1. Investigating the Impact of Intracerebroventricular Streptozotocin on Female Rats with and without Ovaries: Implications for Alzheimer's Disease.

Abstract

To contribute to research on female models of Alzheimer's disease (AD), our aim was to study the effect of intracerebroventricular (ICV) injection of streptozotocin (STZ) in female rats, and to evaluate a potential neuroprotective action of ovarian steroids against STZ. Female rats were either ovariectomized (OVX) or kept with ovaries (Sham) two weeks before ICV injections. Animals were injected with either vehicle (artificial cerebrospinal fluid, aCSF) or STZ (3 mg/kg) and separated into four experimental groups: Sham + aCSF, Sham + STZ, OVX + aCSF and OVX + STZ. Nineteen days post-injection, we assessed different behavioral aspects: burying, anxiety and exploration, object recognition memory, spatial memory, and depressive-like behavior. Immunohistochemistry and Immunoblot analyses were performed in the hippocampus to examine changes in AD-related proteins and neuronal and microglial populations. STZ affected burying and exploratory behavior depending on ovarian status, and impaired recognition but not spatial memory. STZ and ovariectomy increased depressive-like behavior. Interestingly, STZ did not alter the expression of β-amyloid peptide or Tau phosphorylated forms. STZ affected the neuronal population from the Dentate Gyrus, where immature neurons were more vulnerable to STZ in OVX rats. Regarding microglia, STZ increased reactive cells, and the OVX + STZ group showed an increase in the total cell number. In sum, STZ partially affected female rats, compared to what was previously reported for males. Although AD is more frequent in women, reports about the effect of ICV-STZ in female rats are scarce. Abstract

Objectives: Among the complications of parotid surgery, facial palsy is frequent and burdened by high functional and social impact for the patient. There are few data on the efficacy of facial neuromuscular retraining (FNR) in patients with facial palsy after parotid surgery, and no data exist on its impact in timing and extent of recovery. Material and methods: A retrospective study was conducted on patients undergoing FN sparing parotid surgery and suffering from postoperative facial palsy. Among 400 patients undergoing surgery between July 2016 and May 2023, those with the preservation of the FN and onset of facial palsy were selected. Nerve function was evaluated during 2 years follow up using the House-Brackman (H&Bs) and Sunnybrook scales (SBs). Results: A total of 46 patients undergoing partial or total parotidectomy were included. At discharge 18 patients (39,1%) had IV to VI grade paralysis according to the H&Bs and the mean SBs value was 54. At 2 and 6 months after surgery, the average value of Sunnybrook increased to 76.5 and 95.4 respectively. After 12 months no patients with IV to VI grade paralysis were represent in our cohort. Two years after surgery, only five patients (10.9%) had persistent grade II paralysis according to HBs. Conclusions: Our study supports the efficacy of FNR in the rehabilitation of facial paralysis after nerve-sparing parotidectomy. The greater functional improvement is achieved within the first 6 months of rehabilitation. A significant improvement is detected still after 18 months, supporting the importance of long rehabilitation for patients without complete recovery after the first year. [ABSTRACT FROM AUTHOR]

3. The 50th anniversary of the European Society for Muscle Research: a journey through half a century of scientific advances.

Abstract

The European Society for Muscle Research (ESMR) started in 1971 as "European Muscle Club" in a joint initiative of Marcus Schaub, Eduard Jenny and Rudolf Billeter (Zurich), Caspar Rüegg (Heidelberg), Jean Légér (Montpellier), Bernard Swynghedauw (Paris), George Maréchal (Brussels), Gabriel Hamoir (Liège), and Endre Biró (Budapest). Since 1972, local organizers took care of muscle conferences held yearly in different European countries and in Israel in 1987. One of the goals was to establish contacts and collaborations between scientists on both sides of the Iron Curtain. Starting as an informal club, enthusiastically guided by Marcus Schaub as secretary (1971–1995) and later by Ger Stienen (1996–2005), Anders Arner (2006–2017) and Wolfgang Linke (2018-), the ESMR meetings steered international collaborations. The meetings witnessed the remarkable advancement of the insight in skeletal, smooth and cardiac muscle structure and function. In the five decades, the thin and thick filament structure has been resolved to the atomic level, the mechanism of acto-myosin energy transduction and force generation as well as its regulation have been elucidated. The molecular basis of striated and smooth muscle diversity has been found in the existence of multiple protein isoforms. The transcriptional, translational and post-translational regulations which give rise to adaptive responses of muscle tissue have been revealed. Many new players entered the field, such as titin, the ryanodine receptor and several signalling factors. Substantial progress has also been made in the identification of the pathogenesis of many hereditary muscle diseases such as Duchenne MuscularDystrophy and Hypertrophic Cardiac Myopathies. Abstract

This paper investigates whether informative feedback on consumption can nudge water saving. We launched a five-month online information campaign which involved around 1,000 households located in the province of Milan (Italy) with a smart meter. A group of households received monthly reports via email on their per capita daily average water consumption, including a social comparison component. The Intention to Treat (ITT) analysis shows that, compared to a benchmark group, the units exposed to the intervention reduced their per capita water consumption by around 6% (25.8 liters per day or 6.8 gallons). Being able to observe the email opening rate, we find that the ITT effect is mainly driven by complying units. Through an Instrumental Variable approach, we estimated a Local Average Treatment Effect equal to 54.9 liters per day of water saving. A further Regression Discontinuity Design analysis shows that different feedback on consumption class size differentially affected water saving at the margin. We also found that the additional water saving increased with the number of monthly reports, though it did not persist two months after the campaign expired. [ABSTRACT FROM AUTHOR]

Abstract

Skeletal muscle undergoes atrophy and loss of force during long space missions, when astronauts are persistently exposed to altered gravity and increased ionizing radiation. We previously carried out mass spectrometry-based proteomics from skeletal muscle biopsies of two astronauts, taken before and after a mission on the International Space Station. The experiments were part of an effort to find similarities between spaceflight and bed rest, a ground-based model of unloading, focused on proteins located at the costameres. We here extend the data analysis of the astronaut dataset and show compartment-resolved changes in the mitochondrial proteome, remodeling of the extracellular matrix and of the antioxidant response. The astronauts differed in their level of onboard physical exercise, which correlated with their respective preservation of muscle mass and force at landing in previous analyses. We show that the mitochondrial proteome downregulation during spaceflight, particularly the inner membrane and matrix, was dramatic for both astronauts. The expression of autophagy regulators and reactive oxygen species scavengers, however, showed partially opposite expression trends in the two subjects, possibly correlating with their level of onboard exercise. As mitochondria are primarily affected in many different tissues during spaceflight, we hypothesize that reactive oxygen species (ROS) rather than mechanical unloading per se could be the primary cause of skeletal muscle mitochondrial damage in space. Onboard physical exercise might have a strong direct effect on the prevention of muscle atrophy through mechanotransduction and a subsidiary effect on mitochondrial quality control, possibly through upregulation of autophagy and anti-oxidant responses. Abstract

Purpose: A variety of lesions could arise from the GG area, or extend into this region from adjacent sites. The management of perigeniculate lesions includes observation, surgery, and radiation, according to the nature, the size of the lesion, and the accompanying symptoms. Preliminary experiences on the exclusive transcanal endoscopic approach to the GG area have shown safety and feasibility avoiding of any postauricular incision, or brain manipulation. The experience from two referral centers on patients treated for a GG lesion with a totally endoscopic approach is herein reported. Methods: Data about patients who underwent exclusive endoscopic approach to the GG area at the Otolaryngology Departments of the University Hospitals of Modena and Bologna between May 2017 and February 2022 were retrospectively collected. Results: The total number of patients included in our study was 11. 10 patients (91%) had progressive unilateral facial paralysis and 1 patient (11%) presented with chronic otorrhea. The mean largest diameter of the treated lesions was of 8 mm. The resection was extended to the fundus of the IAC in 2 patients (expanded approach). The remaining 9 patients (82%) underwent partial ossicular replacement prosthesis (PORP). No major complications occurred. Facial nerve outcomes were good in all patients and the mean ABG worsened from 12 dB pre-operatively to 22 dB post-operatively. Conclusions: The exclusively endoscopic approach to GG lesions represents a viable alternative to traditional microscopic approaches and may be included in the armamentarium of ear surgeons. Abstract

Non-small-cell lung carcinoma (NSCLC) is the most common lung cancer and one of the pioneer tumors in which immunotherapy has radically changed patients' outcomes. However, several issues are emerging and their implementation is required to optimize immunotherapy-based protocols. In this work, we investigate the ability of the Bromodomain and Extra-Terminal protein inhibitors (BETi) to stimulate a proficient anti-tumor immune response toward NSCLC. By using in vitro, ex-vivo, and in vivo models, we demonstrate that these epigenetic drugs specifically enhance Natural Killer (NK) cell cytotoxicity. BETi down-regulate a large set of NK inhibitory receptors, including several immune checkpoints (ICs), that are direct targets of the transcriptional cooperation between the BET protein BRD4 and the transcription factor SMAD3. Overall, BETi orchestrate an epigenetic reprogramming that leads to increased recognition of tumor cells and the killing ability of NK cells. Our results unveil the opportunity to exploit and repurpose these drugs in combination with immunotherapy. Combination of BET inhibitors (BETi) with immunotherapy has been reported to be synergic for the treatment of non-small cell lung carcinoma (NSCLC). Here, the authors show that BETi-induced epigenetic reprogramming downregulates the expression of NK cell inhibitory receptors on NK cells, increasing their activation and cytotoxicity against NSCLC. Abstract

This study examines the effects of donation incentives on labor supply in an online labor market through a field experiment (n = 944). We manipulate the donation purpose of the incentive to be either unifying or polarizing and the size of the donation relative to the workers' wage. Our experimental design allows us to observe the decision to accept a job (extensive margin) and different dimensions of productivity (intensive margin). We predict and show that a unifying donation purpose attracts more gig workers and improves their productivity compared to a polarizing donation purpose. 10. Assessment of fiber orientation models predictability by comparison with X-ray µCT data in injection-molded short glass fiber-reinforced polyamide.

Abstract

The distribution of fibers in injection-molded thermoplastic-reinforced parts is known to significantly affect both the final mechanical properties and the appearance of defects related to undesired shrinkages and warpages. Even if the numerical modelling of the process is in the state of the art, the selection criteria of the model for the fiber orientation predictability and the influence of the models' parameters are not yet clearly understood. The complexity of the matter increases further considering that, inside the same injection-molded part, the fiber orientation distribution could differ from one region to another depending on the local melt shear flow type. In this context, the aim of this study is to investigate the orientation of fibers in regions of non-simple shear flow in an injection-molded short glass fiber-reinforced part. X-ray microcomputed tomography is used to experimentally investigate fiber distribution and orientation. Furthermore, a number of numerical simulations of the injection molding process are performed in Moldflow® by varying the mesh type (2D/3D), the selected predictive model, and the models' coefficients. The main findings indicate that, in regions with non-simple shear flow, a 2D mesh fails to capture accurate fiber orientation. A 3D mesh is essential for reliable predictions. MRD-specific parameters from the literature reduce prediction error by 37.5% compared to Moldflow's default MRD parameters. Regarding the RSC model, both Wang's optimal retarding rate parameter and AMI's default retarding rate parameter result in similar 5% prediction errors. 12. A MiR181/Sirtuin1 regulatory circuit modulates drug response in biliary cancers.

Abstract

Despite recent advances, biliary tract cancer (BTC) remains one of the most lethal tumor worldwide due to late diagnosis, limited therapeutic strategies and resistance to conventional therapies. In recent years, high-throughput technologies have enabled extensive genome, and transcriptome sequencing unveiling, among others, the regulatory potential of microRNAs (miRNAs). Compelling evidence shown that miRNA are attractive therapeutic targets and promising candidates as biomarkers for various therapy-resistant tumors. The analysis of miRNA profile successfully identified miR-181c and -181d as significantly downregulated in BTC patients. Low miR-181c and -181d expression levels were correlated with worse prognosis and poor treatment efficacy. In fact, progression-free survival analysis indicated poor survival rates in miR-181c and -181d low expressing patients. The expression profile of miR-181c and -181d in BTC cell lines revealed that both miRNAs were dysregulated. Functional in vitro experiments in BTC cell lines showed that overexpression of miR-181c and -181d affected cell viability and increased sensitivity to chemotherapy compared to controls. In addition, by using bioinformatic tools we showed that the miR-181c/d functional role is determined by binding to their target SIRT1 (Sirtuin 1). Moreover, BTC patients expressing high levels of miR-181 and low SIRT1 shown an improved survival and treatment response. An integrative network analysis demonstrated that, miR-181/SIRT1 circuit had a regulatory effect on several important metabolic tumor-related processes. Our study demonstrated that miR-181c and -181d act as tumor suppressor miRNA in BTC, suggesting the potential use as therapeutic strategy in resistant cancers and as predictive biomarker in the precision medicine of BTC. Abstract

The experiences of lesbian, gay, bisexual, and transgender (LGBT +) individuals in Science, Technology, Engineering, and Mathematics (STEM) are still understudied and, despite some improvements, are still characterised by patterns of exclusion, disadvantage, and discrimination. In this article, we explore how visibility is perceived and navigated by LGBT + academics and PhD students in STEM, with a focus on the ways that interlocking systems of oppression impact people and groups who are marginalised and historically excluded. This article draws on a broader research project about the experiences of women and LGBT + people in STEM that was conducted between 2019 and 2020 at a UK university and is framed by intersectionality theory. Based on the thematic analysis of interviews and focus groups with 24 LGBT + participants, findings suggest that visibility is still a risk for LGBT + academics and PhD students in STEM. We found that the labour of navigating visibility was perceived as an unfair disadvantage and that the focus on individuals' visibility in the absence of meaningful and transformative inclusion initiatives by higher education institutions was regarded as tokenistic. The article argues that addressing LGBT + visibility should firstly be an institutional responsibility and not an individual burden and that this work is essential to set the conditions for personal visibility to happen by choice, safely and without retribution. Abstract

Fixed Point-to-Point microwave wireless systems with high spectral efficiency are needed to meet the pervasive and increasing demand for capacity in back-haul networks of mobile radio systems. In this context, spatially multiplexed LoS-MIMO (Line-of-Sight Multiple Input Multiple Output) systems have been studied for about twenty years, particularly in the millimeter wave frequency bands (above 15 GHz). However, their deployment in real networks has been really limited, to the authors' knowledge. This has been due to several factors, i.e. the practical possibility of using extremely high-level modulation formats (nowadays up to 8192-QAM), the joint use of co-channel dual polarization, and the availability of wider channel bands in the new high frequency ranges (e.g. E-Band). In addition, a crucial reason has been the difficulty of installing multiple antennas spaced apart in order to maximize the MIMO spatial multiplexing so providing the maximum capacity gain. This optimal antenna separation, even for the classical MIMO M × N with M = 2 antennas at the receiver and N = 2 antennas at the transmitter, can be several meters, e.g. 5.71 m at 23 GHz on a 5 km link. In this article, we analyze the performance of LoS-MIMO systems where antenna separation is highly sub-optimal, for limiting the array size, and a satisfactory performance is made possible by the exploitation of specific bit loading and power allocation strategies and the setting of the working region of the RF transmitter power amplifiers to operate at a given Signal-to-Inter Modulation Distortion Ratio (SIMDR). The result is an overview of the advantages and drawbacks of compact LoS-MIMO from a wider perspective than in the existing literature, including fundamental aspects for the practical implementation of these systems. Abstract

Irrigation is key to increasing crop yield and meeting the global demand for food. This study reports the assessment of tomato water consumption by bioristor, a new in vivo an Organic ElectroChemical Transistor-based biosensor. Bioristor enables direct, real-time acquisition of biophysical information about the plant's water requirements directly from the plant sap, and thus the water input can be adjusted accordingly. The aim of this study is to demonstrate the efficacy of bioristor in rapidly detecting changes in the plant's water status enhancing water use and irrigation efficiency in tomato cultivation with significant savings in the water supply. To this end, experiments were carried out in 2018 and 2020 in Parma (Italy) in tomato fields under different water regimes. The sensor response index (R) produced by bioristor recorded the real time plant health status, highlighting an excess in the water supplied as well as the occurrence of drought stress during the growing season. In both years, bioristor showed that the amount of water supplied could have been reduced by 36% or more. Bioristor also measured the timing and duration of leaf wetting: 438 h and 409 h in 2018 and 2020, respectively. Abstract

The microstructure of 6xxx aluminum alloys deeply affects mechanical, crash, corrosion and aesthetic properties of extruded profiles. Companies, especially in the transportation sector, require control over the grain structure in order to ensure the quality and the performance of the products. A main challenge for the extrusion companies is to accurately predict the profile microstructure at the design stage, trying to limit the formation of coarse grains and, consequently, the reduction of the mechanical properties of the component. In this work, the modeling of the stored energy, driving force for the recrystallization, was carried out and implemented within the Qform Extrusion FEM code. A novel approach for the evaluation of fibrous and recrystallized microstructures in 6XXX aluminum alloy profiles was proposed and tested in a campaign of experiments involving the extrusion of AA6082 round bars under several die designs and processing conditions. The outcomes proved the good accuracy of the recrystallization predictions during the extrusion of AA6082 aluminum alloy. [ABSTRACT FROM AUTHOR]

Abstract

This research examines the feasibility of using synchronization signals broadcasted by currently deployed fifth generation (5G) cellular networks to determine the position of a static receiver. The main focus lies on the analysis of synchronization among the base stations of a real 5G network in Milan, Italy, as this has a major impact on the accuracy of localization based on time of arrival measurements. Understanding such properties, indeed, is fundamental to characterize the clock drifts and implement compensation strategies as well as to identify the direct communication beam. The paper shows how the clock errors, i.e., inaccurate synchronization, among 5G base stations exhibit a significant bias, which is detrimental for precise cellular positioning. By compensating the synchronization errors of devices' clocks, we demonstrate that it is in principle possible to localize a static user with an accuracy of approximately 8–10 m in non-obstructed visibility conditions, for urban and rural scenarios, using the deployed 5G network operating at 3.68 GHz and relying on broadcast signals as defined by 5G Release 15 standard. 20. Prospective Italian validation of the Vaizey and Wexner and fecal incontinence severity index (FISI) questionnaires.

Abstract

Several objective severity measurement questionnaires of the fecal incontinence (FI), are available to describe type, frequency and degree of FI, and their impact on quality of life, aiming to establish baseline scores measure response to treatment over time and allow comparison among patients treated using different strategies. Presently, despite their widespread use in clinical practice, none of these questionnaire have been validated in the Italian language. The aim is to test the translated Italian version of the Vaizey and Wexner and Fecal Incontinence Severity Index (FISI) questionnaires assessing their reliability and validity among Italian-speaking patients. Two researchers proficient in spoken English and Italian translated both questionnaires in the Italian language. They independently translated the two questionnaires from English and then they met to produce a single version of the two questionnaires, to solve any possible discrepancy. A forward–backward translation was then obtained by a professional bilingual translator, so as to define the final version of the questionnaires. The questionnaires were independently administered twice to 100 Italian-speaking patients by two different and independent raters. Cronbach's α of the first and second Vaizey and Wexner questionnaire was 0.755 and 0.727, respectively. While Cronbach's α of the first and second FISI questionnaire was 0.810 and 0.806, respectively. Spearman correlation and inter-rater reliability were 0.937 and 0.913 for Vaizey and Wexner questionnaire, respectively, and 0.915 and 0.871 for FISI questionnaire, respectively. Italian version of the Vaizey and Wexner and FISI questionnaires proved good consistency, reliability, reproducibility, showing good psychometric properties. [ABSTRACT FROM AUTHOR]

21. Impact of plant extracts on hepatic redox metabolism upon lead exposure: a systematic review of preclinical in vivo evidence.

Abstract

The liver is a central target organ of heavy metals toxicity, and secondary metabolites of several plant species are suggested to attenuate lead (Pb)-induced hepatotoxicity through antioxidant and anti-inflammatory mechanisms. We used a systematic review framework to map the impact of plant extracts and bioactive secondary metabolites on immunological markers and liver redox metabolism in preclinical models of Pb exposure. This is a systematic review performed according to PRISMA guidelines. The structured research of publications was done through PubMed, Scopus, Web of Science, and Embase databases, selecting and analyzing 41 original studies included via the eligibility criteria. Evidence indicates that Pb-exposure increases reactive oxygen/nitrogen species (ROS/RNS) production by δ-aminolevulinic acid auto-oxidation, xanthine dehydrogenase, and xanthine oxidase upregulation. Pb exposure also inhibits antioxidant enzymes, potentiating ROS/NOS levels and reactive cell damage. Plant extracts rich in flavonoids, tannins, alkaloids, anthocyanins, and vitamins exerted hepatoprotective effects by chelating and decreasing Pb bioaccumulation. In addition, plant extracts reinforce exogenous and endogenous antioxidant defenses, attenuating liver oxidative stress and cell death. The lack of blinded evaluators and randomized experimental groups were the main sources of bias identified, which need to be controlled in toxicological studies aimed at identifying natural products applied to the prevention or treatment of Pb poisoning. Abstract

Purpose: During alternate movements across a joint, the changeover from one direction of rotation to the opposite may be influenced by the delay and rate of tension reduction and the compliance to re-lengthening of the previously active muscle group. Given the aging process may affect the above-mentioned factors, this work aimed to compare the dynamics of both the ankle torque decline and muscle re-lengthening, mirrored by mechanomyogram (MMG), in the tibialis anterior because of its important role in gait. Methods: During the relaxation phase, after a supramaximal 35 Hz stimulation applied at the superficial motor point, in 20 young (Y) and 20 old (O) subjects, the torque (T) and MMG dynamics characteristics were measured. Results: The T and MMG analysis provided: (I) the beginning of the decay after cessation of stimulation (T: 22.51 ± 5.92 ms [Y] and 51.35 ± 15.21 ms [O]; MMG: 27.38 ± 6.93 ms [Y] and 61.41 ± 18.42 ms [O]); (II) the maximum rate of reduction (T: − 110.4 ± 45.56 Nm/s [Y] and − 52.72 ± 32.12 Nm/s [O]; MMG: − 24.47 ± 10.95 mm/s [Y] and − 13.76 ± 6.54 mm/s [O]); (III) the muscle compliance, measuring the MMG reduction of every 10% reduction of torque (bin 20–10%: 15.69 ± 7.5[Y] and 10.8 ± 3.3 [O]; bin 10–0%: 22.12 ± 10.3 [Y] and 17.58 ± 5.6 [O]). Conclusion: Muscle relaxation results are different in Y and O and can be monitored by a non-invasive method measuring physiological variables of torque and re-lengthening dynamics at the end of the electromechanical coupling previously induced by the neuromuscular stimulation. Abstract

Small bowel adenocarcinomas (SBAs) are often associated with poor prognosis and have limited therapeutic options. Programmed cell death protein-1 (PD-1)/programmed cell death ligand 1 (PD-L1) pathway blockade is an effective treatment in many microsatellite instability-high (MSI-H) solid tumors. We aimed at investigating PD-L1 and PD-1 expression in non-hereditary, non-ampullary SBAs, associated with celiac disease (CeD), Crohn’s disease (CrD), or sporadic, recruited through the Small Bowel Cancer Italian Consortium. We assessed PD-L1 and PD-1 by immunohistochemistry in a series of 121 surgically resected SBAs, including 34 CeD-SBAs, 49 CrD-SBAs, and 38 sporadic SBAs. PD-L1 and PD-1 expression was correlated with several clinico-pathological features, such as the etiology, microsatellite instability status, and tumor-infiltrating lymphocyte (TIL) density. The prevalence of PD-L1 positivity according to combined positive score (CPS) was 26% in the whole cohort of SBAs, with significantly (p = 0.001) higher percentage (35%) in both CeD-SBAs and CrD-SBAs in comparison with sporadic SBAs (5%). CPS ≥ 1 SBAs were significantly (p = 0.013) more frequent in MSI-H cases (41%) than in non-MSI-H ones (18%); however, 15 CPS ≥ 1 microsatellite stable SBAs were also identified. CPS ≥ 1 SBAs showed higher TIL and PD-1+ immune cell density, more frequently medullary histotype, as well as a better outcome in comparison with CPS < 1 cases. This study demonstrates an increased proportion of PD-L1+ cases in both CeD-SBAs and CrD-SBAs in comparison with sporadic SBAs. In addition, the identification of a subset of PD-L1+ microsatellite stable SBAs supports the need to ascertain additional biomarkers of response to immune checkpoint inhibitors along with MSI-H.

27. Media negativity bias and tax compliance: experimental evidence.

Abstract

We study the impact of the media negativity bias on tax compliance. Through a framed laboratory experiment, we assess how the exposure to biased news about government action affects compliance in a repeated taxation game. Subjects treated with positive news are significantly more compliant than the control group. Instead, the exposure to negative news does not prompt any significant reaction compared to the neutral condition, suggesting that participants may perceive the media negativity bias in the selection and tonality of news as the norm rather than the exception. Overall, our results suggest that biased news provision is a constant source of psychological priming and plays a vital role in taxpayers' compliance decisions. [ABSTRACT FROM AUTHOR]

Abstract

This paper highlights the relevance of connectivity and its architecture as a general conceptual framework which underlies and integrates the concepts of network vulnerability, complexity, and resilience. In particular, it will be pointed out that connectivity architecture can be considered an explicit key element for network vulnerability and shock propagation. While the relevance of the various connectivity configurations is not clearly emphasised in the dynamic complexity models of the space-economy, it appears to play a primary role in network analysis. In this regard, the emerging recognition of connectivity architecture in relation to hubs ‒ and hierarchies of hubs ‒ in a complex network will help the enhancement of network resilience. The paper develops as follows. First, the notion of network vulnerability, which refers not only to the phenomenon of shocks, but also to the propagation of shocks in a network, will be examined. Here it appears that modelling vulnerability and shock propagation, also jointly with cascading disaster models, is strongly based on connectivity issues. The question is: How can conventional (complex) system dynamic modelling, as well as network modelling, take into account these shocks and connectivity dynamics from the methodological viewpoint? A review in this respect shows how connectivity is a 'hidden' element in these complexity models, for example, in chaos or (dynamic) competition models, where interaction parameter values might lead to vulnerable domains and chaotic behaviour. On the contrary, connectivity and its various topologies have a distinct, primary role in network analysis. The issue of network resilience appears therefore to be the 'response' to vulnerability and chaos, calling for robustness and stability of the network in the presence of shocks and disruptions. Resilience analysis refers to the speed at which a network returns to its equilibrium after a shock, as well as to the perturbations/shocks that can be absorbed before the network is induced into some other equilibrium (adaptivity). Connectivity is relevant here, but not often considered in spatial economics. In order to reach a unified methodological framework, attention will finally be paid to a complementary analysis of the (dynamic) concepts of vulnerability and resilience. In this light, chaos models/properties might be seen in a positive perspective, since small changes can lead to uncertain and unstable effects, but also, thanks to connectivity, to new equilibria which are not necessarily negative. Thus, the architecture of connectivity, in its interdisciplinary insights, can be considered as a fundamental (and analytical) approach for identifying vulnerability and resilience patterns in complex networks. Targeting fatty acid amide hydrolase (FAAH) is a promising therapeutic strategy to combat certain forms of pain, including migraine headache. FAAH inhibitors, such as the O-biphenyl-3-yl carbamate URB597, have been shown to produce anti-hyperalgesic effects in animal models of migraine. The objective of this study was to investigate the behavioral and biochemical effects of compounds ARN14633 and ARN14280, two URB597 analogs with improved solubility and bioavailability, in a migraine-specific rat model in which trigeminal hyperalgesia is induced by nitroglycerin (NTG) administration. ARN14633 (1 mg/kg, i.p.) and ARN14280 (3 mg/kg, i.p.) were administered to adult male Sprague-Dawley rats 3 hours after NTG injection. One hour after the administration of either compound, rats were subjected to the orofacial formalin test. ARN14633 and ARN14280 attenuated NTG-induced nocifensive behavior and reduced transcription of genes encoding neuronal nitric oxide synthase, pain mediators peptides (calcitonin gene-related peptide, substance P) and pro-inflammatory cytokines (tumor necrosis factor-alpha, interleukin-1beta and 6) in the trigeminal ganglion, cervical spinal cord and medulla. Finally, both compounds strongly elevated levels of endocannabinoids and/or other FAAH substrates in cervical spinal cord and medulla, and, to a lesser extent, in the trigeminal ganglia. The results indicate that the novel global FAAH inhibitors ARN14633 and ARN14280 elicit significant anti-hyperalgesic effects in a migraine-specific animal model and inhibit the associated peptidergic-inflammatory response. Although the precise mechanism underlying these effects remains to be elucidated, our results support further investigational studies of FAAH blockade as a potential therapeutic strategy to treat migraine conditions. A fully separated bicycle network from vehicular traffic is not realistic even for the most bicycle-friendly cities. Thus, all around the world urban cycling entails switching between streets of different safety, convenience, and comfort levels. As a consequence, the quality of bicycle networks should be evaluated not based on one but multiple factors and by considering the different user preferences regarding these factors. More comprehensive methodologies to assess urban bicycle networks are essential to the operation and planning of modern city transportation. This work proposes a multi-objective methodology to assess—what we refer to as—bikeability between origin–destination locations and over the entire network, useful for evaluation and planning of bicycle networks. We do so by introducing the concept of bikeability curves which allows us to assess the quality of cycling in a city network with respect to the heterogeneity of user preferences. The application of the proposed methodology is demonstrated on two cities with different bike cultures: Amsterdam and Melbourne. 34. Preliminary experience on the use of sucrosomial iron in hemodialysis: focus on safety, hemoglobin maintenance and oxidative stress.

Abstract

Purpose: Iron is usually administered in hemodialysis patients by parenteral route, as oral absorption is poor due to high hepcidin levels. However, administrations of intravenous iron and iron overload are associated with high oxidative stress and systemic inflammation that can affect patient survival. With this study, we evaluated an alternative type of oral iron for the treatment of anemia in hemodialysis patients. The formulation consists in ferric pyrophosphate covered by phospholipids plus sucrose ester of fatty acid matrix, named sucrosomial iron, whose absorption is not influenced by hepcidin. Methods: Twenty-four (24) patients undergoing chronic hemodialysis switched iron supplementation from intravenous (ferric gluconate 62.5 mg weekly) to oral (sucrosomial iron, 90 mg weekly in 3 administrations of 30 mg) route for 3 months. Classical anemia, iron metabolism, inflammation and nutritional biomarkers were monitored, as well as biomarkers of oxidative stress, such as protein-bound di-tyrosines, protein carbonylation, advanced oxidation protein products and protein thiols. Results: Over the 3 months, hemoglobin values remained stable, as the values of hematocrit and mean corpuscular volume. In parallel, other anemia parameters dropped, including ferritin, transferrin saturation and serum iron. On the other side, nutritional biomarkers, such as total proteins and transferrin, increased significantly during the time frame. We also observed a significant decrease in white blood cells as well as a non-significant reduction in C-reactive protein and some oxidative stress biomarkers, such as protein carbonyls and di-tyrosines. Conclusion: Our study demonstrates that a therapy with sucrosomial iron in hemodialysis patients is safe and can maintain stable hemoglobin levels in a three-month period with a possible beneficial effect on oxidative stress parameters. However, the reduction of ferritin and transferrin saturation suggests that a weekly dosage of 90 mg is not sufficient in hemodialysis patients in the long time to maintain hemoglobin. Abstract

Nowadays, the liquid nitrogen cooling in aluminium extrusion is a widely adopted industrial practice to increase the process productivity as well as to improve the extruded profile surface quality by reducing the profile exit temperatures. The cooling channels are commonly designed on the basis of die maker experience only, usually obtaining modest performances in terms of cooling efficiency. Trial-and-error approach is time and cost consuming, thus providing a relevant industrial interest in the development of reliable numerical simulations able to foresee and optimize the nitrogen cooling effect during the die design stage. In this work, an extensive experimental campaign was performed during the extrusion process of an AA6060 industrial hollow profile, in different conditions of nitrogen flow rate and ram speed. The monitored data (die and profile temperatures and extrusion load) were compared with the outputs of a fast and efficient numerical model proposed by the authors, and developed in the COMSOL Multiphysics code, able to compute not only the effect of nitrogen liquid flow but also the gaseous condition. Abstract

The die cooling by means of liquid nitrogen is a widely adopted industrial practice used to increase the production rate in the hot extrusion process of light alloys. The development of a reliable numerical model able to simulate the cooling channel efficiency has become of primary interest for the extrusion sector in order to avoid ineffective die cooling and time-consuming trials and errors. In this work, H13 die inserts with a helicoidally conformal channel were designed and printed by means of the SLM additive technology. Billets of AA6063 aluminum and ZM21 magnesium alloys were extruded at different process speeds under monitored conditions to verify the insert resistance and the cooling effectiveness. A 3D finite element model of the extrusion process coupled with a 1D model of the cooling channel was generated within the COMSOL simulation environment. The experimental outputs were also used to validate the numerical predictions of the developed simulations. The FEM results showed a good matching with the loads and temperatures obtained in the experimental trials. Moreover, the endurance of the AM tool validated the prediction of the stress field, thus proving the reliability of the numerical model for the application in the extrusion of light alloys sector. Abstract

Purpose: To evaluate the post- coronavirus disease-19 (COVID-19) outcome of thyroid function in patients with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2)-related thyrotoxicosis. Methods: This was a single-center prospective study involving 29 patients (11 females, 18 males; median age 64 years, range: 43–85) with thyrotoxicosis diagnosed after hospitalization for COVID-19 and then followed-up for a median period of 90 days (range: 30–120) after hospital discharge. At follow-up, patients were evaluated for serum thyrotropin (TSH), free-thyroxine (FT4), free-triiodiothyronine (FT3), TSH receptor antibodies (TRAb), thyroglobulin antibodies (TgAb), thyroperoxidase antibodies (TPOAb) and ultrasonographic thyroid structure. Results: After recovery of COVID-19, serum TSH values significantly increased (P < 0.001) and FT4 values significantly decreased (P = 0.001), without significant change in serum FT3 (P = 0.572). At follow-up, 28 subjects (96.6%) became euthyroid whereas overt hypothyroidism developed in one case. At the ultrasound evaluation of thyroid gland, hypoecogenicity was found in 10 patients (34.5%) and in these cases serum TSH values tended to be higher than those without thyroid hypoecogenity (P = 0.066). All subjects resulted to be negative for TgAb, TPOAb and TRAb. Conclusion: In a short-term follow-up, thyroid function spontaneously normalized in most subjects with SARS-CoV-2-related thyrotoxicosis. However, thyroid hypoecogenicity was found in a remarkable number of them and future longer-term studies are needed to clarify whether this ultrasonographic alteration may predispose to develop late-onset thyroid dysfunction. Abstract

Caffeine is worldwide used for its power to increase cognitive and physical performance. The ergogenic effects of caffeine, however, do not depend on a direct action on muscles. Actually, the actions of caffeine on skeletal muscles, take place at millimolar concentrations which are far above the micromolar level reached after a regular consumption of coffee or similar drinks, and close to a lethal concentration. At millimolar concentrations caffeine exerts a powerful effect on sarcoplasmic reticulum (SR) activating the release of calcium via ryanodine receptors and, possibly, inhibiting calcium reuptake. For this reason caffeine has become a valuable tool for studying SR function and for diagnostics of SR related muscle disorders. This review aims to briefly describe the effects and the mechanism of action of caffeine on sarcoplasmic reticulum and to focus on its use to study intracellular calcium dynamics in human muscle fibers in physiological and pathological conditions. Abstract

Charge welds are unavoidable defects of the continuous extrusion process whose extension needs to be accurately predicted in order to avoid profile mechanical failures or to minimize unrequired scrap of material. The aim of this work was then to evaluate the accuracy of the charge welds FEM predictions and their applicability in the industrial field. Two different industrial cases involving A6082 and AA6063 aluminium alloys were analysed. The data of charge welds behaviours were experimentally collected, discussed and then innovatively compared to the predicted outcomes of FEM simulations performed using QForm Extrusion® software and of theoretical formulas reported in the literature. As main results, a very good numerical-experimental matching was found, with a peak discrepancy of 122 mm in terms of charge weld extent, while theoretical formulas returned a significant underestimation. Abstract

Background: Prone positioning (PP) has been used to improve oxygenation in patients affected by the SARS-CoV-2 disease (COVID-19). Several mechanisms, including lung recruitment and better lung ventilation/perfusion matching, make a relevant rational for using PP. However, not all patients maintain the oxygenation improvement after returning to supine position. Nevertheless, no evidence exists that a sustained oxygenation response after PP is associated to outcome in mechanically ventilated COVID-19 patients. We analyzed data from 191 patients affected by COVID-19-related acute respiratory distress syndrome undergoing PP for clinical reasons. Clinical history, severity scores and respiratory mechanics were analyzed. Patients were classified as responders (≥ median PaO2/FiO2 variation) or non-responders (< median PaO2/FiO2 variation) based on the PaO2/FiO2 percentage change between pre-proning and 1 to 3 h after re-supination in the first prone positioning session. Differences among the groups in physiological variables, complication rates and outcome were evaluated. A competing risk regression analysis was conducted to evaluate if PaO2/FiO2 response after the first pronation cycle was associated to liberation from mechanical ventilation. Results: The median PaO2/FiO2 variation after the first PP cycle was 49 [19–100%] and no differences were found in demographics, comorbidities, ventilatory treatment and PaO2/FiO2 before PP between responders (96/191) and non-responders (95/191). Despite no differences in ICU length of stay, non-responders had a higher rate of tracheostomy (70.5% vs 47.9, P = 0.008) and mortality (53.7% vs 33.3%, P = 0.006), as compared to responders. Moreover, oxygenation response after the first PP was independently associated to liberation from mechanical ventilation at 28 days and was increasingly higher being higher the oxygenation response to PP. Conclusions: Sustained oxygenation improvement after first PP session is independently associated to improved survival and reduced duration of mechanical ventilation in critically ill COVID-19 patients. Abstract

In this paper a Point Inversion algorithm for NURBS volumes is presented. This algorithm is an extension to 3D of the classic Newton–Raphson iteration, thus implying the calculation of the NURBS volume partial derivatives. Explicit formulas for the derivatives are deduced and reported, in turn requiring the evaluation of the corresponding derivatives of the rational basis functions. Also these derivatives are inferred and shown together with some examples of applications. The method, applicable to any free-form shape NURBS volume is moreover compared with Fast Mapping, a novel approach presented here that, once applied to simple NURBS boxes give performance 20–50 times higher. Abstract

Background: Crohn's disease (CD) and ulcerative colitis, two forms of inflammatory bowel disease (IBD), are chronic and relapsing conditions of the gastrointestinal tract both characterized by long lasting chronic inflammation and increased risk of dysplasia and colorectal cancer (CRC). The aim of our study was to evaluate the interobserver agreement about IBD-associated dysplasia among pathologists belonging to the Italian Group for Inflammatory Bowel Diseases (IG-IBD P). Methods: The present multicenter survey was performed using telepathology, supported by an open source E-learning platform. Biopsy specimens from 30 colonoscopies and from 20 patients were included. The glass slides of any case, including clinical and endoscopic data, were digitalized and uploaded on the E-learning platform. All the digital slides were grouped in 54 diagnostic "blocks". Blinded histopathological evaluation on all the digital slides was performed by 20 gastrointestinal pathologists. Closed-ended questions about (1) the occurrence of IBD; (2) the classification of IBD (as UC or CD); (3) the presence of active versus quiescent disease; (4) the presence of dysplasia; (5) the possible association of dysplasia with the sites of disease (dysplasia-associated lesion or mass—DALM vs adenoma-like mass—ALM); (6) the grading of dysplasia according to the ECCO guidelines (negative, indefinite, low grade, high grade categories) and (7) the presence of associated serrated features, were proposed in each case. Inter-observer agreement was evaluated by mean agreement percentage and kappa statistic, when suitable. Results: The diagnosis of IBD was confirmed in 19 of 20 patients, 17 of 19 being classified as UC, 2 as CD. The mean interobserver agreement percentages about (1) the evidence of IBD, (2) the presence of either UC or CD and (3) the activity grading resulted to be 80%, 69% and 86%, respectively. Dysplasia was detected in 8/20 patients, with moderate agreement between pathologists (mean 72%, k 0.48). Particularly, low grade dysplasia was found in 13 biopsies (combined k 0.38), whereas high grade dysplasia in 8 (combined k 0.47). When the endoscopic and histopathological data were combined, features consistent with DALM were found in 6 of 20 patients with low grade dysplasia and those consistent with ALM in 2 patients with low grade dysplasia in a single biopsy (mean agreement: 86%). An associated serrated pattern was discovered in 4 patients (7 biopsies). Conclusions: Our study showed moderate interobserver agreement about the histopathological detection and classification of IBD-associated dysplasia. Abstract

When multiple charities, social programs and community projects simultaneously vie for funding, donors risk mis-coordinating their contributions leading to an inefficient distribution of funding across projects. Community chests and other intermediary organizations facilitate coordination among donors and reduce such risks. To study this, we extend a threshold public goods framework to allow donors to contribute through an intermediary rather than directly to the public goods. Through a series of experiments, we show that the presence of an intermediary increases public good success and subjects' earnings only when the intermediary is formally committed to direct donations to socially beneficial goods. Without such a restriction, the presence of an intermediary has a negative impact, complicating the donation environment, decreasing contributions and public good success. [ABSTRACT FROM AUTHOR]
