Molybdoenzymes are essential in global nitrogen, carbon, and sulfur cycling. To date, the only known bioavailable source of molybdenum (Mo) is molybdate. However, in the sulfidic and anoxic (euxinic) habitats that predominate in modern subsurface environments and that were pervasive prior to Earth's widespread oxygenation, Mo occurs as soluble tetrathiomolybdate ion and molybdenite mineral that is not known to be bioavailable. This presents a paradox for how organisms obtain Mo to support molybdoenzymes in these environments. Here, we show that tetrathiomolybdate and molybdenite sustain the high Mo demand of a model anaerobic methanogen, Methanococcus maripaludis, grown via Mo-dependent formate dehydrogenase, formylmethanofuran dehydrogenase, and nitrogenase. Cells grown with tetrathiomolybdate and molybdenite have similar growth kinetics, Mo content, and transcript levels of proteins involved in Mo transport and cofactor biosynthesis when compared to those grown with molybdate, implying similar mechanisms of transport and cofactor biosynthesis. These results help to reconcile the paradox of how Mo is acquired in modern and ancient anaerobes and provide new insight into how molybdoenzymes could have evolved prior to Earth's oxygenation. Cough is a common and commonly ignored symptom of lung disease. Cough is often perceived as difficult to quantify, frequently self-limiting, and non-specific. However, cough has a central role in the clinical detection of many lung diseases including tuberculosis (TB), which remains the leading infectious disease killer worldwide. TB screening currently relies on self-reported cough which fails to meet the World Health Organization (WHO) accuracy targets for a TB triage test. Artificial intelligence (AI) models based on cough sound have been developed for several respiratory conditions, with limited work being done in TB. To support the development of an accurate, point-of-care cough-based triage tool for TB, we have compiled a large multi-country database of cough sounds from individuals being evaluated for TB. The dataset includes more than 700,000 cough sounds from 2,143 individuals with detailed demographic, clinical and microbiologic diagnostic information. We aim to empower researchers in the development of cough sound analysis models to improve TB diagnosis, where innovative approaches are critically needed to end this long-standing pandemic. Introduction: Despite advantages for patients with ulcerative colitis, Crohn's disease, and familial adenomatous polyposis, restorative proctocolectomy with ileal pouch-anal anastomosis carries a risk of pouch failure, necessitating pouch excision. The traditional open approach is associated with potential complications. Robotic and laparoscopic techniques are emerging, but comparative outcome data are limited. Methods: We conducted a retrospective study of consecutive adult patients undergoing robotic, laparoscopic, and open ileal pouch excision at Mayo Clinic, Rochester, MN, between January 2015 and December 2023. We analyzed data on patient characteristics, perioperative variables, and postoperative outcomes, focusing on short-term complications. Statistical analysis included appropriate tests. Results: The study included 123 patients: 23 underwent robotic-assisted pouch excision, 12 laparoscopic, and 82 open. The robotic approach had the longest median operative time (334 ± 170 min, p = 0.03). However, it demonstrated significantly lower estimated blood loss than open (150 ± 200 ml vs. 350 ± 300 ml, p = 0.002) and laparoscopic surgery (250 ± 250 ml, p = 0.005). Robotic and laparoscopic groups required fewer preoperative ureteral stents than the open group (p = 0.001). Additionally, the robotic approach utilized fewer pelvic drainages (p < 0.0001) and had a lower rate of lysis of adhesions > 60 min compared to open surgery (p = 0.003). Robotic procedures had significantly lower 30-day postoperative complications than the open approach (30.4% vs. 65.9%, p = 0.002) while also demonstrating fewer 30-day reoperations than the laparoscopic group (p = 0.04). Conclusions: Robotic-assisted pouch excision offered significant benefits, including decreased EBL, reduced need for preoperative ureteral stents, and significantly fewer 30-day postoperative complications compared to open surgery. Background: Fluorine 18-labelled tetrafluoroborate ([18F]TFB) is a substrate for the sodium/iodide symporter. In thyroid cancer, [18F]TFB-PET/CT may be an alternative to iodine imaging to evaluate the extent of disease, eligibility for radioiodine treatment, and success of redifferentiation therapies. We report the results of a pilot study to determine tumor uptake of [18F]TFB and compare its properties to [124I]IodinePET/CT in patients with metastatic thyroid cancer. Methods: Five patients were included in a prospective study. All patients received PET/CT 1 h after injection of 356 ± 12 MBq [18F]TFB and were given 230 ± 9 MBq [124I]Iodine orally on the same day, followed by PET/CT after 48 h. Before redifferentiation therapy, patients underwent an additional baseline [124I]Iodine PET/CT. Cases were analyzed by two board-certified specialists. Detection rates and Spearman correlation for [18F]TFB and [124I]Iodine were calculated. Results: Three patients had poorly differentiated thyroid cancer and received trametinib in a redifferentiation trial. Two patients had papillary thyroid cancer and did not receive redifferentiation therapy. Of the 33 lesions seen before/without redifferentiation therapy, 19 (58%) were visible on [18F]TFB and 30 (91%) on [124I]Iodine imaging. In the patients who underwent redifferentiation therapy, 48 lesions were newly seen on [124I]Iodine PET/CT with a median SUVmax of 3.3 (range, 0.4–285.0). All of these lesions were [18F]TFB-negative. Conclusion: [18F]TFB failed to predict radioactive iodine uptake in patients with poorly differentiated thyroid cancer who underwent redifferentiation therapy with trametinib. It is unclear whether such discrepancies may also occur in other redifferentiation therapies or may even be encountered in redifferentiation-naïve thyroid cancer. Introduction: Acute severe ulcerative colitis (ASUC) is a life-treating presentation of ulcerative colitis (UC) that requires prompt initiation of treatment to avoid complication. Unfortunately, outcomes for ASUC are suboptimal, with as many as 20–30% of patients requiring colectomy. This can be challenging for patients and highlights the need to understand patient experiences and perspectives navigating ASUC. Methods: A qualitative descriptive study utilizing semi-structured interviews was conducted to understand perspectives and experiences of patients navigating ASUC. Adult patients hospitalized for ASUC between January 2017 and March 2024 were eligible. Interviews were conducted both retrospectively among patients with a recent hospitalization and prospectively among patients within 24 h of hospitalization for ASUC. Interviews were analyzed using a well-established hybrid inductive-deductive approach. Results: Thirty-four patients (44.2% response rate) hospitalized for ASUC were interviewed. Hybrid thematic analysis uncovered five major themes: (1) the pervasive impact of UC on QoL and mental health, (2) challenges associated with navigating uncertainty, (3) prioritizing colon preservation, (4) bridging the divide between outpatient expectations and inpatient realities, and (5) balancing rapid symptom improvement with steroid safety. Our findings advocate for transparent approach to care, emphasizing the need for effective communication, education, and better alignment with patient values and expectations. Conclusion: Five key themes were identified, each with significant implications for developing a more patient-centered approach to ASUC care. This article explores the significance of trust in public health response efforts, specifically in relation to the COVID-19 pandemic. Trust in authorities and institutions is crucial for promoting compliance with health protocols and behaviors. Various factors, such as information sources, media credibility, and personal experiences, influence trust. The article focuses on the New York City Test and Trace program and identifies both facilitators and barriers to trust, including the involvement of reputable organizations, adapting to community needs, and forming partnerships. The findings emphasize the importance of trust in public health initiatives and offer insights for enhancing trustworthiness. The article also discusses the implementation of the Test & Trace (T2) program in New York City during the COVID-19 pandemic. The program established partnerships with community-based organizations (CBOs) and community advisory boards (CABs) that had existing relationships with the Department of Health and Mental Hygiene (DOHMH). Trusted sources of information, such as local media outlets, were utilized to effectively communicate COVID-19 information and resources. Hiring frontline staff from local communities who shared similar demographics and identities with community members helped to foster trust. However, barriers to trust included historical mistrust in government and the medical establishment, inconsistent messaging, and misinformation. Recommendations for enhancing trust in public health responses include building relationships with trusted organizations, conducting early media campaigns, and actively addressing misinformation. Purpose: Blood culture (BC) is the gold standard for diagnosing blood stream infections (BSI) but is limited by long turnaround times (TAT) and low detection rate. The T2 Magnetic Resonance method (T2MR) offers a rapid, culture-independent alternative. The objective of this study was to compare the performance of the T2Bacteria assay to BCs in a real-world setting. Methods: Retrospective comparative study consisting of T2Bacteria samples and BCs sampled within 72 h from the T2Bacteria sample. The primary outcome was detections by BC and T2Bacteria, respectively. The secondary outcome was difference in TAT. Results: In total, 640 episodes were included, consisting of 640 T2Bacteria samples and 2,117 BCs. A median of three BCs was collected for each T2Bacteria sample. Overall positivity was 101 (15.8%) by either method. In 29 (28.7%) episodes, both T2Bacteria and BC were concordantly positive. In discordant episodes, 46/101 (45.5%) episodes were T2Bacteria positive/BC negative and 26/101 (25.7%) were T2Bacteria negative/BC positive (McNemar χ2, p < 0,05). In T2Bacteria positive/BC negative episodes, eight had growth of the same microorganism in a non-BC culture. Median (IQR) TAT for BC was 35 h and 30 min (25 h 50 min − 45 h 24 min), compared to 21 h and 3 min (17 h 6 min − 27 h 30 m) for T2Bacteria (p < 0.001), with longer delays for samplings occurring outside work hours. Conclusions: The study highlights a high discordance between T2Bacteria and BC and suggests complementary roles of the methods in BSI diagnostics. Aims: Evaluate the social determinants of health (SDOH) associated with suicidality among female Service Members and Veterans (SMV) in Arizona. Methods: Used data from a statewide Arizona Veteran Survey (n = 1,134) to analyze SDOH associated with suicidality (any self-report of suicidal ideation, suicide attempt, or calling a crisis line). Response data were cross-tabulated and analyzed for statistical significance using a chi-square test with a p-value of p < 0.05. Results: Depression, disability, barriers to treatment, housing instability, feelings of loneliness, and more were associated with increased suicidality among respondents. Substance use within the last 30 days, social support, and Veteran social support were not found to be significantly associated with suicidality. Conclusions: Female SMV populations experiencing social, economic, and health disparities appear to be at higher risk for suicide. These findings suggest that future suicide prevention efforts may be more effective if tailored towards populations in need of additional support and social services. [ABSTRACT FROM AUTHOR]

A comprehensive survey of the octopus fauna around the Americas is presented to facilitate and accelerate the assessment of a full understanding of American octopus biodiversity. Brief accounts are provided summarizing research efforts on octopus species taxonomy, diversity and distribution in different regions of North and South America. Octopus americanus and O. insularis are compared and clearly distinguished from each other and from the closely-related European species, O. vulgaris. The use of genus names Paroctopus and Pinnoctopus is clarified. Included is a discussion of the recent application of genus name Paroctopus to warm-temperate and tropical species of small size, along with a cautionary note about species identifications in the light of past errors in misassigning large-to-giant cold-water species to genus Paroctopus. Related to problems with identifying species of Paroctopus, there is an appended note concerning misidentifications and the importance of thorough species descriptions to obtain species information at the levels of both phenotype and genotype. A lectotype is formally designated for Bathypolypus arcticus (Prosch, 1849); and the status of so-called 'Octopus giganteus' is reviewed briefly. A supplementary online database, AmeriCeph, provides basic information about all known octopus species of the Americas, including the institutional location of type material and the identification of voucher specimens and their depositories. DNA sequences registered in this database are not all fully compatible with barcoding standards. However, a subset of DNA sequences conforming to strict barcode identifications is provided in a second supplementary table, providing barcode sequences directly applicable also to improving standards of seafood traceability. This in turn contributes to building sustainability of exploited octopus fisheries stocks and identification of species suitable for aquaculture trials to meet the increasing commercial demand for octopus worldwide. During infectious disease epidemics, accurate diagnostic testing is key to rapidly identify and treat cases, and mitigate transmission. When a novel pathogen is involved, building testing capacity and scaling testing services at the local level can present major challenges to healthcare systems, public health agencies, and laboratories. This mixed methods study examined lessons learned from the scale-up of SARS-CoV-2 testing services in New York City (NYC), as a core part of NYC's Test & Trace program. Using quantitative and geospatial analyses, the authors assessed program success at maximizing reach, equity, and timeliness of SARS-CoV-2 diagnostic testing services across NYC neighborhoods. Qualitative analysis of key informant interviews elucidated key decisions, facilitators, and barriers involved in the scale-up of SARS-CoV-2 testing services. A major early facilitator was the ability to establish working relationships with private sector vendors and contractors to rapidly procure and manufacture necessary supplies locally. NYC residents were, on average, less than 25 min away from free SARS-CoV-2 diagnostic testing services by public transport, and services were successfully directed to most neighborhoods with the highest transmission rates, with only one notable exception. A key feature was to direct mobile testing vans and rapid antigen testing services to areas based on real-time neighborhood transmission data. Municipal leaders should prioritize fortifying supply chains, establish cross-sectoral partnerships to support and extend testing services, plan for continuous testing and validation of assays, ensure open communication feedback loops with CBO partners, and maintain infrastructure to support mobile services during infectious disease emergencies. On June 1, 2020, NYC Health + Hospitals, in partnership with the NYC Department of Health and Mental Hygiene, other city agencies, and a large network of community partners, launched the New York City Test & Trace (T2) COVID-19 response program to identify and isolate cases, reduce transmission through contact tracing, and provide support to residents during isolation or quarantine periods. In this paper, we describe lessons learned with respect to planning and implementation of case notification and contact tracing. Our findings are based on extensive document review and analysis of 74 key informant interviews with T2 leadership and frontline staff, cases, and contacts conducted between January and September 2022. Interviews elicited respondent background, history of program development, program leadership and structure, goals of the program, program evolution, staffing, data systems, elements of community engagement, trust with community, program reach, timeliness, equity, general barriers and challenges, general facilitators and best practices, and recommendations/improvement for the program. Facilitators and barriers revealed in the interviews primarily revolved around hiring and managing staff, data and technology, and quality of interactions with the public. Based on these facilitators and barriers, we identify suggestions to support effective planning and response for future case notification and contact tracing programs, including recommendations for planning during latent periods, case management and data systems, and processes for outreach to cases and contacts. The COVID-19 pandemic highlighted the importance of addressing social needs in a crisis context. Some US jurisdictions integrated a social service component into case investigation and contact tracing (CI/CT) programs, including the New York City (NYC) Test & Trace (T2) Program; the Take Care initiative referred NYC residents who tested positive or were exposed to COVID-19 to services to support isolation and quarantine and meet basic needs. More research is needed to determine effective implementation strategies for integrating social needs provision into CI/CT programs. To identify barriers and facilitators to the implementation of the Take Care initiative, we conducted key informant interviews with program staff, community-based organization partners, and cases and contacts as part of a larger evaluation of the T2 program. Interviews were recorded, transcribed, and analyzed using rapid qualitative methods. Key facilitators to implementation included utilizing a case management software system, employing strategies to encourage service uptake, leveraging cross-agency collaborations, and partnering with community-based organizations for resource navigation. Barriers identified included external management of the software system, challenges reaching and engaging the public, administrative complications due to shifting collaborations, and management of CBO partners' structure and hiring. Based on our findings, we provide recommendations to support effective planning and implementation of social needs service provision in a crisis context. An open source software package has been developed for Global Navigation Satellite Systems (GNSS) interferometric reflectometry. The gnssrefl package is written in python; it can be installed from the source code, the python packaging index website, or via a docker. It includes modules that download GNSS data and orbit data from global archives. A periodogram is used to retrieve the height of the GNSS antenna over the reflecting surface using signal to noise ratio data. Signals from the Global Positioning System, Glonass, Galileo, and Beidou constellations are supported. Modules are provided to estimate volumetric water content of soil, snow depth/accumulation, and water level. Metastases to the brain are rare in prostate cancer. Here, we describe a patient with two treatment-emergent metastatic lesions, one to the brain with neuroendocrine prostate cancer (NEPC) histology and one to the dural membrane of adenocarcinoma histology. We performed genomic, transcriptomic, and proteomic characterization of these lesions and the primary tumor to investigate molecular features promoting these metastases. The two metastatic lesions had high genomic similarity, including TP53 mutation and PTEN deletion, with the most striking difference being the additional loss of RB1 in the NEPC lesion. Interestingly, the dural lesion expressed both androgen receptor and neuroendocrine markers, suggesting amphicrine carcinoma (AMPC). When analyzing pioneer transcription factors, the AMPC lesion exhibited elevated FOXA1 activity while the brain NEPC lesion showed elevated HOXC10, NFYB, and OTX2 expression suggesting novel roles in NEPC formation or brain tropism. Throughout development, complex networks of cell signaling pathways drive cellular decision-making across different tissues and contexts. The transforming growth factor β (TGF-β) pathways, including the BMP/Smad pathway, play crucial roles in determining cellular responses. However, as the Smad pathway is used reiteratively throughout the life cycle of all animals, its systems-level behavior varies from one context to another, despite the pathway connectivity remaining nearly constant. For instance, some cellular systems require a rapid response, while others require high noise filtering. In this paper, we examine how the BMP-Smad pathway balances trade-offs among three such systems-level behaviors, or "Performance Objectives (POs)": response speed, noise amplification, and the sensitivity of pathway output to receptor input. Using a Smad pathway model fit to human cell data, we show that varying non-conserved parameters (NCPs) such as protein concentrations, the Smad pathway can be tuned to emphasize any of the three POs and that the concentration of nuclear phosphatase has the greatest effect on tuning the POs. However, due to competition among the POs, the pathway cannot simultaneously optimize all three, but at best must balance trade-offs among the POs. We applied the multi-objective optimization concept of the Pareto Front, a widely used concept in economics to identify optimal trade-offs among various requirements. We show that the BMP pathway efficiently balances competing POs across species and is largely Pareto optimal. Our findings reveal that varying the concentration of NCPs allows the Smad signaling pathway to generate a diverse range of POs. Brief, school-based mental health interventions hold promise for reducing barriers to mental health support access, a critical endeavor in light of increasing rates of mental health concerns among youth. However, there is no consensus on whether or not brief school-based interventions are effective at reducing mental health concerns or improving well-being. This systematic review and meta-analysis aims to provide consensus and determine directions for future work. Articles were included if they examined a brief (≤ four sessions or 240 min of intervention time) psychosocial intervention, were conducted within a Pre-K through 12th-grade school setting, included at least one treatment outcome evaluating mental health or well-being, and were published since 2000. A total of 6,702 papers were identified through database searching, of which 81 papers (k studies = 75) were ultimately selected for inclusion. A total of 40,498 students were included across studies and a total of 75 unique interventions were examined. A total of 324 effect sizes were extracted. On average, interventions led to statistically significant improvements in mental health/well-being outcomes versus control conditions up to one-month (g =.18, p =.004), six-month (g =.15, p =.006), and one-year (g =.10, p =.03) post-intervention. There may be benefits to brief school-based interventions from a preventative public health standpoint; future research may focus on how to optimize their real-world utility. Researchers, managers, and policymakers have historically neglected non-game fishes relative to game fishes, and this oversight has extended to invasive non-game fishes in the United States. One such fish—the Eastern Banded Killifish (Fundulus diaphanus diaphanus Lesueur 1817)—has established and rapidly spread in Lake Michigan and connected waters since 2000. Here, we assess potential drivers of the successful invasion of Eastern Banded Killifish, as well as their potential to disrupt native communities and food webs. Specifically, we compare the trophic niche breadth and diet composition between Eastern Banded Killifish and a native subspecies, Western Banded Killifish (Fundulus diaphanus menona Jordan and Copeland 1877), using stable isotope and gut content metabarcoding analyses. Stable isotope analysis showed that Eastern Banded Killifish had a higher variance in littoral dependence and trophic position than Western Banded Killifish, but both stable isotope and gut content metabarcoding analyses revealed an overlap in the diet composition and trophic position between the subspecies. Eastern Banded Killifish may successfully establish outside its native range due to higher feeding variability than Western Banded Killifish, including in habitats historically unused by the native subspecies, but the trophic niche between these two subspecies was similar overall. This study provides insights into the successful invasion of a potentially overlooked non-game fish—Eastern Banded Killifish—while also comparing stable isotope and gut content metabarcoding analyses for an invasive freshwater fish for the first time. Sex differences and age-related changes in the human heart at the tissue, cell, and molecular level have been well-documented and many may be relevant for cardiovascular disease. However, how molecular programs within individual cell types vary across individuals by age and sex remains poorly characterized. To better understand this variation, we performed single-nucleus combinatorial indexing (sci) ATAC- and RNA-Seq in human heart samples from nine donors. We identify hundreds of differentially expressed genes by age and sex and find epigenetic signatures of variation in ATAC-Seq data in this discovery cohort. We then scale up our single-cell RNA-Seq analysis by combining our data with five recently published single nucleus RNA-Seq datasets of healthy adult hearts. We find variation such as metabolic alterations by sex and immune changes by age in differential expression tests, as well as alterations in abundance of cardiomyocytes by sex and neurons with age. In addition, we compare our adult-derived ATAC-Seq profiles to analogous fetal cell types to identify putative developmental-stage-specific regulatory factors. Finally, we train predictive models of cell-type-specific RNA expression levels utilizing ATAC-Seq profiles to link distal regulatory sequences to promoters, quantifying the predictive value of a simple TF-to-expression regulatory grammar and identifying cell-type-specific TFs. Our analysis represents the largest single-cell analysis of cardiac variation by age and sex to date and provides a resource for further study of healthy cardiac variation and transcriptional regulation at single-cell resolution. Analysis of single-nucleus RNA- and ATAC-Seq from healthy human heart revealed age- and sex-specific alterations, defines candidate developmental stage-specific regulatory patterns, and identifies cell type-specific regulatory signatures. Introduction: Penetrating Crohn's disease (CD) often necessitates surgical intervention, with the open approach traditionally favored. Robotic-assisted surgery offers potential benefits but remains understudied in this complex patient population. Additionally, the lack of standardized surgical complexity scoring in CD hinders research and comparisons. Methods: We retrospectively analyzed adult patients with penetrating CD who underwent either robotic-assisted ileocolic resection (RICR) or open ileocolic resection (OICR) at our institution from January 2007 to December 2021. We assessed endpoints, including length of stay, complications, readmissions, reoperations, and other perioperative outcomes. Results: RICR demonstrated safety outcomes comparable to OICR. Importantly, RICR patients experienced significantly reduced estimated blood loss (p < 0.0001), shorter hospital stays (median 4.5 days versus 6.9 days; p = 0.01), lower surgical site infection rates (0% versus 15.4%; p = 0.01), and decreased 30-day readmission rates (0% versus 15.4%; p = 0.01). Linear regression analysis revealed the need for additional strictureplasties (coefficient: 84.8; p = 0.008), colonic resections (coefficient: 41.7; p = 0.008), and estimated blood loss (coefficient: 0.07; p = 0.002) independently correlated with longer operative times). Conclusion: Robotic-assisted surgery appears to be a safe and potentially beneficial alternative for the surgical management of penetrating CD, offering advantages in perioperative outcomes reducing length of stay, blood loss, surgical site infection rates, and readmission rates. High-throughput imaging (HTI) generates complex imaging datasets from a large number of experimental perturbations. Commercial HTI software programs for image analysis workflows typically do not allow full customization and adoption of new image processing algorithms in the analysis modules. While open-source HTI analysis platforms provide individual modules in the workflow, like nuclei segmentation, spot detection, or cell tracking, they are often limited in integrating novel analysis modules or algorithms. Here, we introduce the High-Throughput Image Processing Software (HiTIPS) to expand the range and customization of existing HTI analysis capabilities. HiTIPS incorporates advanced image processing and machine learning algorithms for automated cell and nuclei segmentation, spot signal detection, nucleus tracking, nucleus registration, spot tracking, and quantification of spot signal intensity. Furthermore, HiTIPS features a graphical user interface that is open to integration of new analysis modules for existing analysis pipelines and to adding new analysis modules. To demonstrate the utility of HiTIPS, we present three examples of image analysis workflows for high-throughput DNA FISH, immunofluorescence (IF), and live-cell imaging of transcription in single cells. Background and Objective: Blood-based small molecule metabolites offer easy accessibility and hold significant potential for insights into health processes, the impact of lifestyle, and genetic variation on disease, enabling precise risk prevention. In a prospective study with records of heart failure (HF) incidence, we present metabolite profiling data from individuals without HF at baseline. Methods: We uncovered the interconnectivity of metabolites using data-driven and causal networks augmented with polygenic factors. Exploring the networks, we identified metabolite broadcasters, receivers, mediators, and subnetworks corresponding to functional classes of metabolites, and provided insights into the link between metabolomic architecture and regulation in health. We incorporated the network structure into the identification of metabolites associated with HF to control the effect of confounding metabolites. Results: We identified metabolites associated with higher and lower risk of HF incidence, such as glycine, ureidopropionic and glycocholic acids, and LPC 18:2. These associations were not confounded by the other metabolites due to uncovering the connectivity among metabolites and adjusting each association for the confounding metabolites. Examples of our findings include the direct influence of asparagine on glycine, both of which were inversely associated with HF. These two metabolites were influenced by polygenic factors and only essential amino acids, which are not synthesized in the human body and are obtained directly from the diet. Conclusion: Metabolites may play a critical role in linking genetic background and lifestyle factors to HF incidence. Fishes have spread into previously fishless wetlands, likely affecting other species. In the Prairie Pothole Region of North America, the invasion of fish into wetlands is facilitated by interactions of altered land use, climate, and hydrology. We aimed to understand the effects of fishes on amphipods, which are macroinvertebrates that vertebrates rely on as forage. We hypothesized the presence and abundance of fish, particularly benthivores, would have detrimental effects on amphipod abundance. Our study design targeted a large gradient of amphipod abundances among wetlands, including very high abundances of two amphipod species: Gammarus lacustris and Hyalella azteca. We found that fishless basins had twice as many amphipods as those with fish, on average. Gammarus lacustris were not detected in the presence of Black Bullhead Ameiurus melas. The abundance of both amphipod species had negative associations with the most common fishes, Fathead Minnow Pimephales promelas and Brook Stickleback Culaea inconstans. A multivariate community analysis showed the benthivore-fish functional feeding guild was negatively associated with the amphipod community, as hypothesized. However, our study design captured several wetlands with anomalies of high abundances of both fish and amphipods, obscuring their relationships. Our results aid resource managers by confirming several fish guilds and species are associated with lower abundances of amphipods. These findings can inform resource managers who make decisions about managing for fish and wildlife; for example, they may choose to manage existing fish populations or protect existing wetlands with high amphipod densities from new fish invasions. Safe and effective vaccines have been developed that protect against high-risk strains of HPV, but uptake is relatively low. We previously identified factors such as sexual attitudes and HPV knowledge that impact the intent of Christian parents to vaccinate their children against HPV. We hypothesized that culturally specific interventions in the form of short videos would be effective at improving HPV vaccine intentions and attitudes. We made three short educational videos, one with a Christian focus, one informational about HPV, and one control. Videos were distributed electronically with accompanying surveys, and responses were measured before and after watching a randomly selected video. The religious-focused and educational interventions significantly (p < 0.0001, p = 0.0015) improved intentions towards HPV vaccination. The religiously-focused video also significantly diminished the belief that the HPV vaccine is unnecessary because of a family's values (p = 0.014). Parents significantly credited both interventions with improving their intent to vaccinate their children against HPV (p < 0.001 for both). These results suggest that culturally focused educational interventions are effective at influencing vaccine intentions and attitudes, even when those are based on religious or cultural feelings. Urbanization has important effects on the distribution and persistence of wildlife communities. Urbanization may alter not just the distributions of individual species, but also co-occurrence patterns and thus the potential for interspecific interactions (e.g., competition, predation) that structure wildlife communities. Little is currently known about how urbanization alters species co-occurrence or how these changes shape urban species assemblages. Using tree squirrels as a model functional group, we quantified how urbanization alters species occurrence and co-occurrence patterns to shape species assemblages, and how these effects vary within and among cities. We constructed a multi-species, multi-season occupancy model to identify relationships between tree squirrel occupancy and co-occurrence and local land and tree canopy cover and examined variation in these relationships within and among nine US cities. Species' responses to canopy cover were highly variable among, but less variable within cities, suggesting that even common urban wildlife species may respond differently to urban intensity in different landscape contexts. Species co-occurrence was also highly variable among cities and weakly related to canopy cover within a city. These findings provide important evidence that both environmental attributes and species interactions shape urban wildlife communities. Important for management and conservation, they suggest that tree-canopy cover can particularly support forest species co-occurrence and that managing urban forests to provide high canopy cover could contribute to the diversity of urban wildlife communities in forested ecoregions. Venus, lacking an intrinsic global dipole magnetic field, serves as a textbook example of an induced magnetosphere, formed by interplanetary magnetic fields (IMF) enveloping the planet. Yet, various aspects of its magnetospheric dynamics and planetary ion outflows are complex and not well understood. Here we analyze plasma and magnetic field data acquired during the fourth Venus flyby of the Parker Solar Probe (PSP) mission and show evidence for closed topology in the nightside and downstream portion of the Venus magnetosphere (i.e., the magnetotail). The formation of the closed topology involves magnetic reconnection—a process rarely observed at non-magnetized planets. In addition, our study provides an evidence linking the cold Venusian ion flow in the magnetotail directly to magnetic connectivity to the ionosphere, akin to observations at Mars. These findings not only help the understanding of the complex ion flow patterns at Venus but also suggest that magnetic topology is one piece of key information for resolving ion escape mechanisms and thus the atmospheric evolution across various planetary environments and exoplanets. C–H bond activation enables the facile synthesis of new chemicals. While C–H activation in short-chain alkanes has been widely investigated, it remains largely unexplored for long-chain organic molecules. Here, we report light-driven C–H activation in complex organic materials mediated by 2D transition metal dichalcogenides (TMDCs) and the resultant solid-state synthesis of luminescent carbon dots in a spatially-resolved fashion. We unravel the efficient H adsorption and a lowered energy barrier of C–C coupling mediated by 2D TMDCs to promote C–H activation and carbon dots synthesis. Our results shed light on 2D materials for C–H activation in organic compounds for applications in organic chemistry, environmental remediation, and photonic materials. Background: Individuals with inflammatory bowel disease (IBD) who lack traditional cardiovascular disease (CVD) risk factors, such as young females, are observed to experience adverse CVD outcomes. Whether women with IBD have increased CVD risk after the menopause transition is unclear. Methods: We conducted a survival analysis of Women's Health Initiative (WHI) participants and excluded those with missing IBD diagnosis, model covariate data, follow-up data, or a baseline history of the following CVD outcomes: coronary heart disease (CHD), ischemic stroke, venous thromboembolism (VTE), peripheral arterial disease (PAD). Risk of outcomes between IBD and non-IBD women was performed using Cox proportional hazard models, stratified by WHI trial and follow-up. Models were adjusted for age, socio-demographics, comorbidities (e.g., hypertension, diabetes, hypercholesterolemia, etc.), family history, and lifestyle factors (e.g., smoking, alcohol, physical activity, body mass index, etc.). Results: Of 134,022 WHI participants meeting inclusion criteria, 1367 (1.0%) reported IBD at baseline. Mean baseline age was 63.4 years. After adjusting for age and other confounders, no significant difference was observed between IBD and non-IBD women for the risk of CHD (HR 0.96, 95% CI 0.73–1.24), VTE (HR 1.11, 95% CI 0.81–1.52) or PAD (HR 0.64, 95% CI 0.28–1.42). After adjusting for age, risk of ischemic stroke was significantly higher (HR 1.41, 95% CI 1.06–1.88) in IBD than non-IBD women. With further adjustment, the excess risk of ischemic stroke among IBD women was attenuated and no longer statistically significant (HR 1.31, 95% CI 0.98–1.76). We propose a multistart algorithm to identify all local minima of a constrained, nonconvex stochastic optimization problem. The algorithm uniformly samples points in the domain and then starts a local stochastic optimization run from any point that is the "probabilistically best" point in its neighborhood. Under certain conditions, our algorithm is shown to asymptotically identify all local optima with high probability; this holds even though our algorithm is shown to almost surely start only finitely many local stochastic optimization runs. We demonstrate the performance of an implementation of our algorithm on nonconvex stochastic optimization problems, including identifying optimal variational parameters for the quantum approximate optimization algorithm. Introduction: Presacral neuroendocrine neoplasms (PNENs) are rare tumors, with limited data on management and outcomes. Methods: A retrospective review of institutional medical records was conducted to identify all patients with PNENs between 2008 and 2022. Data collection included demographics, symptoms, imaging, surgical approaches, pathology, complications, and long-term outcomes. Results: Twelve patients were identified; two-thirds were female, averaging 44.8 years of age, and, for the most part, presenting with back pain, constipation, and abdominal discomfort. Preoperative imaging included computed tomography scans and magnetic resonance images, with somatostatin receptor imaging and biopsies being common. Half of the patients had metastatic disease on presentation. Surgical approach varied, with anterior, posterior, and combined techniques used, often involving muscle transection and coccygectomy. Short-term complications affected one-quarter of patients. Pathologically, PNENs were mainly well-differentiated grade 2 tumors with positive synaptophysin and chromogranin A. Associated anomalies were common, with tail-gut cysts prevalent. Mean tumor diameter was 6.3 cm. Four patients received long-term adjuvant therapy. Disease progression necessitated additional interventions, including surgery and various chemotherapy regimens. Skeletal, liver, thyroid, lung, and pancreatic metastases occurred during follow-up, with no mortality reported. Kaplan–Meier analysis showed a 5-year local recurrence rate of 23.8%, disease progression rate of 14.3%, and de novo metastases rate of 30%. Conclusion: The study underscores the complex management of PNENs and emphasizes the need for multicenter research to better understand and manage these tumors. Summary: Long-term physical functioning trajectories following distal forearm fracture are unknown. We found that women with versus those without distal forearm fracture were more likely to experience a 5-year decline in physical functioning, independent of initial physical functioning level. This association was most evident among women 80 years and older. Introduction: Physical functioning trajectory following lower arm or wrist fracture is not well understood. Purpose: This study is to evaluate physical functioning trajectory before vs. after lower arm or wrist fracture, stratified by age. Methods: We performed a nested case–control study of prospective data from the Women's Health Initiative Study (n = 2097 cases with lower arm or wrist fracture, 20,970 controls). Self-reported fractures and the physical functioning subscale of the RAND 36-item Short-Form Health Survey were assessed annually. We examined three physical functioning trajectory groups: stable, improving, and declining. Results: Mean (SD) number of physical functioning measurements was 5.2 (1.5) for cases and 5.0 (1.4) for controls. Declining physical functioning was observed among 20.4% of cases and 16.0% of controls. Compared to women without lower arm or wrist fracture, women with lower arm or wrist fracture were 33% more likely to experience declining physical functioning (adjusted odds ratio [aOR] 1.33 95% confidence interval [CI] 1.19–1.49, reference group stable or improving physical functioning trajectory). Associations varied by age: age ≥ 80 years aOR 1.56 (95% CI 1.29–1.88); age 70–79 years aOR 1.29 (95% CI 1.09–1.52); age < 70 years aOR 1.15 (95% CI 0.86–1.53) (pinteraction = 0.06). Associations between lower arm or wrist fracture and odds of declining physical functioning did not vary by baseline physical functioning or physical activity level. The Europa Thermal Emission Imaging System (E-THEMIS) on the Europa Clipper spacecraft will investigate the temperature and physical properties of Europa using thermal infrared (TIR) images in three wavelength bands centered from 7-14 μm, 14-28 μm and 28-80 μm. E-THEMIS will map >80% of the surface Europa at multiple times of day at a resolution of 8-km per pixel, ∼32% percent of the surface at ≤1 km/pixel resolution, and ∼6% percent at ≤100 m/pixel resolution. The specific objectives of the investigation are to 1) understand the formation of surface features, including sites of recent or current geologic activity, in order to understand regional and global processes and evolution and 2) to identify safe sites for future landed missions. E-THEMIS uses an uncooled microbolometer detector array for the IR focal plane. The E-THEMIS focal plane has 920 cross-track pixels (896 active) and 140 along-track pixels in each of the three spectral bands. The image data are collected at 14-bits per pixel at a frame rate of 60 Hz. The instrument can operate in framing mode, where full frame images are collected, and optionally co-added in time, in each band, or in time-delay-integration (TDI) mode where consecutive rows from each band are offset spatially to remove the spacecraft motion and then summed. In addition, the data in each band can be spatially aggregated from 2 × 2 to 5 × 5 pixels. These modes will be varied throughout each Europa flyby to optimize the data precision while fitting within the E-THEMIS data allocation. The expected temperature precision, measured as the noise equivalent spectral radiance, is 1.2 K at scene temperatures ≥90 K for a TDI of 16 with 4 × 4 pixel coaggregation in Band 2. The absolute accuracy at 90 K is 2−3 K in Band 2. E-THEMIS is an all-reflective, three-mirror anastigmat telescope with a 6.45-cm effective aperture and a speed of f/1.34 cross-track and 1.92 along-track. The mass of instrument Sensor Assembly, mounted on the spacecraft nadir deck, is 11.4 kg, the vault electronics are 1.8 kg, and the two are connected through a 3.1 kg harness. The Sensor volume is 23.7 cm x 31.8 cm x 29.8 cm. E-THEMIS consumes an average operation power of 34.8 W at 28 V. We consider a conjecture that identifies two types of base point free divisors on M ¯ 0 , n . The first arises from Gromov-Witten theory of a Grassmannian. The second comes from first Chern classes of vector bundles associated with simple Lie algebras in type A. Here we reduce this conjecture on M ¯ 0 , n to the same statement for n = 4. We present a cut finite element method for the heat equation on two overlapping meshes: a stationary background mesh and an overlapping mesh that evolves inside/"on top" of it. Here the overlapping mesh is prescribed by a simple discontinuous evolution, meaning that its location, size, and shape as functions of time are discontinuous and piecewise constant. For the discrete function space, we use continuous Galerkin in space and discontinuous Galerkin in time, with the addition of a discontinuity on the boundary between the two meshes. The finite element formulation is based on Nitsche's method. The simple discontinuous mesh evolution results in a space-time discretization with a slabwise product structure between space and time which allows for existing analysis methodologies to be applied with only minor modifications. We follow the analysis methodology presented by Eriksson and Johnson (SIAM J Numer Anal 28(1):43–77, 1991; SIAM J Numer Anal 32(3):706–740, 1995). The greatest modification is the introduction of a Ritz-like "shift operator" that is used to obtain the discrete strong stability needed for the error analysis. The shift operator generalizes the original analysis to some methods for which the discrete subspace at one time does not lie in the space of the stiffness form at the subsequent time. The error analysis consists of an a priori error estimate that is of optimal order with respect to both time step and mesh size. Anthropogenic change is contributing to the rise in emerging infectious diseases, which are significantly correlated with socioeconomic, environmental and ecological factors1. Studies have shown that infectious disease risk is modified by changes to biodiversity2–6, climate change7–11, chemical pollution12–14, landscape transformations15–20 and species introductions21. However, it remains unclear which global change drivers most increase disease and under what contexts. Here we amassed a dataset from the literature that contains 2,938 observations of infectious disease responses to global change drivers across 1,497 host–parasite combinations, including plant, animal and human hosts. We found that biodiversity loss, chemical pollution, climate change and introduced species are associated with increases in disease-related end points or harm, whereas urbanization is associated with decreases in disease end points. Natural biodiversity gradients, deforestation and forest fragmentation are comparatively unimportant or idiosyncratic as drivers of disease. Overall, these results are consistent across human and non-human diseases. Nevertheless, context-dependent effects of the global change drivers on disease were found to be common. The findings uncovered by this meta-analysis should help target disease management and surveillance efforts towards global change drivers that increase disease. Forest growth varies across landscapes due to the intricate relationships between various environmental drivers and forest management. In this study, we analysed the variation of tree growth potential across a landscape scale and its relation to soil moisture. We hypothesised that soil moisture conditions drive landscape-level variation in site quality and that intermediate soil moisture conditions demonstrate the highest potential forest production. We used an age-independent difference model to estimate site quality in terms of maximum achievable tree height by measuring the relative change in Lorey's mean height for a five year period across 337 plots within a 68 km2 boreal landscape. We achieved wall-to-wall estimates of site quality by extrapolating the modelled relationship using repeated airborne laser scanning data collected in connection to the field surveys. We found a clear decrease in site quality under the highest soil moisture conditions. However, intermediate soil moisture conditions did not demonstrate clear site quality differences; this is most likely a result of the nature of the modelled soil moisture conditions and limitations connected to the site quality estimation. There was considerable unexplained variation in the modelled site quality both on the plot and landscape levels. We successfully demonstrated that there is a significant relationship between soil moisture conditions and site quality despite limitations associated with a short study period in a low productive region and the precision of airborne laser scanning measurements of mean height. This is a first-of-its-kind dataset containing detailed purchase histories from 5027 U.S. Amazon.com consumers, spanning 2018 through 2022, with more than 1.8 million purchases. Consumer spending data are customarily collected through government surveys to produce public datasets and statistics, which serve public agencies and researchers. Companies now collect similar data through consumers' use of digital platforms at rates superseding data collection by public agencies. We published this dataset in an effort towards democratizing access to rich data sources routinely used by companies. The data were crowdsourced through an online survey and shared with participants' informed consent. Data columns include order date, product code, title, price, quantity, and shipping address state. Each purchase history is linked to survey data with information about participants' demographics, lifestyle, and health. We validate the dataset by showing expenditure correlates with public Amazon sales data (Pearson r = 0.978, p < 0.001) and conduct analyses of specific product categories, demonstrating expected seasonal trends and strong relationships to other public datasets. Purpose: Infantile hypertrophic pyloric stenosis (IHPS) is suspected to have worse outcomes when length of illness prior to presentation is prolonged. Our objective was to evaluate how social determinants of health influence medical care and outcomes for babies with IHPS. Methods: A retrospective review was performed over 10 years. Census data were used as proxy for socioeconomic status via Geo-Identification codes and correlated with food access and social vulnerability variables. The cohort was subdivided to understand the impact of Medicaid Managed Care (MMC). Results: The cohort (279 cases) was divided into two groups; early group from 2011 to 2015 and late from 2016 to 2021. Cases in the late group were older at the time of presentation (41.5 vs. 36.5 days; p = 0.022) and presented later in the disease course (12.8 vs. 8.9 days; p = 0.021). There was no difference in race (p = 0.282), gender (p = 0.874), or length of stay. Conclusions: Patients who presented with IHPS after implementation of phased MMC were older, had a longer symptomatic course, and shorter pylorus measurements. Patients with public insurance after the implementation of MMC were more likely to follow-up with an outpatient pediatrician within a month of hospitalization.
