Abstract We have recently identified the uncharacterized ZNF555 protein as a component of a productive complex involved in the morbid function of the 4qA locus in facioscapulohumeral dystrophy. Subsequently named DiPRO1 (Death, Differentiation, and PROliferation related PROtein 1), our study provides substantial evidence of its role in the differentiation and proliferation of human myoblasts. DiPRO1 operates through the regulatory binding regions of SIX1, a master regulator of myogenesis. Its relevance extends to mesenchymal tumors, such as rhabdomyosarcoma (RMS) and Ewing sarcoma, where DiPRO1 acts as a repressor via the epigenetic regulators TIF1B and UHRF1, maintaining methylation of cis-regulatory elements and gene promoters. Loss of DiPRO1 mimics the host defense response to virus, awakening retrotransposable repeats and the ZNF/KZFP gene family. This enables the eradication of cancer cells, reprogramming the cellular decision balance towards inflammation and/or apoptosis by controlling TNF-α via NF-kappaB signaling. Finally, our results highlight the vulnerability of mesenchymal cancer tumors to si/shDiPRO1-based nanomedicines, positioning DiPRO1 as a potential therapeutic target.

Abstract Our current understanding of the spectrum of TB and COVID‐19 lesions in the human lung is limited by a reliance on low‐resolution imaging platforms that cannot provide accurate 3D representations of lesion types within the context of the whole lung. To characterize TB and COVID‐19 lesions in 3D, we applied micro/nanocomputed tomography to surgically resected, postmortem, and paraffin‐embedded human lung tissue. We define a spectrum of TB pathologies, including cavitary lesions, calcium deposits outside and inside necrotic granulomas and mycetomas, and vascular rearrangement. We identified an unusual spatial arrangement of vasculature within an entire COVID‐19 lobe, and 3D segmentation of blood vessels revealed microangiopathy associated with hemorrhage. Notably, segmentation of pathological anomalies reveals hidden pathological structures that might otherwise be disregarded, demonstrating a powerful method to visualize pathologies in 3D in TB lung tissue and whole COVID‐19 lobes. These findings provide unexpected new insight into the spatial organization of the spectrum of TB and COVID‐19 lesions within the framework of the entire lung.

There is limited information on utilizing commercially supplied [18F]Fluoride from an off-site cyclotron for the synthesis of radiopharmaceuticals. This study explored the production of the PET hypoxia marker [18F]FMISO, using the Scintomics® GRP module and distantly produced [18F]Fluoride, which lacks published data. A radiochemical yield of 27.4 ± 3.6% (n = 5) and high radiochemical purity were achieved in synthesis time of 48 min. The [18F]FMISO met all Ph. Eur. standards, demonstrating the consistency and reliability of the module in generating a clinical-grade radiopharmaceutical using off-site produced [18F]Fluoride, but available patient doses were severely limited. Deep neural networks (DNNs) are fundamental to modern applications like face recognition and autonomous driving. However, their security is a significant concern due to various integrity risks, such as backdoor attacks. In these attacks, compromised training data introduce malicious behaviors into the DNN, which can be exploited during inference or deployment. This paper presents a novel game-theoretic approach to model the interactions between an attacker and a defender in the context of a DNN backdoor attack. The contribution of this approach is multifaceted. First, it models the interaction between the attacker and the defender using a game-theoretic framework. Second, it designs a utility function that captures the objectives of both parties, integrating clean data accuracy and attack success rate. Third, it reduces the game model to a two-player zero-sum game, allowing for the identification of Nash equilibrium points through linear programming and a thorough analysis of equilibrium strategies. Additionally, the framework provides varying levels of flexibility regarding the control afforded to each player, thereby representing a range of real-world scenarios. Through extensive numerical simulations, the paper demonstrates the validity of the proposed framework and identifies insightful equilibrium points that guide both players in following their optimal strategies under different assumptions. The results indicate that fully using attack or defense capabilities is not always the optimal strategy for either party. Instead, attackers must balance inducing errors and minimizing the information conveyed to the defender, while defenders should focus on minimizing attack risks while preserving benign sample performance. These findings underscore the effectiveness and versatility of the proposed approach, showcasing optimal strategies across different game scenarios and highlighting its potential to enhance DNN security against backdoor attacks. Background: Constipation is prevalent after bariatric surgery and glucagon-like-peptide 1 (GLP-1) analogues. Increasing fat content in the distal small intestine and colon can enhance colonic peristalsis, potentially alleviating symptoms of constipation. Aim: We investigated whether oleic acid can ameliorate constipation in patients undergoing bariatric surgery or receiving GLP-1 analogues. Methodology: Fourteen adults with chronic constipation according to Rome IV criteria following bariatric surgery or GLP-1 analogues were on stable treatment for constipation for more than 4 weeks. This randomized double-blind crossover trial compared microcapsules containing 21.25 g of oleic acid delivered in the distal small intestine or the stomach. The primary outcome was changed in the number of bowel motions over 24 h. Exploratory endpoints included alterations in straining, diarrhoea, faecal leakage over 24 h and hunger, fullness, nausea and calorie intake for the 3 h after ingesting the microcapsules. Findings: Receiving oleic acid into the distal small intestine increased number of bowel movements per day (2.5 vs 1.1, p = 0.009) and caused softer stool consistency (p = 0.03). 9/14 of the control group passed motions and 13/14 of the intervention group passed motions in 24 h (p = 0.059). No significant differences were observed in straining (p = 0.65), rapid bowel movements (p = 0.08), accidental leakage (p = 0.32), hunger, fullness, nausea or food intake between the groups (all p > 0.05). There were no disparities in safety profile between groups. Conclusion: Microcapsules containing oleic acid delivered to the distal small intestine appear to be a safe and effective relief from chronic constipation in patients undergoing bariatric surgery and/or receiving GLP-1 analogues. Mountainous regions typically harbour high plant diversity but are also characterised by low sampling intensity. Coarse-scale species distribution models can provide insights into the distribution of poorly sampled species, but the required bioclimatic data are often limited in these landscapes. In comparison, several environmental factors that vary over relatively fine scales in mountain environments (e.g. measures of topography) can be quantified from remotely-sensed data, and can potentially provide direct and indirect measures of biologically-relevant habitat characteristics in mountains. Therefore, in this study, we combine field-sampled floristic data with environmental predictors derived from remotely-sensed data, to model the ecological niches of 19 montane plant species in the Maloti-Drakensberg mountains, South Africa. The resulting models varied considerably in their performance, and species showed generally inconsistent responses to environmental predictors, with altitude and distance to watershed being most frequently included in models. These results highlight the species-specificity of the forb species' environmental tolerances and requirements, suggesting that environmental change may result in re-shuffling of community composition, instead of intact communities shifting along gradients. Furthermore, while the relatively high importance of altitude (a proxy for temperature) and topographic wetness index (a proxy for soil moisture) suggest that the flora of this region will be sensitive to shifts in temperature and rainfall patterns, several non-climatic environmental variables were also influential. Our findings indicate that local response to climate change in mountains might be especially constrained by soil type and topographic variables, supporting the important influence of non-climatic factors in microclimatic refugia dynamics. High-grade (> 10 g/t) gold mineralization in orogenic gold deposits is of significant economic importance. Understanding the formation of such enriched ore zones is critical for gold exploration success. The world-class Jundee-Bogada gold camp in the Yilgarn Craton of Western Australia comprises both high-grade (avg. > 10 g/t, Jundee deposit) and low-grade (avg. < 3 g/t, Bogada prospect) lodes, despite shared host stratigraphy. The paragenetic framework established for the Jundee gold deposit suggests that the overall gold endowment developed over three deformation events. An early episode of low-grade gold mineralization is associated with colloform-crustiform veins that formed during extensional deformation (DJB2A). A switch to transtensional deformation (DJB2B) resulted in brecciation of the colloform-crustiform veins and coeval deposition of native gold. Late reverse faults record evidence for a third mineralization stage resulting from a NE-SW-directed shortening (DJB3). Mineralization during this late stage was dominantly low-grade, with local occurrences of ultra-high-grade ore zones (> 100 g/t). Each event records transient changes in fluid chemistry during continued hydrothermal activity that spanned local deformation histories. We argue that at the Jundee gold deposit, protracted gold enrichment during three polyphased mineralization episodes resulted in the formation of high-grade gold ores. Whereas the complete metallogenic history is recorded at the Jundee deposit, gold within the Bogada prospect was introduced solely during the late contractional stage (DJB3), resulting in a bulk low-grade endowment. We hypothesize that gold enrichment in high-grade orogenic gold deposits is a direct consequence of the spatial superimposition of protracted ore-forming events. [ABSTRACT FROM AUTHOR]

In hospitalized children, height should be measured. When world health organization (WHO) height measurement gold standards is impossible, the ideal height estimation technique is still unclear. We conducted an international prospective study in eight different pediatric intensive care units to assess the accuracy, precision, practicability, safety, and inter-rater reliability of 12 different height estimation techniques, based on body segment measurement extrapolation, or other calculations using previous or projected heights. All extrapolation techniques were performed on each child, and later compared to their WHO gold standard heights. A total of 476 patients were enrolled. In the < 2-year subgroup, board length use and growth chart extrapolation performed best. In the ≥ 2-year subgroup, growth chart extrapolation and parents' report were the most accurate, followed by height measurement alongside the body with a tape measure. In both groups, body segment extrapolations were poorly predictive and showed mean bias and limits of agreement that varied a lot with age. Most body segment-based techniques presented with frequent measurement difficulties, but children's safety was rarely compromised. The inter-rater reliability of body segment measurement was low in the < 2-year subgroup. Conclusions: To accurately estimate height in hospitalized children, health care professionals should integrate the accuracy, precision, practicability, and reliability of each measurement technique to select the most appropriate one. Body segment-based techniques were the least accurate and should probably not be used. Simple techniques like growth chart extrapolation, or measurement alongside the body (and length board measurement in the youngest) should be implemented in daily practice. The TAR DNA Binding Protein 43 (TDP-43) has been implicated in the pathogenesis of human neurodegenerative diseases and exhibits hallmark neuropathology in amyotrophic lateral sclerosis (ALS). Here, we explore its tractability as a plasma biomarker of disease and describe its localization and possible functions in the cytosol of platelets. Novel TDP-43 immunoassays were developed on three different technical platforms and qualified for specificity, signal-to-noise ratio, detection range, variation, spike recovery and dilution linearity in human plasma samples. Surprisingly, implementation of these assays demonstrated that biobank-archived plasma samples yielded considerable heterogeneity in TDP-43 levels. Importantly, subsequent investigation attributed these differences to variable platelet recovery. Fractionations of fresh blood revealed that ≥ 95% of the TDP-43 in platelet-containing plasma was compartmentalized within the platelet cytosol. We reasoned that this highly concentrated source of TDP-43 comprised an interesting substrate for biochemical analyses. Additional characterization of platelets revealed the presence of the disease-associated phosphoserine 409/410 TDP-43 proteoform and many neuron- and astrocyte-expressed TDP-43 mRNA targets. Considering these striking similarities, we propose that TDP-43 may serve analogous functional roles in platelets and synapses, and that the study of platelet TDP-43 might provide a window into disease-related TDP-43 dyshomeostasis in the central nervous system. The development of immunotherapies has proved to be clinically encouraging to re-establish the immune function modified by the expression of immune inhibitory molecules in tumors. However, there are still patients with poor survival rates following treatment. The elucidation of molecular mechanisms triggered by the neo-expression of particular IC in tumors would constitute a major step toward better understanding tumor evolution and would help to design future clinical protocols. To this end, we investigate the modifications triggered by the neo-expression of the immune checkpoints HLA-G in ccRCC tumor cells. We demonstrate, for the first time, that HLA-G modifies key genes implicated mainly in tumor development, angiogenesis, calcium flow and mitochondria dynamics. The involvement of HLA-G on the expression of genes belonging to these pathways such as ADAM-12, NCAM1 and NRP1 was confirmed by the CRISPR/Cas9-mediated edition of HLA-G. The data reveal multifaceted roles of HLA-G in tumor cells which are far beyond the well-known function of HLA-G in the immune anti-tumor response. This warrants further investigation of HLA-G and these new partners in tumors of different origin so as to propose future new treatments to improve health patient's outcome. [ABSTRACT FROM AUTHOR]

Critically endangered Hooded Vultures (Necrosyrtes monachus Temminck, 1823), like many vulture species globally, are experiencing rapid population declines due to anthropogenic factors such as poisonings, human persecution, trading for belief-based use, and habitat loss/degradation. The Hooded Vulture is widespread across sub-Saharan Africa. Although it is considered one of the most abundant vultures in West Africa, this vulture species is less common in East and southern Africa, with the population at the southern-most edge of the distribution (in South Africa and Eswatini) estimated at only 100–200 mature individuals. The distribution of Hooded Vultures has contracted dramatically in southern Africa, with breeding populations largely confined to protected areas such as the Greater Kruger National Park. This study aimed to investigate the genetic diversity of the southern African range-edge population and assess if the recent contraction in the distribution has resulted in the population experiencing a genetic bottleneck. Sixteen microsatellite loci were amplified for samples collected along the Olifants River in the Greater Kruger National Park (n = 30). The genetic diversity in the South African population was compared to samples (n = 30) collected in Ghana, where Hooded Vultures are more abundant. Contrary to expectations, the South African peripheral Hooded Vulture population showed higher levels of heterozygosity (HO = 0.495) than the Ghanaian population (HO = 0.315). Neither population showed signs of recent bottleneck events when tested using demographic modelling and Approximate Bayesian computation (ABC). However, both populations showed high levels of inbreeding and relatedness. Our results suggest that despite being a small peripheral population, the South African Hooded Vulture population showed a similar level of genetic diversity as individuals sampled from a core population within the species distribution (in Ghana). Background: The Nuclear Factor Y (NF-Y) transcription factor (TF) gene family plays a crucial role in plant development and response to stress. Limited information is available on this gene family in sugarcane. Objectives: To identify sugarcane NF-Y genes through bioinformatic analysis and phylogenetic association and investigate the expression of these genes in response to abiotic and biotic stress. Methods: Sugarcane NF-Y genes were identified using comparative genomics from functionally annotated Poaceae and Arabidopsis species. Quantitative PCR and transcriptome analysis assigned preliminary functional roles to these genes in response to water deficit, cold and African sugarcane borer (Eldana saccharina) infestation. Results: We identify 21 NF-Y genes in sugarcane. Phylogenetic analysis revealed three main branches representing the subunits with potential discrepancies present in the assignment of numerical names of some NF-Y putative orthologs across the different species. Gene expression analysis indicated that three genes, ShNF-YA1, A3 and B3 were upregulated and two genes, NF-YA4 and A7 were downregulated, while three genes were upregulated, ShNF-YB2, B3 and C4, in the plants exposed to water deficit and cold stress, respectively. Functional involvement of NF-Y genes in the biotic stress response were also detected where three genes, ShNF-YA6, A3 and A7 were downregulated in the early resistant (cv. N33) response to Eldana infestation whilst only ShNF-YA6 was downregulated in the susceptible (cv. N11) early response. Conclusions: Our research findings establish a foundation for investigating the function of ShNF-Ys and offer candidate genes for stress-resistant breeding and improvement in sugarcane. Three-dimensional (3D) stacking using hybrid bonding is the most scalable method for 3D integration. As the hybrid bonding pad width is reduced to adopt a higher number of interconnections, the ability to extract the contact resistivity at the bonding interface with high accuracy is critical. Using specific electrical test structures and a dedicated methodology, we extract the contact resistivity for hybrid bonding pad widths down to 400 nm for a Cu/SiO2 hybrid bonding integration. Very low values around 10−11 Ω cm2 were obtained for our reference process, close to the those of Cu grain boundaries. A comprehensive analysis of the experimental contact resistivity is performed to understand its increase with the Cu recess within the bonding pads. Based on thermomechanical simulations and experimental results, the influence of both the pad thickness and initial dishing on the interface closure is discussed, for bonding pad width down to 100 nm. These analyses enable us to propose process conditions to reach low contact resistivity with low sensitivity to wafer-to-wafer overlay for hybrid bonding stacking using bonding pad widths down to 100 nm. Objective: HIV Pre-Exposure Prophylaxis (PrEP) has been used in France since 2016. Its effectiveness is no longer to be demonstrated. However, follow-up and adherence remain the main pitfalls. The main objective of this study was to identify factors associated with persistence or loss of PrEP follow-up. Design: An historic cohort of PrEP users was compiled from the database of consultations in the Indre-et-Loire dedicated sexual health centers (CeGIDD) from June 2016 to June 2021. Methods: Kaplan–Meier curves were performed to compare the group of persistent PrEP users to the discontinuation group. Factors associated with PrEP discontinuation were identified using Cox modelling, considering time-dependent variables. Final variables included in the model were selected based on the Akaike Information Criterion (AIC) and clinical relevance. Results: Over the period, 568 PrEP users were included in the cohort. Median follow-up was 2.3 years. A quarter of users were lost to follow-up within 3 months after PrEP initiation. Sexual risk reduction AIDS community-based support (HR = 0.65[0.42;0.99]), being in a couple (HR = 0.51[0.38;0.68]), and history of syphilis (HR = 0.57[0.40;0.81]) were significantly associated with persistence of follow-up. Remote consultations (HR = 2.74[1.63;4.61]), chemsex practices (HR = 2.01[1.29;3.14]), and side effects (HR=1.72[1.03;2.88]) were significantly associated with a loss of follow-up. Conclusion: These results suggest that more sexual risk reduction AIDS community-based counseling could be a key, necessary for supporting PrEP users in their follow-up pathway. Indeed, AIDS community-based support could be used to build a basis for developing safe pathways. Remote consultations could represent a response to the issue of access to PrEP. To create a significant impact on global HIV incidence, the PrEP offer must be extended, and at-risk PrEP users supported to maintain PrEP use. [ABSTRACT FROM AUTHOR]

Between the sixth and fifteenth c. CE, a vast expanse of central and southern Sudan belonged to the kingdom of Alwa, ruled from the urban metropolis of Soba. Renewed investigation of the city unearthed a small cemetery in the northern part of the site. The heterogeneity of burial practices raised some questions as to the cultural and religious affinities of the deceased and suggested potential multiculturalism of the local urban population. We applied isotopic analyses to investigate the origins of the people buried at Cemetery OS and their concomitant ways of life. Non-concordance of 87Sr/86Sr and δ18O values with local hydro-geological background speaks to the mixing of water sources as a result of residential mobility. The concordance of human and faunal strontium and oxygen results, combined with elevated δ13C values corresponding to almost exclusive reliance on C4 produce, points to the possibility of seasonal movement of people with their herds between the Nile valley and the adjacent grasslands. Despite the turn of the medieval Nubian economy towards settled agriculture, by revealing the granular specificities of human adaptation in challenging ecosystems, our results produce the first insight into the enduring diversification of economic production, even in urbanized settings, and persisting participation of local peoples in agro-pastoral symbiosis. In this theoretical article, we argue that the imminent collapse of earth systems that sustain life forms calls for mathematics education as a field to reflect on and re-evaluate its priorities and thus practices. We consider both what ecological collapse means for mathematics education and whether mathematics education might offer meaningful gestures in response. We explore how the relationship between the social and the ecological is conceptualised in mathematics education (and other relevant) research and what this implies for mathematics education. We read, in this scholarship, a growing focus on the ecological and conceptualisations of socio-ecological relations between existing entities that are dialectical, or mutually dependent. More rarely, are they seen as entangled and monist, and it is in this thought that we locate our contribution of multi-layered gestures of mathematics education. We describe these, in terms of three broad practices: listening for socio-ecological entanglement; attending to the scales of socio-ecological entanglements; and living entanglement as mathematics educators. We exemplify these gestures through examples of curriculum innovation. Acacia longifolia (Andrews) Willd. is a legume native to southeast mainland Australia and Tasmania and has two described subspecies: A. l. subsp. longifolia and A. l. subsp. sophorae. The species has been introduced around the world and is considered invasive in several Mediterranean-type climate regions, including in South America, South Africa, and southern Europe. Previous studies comparing native and invasive populations of A. longifolia have focused on its reproductive ecology and population genetics, and little information exists on the species' early life development and how abiotic factors influence it. Here, we performed a glasshouse experiment to compare the phenotypic responses of native and invasive (in Portugal) A. longifolia seedlings to different levels of water and nutrient availability. We found that seedlings of both subspecies responded similarly to different water and nutrient availability conditions in terms of biomass accumulation, root length, the number of phyllodes produced, phyllode water content, and root-to-shoot ratio. However, compared to native seedlings, invasive seedlings had limited capacity for stress responses. We found that invasive seedlings had lower drought tolerance than native seedlings, and thus the speed of invasion by A. longifolia into drier parts of Portugal may be hindered. Our results also hint of a possible role of seed "imprinting" in this species' early growth responses, resulting in different resource allocation strategies such as favouring early growth and development over drought resistance in the invaded range. Interpretations of Late Pleistocene hominin adaptative capacities by archaeologists have focused heavily on their exploitation of certain prey and documented contemporary behaviours for these species. However, we cannot assume that animal prey-taxa ecology and ethology were the same in the past as in the present, or were constant over archaeological timescales. Sequential isotope analysis of herbivore teeth has emerged as a particularly powerful method of directly reconstructing diet, ecology and mobility patterns on sub-annual scales. Here, we apply 87Sr/86Sr isotope analysis, in combination with δ18O and δ13C isotope analysis, to sequentially sampled tooth enamel of prevalent herbivore species that populated Europe during the Last Glacial Period, including Rangifer tarandus, Equus sp. and Mammuthus primigenius. Our samples come from two open-air archaeological sites in Central Germany, Königsaue and Breitenbach, associated with Middle Palaeolithic and early Upper Palaeolithic cultures, respectively. We identify potential inter- and intra-species differences in range size and movement through time, contextualised through insights into diet and the wider environment. However, homogeneous bioavailable 87Sr/86Sr across large parts of the study region prevented the identification of specific migration routes. Finally, we discuss the possible influence of large-herbivore behaviour on hominin hunting decisions at the two sites. Using multi-isotope analysis of tooth enamel of reindeer, equids and mammoth from the archaeological sites of Königsaue and Breitenbach, the authors identify shifts in ranging behaviour of these hominin prey species during the Last Glacial Period in Central Germany. Acacia longifolia is an aggressive invader in Mediterranean-type ecosystems severely impacting biodiversity and ecosystem functions. The species' invasiveness has been linked to its ability to thrive in nutrient poor soils, high seed production, and quick establishment after fire. In this study, we identify and compare the bacterial endophytes of A. longifolia seeds collected from populations in the species' native (Australia) and invasive (Portugal) ranges. For this, we characterised the morphology (length, width, and weight) of seeds from two sites in each range and isolated and cultivated bacteria from seeds. DNA fingerprinting and cluster analyses revealed slightly higher, and distinct, bacterial diversity associated with seeds collected from native range populations in comparison to those collected from invasive populations. Sequencing of the 16S rDNA gene identified 119 bacterial isolates from 15 genera, with Curtobacterium strains being common in both ranges. Several differences in bacterial genera were found among ranges and sites: Dermacoccus, Frigoribacterium, Kocuria, Pantoea and Phyllobacterium taxa were each unique to seeds from the native populations, while Brevundimonas, Microbacterium, Rhizobium and Sphingomonas taxa were only found in the invasive seeds. The genus Paraburkholderia occurred in all invasive-range seeds but was not isolated from the native-range. Bacillus and Paenibacillus co-occurred in seeds collected from all invaded sites, but the simultaneous presence of both taxa was not found in native-range seeds. We propose that the bacterial endophytes present in invasive-range seeds may be important players for the invasiveness of A. longifolia, due to their role as plant growth promoters, providing extra capabilities helping acacia expansion. Dispersal influences many key aspects of plant ecology at both fine- and broad scales. However, dispersal events are challenging to quantify as they are difficult to observe and measure accurately, despite the importance of understanding species' dispersal capacity. In this study, we quantified dispersal estimates for the dominant vascular flora of sub-Antarctic Marion Island, using a mechanistic model to estimate dispersal potential via anemochory and standardized experiments that simulate dispersal events via zoochory, human activity, and thalassochory, to test if dispersal capacity correlates to contemporary range expansion of the island's native and alien species. Our results demonstrate the broad range of dispersal potential in the island's flora and represent the first quantification of the dispersal potential (via multiple vectors) of the dominant vascular flora in the sub-Antarctic. Dispersal potential was not related to range expansion rates of native or alien species, suggesting that other mechanisms are responsible for the variation observed in species range expansion rates. Although this finding contradicts expectations and evidence from some other studies, it is consistent with research conducted in alpine regions (which may be climatically and physiognomically similar to this sub-Antarctic study site) where factors such as demography and competition are more important predictors of species range expansion than their dispersal ability, dispersal syndromes, or dispersal-related traits. Purpose: Spouses are often the front-line caregivers for colon cancer patients. Providing this support requires a particular set of coping skills. Our objective was to identify key skills that healthcare and medico-social sector professionals could assess in routine practice that would allow them to propose appropriate support to spouses who are accompanying colon cancer patients in their care pathway. Methods: An online two-round Delphi study was conducted among French colon cancer patients, spouses and professionals. The content of the Delphi study was developed from a previously published qualitative study. Results: In the first round of the study, 63% of the participants were professionals (n = 40), 19% spouses (n = 12) and 17% patients (n = 11). In the second round, they were respectively 55% (n = 22), 22% (n = 9) and 22% (n = 9). Twenty-seven of the 75 proposed skills were consensually identified as key skills. Nine were related to emotional and psychological well-being, six to social relations, four to organisation, five to health and three to domestic domains. The three most consensual skills (≥ 90% agreement) for spouses were (1) helping the tired patient in everyday life, (2) stimulating the patient to prevent him/her from giving up and (3) limiting one’s amount of personal time to care for the patient. Conclusion: The study identified the key skills needed by spouses of patients being treated for colon cancer. With their remarkably low thresholds, organic polariton lasers are a promising alternative to organic photonic lasers. However, device stability remains a challenge, in part due to material degradation during deposition of the top dielectric mirror. We demonstrate polariton lasers based on 4,4′-Bis(4-(9H-carbazol-9-yl)styryl)biphenyl (BSBCz) as active material that achieve a low lasing threshold of 8.7 μJ/cm2, and we show that a ZrO2 protection layer between active layer and top mirror significantly improves stability. Optimized devices exhibit minimal degradation after 100,000 excitation pulses at 3.8 times above threshold. Purpose: To assess the effects of Helicobacter pylori (HP) eradication with an omeprazole, clarithromycin, amoxicillin, and metronidazole (OCAM) regimen on the metabolic profile and weight loss 12 months after bariatric surgery (BS). Methods: Retrospective analysis of a prospective cohort of patients with morbid obesity undergoing BS. HP presence was tested preoperatively by gastric biopsy and treated with OCAM when positive. Short-term metabolic outcomes and weight loss were evaluated. Results: HP infection was detected in 75 (45.7%) of the 164 patients included. OCAM effectiveness was 90.1%. HP-negative patients had a greater reduction in glucose levels at 3 (−14.6 ± 27.5 mg/dL HP-treated vs −22.0 ± 37.1 mg/dL HP-negative, p=0.045) and 6 months (−13.7 ± 29.4 mg/dL HP-treated vs −26.4 ± 42.6 mg/dL HP-negative, p= 0.021) and greater total weight loss (%TWL) at 6 (28.7 ± 6.7% HP-treated vs 30.45 ± 6.48% HP-negative, p= 0.04) and 12 months (32.21 ± 8.11% HP-treated vs 35.14 ± 8.63% HP-negative, p= 0.023). Conclusions: Preoperative treatment with OCAM has been associated to poorer glycemic and weight loss outcomes after BS. Introduction: Sleeve gastrectomy (SG) is an effective treatment for obesity in adolescents. The underlying weight loss mechanism may impact the peripheral and central gustatory system along with reward circuits in the brain. This study aims to assess changes in appetitive behavior in short-, medium-, and long-term follow-up. Methods: In this prospective observational study, a total of 8 adolescents with obesity who underwent SG and 9 comparator unoperated participants were studied. Appetitive behaviour towards fat and sweet taste stimuli was assessed using the Progressive Ratio Task (PRT) over a 6 year period. Results: Mean body mass index (BMI) of the surgical patients dropped from 51.5 ± 2.8 kg/m2 to 31.4 ± 1.9 and 30.9 ± 2.3 kg/m2 at 1 and 6 years follow-up, respectively. (p < 0.001). The median (interquartile range) total rewards earned during the PRT was 6 (5–7) pre-surgery, 5 (3–6) after one year and 4 (2–4) after six years from surgery (p = 0.007). Bedrest shifts fasting and postprandial fuel selection towards carbohydrate use over lipids, potentially affecting astronauts' performance and health. We investigated whether this change occurs in astronauts after at least 3 months onboard the International Space Station (ISS). We further explored the associations with diet, physical activity (PA), and body composition. Before and during spaceflight, respiratory quotient (RQ), carbohydrate, and fat oxidation were measured by indirect calorimetry before and following a standardized meal in 11 males (age = 45.7 [SD 7.7] years, BMI = 24.3 [2.1] kg m−²). Postprandial substrate use was determined by 0-to-260 min postprandial incremental area under the curve (iAUC) of nutrient oxidation and the difference between maximal postprandial and fasting RQ (ΔRQ). Food quotient (FQ) was calculated from diet logs. Fat (FM) and fat-free mass (FFM) were measured by hydrometry and PA by accelerometry and diary logs. Spaceflight increased fasting RQ (P = 0.01) and carbohydrate oxidation (P = 0.04) and decreased fasting lipid oxidation (P < 0.01). An increase in FQ (P < 0.001) indicated dietary modifications onboard the ISS. Spaceflight-induced RQ changes adjusted for ground RQ correlated with inflight FQ (P < 0.01). In postprandial conditions, nutrient oxidation and ΔRQ were unaffected on average. Lipid oxidation changes negatively correlated with FFM changes and inflight aerobic exercise and positively with FM changes. The opposite was observed for carbohydrate oxidation. ΔRQ changes were negatively and positively related to FM and FFM changes, respectively. In conclusion, fasting substrate oxidation shift observed during spaceflight may primarily result from dietary modifications. Background and aims: Invasive plants often alter soil abiotic and biotic conditions which can benefit their own growth while harming native species. The impacts on native species may persist as legacy effects after the invasive species has been controlled and removed. This study focused on the Critically Endangered Cumberland Plain Woodland (CPW) vegetation in Australia, where we examined the soil impacts, and their associated legacy effects, associated with the invasion of African olive (Olea europaea subsp. cuspidata). Methods: Seedlings of O. europaea subsp. cuspidata and two native legume species, Acacia implexa and Indigofera australis, were grown in different sterilised and unsterilised soils: uninvaded CPW soil, restored CPW soil where O. europaea subsp. cuspidata was removed approximately 20 years ago, and soil from sites still under O. europaea subsp. cuspidata invasion. We characterised nitrogen-fixing rhizobia in the root nodules of seedlings of the two legumes using next-generation sequencing (NGS) barcoding. Results: Olea europaea subsp. cuspidata did not appear to condition the soil to favour its own growth and grew best in uninvaded CPW soil. The performance of both native legume species, however, was negatively impacted when grown in sterilised and unsterilised invaded soils, relative to their growth in CPW and restored soils. The soils from invaded sites affected the associations between both legume species and their rhizobium mutualists. Nodulation was higher in CPW and restored soils than in invaded soils, indicating that the availability of rhizobia was negatively impacted by O. europaea subsp. cuspidata. This was confirmed by a negative link between nodulation and the abundance of rhizobia that were characteristic of invaded soils. Conclusion: Our findings demonstrate that O. europaea subsp. cuspidata invasion affects the availability of microbial mutualists for native legumes in the CPW. The soil conditions created by O. europaea subsp. cuspidata do not benefit its own performance and these impacts do not persist as legacy effects 20 years after the removal of the invader. Background: Lung ultrasound (US), which is radiation-free and cheaper than chest radiography (CXR), may be a useful modality for the diagnosis of pediatric pneumonia, but there are limited data from low- and middle-income countries. Objectives: The aim of this study was to evaluate the diagnostic performance of non-radiologist, physician-performed lung US compared to CXR for pneumonia in children in a resource-constrained, African setting. Materials and methods: Children under 5 years of age enrolled in a South African birth cohort study, the Drakenstein Child Health Study, who presented with clinically defined pneumonia and had a CXR performed also had a lung US performed by a study doctor. Each modality was reported by two readers, using standardized methodology. Agreement between modalities, accuracy (sensitivity and specificity) of lung US and inter-rater agreement were assessed. Either consolidation or any abnormality (consolidation or interstitial picture) was considered as endpoints. In the 98 included cases (median age: 7.2 months; 53% male; 69% hospitalized), prevalence was 37% vs. 39% for consolidation and 52% vs. 76% for any abnormality on lung US and CXR, respectively. Agreement between modalities was poor for consolidation (observed agreement=61%, Kappa=0.18, 95% confidence interval [95% CI]: − 0.02 to 0.37) and for any abnormality (observed agreement=56%, Kappa=0.10, 95% CI: − 0.07 to 0.28). Using CXR as the reference standard, sensitivity of lung US was low for consolidation (47%, 95% CI: 31–64%) or any abnormality (5%, 95% CI: 43–67%), while specificity was moderate for consolidation (70%, 95% CI: 57–81%), but lower for any abnormality (58%, 95% CI: 37–78%). Overall inter-observer agreement of CXR was poor (Kappa=0.25, 95% CI: 0.11–0.37) and was significantly lower than the substantial agreement of lung US (Kappa=0.61, 95% CI: 0.50–0.75). Lung US demonstrated better agreement than CXR for all categories of findings, showing a significant difference for consolidation (Kappa=0.72, 95% CI: 0.58–0.86 vs. 0.32, 95% CI: 0.13–0.51). Conclusion: Lung US identified consolidation with similar frequency to CXR, but there was poor agreement between modalities. Microbial inoculants are attracting growing interest in agriculture, but their efficacy remains unreliable in relation to their poor survival, partly due to the competition with the soil resident community. We hypothesised that recurrent inoculation could gradually alleviate this competition and improve the survival of the inoculant while increasing its impact on the resident bacterial community. We tested the effectiveness of such strategy with four inoculation sequences of Pseudomonas fluorescens strain B177 in soil microcosms with increasing number and frequency of inoculation, compared to a non-inoculated control. Each sequence was carried out at two inoculation densities (106 and 108 cfu.g soil−1). The four-inoculation sequence induced a higher abundance of P. fluorescens, 2 weeks after the last inoculation. No impact of inoculation sequences was observed on the resident community diversity and composition. Differential abundance analysis identified only 28 out of 576 dominants OTUs affected by the high-density inoculum, whatever the inoculation sequence. Recurrent inoculations induced a strong accumulation of nitrate, not explained by the abundance of nitrifying or nitrate-reducing microorganisms. In summary, inoculant density rather than inoculation pattern matters for inoculation effect on the resident bacterial communities, while recurrent inoculation allowed to slightly enhance the survival of the inoculant and strongly increased soil nitrate content. Background: Obesity is defined as excess adipose tissue causing a deterioration in health, but diagnosing the causes and deciding on treatment can be challenging. Several randomized controlled clinical trials (RCT) have demonstrated the effectiveness of semaglutide as a treatment for obesity. This study investigated the clinical response to semaglutide as a weight loss treatment in a real-world setting. Methods: This observational study investigated the response to injectable semaglutide in the first 3 months during the dose titration phase up to 1 mg. Weight loss after 6 months was also evaluated. The data were collected from the electronic medical records (EMR) from outpatient clinics between July 2021 to March 2023. All participants were older than 18 years, with no history of bariatric surgery within 1 year, and had a least one prescription of injectable semaglutide. The primary outcome was weight change at 3 months. Weight loss in those patients who attended at 6 months was a secondary outcome. Results: A total of 350 patients were included in the study. The vast majority (80.3%) were female. 287 patients (82%) completed 3 months on injectable semaglutide and lost 6.6 ± 3.8% bodyweight. 224 patients (64%) completed 6 months on semaglutide and lost 12 ± 6.1% bodyweight. 188 (65.5%) of patients who completed 3-month follow-up lost ≥5% weight, 39 (13.5%) patients lost ≥10% weight, and 7 (2.4%) patients lost ≥15% weight. While for those patients who completed the 2nd visit (n = 224), 201 (89.7%) lost ≥5% weight, 135 (60.3%) lost ≥10% weight, and 54 (24.1%) lost ≥ 15% body weight. Conclusion: Injectable semaglutide in a real-world setting resulted in similar weight loss and had a similar side effect profile as was observed in randomized controlled trials. Background: Obesity is a chronic and complex disease characterized by the excessive accumulation of adipose tissue, which has detrimental effects on health. Evaluating the changes in quality of life (QoL) after bariatric surgery complements the medical benefits which are documented by healthcare professionals. Purpose: To study the perceived health benefits 1 year after substantial weight loss induced by bariatric surgery. Methods: This pilot study evaluated patients 1 year after bariatric surgery using 13 questions related to the health domains of the KOSS: airway, body mass, cardiovascular risk, diabetes, economic impact, functional, gonadal impact, health status perceived, image, junction of the gastro-esophagus, kidney, liver, and medication. In addition, the patients were asked to score the most significant benefit as "1," while the least beneficial benefit was scored as "13." Results: One hundred fourteen consecutive patients were evaluated (men = 37 and women = 77). The responses were divided into functional, metabolic, and mental/social benefits. Patients ranked the functional question, "I became more active, and I can do more things" as the most important (average score of 3.7 ± 0.2), followed by a question related to metabolic status: "I am less worried about my risk of heart disease" (4.5 ± 0.3), and then a social/mental question, "My clothes fit better" (5.4 ± 0.3). The three least valuable benefits for the cohort were sexual life improvements (8.9 ± 0.3), heartburn improvements (9.0 ± 0.3), and urinary incontinence improvements (9.8 ± 0.3). Conclusions: Our observational pilot study demonstrated that patients value functional benefits after substantial weight loss the most, but that metabolic benefits and social/mental health benefits are also considered important. Phage satellites are bacterial genetic elements that co-opt phage machinery for their own dissemination. Here we identify a family of satellites, named Phage-Inducible Chromosomal Minimalist Islands (PICMIs), that are broadly distributed in marine bacteria of the family Vibrionaceae. A typical PICMI is characterized by reduced gene content, does not encode genes for capsid remodelling, and packages its DNA as a concatemer. PICMIs integrate in the bacterial host genome next to the fis regulator, and encode three core proteins necessary for excision and replication. PICMIs are dependent on virulent phage particles to spread to other bacteria, and protect their hosts from other competitive phages without interfering with their helper phage. Thus, our work broadens our understanding of phage satellites and narrows down the minimal number of functions necessary to hijack a tailed phage. Environmental DNA (eDNA) sampling has attracted worldwide attention over the past few years as an emerging approach to characterising and monitoring biodiversity, and has become particularly important for species that are rare, elusive or endangered. Most animal studies to date have focused on aquatic taxa; studies on other metazoan taxa, particularly wildlife in terrestrial environments, are scarce, with only a handful utilizing soil sources. We aimed to investigate the use of DNA barcoding from soil eDNA in (1) detecting rare/elusive/threatened species and (2) as a tool to investigate and potentially monitor range distributions. Through extensive eDNA sampling along the west coast of South Africa, we aimed to refine the distributions of four golden mole species thought to occur there, and specifically to determine whether De Winton's golden mole, Cryptochloris wintoni (IUCN Critically Endangered; Possibly Extinct), is in fact extant or extinct. Sequences were generated for three barcode markers (mtDNA cyt b, 12S and nuclear GHR) using next-generation amplicon sequencing. Tissue samples from four specimens were used to generate reference sequences for species identification, along with available GenBank sequences. We were able to (1) successfully detect all four species in our data, and (2) improve records of the distributions of these species. Furthermore, we uncovered cryptic diversity in Eremitalpa granti. Our data conclusively reveal the presence of the elusive Cryptochloris wintoni and suggest that this species may in fact be widespread, but not necessarily abundant, and certainly less so in areas subjected to mining activities, which continue to pose a threat to the species. Genetic diversity in over-exploited and threatened species has important, but often neglected, implications for conservation. In South Africa, the bark of the regionally endangered pepper-bark tree, Warburgia salutaris, is extensively harvested for subsistence medicine. We developed 11 polymorphic microsatellite markers to examine the genetic diversity and structure among 227 W. salutaris individuals sampled from across much of its range in South Africa's Kruger National Park (KNP) and 31 trees growing in local nurseries. We also tested the cross-amplification success of these markers in two closely related species, Warburgia elongata (IUCN listed as Endangered) and W. stuhlmannii (IUCN listed as Vulnerable). Contrary to our expectations, we found evidence of clonality and for high genetic diversity in all wild KNP sub-populations of W. salutaris. We identified between 114 and 149 unique multi-locus genotypes in the 258 individuals included in our analyses. Bayesian assignment tests identified three distinct genetic clusters that are represented by the sampled KNP sub-population of W. salutaris, with minimal geographic structure and some admixture. Three genetic clusters were also identified among all sampled individuals, i.e., KNP and nursery trees. Nursery W. salutaris germplasm comprised unique genotypes with similar levels of observed heterozygosity to the wild population. High levels of clonality and reported self-incompatibility, along with short distance pollination, likely explain the lack of recent sexual recruitment (i.e., reduced fruit set and lack of seedlings) in the wild population. These findings will be useful to identify sub-populations where genets may be sampled to maximize genetic diversity within subpopulations for restoration throughout KNP. [ABSTRACT FROM AUTHOR]

In face of the current environmental challenges, developing multifunctional cropping systems is increasingly needed, and crop variety mixtures are particularly interesting since they can deliver diverse services including grain production, yield stability, N2O production regulation, disease control, and reduction of N-fertilizer losses. However, the relationships between intraspecific diversity and ecosystem multifunctionality are poorly understood so far, and practitioners lack science-based guidance to design mixtures. We used a pool of 16 bread wheat varieties classified into 4 functional groups based on 26 below- and aboveground functional traits, to conduct a field trial (88 large plots cultivated with single varieties or mixtures of 2, 4, or 8 varieties), quantifying 15 provisioning and regulating services for each plot. To assess yield stability between local conditions and years, the trial was replicated at 4 other locations and for 2 years, using 2 managements each time. We analyzed how variety number and functional groups predicted the variance in services, and applied in an innovative manner the RLQ co-inertia analysis to relate the (variety × traits) matrix Q to a (plot × services) matrix R, using a (plot × variety) composition matrix L as a link. Our results show that using variety mixtures allowed delivery of baskets of services not reachable when cultivating single varieties, and that mixtures mitigated tradeoffs between different pairs of services. Variety number or functional groups poorly predicted the variance in services, but the RLQ approach allowed the identification of groups of plots delivering consistent baskets of services. Moreover, we demonstrated for the first-time significant relationships between specific baskets of services and bundles of variety traits. We discuss how our results increase our understanding of intraspecific diversity–agroecosystem multifunctionality relationships, and propose the next steps using our new approach to support practitioners for designing variety mixtures that provide particular baskets of services. As one of the key, long-term occupied sites in the Southern Levant, Jericho was one of the most important early Neolithic centres to witness social and economic changes associated with the domestication of plants and animals. This study applies strontium (87Sr/86Sr), oxygen (δ18O) and carbon (δ13C) isotope analyses to the enamel of 52 human teeth from Pre-Pottery Neolithic (PPN) layers of Jericho to directly study human diet and mobility and investigate the degree of consolidation and the flexibility of social organization of Jericho society in the PPN period. The results indicate only two non-local individuals out of the 44 sampled inhabitants identified by strontium isotope analysis and are consistent with the presence of a largely sedentary community at PPN Jericho with no evidence for large-scale migration. We also construct strontium spatial baselines (87Sr/86Sr map) with local 87Sr/86Sr signatures for the sites across the Southern Levant based on systematic compilation and analysis of available data. In addition, we apply proteomic analysis of sex-specific amelogenin peptides in tooth enamel for sex estimation of the sampled individuals (n = 44), the results of which showed a sex-biased ratio (more male than female detected in this sample pool) in Jericho society during the PPN period, which may be due to the limited sample size or selective ritual practices like particular burial zones used for specific groups. We also pretreated a batch of human bone samples recovered from PPNB Jericho for stable carbon and nitrogen isotope analyses for dietary investigations. Background: Obesity is a chronic inflammatory condition and this meta-analysis evaluated the impact of bariatric surgery on SAA. Methods: Studies included all types of bariatric surgery where SAA was measured before and after the surgical procedure. Results: Meta-analysis of 11 clinical studies (n = 394 individuals) confirmed a significant reduction in SAA following bariatric surgery (SMD: − 0.971, 95% CI: − 2.721, 0.779, p < 0.001). Meta-regression did not show any association between the changes in BMI and the absolute difference in SAA levels. No relationship between the changes in SAA and the length of follow-up was found. Conclusion: Bariatric surgery significantly improved SAA. The decrease in SAA was not related to time after surgery or changes in BMI. Bariatric surgery may thus have an independent effect on SAA. Background: The TNM system is used to assess prognosis after colorectal cancer (CRC) diagnosis. Other prognostic factors reported include histopathological assessments of the tumour, tumour mutations and proteins in the blood. As some of these factors are strongly correlated, it is important to evaluate the independent effects they may have on survival. Methods: Tumour samples from 2162 CRC patients were visually assessed for amount of tumour stroma, severity of lymphocytic infiltrate at the tumour margins and the presence of lymphoid follicles. Somatic mutations in the tumour were assessed for 2134 individuals. Pre-surgical levels of 4963 plasma proteins were measured in 128 individuals. The associations between these features and prognosis were inspected by a Cox Proportional Hazards Model (CPH). Results: Levels of stroma, lymphocytic infiltration and presence of lymphoid follicles all associate with prognosis, along with high tumour mutation burden, high microsatellite instability and TP53 and BRAF mutations. The somatic mutations are correlated with the histopathology and none of the somatic mutations associate with survival in a multivariate analysis. Amount of stroma and lymphocytic infiltration associate with local invasion of tumours. Elevated levels of two plasma proteins, CA-125 and PPP1R1A, associate with a worse prognosis. Conclusions: Tumour stroma and lymphocytic infiltration variables are strongly associated with prognosis of CRC and capture the prognostic effects of tumour mutation status. Study design: An ambispective review of consecutive cervical spine surgery patients enrolled in the Canadian Spine Outcomes and Research Network (CSORN) between January 2015 and September 2019. Purpose: To compare complication rates of degenerative cervical spine surgery over time between older (> 65) and younger age groups (< 65). Summary of background data: More elderly people are having spinal surgery. Few studies have examined the temporal nature of complications of cervical spine surgery by patient age groups. Methods: Adverse events were collected prospectively using adverse event forms. Binary logistic regression analysis was utilized to assess associations between risk modifiers and adverse events at the intra-, peri-operative and 3 months post-surgery. Results: Of the 761 patients studied (age < 65, n = 581 (76.3%) and 65 + n = 180 (23.7%), the intra-op adverse events were not significantly different; < 65 = 19 (3.3%) vs 65 + = 11 (6.1%), p < 0.087. Peri-operatively, the < 65 group had significantly lower percentage of adverse events (65yrs (11.2%) vs. 65 + = (26.1%), p < 0.001). There were no differences in rates of adverse events at 3 months post-surgery (< 65 = 39 (6.7%) vs. 65 + = 12 (6.7%), p < 0.983). Less blood loss (OR = 0.99, p < 0.010) and shorter length of hospital stay (OR = 0.97, p < 0.025) were associated with not having intra-op adverse events. Peri-operatively, > 1 operated level (OR = 1.77, p < 0.041), shorter length of hospital stay (OR = 0.86, p < 0.001) and being younger than 65 years (OR = 2.11, p < 0.006) were associated with not having adverse events. Conclusion: Following degenerative cervical spine surgery, the older and younger age groups had significantly different complication rates at peri-operative time points, and the intra-operative and 3-month post-operative complication rates were similar in the groups. As a direct result of laboratory sample manipulation required to identify microplastics (MPs) within a given matrix, some MPs are inevitably lost. The extent of this loss can be quite significant and varies greatly depending on the sample matrix, choice of protocol and target MPs in question. Defining analytical MP recovery is therefore a critical component in ensuring the quality of MP protocols. The potential relationship between particle size and recovery rate has been widely discussed but remains uncertain. To determine whether MP loss correlated with particle size, three aliquots of polyethylene fragments in the 5–50 µm size range and three aliquots of polypropylene fragments in the 50–500 µm size range, were consecutively transferred back and forth from filter to liquid. After each individual transfer the analytical recovery within specified size groups, was evaluated by applying high-resolution darkfield microscopy. Average recovery across the entire size range was estimated at 80% with a standard deviation (std. dev.) of 26%. Notably, particle coverage on filters (A%) showed a more significant impact on recovery than particle size. Maintaining A% below 5% on filters for microscopic analysis is advised to prevent excessive loss due to particle agglomeration. To determine whether the use of red polyethylene fragments in the 5–50 µm size range in combination with darkfield microscopy could potentially improve MP recovery evaluation in environmental samples, three aliquots of 0.5 g of dry brown trout muscle tissue were spiked and treated according to a relevant protocol. This size-discriminating approach accurately determined average recovery at 52% with a std. dev. of 4% and demonstrated the potential for correction of the concentration enhancement of smaller MPs resulting from particle breakup during sample pre-treatment, which would otherwise lead to overestimation of smaller size fractions. Highlights: • Recovery experiment shows no evidence of increasing microplastic loss with decreasing particle size, in the 5–500 µm range. • Investigating colored PE micro-fragments under darkfield illumination enables size-discriminating recovery estimations of microplastics ≥ 5 µm. • Particle area coverage should not exceed 5% of filters intended for microscopic analysis of microplastics (< 500 µm). • For increased relevance to environmental conditions, fragments should replace spheres or beads in microplastic recovery experiments. Obesity is now recognised as a disease associated with significant morbidity and mortality. One of the most common metabolic complications of obesity is type 2 diabetes, because the two disease share similar pathophysiology. Weight loss is known to ameliorate the metabolic abnormalities underlying type 2 diabetes and improve glycemic control. A 15% or greater total body weight loss (TBWL) in patients with type 2 diabetes will have a disease-modifying effect, a result that is incomparable with other hypoglycemic-lowering interventions. Moreover, in patients with diabetes and obesity, weight loss exerts benefits beyond glycemic control and improves cardiometabolic disease risk factors and well-being. We review evidence supporting the role of intentional weight loss in managing type 2 diabetes. We suggest that many people with type 2 diabetes would benefit from an additional weight-based approach to managing their diabetes. Objective: This study aimed to evaluate the long-term stability of surgical maxillary expansion using patient-specific fixation implants (PSFIs) without intraoral retention. Materials and methods: Fifteen patients who had undergone segmented Le Fort I osteotomy and PSFIs with available preoperative (t0) early (t1) and 1-year follow-up computed tomography (CT) scans (t2) were evaluated. The early and 1-year 3D models were superimposed to transfer the bony landmarks; the distances between each pair of landmarks at the different time points were then measured. The distances between the canines and second molars were also measured directly on the CT scans. Results: The achieved maxillary expansions ranged from a median of 4.39 (2.00–6.27) mm at the greater palatine foramina to a median of 2.14 (1.56–2 > 83) mm at the canine level of the palatal bone. One year postoperatively, the changes in skeletal diameters ranged from a median of − 0.53 (− 1.65 to 0.41) mm at the greater palatine foramina (p = 0.12) to 0.17 (− 0.09 to 0.32) mm at the canine level of the palatal bone (p = 0.56). Changes in dental arch diameters ranged from a median of − 0.6 (− 2 to 0.00) mm between the second molars to − 1.3 (− 1.8 to − 0.25) mm between the canines (P < 0.05). Conclusion: This study showed the stability of maxillary expansion osteotomy using PSFIs, even without postoperative intraoral retention. Clinical relevance: PSFIs are a reliable method for the surgical treatment of transverse maxillary discrepancy. PFSIs are easy-to-use and improve surgical accuracy. [ABSTRACT FROM AUTHOR]
