The UK Farm-Scale Evaluations (FSE) compared the effects on biodiversity of management of genetically modified herbicide-tolerant (GMHT) spring-sown crops with conventional crop management. The FSE reported larger weed abundance under GMHT management for fodder maize, one of three crops studied. Increased seed production may be important for the long-term persistence of these arable weeds and may benefit invertebrates, small mammals and seed-eating birds. In three-quarters of FSE maize fields, growers used atrazine on the conventionally managed half, reflecting contemporary commercial practice. Withdrawal of the triazine herbicides atrazine, simazine and cyanazine from approved lists of EU chemicals could therefore reduce or even reverse the reported benefits of GMHT maize. Here we analyse effects of applications of triazine herbicides in conventional maize regimes on key indicators, using FSE data. Weed abundances were decreased greatly relative to all other regimes whenever atrazine was applied before weeds emerged. Here, we forecast weed abundances in post-triazine herbicide regimes. We predict weed abundances under future conventional herbicide management to be considerably larger than that for atrazine used before weeds emerged, but still smaller than for the four FSE sites analysed that used only non-triazine herbicides. Our overall conclusion is that the comparative benefits for arable biodiversity of GMHT maize cropping would be reduced, but not eliminated, by the withdrawal of triazines from conventional maize cropping. The importance of wood ants (Formica rufa) in determining the community structure (defined as the relative abundance of component species) and small-scale distribution of carabids was examined in a mature Scots pine stand in the New Forest, southern England. Carabids and wood ants were sampled by pitfall trapping throughout the forest stand from March to September 1998. The abundance of individual carabid species were modelled using vegetation type (grass or bracken), litter depth and wood ant density as independent explanatory variables. Models were fitted using a maximum-likelihood method (GLIM v.3.77; Baker 1985) with the assumption of a Poisson distribution, using a log-link function. Areas of high wood ant density were characterised by low abundance and species richness of carabids and high percentage dominance by the most commonly sampled species, Abax parallelepipedus. The extent and type of vegetation cover was found to influence the distribution and abundance of certain carabid species but only in areas where the density of wood ants was low. Large-bodied species occurred more frequently in bracken-dominated patches where the litter layer was deeper and the density of potential prey items was higher. The localisation of maize ( Zea mays L.) auxin-binding protein (ABP1) has been studied using a variety of techniques. At the whole-tissue level, tissue printing indicated that ABP1 is expressed to similar levels in all cells of the maize coleoptile and in the enclosed leaf roll. Within cells, the signals from immunofluorescence and immunogold labelling of ultrathin sections both indicated that ABP1 is confined to the endoplasmic reticulum (ER), none being detected in either Golgi apparatus or cell wall. This distribution is consistent with targeting motifs in its sequence. These observations are discussed with reference to the various reports which place a population of ABP1 on the outer face of the plasma membrane, including those suggesting that it is necessary on the cell surface for rapid, auxin-mediated protoplast hyperpolarisation. We have tested one proposed model to account for release of ABP1 from the ER, namely that auxin binding induces a conformational change in ABP1 leading to concealment of the KDEL retention motif. Using double-label immunofluorescence the characteristic auxin-induced rise in Golgi-apparatus signal was found, yet no change in the distribution of the ABP1 signal was detected. Maize suspension cultures were used to assay for auxin-promoted secretion of ABP1 into the medium, but secretion was below the limit of detection. This can be ascribed at least partly to the very active acidification of the medium by these cells and the instability of ABP1 in solution below pH 5.0. In the insect-baculovirus expression system, in which cell cultures maintain pH 6.2, a small amount of ABP1 secretion, less than 1% of the total, was detected under all conditions. Insect cells were shown to take up auxin and no inactivation of added auxin was detected, but auxin did not affect the level of ABP1 in the medium. Consequently, no evidence was found to support the model for auxin promotion of ABP1 secretion. Finally, quantitative glycan analysis was used to determine what proportion of ABP1 might reach the plasma membrane in maize coleoptile tissue. The results suggest that less than 15% of ABP1 ever escapes from the ER as far as the cis-Golgi and less than 2% passes further through the secretory pathway. Such leakage rates probably do not require a specialised mechanism allowing ABP1 past the KDEL retrieval pathway, but we are not able to rule out the possibility that some ABP1 is carried through associated with other proteins. The data are consistent with the presence of ABP1 both on the plasma membrane and in the ER. The relative sizes of the two pools explain the results obtained with immunofluorescence and immunogold labelling and illustrate the high efficiency of ER retention in plants. [ABSTRACT FROM AUTHOR]

Brefeldin A (BFA) induces a major aggregation of the Golgi apparatus (GA) in root cells followed by a complete, but reversible, vesiculation of the Golgi stacks which form two or more BFA compartments in the cytosol. This effect is monitored by the immunofluorescence of a Golgi antigen stained with a monoclonal antibody JIM 84, and by transmission electron microscopy. Depolymerisation of the microtubule cytoskeleton after oryzalin or colchicine treatment does not disturb the three dimensional organisation of the GA in control cells and does not affect any BFA induced Golgi stack movements. After disruption of the actin cytoskeleton with cytochalasin D many aggregates of Golgi stacks can be observed in root cells and these are sensitive to BFA treatment, forming multiple BFA compartments. N-ethyl-maleimide has no effect on the organisation of the GA in control roots but totally inhibits the action of BFA. Thus, it appears that spatial organisation of the GA and the BFA induced Golgi stack movements are actin dependent whilst BFA induced vesiculation of the GA is a structurally separate event. Acidic subcellular structures in the hydra- Chlorella symbiosis were identified by an immunocytochemical analysis of the distribution of the weak base, N-(3-[(2,4-dimitrophenyl)amino]propyl)-N-(3-aminopropyl)methylamine dihydrochloride (DAMP). DAMP accumulated in stenotele nematocysts and gland cell granules, and amorphous vacuoles in the epidermis, but not in the immediate environment of the symbiotic Chlorella, i.e., in the space between each Chlorella cell and the perisymbiont membrane. This paper reports on preliminary investigations into the structure of cell walls of varying complexity as revealed by the rapidfreeze deep-etch technique. Three cell types from different species were examined in order to compare the three-dimensional arrangement of random, polylamellate and helicoidal walls. Each cell type displayed a distinctive level of organisation with respect to the cellulose microfibrils and the matrix material. In polylamellated walls, the microfibrils within each layer were linked to each other by 16-20 nm long side chains regularly spaced along the length of the microfibril. In helicoidal walls, the shifting of the microfibrils could cleary be seen, yet no recognisable structures were observed which could mediate this movement. [ABSTRACT FROM AUTHOR]

The lower epidermis of Selaginella Helvetica leaves has numerous chloroplasts. In the diffuse light of the plant's normal habitat these are distributed over the inner wall of the cell, while in bright sunlight they move to the lateral walls. High voltage electron microscopy of whole critical-point dried cells shows that in the diffuse-light position the chloroplasts are connected by bundles of tightly-packed parallel filaments; these are distinct from, but seem to interconnect with, the filaments of the cytomatrix. In thin sections these appear as conventional microfilament bundles, while staining with rhodamineconjugated phalloidin implies that they are composed of actin. In bright light, when the chloroplasts have moved to the lateral walls, these microfilament bundles completely disappear, while filaments of the cytomatrix system remain attached to the chloroplasts. These results suggest that the function of the microfilament bundles may be to anchor the chloroplasts as much as to move them, and that the cytomatrix system may play a part in the movement; it is possible that actin microfilament bundles may actually dissociate into separate filaments within the cytomatrix. Changes in the abundance and form of endoplasmic reticulum in the three major cell types of the maize root cap were investigated by stereological and stereometric techniques. Quantification from thin sections was by the modification and application of standard morphometric procedures. This revealed dramatic increases in both the volume fraction and surface densities of endoplasmic reticulum as the meristem cells differentiate into starch and secretory cells. A stereometric technique for analysing thick sections was used to assess changes in the types of endoplasmic reticulum as cells differentiate through the root cap. This procedure showed that the proportions of cisternal endoplasmic reticulum to tubular endoplasmic reticulum was highest in the peripheral secretory cells. A technique for producing resin-free sections of plant roots for observation with the high voltage electron microscope is described. The resulting images have high contrast and reveal a three-dimensional filamentous network in the cytoplasm and nucleoplasm. The structure of plastids in the root cap of cress and maize was studied by low- and high-voltage electron microscopy after staining their membranes with a mixture of zinc iodide and osmium tetroxide. In plastids of both species electron-opaque membranes were found in the plastid interior while membranes of lesser electron-opacity comprised the outer envelope and vesicles and cisternae underlying it. Electron-opaque tubules, often in groups attached to the inner membrane of the amyloplast envelope, were found in cress but not in maize. The internal, less-opaque membranes were often found associated with the starch grains. No specific association could be seen between amyloplasts and endoplasmic reticulum (ER); their surfaces showed no regular contact or connexion, though the amyloplasts clearly indented the underlying ER. The ultrastructure and distribution of the Golgi apparatus in developing wheat endosperm was investigated using a zinc iodide-osmium tetroxide staining complex in conjunction with low and high voltage electron microscopy. Dictyosomes were numerous in starchy endosperm and aleurone at 15 days after anthesis, and during the period of rapid storage protein deposition 25 d after anthesis. Fewer dictyosomes were seen in maturing endosperm. Two types of vesicles were associated with the dictyosomes; small, heavily-stained vesicles were sited at the ends of fine tubules which extend from the cisternae, and larger less-stained vesicles were associated with the periphery of the cisternae. Stereo-pairs of micrographs up to 1 μm thick were taken to demonstrate the interconnections between cisternal and tubular endoplasmic reticulum. Elements of tubular ER were closely associated with dictyosomes, but connections were not observed. These results are discussed in relation to the transport of endosperm storage proteins from their site of synthesis on the cisternal ER to their site of storage, the protein bodies. The structure and distribution of cytoplasmic membranes during mitosis and cytokinesis in maize root tip meristematic cells was investigated by low and high voltage electron microscopy. The electron opacity of the nuclear envelope and endoplasmic reticulum (ER) was enhanced by staining the tissue in a mixture of zinc iodide and osmium tetroxide. Thin sections show the nuclear envelope to disassemble at prophase and become indistinguishable from the surrounding ER and polar aggregations of ER. In thick sections under the high voltage electron microscope the spindle is seen to be surrounded by a mass of tubular (TER) and cisternal (CER) endoplasmic reticulum derived from both the nuclear envelope and ER, which persists through metaphase and anaphase. At anaphase strands of TER traverse the spindle between the arms of the chromosomes. The octagonal nuclear pore complexes disappear by metaphase, but irregular-shaped pores persist in the membranes during mitosis. It is suggested that these form a template for pore-complex reformation during telophase. Phragmoplast formation is preceded by an aggregation of TER across the spindle at anaphase. Evidence is presented to suggest that the formation of the desmotubule of a plasmodesma is by the squeezing of a strand of endoplasmic reticulum between the vesicles of the cell plate. Coated vesicles isolated from carrot suspension culture cells were immune-blotted against four antibodies to porcine brain clathrin. Positive cross-reaction was obtained with three antibodies.
