Human genetic studies have linked rare coding variants in microglial genes, such as TREM2, and more recently PLCG2 to Alzheimer's disease (AD) pathology. The P522R variant in PLCG2 has been shown to confer protection for AD and to result in a subtle increase in enzymatic activity. PLCγ2 is a key component of intracellular signal transduction networks and induces Ca2+ signals downstream of many myeloid cell surface receptors, including TREM2. To explore the relationship between PLCγ2 and TREM2 and the role of PLCγ2 in regulating immune cell function, we generated human induced pluripotent stem cell (iPSC)- derived macrophages from isogenic lines with homozygous PLCG2 knockout (Ko). Stimulating TREM2 signalling using a polyclonal antibody revealed a complete lack of calcium flux and IP1 accumulation in PLCγ2 Ko cells, demonstrating a non-redundant role of PLCγ2 in calcium release downstream of TREM2. Loss of PLCγ2 led to broad changes in expression of several macrophage surface markers and phenotype, including reduced phagocytic activity and survival, while LPS-induced secretion of the inflammatory cytokines TNFα and IL-6 was unaffected. We identified additional deficits in PLCγ2- deficient cells that compromised cellular adhesion and migration. We developed nuclear single nucleotide polymorphism (SNP) markers for the Painted Bunting (Passerina ciris) using next-generation sequencing genotyping on the Illumina HiSeq4000 platform. Following the best-RAD approach, we sequenced 96 individuals from five breeding populations occurring in the United States. We assessed the discerning power of 105,000 SNPs and confirmed the quality of these markers in population differentiation studies. We detected outlier SNPs within or near protein coding genes, including sequences coding for olfactory receptors which may relate to migratory navigation. Finally, we provide a novel list of 33 primer pairs flanking SNPs to link individuals to regional populations and improve our understanding of the evolution of this migratory songbird. Recent outbreaks of Ebola virus disease and Zika virus disease highlight the need for disseminating accurate predictions of emerging zoonotic viruses to national governments for disease surveillance and response. Although there are published maps for many emerging zoonotic viruses, it is unknown if there is agreement among different models or if they are concordant with national expert opinion. Therefore, we reviewed existing predictions for five high priority emerging zoonotic viruses with national experts in Cameroon to investigate these issues and determine how to make predictions more useful for national policymakers. Predictive maps relied primarily on environmental parameters and species distribution models. Rift Valley fever virus and Crimean-Congo hemorrhagic fever virus predictions differed from national expert opinion, potentially because of local livestock movements. Our findings reveal that involving national experts could elicit additional data to improve predictions of emerging pathogens as well as help repackage predictions for policymakers. Plasmodium (Novyella) megaglobularis n. sp. was recorded in the olive sunbird Cyanomitra olivacea, and Plasmodium (Novyella) globularis n. sp. and Haemoproteus (Parahaemoproteus) vacuolatus n. sp. were found in the yellow-whiskered greenbul Andropadus latirostris in rainforests of Ghana and Cameroon. These parasites are described based on the morphology of their blood stages and a segment of the mitochondrial cytochrome b gene, which can be used for molecular identification and diagnosis of these species. Illustrations of blood stages of new species are given, and phylogenetic analysis identifies deoxyribonucleic acid (DNA) lineages closely related to these parasites. Traditional taxonomy of avian pigment-forming haemosporidians of the families Plasmodiidae and Haemoproteidae is discussed based on the recent molecular phylogenies of these parasites. We conclude that further work to increase the number of precise linkages between haemosporidian DNA sequences and their corresponding morphospecies is needed before revising the current classification of haemosporidians. Recent studies have demonstrated that changes in scale number are correlated with ecological variables such as precipitation, and this suggests that scale number may be under selection to maintain water balance in reptiles. Here, we present new evidence that variation in scale numbers within and among species of Anolis lizards is under ecologically based natural selection. We measured scalation of the brown anole, Anolis sagrei, in two habitat types on each of five islands in the Bahamas. We also measured scalation for 12 species of anole representing six different ecomorphs from the Greater Antilles. Within populations of A. sagrei, scale numbers increased with increasing precipitation and with decreasing temperature in open arid habitats. Variation measured among species of Anolis from the Greater Antilles showed similar patterns with temperature, precipitation, and elevation. Independent contrasts using scale count data indicated that variation in scale number was congruent within and between species, even after accounting for the influence of phylogeny. We measured natural selection (survival to maturity) on scale number in A. sagrei over two different habitat types in the Bahamas. Patterns of natural selection were congruent with the correlational results described. Finally, results from a breeding experiment in the laboratory provide preliminary evidence that variation in scale number is heritable, and suggests a mechanism for generating these correlations. Our results provide new evidence that the diversification of anoles has been shaped by natural selection and that ecologically based selection pressures help explain diversification at both the population and species levels. Islands are considered to be natural laboratories in which to examine evolution because of the implicit assumption that limited gene flow allows tests of evolutionary processes in isolated replicates. Here we show that this well-accepted idea requires re-examination. Island inundation during hurricanes can have devastating effects on lizard populations in the Bahamas. After severe storms, islands may be recolonized by over-water dispersal of lizards from neighbouring islands. High levels of gene flow may homogenize genes responsible for divergence, and are widely viewed as a constraining force on evolution. Ultimately, the magnitude of gene flow determines the extent to which populations diverge from one another, and whether or not they eventually form new species. We show that patterns of gene flow among island populations of Anolis lizards are best explained by prevailing ocean currents, and that over-water dispersal has evolutionary consequences. Across islands, divergence in fitness-related morphology decreases with increasing gene flow. Results suggest that over-water dispersal after hurricanes constrains adaptive diversification in Anolis lizards, and that it may have an important but previously undocumented role in this classical example of adaptive radiation. Altitudinal migration and dispersal is an important component of the life history of several temperate and tropical birds but remains poorly understood due to the limited success of mark and recapture techniques. Stable isotopes of hydrogen (δD) in rainfall, and to a lesser extent, carbon (δ13C) in plants are known to change with altitude and hence may provide the basis of a technique for tracking the altitudinal movements in birds and other wildlife. We investigated the potential for this technique by measuring δ13C, δD, and δ15N values in tail feathers of eight species of hummingbirds (Phaethornis malaris, P. syrmatophorus, P. guy, Adelomyia melanogenys, Coeligena torquata, C. lutetiae, Metallura baroni, M. williami) along an altitudinal gradient (300–3,290 m asl) in the Andes Mountains of Ecuador. Feather δ13C and δD values were correlated and each changed significantly with elevation above 400 m. In general, we found good agreement between feather δD values and those predicted from a generalized relationship of precipitation and surface water δD with altitude. Similarly, feather δ13C values showed an enrichment of ~1.5‰ per 1,000 m over the linear portion of the elevational response. Stable-nitrogen isotope values were variable, and so did not provide useful information on elevation in birds, apart from trophic effects. Overall there appears to be good potential for using the (δD, δ13C) stable isotope approach to track altitudinal movements and to elucidate previously unrecognized patterns of life history variation in both temperate and tropical species that migrate across elevational isotopic gradients. We studied two species of Ceratogymna hornbills, the black-casqued hornbill, C. atrata, and the white-thighed hornbill, C. cylindricus, in the tropical forests of Cameroon, to understand their movement patterns and evaluate their effectiveness as seed dispersers. To estimate hornbill contribution to a particular tree species' seed shadow we combined data from movements, determined by radio-tracking, with data from seed passage trials. For 13 individuals tracked over 12 months, home range varied between 925 and 4,472 ha, a much larger area than reported for other African avian frugivores. Seed passage times ranged from 51 to 765 min, with C. atrata showing longer passage times than C. cylindricus, and larger seeds having longer gut retention times than smaller seeds. Combining these data, we estimated that seed shadows were extensive for the eight tree species examined, with approximately 80% of seeds moved more than 500 m from the parent plant. Maximum estimated dispersal distances for larger seeds were 6,919 and 3,558 m for C. atrata and C. cylindricus, respectively. The extent of hornbill seed shadows suggests that their influence in determining forest structure will likely increase as other larger mammalian dispersers are exterminated. [ABSTRACT FROM AUTHOR]

This paper presents an empirical analysis of individuals in policy formulating and policy implementing roles. Data for the comparison are from interviews with a random sample of 119 New Zealand middle-level public administrators from 27 government departments. The individuals in the policy formulating roles were slightly younger; however, those in the formulating and implementing roles varied little from each other in terms of social background, educational attainment and career patterns. Few differences were evident in regard to job satisfaction, decisional authority, and hierarchical relations. Significant differences between policy formulators and policy implementors were discovered in terms of work load, career aspirations, and awareness of political influences in governmental policymaking. From the evidence of this study, the New Zealand administrative system does not allocate policy formulating roles to individuals different from those who implement policies. There is an implicit assumption in most policy studies that once a policy has been formulated the policy will be implemented. This assumption is invalid for policies formulated in many Third World nations and for types of policies in Western societies. Third World governments tend to formulate broad, sweeping policies, and governmental bureaucracies often lack the capacity for implementation. Interest groups, opposition parties, and affected individuals and groups often attempt to influence the implementation of policy rather than the formulation of policy. A model of the policy implementation process is presented. Policy implementation is seen as a tension generating force in society. Tensions are generated between and within four components of the implementing process: idealized policy, implementing organization, target group, and environmental factors. The tensions result in transaction patterns which may or may not match the expectations of outcome of the policy formulators. The transaction patterns may become crystallized into institutions. Both the transaction patterns and the institutions may generate tensions which, by feedback to the policymakers and implementors, may support or reject further implementation of the policy. By application of the model, policymakers can attempt to minimize disruptive tensions which can result in the failure of policy outcomes to match policy expectations. [ABSTRACT FROM AUTHOR]

Scalable quantum processors require tunable two-qubit gates that are fast, coherent and long-range. The Heisenberg exchange interaction offers fast and coherent couplings for spin qubits, but is intrinsically short-ranged. Here, we demonstrate that its range can be increased by employing a multielectron quantum dot as a mediator, while preserving speed and coherence of the resulting spin-spin coupling. We do this by placing a large quantum dot with 50-100 electrons between a pair of two-electron double quantum dots that can be operated and measured simultaneously. Two-spin correlations identify coherent spin-exchange processes across the multielectron quantum dot. We further show that different physical regimes of the mediated exchange interaction allow a reduced susceptibility to charge noise at sweet spots, as well as positive and negative coupling strengths up to several gigahertz. These properties make multielectron dots attractive as scalable, voltage-controlled coherent coupling elements. Controllable two-qubit interactions are necessary to build a functional quantum computer. Here the authors demonstrate fast, coherent swapping of two spin states mediated by a long, multi-electron quantum dot that could act as a tunable coupler mediating interactions between multiple qubits. Pyrenestes finches are unique among birds in showing a non-sex-determined polymorphism in bill size and are considered a textbook example of disruptive selection. Morphs breed randomly with respect to bill size, and differ in diet and feeding performance relative to seed hardness. Previous breeding experiments are consistent with the polymorphism being controlled by a single genetic factor. Here, we use genome-wide pooled sequencing to explore the underlying genetic basis of bill morphology and identify a single candidate region. Targeted resequencing reveals extensive linkage disequilibrium across a 300 Kb region containing the insulin-like growth factor 1 (IGF1) gene, with a single 5-million-year-old haplotype associating with phenotypic dominance of the large-billed morph. We find no genetic similarities controlling bill size in the well-studied Darwin’s finches (Geospiza). Our results show how a single genetic factor may control bill size and provide a foundation for future studies to examine this phenomenon within and among avian species. [ABSTRACT FROM AUTHOR]
