Background: The internal strain distribution developing during plastic deformation is important for understanding the mechanical properties of polycrystalline materials. Such distributions may be determined by the microstructural feature tracking method. However, the strain accuracy and the sources of uncertainty of this method are not well quantified yet. Objective: Evaluate the accuracy of local strain determination based on laboratory-based multimodal X-ray tomography measurements. Methods: The plastic deformation behavior of a particle-contained fully recrystallized Al-4mass%Cu alloy was characterized during in-situ tensile testing using absorption contrast tomography to reveal marker particles and diffraction contrast tomography to reveal the grain structure. Results: It was found that the accuracy in strain measurement is inversely proportional to the particle distance, whereas it is independent of the particle size, and the accuracy is not biased by grain boundaries. As an illustrative example, the preliminary analysis of the microstructure-strain relationships reveals significant strain differences both between and within individual grains. Conclusions: The results document the validity and limitations of the microstructural feature tracking method for local strain measurements using a laboratory X-ray source, are of importance for local strain analysis in general and may be considered as an alternative to other methods, e.g. digital image correlation. The measurements allow evaluation of effects of the grain microstructure on the local strain development during plastic deformation. By a preliminary analysis of the strain difference between two selected grains, it is suggested that the grain shape and crystallographic relationships between neighboring grains are of importance for the development of local strains. Purpose: To compare the range of reach of our newly designed omni-directional ureteroscope (URF-Y0016), compared to the commonly used URF-P6, FlexX2s, and LithoVue™ scopes, in the upper, middle, and lower calyces in an ex-vivo pyelocaliceal model. Methods: We fabricated a three-dimensional pyelocaliceal model of the upper, middle, and lower pole calyces using urethane and acrylic resin. The inner surface of the dome of each calyx was engraved with reference lines along eight directions, set at 10° of latitude from the top to the base of the dome, and at angles of 0–90°, to precisely determine the range of reach of each scope. The main feature of the URF-Y0016 scope is the omni-directional bending of the tip of the flexible ureteroscope, with the control of these four directions integrated into a handgun-type control unit with a joystick. The range of reach within each calyx was measured by four expert surgeons. Results: The URF-Y0016 scope provided a greater range of reach along all directions in the lower pole calyx compared to URF-P6, FlexX2s, and LithoVue™ scopes (p < 0.001), particularly along the anterior–posterior direction in the lower lobe calyx. However, the URF-Y0016 scope did not influence the improvement of reach range in the upper and middle pole calyx compared to URF-P6, FlexX2s, and LithoVue™ scopes (p = 0.08, p = 0.296). Strain-rate and temperature dependences of deformation behavior of ultrafine-grained (UFGed) AZ61Mg alloy were examined. For this purpose, AZ61Mg alloy specimens were subjected to multi-directional forging (MDFing) under decreasing temperature conditions to have various grain sizes. The average grain sizes attained by MDFing to 1, 3, 6 and 9 passes were approximately 8, 2, 0.5 and 0.3 μm, respectively. A superior balance of the mechanical properties of strength and ductility at room temperature was achieved by MDFing to three passes and over. The strain-rate sensitivity of mechanical properties of the MDFed specimens increased with decreasing grain size. The specimens with grain sizes of 0.5 and 0.3 μm (0.5 or 0.3 specimen) exhibited stronger strain-rate and temperature dependences of total elongation than those with grain sizes of 8 and 2 μm (8 or 2 specimen). This can be partially ascribed to grain-boundary sliding, since an AFM observation revealed the occurrence of room-temperature grain-boundary sliding in the 0.3 specimen. The activation volume V* for the 8, 2 and 0.5 specimen increased with increasing temperature, while the 0.3 specimen exhibited an inverse temperature dependence of V*. Background:Neoadjuvant chemoradiotherapy (NCRT) for advanced rectal cancer (RC) is a well-evidenced therapy; however, some RC patients have no therapeutic response. Patient selection for NCRT so that non-responsive patients are excluded has been subjective. To date, no molecular markers indicating radiation sensitivity have been reported.Methods:We irradiated six colorectal cancer (CRC) cell lines and identified HCT116 cells as radiation-sensitive and HCT15 and DLD-1 cells as radiation resistant. Using a microarray, we selected candidate radiation sensitivity marker genes by choosing genes whose expression was consistent with a radiation-resistant or sensitive cell phenotype.Results:Among candidate genes, cellular retinol binding protein 1 (CRBP1) was of particular interest because it was not only induced in HCT116 cells by tentative 10 Gy radiation treatments, but also its expression was increased in HCT116-derived radiation-resistant cells vs parental cells. Forced expression of CRBP1 decreased the viability of both HCT15 and DLD-1 cells in response to radiation therapy. We also confirmed that CRBP1 was epigenetically silenced by hypermethylation of its promoter DNA, and that the quantitative methylation value of CRBP1 significantly correlated with histological response in RC patients with NCRT (P=0.031).Conclusions:Our study identified CRBP1 as a radiation-sensitive predictor in RC. Purpose: The aim of this retrospective study was to evaluate the feasibility, safety, and effectiveness of stereotactic body radiotherapy (SBRT) for pulmonary metastases.Patients and Methods: Between April 2007 and March 2011, 87 patients underwent SBRT for pulmonary metastases using the in-house Air-Bag System(TM) to obtain the four-dimensional image for treatment planning and to reduce intrafractional intrathoracic organ motion with abdominal compression to reduce the risk of radiation pneumonitis. Survival and respiratory adverse events were analyzed.Results: The 2- and 3-year overall survival (OS) rates were 47 and 32 %, and the corresponding cause-specific survivals were 52 and 36 %. The 2- and 3-year OS rates were 57 and 49 % for patients in group 1, respectively, while the corresponding OS rates were 48 and 21 %, and 40 and 32 % for patients in groups 2 and 3, respectively. The 2- and 3-year local control (LC) rates were 80 and 80 %, respectively. The corresponding intrathoracic progression-free survival rates were 40 and 32 %, respectively. Concerning adverse respiratory events after SBRT for pulmonary metastases, 14 % were grade 0 (G0), 66 % G1, 13 % G2, 6 % G3, and 1 % G4. Concerning the adverse respiratory events (NCI-CTC) by grade scale, 1- and 2-year cumulative probabilities of radiation pneumonitis were 12 and 20 % for G2 and 4 and 10 % for G3/4, respectively. The mean values for cumulative V20 were 11.6 ± 8.5 %, 29.8 ± 18.6 %, and 25.7 ± 12.8 % in G0/1, G2, and G3/4, respectively. The number of pulmonary metastases that could be safely treated with SBRT was 6 PTVs (or seven gross tumor volumes) within a cumulative V20 of 30 % under the restricted intrafractional respiratory tumor motion using the Air-Bag System(TM).Conclusion: We propose that the number of pulmonary metastases that can be safely treated with SBRT is 6 PTVs with a cumulative V20 of 30 % under the restricted respiratory tumor motion using the Air-Bag System(TM). We report a rare case of life-threatening hemorrhage from the inferior epigastric artery that was injured after stoma construction. The purpose of this study was to examine the incidence of new or recurrent venous thromboembolism (VTE) after retrieval of inferior vena cava (IVC) filters and risk factors associated with such recurrence. Between March 2001 and September 2008, at our institution, implanted retrievable vena cava filters were retrieved in 76 patients. The incidence of new or recurrent VTE after retrieval was reviewed and numerous variables were analyzed to assess risk factors for redevelopment of VTE after filter retrieval. In 5 (6.6%) of the 76 patients, redevelopment or worsening of VTE was seen after retrieval of the filter. Three patients (4.0%) had recurrent deep venous thrombosis (DVT) in the lower extremities and 2 (2.6%) had development of pulmonary embolism, resulting in death. Although there was no significant difference in the incidence of new or recurrent VTE related to any risk factor investigated, a tendency for development of VTE after filter retrieval was higher in patients in whom DVT in the lower extremities had been so severe during filter implantation that interventional radiological therapies in addition to traditional anticoagulation therapies were required (40% in patients with recurrent VTE vs. 23% in those without VTE; p = 0.5866 according to Fisher's exact probability test) and in patients in whom DVT remained at the time of filter retrieval (60% in patients with recurrent VTE vs. 37% in those without VTE; p = 0.3637). In conclusion, new or recurrent VTE was rare after retrieval of IVC filters but was most likely to occur in patients who had severe DVT during filter implantation and/or in patients with a DVT that remained at the time of filter retrieval. A new cloud parameterization based on prognostic equations for the subgrid-scale fluctuations in temperature and total water content is introduced for global climate models. The proposed scheme, called hybrid prognostic cloud (HPC) parameterization, employs simple probability density functions (PDFs) to the horizontal subgrid-scale inhomogeneity, allowing them to vary in shape in response to small-scale processes such as cumulus detrainment and turbulent mixing. Simple tests indicate that the HPC scheme is highly favorable as compared to a diagnostic scheme in terms of the cloud fraction and cloud water content under either uniform or non-uniform forcing. The relevance of the HPC scheme is investigated by implementing it in an atmospheric component model of the climate model MIROC with a coarse resolution of T42. A comparison of the short-term integrations between the T42 model and a global cloud resolving model (GCRM) reveals that the HPC scheme can reproduce, to a certain degree, the subgrid-scale variance and skewness of temperature and total water content simulated in the GCRM. It is also found that the HPC scheme significantly alters the climatological distributions in cloud cover, precipitation, and moisture, which are all improved from the model using a conventional diagnostic cloud scheme. This study evaluated the mid-term results of total knee arthroplasty (TKA) following high tibial osteotomy (HTO), comparing posterior cruciate-retaining prostheses to posterior stabilized prostheses. The Knee Society score for the entire group (20 knees) improved significantly from 62 (median) preoperatively to 87 at the latest follow-up. The postoperative Knee Society score of 85 in posterior cruciate-retaining prostheses (8 knees) was significantly inferior to the 94 score in posterior stabilized prostheses (12 knees). Of Knee Society score, Stability and ROM scores (17 and 21, respectively) in posterior cruciate-retaining TKA were inferior to those in posterior stabilized TKA (25 and 24, respectively). Since postoperative knee instability due to posterior cruciate ligament (PCL) insufficiency is thought to contribute to the inferior results of posterior cruciate-retaining prostheses after HTO, PCL-substituting TKA would be suitable for use after HTO. Purpose: The incidence of a unilateral aplastic distal vertebral artery (VA) has been reported as 0.2% of cases on cerebral angiography. During our daily magnetic resonance (MR) examinations, however, we frequently encounter MR angiograms (MRAs) that do not demonstrate unilateral VA. The purpose of this study was to calculate the frequency of aplastic unilateral distal VA by MR images performed for asymptomatic people.Materials and Methods: Over a time span of 1 year, 237 asymptomatic people (140 men, 97 women; ages 28-67 years, mean 54.4 years) underwent brain MRI during a "brain check-up examination" in our hospital. To identify an aplastic unilateral distal VA, we retrospectively compared three-dimensional time-of-flight MRA with basiparallel anatomic scanning (BPAS)-MRI which was designed for recognition of the arterial outer contour.Results: Aplasia of the unilateral distal VA was confirmed in 11 persons (4.6%). According to our classification, hypoplastic distal VA in 12 (5.1%) and asymptomatic acquired unilateral distal VA occlusion was also proved in 2 (0.8%).Conclusion: We found that the frequency of aplastic unilateral VA was 4.6% in asymptomatic people using a combination of MRA and BPAS-MRI for assessment of an intracranial VA. White-grained wheat cultivars have long been recognized to be less resistant to preharvest sprouting (PHS) than the red-grained ones. Previously two QTLs for grain dormancy, QPhs.ocs-3A.1 ( QPhs-3AS) and QPhs.ocs-4A.1 ( QPhs-4AL) were identified in a highly dormant Japanese red wheat, Zenkoujikomugi (Zen). Aiming at improvement of PHS tolerance in white-grained wheat, the introgression effect of these two QTLs in a white-grained population consisting of 40 recombinant inbred lines (RILs) developed from a cross between Zen and white-grained Spica was examined here. Random 20 RILs with red grains were also developed from the same cross and used as a control population. The RILs were grown in the field and in the glasshouse to evaluate the grain dormancy by germination test. Several SSR markers closely linked to the QPhs-3AS and QPhs-4AL were used to estimate the alleles at the QTLs. Dormancy variation in the RILs was significantly associated with the differences for grain color and the alleles at QPhs-3AS over several years. Although allelic variation was detected in a SSR marker closely linked to QPhs-4AL there was no difference in germination data between the Zen-allele and the Spica-allele groups. As expected, the red-grained RILs with the Zen allele at QPhs-3AS were the most dormant. Some white-grained RILs with the Zen allele at QPhs-3AS showed higher dormancy compared to the red-grained RILs with the alternative allele. These results demonstrated that introgression of the QPhs-3AS gene could contribute to the increased grain dormancy in white-grained wheat. [ABSTRACT FROM AUTHOR]

A major QTL for grain dormancy,QPhs.ocs-3A.1, derived from the highly dormant wheat Zenkoujikomugi (Zen), has been identified in a study made under a controlled environment. Further investigations were needed to dissect the precise position and expression ofQPhs.ocs-3A.1under different field conditions because the ability to detect genetic loci for grain dormancy traits is compromised by environmental effects and genotype/environment interactions. Group 4 chromosomes have also been shown to be possible sites of QTLs for grain dormancy. The objectives of this study were (1) to locate additional molecular markers in theQPhs.ocs-3A.1region, (2) to identify QTLs on the group 4 chromosomes and (3) to elucidate their combined effects. We examined the recombinant inbred lines (RILs) from a cross between Chinese Spring (CS) and Zen over a 3-year period in one location and 1 year in a different location. In an interval mapping studyQPhs.ocs-3A.1was mapped to within the 4.6 cM region flanked byXbarc310andXbcd907at the proximal end of the short arm of chromosome 3A.QPhs.ocs-3A.1was confirmed to be the predominant dormancy QTL since it explained a large portion (11.6-44.8%) of the phenotypic variation, and was strongly displayed under dormancy-breaking conditions or at low germination temperatures. ForQPhs.ocs-4A.1, identified on the long arm of chromosome 4A, andQPhs.ocs-4B.1, on the centromeric region of the long arm of Chr 4B, the LOD peak positions and the desirable allele were consistent between the trials, while the LOD scores and contribution to the phenotypic variation varied. Transgressive segregants were observed among the 125 RILs and most of them had a combination of the three alleles conferring a higher dormancy: the Zen alleles atQPhs.ocs-3A.1andQPhs.ocs-4A.1and the CS allele atQPhs.ocs-4B1. Rationale: Tetrahydrobiopterin (BH4) is a coenzyme of tyrosine hydroxylase (TH) and tryptophan hydroxylase (TPH), rate-limiting enzymes of monoamine biosynthesis. According to the monoamine hypothesis of depression, antidepressants will restore the function of the brain monoaminergic system, and BH4 concentration. Objective: To investigate the effects of fluvoxamine on BH4 levels and dopamine (DA) and serotonin (5-HT) turnover in the mesoprefrontal system, incorporating two risk factors of depression, social isolation and acute environmental change. Methods: Male ddY mice (6W) were divided into two housing groups, i.e. group-housing (eight animals per cage; 35 days), and isolation-housing (one per cage; 35 days), SC injected with fluvoxamine (20 or 40 mg/kg; days 29-35), and exposed to 20-min novelty stress (day 35). The levels of BH4, DA, homovanilic acid (HVA), 5-HT, and 5-hydroxyindoleacetic acid (5-HIAA) were measured in the prefrontal cortex and midbrain. Results: Under the group-housing condition, novelty stress significantly increased BH4 levels in both regions, and the HVA/DA ratio in the midbrain, whereas it did not change any parameters in either region under the isolation-housing condition. In the prefrontal cortex, fluvoxamine significantly decreased the 5-HIAA/5-HT ratio under the group-housing condition, and BH4 levels and the HVA/ DA ratio under the isolation-housing condition. In the midbrain, fluvoxamine significantly decreased all parameters, except for an increasing in the 5-HIAA/5-HT ratio under the isolation-housing condition. Conclusion: Isolation-housing suppressed the increase of BH4 levels and DA turnover elicited by novelty stress. Fluvoxamine suppressed BH4 levels, and DA and 5-HT turnover. The genetic diversity and expression of amoA of autotrophic ammonia oxidizers in wastewater treatment processes were investigated by RT-PCR and denaturing gradient gel electrophoresis (DGGE) in order to identify active components of ammonia-oxidizer populations in a such processes. Ammonia oxidizers, evidenced by the presence of amoA mRNA, were regarded as metabolically active. The DGGE profiles derived from amoA mRNA and from its gene, which were amplified by RT-PCR or PCR using samples collected from a bench-scale reactor treating high concentration of inorganic ammonia, were similar. In contrast, RNA and DNA-derived DGGE profiles from three domestic wastewater treatment facilities were different from each other. These data indicate that the dominant ammonia oxidizers in the bench-scale reactor exhibited ammonia-oxidizing activity, whereas some ammonia oxidizers in the domestic wastewater treatment facilities apparently did not express high levels of amoA mRNA. Copper bicrystals with different [011] symmetric tilt boundaries doped with Sb were tensile tested at several temperatures from 77 to 773 K. The fracture behavior was sensitive to the misorientation angle, Sb doping time, and test temperature. Grain boundaries were more embrittled with increasing the Sb doping time, though their tendency was also dependent on the misorientation angle. The degree of embrittlement of grain boundaries became lower as the test temperature increased to about 500 K, while it became higher again at higher temperatures. The segregation of Sb at [011] tilt boundaries also was systematically measured by energy-dispersive X-ray spectroscopy. A strong correlation was found among the fracture behavior at 77 and 773 K, grain-boundary segregation level of Sb, and grain-boundary energy before Sb doping: a higher-energy boundary has a higher level of Sb segregation and fractures more easily at a lower tensile stress. To find out gene dose effect of each of the three homoeologous Wx genes and their interaction on the production of granule-bound starch synthase (GBSS I) and amylose biosynthesis in the endosperm, Chinese Spring and its near-isogenic waxy types were crossed reciprocally and, obtained a plant population with varying doses of each Wx gene. The amount of GBSSI was increased linearly with increasing gene dose of either of Wx loci. In each of the three Wx loci, the change in amylose content was linear up to 3 doses, with a more potent capacity of Wx-B1a at any dose. Higher level of amylose production was observed in the reciprocal F1 grains than the expected effect of dose/s of each gene or additive effect of different allelic combination by artificially blend starches which have amylose produced by equivalent number of Wx alleles to that of relevant F1 cross. When Wx-B1a and Wx-A1a were combined, increase in amylose content was not in proportion to increase in gene dosage. The enhanced amylose synthesis was shown by 2-gene and 3-gene interaction, indicating that not only type of the three Wx genes and its dose but the interaction among them have significant roles in determining the amylose content. A major component of the observed genetic variation for pre-harvest sprouting in wheat (Triticum aestivum L.) appears to be the level of seed dormancy. Group 3 chromosomes have received attention as carrying the R genes for seed-coat color and the taVp1 genes that are orthologous to the maize Vp1 gene which encode a dormancy-related transcription factor. The objectives of the present study were to map quantitative trait loci (QTLs) for seed dormancy on chromosome 3A and to investigate an association between taVp1 or R-A1 and the QTLs detected. A mapping population in the form of recombinant inbred lines developed from the cross between the highly dormant Zenkoujikomugi (Zen) and Chinese Spring (CS) was utilized. Nineteen marker loci, including taVp1, were mapped on chromosome 3A. The taVp1 locus was located in the middle of the long arm, about 85 cM from the centromere. The population was evaluated in duplicate by growing them under controlled environment conditions. Two QTLs for seed dormancy, designated as QPhs.ocs-3A.1 and QPhs.ocs-3A.2, were identified on the short and long arms, respectively. QPhs.ocs-1 explained 23–38% of the phenotypic variation and the Zen allele had a striking effect on maintaining dormancy. QPhs.ocs-2, with a minor effect, was detectable only at the dormancy-breaking stage. Although QPhs.ocs-2 was loosely linked to taVp1 by around 50 cM, they are clearly distinct genes. Zen and CS carry the white R-A1a allele, and no QTL effect was detected in the vicinity region of R-A1. Hence it was concluded that the high dormancy associated with chromosome 3A of Zen is ascribable to QPhs.ocs-1 on the short arm but is not due to the direct contribution of either the taVp1 or R-A1 locus. The casein kinase CK2 is one of the major multifunctional protein kinases in cells that is expressed ubiquitously and is essential for survival. The α-subunit of CK2 is thought to be involved in light-regulated gene expression and rhythmic expression of genes by circadian rhythm in plants. The rice chromosome-3 region containing the photoperiod-response Hd6 gene, an orthologue of the CK2α genes of Arabidopsis and maize, is in synteny with the wheat chromosome-5A Vrn-A1 region. This evidence proposes two possibilities, first the wheat Vrn-A1 is an orthologue of the rice CK2α, and second the wheat CK2α which has not yet been identified is located independently but tightly linked to Vrn-A1. To clarify whether the wheat CK2α gene is conserved in the Vrn-A1 region and to elucidate the above two possibilities, we attempted to isolate this gene from the wheat cDNA library and to map it on the chromosome-5A region that is syntenous to the rice Hd6 region. The isolated cDNA clone showed an extremely high homology with the Arabidopsis CK2α gene. Using this clone as a probe genomic Southern-blot analyses of the aneuploid lines available in Chinese Spring assigned the wheat homologue of CK2α to the long arm of chromosome 5A. Furthermore, a linkage analysis using an F2 population having recombination in the Vrn-A1 region revealed that the wheat CK2α, designated as tck2a, is tightly linked to Vrn-A1 by 1.1 cM [ABSTRACT FROM AUTHOR]

The granule-bound starch synthase (GBSS I) encoded by the Wxgenes, is involved in amylose synthesis. For analyses of mechanisms of amylose synthesis and associated starch properties in hexaploid wheat, eight possible genotypes having different combinations of the three null alleles at the Wx loci with a common genetic background are a prerequisite. A near-isogenic population of doubled haploid (DH) lines was produced from Chinese Spring × waxy Chinese Spring F1 plants using the wheat × maize method. The Wx protein phenotypes of the DH progeny were examined by sodium dodecyl sulphate-polyacrylamide gel electrophoresis and found that the null alleles at each of the three Wx loci segregated in a Mendelian fashion. A field trial demonstrated no differences between the eight types for ear emergence time, plant height and grain yield traits. Amylose content in the endosperm starch was highest in the wild type while lowest in the waxy type having no Wx proteins. Comparison between single null types and double null types indicated that the amylose synthesis capacity of Wx-A1a allele is the lowest. Pasting properties of starch are the highest in the waxy type, followed by the double null types. Consequently, both peak viscosity and breakdown were negatively correlated with amylose content. The chain-length distribution analysis of amylopectin structure revealed no clear difference among the eight types,suggesting that the reduced GBSS I activity due to introgression of the null Wx alleles does not affect either the chain length or the degree of branching of amylopectin. The purpose of this study is to confirm whether T2-weighted imaging and perfusion imaging, i.e. autoradiogram of 14C-iodoantipyrine, on the course of brain edema correspond to each other or not. Cold injured rat brains were used as a model and were sequentially examined by both methods and compared with each other and with histological specimens. Special focus relies on the time changes in the lesions. High SI of T2-weighted images were observed and the percentages in the high SI area to the total brain area in the same slice were 4.7 +/- 0.31, 5.6 +/- 0.46 and 3.4 +/- 0.42 for 6, 24 and 48 hours, respectively. Three quantitative trait loci (QTLs) controlling seed dormancy were detected on group 4 chromosomes of wheat (Triticum aestivum L.) using 119 doubled haploid lines (DHLs) derived from a cross between AC Domain and Haruyutaka. A major QTL, designated QPhs.ocs-4A.1, was identified within the marker interval between Xcdo795 and Xpsr115 in the proximal region of the long arm of chromosome 4A. Two minor QTLs, QPhs.ocs-4B.2 on 4B and QPhs.ocs-4D.2 on 4D, were flanked by common markers, Xbcd1431.1 and Xbcd1431.2 in the terminal region of the long arms, suggesting a homoeologous relationship. These three QTLs explained more than 80% of the total phenotypic variance in seed dormancy of DHLs grown in the field and under glasshouse conditions. The AC Domain alleles at the three QTLs contributed to increasing seed dormancy. Comparative maps across wheat, barley and rice demonstrated the possibility of a homoeologous relationship between QPhs.ocs-4A.1 and the barley gene SD4, while no significant effects of the chromosome regions of wheat and barley orthologous to rice chromosome 3 region carrying a major seed dormancy QTL were detected. Chromosome 5A of wheat is known to carry a number of genes affecting adaptability and productivity. To localize quantitative trait loci (QTLs) controlling grain yield and its components, an RFLP map was constructed from 118 single-chromosome recombinant lines derived from the F1 between Chinese Spring (Cappelle-Desprez 5A) and Chinese Spring (Triticum spelta 5A). The map was combined with the field-trial data scored over 3 years. A total of five regions in chromosome 5A contributed effects on yield traits. Increases in grain yield, 50-grain weight and spikelet number/ear were determined by complementary QTL alleles from both parents. The effects associated with the vernalization requirement gene Vrn-A1 or a closely linked QTL were significant only in the favorable growing season where the later-flowering vrn-A1 allele from Cappelle-Desprez 5A produced a higher tiller number/plant and spikelet number/ear. The effects of the ear morphology gene q or closely linked QTL(s) were detected for grain yield and ear grain weight. Three other QTLs with minor effects were dispersed along chromosome 5A. These QTLs had large interactions with years due to changes in the magnitude of the significant response. The alleles from T. spelta, however, conferred a higher yield performance. The amylose/amylopectin ratio and the pasting properties of wheat starch are important in producing marketable flour products, especially Japanese noodles. To determine if null mutations at the three Wx loci confer differences in starch-pasting viscosity, we analyzed the variation associated with the null mutations in three separate sets of recombinant substitution lines of chromosomes 7A, 4A and 7D produced from crosses between Chinese Spring and three single-chromosome substitution lines carrying the null Wx alleles. Differential effects of null alleles at the three Wx loci on starch-pasting properties were revealed. With respect to chromosome 4A, the effect of the Wx-B1b allele, giving a higher peak and breakdown viscosity, was unambiguous. In addition, a QTL of minor effect was identified near the centromere on the short arm. The presence or absence of the Wx-A1 protein gave some variation in peak and breakdown viscosity, but the effects of Wx-Alb were much smaller than those of the Wx-Blb allele. Associated effects of the Wx-D1 locus were detected for the breakdown viscosity as the null Wx-D1b allele produced a higher viscosity than the wild-type Wx-D1a. While negative correlations between amylose content and breakdown viscosity were common in the three populations, the null mutations at the Wx loci produced some variation independent of amylose content. The purpose of the present study was to evaluate the clinical usefulness of urinary pyridinoline (Pyr) and deoxypyridinoline (Dpyr) in predicting therapeutic effects of estrogen and alfacalcidol (1alpha-D3) in patients with postmenopausal osteoporosis. We measured urinary excretion of Pyr and Dpyr, and determined bone mineral density (BMD) using a dual-energy x-ray absorptiometry in 48 women with osteoporosis (average age, 55.9+/-8.4 years). Patients were treated with estrogen (HRT, n = 13), 1alpha-D3 (n = 20), or calcium alone (n = 15). Baseline mean levels of urinary Pyr and Dpyr were significantly higher in the 48 patients compared to those in the age-matched postmenopausal women. The levels of urinary Pyr and Dpyr were inversely correlated with BMD. After treatment with estrogen or 1alpha-D3, a significant decrease of urinary Pyr and Dpyr was observed, and elevated urinary Pyr and Dpyr were reduced to the level in premenopausal women. A significant inverse correlation was found in Pyr and Dpyr at 6 months and in lumbar BMD after 24 months of treatment (r = -0.43 to -0.52; P < 0.01). Single crystals of a Cu-Fe alloy, which contained spherical γ-Fe particles, were compressed up to shear strain of 0.17 in liquid nitrogen bath. In the process of straining the structural (optical and TEM) observations in as-deformed and post-deformation annealed samples were provided. The substructure of deformed samples was characterized by slightly developed cell structure and lack of distinct layer-like arrangements of dislocations. Three kinds of particles were found: coherent and semi-coherent f.c.c. γ-Fe and martensiticly transformed b.c.c. α-Fe. The critical diameter for coherency loss was found to be 58 nm at the initial stage of deformation decreasing with strain to 50 nm. These values coincided with the theoretical estimations. It was suggested that relaxation of coherency strains around γ-Fe particles by the generation of interface dislocations might occur prior to the martensitic transformation. Objective: To assess the usefulness of a new axial radiographic technique in knees with patellofemoral arthritis (PF-OA).Design and Patients: After a marking wire had been attached to the skin on the tibial tuberosity so that the wire matched the width of the patellar tendon, an axial radiograph was taken at 30 degrees of flexion in 16 normal knees and 14 PF-OA knees in which computed tomographic analysis had revealed a laterally positioned tibial tuberosity at 30 degrees of flexion. The distance of the marking wire from the lateral condyle and from the patellar groove was compared between the two groups.Results: The marking wire was located significantly laterally in PF-OA knees compared with normal knees.Conclusion: An axial radiograph with a marking wire on the tibial tuberosity is useful for assessing the position of the tibial tuberosity in PF-OA knees. Bile salts regulate the subselection of phosphatidylcholine species secreted into bile and thereby modulate bile metastability. The aim of this study was to determine whether bile salts alter phosphatidylcholine species of the canalicular membrane, and if they do, to clarify whether the cytoprotective action of hydrophilic bile salts is associated with modulation of phosphatidylcholine composition in cell membrane bilayers. Bile salt-pool-depleted rats were infused intravenously with sodium taurocholate at a constant rate (200 nmol/min/100 g body wt) for 2 hr, followed by infusion of either sodium tauroursodeoxycholate, sodium tauroalphamuricholate, or sodium taurobetamuricholate (200 nmol/min/100 g) for 2 hr. Biliary outputs of cholesterol and phosphatidylcholine and phosphatidylcholine hydrophobicity in bile and subcellular fractions were determined. The cytoprotective action of hydrophilic bile salts was determined by the release of canalicular membrane-localizing enzymes (alkaline phosphatase, leucine aminopeptidase) into bile. Tauroursodeoxycholate, taurobetamuricholate, and tauroalphamuricholate decreased the release of these enzymes when compared to values under taurocholate infusion. Bile phosphatidylcholine hydrophobicity was also decreased by the bile salts, whereas the cholesterol/phosphatidylcholine ratio was increased. In contrast, phosphatidylcholine hydrophobicity in the canalicular membrane was increased by these three bile salts. In conclusion, hydrophilic bile salts promote biliary secretion of relatively hydrophilic phosphatidylcholine secretion into bile, and consequently phosphatidylcholine hydrophobicity in canalicular membranes increased. Such an alteration in phosphatidylcholine species within canalicular membrane enhances its lateral packing density with less fluidity, and this may account, in part, for the cytoprotective action of hydrophilic bile salts against hydrophobic bile salts. Chromosome 4A of wheat carries the Wx-B1 gene encoding the granule-bound starch synthase involved in amylose synthesis in the endosperm. To determine the pleiotropic effects of this locus and effects of independent QTLs on agronomic traits, genetical analysis of chromosome 4A was conducted using 98 single-chromosome recombinant substitution lines derived from a cross of Chinese Spring and Chinese Spring (Kanto107 4A) with a low amylose content due to the null Wx-B1b allele. For amylose content, most of the genetic variation was explained by the allelic difference at the Wx-B1 locus. An additional QTL of minor effect was mapped in the 6.2-cM Xbcd1738/Xcdo1387 interval on the short arm, where the allele from Kanto107 led to an increase in amylose content. Field trials over two seasons revealed a pleiotropic effect of Wx-B1, or else the effect of a closely linked QTL, on ear emergence time. A QTL linked to Wx-B1 was detected for plant height. For plant yield and its components, there was no evidence for significant main effects associated with Wx-B1 or adjacent regions. One plant-yield QTL was identified by RFLP markers on the short arm and this was identical to QTLs controlling spikelet number/ear and grain weight/ear. Chromosome 5A of wheat carries major gene loci for agronomic traits including the vernalization requirement ( Vrn-A1) and ear morphology ( Q). To determine whether the genetic variation for ear emergence time and plant height is attributable to either of these major genes as pleiotropic effects or independent QTL, we combined a RFLP map constructed from 120 recombinant substitution lines derived from a cross between ‘Chinese Spring’ (Cappelle-Desprez 5A) and CS( Triticum spelta 5A) with data collected from field trials over 3 years. For ear emergence time the main effects on flowering time were by Vrn-A1 and QEet.ocs-5A.1, the latter a QTL in the 28.6-cM Xcdo584/ Q interval linked to Q by less than 10 cM. The CS( T. spelta 5A) allele at QEet.ocs-5A.1 contributed to an earlier ear emergence time by 2.7–6.0 days, which was approximately equal to the effects of Vrn-A1. For plant height, three QTLs were identified on the long arm and linked in repulsion. The CS( T. spelta 5A) allele at Vrn-A1 or closely linked to Xfba068 contributed to a height reduction of 3.5–6.1 cm, whereas both the Q allele and Qt.ocs-5A.1 allele within the Xcdo1088/Xbcd9 interval from CS(Cappelle-Desprez 5A) produced a shorter plant. When plant height was partitioned into culm length and ear length, the Vrn-A1 allele and CS(Cappelle-Desprez 5A) allele at QCl.ocs-5A.1 within the Xcd1088/Xbcd9 interval were found to contribute to a shorter culm. Objective: To assess the mechanisms contributing to the induction of patellofemoral arthritis (PF-OA).Design and Patients: A computed tomography scan was taken at three levels of the lower extremity in full extension and at 30 degrees of flexion. The cuts were superimposed and 12 parameters were compared in 17 PF-OA knees and 27 normal knees to assess the rotation angle of the tibial tubercle.Results: Although the tibial tubercle was in almost the same position in full extension in the normal and PF-OA knees, it was positioned significantly laterally at 30 degrees of flexion in PF-OA knees. Also the articular surface of the lateral femoral condyle was significantly narrower or steeper in PF-OA knees.Conclusion: Anatomic variations and mechanical abnormalities were identified in the PF-OA knees. Bromosulfophthalein and papaverine have been demonstrated to inhibit biliary lipid secretion without affecting secretion of bile salts in normal rats, so-called uncoupling. Bromosulfophthalein inhibits the capacity of intracanalicular bile salt micelles to induce biliary lipid secretion, and papaverine inhibits vesicular transport within the hepatocyte. We compared the effects of bromosulfophthalein and papaverine on biliary lipid secretion in normal Sprague-Dawley rats and Eizai hyperbilirubinuria rats. The fatty acyl chain saturation in biliary lecithin increased during bromosulfophthalein infusion and decreased during papaverine infusion in Sprague-Dawley rats. Bromosulfophthalein had no effect on biliary lipid secretion in Eizai rats, while papaverine induced uncoupling. The degree of fatty acyl chain saturation in biliary lecithin was unchanged during bromosulfophthalein infusion, but decreased with papaverine in Eizai rats. We deduce that selection of biliary lecithin species occurs at various points in the lipid transport pathway at intracellular and intracanalicular sites. [ABSTRACT FROM AUTHOR]

As a stress model, a forced swimming test was applied to mice; and a typical behavioral change, an immobile posture, was recognized. This affected the brain monoamine levels significantly. The norepinephrine concentration was reduced, while that of its product was increased; and in the case of dopamine, both the amount of the amine and its product were increased. Stress increased the levels of serotonin and its product in the brain. The effects of RS-8359, (±)-4-(4-cyanophenyl)amino-6,7-dihydro-7-hydroxy-5H-cyclopenta[d]-pyrimidine, a new inhibitor of type A monoamine oxidase, on the behavioral and biochemical changes caused by forced swimming were also investigated. RS-8359 significantly improved the immobile posture elicited by the forced swimming test. It reduced the increased turnover of norepinephrine and serotonin systems caused by swimming. These results suggest that the effect of RS-8359 on behavioral and biochemical changes by stress may be mainly due to its effects on norepinephrine and serotonin systems, presumably by the inhibition of type A monoamine oxidase. Chromosome 5B of bread wheat is known to carry two major genes giving rise to genetic disorders, Ne1 for hybrid necrosis and Vg for winter variegation. Additionally, in many european winter wheat varieties this chromosome is represented in a translocated form, with 5BL-7BL, 5BL-7BS chromosomes rather than the normal 5B and 7B forms of the standard variety Chinese Spring. Genetic analysis has been carried out to map these genes and the translocation break point, and to investigate their pleiotropic effects or those of linked quantitative trait loci (qtl) for economically important characters. This was facilitated by the development of single chromosome recombinant lines between a normal and translocated karyotype, and growing these in field experiments over two seasons. There was differential segregation in favour of the translocated karyotype in the population of recombinant lines. Linkage analysis revealed that the two morphological markers and the isozyme locus Ibf-B1 were located on the long arm of 5B with a gene order of: breakpoint - Ne1 - Vg - Ibf-B1. Analysis of quantitative characters using these genes as landmarks showed pleiotropic effects of Ne1 or effects of tightly linked qtl on most of the quantitative characters related to grain yield. An additional qtl determining spikelet and grain number/ear appeared to be linked to the centromere. Effects on ear emergence time were associated with both Ne1 and Vg, and these interacted with environments. Similarly, effects on plant height were associated with Ne1 and Vg. A case is presented of a 64-year-old woman with pulseless disease associated with multiple intracranial aneurysms which were located in the internal carotid circulation at the left internal carotidophthalmic artery and the left internal carotid-posterior communicating junction, and in the basilarvertebral circulation, at the left basilo-anterior inferior cerebellar artery function and at the top of the basilar artery. Autopsy verified that her death was due to rupture of the aneurysm at the top of the basilar artery. From the cerebral angiographic and autopsy findings, it can be presumed that overloaded intracranial hemodynamic forces played some role in the development of the aneurysms. Amylose synthesis in wheat endosperm is mainly controlled by the granule-bound starch synthase of about 60 kDa, the so-called waxy (Wx) protein. The Wx proteins are the product of the Wx genes at a triplicate set of single-copy homoeoloci located on chromosomes 7A ( Wx-A1), 4A ( Wx-B1) and 7D ( Wx-D1). Using 'Chinese Spring' and its aneuploid lines, including nullisomic-tetrasomics, tetrasomics, ditelosomics and deletion stocks, together with single-chromosome substitution lines for these chromosomes, the effects of varying the dosage of whole chromosomes and chromosome arms, as well as the effects of null alleles, upon amylose synthesis were investigated. Nullisomic 4A and the deletion of chromosome segments carrying the Wx-B1 gene reduced the amylose content by more than 3%. A reasonable agreement was found in the substitution lines. This confirms that the absence of the Wx-B1 gene, or else substitution of this gene by its null allele, has the most striking effect on decreasing amylose synthesis. The removal of chromosomes carrying either the Wx-A1 or the Wx-D1 gene reduces the amylose content by less than 2%. A similar reduction was revealed by substitution of these two genes by the null alleles. Double dosages of chromosomes 7A, 4A and 7D did not increase amylose content, while the tetrasomic chromosomes produced more of the respective Wx proteins. The endosperm starch of the wheat grain is composed of amylose and amylopectin. Genetic manipulation of the ratio of amylose to amylopectin or the amylose content could bring about improved texture and quality of wheat flour. The chromosomal locations of genes affecting amylose content were investigated using a monosomic series of Chinese Spring (CS) and a set of Cheyenne (CNN) chromosome substitution lines in the CS genetic background. Trials over three seasons revealed that a decrease in amylose content occurred in monosomic 4A and an increase in monosomic 7B. Allelic variation between CS and CNN was suggested for the genes on chromosomes 4A and 7B. To examine the effects of three Waxy ( Wx) genes which encode a granule-bound starch synthase (Wx protein), the Wx proteins from CS monosomics of interest were analyzed using SDS-PAGE. The amount of the Wx protein coded by the Wx-B1 gene on chromosome arm 4AL was reduced in monosomic 4A, and thus accounted for its decreased amylose content. The amounts of two other Wx proteins coded by the Wx-A1 and Wx-D1 genes on chromosome arms 7AS and 7DS, respectively, showed low levels of protein in the monosomics but no effect on amylose content. For breeding to increase amylose content in the starch of hexaploid wheat, either the introduction of genes from Aegilops squarrosa (DD genome) or the development of high-amylose amphiploids between Triticum durum (AABB genome) and Ae. squarrosa is one of the most efficient ways. The high-amylose genes have not been detected in any species of Triticum and Aegilops. Therefore, we assessed the apparent amylose content in endosperm starch of T. durum and Ae. squarrosa accessions supplied from the major gene banks from world wide, and found several accessions with higher amylose content out of 665 T. durum accessions and out of 732 Ae. squarrosa accessions.
