Organismal functional strategies form a continuum from slow- to fast-growing organisms, in response to common drivers such as resource availability and disturbance. However, whether there is synchronisation of these strategies at the entire community level is unclear. Here, we combine trait data for >2800 above- and belowground taxa from 14 trophic guilds spanning a disturbance and resource availability gradient in German grasslands. The results indicate that most guilds consistently respond to these drivers through both direct and trophically mediated effects, resulting in a 'slow-fast' axis at the level of the entire community. Using 15 indicators of carbon and nutrient fluxes, biomass production and decomposition, we also show that fast trait communities are associated with faster rates of ecosystem functioning. These findings demonstrate that 'slow' and 'fast' strategies can be manifested at the level of whole communities, opening new avenues of ecosystem-level functional classification. Although co-occurring species may differ widely in their response traits, coordinated functional trait shifts may emerge at the community level in response to environmental factors. Soil sorption properties can influence the bioavailability of substances and consequently the toxicity for soil organisms. Current standardised laboratory testing for the exposure assessment of pesticides to soil organisms uses OECD artificial soil that does not reflect the high variation in chemical-physical soil properties found in natural agroecosystems. According to guideline OECD 222, earthworm reproduction tests with Eisenia fetida and the pesticide carbendazim were performed in four natural soils and OECD artificial soil. By using pF 1.6, which ensures a uniformity in actual soil water availability, the control reproduction performance of E. fetida in all natural soils was at the same level as OECD artificial soil. In a principle component analysis, the variation in toxicity between the tested soils was attributable to a combination of two soil properties, namely total organic carbon content (TOC) and pH. The largest difference of 4.9-fold was found between the typical agricultural Luvisol with 1.03% TOC and pH 6.2 (EC10: 0.17 (0.12–0.21) mg a.i. kg−1 sdw, EC50: 0.36 (0.31–0.40) mg a.i. kg−1 sdw) and OECD artificial soil with 4.11% TOC and pH 5.6 (EC10: 0.84 (0.72–0.92) mg a.i. kg−1 sdw, EC50: 1.07 (0.99–1.15) mg a.i. kg−1 sdw). The use of typical agricultural soils in standardised laboratory earthworm testing was successfully established with using the measure pF for soil moisture adjustment. It provides a more application-oriented approach and could serve as a new tool to refine the environmental risk assessment at lower tier testing or in an intermediate tier based approach. Soil respiration is an important pathway of soil organic carbon losses in temperate grasslands; however, it is rarely studied across broad management intensity gradients in a landscape. Using the soda-lime method, we measured in-situ soil CO2 efflux with single measurements of long exposure time (i.e. 3 day long) in 150 grasslands in three German regions in early summer 2018 and 2019. The grasslands ranged from unfertilized and grazed grasslands to intensively fertilized and frequently harvested ones. To assess effects of grazing and fertilization intensities and plant diversity on soil CO2 efflux, we used Structural Equation Modeling to account for direct effects and indirect effects through soil and plant organic matter quantity and quality. Soil CO2 efflux was suppressed by limited water availability caused by naturally occurring droughts in both study years. Under the prevailing environmental conditions, grazing intensity, plant biomass and plant C:N ratio were not related to soil CO2 efflux. In contrast, fertilization intensity was positively associated with soil CO2 efflux (standardized coefficient of net effect: + 0.04 in 2018 and + 0.03 in 2019). This was because fertilization led to lower plant species richness and, thus, to lower C:N ratios in soils, which were associated with higher soil CO2 efflux (plant species richness net effect: −0.09 in 2018 and −0.18 in 2019; soil C:N ratio direct effect: −0.23 in 2018 and −0.33 in 2019). Grassland management intensity influences nutrient cycling both directly, by changing nutrient inputs and outputs from the ecosystem, and indirectly, by altering the nutrient content, and the diversity and functional composition of plant and microbial communities. However, the relative importance of these direct and indirect processes for the leaching of multiple nutrients is poorly studied. We measured the annual leaching of nitrate, ammonium, phosphate and sulphate at a depth of 10 cm in 150 temperate managed grasslands using a resin method. Using Structural Equation Modeling, we distinguished between various direct and indirect effects of management intensity (i.e. grazing and fertilization) on nutrient leaching. We found that management intensity was positively associated with nitrate, ammonium and phosphate leaching risk both directly (i.e. via increased nutrient inputs) and indirectly, by changing the stoichiometry of soils, plants and microbes. In contrast, sulphate leaching risk was negatively associated with management intensity, presumably due to increased outputs with mowing and grazing. In addition, management intensification shifted plant communities towards an exploitative functional composition (characterized by high tissue turnover rates) and, thus, further promoted the leaching risk of inorganic nitrogen. Plant species richness was associated with lower inorganic nitrogen leaching risk, but most of its effects were mediated by stoichiometry and plant community functional traits. Microplastics (MP, plastic particles between 0.1 and 5000 μm) contaminate agricultural soils through the application of organic fertilizers, sewage sludge, and plastic mulch. MP surfaces and the MP-soil interface provide specific habitats for soil microorganisms—the plastisphere. Microorganisms in the plastisphere may benefit from utilizing MP as a carbon (C) source. Hydrolyzable MP with ester bonds are susceptible to enzymatic depolymerization by hydrolysis. In a microcosm experiment, we investigated MP biodegradation of small and large (< 0.5 mm and 0.5–2 mm respectively), hydrolyzable (a poly(lactic acid)/poly(butylene co-adipate terephthalate) blend, PLA/PBAT) and non-hydrolyzable (low-density polyethylene, LDPE) polymers, and the effects of these MP on microorganisms in dry and wet MP-amended soil. MP affected neither abundance and composition of the main soil microbial groups (fungi, Gram-negative, and Gram-positive bacteria), specific activities of ß-glucosidase, ß-xylosidase, lipase, and phenoloxidase, nor respiration in MP-amended soil. Only large PLA/PBAT particles in dry soil were significantly mineralized (15.4% of initial PLA/PBAT-C after 230 days). PLA/PBAT mineralization coincided with enhanced lipase and ß-glucosidase activities on the surfaces of individual PLA/PBAT particles extracted from the soil after incubation (compared to LDPE and non-incubated PLA/PBAT particles). We detected cracks on the surfaces of PLA/PBAT particles using scanning electron microscopy, indicating initiation of MP biodegradation, presumably due to depolymerization by lipases. Results suggest that the PLA/PBAT plastisphere is a polymer-specific habitat for lipase-producing soil microorganisms. Our study demonstrates that analyzing biogeochemical interactions within polymer-specific plastispheres is essential to assess MP fate and their impacts on microbially driven soil processes. Sustainable forest management requires understanding of ecosystem phosphorus (P) cycling. Lang et al. (2017) [Biogeochemistry,https://doi.org/10.1007/s10533-017-0375-0] introduced the concept of P-acquiring vs. P-recycling nutrition strategies for European beech (Fagus sylvatica L.) forests on silicate parent material, and demonstrated a change from P-acquiring to P-recycling nutrition from P-rich to P-poor sites. The present study extends this silicate rock-based assessment to forest sites with soils formed from carbonate bedrock. For all sites, it presents a large set of general soil and bedrock chemistry data. It thoroughly describes the soil P status and generates a comprehensive concept on forest ecosystem P nutrition covering the majority of Central European forest soils. For this purpose, an Ecosystem P Nutrition Index (ENIP) was developed, which enabled the comparison of forest P nutrition strategies at the carbonate sites in our study among each other and also with those of the silicate sites investigated by Lang et al. (2017). The P status of forest soils on carbonate substrates was characterized by low soil P stocks and a large fraction of organic Ca-bound P (probably largely Ca phytate) during early stages of pedogenesis. Soil P stocks, particularly those in the mineral soil and of inorganic P forms, including Al- and Fe-bound P, became more abundant with progressing pedogenesis and accumulation of carbonate rock dissolution residue. Phosphorus-rich impure, silicate-enriched carbonate bedrock promoted the accumulation of dissolution residue and supported larger soil P stocks, mainly bound to Fe and Al minerals. In carbonate-derived soils, only low P amounts were bioavailable during early stages of pedogenesis, and, similar to P-poor silicate sites, P nutrition of beech forests depended on tight (re)cycling of P bound in forest floor soil organic matter (SOM). In contrast to P-poor silicate sites, where the ecosystem P nutrition strategy is direct biotic recycling of SOM-bound organic P, recycling during early stages of pedogenesis on carbonate substrates also involves the dissolution of stable Ca-Porg precipitates formed from phosphate released during SOM decomposition. In contrast to silicate sites, progressing pedogenesis and accumulation of P-enriched carbonate bedrock dissolution residue at the carbonate sites promote again P-acquiring mechanisms for ecosystem P nutrition. Experiments showed that biodiversity increases grassland productivity and nutrient exploitation, potentially reducing fertiliser needs. Enhancing biodiversity could improve P-use efficiency of grasslands, which is beneficial given that rock-derived P fertilisers are expected to become scarce in the future. Here, we show in a biodiversity experiment that more diverse plant communities were able to exploit P resources more completely than less diverse ones. In the agricultural grasslands that we studied, management effects either overruled or modified the driving role of plant diversity observed in the biodiversity experiment. Nevertheless, we show that greater above- (plants) and belowground (mycorrhizal fungi) biodiversity contributed to tightening the P cycle in agricultural grasslands, as reduced management intensity and the associated increased biodiversity fostered the exploitation of P resources. Our results demonstrate that promoting a high above- and belowground biodiversity has ecological (biodiversity protection) and economical (fertiliser savings) benefits. Such win-win situations for farmers and biodiversity are crucial to convince farmers of the benefits of biodiversity and thus counteract global biodiversity loss. Relationships between biodiversity and phosphorus cycling and the underlying processes are complex. To improve our understanding of early microbial colonization of pristine minerals and their group-specific C utilization, we exposed minerals (illite/goethite/quartz) amended with artificial root exudates (ARE, glucose, and citric acid) in grassland soils for a period of 24 weeks. FTIR spectra indicated that mineral-associated ARE were used within the first 2 weeks of exposure and were replaced by other carbohydrates derived from living or dead cells as well as soil-borne C sources transported into the mineralosphere after heavy rain events. Fungi and Gram-positive bacteria incorporated ARE-derived C more rapidly than Gram-negative bacteria. Gram-negative bacteria presumably profited indirectly from the ARE by cross-feeding on mineral-associated necromass of fungi and Gram-positive bacteria. The Gram-negative bacterial phyla Verrucomicrobia, Planctomycetes, Gemmatimonadetes, Armatimonadetes, and Chloroflexi showed a positive correlation with Gram-negative PLFA abundances. After 24 weeks of exposure in the grassland soils, abundances of soil microorganisms in the mineralosphere reached only 3.1% of the population density in soil. In conclusion, both bacteria and fungi slowly colonize new surfaces such as pristine minerals, but quickly assimilate artificial root exudates, creating an active microbial community in the mineralosphere. Aims: An essential task of agricultural systems is to improve internal phosphorus (P) recycling. Cover crops and tillage reduction can increase sustainability, but it is not known whether stimulation of the soil microbial community can increase the availability of soil organic P pools. Methods: In a field experiment in southwest Germany, the effects of a winter cover crop mixture (vs. bare fallow) and no-till (vs. non-inversion tillage) on microbial P-cycling were assessed with soybean as the main crop. Microbial biomass, phospholipid fatty acids (PLFAs), P cycling enzymes, and carbon-substrate use capacity were linked for the first time with the lability of organic P pools measured by enzyme addition assays (using phosphodiesterase, non-phytase-phosphomonoesterase and fungal phytase). Results: Microbial phosphorus, phosphatase, and fatty acids increased under cover crops, indicating an enhanced potential for organic P cycling. Enzyme-stable organic P shifted towards enzyme-labile organic P pools. Effects of no-till were weaker, and a synergy with cover crops was not evident. Conclusions: In this experiment, cover crops were able to increase the microbially mediated internal P cycling in a non-P-limited, temperate agroecosystems. [ABSTRACT FROM AUTHOR]

Tree roots penetrate the soil to several meters depth, but the role of subsoils for the supply of nutrient elements such as phosphorus (P) to the trees is poorly understood. Here, we tested the hypothesis that increased P deficiency in the topsoil results in an increased microbial recycling of P from the forest subsoil. We sampled soils from four German temperate forest sites representing a gradient in total P stocks. We analyzed the oxygen isotopic composition of HCl-extractable phosphate (δ18OP) and identified differences in P speciation with increasing soil depth using X-ray absorption near-edge structure (XANES) spectroscopy. We further determined microbial oxygen demand with and without nutrient supply at different soil depths to analyse nutrient limitation of microbial growth and used nanoscale secondary ion mass spectrometry (NanoSIMS) to visualize spatial P gradients in the rhizosphere. We found that δ18OP values in the topsoil of all sites were close to the isotopic signal imparted by biological cycling when oxygen isotopes in phosphate are exchanged by enzymatic activity. However, with increasing soil depth and increasing HCl-P concentrations, δ18Ο values continuously decreased towards values expected for primary minerals in parent material at depths below 60 cm at sites with high subsoil P stocks and below more than 2 m at sites with low subsoil P stocks, respectively. For these depths, XANES spectra also indicated the presence of apatite. NanoSIMS images showed an enrichment of P in the rhizosphere in the topsoil of a site with high P stocks, while this P enrichment was absent at a site with low P stocks and in both subsoils. Addition of C, N and P alone or in combination revealed that microbial activity in subsoils of sites with low P stocks was mostly P limited, whereas sites with high P stocks indicated N limitation or N and P co-limitation. We conclude that subsoil P resources are recycled by trees and soil microorganisms. With continued weathering of the bedrock and mobilisation of P from the weathered rocks, P cycling will proceed to greater depths, especially at sites characterised by P limitation. Background: Phosphorus (P) is a limiting nutrient in many agroecosystems and costly fertilizer inputs can cause negative environmental impacts. Cover crops constitute a promising management option for sustainable intensification of agriculture. However, their interactions with the soil microbial community, which is a key driver of P cycling, and their effects on the following crop, have not yet been systematically assessed.Scope: We conducted a meta-analysis of published field studies on cover crops and P cycling, focusing on plant-microbe interactions.Conclusions: We describe several distinct, simultaneous mechanisms of P benefits for the main crop. Decomposition dynamics, governed by P concentration, are critical for the transfer of P from cover crop residues to the main crop. Cover crops may enhance the soil microbial community by providing a legacy of increased mycorrhizal abundance, microbial biomass P, and phosphatase activity. Cover crops are generally most effective in systems low in available P, and may access 'unavailable' P pools. However, their effects on P availability are difficult to detect by standard soil P tests, except for increases after the use of Lupinus sp. Agricultural management (i.e. cover crop species selection, tillage, fertilization) can improve cover crop effects. Aims: The aim of our study was to clarify whether inoculating a soil with Pseudomonas sp. RU47 (RU47) bacteria would stimulate the enzymatic cleavage of organic P compounds in the rhizosphere and bulk soil, promoting plant growth. Adding either viable or heat treated RU47 cells made it possible to separate direct from indirect effects of the inoculum on P cycling in soil and plants.Methods: We performed a rhizobox experiment in the greenhouse with tomato plants (Solanum lycopersicum) under low P soil conditions. Three inoculation treatments were conducted, using unselectively grown soil bacteria (bacterial mix), heat treated (HT-RU47) and viable RU47 (RU47) cells, and one not inoculated, optimally P-fertilized treatment. We verified plant growth, nutrient availability, enzyme activities and microbial community structure in soil.Results: A plant growth promotion effect with improved P uptake was observed in both RU47 treatments. Inoculations of RU47 cells increased microbial phosphatase activity (PA) in the rhizosphere.Conclusions: Plant growth promotion by RU47 cells is primarily associated with increased microbial PA in soil, while promotion of indigenous Pseudomonads as well as phytohormonal effects appear to be the dominant mechanisms when adding HT-RU47 cells. Since the development of effective NO mitigation options is a key challenge for future agricultural practice, we studied the interactive effect of tillage systems on fertilizer-derived NO emissions and the abundance of microbial communities involved in NO production and reduction. Soil samples from 0-10 cm and 10-20 cm depth of reduced tillage and ploughed plots were incubated with dairy slurry (SL) and manure compost (MC) in comparison with calcium ammonium nitrate (CAN) and an unfertilized control (ZERO) for 42 days. NO and CO fluxes, ammonium, nitrate, dissolved organic C, and functional gene abundances (16S rRNA gene, nirK, nirS, nosZ, bacterial and archaeal amoA) were regularly monitored. Averaged across all soil samples, NO emissions decreased in the order CAN and SL (CAN = 748.8 ± 206.3, SL = 489.4 ± 107.2 μg kg) followed by MC (284.2 ± 67.3 μg kg) and ZERO (29.1 ± 5.9 μg kg). Highest cumulative NO emissions were found in 10-20 cm of the reduced tilled soil in CAN and SL. NO fluxes were assigned to ammonium as source in CAN and SL and correlated positively to bacterial amoA abundances. Additionally, nosZ abundances correlated negatively to NO fluxes in the organic fertilizer treatments. Soils showed a gradient in soil organic C, 16S rRNA, nirK, and nosZ with greater amounts in the 0-10 than 10-20 cm layer. Abundances of bacterial and archaeal amoA were higher in reduced tilled soil compared to ploughed soils. The study highlights that tillage system induced biophysicochemical stratification impacts net NO emissions within the soil profile according to N and C species added during fertilization. Our understanding of the interactions between minerals, organic matter, and microorganisms at so-called biogeochemical interfaces in soil is still hampered by the inherent complexity of these systems. Artificial soil maturation experiments can help to bridge a gap in complexity between simple abiotic sorption experiments and larger-scale field experiments. By controlling other soil-forming factors, the effect of a particular variable can be identified in a simplified system. Here, we review the findings of a series of artificial soil incubation experiments with the aim of revealing general trends and conclusions. The artificial soils were designed to determine the effect of mineral composition and charcoal presence on the development of abiotic and biotic soil properties during maturation. In particular, the development of soil aggregates, organic matter (OM) composition and turnover, sorption properties, and the establishment of microbial community composition and function were considered. The main objectives of the research were to determine (1) how surface properties and sorption of chemicals modify biogeochemical interfaces; (2) how much time is required to form aggregates from mixtures of pure minerals, OM, and a microbial inoculum; and (3) how the presence of different mineral and charcoal surfaces affects aggregation, OM turnover, and the development of microbial community composition. Characterization of spatial and temporal variation in grassland productivity and nutrition is crucial for a comprehensive understanding of ecosystem function. Although within-site heterogeneity in soil and plant properties has been shown to be relevant for plant community stability, spatiotemporal variability in these factors is still understudied in temperate grasslands. Our study aimed to detect if soil characteristics and plant diversity could explain observed small-scale spatial and temporal variability in grassland productivity, biomass nutrient concentrations, and nutrient limitation. Therefore, we sampled 360 plots of 20 cm × 20 cm each at six consecutive dates in an unfertilized grassland in Southern Germany. Nutrient limitation was estimated using nutrient ratios in plant biomass. Absolute values of, and spatial variability in, productivity, biomass nutrient concentrations, and nutrient limitation were strongly associated with sampling date. In April, spatial heterogeneity was high and most plots showed phosphorous deficiency, while later in the season nitrogen was the major limiting nutrient. Additionally, a small significant positive association between plant diversity and biomass phosphorus concentrations was observed, but should be tested in more detail. We discuss how low biological activity e.g., of soil microbial organisms might have influenced observed heterogeneity of plant nutrition in early spring in combination with reduced active acquisition of soil resources by plants. These early-season conditions are particularly relevant for future studies as they differ substantially from more thoroughly studied later season conditions. Our study underlines the importance of considering small spatial scales and temporal variability to better elucidate mechanisms of ecosystem functioning and plant community assembly. Environmental controls of 2-methyl-4-chlorophenoxyacetic acid (MCPA) degradation are poorly understood. We investigated whether microbial MCPA degraders are stimulated by (maize) litter and whether this process depends on concentrations of MCPA and litter. In a microcosm experiment, different amounts of litter (0, 10 and 20 g kg) were added to soils exposed to three levels of the herbicide (0, 5 and 30 mg kg). The treated soils were incubated at 20 °C for 6 weeks, and samples were taken after 1, 3 and 6 weeks of incubation. In soils with 5 mg kg MCPA, about 50 % of the MCPA was dissipated within 1 week of the incubation. Almost complete dissipation of the herbicide had occurred by the end of the incubation with no differences between the three litter amendments. At the higher concentration (30 mg kg), MCPA endured longer in the soil, with only 31 % of the initial amount being removed at the end of the experiment in the absence of litter. Litter addition greatly increased the dissipation rate with 70 and 80 % of the herbicide being dissipated in the 10 and 20 g kg litter treatments, respectively. Signs of toxic effects of MCPA on soil bacteria were observed from related phospholipid fatty acid (PLFA) analyses, while fungi showed higher tolerance to the increased MCPA levels. The abundance of bacterial tfdA genes in soil increased with the co-occurrence of litter and high MCPA concentration, indicating the importance of substrate availability in fostering MCPA-degrading bacteria and thereby improving the potential for removal of MCPA in the environment. The action mode of sulfonylurea herbicides is the inhibition of the acetohydroxyacid synthase (AHAS) required for the biosynthesis of amino acids valine and isoleucine in plants. However, this enzyme is also present in a range of non-targeted organisms, among which soil microorganisms are known for their pivotal role in ecosystem functioning. In order to assess microbial toxicity of sulfonylurea herbicide nicosulfuron (NS), a tiered microcosm ( Tier I) to field ( Tier II) experiment was designed. Soil bacteria harboring AHAS enzyme tolerant to the herbicide nicosulfuron were enumerated, isolated, taxonomically identified, and physiologically characterized. Results suggested that application of nicosulfuron drives the selection towards NS-tolerant bacteria, with increasing levels of exposure inducing an increase in their abundance and diversity in soil. Tolerance to nicosulfuron was shown to be widespread among the microbial community with various bacteria belonging to Firmicutes ( Bacillus) and Actinobacteria ( Arthrobacter) phyla representing most abundant and diverse clusters. While Arthrobacter bacterial population dominated community evolved under lower (Tier II) nicosulfuron selection pressure, it turns out that Bacillus dominated community evolved under higher (Tier I) nicosulfuron selection pressure. Different NS-tolerant bacteria likewise showed different levels of sensitivity to the nicosulfuron estimated by growth kinetics on nicosulfuron. As evident, Tier I exposure allowed selection of populations able to better cope with nicosulfuron. One could propose that sulfonylureas-tolerant bacterial community could constitute a useful bioindicator of exposure to these herbicides for assessing their ecotoxicity towards soil microorganisms. Soil fungi play an essential role in the decomposition of plant-derived organic material entering soils. The quality and quantity of organic compounds vary seasonally as well as with soil depth. To elucidate how these resources affect fungal communities in an arable soil, a field experiment was set up with two plant species, maize and wheat. Resource availability was experimentally manipulated by maize litter input on one half of these maize and wheat plots after harvest in autumn. Fungal biomass was determined by ergosterol quantification, and community structure was investigated by fungal automated ribosomal intergenic spacer analysis (F-ARISA). An annual cycle was assessed across a depth gradient, distinguishing three soil habitats: the plough layer, rooted soil below the plough layer, and deeper root-free soil. Fungal communities appeared highly dynamic and varied according to soil depth and plant resources. In the plough layer, the availability of litter played a dominant role in shaping fungal communities, whereas in the rooted layer below, community structure and biomass mainly differed between plant species. This plant effect was also extended into the root-free soil at a depth of 70 cm. In winter, the availability of litter also affected fungal communities in deeper soil layers, suggesting vertical transport processes under fallow conditions. These distinct resource effects indicate diverse ecological niches along the soil profile, comprising specific fungal metacommunities. Background and aims: Fine root decomposition contributes significantly to element cycling in terrestrial ecosystems. However, studies on root decomposition rates and on the factors that potentially influence them are fewer than those on leaf litter decomposition. To study the effects of region and land use intensity on fine root decomposition, we established a large scale study in three German regions with different climate regimes and soil properties. Methods In 150 forest and 150 grassland sites we deployed litterbags (100 μm mesh size) with standardized litter consisting of fine roots from European beech in forests and from a lowland mesophilous hay meadow in grasslands. In the central study region, we compared decomposition rates of this standardized litter with root litter collected on-site to separate the effect of litter quality from environmental factors. Results: Standardized herbaceous roots in grassland soils decomposed on average significantly faster (24 ± 6 % mass loss after 12 months, mean ± SD) than beech roots in forest soils (12 ± 4 %; p < 0.001). Fine root decomposition varied among the three study regions. Land use intensity, in particular N addition, decreased fine root decomposition in grasslands. The initial lignin:N ratio explained 15 % of the variance in grasslands and 11 % in forests. Soil moisture, soil temperature, and C:N ratios of soils together explained 34 % of the variance of the fine root mass loss in grasslands, and 24 % in forests. Conclusions: Grasslands, which have higher fine root biomass and root turnover compared to forests, also have higher rates of root decomposition. Our results further show that at the regional scale fine root decomposition is influenced by environmental variables such as soil moisture, soil temperature and soil nutrient content. Microbiologically active biogeochemical interfaces are excellent systems to study soil functions such as pesticide degradation at the micro-scale. In particular, in the detritusphere pesticide degradation is accelerated by input of fresh organic carbon from litter into the adjacent soil. This observed priming effect suggests: (i) pesticide degradation is strongly coupled to carbon turnover, (ii) it is controlled by size and activity of the microbial community and (iii) sorption and transport of dissolved carbonaceous compounds and pesticides might regulate substrate availability and in turn decomposition processes. We present a new mechanistic 1D model ( PEsticide degradation Coupled to CArbon turnover in the Detritusphere, PECCAD) which implements these hypotheses. The new model explicitly considers growth and activity of bacteria, fungi and specific pesticide degraders in response to substrate availability. Enhanced pesticide degradation due to availability of a second source of carbon (dissolved organic carbon) is implemented in the model structure via two mechanisms. First, additional substrate is utilized simultaneously with the pesticide by bacterial pesticide degraders resulting in an increase in their size and activity. Second, stimulation of fungal growth and activity by additional substrates leads directly to higher pesticide degradation via co-metabolism. Thus, PECCAD implicitly accounts for litter-stimulated production and activity of unspecific fungal enzymes responsible for co-metabolic pesticide degradation. With a global sensitivity analysis we identified high-leverage model parameters and input. In the present study, the temporal and spatial variation of the abundance of the alkane monooxygenase gene alkB and 16S rRNA genes in different soil compartments was analysed in the presence or absence of 2-methyl-4-chlorophenoxyacetic acid (MCPA) after the addition of pea litter to soil in a microcosm study. Samples were analysed shortly after litter addition (T0) and 1 week (T1), 3 weeks (T3) and 6 weeks (T6) after the addition of litter. In addition also, the quantity and quality of litter-derived alkanes was analysed and measured. The results revealed a fast and complete degradation of MCPA in all compartments throughout the experiment. Nevertheless, significant changes in the distribution patterns of short- and middle-chained alkanes suggest an interaction of MCPA and alkane degradation. alkB gene copy numbers were highly influenced by the time point of analysis and by the investigated soil compartment. Overall, an increase in alkB gene copy numbers from T0 to T3 was visible in the upper soil compartments whereas a decrease compared to T0 was measured in the deeper soil compartments. MCPA addition resulted in an increase of alkB abundance at T6. Gene copy numbers of 16S rRNA were not influenced by sampling time and soil compartment. In contrast to the control treatments, a slight increase in 16S rRNA gene copy numbers was visible at T1 and T3 compared to T0 in all soil compartments. [ABSTRACT FROM AUTHOR]

Little is known about the changes in abundance of microbial taxa in relation to the chronosequence of receding glaciers. This study investigated how the abundances of ten bacterial phyla or classes varied along successional gradients in two glaciers, Ödenwinkelkees and Rotmoosferner, in the central Alps. Quantitative PCR was used to estimate the abundance of the different bacterial taxa in extended glacier chronosequences, including 10- to 160-year-old successional stages, the surface of the glacier, and a fully established soil. Actinobacteria (15-30%) was the dominant group within the chronosequences. Several taxa showed significant differences in the number of taxa-specific 16S rRNA gene copies per nanogram of DNA and/or in the ratio of taxa-specific to the total bacterial 16S rRNA gene copies (i.e., the relative abundance of the different taxa within the bacterial community) between the established soils or the glacier surface and the 10- to 160-year-old successional stages. A significantly higher proportion of Βetaproteobacteria (20%) was observed on the surface of both glaciers. However, no differences were observed between the 10- to 160-year-old successional stages in the number of taxa-specific 16S rRNA gene copies per nanogram of DNA or in the ratio of taxa-specific to the total bacterial 16S rRNA gene copies for the different taxa. Nevertheless, when the relative abundance data from all the studied taxa were combined and analyzed altogether, most of the sites could be distinguished from one other. This indicates that the overall composition of the bacterial community was more affected than the abundance of the targeted taxa by changes in environmental conditions along the chronosequences. Transport of litter carbon in the detritusphere might determine fungal abundance and diversity at the small scale. Rye residues were applied to the surface of soil cores with two different water contents and incubated at 10°C for 2 and 12 weeks. Fungal community structure was analysed by constructing clone libraries of 18S rDNA and subsequent sequencing. Litter addition induced fungal succession in the adjacent soil and decreased detectable fungal diversity mainly due to the huge supply of substrates. Ergosterol content and N-acetyl-glucosaminidase activity indicated fungal growth after 2 weeks. Simultaneously, the structure of the fungal community changed, with Mortierellaceae proliferating during the initial phase of litter decomposition. Ergosterol measurements were unable to detect this early fungal growth because Mortierellaceae do not produce ergosterol. In the late phase during decomposition of polymeric substrates, like cellulose and chitin, the fungal community was dominated by Trichocladium asperum. Water content influenced community composition only during the first 2 weeks due to its influence on transport processes in the detritusphere and on competition between fungal species.
