Mycotoxins are secondary metabolites produced by specific fungi. More than 400 different mycotoxins are known in the world, and the concentration of these toxins in food and feed often exceeds the acceptable limit, thus causing serious harm to animals and human body. At the same time, modern industrial agriculture will also bring a lot of environmental pollution in the development process, including the increase of heavy metal content, and often the clinical symptoms of low/medium level chronic heavy metal poisoning are not obvious, thus delaying the best treatment opportunity. However, the traditional ways of detoxification cannot completely eliminate the adverse effects of these toxins on the body, and sometimes bring some side effects, so it is essential to find a new type of safe antidote. Trace element selenium is among the essential mineral nutrient elements of human and animal bodies, which can effectively remove excessive free radicals and reactive oxygen species in the body, and has the effects of antioxidant, resisting stress, and improving body immunity. Selenium is common in nature in inorganic selenium and organic selenium. In previous studies, it was found that the use of inorganic selenium (sodium selenite) can play a certain protective role against mycotoxins and heavy metal poisoning. However, while it plays the role of antioxidant, it will also have adverse effects on the body. Therefore, it was found in the latest study that selenium yeast could not only replace the protective effect of sodium selenite on mycotoxins and heavy metal poisoning, but also improve the immunity of the body. Selenium yeast is an organic selenium source with high activity and low toxicity, which is produced by selenium relying on the cell protein structure of growing yeast. It not only has high absorption rate, but also can be stored in the body after meeting the physiological needs of the body for selenium, so as to avoid selenium deficiency again in the short term. However, few of these studies can clearly reveal the protective mechanism of yeast selenium. In this paper, the detoxification mechanism of selenium yeast on mycotoxins and heavy metal poisoning was reviewed, which provided some theoretical support for further understanding of the biological function of selenium yeast and its replacement for inorganic selenium. The conclusions suggest that selenium yeast can effectively alleviate the oxidative damage by regulating different signaling pathways, improving the activity of antioxidant enzymes, reversing the content of inflammatory factors, regulating the protein expression of apoptosis-related genes, and reducing the accumulation of mycotoxins and heavy metals in the body. In this study, a dynamic analysis method for flexible multibody systems using Reissner–Mindlin shells was developed, and an edge center-based strain smoothing mixed interpolation of tensorial components (MITC) method that described the deformation of flexible shells was presented. Three primary achievements were completed. First, a new dynamics equation for Reissner–Mindlin shells based on the floating frame of reference formulation was modeled, in which the coupling of the bending and membrane deformations with rigid motion was considered comprehensively. Second, an edge center-based strain smoothing (ECSS) method that effectively improved the membrane and bending behavior for MITC3 elements was proposed. Third, the ECSS method was designed to construct a linear strain field within an element to ensure the strain consistency at the junction center of the element, resulting in a smoother stress field. The superior performance of the proposed method was verified by convergence analyses, and its advantages for flexible multibody dynamics analyses were also highlighted numerically. This study aimed to explore the effect and mechanism of Bacillus coagulans TL3 (B. coagulansTL3) on ileal inflammatory injury induced by lipopolysaccharide (LPS). Animal models were established wherein male Wistar rats were randomly divided into four groups: a control group, an LPS group, a high-concentration B. coagulans (HBC) group, and a low-concentration B. coagulans (LBC) group. The results showed that the biochemical indices changed, significant pathological changes were found, the number of apoptotic cells increased in the ileal tissue of the LPS group rats; the protein expressions of NFκB, MYD88, TLR4, TNF-α, Il-6, IL-1β, Claudin-1, Occludin, and ZO-1 in the LPS group were significantly decreased. The biochemical indices, pathological changes, and protein expressions in rats subjected to intragastric administration with high or low concentrations of B. coagulans TL3, were significantly reversed compared with the LPS group. These results indicated that TL3 strain could protect rats against ileal oxidative stress and inflammation induced by LPS and the protective mechanism was related to inhibition of the toll-like receptor 4 (TLR4) / myeloid differentiation factor-88 (MyD88) signaling pathway. Zearalenone (ZEA) is a mycotoxin with estrogenic activity whose main effect is to impair the reproductive systems of animals. It leads to reproductive disorders in livestock and thus causes serious losses to agriculture and animal husbandry. This study aims to examine whether ZEA induces toxicity in Leydig cells through the PI3K/AKT signaling pathway and also to investigate the role played by the upstream phosphatase and tensin homolog (PTEN) gene. An adenovirus vector model was constructed to interfere with the PTEN gene to investigate whether ZEA promotes the apoptosis of TM3 cells through the PI3K/AKT pathway. Apoptosis was detected cytometrically and the protein expression levels of PTEN, AKT, p-AKT, Bax, and Bcl-2 were evaluated via western blot analysis. The results show that ZEA induces apoptosis of TM3 cells. PTEN expression is significantly increased (P < 0.01), Bax expression is increased (P < 0.05), AKT and p-AKT expression of anti-apoptotic protein is significantly decreased (P < 0.01), and Bcl-2 protein expression is decreased (P < 0.05) in the ZEA group compared with the control group. In the shRNA+ZEA group, the expression levels of PTEN and Bax proteins are significantly decreased (P < 0.01), AKT protein is significantly increased (P < 0.01), and p-AKT protein is increased (P < 0.05) compared with the ZEA group. This study thus demonstrates that ZEA promotes apoptosis of TM3 cells by targeting PTEN and thus inhibiting the PI3K/AKT signal pathway. [ABSTRACT FROM AUTHOR]

As the main pathogen threatening human and animal health, viruses can affect the immunity and metabolism of bodies. There are innate microbial barriers in the digestive tract of the body to preserve the homeostasis of the animal body, which directly or indirectly influences the host defence against viral infection. Understanding the interaction between viruses and intestinal microbiota or probiotics is helpful to study the pathogenesis of diseases. Here, we review recent studies on the interaction mechanism between intestinal microbiota and viruses. The interaction can be divided into two aspects: inhibition of viral infection by microbiota and promotion of viral infection by microbiota. Zearalenone (ZEN) is a severe contaminant mycotoxin found worldwide. Our previous results indicate that Bacillus velezensis A2 isolated from ZEN-contaminated crop soil can degrade ZEN. Here, we present the complete genomic sequence of B. velezensis A2 (the Genbank accession number: CP053717), which contains 3,929,218 bp in the chromosome, has a GC content of 46.5%, and contains the encoded ZEN-degrading enzyme gene. The aim of the present study was to investigate whether curcumin (CUR) can ameliorate cadmium-induced reproductive toxicity and its mechanism. A total of 48 male mice were equally divided into 4 groups: control, CdCl2 (2 mg/kg, intraperitoneally inject) curcumin (50 mg/kg, intraperitoneally inject), co-treatment with curcumin (50 mg/kg), and CdCl2 (2 mg/kg) for 10 days. The results demonstrated that CdCl2 reduces sperm motility, decreases the sperm density and serum testosterone content, and significantly improves the rate of sperm deformity. CdCl2 increased the level of testicular total superoxide dismutase (T-SOD), glutathione peroxidase (GSH-Px) activity, and glutathione (GSH), and CdCl2 declined the level of malondialdehyde (MDA). However, the semen quality of the mice in the curcumin intervention group was improved. Moreover, the testosterone content and antioxidant capacity were increased. In the Cd group mice, the expression of testicular Nrf2, as well as the mRNA and protein expressions of the downstream target molecules, glutathione peroxidase (GSH-Px), and γ-glutamylcysteine synthetase (γ-GCS) of Nrf2 declined, while the above genetic expressions elevated significantly in the curcumin intervention group. The mechanism of action of zearalenone (ZEA) in inducing germ cell tumors is unclear, and little is known about the change in the transcriptome of germ cells after ZEA exposure. To explore the molecular basis of the ZEA oncogene, we examined the median lethal concentration (50 μmol/L) and pro-apoptotic effect of ZEA on TM3 Leydig cells by MTT and TUNEL assay. Subsequently, we investigated the genetic changes in the transcriptome of TM3 Leydig cells exposed to 50 μmol/L ZEA. The transcriptome sequencing results show that 772 genes are significantly down-regulated, while 204 genes are significantly up-regulated. Gene ontology (GO) enrichment analysis shows that ZEA has a major effect on the connective function, cell composition, cell cycle, and energy metabolism of the TM3 Leydig cells. Using the results of the GO and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses, we select the Ras and Rap1 signaling pathways that are closely related to the occurrence of cancer. The differentially expressed genes visualized in the KEGG pathway were selected for RT-qPCR differential gene expression validation. The results show that the gene expression results are consistent with the transcriptome sequencing results. A total of 95 8-week-old male Kunming mice were randomly assigned into five groups and exposed to zearalenone (ZEA) at doses of 25, 50, and 75 mg/kg delivered by intra-peritoneal (i.p.) injection for 5 days. The testis and epididymis indices involving sperm quality and morphology, testis enzyme activities, serum concentrations of testosterone and estrogen, and the expression levels of the three gene and protein of N-cadherin, vimentin, and claudin 11 related to the blood testis barrier (BTB) were analyzed. Results showed that ZEA significantly decreased body weight and semen quality compared to the control group along with increased activity of alkaline phosphatase (ALP), acid phosphatase (ACP), lactate dehydrogenase (LDH), and reduced serum concentrations of testosterone and estrogen. At the mRNA and protein levels, expression of N-cadherin, vimentin, and claudin 11 significantly increased; however, the mRNA and protein of N-cad expression decreased. These data suggest acute exposure to ZEA reduces sperm quality and significantly decreases the concentration of serum testosterone and estradiol. In addition, the activities of the testis marker enzymes and associated mRNA and protein expressions of the BTB were also significantly affected. Our results demonstrated that ZEA has a significant impact on the reproductive parameters of male mice which showed compensatory response to strengthen the barrier function of the BTB following ZEA exposure. This study evaluated the protective effect of proanthocyanidin (PC) on the cytotoxicity of the Sertoli cell TM4 of mice, as induced by zearalenone (ZEA). Flow cytometry was used to detect the apoptosis rate of cells in each group. The activities of antioxidant enzymes and the content of antioxidant substances were detected by using a proprietary kit; the RT-PCR method was used to detect the expression level of mRNA, the related genes of Nrf2/ARE signal pathway, the nuclear factor E2 related factor 2 (Nrf2), heme oxygenase 1 (HO-1), glutathione peroxidase (GSH-Px), quinone oxidoreductase 1 (NQO1), γ-glutamylcysteine synthetase (γ-GCS) and the expression level of mRNA, the apoptosis-related genes, Bcl-2 and Bax; the Western-blot method was used to detect the protein expression levels of Nrf2, GSH-Px, HO-1, γ-GCS and NQO1 in each group. Our results showed that PC could reduce the apoptosis rate of the TM4 cells exposed to ZEA ( p < 0.01); PC could enhance the decrease in the activities of T-SOD and GSH-Px induced by ZEA ( p < 0.05), reduce the increase in the content of MDA, as caused by ZEA; PC could significantly up-regulate the down-regulation levels of the mRNA and protein of Nrf2, GSH-Px, HO-1, γ-GCS and NQO1 induced by ZEA. PC could enhance the decrease in the mRNA expression level of Bcl-2 and down-regulate the mRNA expression of Bax induced by ZEA ( p < 0.05). These results demonstrated that PC conferred protective effects against oxidative damage and apoptosis of TM4 cells induced by ZEA. Zearalenone (ZEN) exerts a major effect on human and animal health and has led to serious worldwide economic problems. In this study, we investigated whether proanthocyanidin (PC) can prevent ZEN-induced testicular oxidative damage in male mice and explored the underlying mechanism. Kunming mice were injected with ZEN (40 mg kg) on the 11th day after intragastric administration of PC (75 or 150 mg/kg) for 10 days; the sperm quality of mice was then analysed statistically. Additionally, testicular morphology parameters related to oxidative damage, apoptosis and the expression of endoplasmic reticulum (ER) stress-related genes (GRP78, CHOP and XBP-1) were all measured. Results showed that ZEN exposure significantly reduced the sperm density, improved the sperm aberration rate, increased the MDA level and reduced SOD and GSH-Px activities. Meanwhile, ZEN was attributed to the downregulation of the expressions of the gene and protein of Bcl-2 and upregulation of the expressions of the gene and protein of Bax and caspase-3. ZEN exposure also upregulated the mRNA expression of GRP78, CHOP and XBP-1; however, PC pre-treatment reduced ZEN-induced oxidative damage and tended to maintain normal testicular morphology. Furthermore, PC pre-treatment substantially downregulated the expressions of the GRP78, CHOP and XBP-1 and upregulated the expression of the Bcl-2 gene. In conclusion, PC, due to its anti-oxidative ability, could ameliorate ZEN-induced testicular reproductive toxicity in male mice by decreasing ER stress and testicular cell apoptosis. Healthy male Kunming mice received selenium yeast for 14 days prior to a single oral administration of zearalenone (ZEN). After 48 h, blood samples were collected for analysis and showed that mice in the ZEN-treated group has significantly decreased lymphocytes ( P < 0.05) and platelets ( P < 0.05) along with an increased white blood cell (WBC) count and other constituents ( P < 0.05). The serum biochemistry analysis of the ZEN group indicated that glutamic pyruvic transaminase (ALT), glutamic oxaloacetic transaminase (AST), urea, and uric acid were significantly increased ( P < 0.05), whilst total bilirubin (TB) and albumin (ALB) were decreased along with serum testosterone and estrogen ( P < 0. 05). The level of malondialdehyde (MDA) in the serum of the ZEN group was significantly increased whilst glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) had significantly decreased ( P < 0.05). Treatment with selenium yeast had a significant effect on response with most of the experimental parameters returning to levels similar to those observed in the untreated control mice. From these data, it can be concluded that ZEN is highly poisonous in Kunming mice with high levels of toxicity on the blood, liver, and kidneys. Objectives: To investigate the value of diffusion tensor imaging (DTI) and tractography in renal allografts at the early stage after kidney transplantation.Methods: This study was approved by the institutional ethical review committee, and written informed consent was obtained. A total of 54 renal allograft recipients 2-3 weeks after transplantation and 26 age-matched healthy volunteers underwent renal DTI with a 3.0-T magnetic resonance imaging (MRI) system. Recipients were divided into three groups according to the estimated glomerular filtration rate (eGFR). Apparent diffusion coefficient (ADC) and fractional anisotropy (FA) of the cortex and medulla were measured and compared among the groups. Whole-kidney tractography was performed. Correlation of eGFR with diffusion parameters was evaluated.Results: In allografts with stable function, the medullary ADC was higher and the cortical FA was lower (p < 0.001) than in healthy kidneys. The cortical ADC, medullary ADC and FA decreased as the allograft function declined, and with a positive correlation with eGFR (p < 0.001); cortical FA did not. The major role of insulin-like growth factor-1 (IGF-1) in the liver is to mediate glucose uptake in hepatocytes to synthesize glycogen and maintain blood glucose homeostasis. In this study, to evaluate the role of IGF-1 on gluconeogenesis and nutrient metabolism in dairy cattle, pyruvate carboxylase (PC) and phosphoenolpyruvate carboxykinase (PEPCK) expression and enzyme activity were evaluated in primary cultures of bovine hepatocytes treated with different concentrations of IGF-1 by quantitative polymerase chain reaction and spectrophotometry, respectively. The results showed that expression of PC and PEPCK were significantly lower in bovine hepatocytes by IGF-1 treatment in test group compare to the control group ( P < 0.01). As IGF-1 concentration increased, PC and PEPCK enzyme activity in bovine hepatocytes decreased. Evaluating PC and PEPCK mRNA levels and enzyme activity may thus be useful to monitor subclinical ketosis in dairy cows. [ABSTRACT FROM AUTHOR]

The δ18O data obtained from an 18.7 m ice core drilled in Chongce Ice Cap at an elevation of 6,530 m a.s.l. in the West Kunlun Mountains on the northern Tibetan Plateau show a strong correlation with the summer temperature of the middle to upper troposphere over the mid-high latitudes of Eurasia. Based on this, the δ18O record can be used as a proxy of the June–September mean temperature of the mid-upper troposphere (MUT) from 1903 to 1992. The time span of the ice core record is much longer than the meteorological data available only after 1948. Using the empirical mode decomposition method (EMD), the δ18O record is decomposed into various frequency components and compared with the solar irradiance variations of the same period. The results show that (1) The June–September mean temperature of the MUT over Eurasian mid-high latitudes is completely decomposed into four IMF (intrinsic mode function) components and an increasing trend. (2) Solar irradiance is decomposed into the Schwabe cycle, the Hale cycle, the Gleissberg cycle, and an increasing trend. (3) The correlation coefficients between the June and September mean temperatures of the MUT over Eurasian mid-high latitudes and solar irradiance on the longer timescales (at least more than 11-year) show the significant correlations; their phase changes are basically identical in general, and (4) the 11-year Schwabe cycle exists in the June–September mean temperature of the MUT over Eurasian mid-high latitudes during most of the time from 1903 to 1992, and only in the two high-temperature phases (1929–1944 and from 1975 to the present) may global warming disturb this relation. Sand-dust storm is a special natural disaster that frequently occurs in deserts and their surrounding areas. With the data published on Surface Meteorological Monthly Bulletin and Surface Chart during 1971–1996, the temporal-spatial distribution and annual variation of sand-dust storms are analyzed on the basis of the case study of atmospheric processes. Furthermore, the tracks and source areas of sand-dust storms are determined with the aid of GIS. The results show that except some parts of Qinghai Province and Inner Mongolia as well as Beijing, sand-dust storms decrease apparently in time and space in recent decades in China. Sand-dust storms occur most frequently in spring, especially in April. According to their source areas, sand-dust storms are classified into two types, i.e., the inner-source and outer-source sand-dust storms. Most of the outer-source sand-dust storms move along the north and west tracks. The north-track outer-source sand-dust storms always intrude into China across the Sino-Mongolian border from Hami, a city in the eastern part ofXinjiang, to Xilin Gol, a league in Inner Mongolia, while the west-track ones intrude into China from both southern and northern Xinjiang. The source lands of inner-source sand-dust storms concentrate in the Taklimakan Desert and its surrounding areas in southern Xinjiang, southern part of the Junggar Basin in north of Xinjiang, the Hexi Corridor in western Gansu Province, the dry deserts of Inner Mongolia and the Qaidam Basin in Qinghai.
