The influence of chimeric antigen receptor T (CAR-T) cell therapy on platelet function in relapsed/refractory (R/R) multiple myeloma (MM) has not been thoroughly investigated. Our cohort comprised fifty MM patients treated with CAR-T cells. The mean platelet closure time (PCT) induced by collagen/adenosine diphosphate (CADP) in peripheral blood was significantly prolonged before lymphodepletion (195.24 ± 11.740 s) and notably reduced post-CAR-T cell therapy (128.02 ± 5.60 s), with a statistically significant improvement (67.22, 95% CI 46.91–87.53, P < 0.001). This post-treatment PCT was not significantly different from that of healthy controls (10.64, 95% CI 1.11–22.40, P > 0.05). Furthermore, a pronounced enhancement in PCT was observed in patients with a response greater than partial remission (PR) following CAR-T cell infusion compared to pre-treatment values (P < 0.001). An extended PCT was also associated with a less favorable remission status. In patients with cytokine release syndrome (CRS) grades 0–2, those with a PCT over 240.5 s exhibited a shorter progression-free survival (PFS), with median PFS times of 10.2 months for the PCT > 240.5 s group versus 22.0 months for the PCT ≤ 240.5 s group. Multivariate analysis revealed that a PCT value exceeding 240.5 s is an independent prognostic factor for overall survival (OS) in R/R MM patients after CAR-T cell therapy. The study demonstrates that CAR-T cell therapy enhances platelet function in R/R MM patients, and PCT emerges as a potential prognostic biomarker for the efficacy of CAR-T cell therapy. Few studies have reported the associations of granulocyte colony-stimulating factor (G-CSF) with cytokine release syndrome (CRS), neurotoxic events (NEs) and efficacy after chimeric antigen receptor (CAR) T-cell therapy for relapsed or refractory (R/R) B-cell acute lymphoblastic leukemia (B-ALL). We present a retrospective study of 67 patients with R/R B-ALL who received anti-CD19 CAR T-cell therapy, 41 (61.2%) patients received G-CSF (G-CSF group), while 26 (38.8%) did not (non-G-CSF group). Patients had similar duration of grade 3–4 neutropenia between the two groups. The incidences of CRS and NEs were higher in G-CSF group, while no differences in severity were found. Further stratified analysis showed that the incidence and severity of CRS were not associated with G-CSF administration in patients with low bone marrow (BM) tumor burden. None of the patients with low BM tumor burden developed NEs. However, there was a significant increase in the incidence of CRS after G-CSF administration in patients with high BM tumor burden. The duration of CRS in patients who used G-CSF was longer. There were no significant differences in response rates at 1 and 3 months after CAR T-cell infusion, as well as overall survival (OS) between the two groups. In conclusion, our results showed that G-CSF administration was not associated with the incidence or severity of CRS in patients with low BM tumor burden, but the incidence of CRS was higher after G-CSF administration in patients with high BM tumor burden. Despite the high therapeutic response achieved with B-cell maturation antigen (BCMA)-specific chimeric antigen receptor (CAR) T-cell therapy in relapsed and refractory multiple myeloma (R/R MM), primary resistance and relapse exist with single-target immunotherapy. Here, we design bispecific BC19 CAR T cells targeting BCMA/CD19 and evaluate antimyeloma activity in vitro and in vivo. Preclinical results indicate that BC19 CAR specifically recognize target antigens, and BC19 CAR T cells mediate selective killing of BCMA or CD19-positive cancer cells. BC19 CAR T cells also exhibit potent antigen-specific anti-tumor activity in xenograft mouse models. We conduct an open-label, single-arm, phase I/II study of BC19 CAR T cells in 50 patients with R/R MM (ChiCTR2000033567). The primary endpoint was safety. BC19 CAR T cells are well tolerated with grade 3 or higher cytokine release syndrome in 8% of patients and grade 1 neurotoxic events in 4% of patients, which meet the pre-specified primary endpoint. Secondary endpoints include overall response rate (92%), median progression-free survival (19.7 months), median overall survival (19.7 months) and median duration of response (not reached). Our study demonstrates that bispecific BC19 CAR T cells are feasible, safe and effective in treating patients with R/R MM. CAR-T cell therapies targeting BCMA have shown promising responses in patients with multiple myeloma (MM), however primary resistance and relapse are frequently observed. Smart materials that integrate multi-stimuli response, full reversibility, and dual-visual read-out channel are highly desired for anti-counterfeiting and information encryption applications. Herein, we developed a multiresponsive perchlorate terpyridyl Pt(II) nano complex which could undergo fully reversible conversion between three forms stimulated by water or formaldehyde molecule due to the extent of Pt–Pt interaction. Meanwhile, a dual-visual channel, i.e., the colorimetric channel changed from yellow to orange or red and the corresponding luminescent channel from orange to orange-red or red, has also been found. The weak and equivalent strength of ion-dipole interaction and hydrogen bond that generated between formaldehyde/water and Pt(II) salt result in the easy-control reversibility between the three forms. Furthermore, by introducing different polymer matrices, 1Cl·ClO4@PMMA (1Cl·ClO4: [Pt(tpy)Cl]·ClO4, tpy: 2,2′:6′,2″-terpyridine), PMMA: poly(methyl methacrylate)) and 1Cl·ClO4@PVA (PVA: polyvinyl alcohol) are successfully constructed, which exhibit different reversible behaviors since the PMMA and PVA matrix exert different influences on the strength of hydrogen-bond. Phytoremediation is considered an effective method for indoor air pollution control. The removal rate and mechanism of benzene in air by two plants, Tradescantia zebrina Bosse and Epipremnum aureum (Linden ex André) G. S. Bunting, were investigated through fumigation experiments under the condition of plant hydroponics culturing. Results showed that the plant removal rates increased with increase in benzene concentration in air. When the benzene concentration in air was set at 432.25–1314.75 mg·m−3, the removal rates of T. zebrina and E. aureum ranged from 23.05 ± 3.07 to 57.42 ± 8.28 mg·kg–1·h–1 FW and from 18.82 ± 3.73 to 101.58 ± 21.20 mg·kg–1·h–1 FW, respectively. The removal capacity was positively related to the transpiration rate of plants, indicating that gas exchange rate could be a key factor for the evaluation of removal capacity. There existed fast reversible transport of benzene on air-shoot interface and root-solution interface. After shoot exposure to benzene for 1 h, downward transport was the dominant mechanism in the removal of benzene in air by T. zebrina, while in vivo fixation was the dominant mechanism at exposure time of 3 and 8 h. Within 1–8 h of shoot exposure time, in vivo fixation capacity was always the key factor affecting the removal rate of benzene in the air by E. aureum. Contribution ratio of in vivo fixation in the total benzene removal rate increased from 6.29 to 92.29% for T. zebrina and from 73.22 to 98.42% for E. aureum in the experimental conditions. Reactive oxygen species (ROS) burst induced by benzene exposure was responsible for the contribution ratio change of different mechanisms in the total removal rate, which also was verified by the change of activities of antioxidant enzymes (CAT, POD, and SOD). Transpiration rate and antioxidant enzyme activity could be considered parameters to evaluate the plant removal ability to benzene and to screen plants for establishment of plant-microbe combination technology. Response surface methodology (RSM) was used to optimize the oxidizing the omeprazole sulfide to (S)-omeprazole catalyzed by environmentally friendly catalyst soybean pod peroxidase (SPP) in cetyltrimethylammonium bromide (CTAB)/isooctane/n-butyl alcohol/water water-in-oil microemulsions. With the initial concentration of SPP of 3200 U ml−1, the conversion of the omeprazole sulfide, the (S)-omeprazole yield and ee were 93.75%, 91.56% and 96.08%, respectively, under the optimal conditions: Wo of 15.85, the concentration of H2O2 of 22.44 mM and reaction temperature of 49.68 °C, respectively. The proposed mechanism of asymmetric sulfoxidations catalyzed by SPP involves three concomitant mechanisms as follows: (1) a two-electron reduction of SPP-I, (2) a single-electron transfer to SPP-I and (3) nonenzymatic reactions. Based on the proposed mechanism which is reasonable and can express the oxidations, the reaction system includes five enzymatic and two nonenzymatic reactions. With 5.44% of the average relative error, a kinetic model based on the mechanisms fitting observed data very well was established, and the SPP-catalyzed reactions including both the two-electron reduction and the single-electron transfer mechanisms obey ping-pong mechanism with substrate and product inhibition, while nonenzymatic reactions follow a power law. (Ti,W)C-based cement tool materials with c-BN particles as additive phase are fabricated by the hot press sintering. The effect of c-BN addition on the microstructure and mechanical properties of (Ti,W)C-based cement tool materials are investigated. With the addition of c-BN, the fracture mode of composite transform from intergranular to transgranular fracture. The main toughening mechanism is particle bridging, crack deflection and crack bifurcation, which improve the fracture toughness of cement tool material. The proper addition of c-BN can improve the mechanical properties of the composites. When the content of c-BN is 1.5 wt %, (Ti,W)C-based cement tool materials reaches the optimum comprehensive mechanical properties. (S)-Omeprazole is a very effective anti-ulcer medicine that is difficult to be prepared using whole cells at elevated substrate concentrations. In the chloroform–water biphasic system, resting cells of the mutant QZ-3 of Rhodococcus rhodochrous (R. rhodochrous) ATCC 4276 were used to catalyze the bio-oxidation of omeprazole sulfide for preparation of (S)-omeprazole. Using response surface methodology (RSM), the reaction was optimized to work at a substrate concentration of 180 mM and a cell concentration of 100 g/L. The optimal yield of (S)-omeprazole obtained was 92.9% with enantiomeric excess (ee) (> 99%), and no sulfone by-product was detected under the optimal working conditions; reaction temperature 37 °C, pH 7.3 and reaction time, 43 h. A quadratic polynomial model was established, which predicts the experimental data with very high accuracy (R2 = 0.9990). The chloroform–water biphasic system may contribute to the significant improvement in substrate tolerance because almost all substrates are partitioned in the organic phase (water solubility of omeprazole sulfide is only about 0.5 mg/mL), resulting in little damage and inhibition to cells by substrates. The mutant QZ-3 of R. rhodochrous ATCC 4276 exhibited high enantioselectivity, activity and substrate and product tolerance. Enantiopure esomeprazole is an important drug in the treatment of gastric ulcer. The asymmetric sulfoxidation of omeprazole thioether was catalyzed by immobilized cells of a mutant of Rhodococcus rhodocrous ATCC 4276 to synthesize esomeprazole. The bioreaction was carried out in a biphasic system (chloroform-water), at a high substrate concentration (200 mM), and optimized using response surface methodology (RSM). The optimal yield of esomeprazole obtained was 94.8% with e.e. (>99%) without the formation of the sulfone form as a byproduct, under the optimal conditions: the concentration of immobilized cells, 283.5 g/L, the incubation temperature, 37.05 °C, and pH of phosphate buffer, 7.35, respectively. A quadratic polynomial model was developed with R2 of 0.9998, which indicates that the model predicts the observed data with very high accuracy. The mutant exhibited a high enantioselective activity and substrate and product tolerance. The small size of immobilized cell beads (0.5–1 mm) creates a large reaction interface. The aerated flask provides enough oxygen for a high concentration of cells. The significant improvement of substrate tolerance may mainly be attributed to employing the chloroform-water biphasic system because organic substrates may be partitioned in the organic phase, eliminating potential damage and inhibition to cells. The formaldehyde dehydrogenase (FADH) activity in plants is essential to the removal of airborne formaldehyde (FA) by plants. A rapid and efficient method was established to assess the FADH activity in plants by analyzing the efficiencies of the extracts of fresh and enzyme-inactivated leaves to degrade FA, with the enzyme-inactivated leaves prepared by freezing with liquid nitrogen. The efficiencies of airborne FA dissipated by different plants were evaluated through the FA fumigation experiments using four selected plants, with the results analyzed against the calculated leaf FADH activities. Fresh and enzyme-inactivated leaf extracts degraded FA to different extents. The degradative efficiencies of leaf extracts were positively related to the initial FA test levels at 6–18 mg l−1. The relative plant-leaf FADH activities formed the order of Chenopodium album L. > Atenia cordifolia > Plantain > Aloe, which was in line with the observed FA dissipating efficiencies of the plants exposed to 0.72 mg m−3 airborne FA for 24 h. Other dominant degrading mechanisms in plant leaves resulted in higher dissipating efficiencies of Plantain over that of Atenia cordifolia when exposed to 1.56 mg m−3 FA for 24 h. This paper is devoted to the Lp (p > 1) solutions of one-dimensional backward stochastic differential equations (BSDEs for short) with general time intervals and generators satisfying some non-uniform conditions in t and ω. An existence and uniqueness result, a comparison theorem and an existence result for the minimal solutions are respectively obtained, which considerably improve some known works. Some classical techniques used to deal with the existence and uniqueness of Lp (p > 1) solutions of BSDEs with Lipschitz or linear-growth generators are also developed in this paper. G-CSF is the most often used agent in clinical hematopoietic stem and progenitor cell (HSPC) mobilization. However, in about 10 % of patients, G-CSF does not efficiently mobilize HSPC in clinically sufficient amounts. Cdc42 activity is involved in HSPC mobilization. In the present study, we explore the impact of Cdc42 inhibitor ML141 on G-CSF-mediated HSPC mobilization in mice. We found that the use of ML141 alone only triggered modest HSPC mobilization effect in mice. However, combination of G-CSF and ML141 significantly promoted HPSC counts and colony forming units in peripheral blood, as compared to mice treated with G-CSF alone. ML141 did not significantly alter the levels of SDF-1 and MMP-9 in the bone marrow, when used alone or in combination with G-CSF. We also found that G-CSF administration significantly increases the level of GTP-bound Cdc42, but does not alter the expression of Cdc42 in the bone marrow. Our data indicate that the Cdc42 signal is a negative regulator in G-CSF-mediated HSPC mobilization, and that inhibition of the Cdc42 signal efficiently improves mobilization efficiency. Thymol is a natural monoterpene phenol primarily found in thyme, oregano, and tangerine peel. It has been shown to possess anti-inflammatory property both in vivo and in vitro. In the present paper, we studied the anti-inflammatory effect of thymol in lipopolysaccharide (LPS)-stimulated mouse mammary epithelial cells (mMECs). The mMECs were stimulated with LPS in the presence or absence of thymol (10, 20, 40 μg/mL). The concentrations of tumor necrosis factor α (TNF-α), interleukin (IL)-6, and IL-1β in the supernatants of culture were determined using enzyme-linked immunosorbent assay. Cyclooxygenase-2 (COX-2), inducible nitric oxide synthase (iNOS), extracellular signal-regulated protein kinase (ERK), c-Jun N-terminal kinase (JNK), nuclear factor-κB (NF-κB), and inhibitor protein of NF-κB (IκBα) were measured using western blot. The results showed that thymol markedly inhibited the production of TNF-α and IL-6 in LPS-stimulated mMECs. The expression of iNOS and COX-2 was also suppressed by thymol in a dose-dependent manner. Furthermore, thymol blocked the phosphorylation of IκBα, NF-κB p65, ERK, JNK, and p38 mitogen-activated protein kinases (MAPKs) in LPS-stimulated mMECs. These results indicate that thymol exerted anti-inflammatory property in LPS-stimulated mMECs by interfering the activation of NF-κB and MAPK signaling pathways. RP105 is a member of the toll-like receptor family of proteins that transmits an activation signal in B cells, playing a role in regulation of B cell growth and death; in macrophages and dendritic cells, RP105 is a specific inhibitor of TLR4 signaling. RP105 is uniquely important for regulating TLR4-dependent signaling. It also proved that RP105 is closely related to TLR2 in macrophage activation by Mycobacterium tuberculosis lipoproteins. The aim of our study is to investigate the role of RP105 in mouse macrophages activation of TLR4 and TLR2 signaling by lipopolysaccharides (LPS) and Pam3CysSerLys4 (Pam3CSK4) alone or in combination, and the interaction between TLR2 and TLR4 signaling through RP105. Our results indicate that besides exhibiting negative regulation of TNF-α and IL12-p40 secretion in macrophage activated by LPS, RP105 is also involved in macrophages activation by Pam3CSK4 through TLR2 signaling and exhibited regulation to IL-10 and RANTES production by mouse peritoneal macrophage activated by Pam3CSK4. In macrophages activation by LPS and Pam3CSK4 in combination, TLR2 signaling can overcome RP105-mediated regulation of TLR4 signaling. Thus, our data demonstrate that not only TLR4 signaling, but also RP105 appears to be an essential accessory for immune responses through TLR2 signaling. The function of TLR2 and TLR4 in response to TLR ligands could be associated with each other by RP105. Background and objective: Mastitis is defined as inflammation of the mammary gland in domestic dairy animals and humans. Salidroside, a major component isolated from Rhodiola rosea L., has potent anti-inflammatory properties, but whether it can be used in mastitis treatment has not yet been investigated. The aim of this study was to assess the protective effects of salidroside against lipopolysaccharide (LPS)-induced mastitis in mice and the mechanism of action. Methods and results: We used a mouse mastitis model in which mammary gland inflammation was induced by LPS challenge. Salidroside administered 1 h before LPS infusion significantly attenuated inflammatory cell infiltration, reduced the activity of myeloperoxidase in mammary tissue, and decreased the concentration of tumor necrosis factor-α, interleukin (IL)-1β, and IL-6 in a dose-dependent manner. Further studies revealed that salidroside down-regulated phosphorylation of LPS-induced nuclear transcription factor-kappaB (NF-κB) p65 and inhibitor of NF-κB α (IκBα) in the NF-κB signal pathway, and suppressed phosphorylation of p38, extracellular signal-regulated kinase (ERK) and c-jun NH-terminal kinase (JNK) in MAPKs signal pathways. To explore whether the graft-versus-leukemia (GVL) effects could be enhanced and acute graft-versus-host disease (aGVHD) could be relieved by syngeneic bone marrow mixed with G-CSF-primed H-2 haploidentical marrow grafting. Female L615 (H-2k) mice were recipient mice and male (615 × BALB/c) F1 (6BF1) (H-2k × H-2d) mice were donors respectively. Donor mice were injected subcutaneously with G-CSF daily at 0.01 μg/g for 6 days, and splenocytes were harvested on day 7. A total of 615 mice were loaded with L615 tumor cells and received 8.5 Gy (60Co γ-ray) irradiation three days later, followed by a mixed bone marrow transplantation (MBMT). The allo-grafts consisted of a mixture of syngenetic plus G-CSF-mobilized (control diluents) H-2 haploidetical marrow cells. GVL effects were monitored by survival time and survival rate of recipient mice. GVHD was assessed by clinical signs of weight loss, ruffled fur, diarrhea and histological changes of skin, liver and small intestines. Allogeneic chimerism was detected using cytogenetic analysis. Enzyme-linked immunosorbent assay (ELISA) method was used to detect cytokines (IL-2, IL-4 and IFN-γ). Fluorescence-activated cell sorting (FACS) analysis was used to detect T-cell phenotype. (1) The mice merely received L615 leukemia cells were all died of leukemia. The L615 mice of 3:1 and 4:1 MBMT groups survived longer than those post syngeneic BMT ( P < 0.01). (2) The survival time of mice in the G-CSF-treated MBMT groups was longer than that of non-primed MBMT groups ( P < 0.05). Administration of G-CSF-treated 6BF1 mice could markedly increase the survival rate of 3:1 and 4:1 MBMT mice ( P < 0.01) with little or a little GVHD. (3) As the post-transplanted time prolonged, the rates of allogeneic chimerism were decreased. The chimerism rates decreased to zero when the mice died of leukemia relapse. (4) L3T4+ cells and relative ratio in both subsets were significantly reduced in G-CSF-treated donor mice. After G-CSF-treated donors, splenocytes from recipients at day 14 post-MBMT showed an increased production of IL-4 and a decreased production of IL-2 and IFN-γ. Syngeneic bone marrow mixed with H-2 haploidentical marrow grafts had a potential way to increase GVL effects, and this GVL effects could be enhanced with little or a little GVHD by G-CSF pretreatment of donors. How to reduce the incidence and severity of acute graft-versus-host disease (aGVHD) is a crucial step to improve the overall survival of allogeneic bone marrow transplantation (allo-BMT). The low incidence of severe aGVHD observed in allogeneic peripheral blood stem cell transplantation (allo-PBSCT), which may be related to modulating immune function of T lymphocytes by granulocyte colony-stimulating factor (G-CSF) primed donors. The study aimed to explore whether aGVHD could be alleviated by syngeneic bone marrow mixed with G-CSF-mobilized H-2 haploidentical marrow grafting. Female BALB/c mice and neonatal BALB/c mice were recipients and male (BALB/c × C57BL/6)F1(BCF1) mice were donor mice respectively. Donor mice were injected subcutaneously with G-CSF daily at 0.01 μg/g body weight or saline for 6 days, and splenocytes were harvested on day 6. Spleen index (SI) represented GVHD in neonatal mice after the intraperitoneal injection of mixed spleen cells. Lethally irradiated (60Co, 8.5 Gy) adult mice were transplanted with a mixture of syngeneic plus G-CSF-mobilized (control diluents) H-2 haploidentical marrow cells. Survival time and survival rate of the recipients were observed after mixed marrow transplantation (MBMT). GVHD was assessed by observing signs of weight loss, ruffled fur, diarrhea and histological change of skin, liver and small intestines. Enzyme-linked immunosorbent assay (ELISA) method was used to detect cytokines (IL-2, IL-4 and INF-γ). Fluorescence-activated cell sorting (FACS) analysis was used to detect T cells phenotype. (1) The neonatal mice subject to injection of 2:1 and 1:1 mixed spleen cells and H-2 haploidentical spleen cells all suffered from aGVHD. The severity of aGVHD in recipient mice receiving G-CSF-mobilized splenocytes was dramatically reduced. (2) The aGVHD signs and histological change were observed in most mice of 2:1 and 1:1 MBMT groups. However, the survival time of G-CSF-mobilized MBMT was longer than in control groups and these mice had signs of moderate GVHD. (3) L3T4 + cells and relative ratio in both subsets was significantly reduced in G-CSF-treated donor mice. The total number of Thy1.2 and lyt2+ cells was increased after G-CSF pretreatment of donors, but no statistical difference. (4) The supernatants from a primary MLR were collected at 48 h for cytokine measurement. The results showed an increased production of IL-4 and a decreased production of IL-2 and INF-γ after stimulating with concanavalin A for 48 h.
