Analyses of food-borne pathogens are of great importance in order to minimize the risk of infection for customers. These analyses should be as fast as possible. Any detection method requires enrichment and quantitative analysis of the enriched microbes. Conventional enrichment methods, which take several days, need to be replaced by faster techniques such as biomagnetic separation (BMS). This technique involves the use of paramagnetic microspheres coated with ligands that have special affinity to the microbes that have to be detected. In the studies reported here, enrichment experiments by BMS were carried out using the non-pathogenic E. coli DSM 498 as a model strain and beads coated with a polyethylenimine (PEI) anion-exchange material. The results show that the number of cells separated, as a proportion of the total, was positively correlated with the bead concentration and the length of the period they were mixed together. In addition, a mathematical model, based on the rate of impact between two different sorts of particles, was developed to describe the proportion of separated cells as a function of incubation time and the concentration, size and density of the beads and cells. This is the first mathematical description of cell–bead interactions to be based on well-understood physicochemical principles. The model was confirmed by separation experiments in which the concentration of beads and the incubation period were varied. Weisse Elster River sediment from the Leipzig Lowlands region (Saxony, Germany) is anthropogenically polluted by heavy metals. Sediment dredged from a trap to the south of Leipzig was characterized in detail. When freshly dredged sediment contacts air, the material turns acidic because of oxidation processes, the heavy metals become soluble and the sediment poses an environmental risk. We are therefore developing a sediment-treatment process based on heavy metal removal by bioleaching. Leaching experiments were carried out in suspension and in the solid bed. The heavy metals were solubilized to nearly the same extent by H2SO4 dosage (pure chemical extraction) and addition of elemental sulphur (microbial oxidation of S0 to H2SO4). With increasing dosage of the leaching agent, Zn, Cd, Ni, Cu and Cr were more and more solubilized, whereas Pb was only dissolved in small amounts. The addition of 2% S0 is considered an optimum dosage. When 5% S0 was added to the sediment, the pH dropped to 1.76 and large amounts of undesirable compounds such as Ca, Al and Fe were solubilized. The higher the temperature, the faster the metals were solubilized in both suspension and the solid bed. The temperature optimum for activating the indigenous S0-oxidizing microbes of the sediment lies between 30 and 40 °C. Conditioning of freshly dredged sediment with plants makes it suitable for solid-bed leaching; the kinetics of heavy metal solubilization from sediment conditioned for 6 months with Phragmites australis was the same as from long-term stored sediment. After PEG placement at the Medical Department of the University Hospital in Kiel, 210 patients (mean age 61.3 years; 137 men, 73 women) were prospectively followed-up for 133+/-181 days. Close-meshed evaluations of the development of nutritional status, long-term outcome, complications, subjective acceptability, patient care after discharge from the hospital, survival, and nutritional long-term problems were performed. The PEG procedure (duration 13.3+/-4.2 min) was carried out for neurological (42%), ear-nose-throat (28%), and internal medical (30%) indications. Procedure-related mortality was 0%, while altogether 3.8% severe and 20.0% mild complications were observed. Body weight decreased by a mean of 11.4+/-1.5 kg in the three months before and increased by 3.5+/-1.7 kg one year after PEG placement with no significant differences between malignant or benign underlying diseases. Individual subjective acceptability was excellent in 83%, sufficient in 15%, and poor in 2% of patients only. One-year survival rate was 34.3%. The various results of the present prospective study demonstrate that long-term enteral feeding via PEG is a safe, effective, easy-to-practice, and highly acceptable method with excellent long-term results and distinct improvement of nutritional status. Studies of microbial purification of a model waste water containing 4-nitrophenol were carried out in a continuously working aerobic solid-bed reactor. The main emphasis was on the dynamic behaviour of the system after a sudden change in cultivation conditions and on the steady-state performance of the reactor as a function of the pollution load. A change from ammonium-free to ammonium-containing medium hardly influenced the nitrophenol degradation. The reactor responded differently to an increase in pollutant load, which was brought about by increasing either the 4-nitrophenol content or the flow of the waste water. Up to a load of 270 mg l−1 h−1 the pollutant was stably and almost completely degraded. At a higher load, only a partial 4-nitrophenol degradation took place. A mathematical model was derived to describe the processes that occurred in the reactor. By segregation into two compartments – the aqueous phase and the biofilm – account was taken of the fact that the pollutant is carried into the biofilm by diffusion and is degraded there. The observed relations between the pollutant load, the pollutant concentration in the outlet of the reactor and the reactor performance agreed with the simulated process behaviour. The microbial degradation of hydrocarbons was studied in an artificially contaminated sandy soil, using a pilot-scale percolator system. After a short lag period, an intensive degradation occurred, which diminished in time and completely stopped in the end, despite large residual contaminations (residues of 56% diesel fuel, 20% n-hexadecane and 3.5% phenanthrene at the initial loadings of each 3000 mg/kg). The remaining pollutant content was influenced by the kind of hydrocarbon but was nearly independent of its initial loading. According to a model-aided analysis of the carbon dioxide production during remediation, the observed stagnation of degradation was caused by a limited bioavailability of the pollutants. The degradation in the soil-free aqueous phase was more extensive than in the soil, which suggests that the limited bioavailability in the soil can be attributed mainly to matrix-dependent rather than substrate-dependent influences. Generally, fine particles and organic matter are mainly responsible for the adsorption of pollutants to the soil matrix. Our sandy soil also bound hydrocarbons adsorptively although it contained neither silty material nor significant amounts of organic matter. As shown by Brunauer Emmett Teller (BET) analysis, the soil particles were covered by micropores, which enlarged the soil surface by a factor of 120 in comparison with the macroscopic surface area. As an alternative to chemical thrombolysis, an acutely occluded transjugular intrahepatic portosystemic stent shunt in a 72-year-old woman was successfully recanalized using a 5 Fr rheolytic catheter system. No adjunctive thrombectomy treatment was necessary. The underlying stenotic leason was treated with percutaneous transluminal angioplasty and a Palmaz stent. Aim of the present study was to evaluate the role of cellular uptake of dietary [3H]putrescine for the regulation of pancreatic, hepatic, and small intestinal polyamine metabolism during normal and camostate-induced pancreatic growth in rats in vivo. Initially dose-response and time-course studies of [3H]putrescine uptake were performed. Male Wistar rats were either treated with the synthetic trypsin inhibitor camostate (200 mg/kg body wt orally twice daily), camostate plus the ornithine decarboxylase inhibitor alpha-difluoromethylornithine (DFMO) (2% in drinking water plus 3 x 300 mg/kg body wt intraperitoneally during daytime) or saline as controls. After 4, 8, 12, 24, 36, 48, or 120 hr, five to seven animals per group were killed, respectively. Orally fed [3H]putrescine (10 nmol/kg body wt. 2 hr prior to death) is rapidly taken up and further metabolized to spermidine in normal growing pancreas, liver, and small intestine. Feeding of camostate significantly enhanced dietary [3H]putrescine uptake, while simultaneous inhibition of de novo synthesis of intracellular polyamines by DFMO resulted in a highly significant further increase in cellular uptake of orally fed [3H]putrescine, which is immediately metabolized to spermidine. The present in vivo data confirm the important role of dietary putrescine uptake for the maintenance of intracellular polyamine pool in normal and stimulated pancreatic growth. Furthermore, dietary putrescine uptake is an important regulatory mechanism to maintain the normal and growth-stimulated cellular polyamine pool in the pancreas after potent simultaneous inhibition of intracellular de novo polyamine synthesis. The patterns of activity of the nucleolus organizer regions (NORs) in the spermatogeneses of ten species of all non-mammalian classes of vertebrates and one species of the cephalochordates were investigated with the silver (Ag)-staining technique. The Ag-stainability of the NORs is a measure of the transcriptional activity of the ribosomal RNA genes. In all species, there is a very similar pattern of NOR-activity in the various stages of Spermatogenesis. The qualitative analysis of the Ag-stainability of the NORs was in very good agreement with the results obtained for mammals: Ag-stained NORs are detectable during the entire meiotic prophase up to the pachytene stage, completely absent in the meiotic metaphases I and II, and again demonstrable in early spermatid nuclei. The results confirm the occurrence of postmeiotic reactivation of the RNA genes. The preferential inhibition of rRNA synthesis by low doses of actinomycin D induced a rapid decline of the Ag-stainability of the postmeiotically reactivated NORs. Microbial hydrocarbon degradation in soil was studied during periodical aerobic/anaerobic switching and under purely aerobic conditions by using a pilot-scale plant with diesel-fuel-contaminated sand. The system worked according to the percolation principle with controlled circulation of process water and aeration. Periodical switching between 4 h of aerobic and 2 h of anaerobic conditions was achieved by repeated saturation of the soil with water. Whatever the cultivation mode, less than 50% of the diesel was degraded after 650 h because the hydrocarbons were adsorbed. Contrary to expectations, aerobic/anaerobic changes neither accelerated the rate of degradation nor reduced the residual hydrocarbon content of the soil. Obviously the pollutant degradation rate was determined mainly by transport phenomena and less by the efficiency of microbial metabolism. The total mass of oxygen consumed and carbon dioxide produced was greater under aerobic/anaerobic changing than under aerobic conditions, although the mass of hydrocarbons degraded was nearly the same. As shown by an overall balance of microbial growth and by a carbon balance, the growth yield coefficient was smaller during aerobic/anaerobic changes than under aerobic conditions. An analytical method for the quantitative determination of Al, Ti, V, Cr, Co, Ni, Cu, Zn, Mo, Cd, Ti and Pb in plant samples by ICP-MS has been developed. Spectral interferences, plant matrix effects, precision and accuracy are discussed.
