Purpose: Management of scrotal hernias presents as a common challenge, with operative interventions to address these hernias associated with higher rates of morbidity compared to those of less-complex pathology. Surgeons have advocated for the use of techniques such as primary abandonment of the distal sac as a potential means to reduce complications for operative intervention, with preliminary findings demonstrating feasibility. We sought to assess outcomes related to primary sac abandonment among patients undergoing minimally invasive (MIS) repair of scrotal hernias. Methods: A review of prospectively maintained databases among two academic hernia centers was conducted to identify patients who underwent MIS inguinal hernia repairs with primary sac abandonment. Patient demographics, hernia risk factors, intraoperative factors, and postoperative outcomes were evaluated. Short-term outcomes related to patient-reported experiences and surgical-site occurrences requiring procedural intervention were queried. Results: Sixty-seven male patients [median age: 51.6 years; interquartile range (IQR): 45–65 years] underwent inguinal hernia repair with primary sac abandonment. Anatomic polypropylene mesh was used in 98.5% cases. Rates of postoperative complications were low and included postoperative urinary retention (6%), clinically identified or patient-reported seromas/hematomas within a 30-day follow-up period (23.9%), deep venous thrombosis (1.5%), and pelvic hematoma (1.5%). No seromas or hematomas necessitated procedural interventions, with resolution of symptoms within three months of their operation date. Conclusion: We report a multi-center experience of patients managed with primary abandonment of the sac technique during repair of inguinoscrotal hernias. NADH:ubiquinone oxidoreductase, respiratory complex I, plays a major role in cellular energy metabolism by coupling electron transfer with proton translocation. Electron transfer is catalyzed by a flavin mononucleotide and a series of iron-sulfur (Fe/S) clusters. As a by-product of the reaction, the reduced flavin generates reactive oxygen species (ROS). It was suggested that the ROS generated by the respiratory chain in general could damage the Fe/S clusters of the complex. Here, we show that the binuclear Fe/S cluster N1b is specifically damaged by H2O2, however, only at high concentrations. But under the same conditions, the activity of the complex is hardly affected, since N1b can be easily bypassed during electron transfer. [ABSTRACT FROM AUTHOR]

Structural snapshots of protein/ligand complexes are a prerequisite for gaining atomic level insight into enzymatic reaction mechanisms. An important group of enzymes has been deprived of this analytical privilege: members of the protein tyrosine phosphatase (PTP) superfamily with catalytic WPD-loops lacking the indispensable general-acid/base within a tryptophan-proline-aspartate/glutamate context. Here, we provide the ligand/enzyme crystal complexes for one such PTP outlier: Arabidopsis thaliana Plant and Fungi Atypical Dual Specificity Phosphatase 1 (AtPFA-DSP1), herein unveiled as a regioselective and efficient phosphatase towards inositol pyrophosphate (PP-InsP) signaling molecules. Although the WPD loop is missing its canonical tripeptide motif, this structural element contributes to catalysis by assisting PP-InsP delivery into the catalytic pocket, for a choreographed exchange with phosphate reaction product. Subsequently, an intramolecular proton donation by PP-InsP substrate is posited to substitute functionally for the absent aspartate/glutamate general-acid. Overall, we expand mechanistic insight into adaptability of the conserved PTP structural elements. Inositol pyrophosphates (PP-IPs) regulate immunity and phosphate homeostasis. Condensed phosphates may exist as linear, cyclic or branched structures. Due to their important role in nature, linear polyphosphates have been well studied. In contrast, branched phosphates (ultraphosphates) remain largely uncharacterised, because they were already described in 1950 as exceedingly unstable in the presence of water, epitomized in the antibranching-rule. This rule lacks experimental backup, since, to the best of our knowledge, no rational synthesis of defined ultraphosphates is known. Consequently, detailed studies of their chemical properties, reactivity and potential biological relevance remain elusive. Here, we introduce a general synthesis of monodisperse ultraphosphates. Hydrolysis half-lives up to days call the antibranching-rule into question. We provide evidence for the interaction of an enzyme with ultraphosphates and discover a rearrangement linearizing the branched structure. Moreover, ultraphosphate can phosphorylate nucleophiles such as amino acids and nucleosides with implications for prebiotic chemistry. Our results provide an entry point into the uncharted territory of branched condensed phosphates. The "anti-branching rule", introduced in 1950, excludes branched polyphosphates from biological relevance due to their supposedly rapid hydrolysis. Microwave Kinetic Inductance Devices (MKIDs) are poised to allow for massively and natively multiplexed photon detectors arrays and are a natural choice for the next-generation CMB Stage 4 experiment which will require 10 5 detectors. In this article, we discuss what noise performance of present generation MKIDs implies for CMB measurements. We consider MKID noise spectra and simulate a telescope scan strategy which projects the detector noise onto the CMB sky. We then analyze the simulated CMB + MKID noise to understand particularly low-frequency noise affects the various features of the CMB, and thusly set up a framework connecting MKID characteristics with scan strategies, to the type of CMB signals we may probe with such detectors. Hop with powdery mildew [HPM: caused by Podosphaera macularis (Wallr.) U. Braun & S. Takam.] results in significant losses in hop production by reducing yield and quality. One means of increasing yield and quality is the production of resistant hop lines. Breeding for resistance can be significantly improved and accelerated by use of marker-assisted selection. The objective of this preliminary study was to identify QTLs and markers for genetic resistance to HPM. A bi-parental mapping population between the resistant line “Newport” and susceptible line ‘21110M’. Phenotypic data was scored under controlled greenhouse conditions. Significant differences among offspring were observed and disease resistance did not follow a distinct binomial distribution, suggesting quantitative genetic control. Genotyping-by-sequencing resulted in approximately 375 K SNP markers, which were filtered down to 2263 markers mapped to 10 linkage groups. Interval Mapping identified four QTLs with one on linkage group 1 and three located on linkage group 6. Composite interval mapping identified three QTLs, all located on linkage group 6. Mixed linear models identified 15 markers associated with expression of resistance to HPM. Three of these 15 SNPs were also identified in QTL-CIM analysis. Evaluation of the scaffolds containing the significant SNP markers identified seven putative genes—several of which appear involved in disease resistance in other plant species. Increased labor costs and reduced labor pools for hop production necessitate the development of strategies that improve efficiency and automation of hop production. One solution for reducing labor inputs is the use of ‘‘short-trellis’’ hop varieties. Unfortunately, little information exists on the genetic control of this trait in hop, and there are no known molecular markers available for selection. This preliminary study was enacted to identify QTLs associated with expression of short-stature growth phenotype using SNPs identified within genome-assembled scaffolds. A bi-parental mapping population of 87 offspring was obtained from the cross, ‘‘Pioneer 9 25/95/15’’. Genotyping-by-sequencing was performed on parents and offspring. SNPs were identified using TASSEL v3.0 with either ‘Teamaker’ reference genome or ‘Shinsuwase’ genome. The genetic map derived from ‘Teamaker’ SNPs was far superior and was used for all further analysis. QTL analysis identified eight QTLs linked to short stature with five showing strong statistical association based upon three different statistical analyses. All eight QTLs were found on linkage group one. Evaluation of scaffolds containing SNP markers located at or surrounding QTL regions (±1 cM) identified 67 putative genes—several of which are known structural genes. A genome-wide scan of SNP markers identified an additional marker found on a scaffold containing a putative gene (Aspartyl protease family protein) known to induce dwarf characteristics in other species. Hop is one of the few dioecious plants with dimorphic sex chromosomes. Because the entire Cannabaceae family is dioecious, hop and other members of this family are thought to have a relatively older sex chromosomal system than other plant species. Hop cones are only produced in female hops with or without fertilization. This has lead to most genomic research being directed toward female plants. The work we present provides genomic resources surrounding male plants. We have produced a draft genome for the male hop line USDA 21422M using a novel genome assembly method. In addition, we identified a 1.3 Mb set of scaffolds, which appear to be the male specific region based upon specificity with male hop accessions. This set includes a smaller high confidence total length 18 Kb set of scaffolds, which are supported by over 500 individuals, including the USDA world collection of hop varieties and two mapping populations, with genotyping by sequencing. We also have identified a portion of the Teamaker × 21422M linkage map to be associated with the pseudo-autosomal region (PAR). Downy mildew (caused by Pseudoperonospora humuli) causes significant losses in hop ( Humulus lupulus L.) cone yield and quality, and potential crown death. Breeding for resistance has proved difficult presumably because of the highly quantitative nature of genetic control over expression. The objective of this study was to utilize multiple environments to ascertain downy mildew resistance and identify molecular markers linked to resistance. A widely segregating population, 'Teamaker' × USDA 21422M was inoculated and evaluated for downy mildew in field studies (Corvallis, OR and Yakima, WA) and a greenhouse study. Next generation sequencing using an Illumina HiSeq 2000 was performed on this population. Resulting reads were imported into UNEAK TASSEL 4.0 pipeline for processing and SNP-calls. Approximately 120,435 unfiltered SNP markers were identified and of these markers, 9081 high quality markers chosen for association analysis. Data analyses for field studies in OR and WA were performed separately using trait values averaged across blocks. Differential trait associated marker sets were identified across each physical environment with partial overlap between field environments. Mixed linear model analyses identified 39 markers (21 from OR, 10 from WA and eight overlapping) associated with response to downy mildew infection (p < 1 × 10). The most significant 17 markers were validated using high-resolution melting curve analysis and the resistant allele identified. Four SNP markers, along with the respective 'resistant' alleles, were classified as highly selective for resistance (model adjusted R = 0.437). Following the SARS outbreak, extensive surveillance was undertaken globally to detect and identify coronavirus diversity in bats. This study sought to identify the diversity and prevalence of coronaviruses in bats in the Australasian region. We identified four different genotypes of coronavirus, three of which (an alphacoronavirus and two betacoronaviruses) are potentially new species, having less than 90% nucleotide sequence identity with the most closely related described viruses. We did not detect any SARS-like betacoronaviruses, despite targeting rhinolophid bats, the putative natural host taxa. Our findings support the virus-host co-evolution hypothesis, with the detection of Miniopterus bat coronavirus HKU8 (previously reported in Miniopterus species in China, Hong Kong and Bulgaria) in Australian Miniopterus species. Similarly, we detected a novel betacoronavirus genotype from Pteropus alecto which is most closely related to Bat coronavirus HKU9 identified in other pteropodid bats in China, Kenya and the Philippines. We also detected possible cross-species transmission of bat coronaviruses, and the apparent enteric tropism of these viruses. Thus, our findings are consistent with a scenario wherein the current diversity and host specificity of coronaviruses reflects co-evolution with the occasional host shift. [ABSTRACT FROM AUTHOR]

Hop downy mildew (DM) is an obligate parasite causing severe losses in hop if not controlled. Resistance to this pathogen is a primary goal for hop breeding programs. The objective of this study was to identify QTLs linked to DM resistance. Next-generation-sequencing was performed on a mapping population segregating for DM resistance levels. Cloned plants were grown in a RCBD with three replicates under three environments: greenhouse (GH), field plots in Oregon (OR), Corvallis field plots in Washington (WA), Yakima). The linkage map of 3,341 SNP markers was determined with a four-stage process using Rqtl, TMAP, Joinmap v 4.0 and MERGEMAP. QTL analysis was performed using JMP Genomics and TASSEL 5.0. SNP markers were distributed across 11 linkage groups (LGs) with an average distance between markers of 0.2 cM and total distance of 745.9 cM. QTLs for all three environments were identified using multiple interval mapping. Overall heritability across the three environments varied from h = 0.38 (GH) to 0.57 (OR). A total of 22 QTLs across 8 LG were identified for DM resistance: 5 identified from OR field data, 12 using WA data and five from GH DM data. No epistasis was observed. This study points out the complexity of genetic control of DM resistance in hop and identifies several markers that can be potentially be used to select for DM resistance in hop. The stability of Al-Mn transition edge sensor (TES) bolometers is studied as we vary the engineered TES transition, heat capacity, and/or coupling between the heat capacity and TES. We present thermal structure measurements of each of the 39 designs tested. The data is accurately fit by a two-body bolometer model, which allows us to extract the basic TES parameters that affect device stability. We conclude that parameters affecting device stability can be engineered for optimal device operation, and present the model parameters extracted for the different TES designs. Upcoming experiments aim to produce high fidelity polarization maps of the cosmic microwave background. To achieve the required sensitivity, we are developing monolithic, feedhorn-coupled transition edge sensor polarimeter arrays operating at 150 GHz. We describe this focal plane architecture and the current status of this technology, focusing on single-pixel polarimeters being deployed on the Atacama B-mode Search (ABS) and an 84-pixel demonstration feedhorn array backed by four 10-pixel polarimeter arrays. The feedhorn array exhibits symmetric beams, cross-polar response <−23 dB and excellent uniformity across the array. Monolithic polarimeter arrays, including arrays of silicon feedhorns, will be used in the Atacama Cosmology Telescope Polarimeter (ACTPol) and the South Pole Telescope Polarimeter (SPTpol) and have been proposed for upcoming balloon-borne instruments. Implementation of molecular methods in hop ( Humulus lupulus L.) breeding is dependent on the availability of sizeable numbers of polymorphic markers and a comprehensive understanding of genetic variation. However, use of molecular marker technology is limited due to expense, time inefficiency, laborious methodology and dependence on DNA sequence information. Diversity arrays technology (DArT) is a high-throughput cost-effective method for the discovery of large numbers of quality polymorphic markers without reliance on DNA sequence information. This study is the first to utilise DArT for hop genotyping, identifying 730 polymorphic markers from 92 hop accessions. The marker quality was high and similar to the quality of DArT markers previously generated for other species; although percentage polymorphism and polymorphism information content (PIC) were lower than in previous studies deploying other marker systems in hop. Genetic relationships in hop illustrated by DArT in this study coincide with knowledge generated using alternate methods. Several statistical analyses separated the hop accessions into genetically differentiated North American and European groupings, with hybrids between the two groups clearly distinguishable. Levels of genetic diversity were similar in the North American and European groups, but higher in the hybrid group. The markers produced from this time and cost-efficient genotyping tool will be a valuable resource for numerous applications in hop breeding and genetics studies, such as mapping, marker-assisted selection, genetic identity testing, guidance in the maintenance of genetic diversity and the directed breeding of superior cultivars. Men's health issues have increasingly gained attention not only in the mass media, but also among most health-care providers. The diagnosis and treatment of male-related health problems, unfortunately, can lead to complications and error-related injuries resulting in claims of medical malpractice. This review article will look at the most common claims relating to complications and injuries in the management of men's health issues. Reviews of the literature over the past three decades using multiple search engines including PubMed were utilized. The most pertinent articles were selected on the basis of their relevance to men's health issues, complications and medico-legal ramifications. An evaluation of the literature reveals that although the number of claims against urologists has not increased over the past several decades, indemnity payments have continued to rise significantly. Claims can be divided into those relating to diagnosis and those relating to treatment. Providers of men's health care may become involved in claims of medical malpractice at some time during their careers. Patients’ care can result in complications and injuries, most of which do not lead to claims. Certain areas of men's health lead to more claims than others. Eight-hundred forty-nine patients with symptomatic nonerosive GERD from two clinical trials of lansoprazole 15 mg daily (LAN 15) and lansoprazole 30 mg daily (LAN 30) vs ranitidine 150 mg twice a day (RAN 150) completed a health-related quality-of-life (HRQoL) questionnaire at baseline and four and eight weeks after treatment. The questionnaire included the Short-Form 12, GERD symptoms, eating symptoms, social restrictions, problems with sleep, work disability, treatment satisfaction, and associated importance weights items. Both LAN groups reported greater, although not significant, improvement from baseline to week 8 versus RAN 150 in the majority of HRQoL scales. Treatment satisfaction was significantly higher at week 8 in both LAN groups. Quality-days incrementally gained analysis showed that both LAN groups gained significantly more quality days than RAN 150. Patients taking lansoprazole 15 or 30 mg daily reported better outcomes than those receiving ranitidine 150 twice a day over the eight-week study. [ABSTRACT FROM AUTHOR]

The current study determined the extracellular content of glutamate, 10 additional amino acids, lactate, glucose and some antioxidants in a rodent model of malignant glioma, its peritumoral space and the adjacent cortex. RG2 tumors were induced in the right frontal cortex of Fischer-344 rats ( n=10) by a standardized procedure to obtain a maximum sagittal tumor width of 3–4 mm diameter. After confirmation of tumor growth and localization by contrast enhanced MRI three microdialysis probes were implanted simultaneously in the cortex: at the tumor implantation site ( tumor), 2 mm caudally, brain around tumor ( BAT) and 4 mm caudally ( cortex) to the site of implantation. Dialysate concentrations of glutamate were increased 3.9-fold in tumor and 2-fold in BAT compared with cortex. Glycine was elevated 11.4-fold in tumor and 2.6-fold in BAT. Lactate was increased 1.7-fold in tumor, 1.2-fold in BAT. Levels of glucose, ascorbic acid and uric acid were not significantly different in tumor, BAT and cortex. The increased dialysate levels of glutamate and glycine in the peritumoral space may contribute to impaired neuronal function and epileptiform activity associated with this tumor type in humans. This study was conducted to test the psychometric properties of a newly developed health-related quality-of-life (HRQoL) questionnaire. A total of 172 symptomatic GERD patients completed a 57-item questionnaire (containing the SF-12; symptom frequency/bothersomeness; problems related to activities, sleep, work disability; overall HRQoL; and treatment satisfaction) at baseline, week 1 [retest (N = 25)], and week 4 [follow-up (N = 100)]. Internal-consistency reliability was acceptable for most scales (range: 0.74-0.92). Test-retest reliability was acceptable for most scales (ICC: 0.74-0.85). Construct validity was demonstrated based on observed correlations. Known-groups validity was upheld, as patients who experienced more symptom days and patients who reported higher pain reported worse HRQoL than those with less symptoms or less severe pain. When categorized according to change in pain severity, Guyatt's statistic for the "improved" and "worse" groups demonstrated responsiveness, although many of the scales for the "stable" group were also responsive. Objective: Patient-centered methods for evaluating treatments require validated preference-elicitation techniques. We describe the validation of two preference-elicitation approaches for use in an Extended Q-TWiST treatment evaluation. The first method was an "idiographic" approach, which attempts to capture intra-individual differences in the degree to which each domain distracted from and interfered with life activities. The second method, a Likert-scaled approach, asks patients to evaluate the importance of each quality-of-life (QOL) domain.Methods: Patient-reported QOL and preferences were assessed in participants with gastroesophageal reflux disease at baseline (n = 172), one week (n = 25), and 4 weeks after baseline (n = 100).Results: Both approaches demonstrated high internal consistency and the ability to discriminate known groups based on reported pain and number of days with symptoms. The idiographic approach exhibited responsiveness, although it was more highly correlated with QOL than the Likert-scaled approach. The Likert-scaled approach had good face validity but demonstrated low reliability compared to the idiographic approach.Conclusions: Both preference-elicitation methods exhibited promise as well as limitations. Future research should focus on increasing the reliability of the Likert-scaled approach, reducing the overlap between the idiographic approach and QOL, and examining the relationship between reliability and responsiveness for a range of illness trajectories. The amphibian pupillary response is mediated by two mechanisms. One is restricted to the iris itself, the other involves the central nervous system. In the present study we investigated the dynamic of the pupillary response and separated the parts by experiments with lesions of the optic and oculomotor nerve. The pupillary response of urodeles is comparable to that in other vertebrates. Differences are observable in amplitude and time course. Furthermore there are no binocular summation or consensual reactions. The central control of the pupillary response increases amplitude and velocity of the pupillary response in sudden luminance changes, compared to the pure iris related reaction. The separation of central and peripheral pupillary reflex control was shown more quantitatively through the introduction of a theoretical model. The investigation of the dark response gives evidence that there is no dilatator pupillae in urodele amphibians, since comparison to results in anuran species show that this is not a general feature in amphibians. This paper presents a study of the rectifying properties of heavily doped polycrystalline silicon (polysilicon) on 4H silicon carbide (4H-SiC). Current properties and barrier heights were found using analysis of the heterojunction. This revealed that Schottky analysis would be valid for the large barrier height devices. Isotype and an-isotype devices were fabricated on both p-type and n-type SiC and the electrical characteristics were investigated using capacitance vs voltage measurements, current vs voltage measurements (I-V), and temperature I-V measurements. Extraction of the barrier height, built-in potential, and Richardson constant were made and then compared to theoretical values for the heterojunction. Temperature I-V measurements demonstrated that the current transport mechanism is thermionic emission, confirming the validity of the Schottky diode model. The I-V characteristics show near ideal diode rectifying behavior and the capacitance-voltage characteristics show ideal junction space charge modulation for all polysilicon/SiC combinations. These experimental results match well with heterojunction band-offset estimated barrier heights and demonstrate that the barrier height of the polysilicon/4H SiC interface may be controlled by varying the polysilicon doping type. A novel method for measuring magnetostriction constants is presented. A strain, periodic in time, applied to the sample, causes a modulation of the ferromagnetic resonance line position. The height of the signal obtained after phase-sensitive detection is proportional to the strain modulation depth. The appropriate magnetostriction constant λ is obtained by comparing the height of the SMFMR signal with that of the FMR line, as recorded by means of magnetic field modulation. This paper reviews the reliability and validity of an instrument for assessing health-related quality of life (HRQOL) in CaPSURE(tm) (Cancer of the Prostate Strategic Urologic Research Endeavor), an observational database of men with biopsy-confirmed prostate cancer. The questionnaire includes the RAND 36 item Health Survey 1.0, the UCLA Prostate Cancer Index and items measuring self-esteem and the impact of prostate cancer in general and on the family. The reliability and validity of this instrument are reported for a group of 2,382 men enrolled in CaPSURE(tm). Individuals complete HRQOL questionnaires at enrolment and quarterly thereafter. The subscales of the instrument resulted in a high internal-consistency reliability across all subscales (range=0.75-0.94). The test-retest reliability was good with the exception of the comparative health item (intraclass correlation coefficient (ICC)=0.55). The concurrent validity data included moderately strong associations with subscales of similar concepts and the ability to distinguish between patients who are known to be different - newly diagnosed (within 6 months) and patients diagnosed and treated an average of 3 years ago. Overall, this instrument demonstrated good reliability and validity and supported the need for incorporating HRQOL as a component of outcomes assessment in the management of patients with prostate cancer. Species in the Laxa and Grandia groups of the genus Panicum are adapted to low, wet areas of tropical and subtropical America. Panicum milioides is a species with C photosynthesis and low apparent photorespiration and has been classified as a C/C intermediate. Other species in the Laxa group are C with normal photorespiration. Panicum prionitis is a C species in the Grandia group. Since P. milioides has some leaf characteristics intermediate to C and C species, its photosynthetic response to irradiance and temperature was compared to the closely related C species, P. laxum and P. boliviense and to P. prionitis. The response of apparent photosynthesis to irradiance and temperature was similar to that of P. laxum and P. boliviense, with saturation at a photosynthetic photo flux density of about 1 mmol m s at 30°C and temperature optimum near 30°C. In contrast, P. prionitis showed no light saturation up to 2 mmol m s and an optimum temperature near 40°C. P. milioides exhibited low CO loss into CO-free air in the light and this loss was nearly insensitive to temperature. Loss of CO in the light in the C species, P. laxum and P. boliviense, was several-fold higher than in P. milioides and increased 2- to 5-fold with increases in temperature from 10 to 40°C. The level of dark respiration and its response to temperature were similar in all four Panicum species examined. It is concluded that the low apparent photorespiration in P. milioides does not influence its response of apparent photosynthesis to irradiance and temperature in comparison to closely related C Panicum species. The normalized form of the amputated vertex functions of an unstable particle is derived from the requirement that the general Ansatz (1) for the scattering amplitude of two stable particles with a resonance in the energy-variable is unitary in the energy interval below the threshold for three particle production. Necessary and sufficient conditions for the convergence of the improper-graph-series (damping series) in certain regions of the p-plane are derived for the cases of both stable and unstable particles. The field-theoretical model - see chapter 2 - underlying the discussion is perturbation-theoretic. Non-observables (in the instable case) are treated in the familiar way only after suitable modifications of the Dyson-prescriptions have been made. The analysis of myo-inositol phosphates (InsPs) and myo-inositol pyrophosphates (PP-InsPs) is a daunting challenge due to the large number of possible isomers, the absence of a chromophore, the high charge density, the low abundance, and the instability of the esters and anhydrides. Given their importance in biology, an analytical approach to follow and understand this complex signaling hub is desirable. Here, capillary electrophoresis (CE) coupled to electrospray ionization mass spectrometry (ESI-MS) is implemented to analyze complex mixtures of InsPs and PP-InsPs with high sensitivity. Stable isotope labeled (SIL) internal standards allow for matrix-independent quantitative assignment. The method is validated in wild-type and knockout mammalian cell lines and in model organisms. SIL-CE-ESI-MS enables the accurate monitoring of InsPs and PP-InsPs arising from compartmentalized cellular synthesis pathways, by feeding cells with either [13C6]-myo-inositol or [13C6]-D-glucose. In doing so, we provide evidence for the existence of unknown inositol synthesis pathways in mammals, highlighting the potential of this method to dissect inositol phosphate metabolism and signalling. Koalas are an iconic Australian marsupial undergoing precipitous population reduction in South-East Queensland from complex interacting threats. To investigate the causes of death and the interaction of comorbidities with demography in South-East Queensland koalas, a large scale, high-throughput prospective necropsy survey was conducted spanning 2013–2016. During this period, 519 necropsies were conducted in 155 young/subadult koalas, 235 mature, 119 old koalas and 10 of unknown age. Similar numbers of males and females were assessed. Trauma and infectious disease at were the most common single diagnoses. However, comorbidity was frequent, including multicentric infection or infectious disease in combination with trauma or senescence. Female koalas had proportionally more reproductive chlamydiosis compared to males in which the ocular and urinary systems were more commonly affected. Comorbidity and disease were strongly associated with poor body condition, and trauma was associated with good body condition. Animals affected by motor vehicle trauma were often in better body condition than those affected by animal attack, tree fall or other causes of trauma. This study identified a higher frequency of infections and comorbidity then previously reported, confirming the complex nature of interacting threats to the koala population. [ABSTRACT FROM AUTHOR]
