the understanding of and the search for paramagnetic topological materials5,6. Using magnetic topological indices obtained from magnetic topological quantum chemistry (MTQC)7, here we perform a high-throughput search for magnetic topological materials based on first-principles calculations. We use as our starting point the Magnetic Materials Database on the Bilbao Crystallographic Server, which contains more than 549 magnetic compounds with magnetic structures deduced from neutron-scattering experiments, and identify 130 enforced semimetals (for which the band crossings are implied by symmetry eigenvalues), and topological insulators. For each compound, we perform complete electronic structure calculations, which include complete topological phase diagrams using different values of the Hubbard potential. Using a custom code to find the magnetic co-representations of all bands in all magnetic space groups, we generate data to be fed into the algorithm of MTQC to determine the topology of each magnetic material. Several of these materials display previously unknown topological phases, including symmetry-indicated magnetic semimetals, three-dimensional anomalous Hall insulators and higher-order magnetic semimetals. We analyse topological trends in the materials under varying interactions: 60 per cent of the 130 topological materials have topologies sensitive to interactions, and the others have stable topologies under varying interactions. We provide a materials database for future experimental studies and open-source code for diagnosing topologies of magnetic materials. High-throughput calculations are performed to predict approximately 130 magnetic topological materials, with complete electronic structure calculations and topological phase diagrams.
