2. Determinants of hand pulse wave velocity and hand pulse transit time in healthy adults.

- Abstract
Arterial pulse wave velocity (PWV) is recognized as a convenient method to assess peripheral vascular stiffness. This study explored the clinical characteristics of hand PWV (hPWV) and hand pulse transit time (hPTT) in healthy adults (sixty males = 42.4 ± 13.9 yrs; sixty-four females = 42.8 ± 13.9 yrs) voluntarily participated in this study. The arterial pulse waveform and the anatomical distance from the radial styloid process to the tip of the middle finger of both hands were recorded in the sitting position. The hPWV was calculated as the traversed distance divided by hPTT between those two points. Male subjects showed significantly greater hPWV, systolic blood pressure, and pulse pressure than age-matched female subjects, while the hPTT was not significantly different between genders. Multiple linear regression analysis showed that gender is a common determinant of hPWV and hPTT, and that age and heart rate (HR) were negatively correlated with hPWV and hPTT, respectively. We conclude that male subjects have greater hPWV than female subjects. Ageing is associated with decreased hPWV, while increased HR is associated with a smaller hPTT. The hPWV and hPTT might be used as non-invasive indices to characterise the ageing and arterial stiffness of peripheral blood vessels. [ABSTRACT FROM AUTHOR]

3. A Retrospective Analysis of Robertsonian Translocations from a Single Center in China.

- Abstract
Robertsonian translocations (ROBs) are the most common structural chromosomal abnormalities in the general population, with an estimated incidence rate of 1/1000 births. In this study, we retrospectively analyzed the cases of ROBs from September 2015 to August 2022 and totally identified ROB carriers from 84,569 specimens karyotyped in a single accredited laboratory in China, including 189 cases of balanced ROBs and 3 of mosaic ROBs. Microsoft Excel and descriptive statistics were used to record and analyze the collected data. The male/female ratio of ROBs is 1/1.29, with der(13;14) and der(14;21) being the main karyotypes. Among the 192 patients, 7 were lost to follow-up, 82 had given birth, and 103 were childless (such as miscarriage, fetal chromosomal abnormalities, in vitro fertilization (IVF) failure, or divorce). A total of 44 amniocenteses were performed in 42 couples; ROB cases with natural pregnancies showed that the normal karyotype and balanced ROBs of fetal accounted for 66.67% (16/24), while the results of assisted pregnancies showed 90.00% (18/20). This study represents the largest collections of ROBs in Jiangxi population and reminder that the ROB carriers can achieve the ideal outcome for pregnancy with the appropriate genetic guidance and assisted reproductive technologies (ART). Background: Phospholipase A2 receptor (PLA2R)-associated membranous nephropathy (MN) was manifested as seropositive for PLA2R antibodies (SAb) and/or glomerular PLA2R antigens' (GAg) deposits. According to the test of SAb and GAg, PLA2R-associated MN can be divided into SAb + /GAg−, SAb−/GAg + , and SAb + /GAg + groups. The clinical characteristics and outcomes of the three groups need to be further evaluated. Methods: 184 PLA2R-associated MN patients were enrolled. SAb was measured by enzyme-linked immunosorbent assay with a cut-off value of 14 RU/mL. GAg was detected by immunofluorescence using a paraffin section of renal biopsy samples. Clinical characteristics and the decline of eGFR were compared among the 3 groups. Results: There were 33 SAb + /GAg−, 46 SAb−/GAg +, and 105 SAb + /GAg + PLA2R-associated MN patients reviewed. Clinical characteristics, such as the level of proteinuria, serum albumin, as well as eGFR, were comparable between the SAb + /GAg− and SAb + /GAg + patients. While SAb−/GAg + patients exhibited mild clinical manifestations as evidenced by higher serum albumin (P < 0.001) and lower proteinuria (p = 0.049) compared with SAb + /GAg + patients. After 21.96 ± 7.39 month follow-up, the eGFR decrease was no difference between the SAb + /GAg− and SAb + /GAg + patients. SAb−/GAg + patients had a lower rate of the > 20% eGFR decline as well as a 50% eGFR decline compared with the SAb + /GAg + patients (10.87% vs 30.48%, p = 0.013; 0.00% vs 4.76%, p = 0.324). Conclusions: Our study showed that the clinical manifestations of SAb + /Gag− patients were the same as those of double-positive patients, while SAb−/GAg + patients exhibited mild clinical manifestations and slower eGFR decline compared to the double-positive patients. 5. Association between aging-related biomarkers and longitudinal trajectories of intrinsic capacity in older adults.

Intrinsic capacity (IC), the composite of physical and mental capacities, declines with age at different rates and patterns between individuals. We aimed to investigate the association between longitudinal IC trajectories and plasma biomarkers of two hallmarks of aging—chronic inflammation and mitochondrial dysfunction—in older adults. From the Multidomain Alzheimer Preventive Trial (MAPT), we included 1271 community-dwelling older people (mean [SD] age = 76.0 [4.3] years) with IC data over four years. Group-based multi-trajectory modeling was performed to identify clusters of the participants with similar longitudinal patterns across four IC domains: cognition, locomotion, psychology, and vitality. Five IC multi-trajectory groups were determined: low in all domains (8.4%), low locomotion (24.6%), low psychological domain (16.7%), robust (i.e., high in all domains except vitality; 28.3%), and robust with high vitality (22.0%). Compared to the best trajectory group (i.e., robust with high vitality), elevated levels of plasma interleukin-6 (IL-6), tumor necrosis factor receptor-1 (TNFR-1), and growth differentiation factor-15 (GDF-15) were associated with a higher risk of belonging to the "low in all domains" group (IL-6: relative risk ratio (RRR) [95% CI] = 1.42 [1.07 – 1.88]; TNFR-1: RRR = 1.46 [1.09 – 1.96]; GDF-15: RRR = 1.99 [1.45 – 2.73]). Higher IL-6 and GDF-15 also increased the risk of being in the "low locomotion" group. GDF-15 outperformed other biomarkers by showing the strongest associations with IC trajectory groups. Our findings found that plasma biomarkers reflecting inflammation and mitochondrial impairment distinguished older people with multi-impaired IC trajectories from those with high-stable IC. Highlights: A virus-like nanoparticle (reBiosome) was developed via site-specific codon mutation for displaying unnatural amino acid (Azi) on virus envelope protein at a rational site, followed by conjugating weak acid-responsive polyethylene glycol polymer on Azi via bioorthogonal chemistry. The reBiosome exhibited reduced virus-like immunogenicity, prolonged blood circulation and enhanced delivery to weakly acidic disease foci. The reBiosome enabled efficient delivery of gene editing and gene silencing system, demonstrating remarkable therapeutic efficacy in breast cancer and arthritis, respectively. Gene therapy offers potentially transformative strategies for major human diseases. However, one of the key challenges in gene therapy is developing an effective strategy that could deliver genes into the specific tissue. Here, we report a novel virus-like nanoparticle, the bioorthgonal engineered virus-like recombinant biosome (reBiosome), for efficient gene therapies of cancer and inflammatory diseases. The mutant virus-like biosome (mBiosome) is first prepared by site-specific codon mutation for displaying 4-azido-L-phenylalanine on vesicular stomatitis virus glycoprotein of eBiosome at a rational site, and the reBiosome is then prepared by clicking weak acid-responsive hydrophilic polymer onto the mBiosome via bioorthogonal chemistry. The results show that the reBiosome exhibits reduced virus-like immunogenicity, prolonged blood circulation time and enhanced gene delivery efficiency to weakly acidic foci (like tumor and arthritic tissue). Furthermore, reBiosome demonstrates robust therapeutic efficacy in breast cancer and arthritis by delivering gene editing and silencing systems, respectively. 7. Role of TPEN in Amyloid-β25–35-Induced Neuronal Damage Correlating with Recovery of Intracellular Zn2+ and Intracellular Ca2+ Overloading.

The overproduction of neurotoxic amyloid-β (Aβ) peptides in the brain is a hallmark of Alzheimer's disease (AD). To determine the role of intracellular zinc ion (
i Zn2+ ) dysregulation in mediating Aβ-related neurotoxicity, this study aimed to investigate whether N, N, N′, N′‑tetrakis (2‑pyridylmethyl) ethylenediamine (TPEN), a Zn2+ ‑specific chelator, could attenuate Aβ25–35 ‑induced neurotoxicity and the underlying mechanism. We used the 3-(4, 5-dimethyl-thiazol-2-yl)-2, 5-diphenyltetrazolium bromide assay to measure the viability of primary hippocampal neurons. We also determined intracellular Zn2+ and Ca2+ concentrations, mitochondrial and lysosomal functions, and intracellular reactive oxygen species (ROS) content in hippocampal neurons using live-cell confocal imaging. We detected L-type voltage-gated calcium channel currents (L-ICa ) in hippocampal neurons using the whole‑cell patch‑clamp technique. Furthermore, we measured the mRNA expression levels of proteins related to thei Zn2+ buffer system (ZnT-3, MT-3) and voltage-gated calcium channels (Cav1.2, Cav1.3) in hippocampal neurons using RT-PCR. The results showed that TPEN attenuated Aβ25–35 ‑induced neuronal death, relieved the Aβ25–35 ‑induced increase in intracellular Zn2+ and Ca2+ concentrations; reversed the Aβ25–35 ‑induced increase in ROS content, the Aβ25–35 ‑induced increase in the L-ICa peak amplitude at different membrane potentials, the Aβ25–35 ‑induced the dysfunction of the mitochondria and lysosomes, and the Aβ25–35 ‑induced decrease in ZnT-3 and MT-3 mRNA expressions; and increased the Cav1.2 mRNA expression in the hippocampal neurons. These results suggest that TPEN, the Zn2+ -specific chelator, attenuated Aβ25–35 ‑induced neuronal damage, correlating with the recovery of intracellular Zn2+ and modulation of abnormal Ca2+ -related signaling pathways. 8. Determination of the ambience duration of lavender essential oil with three perfume fixatives using the thermokinetics approach.

Various cosmetic perfumes have distinct fragrance residence times. Changes in the temperature of the surroundings can decrease thermal stability. Therefore, the determination of effective stabilisers for perfumes is indispensable. In this study, a thermogravimetric analyser was used to individually determine the thermal stability of three perfume fixatives added to the lavender essential oil. The apparent activation energy (Ea) was calculated using three thermokinetic equations: the Friedman differential iso-conversional, Flynn–Wall–Ozawa, and ASTM E698-18 kinetic equations. The thermal analysis test results revealed that trans-cinnamic acid exhibited the highest thermal stability among the four test samples. The final reaction temperature and the peak temperature of trans-cinnamic acid were approximately 80 °C more than that of lavender essential oil, which exhibited the lowest thermal stability. The Ea was calculated using three thermokinetic equations. The Ea estimation of trans-cinnamic acid was 85.2–95.1 kJ mol–1. In the thermal analytical experiments of the perfume samples, the sample of lavender essential oil with ethanol exhibited the highest mass loss derivative, which indicated that perfume fixatives added to lavender essential oils with ethanol increased the fragrance residence time. Accurate thermokinetic parameters of lavender essential oils determined in the experiment can optimise cosmetic perfume manufacturing processes and related research areas. [ABSTRACT FROM AUTHOR]

We investigated combining a core AD neuropathology measure (plasma amyloid-beta [Aβ]
42/40 ) with five plasma markers of inflammation, cellular stress, and neurodegeneration to predict cognitive decline. Among 401 participants free of dementia (median [IQR] age, 76 [73–80] years) from the Multidomain Alzheimer Preventive Trial (MAPT), 28 (7.0%) participants developed dementia, and 137 (34.2%) had worsening of clinical dementia rating (CDR) scale over 4 years. In the models utilizing plasma Aβ alone, a tenfold increased risk of incident dementia (nonsignificant) and a fivefold increased risk of worsening CDR were observed as each nature log unit increased in plasma Aβ levels. Models incorporating Aβ plus multiple plasma biomarkers performed similarly to models included Aβ alone in predicting dementia and CDR progression. However, improving Aβ model performance for composite cognitive score (CCS) decline, a proxy of dementia, was observed after including plasma monocyte chemoattractant protein 1 (MCP1) and growth differentiation factor 15 (GDF15) as covariates. Participants with abnormal Aβ, GDF15, and MCP1 presented higher CCS decline (worsening cognitive function) compared to their normal-biomarker counterparts (adjusted β [95% CI], − 0.21 [− 0.35 to − 0.06], p = 0.005). In conclusion, our study found limited added values of multi-biomarkers beyond the basic Aβ models for predicting clinically meaningful cognitive decline among non-demented older adults. However, a combined assessment of inflammatory and cellular stress status with Aβ pathology through measuring plasma biomarkers may improve the evaluation of cognitive performance. 11. Next-generation sequencing: a follow-up of 36,913 singleton pregnancies with noninvasive prenatal testing in central China.

Purpose: To evaluate the noninvasive prenatal testing (NIPT) results of 36,913 cases in Jiangxi province of central China and explore its application value in prenatal screening and diagnosis. Methods: This retrospective analysis included 36,913 singleton pregnant women who underwent NIPT because of moderate-/high-risk pregnancy or voluntary requirements between January 2017 and December 2019 in our hospital. Chromosomal abnormalities such as trisomies 21, 18, and 13 (T21, T18, T13) and sex chromosome aneuploidies (SCAs) were judged by standard Z-score analysis. Positive NIPT results were confirmed by amniocentesis and karyotyping. Pregnancy outcomes were followed up via telephone interview. Results: A total of 1.01% (371/36,913) positive cases were detected by NIPT, comprising 137, 46, 31, and 157 cases of T21, T18, T13, and SCAs, respectively. A total of 116 of T21, 27 of T18, 13 of T13, and 51 of SCAs were confirmed to be true positive; all normal cases that had been followed up were verified to be true negative. The NIPT sensitivity in T21, T18, T13, and SCAs was 100.00% individually, whereas the specificity was 99.94% (36,488/36,509), 99.95% (36,579/36,598), 99.95% (36,594/36,612), and 99.72% (36,472/36,574), respectively. Furthermore, the negative predictive values of T21, T18, T13, and SCAs were all 100%, while the positive predictive values were 84.67%, 58.70%, 41.94%, and 33.33%, respectively. Conclusion: NIPT is highly sensitive and has a low false positive rate in testing clinically significant fetal aneuploidies of general reproductive women. However, this technique cannot substitute for amniocentesis and karyotyping, and detailed genetic counseling is also essential for the high-risk group of NIPT. [ABSTRACT FROM AUTHOR]

NADPH oxidase (NOX) enzymes are involved in a various physiological and pathological processes such as platelet activation and inflammation. Interestingly, we found that the pan-NOX inhibitors VAS compounds (VAS2870 and its analog VAS3947) exerted a highly potent antiplatelet effect. Unlike VAS compounds, concurrent inhibition of NOX1, 2, and 4 by treatment with ML171, GSK2795039, and GKT136901/GKT137831 did not affect thrombin and U46619-induced platelet aggregation. These findings suggest that VAS compounds may inhibit platelet aggregation via a NOX-independent manner. Thus, we aimed to investigate the detailed antiplatelet mechanisms of VAS compounds. The data revealed that VAS compounds blocked various agonist-induced platelet aggregation, possibly via blocking PKC downstream signaling, including IKKβ and p38 MAPK, eventually reducing platelet granule release, calcium mobilization, and GPIIbIIIa activation. In addition, VAS compounds inhibited mouse platelet aggregation-induced by collagen and thrombin. The in vivo study also showed that VAS compounds delayed thrombus formation without affecting normal hemostasis. This study is the first to demonstrate that, in addition to inhibiting NOX activity, VAS compounds reduced platelet activation and thrombus formation through a NOX-independent pathway downstream of PKC. These findings also indicate that VAS compounds may be safe and potentially therapeutic agents for treating patients with cardiovascular diseases. [ABSTRACT FROM AUTHOR]

In the staging of many sport events, ranging from grassroots to mega-events, volunteers have become a vital component of the sport industry. This study aimed to examine the sport volunteering behaviour and well-being of young adults within the framework of the theory of planned behaviour. Participants were 347 college students (61% males; M age = 19.80 years, SD = 1.69) majoring in sport and physical education. Attitude, subjective norms, perceived behavioural control, intention, behaviour, and well-being were measured in different ways with a two-time-point design. The results revealed that perceived behavioural control positively influenced both volunteering intention and behaviour and that sport volunteering behaviour effectively predicted well-being. However, attitude and subjective norms did not influence intention. This study provides practical implications not only for sport volunteerism researchers but also for managers in developing strategies for recruiting and managing sport volunteers. A study in eastern Taiwan evaluated the importance of montane water contribution (MC) to adjacent valley-plain groundwater (VPG) in a tectonic suture zone. The evaluation used a ternary natural-tracer-based end-member mixing analysis (EMMA). With this purpose, VPG and three end-member water samples of plain precipitation (PP), mountain-front recharge (MFR), and mountain-block recharge (MBR) were collected and analyzed for stable isotopic compositions (δ

After evaluation, Cl− is deemed unsuitable for EMMA in this study, and the contribution fractions of respective end members derived by the δ18 O-EC pair are similar to those derived by the δ2 H-EC pair. EMMA results indicate that the MC, including MFR and MBR, contributes at least 70% (679 × 106 m3 water volume) of the VPG, significantly greater than the approximately 30% of PP contribution, and greater than the 20-50% in equivalent humid regions worldwide. The large MC is attributable to highly fractured strata and the steep topography of studied catchments caused by active tectonism. Furthermore, the contribution fractions derived by EMMA reflect the unique hydrogeological conditions in the respective study sub-regions. A region with a large MBR fraction is indicative of active lateral groundwater flow as a result of highly fractured strata in montane catchments. On the other hand, a region characterized by a large MFR fraction may possess high-permeability stream beds or high stream gradients. Type 2 diabetes mellitus (T2DM) is a known cause of cognitive dysfunction, and brain-derived neurotrophic factor (BDNF) is a key protein in promoting memory growth and survival of neurons. However, the relationship between plasma BDNF and diabetic cognitive dysfunction is still elusive. A total of 89 patients over 60 years with T2DM and 40 well-matched health controls were enrolled. All participants received a set of multi-dimensional neuropsychological tests for the cognitive assessment. The subjects were divided into amnesic mild cognitive impairment (aMCI) and non-aMCI groups. An enzyme-linked immunosorbent assay (ELISA) was used to measure plasma BDNF concentrations for all subjects. No significant difference was found between T2DM patients and healthy control in MMSE scores. The T2DM patients performed significantly worse in four cognitive domains (including episodic memory, executive function, visuospatial function, and information processing speed) compared with the controls (all p < 0.05). The prevalence of aMCI in T2DM population was higher [OR = 4.032 (1.536~10.582), 37/89-6/40]. Additionally, the plasma concentration of BDNF in T2DM patients was significantly lower than that in controls (p < 0.01). However, no significant correlation was found between plasma BDNF and cognitive function in T2DM. Our results suggested that T2DM have a higher prevalence of cognitive impairment. The plasma BDNF concentration in T2DM patients was significantly lower than that in controls, but low BDNF was not a biomarker for cognitive dysfunction in T2DM patients. [ABSTRACT FROM AUTHOR]

The Mesoproterozoic Kawabulake Group, which is unconformably overlain by the Lower Cambrian Huangshan Formation and conformably overlies the Mesoproterozoic Xingxingxia Group in the Eastern Tianshan area, NW China, is comprised mainly of siltstone, slate, sandstone and phyllite. New geochemical data for the clastic rocks from the Kawabulake Group were investigated to constrain the provenance and weathering history of the source rocks, in order to evaluate the tectonic evolution of the Eastern Tianshan area. Kawabulake Group rocks are compositionally similar to PAAS (average Post-Archean Australian Shale), indicating derivation from a felsic source that is characterized by depletion in some HFSEs such as Nb, Ta and Ti. The Chemical Index of Alteration (CIA) for the sandstone, siltstone and slate samples (CIA = 60 on average) suggests intensely weathered sources. Light REE-enrichment patterns ((La/Yb) = 4-20) coupled with negative Eu anomalies (Eu/Eu* between 0.44 and 0.89 with an average of 0.62) are similar to those of PAAS, consistent with cratonic sources. The major and trace element compositions imply a dominantly Precambrian felsic source region with a minor contribution from mafic materials. The Sr-Nd isotopic compositions and the ages of T (~3.4, 2.5-1.8, 1.2 and 1.0 Ga) are consistent with the evolutionary history of Kuluketage in the northern Tarim, suggesting that the Tarim Craton was the main source area for the Kawabulake Group. The precise relation between heart rate variability (HRV) and autonomic re-innervation has not been established explicitly in patients after orthotopic heart transplantation (OHT), but can be inferred from the fact that the HRV is reduced immediately after OHT and may increase gradually with time. The aim of this study was to investigate the residual HRV in patients about 1-2 years after OHT, as compared with patients after coronary artery bypass graft (CABG) surgery. Thirteen patients who had received OHT and 14 patients who had received CABG surgery were recruited. HRV analysis was performed and the HRV measures in supine position were compared between these two groups of patients. We found that the mean (mRRI), standard deviation and coefficient of variation of RR intervals, total power, very low frequency power (VLFP), low frequency power, high frequency power (HFP), normalized VLFP (nVLFP) and low-/high-frequency power ratio in the OHT group were all significantly decreased, while the heart rate (HR) and normalized HFP (nHFP) were significantly increased, as compared with the CABG group. The decrease in HRV was more severe in the VLFP region. A smaller nVLFP and a greater nHFP were associated with a smaller mRRI and a larger HR in the OHT patients. The slope of the power law relation of HRV became positive in OHT patients, instead of negative in CABG patients. We conclude that patients after OHT have residual HRV which were characterized by severely depressed time and frequency domain HRV, increased HR and nHFP, decreased nVLFP, and positive slope of the power-law relation of HRV. The use of nHFP as the indicator of vagal modulation and the use of nVLFP as the indicator of renin-angiotensin modulation, thermoregulation and vagal withdrawal must be careful in the OHT patients. As the submitted articles increase the burden of paper reviewing process gets heavy. It is not only time-consuming and laborious, but cannot find proper reviewers to review the specific manuscripts in some cases. Sometimes, the assignment of papers to specific reviewers is not objective enough. Thus, this study is based on the design concept of content management, CMS, to develop an online collaborative electronic journal paper review system. We adopt rich internet application, RIA, technology to web application development and apply text mining technologies to articles classification. We propose an assignment mechanism of paper reviews to achieve the automatic, fair and efficient paper reviewers΄ assignment. Research about organizations has consistently revealed that proactive personality is a relatively stable disposition and a significant antecedent of self-efficacy, which generates employees' proactive behavior and thus benefits individuals and firms. Consequently, the present study aimed to examine whether the relationship between proactive personality, a general temperamental tendency, and specific self-efficacy that is focused on certain activities or tasks might emerge across contexts in an education setting with a directional effect. From a sample of 123 students, we employed a longitudinal approach using two waves of surveys to examine the directional relations between students' proactive personalities and their academic self-efficacy in education. The results showed that students' proactive personalities at Time 1 predicted change in their academic self-efficacy at Time 2, suggesting a top-down relationship. On the contrary students' academic self-efficacy at Time 1 could not predict their proactive personalities at Time 2. In short, the directional effect of proactive personality on self-efficacy across contexts was captured in this study. Implications and limitations were discussed. [ABSTRACT FROM AUTHOR]

gamma-Aminobutyric acid (GABA) is the major inhibitory neurotransmitter in the central nervous system, and it also appears in peripheral tissues. Platelets are anuclear blood cells that play a central role in hemostatic processes. Although platelets possess a GABA uptake system, the functional activity of GABA in platelets has remained unclear. We determined that GABA is abundantly distributed in the platelets at a concentration of approximately 1.03 ng/10 cells. GABA (0.5 μM) specifically inhibited collagen-induced platelet activation accompanied by [Ca] i mobilization, phospholipase Cγ2, protein kinase C, Akt phosphorylation, and hydroxyl radical formation. In addition, GABA interfered with fluorescein isothiocyanate-collagen binding to platelet membranes and produced a concentration-dependent shift in the collagen concentration-response curve and a Schild plot slope of −0.96 ± 0.11, indicating competitive inhibition. Platelet activation induced by convulxin, a glycoprotein VI agonist, was inhibited by GABA, whereas activation induced by the integrin αβ agonist, aggretin, was not. Immunoprecipitation and surface plasmon resonance revealed that GABA binds directly to glycoprotein VI in human platelets with equilibrium dissociation (binding) constant ( K) of 41.4 nM. The closure time of whole blood and the occlusion time of platelet plug formation were significantly prolonged by GABA in vivo. In this study, GABA is a specific inhibitor of collagen glycoprotein VI and may be involved in an endogenous negative feedback mechanism for platelet activation. Thus, GABA may represent a potential target for the development of novel interventions for the treatment of cardiovascular diseases associated with platelet activation, such as stroke and myocardial infarction. Objective: This study investigates the breathing frequency (BF)-independent effect of Tai Chi Chuan (TCC) on autonomic nervous modulation in TCC practitioners. Methods: Twenty-five TCC practitioners and 25 sedentary normal controls were recruited. The stationary heart rate variability (HRV) measures of TCC practitioners and controls were compared. The same HRV measures in TCC practitioners and among the controls, TCC practitioners before TCC and TCC practitioners 30 min after TCC were compared. Results: In TCC practitioners, the BF, normalized high-frequency power (nHFP), and normalized very low-frequency power were significantly increased, while the normalized low-frequency power (nLFP) was significantly decreased 30 min after TCC. The BF correlated significantly and negatively with heart rate (HR), nHFP and nLFP, and correlated significantly and positively with mean RR interval (Mn) before TCC in TCC practitioners. A slower BF is associated with a higher HR, a greater vagal modulation, and a greater combined sympatho-vagal modulation before TCC. To remove the effect of BF on HRV measures, new indices such as HR*BF, nHFP*BF, nLFP*BF, and Mn/BF were introduced for comparison among the controls, TCC practitioners before TCC, and TCC practitioners 30 min after TCC. Thirty minutes after TCC, the Mn/BF of TCC practitioner was smaller whereas HR*BF and nHFP*BF were greater than those before TCC. Interpretation: The BF-independent effects of TCC on the autonomic nervous modulation of TCC practitioners are an increase in vagal modulation and HR, and a decrease in mean RR interval. 26. A comparative study of pulse rate variability and heart rate variability in healthy subjects.

Both heart rate variability (HRV) and pulse rate variability (PRV) are noninvasive means for the assessment of autonomic nervous control of the heart. However, it is not settled whether or not the PRV obtained from either hand can be the surrogate of HRV. The HRV measures obtained from electrocardiographic signals and the PRV measures obtained from the pulse waves recorded from the index fingers of both hands were compared in normal subjects by using linear regression analysis and Bland and Altman method. Highly significant correlations ( P < 0.001, 0.89 < r < 1.0) were found between all HRV measures and the corresponding PRV measures of both hands. However, there were insufficient agreements in some measures between pairwise comparisons among HRV, right PRV and left PRV except heart rate and ultra-low frequency power (ULFP). The PRV of either hand is close to, but not the same as the HRV in healthy subjects. The HRV, right PRV and left PRV are not surrogates of one another in normal subjects except heart rate and ULFP. Since HRV is generally accepted as the standard method for the assessment of the autonomic nervous modulation of a subject, the PRV of either hand may not be suitable for the assessment of the cardiac autonomic nervous modulation of the subject. 27. Appropriateness of ambulatory prescriptions in Taiwan: translating claims data into initiatives.

Background Appropriate prescribing is fundamental to successful pharmacotherapy. The status of current ambulatory medication practices in medicine and pharmacy would be better understood through an analysis of community pharmacy prescription claims. Objective The aims of the study were to investigate patterns of the types of prescriptions claimed by community pharmacies, undetected prescription errors by community pharmacists, and associated factors of prescription errors. Setting A population-based claims database of prescriptions dispensed by community pharmacies in Taiwan. Methods Ambulatory prescriptions were randomly sampled and reassessed for prescribing appropriateness by medical center pharmacists using explicit criteria. Demographics of patients, physicians, care facilities, and prescription/dispensing details were assessed and used to identify associated factors for prescription errors using descriptive analyses as well as logistic regression. Main outcome measures Erroneous prescriptions prescribed by physicians, and dispensed and claimed through community pharmacies. Results The study included analyses of 3065 prescriptions dispensed in community pharmacies resulting from 1003 patient visits, mostly to physician or dental clinics (99.5%). Prescribing characteristics, patterns, and examples of prescription errors are described. Prescription errors were identified in 18.3% ( n = 560) of prescriptions and 34.9% ( n = 350) of patient visits. Potential prescribing errors included errors of omission (25.5%), errors of commission (53.4%), and others (21.1%). The top three errors were incorrect dosage (27.5%), missing indication (23.6%), and insufficient or unavailable drug information (18.9%). Drugs most frequently associated with prescription errors included antihistamines, hormones, and gastrointestinal agents. Prescription were also higher in the central and eastern regions of Taiwan. Pediatricians accounted for a disproportionate number of prescription errors. Conclusion Prescription errors are prevalent in ambulatory care in Taiwan, and differential practice standards exist between community and hospital services. This disparity needs to be reconciled by pertinent initiatives to enhance community-hospital and pharmacist-general practitioner communication and interprofessional educational efforts to improve medication use and safety. Andrographolide is a novel NF-κB inhibitor from the leaves of Andrographis paniculata. Platelet activation is relevant to a variety of thrombotic diseases. However, no data are available concerning the effects of andrographolide in platelet activation. The aim of this study was to examine the mechanisms of andrographolide in preventing platelet activation. Andrographolide (25-75 μΜ) exhibited a more potent activity of inhibiting platelet aggregation stimulated by collagen. Andrographolide inhibited collagen-stimulated platelet activation accompanied by relative Ca mobilization; thromboxane A formation; and phospholipase C (PLC)γ2, protein kinase C (PKC), mitogen-activated protein kinase (MAPK), and Akt phosphorylation. Andrographolide markedly increased cyclic GMP, but not cyclic AMP levels. Andrographolide also stimulated endothelial nitric oxide synthase (eNOS) expression, NO release, and vasodilator-stimulated phosphoprotein (VASP) phosphorylation. ODQ, an inhibitor of guanylate cyclase, markedly reversed the andrographolide-mediated inhibitory effects on platelet aggregation, p38 MAPK and Akt phosphorylation, and the andrographolide-mediated stimulatory effect on VASP phosphorylation. Furthermore, a PI3 kinase inhibitor (LY294002) but not a PKC inhibitor (Ro318220) significantly diminished p38 MAPK phosphorylation; nevertheless, a p38 MAPK inhibitor (SB203580) and LY294002 diminished PKC activity stimulated by collagen. Andrographolide also reduced collagen-triggered hydroxyl radical (OH) formation. In vivo studies revealed that andrographolide (22 and 55 μg/kg) is effective in reducing the mortality of ADP-induced acute pulmonary thromboembolism and significantly prolonged platelet plug formation in mice. This study demonstrates for the first time that andrographolide possesses a novel role of antiplatelet activity, which may involve the activation of the eNOS-NO/cyclic GMP pathway, resulting in the inhibition of the PI3 kinase/Akt-p38 MAPK and PLCγ2-PKC cascades, thereby leading to inhibition of platelet aggregation. The proton resonant properties in Ne, which determine the reaction rate of the key stellar O(α,p)F reaction, have been studied by using a technique of proton resonant elastic scattering of F+p. A 4.22 MeV/nucleon F radioactive ion (RI) beam was produced via a projectile-fragmentation reaction, and separated by a Radioactive Ion Beam Line in Lanzhou (RIBLL). By bombarding a thick (C2) target, the energy spectra of the recoiled protons were measured by two Δ E- E silicon telescopes at the center-of-mass scattering angles of θ≈175°±5°, θ≈152°±8°, respectively. Several proton resonances in Ne were observed clearly. A further R-matrix analysis of the experimental data is under way to determine the resonant parameters. Objective: To investigate the effect of pregnancy on the pulse wave of the mother.Patients and Methods: Pulse waveforms recorded via a pulse oximeter from the left index finger of non-pregnant women and pregnant women in the three trimesters of pregnancy were Fourier transformed into power spectra. Spectral indices of the power spectra of pulse waveform were obtained and compared among non-pregnant women and pregnant women in the three trimesters of pregnancy.Results: The power of harmonics of pulse wave decayed exponentially with respect to the order of harmonics. The exponent and initial value of exponential decay for the power of harmonics and the power of the 2nd harmonic were increased, whereas the total power of pulse and the powers of higher order harmonics were decreased during pregnancy.Conclusion: The power of harmonics of pulse wave can be described by an exponential decay function with respect to the order of harmonics in both non-pregnant and pregnant women. The effects of pregnancy on the pulse wave are the reduction in the total power of pulse and the power of higher order harmonics, and the increase in the power of lower order harmonics in the power spectrum of pulse wave. This effect of pregnancy on the pulse wave might be caused by the decrease in vascular resistance during pregnancy, the increase in workload on the heart due to increased demand of the growing fetus, and the aortocaval compression caused by the progressively enlarged gravid uterus and fetus. The purpose of this study is to explore the relationships between perceived coworker loafing and counterproductive work behaviors toward the organization (CWB-O) and toward the coworkers (CWB-I). Data were collected from 184 supervisor–employee pairs from multiple sources (i.e., self-rated and supervisor-rated). Structural equation modeling (SEM) analyses were conducted to test our hypotheses. The results of SEM showed that perceived loafing was positively related to CWB-O (self-rated) and CWB-I (self-rated and supervisor-rated). Moreover, a revenge motive toward the organization fully mediated the relationship between perceived loafing and CWB-O, whereas a revenge motive toward coworkers fully mediated the relationship between perceived loafing and CWB-I. This study advances our understanding as to how and why perceived coworker loafing increases employees’ CWB-I and CWB-O. Our investigation also highlights the important cognitive mediator: revenge motive in the perceived loafing–CWB linkage. This is one of the first studies which examines the relationships between perceived coworker loafing and two facets of CWB, and investigates a cognitive mediator (i.e., a revenge motive) that underlines the perceived loafing–CWB linkage. - Abstract
Abstract  It has been reported that over-expression of vascular endothelial growth factor-C (VEGF-C) in tumors leads to increased lymphangiogenesis and resistance to chemotherapy. Therefore, we hypothesized that VEGF-C would be a good molecular target for cancer gene therapy. In this study, we silenced the expression of VEGF-C with the highly specific post-transcriptional suppression of RNA interference (RNAi) in human breast cancer MCF-7 cell line. The expression of VEGF-C was examined by reverse transcription-polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay (ELISA), and the effect of plasmid on human lymphatic endothelial cells (HLECs) in vitro was analyzed by migration and 3-(4, 5-dimethylt-hiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. The sensitivity to anticancer agents was evaluated by MTT and apoptosis assay, and apoptosis-related genes bcl-2/bax ratio was determined by Western Blotting. Results showed that of three siRNA-expressing vectors, P-1/siRNA most significantly suppressed the expression of VEGF-C mRNA and protein (38.1% of control and 117.8 ± 24.2 pg/ml, respectively) and interfered with proliferation and migration of HLECs in vitro. Moreover, transfection of VEGF-C/siRNA combined with Epirubicin markedly decreased breast cancer cells viability, reaching up to 38.5%, and increased apoptosis rate from 13.1% to 38.9%, as determined by decrease of bcl-2/bax ratio. In summary, VEGF-C would be a good molecular target, and a combination of Epirubicin and RNAi targeting VEGF-C could be an effective means for suppressing lymphatic metastasis and enhancing chemosensitivity of human breast cancer cells. Background Despite the numerous in-vitro studies on the mechanical properties and simulated injury mechanisms of the anterior talofibular ligament (ATFL) and calcaneofibular ligament (CFL), the in-vivo biomechanical behavior of these two ligaments has not yet been described. Methods Apparent length of the ATFL and CFL was measured in four ankles in healthy male subjects between 32 and 45 years of age (two left and two right) during a dorsiflexion-plantarflexion and supination-pronation arc of motion using a combined dual-orthogonal fluoroscopic and magnetic resonance imaging technique. Results The ATFL elongated from the neutral position at 16.3 +/- 3.0 mm to 20.8 +/- 2.7 mm at maximal plantarflexion and shortened significantly from the neutral position to 13.9 +/- 2.9 mm at maximal dorsiflexion (p = 0.01). The CFL shortened from the neutral position at 28.0 +/- 2.9 mm to 26.6 +/- 2.2 mm at maximal plantarflexion (p = 0.08) and elongated significantly from the neutral position to 29.9 +/- 3.0 mm at maximal dorsiflexion (p = 0.003). The ATFL elongated significantly from 14.8 +/- 2.5 mm at maximal pronation to 17.4 +/- 3.0 mm at maximal supination (p = 0.08). At the same time, the CFL shortened from 31.0 +/- 3.8 mm at maximal pronation to 26.9 +/- 3.6 mm at maximal supination (p = 0.02). Conclusion The results showed that the ATFL elongates more during plantarflexion and supination whereas the CFL increases in length with dorsiflexion and pronation. Concurrently, these data also demonstrated the reciprocal function between the two ligaments. While one shortens, the other one elongates. The different elongation of the ATFL and CFL during the same motion arc suggests that under excessive loading conditions the ATFL might be more vulnerable in plantarflexion and supination while the CFL might be more susceptible to injury in dorsiflexion and pronation. Furthermore, in the case of surgical reconstruction the grafts used to reconstruct the two ligaments may need to be tensioned at different positions of the ankle in order to reproduce their natural in vivo function.

Honokiol, derived from Magnolia officinalis, has various pharmacological properties. Platelet activation plays a critical role in cardiovascular diseases. Honokiol has been reported to inhibit collagen-stimulated rabbit platelet aggregation. However, detailed further studies on the characteristics and functional activity of honokiol in platelet activation are relatively lacking. In the present study, honokiol specifically inhibited platelet aggregation and Ca
+2 ion mobilization stimulated with collagen or convulxin, an agonist of glycoprotein (GP) VI, but not with aggretin, an agonist of integrin α2 β1 . Honokiol also attenuated the phosphorylation of Lyn, PLCγ2, PKC, MAPKs, and Akt after convulxin stimulation. Honokiol have no cytotoxicity in zebrafish embryos. Honokiol diminished the binding of anti-GP VI (FITC-JAQ1) mAb to human platelets, and it also reduced the coimmunoprecipitation of GP VI-bound Lyn after convulxin stimulation. The surface plasmon resonance results revealed that honokiol binds directly to GP VI, with a KD of 289 μM. Platelet function analysis revealed that honokiol substantially prolonged the closure time in human whole blood and increased the occlusion time of thrombotic platelet plug formation in mice. In conclusion, honokiol acts as a potent antagonist of collagen GP VI in human platelets, and it has therapeutic potential in the prevention of the pathological thrombosis. Positron emission tomography (PET), a cornerstone in cancer diagnosis and treatment monitoring, relies on the enhanced uptake of fluorodeoxyglucose ([
18 F]FDG) by cancer cells to highlight tumors and other malignancies. While instrumental in the clinical setting, the accuracy of [18 F]FDG-PET is susceptible to metabolic changes introduced by radiation therapy. Specifically, radiation induces the formation of giant cells, whose metabolic characteristics and [18 F]FDG uptake patterns are not fully understood. Through a novel single-cell gamma counting methodology, we characterized the [18 F]FDG uptake of giant A549 and H1299 lung cancer cells that were induced by radiation, and found it to be considerably higher than that of their non-giant counterparts. This observation was further validated in tumor-bearing mice, which similarly demonstrated increased [18 F]FDG uptake in radiation-induced giant cells.
