1. On the basis of new theoretical considerations developed by us a new and reliable method has been elaborated for the rapid determination of water deficit in plant cultures. 2. In paper-chromatograms of plants optimally supplied with water free proline is shown to be present in very small amounts, usually only as traces. Comparative experiments showed that in cases of water shortage the proline content of the leaves of the plant strikingly increases. This commonly occurs in every phase of development of mono- and dicotyledons. 3. Drought-resistant plants synthetize more proline than varieties less resistant to water deficit when equally water deficient. 4. By a method which does not require chromatography the procedure of tests has been accelerated so that leaf extract containing amino acid mixture was dropped onto a chromatogram paper and immediately treated with isatine. In this test the effect of slight, permanent or medium water deficit upon proline content is so great that extracts react with isatine to give an intense dark blue colour which suppresses the reddish-brown colour of other amino acids. 5. To avoid the necessity of preparing isatine reagent frequently, because it loses its amino acid sensitivity very rapidly, a new isatine solution has been formulated. Our isatine-paper indicator prepared with this solution keeps its reactivity with amino acids 5 to 6 months. 6. An unusually high proline concentration occurs also if the salt content of irrigation water or also of the soil is high. The increase of the proline level indicates therefore physiological dryness. During excavations carried out at the necropolis of San Genesio, taphonomic analysis permitted the detection of a small nucleus of distinctive graves dated to the sixth century and set within the wider context of autochthonous burials. This group was characterized by features such as wooden coffins and tree trunk burials, elements typical of the Germanic cultural milieu. Furthermore, anthropological analysis of the skeletal remains identified a case of Artificial Cranial Deformation (ACD), a distinctive element of Gothic ethnic groups. Individuals with ACD are extremely rare in the Italian archeological record and are related to the period in which the Gothic migrations took place. The subject, buried in a wooden casket, was a male individual with a deformed skull, polytraumatized, and most likely killed in combat, possibly during the Gothic War. This is the first Italian case of an individual with ACD, who died a certified violent death. The present study, despite the absence of traditional Germanic grave goods, has allowed to hypothesize the existence at San Genesio of an allochthonous group, a theory further supported by strontium isotope analysis. Introduction: Ovarian cancer has a high mortality rate due to difficulties in early detection and chemotherapy resistance. Human epididymal protein 4 (HE4) has been adopted as a novel serum biomarker for early ovarian cancer diagnosis, and the presence of Lewis y antigen modifications on HE4 in ovarian cancer cell lines has been detected in previous studies. The aim of this study was to analyze the expression of HE4 and Lewis y antigen in human ovarian cancer in order to find a correlation between them, as well as with the clinical pathological parameters of patients with ovarian cancer. Methods: Immunohistochemistry was used to detect the respective expression of these compounds in two patient groups (chemotherapy-resistant and chemotherapy-sensitive) containing a total of 95 patients. Then, a bioinformatic approach was adopted and online large sample databases (TCGA, CCLE, and GTEx; Metascape, Cytoscape) were used to explore the potential mechanisms of action of these compounds. Results: The results of this study demonstrate that high HE4 and Lewis y expression could be used as markers for chemotherapy resistance and poor prognosis in patients with ovarian cancer. These two expression events were widely correlated in various cancer tissues and are thought to act by activating the p38 mitogen-activated protein kinases (MAPK) pathway and inducing Vascular Endothelial Growth Factor A (VEGFA), Prostaglandin-Endoperoxide Synthase 2 (PTGS2), Early Growth Response 1 (EGR1), and Hypoxia-Inducible Factor 1-Alpha (HIFI1A), thereby promoting malignant biological behavior and resistance in ovarian cancer. Conclusions: These findings not only reveal the possible mechanism by which HE4 and Lewis y antigen affect ovarian cancer but also identify a four-gene signature that could be very useful in ovarian cancer detection and/or the development of new targeted therapies. The La Chapelle-aux-Saints 1 skeleton of an old (>60-year-old) male Neanderthal is renowned for the advanced osteoarthritis of its spinal column and hip joint, and their implications for posture and lifestyle in these Mid- to Late Pleistocene humans. Reassessment of the pathologic lesions reveals erosions at multiple non-contiguous vertebrae and reactive bone formation extending far beyond the left hip joint, which suggests the additional diagnosis of brucellosis. This implies the earliest secure evidence of this zoonotic disease in hominin evolution. Brucellosis might have been transmitted via butchering or eating raw meat and is well compatible with the range of prey animals documented for Neanderthals. Purpose: Aging is known to play a critical role in the etiopathogenesis of several diseases. Among them, cardiovascular disorders are especially relevant since they are becoming the first cause of death in western countries. Resveratrol is a polyphenolic compound that has been shown to exert beneficial effects at different levels, including neuronal and cardiovascular protection. Those effects of resveratrol are related, at least in part, to its antioxidant and anti-inflammatory properties. In the current investigation we were interested in exploring whether the positive effects of resveratrol at cardiac level were taking place even when the supplementation started in already old animals. Methods: Old male rats were supplemented with resveratrol during 10 weeks. Using RT-PCR, we analyzed the effects of resveratrol supplementation on the expression of different genes related to inflammation, oxidative stress and apoptosis in rat heart. Results: Resveratrol reverted age-related changes in inflammatory, oxidative and apoptotic markers in the rat heart. Among others, the expression of two major inflammatory markers, INF-γ and TNF-α and two oxidative markers, heme oxygenase-1 and nitric oxide synthase, were increased with aging, and resveratrol supplementation reduced the level of some of these to those observed in the heart of young animals. Moreover, age-related changes in apoptotic markers in rat heart tend to be also reverted by resveratrol treatment. Summary: Background: In tertiary syphilis, Treponema pallidum triggers the formation of granulomatous nodules in various organs of the human body. Within the skeleton, predominantly in the skull and long bones, these characteristic syphilitic lesions cause typical patterns of bone damage. In this study, micro-computed tomography (µ-CT) was used to assess the microarchitecture of these osseous defects in untreated syphilitic skull bones. Material and methods: Bone structure of 30 macerated human skulls was noninvasively examined by means of µ-CT images (Viscom X8060 NDT). A total of 20 specimens showing typical morphological signs of syphilis were provided by the Collection of Anatomical Pathology of the Museum of Natural History in Vienna. They were compared to 10 macerated control skulls provided by the Division of Anatomy of the Medical University of Vienna. Results: All samples affected by syphilis showed perforating defects and increased porosity. Furthermore, we observed sclerotic reorganization and complete loss of the cortical bone in 80% of infected cases. Cortical thinning occurred in 75%. Conclusion: Our findings revealed extensive micromorphological bone destruction and a broad variability of osseous manifestations of (tertiary) syphilis. [ABSTRACT FROM AUTHOR]

This research focuses on the Neolithic populations of Western Switzerland (5500–2200 BCE) and on establishing their paleopathological profile. This chronological period presents varying burial practices, and the aim was to be able to synthesise all information regarding the pathologies these populations exhibited, in such a way that statistical tests could be performed to determine whether geographical (site location), biological (age, sex, stature) and chronological factors played a role in lesion distribution. As a result, a methodology was devised to consider bone lesions in both a quantified and qualified manner, and able to handle individualised, multiple and commingled remains. The statistical analysis showed that pathological profiles differed depending on site location, with some exhibiting patterns that raise further questions as to the socio-economical functioning of these prehistoric societies. Biological factors also appear to play a role in lesion distribution, with immature individuals exhibiting more metabolic deregulations, whilst the adults—as expected—showed more signs of degenerative pathologies. Some chronological evolution of health is also perceivable; however, the small sample size available for some periods means these results are tentative at best. These remains, however, proved more than an accumulation of bone lesions, and the qualified data collected from them also yielded individual and collective narratives of health. Instances of infectious diseases were discovered, indicating that pathologies such as tuberculosis were a persistent presence at the time, with all the consequences this entails. The ancient geographical origins of Brahmins—a prominent ethnic group in the Indian subcontinent—have remained controversial for a long time. This study employed the AMOVA (analysis of molecular variance) test to evaluate genetic affinities of this group with thirty populations of Central Asia and Europe. A domestic comparison was performed with fifty non-Brahmin groups in India. The results showed that Brahmins had genetic affinities with several foreign populations and also shared their genetic heritage with several domestic non-Brahmin groups. The study identified the deep ancient origins of Brahmins by tracing their Y-chromosome haplogroups and genetic markers on the Y-DNA phylogenetic tree. It was confirmed that the progenitors of this group emerged from at least 12 different geographic regions of the world. The study concluded that about 83% of the Brahmins in the dataset belonged to four major haplogroups, of which two emerged from Central Asia, one from the Fertile Crescent, and one was of an indigenous Indian origin. Hypnosis and hypnotic suggestions are gradually gaining popularity within the consciousness community as established tools for the experimental manipulation of illusions of involuntariness, hallucinations and delusions. However, hypnosis is still far from being a widespread instrument; a crucial hindrance to taking it up is the amount of time needed to invest in identifying people high and low in responsiveness to suggestion. In this study, we introduced an online assessment of hypnotic response and estimated the extent to which the scores and psychometric properties of an online screening differ from an offline one. We propose that the online screening of hypnotic response is viable as it reduces the level of responsiveness only by a slight extent. The application of online screening may prompt researchers to run large-scale studies with more heterogeneous samples, which would help researchers to overcome some of the issues underlying the current replication crisis in psychology. In this study, we aimed to develop an approach for genetic material extraction and sample preparation of ancient DNA for next generation sequencing that minimizes intra-laboratory contamination. A special module system consisting of four sterile gloveboxes connected by antechambers, which maintain a high purity atmosphere and allow conducting all stages of sample preparation in an isolated clean environment, starting with the processing of archaeological material, was developed. Furthermore, a fast and cost-effective double-stranded library preparation protocol for massive parallel sequencing was developed using non-standard modified sequencing adaptors, which enabled obtaining a library of fragments from extremely low amounts of starting degraded material. Bioinformatics data processing showed that the distribution of substitution frequencies in a subset of reads, mapped to the human reference genome, completely coincides with the pattern of postmortem modifications expected when the double-stranded library preparation method is employed. This is a strong evidence for the endogenous origin of the sequenced fragments of the human genome. Lions (Panthera leo) are of particular conservation concern due to evidence of recent, widespread population declines in what has hitherto been seen as a common species, robust to anthropogenic disturbance. Here we use non-invasive methods to recover complete mitochondrial genomes from single hair samples collected in the field in order to explore the identity of the Gabonese Plateaux Batéké lion. Comparison of the mitogenomes against a comprehensive dataset of African lion sequences that includes relevant geographically proximate lion populations from both contemporary and ancient sources, enabled us to identify the Plateaux Batéké lion as a close maternal relative to now extirpated populations found in Gabon and nearby Congo during the twentieth century, and to extant populations of Southern Africa. Our study demonstrates the relevance of ancient DNA methods to field conservation work, and the ability of trace field samples to provide copious genetic information about free-ranging animals. Males and females display biological differences that lead to a higher variance of offspring number in males, and this is frequently exacerbated in human societies by mating practices, and possibly by past socio-cultural circumstances. This implies that the genetic record might contain the imprint of past male-mediated expansions, which can be investigated by analysing the male-specific region of the Y chromosome (MSY). Here, we review studies that have used MSY data to infer such expansions. Sets of short-tandem repeats define haplotypes of very low average frequencies, but in a few cases, high-frequency haplotypes are observed, forming the core of descent clusters. Estimates of the ages of such clusters, together with geographical information, have been used to propose powerful historical founders, including Genghis Khan, although without direct supporting evidence. Resequencing of multi-megabase segments of MSY has allowed the construction of detailed phylogenies in which branch lengths are proportional to time, leading to the identification of lineage expansions in the last few millennia as well as the more distant past. Comparisons with maternally-inherited mitochondrial DNA sequence data allow the male specificity of some of these expansions to be demonstrated. These include expansions in Europe in the last ~5000 years that may be associated with a cultural shift during the Bronze Age, as well as expansions elsewhere in the world for which explanations from archaeological evidence are not yet clear. We discuss the relations of processed foods, especially cooked foods, in the human diet to digestive tract form and function. The modern consumption of over 70 % of foods and beverages in highly refined form favours the diet-related classification of humans as cucinivores, rather than omnivores. Archaeological evidence indicates that humans have consumed cooked food for at least 300-400,000 years, and divergence in genes associated with human subpopulations that utilise different foods has been shown to occur over periods of 10-30,000 years. One such divergence is the greater presence of adult lactase persistence in communities that have consumed dairy products, over periods of about 8,000 years, compared to communities not consuming dairy products. We postulate that 300-400,000 years, or 10,000-14,000 generations, is sufficient time for food processing to have influenced the form and function of the human digestive tract. It is difficult to determine how long humans have prepared foods in other ways, such as pounding, grinding, drying or fermenting, but this appears to be for at least 20,000 years, which has been sufficient time to influence gene expression for digestive enzymes. Cooking and food processing expands the range of food that can be eaten, extends food availability into lean times and enhances digestibility. Cooking also detoxifies food to some extent, destroys infective agents, decreases eating time and slightly increases the efficiency of assimilation of energy substrates. On the other hand, cooking can destroy some nutrients and produce toxic products. The human digestive system is suited to a processed food diet because of its smaller volume, notably smaller colonic volume, relative to the intestines of other species, and because of differences from other primates in dentition and facial muscles that result in lower bite strength. There is no known group of humans which does not consume cooked foods, and the modern diet is dominated by processed foods. Grapevine is moderately sensitive to salinity and accumulation of toxic levels of Cl in leaves is the major reason for salt-induced symptoms. In this study, apoplastic Cl uptake and transport mechanism(s) were investigated in two grapevine ( Vitis sp.) rootstock hybrids differing in salt tolerance; 1103 Paulsen (salt tolerant) and K 51-40 (salt sensitive). Increased external salinity caused high Cl accumulation in shoots of the salt sensitive K 51-40 in comparison to Paulsen. Measurement of NO net fluxes under high salinity showed that by increasing external Cl concentrations K 51-40 roots showed reduced NO accumulation. This was associated with increased accumulation of Cl. In comparison to Paulsen, K 51-40 showed reduced NO/Cl root selectivity with increased salinity, but Paulsen had lower selectivity over the whole salinity range (0-45 mM). To examine if root hydraulic and permeability characterisations accounted for differences between varieties, the root pressure probe was used on excised roots. This showed that the osmotic Lp was significantly smaller than hydrostatic Lp, but no obvious difference was observed between the rootstocks. The reflection coefficient ( σ) values (0.48-0.59) were the same for both rootstocks, and root anatomical studies showed no obvious difference in apoplastic barriers of the main and lateral roots. Comparing the uptake of Cl with an apoplastic tracer, PTS (3-hydroxy-5,8,10-pyrentrisulphonic acid), showed that there was no correlation between Cl and PTS transport. These results indicated that bypass flow of salts to the xylem is the same for both rootstocks (0.77 ± 0.2 and 1.05 ± 0.12 %) and hence pointed to differences in membrane transport to explain difference in Cl transport to the shoot. The blood group antigen Lewis Y is expressed on epithelial tumors of the respiratory, digestive and reproductive system. Despite being regarded as an attractive target for immunotherapy, its function is still not well defined and its prognostic value remains a subject of discussion. Eighty-three paraffin-embedded tissue sections of non-small cell lung cancer (NSCLC) patients in stage I–IIIa, who underwent surgical resection of the primary tumor (73% male; 43% adenocarcinoma), were stained with a new, highly specific monoclonal antibody against Lewis Y (clone A70-C/C8). A positive Lewis Y expression was observed in 51% of patients; adenocarcinomas were favorably stained (67%). Multivariate analysis identified stage I, blood group A or AB and Lewis Y expression on tumor cells to be independent markers for improved survival after tumor resection (p = 0.024, 0.043, 0.003, respectively). In summary, unlike in several previous studies the presence of Lewis Y on tumor cells is a favorable prognostic factor in this cohort of resected NSCLC patients. Coexisting blood group antigen A may be of additional positive prognostic impact. We hypothesize that related blood group antigens both on tumor cells and in peripheral blood may have an underestimated function for progression in resected NSCLC. Copyright © 2008 S. An improved genetic linkage map was constructed from a peach Ferjalou Jalousia® × Fantasia (J×F) F2 population. Ferjalou Jalousia® is a flat low-acidity clingstone peach, and Fantasia is a round, normally acidic freestone peach. This population is segregated for six Mendelian characters: pollen sterility, peach or nectarine fruit, flat or round fruit, clingstone, or freestone fruit. It also segregates for the D major gene controlling the fruit’s low acidity. A new character is reported here for the first time that segregates as a Mendelian character: trees bearing aborting fruits. These trees have flowers, but fruits start to fall 2 months after blooming. This recessive character has been named Af. We demonstrate that it is linked to the flat shape of the fruit. The previous map obtained from this cross was constructed using 63 individuals, whereas the present map was constructed using 207 individuals. Moreover, 82 simple-sequence repeat (SSR) markers, including 10 expressed sequence tag-SSRs, and 43 amplified fragment length polymorphism (AFLP) markers were added. Molecular markers linked to the six Mendelian characters were identified, and one of them has already been used for marker-assisted selection. The hypothesis of disease-related large mammal extinction has new support. A unique pathologic zone of resorption was first noticed in a Hiscock Mammut americanum metacarpal. The pathognomonic zone of resorption was present in fifty-nine (52%) of 113 skeletons with feet available for examination. Metacarpals and metatarsals were most commonly affected. Associated rib periosteal reaction is highly suggestive of tuberculosis and the foot lesions were identical to that documented in Bison as pathognomonic for tuberculosis. Recognizing that only a portion of animals infected by infectious tuberculosis develop bone involvement, the high frequency of the pathology in M. americanum suggests that tuberculosis was not simply endemic, but actually pandemic, a hyperdisease. Pathognomonic metacarpal undermining is a skeletal pathology that has been associated with Mycobacterium tuberculosis in bovids. Postcranial artiodactyl, perissodactyl, and carnivore skeletons were examined in major university and museum collections of North America and Europe for evidence of this and other pathology potentially attributable to tuberculosis. Among nonproboscidean mammals from pre-Holocene North America, bone lesions indicative of tuberculosis were restricted to immigrant bovids from Eurasia. No bone lesions compatible with diagnosis of tuberculosis were found in large samples of other pre-Holocene (164 Oligocene, 397 Miocene, and 1,041 Plio–Pleistocene) North American mammals, including 114 antilocaprids. Given the unchanged frequency of bovid tubercular disease during the Pleistocene, it appears that most did not die from the disease but actually reached an accommodation with it (as did the mastodon) (Rothschild and Laub 2006). Thus, they were sufficiently long-lived to assure greater spread of the disease. The relationships of the proboscidean examples need further study, but present evidence suggests a Holarctic spread of tuberculosis during the Pleistocene, with bovids acting as vectors. While the role of other animals in the transmission of tuberculosis could be considered, the unique accommodation achieved by bovids and mastodons makes them the likely “culprits” in its spread. Palaeomicrobiology is an emerging field that is devoted to the detection, identification and characterization of microorganisms in ancient remains. Data indicate that host-associated microbial DNA can survive for almost 20,000 years, and environmental bacterial DNA preserved in permafrost samples has been dated to 400,000–600,000 years. In addition to frozen and mummified soft tissues, bone and dental pulp can also be used to search for microbial pathogens. Various techniques, including microscopy and immunodetection, can be used in palaeomicrobiology, but most data have been obtained using PCR-based molecular techniques. Infections caused by bacteria, viruses and parasites have all been diagnosed using palaeomicrobiological techniques. Additionally, molecular typing of ancient pathogens could help to reconstruct the epidemiology of past epidemics and could feed into current models of emerging infections, therefore contributing to the development of appropriate preventative measures. The publication of The Osteological Paradox (Wood et al. , 1992, Current Anthropology, 33:343–370) a decade ago sparked debate about the methods and conclusions drawn from bioarchaeological research. Wood et al. (1992, Current Anthropology, 33:343–370) highlighted the problematic issues of selective mortality and hidden heterogeneity in frailty (susceptibility to illness), and argued that the interpretation of population health status from skeletal remains is not straightforward. Progress in bioarchaeology over the last few years has led to the development of tools that will help us grapple with the issues of this “osteological paradox.” This paper provides a review of recent literature on age and sex estimation, paleodemography, biodistance, growth disruption, paleopathology, and paleodiet. We consider how these advances may help us address the implications of hidden heterogeneity in frailty and selective mortality for studies of health and adaptation in past societies. Summary. Peptides of the V3 loop of the HIV-1 envelope glycoprotein gp120 have been shown to bind with high affinity to the immunophilins cyclophilin (Cyp) A, CypB and the FK506-binding protein 12 (FKBP12) [10]. We investigated whether immunophilins affect HIV-1 infection by assuming they are able to bind to the V3 loop of gp120. T cells and peripheral blood mononuclear cells were infected with T-cell-tropic or macrophage-tropic HIV-1 strains, respectively, in the presence of different concentrations of immunophilins. P24 antigen ELISA and real-time PCR measurements demonstrated that exogenously added immunophilins do not influence HIV-1 infection. CypA is known to interact with the HIV-1 Gag polyprotein and to be incorporated into the virions. This incorporation can be prevented by cyclosporin A (CsA) resulting in a decreased yield of infectious virus, the mechanism of which is unknown. We measured a normal production of proviral DNA in the first round of infection in CsA treated cells but afterwards, infection was decreased if CsA was present. Pre-treatment of the HIV-1 inocula with CsA, blocking the function of virus-associated CypA, did not inhibit the ensuing yield of infection. We therefore may conclude that endogenous CypA exerts its action after reverse transcription but before virus maturation, probably during capsid formation. Skeletons represent the most direct evidence of the biology of past populations, and their study provides insight into health and well-being, dietary history, lifestyle (activity), violence and trauma, ancestry, and demography. These areas help inform our understanding of a range of issues, such as the causes and consequences of adaptive shifts in the past (e.g., foraging to farming, sedentarism), the biological impact of invasion and colonization, differential access to food and other resources (e.g., by gender or status), and conflict and warfare. Central to bioarchaeological inquiry are the interaction between biology and behavior and the role of environment on health and lifestyle. Bioarchaeological analysis has traditionally focused on local settings. However, important perspective on general questions of human adaptation is possible both regionally and globally. The present article reviews the various definitions and terminology of pollen viability and longevity as well as the various tests of its assessment. We compare the advantages and the disadvantages of each method and suggest some practical implications as revealed by the extensive data. We recognize eight main hypotheses concerning the ecology and the evolution of pollen longevity and critically evaluated them according to the literature. The hypotheses are grouped as follows: (1) Desiccation risk-carbohydrate content; (2) Pollen packaging; (3) Pollen competitive ability; (4) Pollinator activity-stigma receptivity duration; (5) Self-pollination chance; (6) Pollen exposure schedule; (7) Pollen travel distance, and (8) Pollen removal chance. The effects of sodium chloride (NaCl) and abscisic acid (ABA) on protein synthesis, protein hydrolysis, glutamine synthetase, Δ-pyrroline-5-carboxylate reductase and amino acid composition have been studied in aerial parts of Pennisetum typhoides (Stapf et Hubbard) seedlings. Whereas protein synthesis is inhibited by NaCl and ABA, the activity or the synthesis of leucine-arylamidase, glutamine synthetase and Δ-pyrroline-5-carboxylate reductase is promoted by both the substances. The results of the amino acid analysis showed that in the presence of NaCl and ABA the amounts of alanine, aspartate and ammonia decreased, while the concentrations of glutamate, and especially that of proline increased greatly. The effect of water stress on growth (fresh weight, dry weight), water relations (water saturation deficit, water potential, osmotic pressure), and proline metabolism in Phaseolus vulgaris were studied. Experimentally, water deficit was produced by reduced watering of the bean plants. This resulted in a decrease in water potential and leaf fresh and dry weight. Increases in the water saturation deficit and the osmotic pressure of the sap were, however, recorded. Water stress was also induced by treatment of the plants with polyethylene glycol, but its effects on the above mentioned parameters were different. In addition, necrosis of the foliage was observed. According to the present results, polyethylene glycol seems to be suitable only for the induction of short-termed water stress conditions. The effects of water stress on growth and on water relations of the plants were accompanied by a marked increase in the free amino acid content, especially that of the free proline content of the plants. The activities of the proline dehydrogenase and glutamate dehydrogenase were stimulated under water stress conditions, indicating that proline accumulation in water stressed plants is not attributed to an inhibited proline breakdown. The accumulated proline was metabolized rapidly once the water deficit of the plant was relieved by watering. The ability of the plant to accumulate proline might be of ecological importance for the plant and might be an adaption mechanism of the plant to overcome short periods of drought. Sugarcane var. CO 740 is known to be salt-sensitive. Effect of increasing concentrations of NaCl and NaSO as prolonged salt stress, and of −10 bar NaCl and PEG as well as 10 EC NaCl NaSO, MgCl and MgSO as abrupt stress in base-nutrient solution has been investigated on proline metabolism. Neither of these stress conditions could effectively stimulate proline accumulation in the leaves of treated sugarcane plants, in spite of the abrupt drop in leaf osmotic potential. Tobacco varieties carrying the N gene from Nicotiana glutinosa respond to infection by Tobacco Mosaic Virus (TMV) by forming necrotic local lesions (hypersensitive reaction), thereby localizing the infection. In this study, infected mesophyll leaf tissue of N. tabacum Samsun NN was treated with the non-permeating, non-metabolizable carbohydrate mannitol. The local lesions developed under iso-osmotic conditions (0.28 M mannitol), though with a slight delay and with a reduced rate of growth, as compared to those on attached leaves. At increasing plasmolysing concentrations of mannitol, necrotization was progressively inhibited, but not completely suppressed. The leaf tissue produced tiny translucent zones, with a delay that increased between the virus inoculation and application of the plasmolytica. Activities of phenylalanine ammonia-lyase (PAL, EC 4.3.1.5) and O-methyltransferase (OMT, EC 2.1.1.6) are strongly stimulated in hypersensitively reacting tobacco and were used as biochemical markers in the present study. This study was done to determine whether the inhibitory effect of plasmolysis on the elicitation of the hypersensitive response is due to a decrease in virus spread, resulting from the rupture of plasmodesmata or, at least in part, to metabolic alterations of the host cell exposed to osmotic stress. Since necrotization is normally preceded by intense virus multiplication, the inhibitory effects found for early applications (i.e., before local lesion appearance) of plasmolytica could easily be related to an inhibition of virus spread which also occurred in similarly treated leaf tissue of the systemically reacting variety Samsun. The most meaningful data were obtained from mannitol treatments performed on leaf tissue already carrying local lesions, i.e., in which the elicitor(s) and/or the factor(s) of necrotization were already operating. Under iso-osmotic conditions, we found the stimulated PAL and OMT activities characteristic of the hypersensitive response. At plasmolysing concentrations of mannitol, we observed the counteracting effects of two different mechanisms controlling the phenylpropanoid enzymes. Floating the leaf material on the liquid medium induced an ageing-like effect with a continuous increase in enzyme activities that was independent on osmotic pressure and sensitive to cycloheximide. At the same time, the stimulated enzyme activities related to hypersensitivity decreased at a rate related to osmotic pressure. Since PAL and OMT of tobacco leaves are long-lived enzymes, it is likely that the increased de novo synthesis of the enzymes was suppressed by plasmolysis while their degradation and/or inactivation was maintained or even increased. From these results it is concluded that the apparent inhibition of the hypersensitive response by plasmolysis is due to both a decrease in virus spead (artificially caused by the rupture of connections between cells) and to drastic metabolic alterations of the host cell exposed to high osmotic pressure. Axenic cultures of bacteroid-containing protoplasts were isolated from root nodules of Vigna unguiculata L. Walp. Dimensions of the protoplasts were 35 to 135 μm long x 35 to 95 μm wide. Yields were about 30 to 50 mg dry weight per gram fresh weight of nodules. About 5x10 protoplasts packed into 1 ml of basal medium under the influence of gravity. When incubated in hypertonic, nitrogen-free media, freshly isolated protoplasts began to reduce acetylene to ethylene after a lag period of 24 to 48 h. Various additions to the basal medium showed that the system possessed functional glycolytic and tricarboxylic acid pathways. Endogenous application of various intermediary metabolites stimulated both acetylene reduction and respiration, though not often equally. As acetylene reduction, but not respiration, was inhibitable by both asparagine and glutamine, the system appears suitable for the study of mechanisms controlling symbiotic nitrogen fixation. The biosynthesis of Fraction I protein in isolated protoplasts is compared with that in the plant. Radioactive precursors were incorporated into isolated protoplasts ('in vitro' labeling) and into leaves, from which the protoplasts were isolated later ('in situ' labeling). The formation of cell wall fibres at the surface of isolated leaf protoplasts has been studied by scanning electron microscopy. Fibres are not formed on incubated protoplasts until a lag period has elapsed. This period is about 8 h for leaf protoplasts of Nicotiana tabacum and about 45 h for leaf protoplasts of Antirrhinum majus. In the case of Antirrhinum protoplasts the length of the lag period is dependent on the concentration of osmoticum present during the incubation period. If regenerating protoplasts are briefly treated with dilute cellulase, the newly formed wall is completely digested. Such protoplasts are capable of producing new fibres at the surface within minutes of their return to a nutrient medium. It has previously been postulated that when plants are stressed by certain changes in patterns of weather they become a better source of food for invertebrate herbivores because this stress causes an increase in the amount of nitrogen available in their tissues for young herbivores feeding on them. And this may cause outbreaks of such phytophagous invertebrates. Evidence is now presented that a similar physiological mechanism appears to operate when a wide variety of apparently unrelated environmental factors impinge on plants or parts of plants in such a way as to perturb their metabolism. A broken branch, lightning strike, fire, nutrient deficiencies or an otherwise adverse site; all may have this effect. With the advent of modern man the available agencies increase and diversify to include pesticides, irradiation and air pollutants. One common metabolic response by plants to all such agents impinging on them seems to be equivalent to that found in senescing plant tissues - the breakdown and mobilization of nitrogen in soluble form away from the senescing/stressed tissues. Young herbivores which chance to feed on such stressed/senescing tissues have a greater and more readily available supply of nitrogen in their food than they would have had if feeding on unstressed plants. As a result many more of them survive, and there is an increase in abundance of their kind. Such increases may be quite localised and short-lived or more widespread and persistent, depending on the extent and duration of the stress experienced by the plants. And in the face of this improved nutrition and survival of the very young, predators and parasites seem to have only a minor influence on subsequent changes in abundance of their herbivorous prey. Another effect of increased mobilization of nitrogen in stressed plants is an increase in the quantity of the seed that they set. This has led to the conclusion that increased abundance of some species of birds at such times is due to a greater supply of seeds as winter food for recent fledglings. But it may be that the increased abundance is due to the synchronous increase in phytophagous insects providing a richer source of protein food for laying hens and growing nestlings. The usefulness of radioiodinated fibrinogen in tumor localization and of coagulation in 80 cancer patients was investigated. Tumors externally detected demonstrated positive localization in 63% of all cases and in 100% of osteosarcomas of limbs. Fibrinogen half-life was reduced in cancer patients particularly in cases with lymphoma, Hodgkin's disease and osteosarcoma irrespectively of basal plasma fibrinogen levels. In lymphoma and ovarian cancer, fibrinogen degradation products were correlated to radiofibrinogen T/2 reduction. In patients with the lowest fibrinogen T/2 and platelet count, heparin infusion therapy returned the fibrinogen half-life to normal range. Present results suggest the existence of enhanced coagulation both inside and around the tumor and/or dissemination in early cancer as well as in the more advanced stage. [ABSTRACT FROM AUTHOR]

For years, the issues related to the origin of the Goths and their early migrations in the Iron Age have been a matter of hot debate among archaeologists. Unfortunately, the lack of new independent data has precluded the evaluation of the existing hypothesis. To overcome this problem, we initiated systematic studies of the populations inhabiting the contemporary territory of Poland during the Iron Age. Here, we present an analysis of mitochondrial DNA isolated from 27 individuals (collectively called the Mas-VBIA group) excavated from an Iron Age cemetery (dated to the 2nd-4th century A.D.) attributed to Goths and located near Masłomęcz, eastern Poland. We found that Mas-VBIA has similar genetic diversity to present-day Asian populations and higher diversity than that of contemporary Europeans. Our studies revealed close genetic links between the Mas-VBIA and two other Iron Age populations from the Jutland peninsula and from Kowalewko, located in western Poland. We disclosed the genetic connection between the Mas-VBIA and ancient Pontic-Caspian steppe groups. Similar connections were absent in the chronologically earlier Kowalewko and Jutland peninsula populations. The collected results seem to be consistent with the historical narrative that assumed that the Goths originated in southern Scandinavia; then, at least part of the Goth population moved south through the territory of contemporary Poland towards the Black Sea region, where they mixed with local populations and formed the Chernyakhov culture. Finally, a fraction of the Chernyakhov population returned to the southeast region of present-day Poland and established the archaeological formation called the "Masłomęcz group".
