Objective: Malnutrition is a common issue in patients with head and neck squamous cell carcinoma (HNSCC), leading to compromised immune function and diminished treatment efficacy. This is particularly concerning for therapies targeting the PD-1 pathway, which are pivotal in cancer treatment. The aim of this retrospective study was to explore the potential benefits of immunonutrition in improving immune responses and clinical outcomes for these patients. Methods: In the study, 49 individuals diagnosed with HNSCC were enrolled and divided into two groups: one group received specialized immunonutrition support designed to enhance immune function, while the other group received standard nutritional care. The researchers assessed immune function by evaluating CD4+ and CD8+ T cell counts, which are critical indicators of immune health. Additionally, clinical outcomes were monitored, focusing on infection rates and the duration of hospital stays. Results: Patients who received immunonutrition showed a significant improvement in immune function, as indicated by higher levels of CD4+ and CD8+ T cells. Furthermore, these patients experienced shorter recovery times and lower infection rates compared to those receiving standard nutrition. Conclusion: These findings suggest that immunonutrition may play a vital role in enhancing the effectiveness of cancer therapies, including PD-1 inhibitors. By supporting immune health, immunonutrition could potentially improve patient outcomes and quality of life during and after treatment. This study aimed to investigate the effect of hetrombopag combined with conventional treatment on immune function in patients with severe aplastic anemia (SAA). Patients were categorized into the control group (n = 50, receiving conventional treatment only) and experimental group (n = 50, receiving hetrombopag combined with conventional treatment). Before treatment and at weeks 18, 24, and 52 after treatment, the two groups were compared in routine blood test indicators, natural killer (NK) cell activity, and peripheral blood inflammatory factor levels. The overall remission rate and incidence of adverse events were also compared between the two groups. Outpatient or telephone follow-up was performed before treatment and at weeks 18, 24, and 52 after treatment to observe patients' immune function, treatment outcome, quality of life, and adverse events. Hemoglobin (Hb), and platelet count (PLT) (P < 0.05), and a rise in NK cell activity (P < 0.05). Interleukin (IL-10) levels were significantly higher, while IL-6 levels were significantly lower in the experimental group compared to the control group (P < 0.05). After receiving the treatment, all scores of SF-36 domains in both groups were higher than before treatment, particularly with higher scores in the experimental group (P < 0.05). To assess the safety and efficacy of fecal microbiota transplantation (FMT) as an adjunctive therapeutic intervention for IgA nephropathy (IgAN). Fifteen patients with IgA nephropathy were recruited based on inclusion and exclusion criteria and underwent FMT using enteric microbial capsules. Clinical indicators, intestinal microbiota and metabolomic profiles, as well as changes in serum immune cells and cytokines, were monitored before and after FMT. No severe adverse reactions were observed in the subjects. After FMT, there was a reduction in the 24-h urinary protein quantification in subjects. The relative abundances of Phocaeicola_vulgatus, Bacteroides_uniformis, Prevotella_copri, Phocaeicola_dorei, Bacteroides_ovatus, Bacteroides_xylanisolvens, Parabacteroides _distasonis, Bifidobacterium_pseudocatenulatum, Bacteroides_sp._HF-162, and Bifidobacterium_longum changed after FMT. In terms of intestinal metabolites, the levels of acylcarnitine18:0 (ACar.18:0), cotinine, N-arachidonoyl-L-serine, phosphatidylcholine (PC. (18:3e/22:6)), serotonin, and fumagillin showed significant changes. Flow cytometry analysis showed the absolute count of plasma B cells decreased in subjects, and this change correlated with alterations in the intestinal microbiota and metabolites. This study preliminarily evaluates the safety and efficacy of FMT in patients with IgAN. No significant adverse reactions were observed, and the administration of FMT alongside ACEI/ARB therapy was effective in reducing urinary protein levels in patients with IgAN, a process that may be associated with B-cell immunity. As a member of the small G protein superfamily, ras-related nuclear proteins (Ran) mainly function in regulating nucleocytoplasmic transport and microtubule formation. Ran is reported to have an immune function in invertebrate defense against pathogen invasion. This study obtained the full-length sequence of the Ran cDNA (PcRan) of Procambarus clarkii and investigated changes in its expression after pathogenic infection and its role in the clearance of pathogenic bacteria. Phylogenetic tree analysis revealed that PcRan and Ran of other crustaceans belong to one branch and have a close genetic relationship. Multiple sequence alignment analyses revealed that PcRan has a highly conserved G domain and contains all the G1–G5 motifs. PcRan expression was widely distributed in various tissues in P. clarkii, with the highest expression levels being found in the muscle and gills. The levels of PcRan expression in several major immune organs, including hepatopancreas, gills, hemocytes, and intestines, were significantly increased after infection with white spot syndrome virus or Aeromonas veronii. After PcRan overexpression, the bacterial clearance ability and survival rate of P. clarkii infected with A. veronii was significantly higher than that of the control group. This study showed that PcRan is involved in the innate immune process of P. clarkii. The results of this study provide theoretical significance and application value for understanding the immune function of Ran after crayfish pathogen infection, and provide guidance for the prevention and control of crayfish diseases. The first 1000 days of life is a critical period of development in which adverse circumstances can have long-term consequences for the child’s health. Maternal immune activation is associated with increased risk of neurodevelopmental disorders in the child. Aberrant immune responses have been reported in individuals with neurodevelopmental disorders. Moreover, lasting effects of maternal immune activation on the offspring’s immune system have been reported. Taken together, this indicates that the effect of maternal immune activation is not limited to the central nervous system. Here, we explore the impact of maternal immune activation on the immune system of the offspring. We first describe the development of the immune system and provide an overview of reported alterations in the cytokine profiles, immune cell profiles, immune cell function, and immune induction in pre-clinical models. Additionally, we highlight recent research on the impact of maternal COVID-19 exposure on the neonatal immune system and the potential health consequences for the child. Our review shows that maternal immune activation alters the offspring’s immune system under certain conditions, but the reported effects are conflicting and inconsistent. In general, epigenetic modifications are considered the mechanism for fetal programming. The available data was insufficient to identify specific pathways that may contribute to immune programming. As a consequence of the COVID-19 pandemic, more research now focuses on the possible health effects of maternal immune activation on the offspring. Future research addressing the offspring’s immune response to maternal immune activation can elucidate specific pathways that contribute to fetal immune programming and the long-term health effects for the offspring. The Rab family of proteins is involved in organelle regulation, trafficking, signal transduction, mitosis, phagocytosis, growth differentiation, cytoskeleton protein depolymerization and other cellular functions and abnormal expression has been associated with many pathologies, including cancer. Previous work has focused on Rab function in higher animals but fish, crustaceans and other aquatic animals also express the Rab proteins. The current review summarizes research progress on Rab proteins, focusing on immune defense against pathogens in aquatic animals. Some differences in Rab protein function between aquatic and higher animals were found and greater involvement in the fish and crustacean immune system than that of higher animals. To reveal the variation of gut microbiota and its association with immune function in cured patients with coronavirus 2019 (COVID-19) disease, gut microbiota of patients discharged from hospital for 20 ~ 23 months and healthy volunteers was analyzed by high throughput 16S rRNA sequencing. The diversity and abundance were compared, and the correlation with immunity factors was investigated, and changes in the content of 6 genera microorganisms with proportion higher than 0.1% were revealed in patients with COVID-19 disease: reduced content of Subdoligranulum, Haemophilus, Coprococcus, Eubacterium vertriosum group, and Lachnospiraceae ND3007 group and increased content of Hungatella. NK cells were negatively correlated to Subdoligranulum, while CD8 cells were positively correlated to Subdoligranulum but negative to Hungatella. IL-8 concentration was negatively correlated to Subdoligranulum, Haemophilus, Coprococcus, Eubacterium vertriosum group, and Lachnospiraceae ND3007 group but positively to Hungatella, while IL-1β concentration was negatively correlated to Haemophilus and Eubacterium ventriosum group but positively to Hungatella. The variation of probiotics and potential pathogenic bacteria implies a higher risk in diseases and inflammation, and the modulation of the gut microbiota may help the healing of COVID-19 patients. Eucommia ulmoides is a traditional Chinese herbal medicine, with pharmacological effects such as lowering blood pressure and enhancing immune function. The effects of dietary Eucommia ulmoides polysaccharide (EUP) on immune function and meat quality were studied in Songliao Black Pigs. Blood lymphocyte counts and percentage, concentrations of serum total protein and of albumin increased, whereas those of urea nitrogen and triglyceride decreased. White blood cell and lymphocyte counts, and serum IgA, IgE, IgG2 a and IFN-γ increased. Average daily weight gain, slaughter weight, lean meat rate and cooked meat rate increased, whereas pH24, feed-weight ratio, fat rate, yellowness (b#) and centrifugal dehydration rate decreased. Transcriptome sequencing of longissimus dorsi muscle detected 32 differentially expressed genes (DEGs), of which 26 were up-regulated and 6 down-regulated. A total of 19 genes were differentially expressed in the four groups, 18 of which were up-regulated. The DEGs included ADAMTS4, PER1, STAC, SERPINE1, FASN, THRSP, SP7 and KRT80 and the protein interaction network showed 20 up-regulated nodes, three down-regulated nodes and 14 DEGs. GO functional annotation and enrichment analysis showed that 34 items were significantly enriched, including transferase activity, actin binding, acetyl coenzyme A, acyl coenzyme A metabolism, adipose tissue development and acyl glycerol homeostasis. KEGG pathway analysis showed that the AMPK and PPAR signaling pathways were enriched. Dietary Eucommia polysaccharide enhanced immune function in Songliao Black Pigs, improved growth and carcass performance, increased the expression of genes related to meat quality traits and improved meat quality. Background: Spontaneous clearance of chronic hepatitis C virus (HCV) is rare in adults. A T-lymphocyte response is thought to be involved in HCV-RNA clearance. Splenectomy reportedly has a beneficial effect on T cell immune function in patients with cirrhosis. To the best of our knowledge, the present report is the first to describe spontaneous clearance of serum HCV-RNA within 1 year after splenectomy in a patient with cirrhosis. Case presentation: A 55-year-old man with HCV cirrhosis was transferred to our institution with advanced pancytopenia, splenomegaly, and gastric varices. He had a 1-year history of ascites, edema, and general fatigue. The patient had a Child–Pugh score of 8 and serological type 1 HCV; the HCV-RNA level was 4.7 log IU/mL. Contrast-enhanced computed tomography showed gastric varices and marked splenomegaly (estimated spleen volume of 2175 mL). Esophagogastroduodenoscopy revealed enlarged gastric varices with no red color sign, and the varices were larger than those 1 year prior. He was diagnosed with decompensated HCV-related liver cirrhosis and portal hypertension. We considered direct-acting antiviral (DAA) therapy; however, DAA therapy was not approved in Japan for patients with decompensated cirrhosis at that time. Hand-assisted laparoscopic splenectomy was performed to improve the worsening portal hypertension. Further, we planned the initiation of DAA therapy after surgery, when such therapy would become available. DAA therapy was approved 1 year after splenectomy. At that time, we measured the HCV-RNA level before the initiation of DAA therapy; unexpectedly, however, serum HCV-RNA was not detectable, and the virus continued to disappear during the following 4 years. His liver function (total bilirubin, albumin, and prothrombin time) and pancytopenia improved during the 5 years postoperatively. The serum aspartate and alanine aminotransferase levels normalized between 1 and 5 years postoperatively. Esophagogastroduodenoscopy showed no change in the gastric varices during the 5 years after surgery. The patient remained asymptomatic and continued to do well. Conclusions: We have presented a case of spontaneous clearance of HCV-RNA after splenectomy in a patient with cirrhosis and portal hypertension. Rab5A play important roles in regulating trafficking organelles, especially in phagosome formation. In the present study, full-length cDNA sequences of Rab5A were cloned from Yellow River Carp Cyprinus carpio, which was designated as CcRab5A. The full-length cDNA of the CcRab5A cDNA sequence is 2434 bp and included an open reading frame (ORF) encoding 216 amino acids polypeptide with an estimated molecular weight of 23.47 kDa. Bioinformatics analysis showed that the CcRab5A protein was highly conserved during evolution. CcRab5A's deduced amino acid sequence showed high identity to Cyprinus carpio (99.54%) in comparison. The guanine-base binding motif (G), phosphate/magnesium-binding motif (PM), and Rab family motif (Rab F) of CcRab5A are highly conserved among various species, but the N- and C-terminal regions were hypervariable, according to the results of multiple sequence alignment and phylogenetic analysis. Additionally, 11 tissues of Yellow River Carp were examined using Real-time Fluorescence Quantitative PCR (qRT-PCR) to determine the expression levels, with the highest expression levels in head kidney and blood. Followed by heart, liver, muscle, brain, gill, skin, spleen and intestine; The expression level in body kidney was the lowest. Yellow River Carp was immunized with Aeromonas hydrophila and Spring viremia of carp virus (SVCV) respectively, and the expression changes of CcRab5A in gill, spleen, liver, intestine and skin of Yellow River Carp were detected. The results showed that the expression level of the gene was obviously up-regulated at different time points. The eukaryotic recombinant plasmids of CcRab5A, pEGFP-N3 were constructed and transfected into GCO cells for subcellular localization. The results showed that CcRab5A were mainly distributed in nuclear membrane and various endosome membranes. This study aimed to explore the amelioration of immune function and clinical symptoms in epilepsy patients following the implementation of topiramate therapy. We retrospectively analyzed 90 epilepsy patients treated at our institution from September 2020 to July 2022, segregating them into two groups: the observation group (n = 50, administered with a combination of carbamazepine and topiramate), and the control group (n = 40, treated with carbamazepine alone). The study evaluated disparities in clinical outcomes, immune function modifications, epilepsy score variances, quality of life indices, and incidence of adverse reactions before and after treatment in both groups. The observation group demonstrated a total treatment efficacy rate of 94.00% (47/50), significantly exceeding that of the control group, which stood at 75.00% (30/40) (P < 0.05). Pre-treatment epilepsy and QOLIE-31 scores showed no statistically meaningful difference between the two groups (P > 0.05); however, post-treatment evaluation after 3 months indicated that the observation group had significantly lower epilepsy scores and notably higher QOLIE-31 scores (P < 0.05). Although pre-treatment immune function indices (including CD3+, CD4+, and IgA levels) did not show a significant difference between the groups (P > 0.05), post-treatment levels of CD3+ and CD4+ were markedly higher, and IgA levels were lower in the observation group (P < 0.05). The observation group also exhibited a reduced overall incidence of adverse reactions (12.00%, 6/50), significantly lower than the control group (32.50%, 13/40) (P < 0.05). The adjunctive use of topiramate with standard therapy can enhance the clinical efficacy in epilepsy patients, ameliorating immune function, augmenting the quality of life, and escalating treatment safety, thereby presenting substantial value for clinical application. The significance of Vitamin D has been appreciated beyond bone health and calcium metabolism. The importance of Vitamin D in respiratory health has been recognized due to its immunomodulatory and anti-microbial properties. The hypothesis is that Vitamin D could have a significant role in the pathogenesis of respiratory diseases and may represent a novel preventive and therapeutic strategy. Furthermore, enumerable observational studies established the association of Vitamin D deficiency with respiratory diseases such as asthma, bronchiolitis, pneumonia, tuberculosis, etc. However, experimental studies have not shown the encouraging results. This brief review will summarize and discuss the synthesis and metabolism of Vitamin D, the prevalence of Vitamin D deficiency in children, its role in the pathogenesis of various childhood respiratory diseases, and an overview of the therapeutic trials assessing the role of Vitamin D supplementation in childhood respiratory diseases. The autonomic nervous system (ANS) plays a complex role in the regulation of the immune system, with generally inhibitory effects via activation of β-adrenergic receptors on immune cells. We hypothesized that HIV-associated autonomic neuropathy (HIV-AN) would result in immune hyperresponsiveness which could be depicted using network analyses. Forty-two adults with well-controlled HIV underwent autonomic testing to yield the Composite Autonomic Severity Score (CASS). The observed range of CASS was 2–5, consistent with normal to moderate HIV-AN. To construct the networks, participants were divided into 4 groups based on the CASS (i.e., 2, 3, 4 or 5). Forty-four blood-based immune markers were included as nodes in all networks and the connections (i.e., edges) between pairs of nodes were determined by their bivariate Spearman's Rank Correlation Coefficient. Four centrality measures (strength, closeness, betweenness and expected influence) were calculated for each node in each network. The median value of each centrality measure across all nodes in each network was calculated as a quantitative representation of network complexity. Graphical representation of the four networks revealed greater complexity with increasing HIV-AN severity. This was confirmed by significant differences in the median value of all four centrality measures across the networks (p ≤ 0.025 for each). Among people with HIV, HIV-AN is associated with stronger and more numerous positive correlations between blood-based immune markers. Findings from this secondary analysis can be used to generate hypotheses for future studies investigating HIV-AN as a mechanism contributing to the chronic immune activation observed in HIV. Background: Sepsis is a condition of severe septicemia and septic shock caused by infection, accompanied by multiple organ dysfunction often leading to patients' admission to the intensive care unit (ICU) and death. Objective: This study aims to investigate the efficacy of urinary trypsin inhibitor in the treatment of rats with severe sepsis, and to analyze its effects on coagulation and immunity. Sixty Sprague Dawley rats were randomly divided into a model control group (MCG), an ulinastatin group (UG), and a normal control group (n = 20 each). Rats in the model CG and UG were modeled for severe sepsis through cecal ligation and puncture, and the 72 h survival of the rats in the three groups was recorded. The expression of IL-18 and TNF-α before urinary trypsin inhibitor administration and 72 h after administration was detected by an ELISA. Activated partial thromboplastin time (APTT), prothrombin time (PT), thrombin time (TT), and fibrinogen (FIB) were determined using a fully automatic coagulation analyzer. CD4 + , CD8 + , and CD4 + /CD8 + levels were quantitated by flow cytometry. Results: After administration, compared with those in the MCG, rats in the UG showed higher survival rates (P < 0.05), lower IL-18 and TNF-α expression (P < 0.05), lower APTT, PT, and TT (P < 0.05), higher FIB (P < 0.05), higher CD4 + and CD4 +/CD8 + levels (P < 0.05), and significantly lower CD8 + levels (P < 0.05). Conclusion: Urinary trypsin inhibitor can increase the survival rate of rats with severe sepsis, reduce their inflammatory responses, and ameliorate coagulation disorders and immune dysfunction. Pathological conditions and harmful drugs cause many gastrointestinal diseases in broiler chicken. The current study was conducted to investigate the effect of trace elements zinc (Zn) and selenium (Se) supplementation on histomorphology, immunological role, and functional activity of goblet cells (GCs) of the small intestine. The Alcian blue-periodic acid-Schiff (AB-PAS) was performed to assess the histomorphological changes in GCs, which revealed the regular dispersion with high electron density of GCs throughout the mucosal surface in the supplemented group. However, irregular dispersion with low electron density of GCs was present in the control group. The immunological functional role of GCs within the small intestine was examined by mucicarmine staining, immunohistochemistry, and immunofluorescence. The results showed a high mucin glycol protein secretion in the supplemented group, whereas limited mucin glycol protein secretion in the control group. Furthermore, the biological significance showed a high and low immunoreactivity of Muc2 and Muc13 in the supplemented and control groups, respectively. Immunofluorescence was used to confirm the immunosignaling of Muc2. Results revealed high immunosignaling of Muc2 at the apical part of the small intestine in the supplementation group, while low immunosignaling of Muc2 in the control group. Results suggest that trace element supplementation had significant effect on morphology and immunological role of GCs, which might be essential for immune function and health status of broiler chicken. Abstract Group A streptococci have evolved multiple strategies to evade human antibodies, making it challenging to create effective vaccines or antibody treatments. Here, we have generated antibodies derived from the memory B cells of an individual who had successfully cleared a group A streptococcal infection. The antibodies bind with high affinity in the central region of the surface‐bound M protein. Such antibodies are typically non‐opsonic. However, one antibody could effectively promote vital immune functions, including phagocytosis and in vivo protection. Remarkably, this antibody primarily interacts through a bivalent dual‐Fab cis mode, where the Fabs bind to two distinct epitopes in the M protein. The dual‐Fab cis‐binding phenomenon is conserved across different groups of M types. In contrast, other antibodies binding with normal single‐Fab mode to the same region cannot bypass the M protein's virulent effects. A broadly binding, protective monoclonal antibody could be a candidate for anti‐streptococcal therapy. Our findings highlight the concept of dual‐Fab cis binding as a means to access conserved, and normally non‐opsonic regions, regions for protective antibody targeting.

Cold-inducible RNA-binding protein (CIRBP) responds to a wide array of cellular stresses such as cold shock, hypoxia, and inflammatory responses. However, functional studies of CIRBP in jawless vertebrates are limited. In this study, a CIRBP homolog from the jawless vertebrate lamprey (Lethenteron reissneri) was cloned and characterized (named Lr-CIRBP). The cDNA fragment of Lr-CIRBP has a 516 bp open reading frame (ORF) that encodes 171 amino acids, comprising a glycine-rich region at the C-terminal, similar to higher vertebrates but slightly shorter, and an RNA recognition motif (RRM) domain at the N-terminus. The predicted Lr-CIRBP sequence had 51.4 ~ 70.6% similarity with CIRBPs from other vertebrates. Further phylogenetic analysis revealed that Lr-CIRBP is located in the outgroup of vertebrates and is the ancestor of vertebrates. Based on real-time quantitative PCR experimental analysis, Lr-CIRBP expression was highest in leukocytes and increased significantly after multi-stimulation, peaking at 12 h. RNA interference showed that Lr-CIRBP knockdown can down-regulate the expression of inflammatory factors in Lethenteron reissneri. In conclusion, our study successfully clarifies the ancestral features and functions of CIRBP, while revealing valuable insight into how the protein is involved in the immune responses of a jawless vertebrate. Neuronal nitric oxide synthase (nNOS) was the producer of nitric oxide (NO) which played important gas messenger molecules in biological process. It also can take effect as immune regulation molecule in organism. Black rockfish (Sebastes schlegelii) is an important economic fish which were widely farmed in East Asia countries. Meanwhile, the pathogenic bacteria such as the Edwardsiella tarda and Vibrio anguillarum in seawater always brought serious obstacles to their healthy growth. In order to explore the expression pattern of nNOS gene under the pathogen stimulation and predict its immune function, the nNOS gene in black rockfish named SsnNOS was identified. It was 3780 bp in length, located on chromosome 6, and contained 27 coding domain sequence (CDs). According to the phylogenetic analysis, the SsnNOS showed closest relative to the counterpart gene of swamp eel (Monopterus albus). Meanwhile, analysis of SsnNOS expression in various healthy tissues showed that SsnNOS expression level was highest in healthy brain tissues, followed by intestinal tissues. In addition, SsnNOS showed significant expression changes in response to stimulation by two pathogens. Particular in gill, the expression of SsnNOS after pathogenic stimulation increased significantly. The Elisa analysis showed the SsnNOS content in gills was much higher than that in other tissues at all time points. Moreover, the expression patterns of SsnNOS in brain, intestine and kidney after stimulation by pathogens showed a distinct expression pattern which first down-regulated and then up-regulated. Simultaneously defending against predators, stymieing competitors, and generating immune responses can impose conflicting demands for host species caught in the entanglement of a food web. Host immunity is not only shaped by direct interactions among species, but also many indirect cascading effects. By reducing competition, predators in particular can affect resource acquisition necessary for hosts to mount energetically costly immune responses. However, identifying the links between predators and host immune responses determined by resource acquisition is a complex affair, because predators can (1) reduce host density and thus competition among hosts, (2) exert non-consumptive trait-mediated effects on host resource acquisition behavior, and (3) generate natural selection on host resource acquisition behavior. To examine the relative contributions of these potential predator driven density- and trait-mediated effects on a key aspect of immune function (total phenoloxidase activity, total PO), we conducted mesocosm and field experiments with larval damselflies (Enallagma signatum) and their dominant fish predator (Lepomis macrochirus). Although we expected to observe declines in total PO activity with increases in damselfly density, we found no relationship between density and total PO activity. We also found no support for the prediction that total PO activity would vary as a result of either non-consumptive trait-mediated effects or selection on damselfly foraging activity underlying resource acquisition. Despite the lack of trait- or density-mediated effects, we did find that total PO activity increased with damselfly prey density among lakes, implying resource limitation for this aspect of immune function. These unexpected results point to the need to better understand the ecological conditions whereby predators and competitors constrain immune functions necessary for species to defend themselves in complex food webs. This study investigated the effects of synbiotics supplementation on growth performance, antioxidant status, immune function, and intestinal barrier function in broilers subjected to cyclic heat stress. One hundred and forty-four 22-day-old male broilers were randomly assigned to one of three treatment groups of six replicates each for a 21-day study, with eight birds per replicate. Broilers in the control group were reared at a thermoneutral temperature and received a basal diet. Broilers in the other two heat-stressed groups were fed a basal diet supplemented without (heat-stressed group) and with 1.5 g/kg synbiotic (synbiotic group). One and a half gram of the synbiotic consisted with 3 × 109 colony forming units (CFU) Clostridium butyricum, 1.5 × 109 CFU Bacillus licheniformis, 4.5 × 1010 CFU Bacillus subtilis, 600 mg yeast cell wall, and 150 mg xylooligosaccharide. Compared with the control group, heat stress increased rectal temperatures at 28, 35, and 42 days of age, respectively (P < 0.05). Birds subjected to heat stress had reduced weight gain, feed intake, and feed efficiency during 22 to 42 days (P < 0.05). In contrast, supplementation with the synbiotic decreased rectal temperature at 42 days of age and elevated weight gain of heat stress–challenged broilers (P < 0.05). Heat-stressed broilers exhibited a lower superoxide dismutase (SOD) activity in jejunal mucosa and a higher malondialdehyde accumulation in serum, liver and jejunal mucosa (P < 0.05), and the regressive SOD activity was normalized to control level when supplementing synbiotic (P < 0.05). Heat stress increased interleukin-1β (IL-1β) and interferon-γ (IFN-γ) levels in serum and IL-1β content in jejunal mucosa of broilers (P < 0.05). Synbiotic reduced IL-1β level in serum of broilers subjected to heat stress (P < 0.05). Compared with the control group, elevated serum diamine oxidase activity and reduced jejunal villus height were observed in broilers of the heat-stressed group (P < 0.05), and the values of these two parameters in the synbiotic group were intermediate (P > 0.05). Heat stress upregulated mRNA abundance of IL-1β and IFN-γ and downregulated gene expression levels of occluding and zonula occluden-1 (ZO-1) in jejunal mucosa of broilers (P < 0.05). The alterations in the mRNA expression levels of jejunal IL-1β and ZO-1 were reversed by the synbiotic (P > 0.05). To investigate the effects of nano-selenium (nano-Se) and Macleaya cordata extracts (MCE) on immune function and oxidative damage of sows and intrauterine growth retardation (IUGR) piglets exposed to heat stress (HS) in large-scale farms, a 2 × 2 factorial design was adopted in this test, and the two factors were nano-Se (0, 0.50 mg/kg) and MCE (0, 500 mg/kg). A total of 80 sows ([Landrace × Yorkshire] × Duroc, parity 2) were used in a 25-day trial from day 90 of gestation to delivery with 20 replications per group and 1 sow per replication. The dietary treatments of sows were as follows: (1) CON group, basic diet (0.30 mg/kg added Se, sodium selenite); (2) Nano-Se group, basic diet (0.00 mg/kg added Se) + 0.50 mg/kg added nano-Se; (3) MCE group, basic diet (0.00 mg/kg added Se) + 500 mg/kg added MCE; and (4) Combined group, basic diet (0.00 mg/kg added Se) + 0.50 mg/kg added nano-Se and 500 mg/kg added MCE. The activities of serum SOD, CAT, and GSH-Px of sows and IUGR piglets were significantly increased in MCE group and combined group, and the MDA content was extremely decreased. There were extreme differences in serum IgG level of sows and IUGR piglets, colostrum, and serum IgM level of IUGR piglets in MCE group and combined group compared with CON group. Maternal combined diets increased greatly the levels of serum IL-10 and IFN-γ of sows and IUGR piglets, and decreased extremely the contents of serum IL-1β and TNF-α. MCE alone or combination with nano-Se in sow diets decreased greatly mRNA level of Hsp70 and increased mRNA level of Hsp27 in sows and IUGR piglets. Salinity and alkalinity are important indicators of the aquatic environment, and saline-alkali stress has negative effects on aquatic organisms. Dietary supplementation with probiotics can improve the overall health and productivity of aquatic animals. Bacillus subtilis is a widely used probiotic in aquaculture. A total of 240 Carassius auratus gibelio (13.85 ± 0.15 g) were assigned to four groups. This experiment used a completely randomized design with three replicates and a 2 × 2 factorial scheme, with two saline-alkali concentrations (0 g/L NaCl + 0 g/L NaHCO3 and 5 g/L NaCl + 2.52 g/L NaHCO3) and two diets (control and supplemented with 108 CFU/g B. subtilis). After 12, 24, 48, 72, and 96 h of saline-alkali exposure, fish blood samples were collected and serum levels of immunoglobulin M, alkaline phosphatase, and acid phosphatase were measured. Gills and kidneys were sampled to analyze superoxide dismutase activity, catalase activity, malondialdehyde content, and histopathological alterations. The results suggested that a B. subtilis-supplemented diet may enhance immunity, improve antioxidant ability, and reduce tissue damage in C. gibelio exposed to saline-alkali stress. Adult prematurely aging mice (PAM), characterized by inadequate stress response, show premature immunosenescence and shorter lifespan compared to adult exceptional non-PAM (E-NPAM). Aging can be influenced by lifestyle factors, such as social environment. The continuous cohabitation of female PAM with E-NPAM improved behavioral responses, immunity, redox state, and longevity of PAM, but caused deterioration in E-NPAM. The objective of this study was to investigate whether the social interaction of only 15 min/day for 2 months of PAM with E-NPAM, can produce that improvement in PAM without causing deterioration in E-NPAM. After that short social interaction PAM and E-NPAM were submitted to behavioral tests and peritoneal leukocytes were collected to assess immune functions, oxidative and inflammatory state parameters as well as catecholamine concentrations. The lifespan of each animal was recorded. Plasmatic concentration of oxytocin was also studied. Results showed that PAM presented better behavioral responses, immunity and oxi-inflammatory state after interacting with E-NPAM, and consequently a longer lifespan. E-NPAM, in general, did not show any changes after interaction with PAM, not affecting their longevity either. In conclusion, a short social interaction between PAM and E-NPAM could be an excellent strategy for improving the health state and longevity of PAM without causing deleterious effect on E-NPAM. Background: Exercise training can positively impact the immune system and particularly natural killer (NK) cells, at least in healthy people. This effect would be of relevance in the context of cancer given the prominent role of these cells in antitumor immunity. In this systematic review and meta-analysis, we aimed to summarize current evidence on the effects of exercise training on the levels and function of NK cells in cancer survivors (i.e., from the time of diagnosis until the end of life). Methods: Relevant articles were searched in PubMed, Scopus, Web of Science and Cochrane Central Register of Controlled Trials (until January 11, 2022). Randomized controlled trials (RCT) of exercise training (i.e., non-acute) interventions vs usual care conducted in cancer survivors and assessing NK number and/or cytotoxic activity (NKCA) before and upon completion of the intervention were included. Methodological quality of the studies was assessed with the PEDro scale, and results were meta-analyzed using a random effects (Dersimoian and Laird) model. Results: Thirteen RCT including 459 participants (mean age ranging 11–63 years) met the inclusion criteria. Methodological quality of the studies was overall fair (median PEDro score = 5 out of 10). There was heterogeneity across studies regarding cancer types (breast cancer, non-small cell lung cancer and other solid tumors), treatment (e.g., receiving vs having received chemotherapy), exercise modes (aerobic or resistance exercise, Tai Chi, Yoga) and duration (2–24 weeks). No consistent effects were observed for NK number in blood (mean difference [MD]: 1.47, 95% confidence interval [CI] − 0.35 to 3.29, p = 0.113) or NKCA as assessed in vitro (MD: − 0.02, 95%CI − 0.17 to 0.14, p = 0.834). However, mixed results existed across studies, and some could not be meta-analyzed due to lack of information or methodological heterogeneity. Conclusions: Current evidence does not support a significant effect of exercise training intervention on NK cells in blood or on their 'static response' (as assessed in vitro) in cancer survivors. Light at night is a pervasive problem in our society; over 80% of the world's population experiences significant light pollution. Exacerbating this issue is the reality that artificially lit outdoor areas are growing by 2.2% per year and continuously lit areas brighten by 2.2% each year due to the rapid growths in population and urbanization. Furthermore, the increase in the prevalence of night shift work and smart device usage contributes to the inescapable nature of artificial light at night (ALAN). Although previously assumed to be innocuous, ALAN has deleterious effects on the circadian system and circadian-regulated physiology, particularly immune function. Due to the relevance of ALAN to the general population, it is important to understand its roles in disrupting immune function. This review presents a synopsis of the effects of ALAN on circadian clocks and immune function. We delineate the role of ALAN in altering clock gene expression and suppressing melatonin. We review the effects of light at night on inflammation and the innate and adaptive immune systems in various species to demonstrate the wide range of ALAN consequences. The objective of this study was to investigate the effect of Chinese herbal medicines (CHMs) on the productive performance, egg quality, nutrient retention and immune function of Hy-Line Brown laying hens. A total of 480 23-week-old Hy-Line Brown laying hens were randomly allotted to four groups with six replicates of twenty birds each. The dietary treatments were control (basal diet), and CHMs groups (basal diet supplemented with 4, 8 and 12 g kg−1 dry weight CHMs). After 42 days, 8 and 12 g kg−1 dry weight CHMs significantly increased the productive performance, retention of crude protein, egg albumen height, egg yolk, yolk color, haugh unit, egg yolk cholesterol concentration and indexes of immune organs when compared with control groups (p < 0.05). However, no statistically significant differences were observed between 8 and 12 g kg−1 dry weight CHMs. Moreover, there were no significant differences in the retention of dry matter and gross energy, egg specific gravity and shell thickness between treatment and control groups. Furthermore, cytokine levels in the jejunum and ileum mucosa of birds fed the 8 g kg−1 dry weight CHMs diet were greater than those of birds fed the 4 and 12 g kg−1 dry weight CHMs and control diets. The study indicated that dietary supplementation of CHMs is a viable strategy for improving production performance, egg quality, nutrient retention, immune function of laying hens. Selenium is an essential trace element that can regulate the function of immnue cells via selenoproteins. However, the effects of selenium on human dendritic cell (DCs) remain unclear. Thus, selenoprotein levels in monocytes, immature DCs (imDCs) and mature DCs (mDCs) treated with or without Na2SeO3 were evaluated using RT-PCR, and then the immune function of imDCs and mDCs was detected by flow cytometry, cell counting and the CCK8 assay. In addition, the effects of Se on cytokine and surface marker expression were investigated by RT-PCR. The results revealed different expression levels of selenoprotein in monocytes, imDCs and mDCs, and selenoproeins could be regulated by Se. Moreover, it was indicated that anti-phagocytic activity was improved by 0.1 µM Se, whereas it was suppressed by 0.2 µM Se in imDCs; The migration of imDCs and mDCs was improved by 0.1 µM Se, whereas their migration was inhibited by treatment with 0.05 or 0.2 µM Se; The mixed lymphocyte reaction of mDCs was improved by 0.1 µM Se, and it was inhibited by 0.05 and 0.2 µM Se. In addition, 0.1 µM Se improved the immune function of DCs through the regulation of CD80, CD86, IL12-p35 and IL12-p40. Wheres 0.05 and 0.2 µM Se impaired immune function of DCs by up-regulation of interleukin (IL-10) in imDCs and down-regulation of CD80, CD86, IL12-p35 and IL12-p40 in mDCs. Objective: To explore the antitumor effects of ethanol extract from Ventilago leiocarpa Benth (EEVLB) on sarcoma 180 (S180) tumor-bearing mice and the potential mechanism. Methods: Sixty mice were randomly assigned to 6 groups according to a random number table: normal group, model group, 5-fluorouracil (5-FU) group (0.02 g·kg−1), and high-, medium-, low-dose EEVLB groups (100, 84, and 56 g of raw material·kg−1 body weight, respectively), with 10 mice each group. All treatments were given once daily for 10 consecutive days. Effects of EEVLB on inhibiting tumor growth and immune function in mice were evaluated among all groups after the treatments by detecting tumor inhibition rate, organ index, serum levels of interleukin (IL)-2, -6, -10, CD3+CD4+ T lymphocytes, CD4+/CD8+ ratio, caspase-3 and Bcl-2. Results: EEVLB with different concentrations achieved inhibition of tumor growth in vivo, wherein the high-dose group showed the most significant reduction in tumor weight and increased apoptosis of tumor cells (P<0.05). In addition, both net weight gain and spleen index of mice showed uptrend in EEVLB treatment groups (P<0.05). Besides, serum levels of IL-2 and IL-6, percentages of CD3+CD4+ T lymphocytes and ratio of CD4+/CD8+ in peripheral blood were elevated in high- and medium-dose EEVLB groups compared with the model group (P<0.05). Also, upregulation of caspase-3 and downregulation of Bcl-2 were observed at protein levels in the high-dose EEVLB group (P<0.01). Conclusions: EEVLB exhibits promising antitumor activity in vivo. Seasonal variation in immune function can be attributed to life history trade-offs, and to variation in environmental conditions. However, because phenological stages and environmental conditions co-vary in temperate and arctic zones, their separate contributions have not been determined. We compared immune function and body mass of incubating (female only), chick-feeding (female and male), and non-breeding (female and male) red-capped larks Calandrella cinerea breeding year-round in three tropical equatorial (Kenya) environments with distinct climates. We measured four immune indices: haptoglobin, nitric oxide, agglutination, and lysis. To confirm that variation in immune function between breeding (i.e., incubating or chick-feeding) and non-breeding was not confounded by environmental conditions, we tested if rainfall, average minimum temperature (Tmin), and average maximum temperature (Tmax) differed during sampling times among the three breeding statuses per location. Tmin and Tmax differed between chick-feeding and non-breeding, suggesting that birds utilized environmental conditions differently in different locations for reproduction. Immune indices did not differ between incubating, chick-feeding and non-breeding birds in all three locations. There were two exceptions: nitric oxide was higher during incubation in cool and wet South Kinangop, and it was higher during chick-feeding in the cool and dry North Kinangop compared to non-breeding birds in these locations. For nitric oxide, agglutination, and lysis, we found among-location differences within breeding stage. In equatorial tropical birds, variation in immune function seems to be better explained by among-location climate-induced environmental conditions than by breeding status. Selenium levels can regulate the function of T cells, macrophages, B cells, natural killer cells and other immune cells. However, the effect of selenium on the immune function of dendritic cells (DCs) isolated from selenium-supplemented mice is unknown. In this study, C57BL/6J mice were randomly divided into three groups and fed diets containing low (0.08 ppm), medium (0.25 ppm) or high (1 ppm) selenium levels for 8 weeks. Immature (imDCs) and mature (mDCs) dendritic cells were then isolated from the bone marrow. Next, the migration, phagocytic capacity and mixed lymphocyte reaction (MLR) for imDCs and mDCs were detected by transwell and flow cytometry. The levels of C-C chemokine receptor type 7 (CCR7), major histocompatibility complex II (MHCII) and reactive oxygen species (ROS) were assayed by flow cytometry. F-actin and superoxide dismutase (SOD) activity was detected by fluorescence microscopy and SOD assay kit, respectively. In addition, the extracellular signal-regulated kinase (ERK), Akt, Ras homolog gene family member A/Rho-associated protein kinase (RhoA/ROCK) signalling, selenoprotein K (SELENOK) and glutathione peroxidase 1 (GPX1) levels were measured by western blot analysis. The results indicated that selenium deficiency enhanced the migration of imDCs by ROS and SELENOK-mediated ERK, Akt and RhoA/ROCK pathways but impaired the antigen uptake of imDCs. Although a high selenium level inhibited the migration of imDCs, it had no effect on phagocytic capacity. For mDCs, low selenium levels impaired free migration, and high levels inhibited the chemotactic migration involved in F-actin and CCR7, respectively. Low and high selenium levels impaired the MLR by inhibiting MHCII surface localisation, which might be related to ROS- and SELENOK-mediated ERK, Akt and RhoA/ROCK signalling pathways. The Wumeng semi-fine wool sheep is vital to the production system in Southwest China. To study the effects of nano-selenium (Nano-Se) poisoning in the Wumeng semi-fine wool sheep, poisoning model was established. A total of 20 animals with an average initial body weight (BW) of 35.57 ± 3.31 kg were used in this study. After 1-week-long acclimatization period, all animals were randomly divided into two groups: (1) control, with no any treatment; (2) Nano-Se group, orally administered 5 mg Nano-Se (BW/kg/day). There were 10 replications/group and one sheep/replication. The experiment lasted for 30 days. Compared with the control, hemoglobin, erythrocyte count, and packed cell volume in Nano-Se group markedly decreased (P < 0.01), and the activities of serum lactate dehydrogenase, glutamic oxaloacetic transaminase, cereal third transaminase, and alkaline phosphatase in Nano-Se group significantly increased (P < 0.01). T3 and FT3 in serum in Nano-Se group were greatly lower than those in the control (P < 0.01). Contents of T4, FT4, and TSH in serum in Nano-Se group were extremely higher than those in the control (P < 0.01). Levels of IgG, IgM, IgA, interleukin 2 (IL-2), and tumor necrosis factor-alpha (TNF-α) in Nano-Se group were much lower than those of the control (P < 0.01), but levels of interleukin 6 (IL-6) and interleukin-1β (IL-1β) were greatly higher than those in the control (P < 0.01). Compared with the control, serum superoxide dismutase, glutathione peroxide, total antioxidant capacity, and catalase from Nano-Se group extremely decreased (P < 0.01), and content of serum malondialdehyde in Nano-Se group markedly increased (P < 0.01). The current results indicated that the blood parameters in the Wumeng semi-fine wool sheep were affected by Nano-Se poisoning, and the immune function and antioxidant capacity were greatly reduced too. Lung squamous cell carcinoma (LUSC) kills more than four million people yearly. Creating more trustworthy tumor molecular markers for LUSC early detection, diagnosis, prognosis, and customized treatment is essential. Cuproptosis, a novel form of cell death, opened up a new field of study for searching for trustworthy tumor indicators. Our goal was to build a risk model to assess drug sensitivity, monitor immune function, and predict prognosis in LUSC patients. The 19 cuproptosis-related genes were found in the literature, and patient genomic and clinical information was collected using the Cancer Genomic Atlas (TCGA) database. The LUSC patients were grouped using unsupervised clustering techniques, and 7626 differentially expressed genes were identified. Using univariate COX analysis, LASSO regression analysis, and multivariate COX analysis, a prognostic model for LUSC patients was developed. The tumor immune escape was evaluated using the Tumor Immune Dysfunction and Exclusion (TIDE) method. The R packages ‘pRRophetic,’ ‘ggpubr,’ and ‘ggplot2’ were utilized to examine drug sensitivity. For modeling, a 6-cuproptosis-based gene signature was found. Patients with high-risk LUSC had significantly worse survival rates than those with low-risk conditions. The possibility of tumor immunological escape was increased in patients with higher risk scores due to more immune cell inactivation. For patients with high-risk LUSC, we discovered seven potent potential drugs (AZD6482, CHIR.99021, CMK, Embelin, FTI.277, Imatinib, and Pazopanib). In conclusion, the cuproptosis-based genes predictive risk model can be utilized to predict outcomes, track immune function, and evaluate medication sensitivity in LUSC patients. 17α-Ethynylestradiol (EE2) is a synthetic hormone that has become a widespread problem in the aquatic environments. The immunomodulatory effects of EE2 in fish have attracted much attention in recent years; however, little is known about its immunotoxicological impacts. This study was conducted to investigate the immune function of female Siamese fighting fish exposed to an environmental concentration of EE2, 10 ng/L. Aggressive reproductive behavior in this species often results in injuries to females after spawning, and thus they are a good candidate in immunotoxic actions of EE2. Fish were exposed to EE2 for 15 days and then distributed for breeding. Immune function, pathogen resistance, and survival rate were assessed after spawning. Immune responses including serum concentrations of IgM, lysozyme, complement, and bactericidal activity were lower in EE2-exposed females than the control fish. Also, EE2-exposed females showed higher mortality rate regardless of whether they were challenged with the bacterium. These results suggest that EE2 suppressed immune responses of spawned female fighting fish, thereby reducing its potential to resist against opportunistic pathogens. [ABSTRACT FROM AUTHOR]

This study explored the relationship between immunological status and clinical characteristics of aplastic anemia (AA) patients to plasma aluminum levels, which were increased after constant exposure to high levels of this metal. Sixty-two AA patients (33 cases with high and 29 cases with low or no exposure to aluminum) and 30 healthy controls were selected for this study. Aluminum in human albumin solution was measured by inductivity coupled plasma mass spectrometry. IL-10, IL-12, IL-17, and INF-γ levels were measured by enzyme-linked immunosorbent assay. The distribution of lymphocyte subsets were determined by flow cytometry. The expression levels of immunoglobulins and complement C3 and C4 were also measured. Exposure to high aluminum raised the levels of serum aluminum in AA patients (P < 0.01). The levels of hemoglobin and complement C4 were lower in AA patients with high aluminum exposure (P < 0.05 and < 0.01, respectively). The percentage of CD4+ T cells and the ratio of CD4+/ CD8+T cells in peripheral blood in AA patients with high aluminum exposure were higher compared with control AA patients (P < 0.05 in both cases), while the percentage of CD8+ T cells was significantly lower than that in non-aluminum–exposed AA patients (P < 0.05). Compared with non-aluminum–exposed AA patients, the level of IL-10 in the high aluminum–exposed AA group was significantly higher (P < 0.05 in both cases). The immunological and clinical characteristics of AA patients from regions of high aluminum exposure are different to those in from non-aluminum areas. Avian obligate brood parasites lay their eggs in the nests of other species that may provide care for the foreign offspring. Brood parasitism often imparts substantial fitness losses upon host nestlings when they are raised alongside the typically more competitive, larger, and older parasitic chick(s). Whereas fitness costs due to reduced host offspring survival in parasitized broods have been studied in detail, the physiological changes in host nestlings caused by parasitic nestmate(s) are less well known. We compared prothonotary warbler (Protonotaria citrea) nestlings, a host of the nest-sharing brown-headed cowbird (Molothrus ater), in experimentally parasitized vs. non-parasitized broods. Our aim was to determine whether cohabitation with brood parasitic young impacted host nestling baseline corticosterone plasma concentrations, immune responses, body condition, and mortality. Corticosterone levels and body condition of host nestlings were similar between nests with or without a cowbird nestmate, whereas host immune responses were lower and nestling mortality was greater in parasitized broods, irrespective of variation in brood size or total brood mass. We detected no trade-offs of baseline corticosterone levels with either immune responses or with body condition. These results suggest that this host species' nestlings experience some adverse fitness-relevant physiological effects in parasitized broods, but are also resilient in other aspects when coping with brood parasitism. Sexual minority (SM) adults (those who are lesbian, gay, or bisexual) consistently report more health problems compared to heterosexuals, and they tend to experience excess social stress. Although numerous studies have established links between social stress and clinical outcomes in SM adults, few studies have examined biological factors that may help explain how social stress leads to health disparities among SM adults. We used data from the Midlife in the United States Study (MIDUS) to examine whether two inflammatory markers that have been commonly associated with social stress—C-reactive protein (CRP) and interleukin-6 (IL-6)—differed by sexual orientation and whether any differences were explained by perceptions of discrimination. Participants self-identified as heterosexual (n = 1956) or lesbian, gay, or bisexual (n = 81). After controlling for age, gender, race, and education, SM individuals had higher CRP and IL-6 than heterosexuals on average and these differences were partially explained by perceptions of discrimination. The study was conducted to evaluate the effects of heat stress on antioxidant status, immune response, and related gene expression of lambs. Eighteen male lambs were randomly allocated into three treatment groups that were as follows: indoor temperature control group furnished with an air-conditioner (ITC), indoor temperature non-control group suffered intermittent and varying degrees of heat stress (ITNC), outdoor temperature non-control group in the external natural environment (OTNC). ITNC group presented a more severe and prolonged exposure to thermal stress than the other two groups. The trial lasted 28 days. Blood samples were collected on days 14 and 28 to analyze total superoxide dismutase (T-SOD), catalase (CAT), glutathione peroxidase (GPx) activities, malondialdehyde (MDA) concentrations, total antioxidant capacity (T-AOC), interleukin-1 (IL-1), interleukin-2 (IL-2), tumor necrosis factor-alpha (TNF-α), immunoglobulin A (IgA), immunoglobulin G (IgG), and immunoglobulin M (IgM) concentrations and gene expressions of SOD1, SOD2, GPx, CAT, nuclear factor erythroid 2-related factor 2 (Nrf2), IL-1β, IL-2, and TNF-α. Results showed that on day 14 an elevated temperature reduced (p < 0.05) the level of GPx, T-SOD, T-AOC, and IgG, whereas significantly increased (p < 0.05) CAT, MDA, IgA, and TNF-α levels. Gene expressions of SOD1 and GPx were down-regulated (p < 0.05). On day 28, ITNC group significantly decreased (p < 0.05) CAT, GPx, T-SOD, T-AOC, IgG, and IL-2 levels and increased (p < 0.05) MDA, IL-1, and TNF-α levels, accompanying by the reducing or increasing (p < 0.05) of their relative gene expression apart from CAT and IL-1β gene (p > 0.10). In addition, in ITNC and OTNC groups, the level of CAT, T-SOD, T-AOC, IgG, IgM, and IL-2 and the expression of CAT, SOD2, and IL-2 reduced (p < 0.05), whereas IL-1 and TNF-α levels and IL-1β expression increased (p < 0.05) on day 28 compared with day 14. In ITC group, the level of IgA, IL-1, and TNF-α and the expression of IL-1β and TNF-α increased (p < 0.05), while the content of IgG and IgM reduced (p < 0.05) on day 28 compared with day 14. These results indicated that heat stress negatively affected the antioxidant status and immune response of lambs, and the negative effects of heat stress are not only related to the stress duration but also associated with the stress severity. People living with HIV (PLWH) in the antiretroviral therapy (ART) era may lose more life-years to tobacco use than to HIV. Yet, smoking rates are more than twice as high among PLWH than the general population, contributing not just to mortality but to other adverse health outcomes, including neurocognitive deficits (neuroHIV). There is growing evidence that synergy with chronic inflammation and immune dysregulation that persists despite ART may be one mechanism by which tobacco smoking contributes to neuroHIV. This review will summarize the differential effects of nicotine vs tobacco smoking on inflammation in addition to the effects of tobacco smoke components on HIV disease progression. We will also discuss biomarkers of inflammation via neuroimaging as well as biomarkers of nicotine dependence (e.g., nicotine metabolite ratio). Tobacco smoking and nicotine may impact ART drug metabolism and conversely, certain ARTs may impact nicotine metabolism. Thus, we will review these bidirectional relationships and how they may contribute to neuroHIV and other adverse outcomes. We will also discuss the effects of tobacco use on the interaction between peripheral organs (lungs, heart, kidney) and subsequent CNS function in the context of HIV. Lastly, given the dramatic rise in the use of electronic nicotine delivery systems, we will discuss the implications of vaping on these processes. Despite the growing recognition of the importance of addressing tobacco use among PLWH, more research is necessary at both the preclinical and clinical level to disentangle the potentially synergistic effects of tobacco use, nicotine, HIV, cognition and immune dysregulation, as well as identify optimal approaches to reduce tobacco use. The Wumeng semi-fine wool sheep is vital to the production system of the Wumeng mountainous area in Southwest China. To study the effect of nano-copper poisoning in the Wumeng semi-fine wool sheep, nano-copper poisoning model was established in sheep. We determined blood physiological and biochemical parameters, immune indexes, and antioxidant indicators. The results showed that Hb, RBC, and PCV levels in blood from the nano-copper group were markedly lower (P < 0.01) than those in the control group. Serum LDH, AST, ALT, CPK, and Cp from the nano-copper group were significantly higher (P < 0.01) than those in control animals. Serum SOD, GSH-Px, CAT, and T-AOC from the nano-copper group were significantly lower (P < 0.01) than those in control sheep, and MDA content in serum from the nano-copper group were markedly higher (P < 0.01) than those in control sheep. The levels of IL-2, IL-6, IL-1β, IgA, IgM, and IgG from the nano-copper group were significantly lower (P < 0.01) than those in the control group. Upstream stimulatory factors are kinds of multi-functional transcription factors, which are expressed in eukaryotes widely, including Upstream stimulatory factor 1 (USFl) and upstream stimulatory factor 2 (USF2). USF protein has a typical basic helix-loop-helix leucine zipper (b-HLH-LZ) structure, which is involved in cell cycle, cell proliferations, glucose and lipid metabolism, and other biochemical processes. Although the USF family is an important regulator of cellular processes, little is known about the USF genes of lampreys, especially their evolutionary relationships, expression profiles, and biological functions. Here, an upstream stimulatory factor 2 (USF2) homolog from lamprey (Lethenteron reissneri) was identified and characterized (designated as L-USF2) because it is closer to USF2 subfamily than to USF1 subfamily. The cDNA fragment of L-USF2 has an open reading frame (ORF) of 765-bp length, encodes 254 amino acids, and contains an HLH domain at the c-terminal of amino acids. Meanwhile, motifs and genetic structure analysis reveal that USF2 gene exons are conserved. Moreover, the 3D structure analysis indicates that L-USF2 adopts the general USF2 folding and has a high structural similarity with H-USF2. The synteny results showed that the L-USF2 adjacent gene changed greatly compared with the jaw vertebrates. By real-time quantitative experiment and Western blot analysis, we found that L-USF2 gene played a significant role in the immune responses. This study not only provides us with a further understanding of the evolution and function of the USF gene family but also provides a basis for exploring its immune responses and immune defenses in lampreys. Purpose: There are limited data regarding postoperative complications and autoimmune reactions caused by surgery in early-stage cervical cancer patients who underwent laparoscopic radical resection (LRR). This study aimed to investigate the therapeutic effect of LRR of cervical cancer patients and its effect on cytokines.Methods: 168 patients with cervical cancer were enrolled. The patients were divided into open group and laparoscopic group according to the random number table method, with 84 cases in each group. The surgical-related indexes and the incidence of complications of the two groups were observed, and the IFN-γ, TNF, and IL-1/2/4/6/8/10/12 levels in peripheral blood were compared before and after surgery in both groups.Results: The operation time of the patients in the laparoscopic group was significantly shorter than that in the open group (119.56 ± 45.26 vs. 206.36 ± 54.39, P < 0.01). The intraoperative blood loss in the laparoscopic group was significantly less than that in the open group (155.29 ± 57.58 vs. 529.58 ± 162.4, P < 0.01). The postoperative visual analog scale (VAS) score was also significantly lower than that in the open group (3.65 ± 0.88 vs. 6.32 ± 1.12, P < 0.01). There was no significant difference in the incidence of complications between the two groups. The degree of inflammatory cytokines changes caused by LRR was less than that of open radical surgery (P < 0.001).Conclusions: LRR surgery has less stress on patients with early cervical cancer than open surgery within 5 days after surgery, which has certain reference value for early cervical cancer treatment. Se-methylselenocysteine (MeSeCys) is a natural organic selenium (Se) supplement. However, its effects on animal nutrition are poorly understood. This study compared the effects of sodium selenite (SeNa), MeSeCys, and selenomethionine (SeMet) on immune function, tissue Se concentration, meat quality, and selenoprotein gene expression in pigs. A total of 72 finishing pigs were divided into four groups, which received a basal diet (BD, 0.1 mg Se/kg) without Se supplementation or one supplemented with SeNa, MeSeCys, or SeMet at a concentration of 0.25 mg Se/kg. Organic Se supplementation significantly increased the immune globulin A (IgA), IgG, and IgM serum levels compared with BD and SeNa groups (P < 0.05). There were no statistically significant differences in growth performance among the four groups. SeMet was more efficient in increasing Se concentrations in the heart, muscle, and liver than MeSeCys and SeNa (P < 0.05), while no statistically significant differences were observed between MeSeCys and SeNa. Se supplementation significantly decreased the pressing muscle loss compared with the BD group (P < 0.05). Meat color and pH were not significantly affected. Se supplement effects on liver selenoprotein gene mRNA level enhancement were ranked as follows: MeSeCys > SeMet > SeNa (P < 0.05). In muscle tissues, only the SELENOW mRNA level was significantly increased by the MeSeCys and SeMet treatment, compared with the SeNa group. In conclusion, SeMet was more efficient in increasing Se concentrations than MeSeCys and SeNa in pigs, while MeSeCys was more efficient in enhancing selenoprotein gene expression than SeMet and SeNa. Monocarboxylate transporter 1 (MCT1) represents a potential therapeutic target in cancer. The objective of this study was to determine the efficacy of AZD3965 (a specific inhibitor of MCT1) and α-cyano-4-hydroxycinnamic acid (CHC, a nonspecific inhibitor of MCTs) in the murine 4T1 tumor model of triple-negative breast cancer (TNBC). Expression of MCT1 and MCT4 in 4T1 and mouse mammary epithelial cells were determined by Western blot. Inhibition of MCT1-mediated l-lactate uptake and cellular proliferation by AZD3965 and CHC was determined. Mice bearing 4T1 breast tumors were treated with AZD3965 100 mg/kg i.p. twice-daily or CHC 200 mg/kg i.p. once-daily. Tumor growth, metastasis, intra-tumor lactate concentration, immune function, tumor MCT expression, and concentration-effect relationships were determined. AZD3965 and CHC inhibited cell growth and l-lactate uptake in 4T1 cells. AZD3965 treatment resulted in trough plasma and tumor concentrations of 29.1 ± 13.9 and 1670 ± 946 nM, respectively. AZD3965 decreased the tumor proliferation biomarker Ki67 expression, increased intra-tumor lactate concentration, and decreased tumor volume, although tumor weight was not different from untreated controls. CHC had no effect on tumor volume and weight, or intra-tumor lactate concentration. AZD3965 treatment reduced the blood leukocyte count and spleen weight and increased lung metastasis, while CHC did not. These findings indicate AZD3965 is a potent MCT1 inhibitor that accumulates to high concentrations in 4T1 xenograft tumors, where it increases tumor lactate concentrations and produces beneficial effects on markers of TNBC; however, overall effects on tumor growth were minimal and lung metastases increased. The maturation and development of T cells were not completed until T cells were selected in thymus. It was not until the early 1960s that j.f.a.p. discovered the importance of thymus in T cell development. Twelve healthy piglets were randomly divided into two groups, the experimental group (LGG group) and the control group (saline group). The LGG group piglets were given 1 ml LGG (6 × 109 CFU/ml) per day. The saline group was given 1 ml of normal saline per day. The piglets were slaughtered at 30 days and 45 days, respectively, and the MLN, jejunum and ileum PPs, LP of the piglets were taken. The expression of CD3+CD4+ T lymphocytes was detected by flow cytometry, and intestinal villi development was observed by intestinal paraffin section. The results showed that the flow cytometry results at 30 days and 45 days showed that the CD3+CD4+ T lymphocytes in the MLN group were significantly different from those in the saline group (P < 0.05, P < 0.01).The CD3+CD4+ T lymphocytes in the jejunum PP of piglets in LGG group were significantly different from those in saline group (P < 0.05). The CD3+CD4+ T lymphocytes in the ileum PP of the LGG group were significantly different from those in the saline group (P < 0.05, P < 0.01). The long-standing interest in the effects of religiosity and spirituality (R/S) on health outcomes has given rise to a large and diverse literature. We conducted a meta-analysis on research involving R/S and physiological markers of health to elucidate both the scope and mechanism(s) of this phenomenon. A combined analysis found a significant, but small, beneficial effect. Subgroup analyses found that some measures of both extrinsic and intrinsic religiosity were significantly associated with health. Several outcome measures, including blood pressure, C-reactive protein, and cardiovascular health markers, were significantly associated with R/S. Our findings suggest that R/S benefits health, perhaps through minimizing the disruptive effects of stress/depression on inflammation. We hope that researchers can use these results to guide efforts aimed at elucidating the true mechanism(s) linking religious/spiritual beliefs and physical health. [ABSTRACT FROM AUTHOR]
