Abstract The increasing use of artificial intelligence (AI) systems in our daily lives through various applications, services, and products highlights the significance of trust and distrust in AI from a user perspective. AI-driven systems have significantly diffused into various aspects of our lives, serving as beneficial “tools” used by human agents. These systems are also evolving to act as co-assistants or semi-agents in specific domains, potentially influencing human thought, decision-making, and agency. Trust and distrust in AI serve as regulators and could significantly control the level of this diffusion, as trust can increase, and distrust may reduce the rate of adoption of AI. Recently, a variety of studies focused on the different dimensions of trust and distrust in AI and its relevant considerations. In this systematic literature review, after conceptualizing trust in the current AI literature, we will investigate trust in different types of human–machine interaction and its impact on technology acceptance in different domains. Additionally, we propose a taxonomy of technical (i.e., safety, accuracy, robustness) and non-technical axiological (i.e., ethical, legal, and mixed) trustworthiness metrics, along with some trustworthy measurements. Moreover, we examine major trust-breakers in AI (e.g., autonomy and dignity threats) and trustmakers; and propose some future directions and probable solutions for the transition to a trustworthy AI.

Background: Prior studies have established that macroaggregated albumin (MAA)-SPECT/CT offers more robust lung shunt fraction (LSF) and lung mean absorbed dose (LMD) estimates in 90Y radioembolization in comparison to planar imaging. However, incomplete SPECT/CT coverage of the lungs is common due to clinical workflows, complicating its potential use for LSF and LMD calculations. In this work, lung truncation in MAA-SPECT/CT was addressed via correction strategies to improve 90Y treatment planning. Methods: Lung truncation was simulated in 56 cases with adequate (> 90%, mean: 98%) lung coverage in MAA-SPECT/CT by removing slices in ~ 5 mm increments from the lung apices to the diaphragm. A wide range of lung coverages from 100% to < 1% in ~ 2% increments were created. LSF and LMD were calculated with four methods. (1) 2D planar imaging standard (not truncated), truncated lung SPECT/CT data was: (2) used with no corrections (SPECTTrunc), (3) uniformly extrapolated to full lung coverage (SPECTUniform), (4) fit with an empirical model to predict lung counts at full lung coverage (SPECTFit). To determine counts for LSF, full lung volumes, those modified at the lung/liver boundary (Lungs 2-cm), and those isolated to the left lung (Left Lung) were used. The correction methods were then applied to 31 independent cases without full lung coverage (< 90%, mean: 74%). The variations in LSF and LMD estimates from each correction method were analyzed. Results: Averaged across simulated lung coverages from 40 to 80%, percent errors relative to non-truncated data for SPECTTrunc were (mean ± σ) − 22% ± 15% for LSF and 34% ± 29% for LMD. SPECTUniform had similar errors with 29% ± 26% for both LSF and LMD. SPECTFit yielded the most accurate and precise estimates for LSF and LMD, with errors of 11% ± 20% for both. The Left Lung approach equalized LMD errors in all three correction methods, with percent errors of 3% ± 17% (SPECTTrunc), 2% ± 17% (SPECTUniform), and 4% ± 13% (SPECTFit). In the 31 cases without ground truth LSF or LMD, Left Lung produced highly comparable LMD estimates, with a mean (max) coefficient of variation across the three correction methods of 4% (20%). Conclusion: LSF and LMD can be estimated for 90Y radioembolization using truncated lung coverage data in MAA-SPECT/CT. Empirical models to predict lung counts at full lung coverage produced LSF and LMD estimates with minimal bias and uncertainty. With lung/liver boundary adjustments, all SPECT/CT methods assessed in this work yielded LMD estimates comparable to ground truth, even down to 50% lung coverage. [ABSTRACT FROM AUTHOR]

Social justice has become a common goal in school psychology, including in practice guidelines. However, there is a paucity of research reflecting a representative sample of US practitioners providing input on key social justice competency areas and how frequently they employ these competencies. Utilizing a national sample of 145 practitioners, this study addresses this gap. Framed around the National Association of School Psychologists' Practice Model, respondents indicated a wide range of competency areas as important to the role of school psychologist, as well as widespread efforts to utilize these strategies, particularly actions that were more individualistic and under their direct control. Introduction: Black women's collective wisdom has informed Black women's sexual development for decades. Yet, empirical collective wisdom regarding what is needed to reduce their experiences of sexual pain and enhance sexual pleasure has been omitted. This study fills this gap by centering the voices of Black women living in the Southern region of the USA who have experienced sexual difficulties. Methods: We used thematic analysis to find themes across n = 21 in-depth interviews with Black women living in the Southern USA who consented to participate in a mixed methods study focused on Black women's experiences of sexual pain and pleasure. We interviewed participants between January and March 2021. Results: There were five main themes: 1) Talk that talk, 2) On my mama and big mama, 3) Take me to church, 4) Put yourself first, and 5) What about the men? Conclusions: Findings of this study reveal a common desire among Black women who have experienced sexual pain to have safe spaces to talk about sexual health through multigenerational sister circles and in churches. Heterosexual participants also stressed the importance of sexuality education opportunities for Black men. Policy Implications: It is important to understand the systems of oppression that present obstacles to Black women's sexual development and sexual health knowledge acquisition. Equitable funding for Black women's sexual health programs, and using multiplatform approaches to reach multiple generations and center the voices of Black women, are crucial to advancement. Over the last decades, a multitude of results in educational and psychological research have shown that the implementation of multiple external representations (MERs) in educational contexts represents a valuable tool for fostering learning and problem-solving skills. The context of science, technology, engineering, and mathematics (STEM) education has received great attention because it necessitates using various symbolic (e.g., text and formula) and graphical representations (e.g., pictures and graphs) to convey subject content. Research has mainly explored effects of combining two representations, but the potential benefits of integrating more than two representations on students’ learning remain underexplored. This gap limits our understanding of promising educational practices and restricts the development of effective teaching strategies catering to students’ cognitive needs. To close this gap, we conducted a systematic review of 46 studies and a meta-analysis that included 132 effect sizes to evaluate the effectiveness of using more than two representations in STEM education and to identify moderating factors influencing learning and problem-solving. A network diagram analysis revealed that the advantages of learning and problem-solving with MERs are also applicable to more than two representations. A subsequent meta-analysis revealed that the learning with more than two representations in STEM can have advantageous effects on students cognitive load ( Hedges ′ g = 0.324 , p <. 001 , 95 % CI [ 0.164 , 0.484 ] ) and performance ( Hedges ′ g = 0.118 , p <. 001 , 95 % CI [ 0.050 , 0.185 ] ) compared to learning with two representations without notable differences in learning time. The pervasive digitization of society underscores the crucial role of data and its significant impact on decision-making across various domains. As a result, it is essential for individuals to acquire competencies in handling data. This need is particularly pertinent in K-12 education, where early engagement with data and statistics can lay a foundational understanding for future academic and professional endeavors. Additionally, K-12 education should provide students with critical skills necessary for navigating the complexities of daily life and making informed decisions in a data-rich society. This systematic review examines the state of research on statistical and data literacy in K-12 STEM (Science, Technology, Engineering, and Mathematics) education. It focuses specifically on cognitive, affective, and behavioral metrics and pedagogical approaches empirically investigated in this context. Using a rigorous selection process, we identified and synthesized 83 original empirical papers. Additionally, we invited the authors of these studies to share their perspectives on future strategies for addressing statistical and data literacy. The results indicate that the included studies primarily focus on the construct of statistical literacy, which is operationalized through a diverse array of metrics, predominantly within the context of mathematics education. We identified effective pedagogical approaches, such as authentic problem-solving and the integration of real-world data. The researchers surveyed emphasized the importance of interdisciplinary teaching, adapted curricula, and improved professional development for pre- and in-service teachers. Our findings underscore the growing relevance of this field, but suggest that integrated perspectives on statistical and data literacy within STEM subjects are limited. [ABSTRACT FROM AUTHOR]

Phenotypic plasticity allows a plant cell to alter its structure and function in response to external pressure. This adaptive phenomenon has also been important in the evolution of plants including the emergence of land plants from a streptophyte alga. Penium margaritaceum is a unicellular zygnematophyte (i.e., the group of streptophyte algae that is sister to land plants) that was employed in order to study phenotypic plasticity with a focus on the role of subcellular expansion centers and the cell wall in this process. Live cell fluorescence labeling, immunofluorescence labeling, transmission electron microscopy, and scanning electron microscopy showed significant subcellular changes and alterations to the cell wall. When treated with the actin-perturbing agent, cytochalasin E, cytokinesis is arrested and cells are transformed into pseudo-filaments made of up to eight or more cellular units. When treated with the cyclin-dependent kinase (CDK) inhibitor, roscovitine, cells converted to a unique phenotype with a narrow isthmus zone. [ABSTRACT FROM AUTHOR]

Materials based on iron oxide were prepared via a closed system solution combustion synthesis (IOP), evaluating the effect of adding mango seed husk (T), peel (C), and a mixture of the portions (M) on the material properties obtained (IOT, IOC, and IOM, respectively). The structural (XRD, Mössbauer), morphological (SEM and TEM), optical (UV-VIS diffuse reflectance), textural (BET), and magnetic (VSM) properties of the materials obtained were evaluated. Different phase compositions (hematite, magnetite, and maghemite) were identified in the structure of the materials. Adding mango residue favored the formation of magnetite with superparamagnetic particles in the IOC sample. The solids were evaluated in the methylene blue photocatalysis at different concentrations, observing different effects on the discoloration rate. The IOP sample performed better at the lowest concentration (50 mg L−1) of the dye, with 98% discoloration after 90 min and a specific speed of 44.7 × 10−3 min−1, which can be explained by the heterojunction effect, making electronic recombination harder and increasing the quantum yield of the photocatalyst. At 70 mg L−1 concentration, the best catalyst was the IOM with 82% discoloration and a specific speed of 15.8 × 10−3 min−1, explained by the increase in the specific area due to the insertion of biomass, which improved adsorptive capacity, maintaining the observed heterojunction effects. Under sunlight, the best composite was IOC, with 90.5% discoloration and activity of 29.8 × 10−3 min−1, justified by combining favorable textural and optical properties. [ABSTRACT FROM AUTHOR]

Background: The approved dose of Selinexor, 60 mg twice-weekly, is associated with several clinically relevant toxicities. Preclinical studies show that a sustained-release formulation of selinexor results in a lower toxicity profile. Objective: The phase 1b METSSAR trial assessed the safety and tolerability of an alternative dosing schedule of selinexor (to mimic the sustained-release formulation) in advanced soft tissue sarcoma (STS) patients. Patients and methods: Selinexor was administered in a split-dose schedule (40 mg, 20 mg, 20 mg in the morning, afternoon, and evening, respectively) on days 1, 8, 15, and 22 of a 28-day cycle, until unacceptable toxicity or disease progression. The primary endpoint was the rate of grade ≥ 3 treatment-related adverse events (TRAEs). Secondary objectives were EORTC QLQ-C30 quality of life (QoL) assessment, and preliminary efficacy. Results: Twenty patients with 12 STS subtypes were enrolled and received a median of four cycles of treatment. There were no grade ≥ 3 TRAEs. Dysgeusia, nausea, fatigue, and thrombocytopenia were the most common grade ≤ 2 TRAEs. No treatments were discontinued due to TRAE, but four patients (20%) required dose reduction. Median change in global health status (GHS) score from baseline to cycle 2 (by QLQ-C30 v3.0) was – 8.33, and only 39% of patients reported a clinically meaningful decline in GHS score (≥ 10 points). Median symptom scale scores on treatment were increased for fatigue (+12.35), nausea/vomiting (+18.52), and anorexia (+16.67), but reduced for pain (– 3.70). The median progression-free survival (PFS) was 4.0 months (95% confidence interval 1.9–7.5). Conclusions: Split-dose once-weekly selinexor was reasonably well tolerated in this heterogeneous group of advanced STS patients with a better, or at least similar, clinician- and patient-reported toxicity profile compared to the standard dosing regimen. Genes encoding subunits of SWI/SNF (BAF) chromatin remodeling complexes are mutated in nearly 25% of cancers. To gain insight into the mechanisms by which SWI/SNF mutations drive cancer, we contributed ten rhabdoid tumor (RT) cell lines mutant for SWI/SNF subunit SMARCB1 to a genome-scale CRISPR–Cas9 depletion screen performed across 896 cell lines. We identify PHF6 as specifically essential for RT cell survival and demonstrate that dependency on Phf6 extends to Smarcb1-deficient cancers in vivo. As mutations in either SWI/SNF or PHF6 can cause the neurodevelopmental disorder Coffin-Siris syndrome, our findings of a dependency suggest a previously unrecognized functional link. We demonstrate that PHF6 co-localizes with SWI/SNF complexes at promoters, where it is essential for maintenance of an active chromatin state. We show that in the absence of SMARCB1, PHF6 loss disrupts the recruitment and stability of residual SWI/SNF complex members, collectively resulting in the loss of active chromatin at promoters and stalling of RNA Polymerase II progression. Our work establishes a mechanistic basis for the shared syndromic features of SWI/SNF and PHF6 mutations in CSS and the basis for selective dependency on PHF6 in SMARCB1-mutant cancers. Here, the authors identify PHF6 as a dependency in SMARCB1-deficient rhabdoid cancers. Production of hard X-ray via inverse Compton scattering at photon energies below 100 keV range aimed at potential applications in medicine and material research is reported. Experiments have been performed at the Brookhaven National Laboratory, Accelerator Test Facility, employing the counter collision of a 70 MeV, 0.3 nC electron beam with a near infra-red Nd: YAG laser (1064 nm wavelength) pulse containing ~ 100 mJ in a single shot basis. Human immunodeficiency virus-associated neurocognitive disorders persist in the combination antiretroviral therapy era. CD4 nadir is a well-established predictor of cognition cross-sectionally, but its impact on longitudinal neurocognitive (NC) trajectories is unclear. The few studies on this topic examined trajectories of global cognition, rather than specific NC domains. The current study examined CD4 nadir in relation to domain-specific NC decline. 132 HIV + adults from the Temple/Drexel Comprehensive NeuroHIV Center, Clinical and Translational Research Support Core Cohort were administered comprehensive NC assessments longitudinally, with last visit occurring an average of 12 years after CD4 nadir. Linear mixed models were used to examine CD4 nadir in relation to longitudinal NC trajectories in three empirically identified NC domains: speed/executive function (S/EF), visuospatial memory (VM), and verbal fluency (VF). CD4 nadir was associated with change in VF (p = 0.020), but not with S/EF or VM. Specifically, those with CD4 nadir < 200 demonstrated increasing VF over time (p =.002), whereas those with CD4 nadir > 200 demonstrated stable VF (p =.568), though these differing trajectories may partly reflect regression to the mean or differential practice effect. CD4 dynamics over time were analyzed as potential mechanisms for the identified associations, with mixed findings. While low CD4 nadir has been associated with weaker neurocognition among people living with HIV, the results of this study suggest that low CD4 nadir is not associated with ongoing decline a decade later. Nadir-related deficits in VF may be stable or even improve over time, possibly reflecting the beneficial cognitive effects of long-term treatment and immune reconstitution. [ABSTRACT FROM AUTHOR]

G-quadruplexes (G4s) formed by guanine-rich nucleic acids induce genome instability through impeding DNA replication fork progression. G4s are stable DNA structures, the unfolding of which require the functions of DNA helicases. Pif1 helicase binds preferentially to G4 DNA and plays multiple roles in maintaining genome stability, but the mechanism by which Pif1 unfolds G4s is poorly understood. Here we report the co-crystal structure of Saccharomyces cerevisiae Pif1 (ScPif1) bound to a G4 DNA with a 5′ single-stranded DNA (ssDNA) segment. Unlike the Thermus oshimai Pif1-G4 structure, in which the 1B and 2B domains confer G4 recognition, ScPif1 recognizes G4 mainly through the wedge region in the 1A domain that contacts the 5′ most G-tetrad directly. A conserved Arg residue in the wedge is required for Okazaki fragment processing but not for mitochondrial function or for suppression of gross chromosomal rearrangements. Multiple substitutions at this position have similar effects on resolution of DNA duplexes and G4s, suggesting that ScPif1 may use the same wedge to unwind G4 and dsDNA. Our results reveal the mechanism governing dsDNA unwinding and G4 unfolding by ScPif1 helicase that can potentially be generalized to other eukaryotic Pif1 helicases and beyond. Emergent marsh and open water have been identified as alternate stable states in tidal marshes with large, relative differences in hydrogeomorphic conditions. In the Florida coastal Everglades, concern has been raised regarding the loss of non-tidal, coastal peat marsh via dieback of emergent vegetation and peat collapse. To aid in the identification of alternate stable states, our objective was to characterize the variability of hydrogeomorphic and biologic conditions using a field survey and long-term monitoring of hydrologic and geomorphic conditions across a range of vegetated (emergent, submerged) and unvegetated (open water) communities, which we refer to as "ecosystem states," in a non-tidal, brackish peat marsh of the coastal Everglades. Results show (1) linear relationships among field-surveyed geomorphic, hydrologic, and biologic variables, with a 35-cm mean difference in soil surface elevation between emergent and open water states, (2) an overall decline in soil elevation in the submerged state that was related to cumulative dry days, and (3) a 2× increase in porewater salinity during the dry season in the emergent state that was also related to the number of dry days. Coupled with findings from previous experiments, we propose a conceptual model that describes how seasonal hydrologic variability may lead to ecosystem state transitions between emergent and open water alternate states. Since vegetative states are only moderately salt tolerant, as sea-level rise pushes the saltwater front inland, the importance of continued progress on Everglades restoration projects, with an aim to increase the volume of freshwater being delivered to coastal wetlands, is the primary management intervention available to mitigate salinization and slow ecosystem state shifts in non-tidal, brackish peat marshes. Objective Structured Clinical Examinations (OSCEs) and Work Based Assessments (WBAs) are the mainstays of assessing clinical competency in health professions' education. Underpinned by the extrapolation inference in Kane's Validity Framework, the purpose of this study is to determine whether OSCEs translate to real life performance by comparing students' OSCE performance to their performance in real-life (as a WBA) using the same clinical scenario, and to understand factors that affect students' performance. A sequential explanatory mixed methods approach where a grade comparison between students' performance in their OSCE and WBA was performed. Students were third year pharmacy undergraduates on placement at a community pharmacy in 2022. The WBA was conducted by a simulated patient, unbeknownst to students and indistinguishable from a genuine patient, visiting the pharmacy asking for health advice. The simulated patient was referred to as a 'mystery shopper' and the process to 'mystery shopping' in this manuscript. Community pharmacy is an ideal setting for real-time observation and mystery shopping as staff can be accessed without appointment. The students' provision of care and clinical knowledge was assessed by the mystery shopper using the same clinical checklist the student was assessed from in the OSCE. Students who had the WBA conducted were then invited to participate in semi-structured interviews to discuss their experiences in both settings. Overall, 92 mystery shopper (WBA) visits with students were conducted and 36 follow-up interviews were completed. The median WBA score was 41.7% [IQR 28.3] and significantly lower compared to the OSCE score 80.9% [IQR 19.0] in all participants (p < 0.001). Interviews revealed students knew they did not perform as well in the WBA compared to their OSCE, but reflected that they still need OSCEs to prepare them to manage real-life patients. Many students related their performance to how they perceived their role in OSCEs versus WBAs, and that OSCEs allowed them more autonomy to manage the patient as opposed to an unfamiliar workplace. Purpose: The COVID-19 pandemic has caused great strain on older adults with cancer and their healthcare providers. This study explored healthcare providers' reported changes in cancer care, clinical barriers to care, patient questions, and the overall experiences of caring for older adults with cancer during the COVID-19 crisis. Methods: The Advocacy Committee of the Cancer and Aging Research Group and the Association of Community Cancer Centers developed a survey for healthcare providers of adults with cancer, inquiring about their experiences during the pandemic. Responses from the survey's four open-ended items were analyzed by four independent coders for identification of common themes using deductive and inductive methods. Results: Participants (n = 137) represented a variety of demographic and clinical experiences. Six overall themes emerged, including (1) telehealth use, (2) concerns for patient mental health, (3) patient physical and social isolation, (4) patient fear of contracting COVID-19, (5) continued disruptions to cancer care, and (6) patients seeking guidance, particularly regarding COVID-19 vaccination. Questions fielded by providers focused on the COVID-19 vaccination's safety and efficacy during older adults' cancer treatment. Conclusions: Additional resources (e.g., technology support, established care guidelines, and sufficient staffing) are needed to support older adults with cancer and healthcare providers during the pandemic. Future research should explore universally effective in-person and virtual treatment strategies for older adults with cancer. Implications for Cancer Survivors: Persistence of telehealth barriers, particularly a lack of infrastructure to support telehealth visits, social isolation, and restrictive visitor policies as a result of COVID-19, negatively impacted the mental health of older adults with cancer. Introduction: Studies have reported health-related quality-of-life impacts of Duchenne muscular dystrophy (DMD); however, further research is needed to understand how those with DMD experience their condition and how psychosocial impacts evolve over time in response to disease progression. This qualitative study explores the social and emotional implications of key transitions, challenges and adaptations throughout the disease course from the perspective of patients and family caregivers. Methods: Semi-structured interviews were conducted with men and boys with DMD, and/or their caregivers, in the USA. Thematic analysis was used to examine patterns in data collected across the interviews. Results: Nineteen participants were included. Three major themes were identified: (1) barriers to participation are multifaceted; (2) an emotional journey shaped by 'inevitable progression;' (3) family provides critical tangible and emotional support. This study illustrates that psychosocial impacts of DMD are shaped by knowledge of the condition's natural history alongside other factors including the extent of social barriers, personal growth and adaptation, and family support. Conclusions: Findings provide insight into the strength and resilience with which individuals and their families respond to daily challenges and major clinical milestones and highlight the relative importance of loss of upper limb function as a transition in DMD affecting health-related quality-of-life. Microplastics and nanoplastics (collectively, MNPs) are increasingly entering soils, with potential adverse impacts to agriculture and groundwater. Environmental detection, characterization, and quantification of MNPs is difficult and subject to artifacts, often requiring labor-intensive separation from environmental matrices. These analytical challenges make it difficult to conduct experiments investigating specific MNP characteristics influencing their transport and fate, particularly when examining multiple plastic types at low concentrations. By synthesizing a suite of metal-tagged polymers, which are cryomilled to create polydisperse fragmented particle suspensions, single particle ICP-MS (spICP-MS) can be used to quantify MNP particle size and concentration in controlled fate and transport studies. Use of unique metal-polymer pairs enables accurate, simultaneous analysis of multiple MNP types which can be used to track total particle transport and retention within a variety of environmental matrices. This was demonstrated using saturated sand column transport experiments to quantify the movement of two plastics having different properties: tin-tagged polystyrene (Sn-PS) and tantalum-tagged polyvinylpyrrolidone (Ta-PVP). The behavior of these polydisperse, fragmented MNPs was compared to that of fluorescent, carboxylated monodisperse PS spherical microspheres (Fl-PS). Mobility of all MNP types increased with decreasing particle size, and hydrophilic Ta-PVP particles migrated more effectively than the hydrophobic Sn-PS particles. Furthermore, the addition of humic acid (HA) to the carrier solution increased the colloidal stability of both metal-tagged MNP suspensions, resulting in much greater elution from the column than in HA-free deionized water or moderately- hard water (ionic strength = 5mM). This combination of particle synthesis and spICP-MS analysis provides insights into the transport of MNP having physical properties that are representative of environmental MNPs and opens up a broad range of applications for study of MNP environmental fate and transport. Objectives: Previous research has shown the capacity for mindfulness to be strongly associated with psychological well-being, that components of mindfulness show significant growth through young adulthood, and that this developing, malleable capacity is vital as individuals learn to deal appropriately with negative thoughts and unwelcome emotions. Smartphones, typically used in an automatic or experientially avoidant way, can undermine this development, leading to a decreased capacity for mindfulness. The purpose of these studies were to examine the extent to which smartphone use is negatively associated with young adults' mindfulness and the degree to which increased cognitive and behavioral involvement with smartphones may exacerbate this relation using a newly developed conceptual model. Method: Study 1 was conducted using self-report measures of mindfulness among a cross-sectional sample of university students aged 18–20 years (n = 668). Study 2 augmented Study 1 using objective measures of smartphone screen time and the cognitive regulatory components of mindfulness in a planned missingness design. Results: Results indicate smartphone involvement (a compulsive pattern of use and cognitive preoccupation with one's smartphone) to be significantly associated with lower trait mindfulness. Additionally, exploratory analysis of smartphone involvement as a mediator of the effect of smartphone use on mindfulness demonstrated a significant estimated indirect effect. Conclusion: These results provide preliminary empirical support for the newly proposed conceptual model which posits associations between mindfulness and the use of smartphones in a cognitively and behaviorally involved way. Renal cell carcinoma (RCC) is an uncommon malignancy whose incidence has been increasing over the past few decades, posing treatment challenges for elderly or infirm patients who are not surgical candidates. Stereotactic ablative radiotherapy (SABR) has emerged as a promising non-invasive treatment modality for RCC. The high dose-per-fraction used in SABR overcomes some of the mechanisms of radioresistance that has hindered the effective treatment of RCC with conventional radiotherapy. For primary RCC, local control rates for SABR exceed 90%, with typically minimal grade 3 or higher toxicities, offering a viable alternative for inoperable patients and those not eligible for or unable to tolerate radiofrequency or cryotherapy ablation. SABR can also be used in patients with a solitary kidney as a strategy for renal preservation to avoid need for dialysis. Given its excellent local control rates, low toxicity and preservation of renal function, SABR offers an attractive alternative to more invasive modalities for treatment of localized RCC. Purpose: After cessation of androgen deprivation therapy (ADT), testosterone gradually recovers to supracastrate levels (> 50 ng/dL). After this, rises in prostate-specific antigen (PSA) are often seen. However, it remains unknown whether early PSA kinetics after testosterone recovery are associated with subsequent biochemical recurrence (BCR). Methods: We performed a secondary analysis of a phase III randomized controlled trial in which newly diagnosed localized prostate cancer patients were randomly allocated to ADT for 6 months starting 4 months prior to or simultaneously with prostate RT. We calculated the PSA doubling time (PSADT) based on PSA values up to 18 months after supracastrate testosterone recovery. Competing risk regression was used to evaluate the association of PSADT with relative incidence of BCR, considering deaths as competing events. Results: Overall, 313 patients were eligible. Median PSADT was 8 months. Cumulative incidence of BCR at 10 years from supracastrate testosterone recovery was 19% and 11% in patients with PSADT < 8 months and ≥ 8 months (p = 0.03). Compared to patients with PSADT of < 4 months, patients with higher PSADT (sHR for PSADT 4 to < 8 months: 0.36 [95% CI 0.16–0.82]; 8 to < 12 months: 0.26 [0.08–0.91]; ≥ 12 months: 0.20 [0.07–0.56]) had lower risk of relative incidence of BCR. Conclusions: Early PSA kinetics, within 18 months of recovery of testosterone to a supracastrate level, can predict for subsequent BCR. Taking account of early changes in PSA after testosterone recovery may allow for recognition of potential failures earlier in the disease course and thereby permit superior personalization of treatment. Introduction: Neoadjuvant therapy is an essential component of multimodality therapy for locally advanced breast adenocarcinoma (BC). Complete pathologic response (pCR) is a useful surrogate for long-term oncologic outcome. Aim: To assess the association between clinicopathologic, molecular and immunological markers and treatment response to neoadjuvant therapy in BC. Methods: BC patients undergoing neoadjuvant therapy were identified from a prospectively maintained institutional database. Serum haematological/biochemical values, histopathologic, immunohistochemical data and TNM stage were obtained from electronic records. Patients were categorised into complete responders vs non-complete responders and responders vs non-responders. Statistical analysis was performed via SPSS. Results: Overall, 299 BC patients were included. The average age was 49.8 ± 11.5 years. A pCR was evident in 22.6% (n = 69). pCR was associated with early T stage and non-luminal subtypes (HER2 enriched [HER2 +] and triple negative [TNBC]). The neutrophil–lymphocyte ratio (NLR) pre-operatively was lower in patients with a pCR (p = 0.02). The lymphocyte-CRP ratio (LCR) was also slightly reduced in responders (p = 0.049) at diagnosis. A pre-op NLR greater than 2 was not found to be a significant predictive factor (p = 0.071) on multivariable logistic regression analysis. T stage at diagnosis (p = 0.024), N stage (p = 0.001) and breast cancer subtype (p = 0.0001) were also determined to be significant predictive factors of complete response. Conclusion: pCR was more likely in patients with less advanced disease in BC. The presence of HER2 + or TNBC in BC also increases the likelihood of pCR. Neoadjuvant therapy stimulates the systemic inflammatory response; however, a reduced baseline NLR may be associated with increased pCR. Confirmation with larger datasets is required. The medial entorhinal cortex (MEC) is hypothesized to function as a cognitive map for memory-guided navigation. How this map develops during learning and influences memory remains unclear. By imaging MEC calcium dynamics while mice successfully learned a novel virtual environment over ten days, we discovered that the dynamics gradually became more spatially consistent and then stabilized. Additionally, grid cells in the MEC not only exhibited improved spatial tuning consistency, but also maintained stable phase relationships, suggesting a network mechanism involving synaptic plasticity and rigid recurrent connectivity to shape grid cell activity during learning. Increased c-Fos expression in the MEC in novel environments further supports the induction of synaptic plasticity. Unsuccessful learning lacked these activity features, indicating that a consistent map is specific for effective spatial memory. Finally, optogenetically disrupting spatial consistency of the map impaired memory-guided navigation in a well-learned environment. Thus, we demonstrate that the establishment of a spatially consistent MEC map across learning both correlates with, and is necessary for, successful spatial memory. The selection of an inhaler device is a key component of respiratory disease management. However, there is a lack of clarity surrounding inhaler resistance and how it impacts inhaler selection. The most common inhaler types are dry powder inhalers (DPIs) that have internal resistance and pressurised metered dose inhalers (pMDIs) that use propellants to deliver the drug dose to the airways. Inhaler resistance varies across the DPIs available on the market, depending largely on the design geometry of the device but also partially on formulation parameters. Factors influencing inhaler choice include measures such as flow rate or pressure drop as well as inhaler technique and patient preference, both of which can lead to improved adherence and outcomes. For optimal disease outcomes, device selection should be individualised, inhaler technique optimised and patient preference considered. By addressing the common clinically relevant questions, this paper aims to demystify how DPI resistance should guide the selection of the right device for the right patient. Statistical and data literacy have emerged as key competencies in the 21st-century. Science, technology, engineering, and mathematics (STEM) curricula have been adapted in various countries to place increased emphasis on proficient data use and statistical comprehension. Consequently, there is a need for an adequate education and professional training of STEM teachers. This systematic review analyzes and synthesizes research on teachers' statistical and data literacy in K-12 STEM education. A total of 42 articles were analyzed, with a specific focus on teacher variables, teacher education, and their impact on classroom practice. The findings emphasize the recent emergence of research in this domain, particularly focusing on statistical literacy, with mathematics being the dominant STEM domain explored. Most studies examined cognitive variables of both pre-service and in-service teachers, revealing relevant gaps in knowledge and skills related to statistical and data literacy. Limited attention was given to teachers' affective variables. Research on the complex relationships between teacher variables, their teaching practices, and their students' learning is scarce, necessitating further investigation. Several pedagogical approaches to prepare STEM teachers for promoting statistical and data literacy in their classrooms were identified. Positive impacts on teachers' cognitive and affective variables were observed, but longitudinal effects and transfer to classroom practice require further inquiry. As curricula worldwide increasingly emphasize statistical and data literacy, this review underscores the necessity of enhancing teachers' competencies, advocates for tailored pedagogical approaches, and emphasizes the need for further research to shape effective instruction in K-12 STEM education. Peripheral vascular diseases (PVDs) represent a significant burden on global human health and healthcare systems. With continued growth in obesity and diabetes, it is likely that the incidence of these conditions will increase. As many PVDs remain undiagnosed, low-cost and easy to use diagnostic methods are required. This work uses newly developed wearable electro-resistive morphic sensors to assess venous and arterial competence in the lower limbs of 36 healthy subjects. Comparison of this HeMo device was made to currently available benchtop light reflection rheography and photoplethymography devices. Results indicate that HeMo can detect the physiological signals of interest for both chronic venous insufficiency and peripheral arterial disease and all subjects were interpreted as healthy by each system. However, measurement repeatability of HeMo was highlighted as an issue that requires further system development. Furthermore, as HeMo captures changes in a section of limb circumference due to changes in underlying blood movement, rather than at a single point, the recorded signal is typically damped by comparison. To understand patterns in CO2 partial pressure (PCO2) over time in wetlands' surface water and porewater, we examined the relationship between PCO2 and land–atmosphere flux of CO2 at the ecosystem scale at 22 Northern Hemisphere wetland sites synthesized through an open call. Sites spanned 6 major wetland types (tidal, alpine, fen, bog, marsh, and prairie pothole/karst), 7 Köppen climates, and 16 different years. Ecosystem respiration (Reco) and gross primary production (GPP), components of vertical CO2 flux, were compared to PCO2, a component of lateral CO2 flux, to determine if photosynthetic rates and soil respiration consistently influence wetland surface and porewater CO2 concentrations across wetlands. Similar to drivers of primary productivity at the ecosystem scale, PCO2 was strongly positively correlated with air temperature (Tair) at most sites. Monthly average PCO2 tended to peak towards the middle of the year and was more strongly related to Reco than GPP. Our results suggest Reco may be related to biologically driven PCO2 in wetlands, but the relationship is site-specific and could be an artifact of differently timed seasonal cycles or other factors. Higher levels of discharge do not consistently alter the relationship between Reco and temperature normalized PCO2. Our understanding of the complex pathophysiology of Heart failure with preserved ejection fraction (HFpEF) is limited by the lack of a robust in vivo model. Existing in-vivo models attempt to reproduce the four main phenotypes of HFpEF; ageing, obesity, diabetes mellitus and hypertension. To date, there is no in vivo model that represents all the haemodynamic characteristics of HFpEF, and only a few have proven to be reliable for the preclinical evaluation of potentially new therapeutic targets. HFpEF accounts for 50% of all the heart failure cases and its incidence is on the rise, posing a huge economic burden on the health system. Patients with HFpEF have limited therapeutic options available. The inadequate effectiveness of current pharmaceutical therapeutics for HFpEF has prompted the development of device-based treatments that target the hemodynamic changes to reduce the symptoms of HFpEF. However, despite the potential of device-based solutions to treat HFpEF, most of these therapies are still in the developmental stage and a relevant HFpEF in vivo model will surely expedite their development process. This review article outlines the major limitations of the current large in-vivo models in use while discussing how these designs have helped in the development of therapy devices for the treatment of HFpEF. Background: The progression of Duchenne muscular dystrophy (DMD) is characterized by loss of ambulation, respiratory insufficiency, cardiomyopathy, and early mortality. DMD profoundly impacts health-related quality-of-life (HRQoL). However, few health state utility data exist; published utilities tend to be derived from small samples for a limited number of health states and are often based on caregiver-reported patient health status. This study estimated utility values for varied clinical and functional health states in DMD, based on patient-reported health status. Methods: Individuals with DMD in the US aged 12–40 years completed the EQ-5D (5-level) and Health Utilities Index (HUI) preference-based instruments. Based on responses to a clinical questionnaire, participants self-classified into functional health states according to level of lower and upper limb function, use of respiratory support, and presence of cardiomyopathy. Mean [standard deviation (SD)] utility and EQ-5D visual analogue scale (VAS) scores were estimated according to health state; and median (interquartile range) attribute levels calculated to understand which domains of health are most severely affected in DMD. Results: Of 63 males with DMD, mean (SD) age was 19.8 (6.1) years and 11 (17.5%) were ambulatory. Mean (SD) utility values were 0.92 (0.08; HUI2), 0.84 (0.20; HUI3), and 0.84 (0.13; EQ-5D) for ambulatory patients without cardiomyopathy (n = 10). For non-ambulatory patients with moderately impaired upper limb function, night and daytime ventilation without cardiomyopathy, mean (SD) utilities were 0.49 (0.07) for the HUI2, 0.16 (0.15) for the HUI3 and 025 (0.14) for the EQ-5D. Mean (SD) VAS scores for the same health states were 91 (9) and 83 (21), respectively. In addition to impairments in mobility/ambulation, and self-care, attributes like usual activities and pain also showed notable effects of DMD. Conclusions: In DMD, although a relationship between disease progression and HRQoL is observed, there is large variability in utility within functional health states, and across instruments. Utility values for less severe non-ambulatory health states described by level of upper limb function are novel. These utility values, derived based on direct patient feedback rather than from caregiver report, are relevant to individuals of varying functional statuses and augment scarce DMD-specific utility data. Nonlinear acoustic methods demonstrate high sensitivity for concrete damage evaluation. Among these methods, nonlinear resonance acoustic methods have been widely used in laboratory tests on small scale test samples. In this study, a nonlinear impact-echo (IE) method for concrete damage evaluation was introduced, in which the IE frequency shift was measured at different impact force levels. The nonlinear IE test and the nonlinear impact resonance acoustic spectroscopy (NIRAS) test share similarities in the experimental setup and analysis method. However, the nonlinear IE test excites a local thickness resonance mode of a member instead of the global resonance vibration. Therefore, the analyzed mode is unaffected by member boundary conditions and is applicable to large concrete structural members. To identify the fundamental IE frequencies, multiple impacts were applied along the members. Once the fundamental IE frequencies were determined, the effectiveness of the nonlinear IE method was evaluated by testing seven concrete beam specimens with varying levels of alkali-silica reaction (ASR) damage. Purpose: The synergy of combining androgen receptor-signaling inhibition (ARSI) to radiotherapy (RT) in prostate cancer has been largely attributed to non-homologous end joining (NHEJ) inhibition. However, this mechanism is unlikely to explain recently observed trial results that demonstrated the sequencing of ARSI and RT significantly impacts clinical outcomes, with adjuvant ARSI following RT yielding superior outcomes to neoadjuvant/concurrent therapy. We hypothesized this is driven by differential effects on AR-signaling and alternative DNA repair pathway engagement based on ARSI/RT sequencing. Methods: We explored the effects of ARSI sequencing with RT (neoadjuvant vs concurrent vs adjuvant) in multiple prostate cancer cell lines using androgen-deprived media and validation with the anti-androgen enzalutamide. The effects of ARSI sequencing were measured with clonogenic assays, AR-target gene transcription and translation quantification, cell cycle analysis, DNA damage and repair assays, and xenograft animal validation studies. Results: Adjuvant ARSI after RT was significantly more effective at killing colony forming cells and decreasing the transcription and translation of downstream AR-target genes across all prostate cancer models evaluated. These results were reproduced in xenograft studies. The differential effects of ARSI sequencing were not fully explained by NHEJ inhibition alone, but by the additional disruption of homologous recombination specifically with adjuvant sequencing of ARSI. Conclusion: We demonstrate that altered sequencing of ARSI and RT mediates differential anti-AR-signaling and anti-cancer effects, with the greatest benefit from adjuvant ARSI following RT. This paper explores how schoolteachers in Kazakhstan engaged with the Renewed Content of Education (RCE) that has been introduced by the Government, and how changes in their beliefs and understandings influenced classroom practice. The study draws on the ecological model of teacher agency and elaborates on factors that contribute to the formation of teacher agency. The study used a mixed methods research design and is based on data collected over two years in rural and urban schools across three regions of Kazakhstan. Altogether, 227 teachers having different levels of experience with the new curriculum were involved in focus group discussions. The findings demonstrate that the majority of teachers acknowledged the value of the RCE, its short- and long-term benefits for students, and the broader aim of boosting the economic competitiveness of the country. At the same time, the findings suggest that, while a surface change occurred in teachers' beliefs, their pedagogical practices, and the learning context, there is limited evidence that the teachers moved fully to new ways of teaching and embedded the principles of the RCE in practice. Through our findings, we verified the centrality of socially dynamic relationships in educational change. Teachers shared agency in developing their own rules and routines for collaboration. This paper adds to research on educational change in an international context by showing that the scope for teacher agency in reform implementation increases when teachers are able to develop deep reform-oriented beliefs, discourses, and pedagogical understanding. According to Fitts' Law, the time to reach a target (movement time, MT) increases with distance. A violation of Fitts' Law occurs when target positions are outlined before and during movement, as MTs are not different when reaching to the farthest and penultimate targets. One hypothesis posits that performers cognitively process the edges of a target array before the center, allowing for corrective movements to be completed more quickly when moving to edge targets compared to middle targets. The objective of this study was to test this hypothesis by displaying a target range rather than outlines of individual targets in an effort to identify the effects of array edges. Using a touch-screen laptop, participants (N = 24) were asked to reach to one of three targets which would appear within a presented range. Separately, targets were also presented without a range to determine if the display protocol could evoke Fitts' Law. Movements were assessed with the touch screen and optical position measurement. A main effect was found for relative position within a range (touch: F2,44 = 15.4, p < 0.001, η2p = 0.412; position: F2,40 = 15.6, p < 0.001, η2p = 0.439). As hypothesised, MT to the farthest target in a range was not significantly different than MT to the middle target (touch: p = 0.638, position: p = 0.449). No violation was found when a target range was not presented (touch: p = 0.003, position: p = 0.001). Thus, a target range reproduces the Fitts' Law violation previously documented with individually outlined targets, which supports and extends the discussed hypothesis. [ABSTRACT FROM AUTHOR]

Purpose: The current investigation aimed to examine the running performance of elite Ladies Gaelic football (LGF) match-play and establish the within game positional profile, in addition to the running performance of players across halves of play. Methods: GPS technology was used to examine the running performance of thirty-three (n = 33) elite LGF players (age; 23 ± 5 years, height; 173 ± 5 cm, body mass; 63 ± 4 kg). Across the duration of the observational period, one hundred and thirty-one (n = 131) individual samples were collected over 15 competitive matches. Data were classified based on positional line and across halves of play. Running performance was determined across the following performance variables of total distance covered (m) (TD), relative distance (m·min−1), HSR (≥ 4.4 m·s−1), RHSR (HSR; m·min−1), percentage HSR (% HSR), VHSR (≥ 5.5 m·s−1), peak velocity (m s−1), percentage peak velocity (% PeakV), accelerations (n; ≥ 3 m·s−2) and decelerations (n; ≤ − 3 m·s−2), Results: Independent of position the mean distance covered during match play was 7319 ± 1021 m which equates to a relative work rate of 116 ± 9 m·min−1. The high-speed distance of players was 1547 ± 432 m, which equates to a relative high-speed running distance of 25 ± 11 m·min−1. The maximal velocity of players was 7.17 ± 0.41 m·s−1 reflective of a relative maximal velocity of 86 ± 4 percent. The greatest distances were covered by half-backs, midfielders, and half-forwards, with these positions covering significantly greater distances than full-backs (p ≤ 0.05; ES: 0.59–1.25; small-moderate) and full-forwards (p ≤ 0.05; ES: 0.44–1.21; small-moderate). While TD and maximal velocity was shown not to change across halves of play, significant reductions (p ≤ 0.05) were observed across HSR (ES: 0.64–1.02; moderate), accelerations and decelerations (ES: 0.59–1.20; moderate). Conclusion: The current investigation is the first of its kind within elite LGF, the data within the investigation can aid the coaching process by allowing for the development of sport-specific training regimen specific to the positional needs of elite LGF players. Invasion risks may be influenced either negatively or positively by climate change, depending on the species. These can be predicted with species distribution models, but projections can be strongly affected by the source of the environmental data (climate data source, Global Circulation Models GCM and Shared Socio-economic Pathways SSP). We modelled the distribution of Phelsuma grandis and P. laticauda, two Malagasy reptiles that are spreading globally. We accounted for drivers of spread and establishment using socio-economic factors (e.g., distance from ports) and two climate data sources, i.e., Climatologies at High Resolution for the Earth's and Land Surface Areas (CHELSA) and Worldclim. We further quantified the degree of agreement in invasion risk models that utilised CHELSA and Worldclim data for current and future conditions. Most areas identified as highly exposed to invasion risks were consistently identified (e.g. in Caribbean and Pacific Islands). However, projected risks differed locally. We also found notable differences in quantitative invasion risk (3% difference in suitability scores for P. laticauda and up to 14% for P. grandis) under current conditions. Despite both species native distributions overlapping substantially, climate change will drive opposite responses on invasion risks by 2070 (decrease for P. grandis, increase for P. laticauda). Overall, projections of future invasion risks were the most affected by climate data source, followed by SSP. Our results highlight that assessments of current and future invasion risks are sensitive to the climate data source, especially in islands. Anxiety is one of the most frequently reported co-occurring conditions for autistic children and adolescents. The relationship between anxiety and social outcomes in autistic youth has been the focus of a range of studies, with mixed results. This meta-analysis aimed to identify the strength of the association between anxiety and a frequently researched social outcome (social competence) in autistic young people and whether that association is influenced by individual or research design factors. A previous preregistered systematic review was updated with a search of the same three databases (CINAHL, ERIC, and PsycINFO) as the original review. Through this, 20 studies with sufficient data on a neurotypically-defined measure of social competence and anxiety were identified. Results were synthesised using a mixed effects model. The meta-analysis on 2,321 participants (from 22 samples) highlighted wide heterogeneity in results. The findings show that anxiety has a significant, small negative impact on social competence (d = − 0.48; 95% CI = − 0.71, − 0.26), meaning that as scores on measures of anxiety increase, scores on measures of social competence decrease. This relationship between anxiety and social competence was moderated by age, becoming weaker as age increased. Whilst this is an important finding for supporting mental health and well-being of autistic young people, the large amount of variance left unexplained suggests that multiple factors, including the use of measures designed for neurotypical people and the potential impact of camouflaging on such measures, need to be considered in future designs. Background : Developmental pulmonary vein pulmonary vein stenosis in the setting of prematurity is a rare and poorly understood condition. Diagnosis can be challenging in the setting of chronic lung disease of prematurity. High-resolution non-contrast chest computed tomography (CT) is the conventional method of evaluating neonates for potential structural changes contributing to severe lung dysfunction and pulmonary hypertension but may miss pulmonary venous stenosis due to the absence of contrast and potential overlap in findings between developmental pulmonary vein pulmonary vein stenosis and lung disease of prematurity. Objective: To describe the parenchymal changes of pediatric patients with both prematurity and pulmonary vein stenosis, correlate them with venous disease and to describe the phenotypes associated with this disease. Materials and methods: A 5-year retrospective review of chest CT angiography (CTA) imaging in patients with catheterization-confirmed pulmonary vein stenosis was performed to identify pediatric patients (< 18 years) who had a history of prematurity (< 35 weeks gestation). Demographic and clinical data associated with each patient were collected, and the patients' CTAs were re-reviewed to evaluate pulmonary veins and parenchyma. Patients with post-operative pulmonary vein stenosis and those with congenital heart disease were excluded. Data was analyzed and correlated for descriptive purposes. Results: A total of 17 patients met the inclusion criteria (12 female, 5 male). All had pulmonary hypertension. There was no correlation between mild, moderate, and severe grades of bronchopulmonary dysplasia and the degree of pulmonary vein stenosis. There was a median of 2 (range 1–4) diseased pulmonary veins per patient. In total, 41% of the diseased pulmonary veins were atretic. The right upper and left upper lobe pulmonary veins were the most frequently diseased (n = 13/17, 35%, n = 10/17, 27%, respectively). Focal ground glass opacification, interlobular septal thickening, and hilar soft tissue enlargement were always associated with the atresia of an ipsilateral vein. Conclusion: Recognition of the focal parenchymal changes that imply pulmonary vein stenosis, rather than chronic lung disease of prematurity changes, may improve the detection of a potentially treatable source of pulmonary hypertension, particularly where nonangiographic studies result in a limited direct venous assessment. Human chosen passwords are often predictable. Research has shown that users of similar demographics or choosing passwords for the same website will often choose similar passwords. This knowledge is leveraged by human password guessers who use it to tailor their attacks. In this paper, we demonstrate that a learning algorithm can actively learn these same characteristics of the passwords as it is guessing and that it can leverage this information to adaptively improve its guessing. Furthermore, we show that if we split our candidate wordlists based on these characteristics, then a multi-armed bandit style guessing algorithm can adaptively choose to guess from the wordlist which will maximise successes. Provenance studies demonstrate the important control of plate boundary mountain building on continental sediment routing systems. Less well understood is if subsidence and uplift in cratons also has the potential to affect the organization of sediment routing systems on continental scales. New detrital zircon provenance data from the Michigan Basin in the Midcontinent of North America preserve evidence of intrabasin provenance heterogeneity in Cambrian, Ordovician, and middle Devonian strata. These results suggest that cratonic basins serve as effective sediment barriers that prevent mixing within and across basins from 10 to 100 s of millions of years. Internal sediment mixing, sorting, and dispersal may be achieved by a combination of sedimentary processes and inherited low relief topography. These observations are consistent with provenance data sets from eastern Laurentian Midcontinent basins that show locally and regionally variable provenance signatures during the early Paleozoic. By the late Devonian, provenance signatures throughout the basins homogenized, consistent with the emergence of transcontinental sediment transport systems associated with Appalachian orogenesis at the plate margin. These results demonstrate the importance of cratonic basins on local and regional sediment routing systems suggesting that these features may impede the integration of continental-scale sediment routings systems, particularly during periods of plate margin quiescence.
