Logistical constraints concerning parasite sample storage can hinder progress with the discovery of genetic variation on a global scale. New storage methods are being developed to address this, but require testing in order to understand how widely applicable these methods are. Whatman FTA cards have been tested previously under laboratory conditions for storing low-concentration Trichomonas gallinae isolates with the conclusion that they are not suitable, but have not been tested under field conditions. Here, we conducted a field-test, comparing FTA cards with storage in ethanol for T. gallinae samples collected and cultured from wild Columbiformes in Africa using standard field methods, before transportation to the UK. After 6 months storage, both methods resulted in an overall prevalence of 100% following PCR amplification (n = 59), suggesting that FTA cards are suitable for estimation of T. gallinae prevalence. However, samples stored in ethanol produced more, and longer, sequences than those stored on FTA cards. These data suggest storage in ethanol is preferable for the acquisition of high quality genetic strain data, but that FTA cards can be used successfully to ascertain infection prevalence and identify parasite strains under field conditions. Previous palaeomagnetic studies have allowed the recognition of a distinctive area of Neotethyan oceanic rocks, including the Troodos ophiolite in Cyprus and the Hatay ophiolite to the east in southern Turkey, that underwent 90° of anticlockwise rotation between Late Cretaceous (Campanian) and Early Eocene time. The southern and western boundaries of this rotated Troodos-Hatay microplate have been inferred to lie within, or adjacent to, zones of deformed oceanic and continental margin rocks that are now exposed in southern and western Cyprus; however, the northern boundary of the microplate remains undefined. Relevant to this problem, palaeomagnetic data are presented here from basaltic lavas exposed along the Kyrenia Range, mostly from Late Cretaceous (Maastrichtian) sites and one Eocene site. A positive inclination-only fold test demonstrates that remanences are pre-deformational in age, and positive conglomerate tests show that magnetic remanences were acquired before Late Eocene-Early Oligocene time, together suggesting that primary magnetizations are preserved. Data from the eastern Kyrenia Range and the Karpas Peninsula (the easternmost extension of the Kyrenia Range) document significant relative tectonic rotation between these localities, with no rotation in the eastern range versus 30° of anticlockwise rotation of the Karpas Peninsula. Unfortunately, palaeomagnetic sites from the western Kyrenia Range did not yield tectonically interpretable magnetization directions, probably due to complex poly-phase thrusting and folding, and the central range also yielded no interpretable data. However, the available palaeomagnetic data are sufficient to demonstrate that the Kyrenia terrane underwent a separate rotation history to the Troodos-Hatay microplate and also implies that the northern boundary of the Troodos-Hatay microplate was located between the Troodos ophiolite and the Kyrenia Range. The former microplate margin has since been overridden and concealed by two phases of southwards thrusting and folding of the Kyrenia Range units (Mid-Eocene; latest Miocene-earliest Pliocene). The likely cause of the anticlockwise rotation affecting the Karpas Peninsula, and by implication the curvature of the Kyrenia Range as a whole, relates to regional late-stage subduction and diachronous continental collision. The Southern Neotethys sutured in SE Turkey during the Early Miocene, whereas a relict ocean basin remained further west in the easternmost Mediterranean, allowing a remnant N-dipping subduction zone to retreat southwards and so induce the present-day arcuate shape of the Kyrenia Range. Large-offset oceanic detachment faults are a characteristic of slow- and ultraslow-spreading ridges, leading to the formation of oceanic core complexes (OCCs) that expose upper mantle and lower crustal rocks on the seafloor. The lithospheric extension accommodated by these structures is now recognized as a fundamentally distinct "detachment-mode" of seafloor spreading compared to classical magmatic accretion. Here we demonstrate a paleomagnetic methodology that allows unequivocal recognition of detachment-mode seafloor spreading in ancient ophiolites and apply this to a potential Jurassic detachment fault system in the Mirdita ophiolite (Albania). We show that footwall and hanging wall blocks either side of an inferred detachment have significantly different magnetizations that can only be explained by relative rotation during seafloor spreading. The style of rotation is shown to be identical to rolling hinge footwall rotation documented recently in OCCs in the Atlantic, confirming that detachment-mode spreading operated at least as far back as the Jurassic. Trichomonosis, caused by the protozoan Trichomonas gallinae, is an emerging infectious disease in finches, and is more commonly found in columbids and raptors. Infections can be sub-clinical or cause morbidity and mortality, but the parasite is currently only detectable by incubation of an oral swab. Here, we test whether T. gallinae parasites can be detected by PCR from faecal or non-cultured samples from the oral cavity and gastrointestinal tract of infected Turtle Doves ( Streptopelia turtur). PCR did not detect T. gallinae parasites in any faecal samples screened, and in only 1 of 11 oral/gastrointestinal samples (from the mouth of a nestling suspected to have died from trichomonosis). We conclude that both oral swabs and parasite culture are still necessary to detect the sub-clinical presence of T. gallinae infection in birds. [ABSTRACT FROM AUTHOR]
