Purpose: We sought to evaluate the incidence, natural history, and management of cystic spinal lesions following myelomeningocele/myeloschisis closure. Methods: We performed a single-center retrospective review of all patients who underwent myelomeningocele/myeloschisis closure from 2013 to 2018 with follow-up to 5 years old. Results: We analyzed 100 fetal repairs and 81 postnatal closures from 305 total surgeries. Patients within this cohort systematically underwent serial MRI scans of the lumbar spine and had clinical follow-up until at least 5 years of age. Ninety-three (51.2%) developed radiographic evidence of cystic lesions with 28 (30.1%) requiring surgical intervention. Presence of cysts was higher in fetal repair (67/100, 67%) compared with postnatal (26/81, 32.1%; p < 0.01). Of the 93 patients with radiographic cysts, 28 (30.1%) underwent surgical resection at a median age of 27.6 months old ([Q1, Q3], [13.0, 48.6 months]). Fetal repair patients had a higher rate (26/67, 38.8%) of cysts requiring surgical resection compared with postnatal closure (2/26, 7.7%, p value < 0.01). Pathology demonstrated 18 of resected cysts were dermoid, 8 were epidermoid, and 2 were fibrous tissue. Post-operatively, no patients experienced a worsened ambulation status. Bladder compliance showed a non-significant trend toward improvement. Conclusions: Cystic lesions in myelomeningocele/myeloschisis patients are common findings that result in nerve root tethering. We propose regular screening in both symptomatic and asymptomatic patients to circumvent nerve injury. Most cystic lesions do not require surgical resection though fetal repair is associated with a higher incidence of operative cysts. One longitudinal study of married couples and one experiment tested the hypothesis that the experience of sexual desire for an alternative sexual partner might heighten feelings of desire for one's long-term romantic partner, and conversely, sexual desire for one's long-term partner might heighten desire for alternative partners. A daily-diary study of newlywed couples revealed that (a) on days people reported heightened interest in alternative partners, they also reported increased desire to have sex with their partner and (b) on days people reported heightened desire to have sex with their partner, they also reported increased interest in alternative partners. An experimental study of partnered individuals revealed that people primed with sexual desire for an alternative partner reported increased sexual desire for their romantic partner (relative to a control condition). People primed with sexual desire for their romantic partner, however, did not report increased sexual desire for alternatives. Taken together, these findings support evolutionary perspectives on the function of sexual desire. Findings are consistent with the broader hypothesis that sexual desire is not partner-specific. Purpose: Historically, the presence of gray matter heterotopia was a concern for adverse postnatal neurocognitive status in patients undergoing fetal closure of open spinal dysraphism. The purpose of this study was to evaluate neurodevelopmental outcomes and the onset of seizures during early childhood in patients with a prenatal diagnosis of myelomeningocele/myeloschisis (MMC) and periventricular nodular heterotopia (PVNH). Methods: All patients evaluated at the Center for Fetal Diagnosis and Treatment with a diagnosis of MMC between June 2016 to March 2023 were identified. PVNH was determined from prenatal and/or postnatal MRI. The Bayley Scales of Infant and Toddler Development (edition III or IV) were used for neurodevelopmental assessments. Patients were screened for seizures/epilepsy. Results: Of 497 patients evaluated with a prenatal diagnosis of MMC, 99 were found to have PVNH on prenatal MRI, of which 35 had confirmed PVNH on postnatal imaging. From the 497 patients, 398 initially did not exhibit heterotopia on prenatal MRI, but 47 of these then had confirmed postnatal PVNH. The presence of PVNH was not a significant risk factor for postnatal seizures in early childhood. The average neurodevelopmental scores were not significantly different among heterotopia groups for cognitive, language, and motor domains. Conclusion: The presence of PVNH in patients with a prenatal diagnosis of MMC does not indicate an increased risk for neurodevelopmental delay at 1 year of age. We did not demonstrate an association with seizures/epilepsy. These findings can aid clinicians in prenatal consultation regarding fetal repair of open spinal dysraphism. The current wave of the opioid epidemic has contributed to a record number of drug-related overdoses and a significant proportion of people who experience opioid use disorder are admitted to local jails. These correctional facilities serve as the principal entry point to the criminal justice system as nearly every person who is taken into custody is admitted to a local detention center. Although jails are recognized as primary intervention points for people who may require treatment for opioid use disorder, services in these facilities remain deficient. The absence of jail-based treatment has become a pressing concern as the number of drug-related deaths in custody continues to rise and the risk of post-release overdose also remains high. The present study draws on the opioid-related module of the 2019 Bureau of Justice Statistics' Census of Jails to assess the relationships between the characteristics of 2588 local detention centers and the availability of treatment services. These specific approaches included screening for opioid use disorder, providing medication to manage withdrawal symptoms, administering medication for opioid use disorder (MOUD), providing overdose reversal medication at the time of release, and linking people with community-based care following release from the detention center. The results demonstrate facilities located in the Northeast, larger jails, those in urban areas, and detention centers with higher turnover rates are significantly more likely to provide a wider variety of opioid treatment services. These findings have important implications for the prioritization of policies and the allocation of resources to support the adoption of opioid treatment services in local jails. Climate change can exacerbate the effects of environmental pollutants on aquatic organisms. Pollutants such as human antidepressants released from wastewater treatment plants have been shown to impact life-history traits of amphibians. We exposed tadpoles of the wood frog Lithobates sylvaticus to two temperatures (20 °C and 25 °C) and two antidepressants (fluoxetine and venlafaxine), and measured timing of metamorphosis, mass at metamorphosis, and two behaviors (startle response and percent motionless). Antidepressants significantly shortened time to metamorphosis at 20 °C, but not at 25 °C. At 25 °C, tadpoles metamorphosed significantly faster than those at 20 °C independent of antidepressant exposure. Venlafaxine reduced body mass at 25 °C, but not at 20 °C. Temperature and antidepressant exposure affected the percent of tadpoles showing a startle response. Tadpoles at 20 °C displayed significantly more responses than at 25 °C. Exposure to fluoxetine also increased the percent of tadpoles showing a startle response. Venlafaxine reduced the percent of motionless tadpoles at 25 °C but not at 20 °C. While our results showed that antidepressants can affect the timing of metamorphosis in tadpoles, warmer temperatures overrode these effects and caused a reduction in an important reaction behavior (startle response). The carbohydrate fraction of most mammalian milks contains a variety of oligosaccharides that encompass a range of structures and monosaccharide compositions. Human milk oligosaccharides have received considerable attention due to their biological roles in neonatal gut microbiota, immunomodulation, and brain development. However, a major challenge in understanding the biology of milk oligosaccharides across other mammals is that reports span more than 5 decades of publications with varying data reporting methods. In the present study, publications on milk oligosaccharide profiles were identified and harmonized into a standardized format to create a comprehensive, machine-readable database of milk oligosaccharides across mammalian species. The resulting database, MilkOligoDB, includes 3193 entries for 783 unique oligosaccharide structures from the milk of 77 different species harvested from 113 publications. Cross-species and cross-publication comparisons of milk oligosaccharide profiles reveal common structural motifs within mammalian orders. Of the species studied, only chimpanzees, bonobos, and Asian elephants share the specific combination of fucosylation, sialylation, and core structures that are characteristic of human milk oligosaccharides. However, agriculturally important species do produce diverse oligosaccharides that may be valuable for human supplementation. Overall, MilkOligoDB facilitates cross-species and cross-publication comparisons of milk oligosaccharide profiles and the generation of new data-driven hypotheses for future research. The Dresden-II reactor experiment has recently reported a suggestive evidence for the observation of coherent elastic neutrino-nucleus scattering, using a germanium detector. Given the low recoil energy threshold, these data are particularly interesting for a low-energy determination of the weak mixing angle and for the study of new physics leading to spectral distortions at low momentum transfer. Using two hypotheses for the quenching factor, we study the impact of the data on: (i) The weak mixing angle at a renormalization scale of ~ 10 MeV, (ii) neutrino generalized interactions with light mediators, (iii) the sterile neutrino dipole portal. The results for the weak mixing angle show a strong dependence on the quenching factor choice. Although still with large uncertainties, the Dresden-II data provide for the first time a determination of sin2θW at such scale using coherent elastic neutrino-nucleus scattering data. Tight upper limits are placed on the light vector, scalar and tensor mediator scenarios. Kinematic constraints implied by the reactor anti-neutrino flux and the ionization energy threshold allow the sterile neutrino dipole portal to produce up-scattering events with sterile neutrino masses up to ~ 8 MeV. In this context, we find that limits are also sensitive to the quenching factor choice, but in both cases competitive with those derived from XENON1T data and more stringent that those derived with COHERENT data, in the same sterile neutrino mass range. Sex is integral to maintaining a satisfying long-term romantic relationship such as marriage. It is thus important to identify the factors that promote sexual satisfaction in these relationships. To this end, we examined the extent to which a crucial evolved individual difference—sexual disgust sensitivity—impacts people's sexual satisfaction and relationship satisfaction. Using a two-year longitudinal study of 102 newlywed couples (204 individuals), we demonstrated that, rather than exerting main effects, the interaction of both couple members' sexual disgust sensitivities was indirectly associated with marital satisfaction through sexual satisfaction. People whose partners' sexual disgust sensitivities were relatively similar (versus dissimilar) to their own maintained higher levels of sexual satisfaction across the first two years of marriage, which was associated with similarly elevated marital satisfaction. Not only do these findings highlight the importance of integrating evolutionary perspectives and relationship science, they underscore the value of conducting dyadic research to examine the unique intersection of both couple members' characteristics for people's relationship outcomes. Introduction: Zoledronic acid (ZA) is an intravenous bisphosphonate used to treat pediatric osteoporosis. Adverse events including hypocalcemia and acute phase reaction (APR) are common following first-infusion. The purpose of this report is to describe implementation of a ZA clinical practice guideline and the subsequent process changes to improve adherence to aspects of the protocol related to safety and efficacy. Methods: Quality assurance was evaluated by chart review over a 5-year period to compare the prevalence of hypocalcemia and APR to published data. A quality improvement (QI) initiative consisting of process changes including the addition of an endocrine RN to coordinate infusions and a shift to patient/family self-scheduling of infusions was conducted. The effect of the interventions on safety (completion of pre- and post-infusion bloodwork) and efficacy (receipt of all prescribed infusions) outcomes was evaluated. Results: Seventy-two patients received 244 infusions over the period. The frequency of hypocalcemia (22%) and APR (31%) was consistent with prior reports. 99% of patients received pre-infusion bloodwork, 78% received post-first-infusion bloodwork, and 47% received all prescribed infusions. QI initiatives increased the percentage of patients receiving post-first-infusion bloodwork from 67 to 79% and those receiving all infusions from 62 to 74%, but fell short of the goal of 90%. Conclusions: The implementation of a standardized protocol for ZA use in children was successful in confirming patient eligibility with pre-infusion bloodwork but failed to ensure that patients obtained post-first-infusion bloodwork and received all prescribed infusions. Reactor neutrino experiments provide a rich environment for the study of axionlike particles (ALPs). Using the intense photon flux produced in the nuclear reactor core, these experiments have the potential to probe ALPs with masses below 10 MeV. We explore the feasibility of these searches by considering ALPs produced through Primakoff and Compton-like processes as well as nuclear transitions. These particles can subsequently interact with the material of a nearby detector via inverse Primakoff and inverse Compton-like scatterings, via axio-electric absorption, or they can decay into photon or electron-positron pairs. We demonstrate that reactor-based neutrino experiments have a high potential to test ALP-photon couplings and masses, currently probed only by cosmological and astrophysical observations, thus providing complementary laboratory-based searches. We furthermore show how reactor facilities will be able to test previously unexplored regions in the ∼MeV ALP mass range and ALP-electron couplings of the order of gaee ∼ 10−8 as well as ALP-nucleon couplings of the order of g ann 1 ∼ 10−9, testing regions beyond TEXONO and Borexino limits. With large active volume sizes dark matter direct detection experiments are sensitive to solar neutrino fluxes. Nuclear recoil signals are induced by 8B neutrinos, while electron recoils are mainly generated by the pp flux. Measurements of both processes offer an opportunity to test neutrino properties at low thresholds with fairly low backgrounds. In this paper we study the sensitivity of these experiments to neutrino magnetic dipole moments assuming 1, 10 and 40 tonne active volumes (representative of XENON1T, XENONnT and DARWIN), 0.3 keV and 1 keV thresholds. We show that with nuclear recoil measurements alone a 40 tonne detector could be as competitive as Borexino, TEXONO and GEMMA, with sensitivities of order 8.0 × 10−11μB at the 90% CL after one year of data taking. Electron recoil measurements will increase sensitivities way below these values allowing to test regions not excluded by astrophysical arguments. Using electron recoil data and depending on performance, the same detector will be able to explore values down to 4.0 × 10−12μB at the 90% CL in one year of data taking. By assuming a 200-tonne liquid xenon detector operating during 10 years, we conclude that sensitivities in this type of detectors will be of order 10−12μB. We classify new physics signals in coherent elastic neutrino-nucleus scattering (CEνNS) processes induced by 8B solar neutrinos in multi-ton xenon dark matter (DM) detectors. Our analysis focuses on vector and scalar interactions in the effective and light mediator limits after considering the constraints emerging from the recent COHERENT data and neutrino masses. In both cases we identify a region where measurements of the event spectrum alone suffice to establish whether the new physics signal is related with vector or scalar couplings. We identify as well a region where measurements of the recoil spectrum are required so to establish the nature of the new interaction, and categorize the spectral features that enable distinguishing the vector from the scalar case. We demonstrate that measurements of the isospin nature of the new interaction and thereby removal of isospin related degeneracies are possible by combining independent measurements from two different detectors. The expression of CD64 in neutrophils (nCD64) has shown utility in the diagnosis of sepsis. The aim of this study was to assess the usefulness of nCD64 expression to identify patients with community-acquired pneumonia (CAP) at risk of a poor outcome. A prospective study of nCD64 expression (determined by flow cytometry) in patients with CAP was performed. The sensitivity/specificity of nCD64 in predicting poor outcome [defined as intensive care unit (ICU) admission and/or clinical deterioration after arrival at the emergency department] was calculated. Eighty-three adults with CAP were included; 14.5 % had septic shock, 19.3 % required ICU admission, and 10.8 % presented clinical deterioration after admission. The mean of the median fluorescence intensity (MFI) of nCD64 expression was 1140 (±1097). Patients with nCD64 expression ≥2700 MFI had more clinical deterioration (36.4 vs. 7.2 %, p = 0.015) and more ICU admission (45.5 vs. 14.5 %, p = 0.028). To identify clinical deterioration and ICU admission, nCD64 expression showed a sensitivity of 44.4 and 33.3 % and a specificity of 90.1 and 90.8 %, respectively. The addition of nCD64 expression to the Pneumonia Severity Index and CURB-65 severity scores did not improve the accuracy of predicting these outcomes. Although nCD64 expression is associated with an increased risk of ICU admission or clinical deterioration after admission, its accuracy in predicting these poor outcomes is modest and does not significantly improve the predictive ability of the PSI and CURB-65 severity scores. In this work, we performed the rational design of a cationic antimicrobial peptide, GIBIMPY4, using the software DEPRAMPs developed at the GIBIM research group. GIBIMPY4 has a length of 17 amino acids, it is amphipathic, its structure is α-helix and it has a net charge of (+5). Solid-phase peptide synthesis was performed using the Fmoc strategy in acid medium. The primary structure was confirmed by MALDI-TOF mass spectrometry. The antimicrobial activity of the peptide was evaluated by broth microdilution method by measuring optical density in 96-well microplates. The minimal inhibitory concentration of GIBIMPY4 to kill 50 % of the bacterial cells (MIC) was 6.20 ± 0.02 µM for MRSA and 4.55 ± 0.02 µM for E. coli O157:H7, while also reporting a bacteriostatic effect for the later. GIBIMPY4 activity was sensitive to salt concentration in E. coli but insignificant effect in its activity against MRSA. The peptide seems to be a broad-spectrum antimicrobial agent based on the results against Gram-positive and Gram-negative bacteria and was specific for bacterial cells E. coli O157:H7 with index of specificity equal to 9.01 in vitro assays. The Cocinetas Basin of Colombia provides a valuable window into the geological and paleontological history of northern South America during the Neogene. Two major findings provide new insights into the Neogene history of this Cocinetas Basin: (1) a formal re-description of the Jimol and Castilletes formations, including a revised contact; and (2) the description of a new lithostratigraphic unit, the Ware Formation (Late Pliocene). We conducted extensive fieldwork to develop a basin-scale stratigraphy, made exhaustive paleontological collections, and performed Sr/Sr geochronology to document the transition from the fully marine environment of the Jimol Formation (ca. 17.9-16.7 Ma) to the fluvio-deltaic environment of the Castilletes (ca. 16.7-14.2 Ma) and Ware (ca. 3.5-2.8 Ma) formations. We also describe evidence for short-term periodic changes in depositional environments in the Jimol and Castilletes formations. The marine invertebrate fauna of the Jimol and Castilletes formations are among the richest yet recorded from Colombia during the Neogene. The Castilletes and Ware formations have also yielded diverse and biogeographically significant fossil vertebrate assemblages. The revised lithostratigraphy and chronostratigraphy presented here provides the necessary background information to explore the complete evolutionary and biogeographic significance of the excellent fossil record of the Cocinetas Basin. Models for Majorana neutrino masses can be classified according to the level in perturbation theory at which the effective dimension five operator LLHH is realized. The possibilities range from the tree-level up to the three-loop level realizations. We discuss some general aspects of this approach and speculate about a model independent classification of the possible cases. Among all the realizations, those in which the effective operator is induced by radiative corrections open the possibility for lepton number violation near-or at-the electroweak scale. We discuss some phenomenological aspects of two generic realizations: the Babu-Zee model and supersymmetric models with bilinear R-parity violation. [ABSTRACT FROM AUTHOR]

This chapter of the “Flavor in the era of LHC” workshop report discusses flavor-related issues in the production and decays of heavy states at the LHC at high momentum transfer Q, both from the experimental and the theoretical perspective. We review top quark physics, and discuss the flavor aspects of several extensions of the standard model, such as supersymmetry, little Higgs models or models with extra dimensions. This includes discovery aspects, as well as the measurement of several properties of these heavy states. We also present publicly available computational tools related to this topic. The thyroid functions as an apex endocrine organ that controls growth, differentiation and metabolism 1 , and thyroid diseases comprise the most common endocrine disorders 2 . Nevertheless, high-resolution views of the cellular composition and signals that govern the thyroid have been lacking 3,4 . Here, we show that Notch signalling controls homeostasis and thermoregulation in adult mammals through a mitochondria-based mechanism in a subset of thyrocytes. We discover two thyrocyte subtypes in mouse and human thyroids, identified in single-cell analyses by different levels of metabolic activity and Notch signalling. Therapeutic antibody blockade of Notch in adult mice inhibits a thyrocyte-specific transcriptional program and induces thyrocyte defects due to decreased mitochondrial activity and ROS production. Thus, disrupting Notch signalling in adult mice causes hypothyroidism, characterized by reduced levels of circulating thyroid hormone and dysregulation of whole-body thermoregulation. Inducible genetic deletion of Notch1 and 2 in thyrocytes phenocopies this antibody-induced hypothyroidism, establishing a direct role for Notch in adult murine thyrocytes. We confirm that hypothyroidism is enriched in children with Alagille syndrome, a genetic disorder marked by Notch mutations, suggesting that these findings translate to humans., (© 2023.
