Background: Acute Lymphoblastic Leukemia (ALL) is a neoplasm of the hematopoietic system characterized by a clonal expansion of abnormal lymphocyte precursor cells. ALL is the most common form of cancer in children, but despite advances in treatment, it can still be fatal. Ethnic differences influence survival rates, and genomic ancestry plays an important role, especially in mixed-race populations such as Latin America. This study aims to analyze the influence of genomic ancestry on toxicity in children with ALL in the Amazon region. Methods: The study included 171 patients (protocol number 119,649/2012—Ethics Committee) with ALL treated at a pediatric treatment center in Belém do Pará, in the Brazilian Amazon. The patients were submitted to the BFM protocol of induction therapy for ALL. Toxicity was assessed based on laboratory tests and adverse events, classified according to the CTC-NCI guide. Genomic ancestry was determined using autosomal informative markers. Results: The majority of children (94.74%) developed some type of toxicity during treatment, 87.04% of which were severe. Infectious toxicity was the most common, present in 84.8% of cases, 77.24% of which were severe. Amerindian ancestry showed an association with the risk of severe general toxicity and severe infectious toxicity, with a contribution of 35.0% demonstrating a significant increase in risk. In addition, post-induction refractoriness and relapse were also associated with an increased risk of death. Conclusion: This study highlights the influence of Amerindian genomic ancestry on response to therapy and toxicity in children with ALL in the Amazon region. The Japanese beetle, Popillia japonica Newman (Coleoptera: Scarabaeidae), is a major pest of turfgrasses, pastures and hay meadows in the invaded areas of North America and Europe. This study evaluated the efficacy of two biocontrol agents (the fungus Metarhizium anisopliae (Metschn.) Sorokin and the nematode Heterorhabditis bacteriophora Poinar) and the insecticide chlorantraniliprole against P. japonica larvae using a soil seeder modified for soil injection with minimum grass disturbance (“Eco Defender 25”). In 2021, trials were carried out at two hay meadows in the Lombardy region of Italy, using water and undisturbed plots as controls. Both biological agents and the insecticide reduced larval densities at one or the other site, but not at both sites, compared to the control. In the following spring, drone surveys conducted over the hay meadows did not show differences in grass quality between treatments, including the undisturbed plots. Recommendations and guidelines for management of SARS-COV-2 infection in hematologic patients were developed in the very difficult context of dealing with novel viral variants from one pandemic wave to another, with different susceptibility to available drugs and vaccines. Moreover, the largest SARS-COV-2 case series in patients treated for hematologic malignancies, including stem cell transplant recipients, was published before the Omicron surge, and refers mainly to Alpha and Delta viral variants. These infections had very high mortality, in a period when antivirals and monoclonal antibodies were mostly unavailable. Here, we report for the first time a SARS-COV-2 Omicron variant outbreak inside a Bone Marrow Transplant (BMT) Unit, describing the characteristics, clinical course, and infection outcomes shortly before and shortly after myeloablative transplantation. We detail how infections were treated off-label and managed inside the BMT ward, to guarantee the best possible outcomes while avoiding risks for non-infected inpatients. The positive outcomes observed suggest that it may not be absolutely necessary to obtain SARS-CoV-2 PCR negativity before BMT in hematologic patients after treated infection, in cases with long-term PCR positivity and high-risk hematologic disease. The fall armyworm, Spodoptera frugiperda (J.E. Smith), is an invasive pest threatening crop production and food security worldwide. High concerns are linked to the potential establishment of the species in Europe. The high migratory capacity of S. frugiperda causes concerns about the potential impacts of transient populations invading new areas from suitable hotspots. In the present work, we developed and used a physiologically-based demographic model to quantitatively assess the risks of S. frugiperda in Europe. The risks were assessed considering a best-, a median-, and a worst-case scenario. The Mediterranean coastal areas of Southern Europe resulted particularly suitable for the establishment of the species, with suitable areas reaching even higher latitudes, in the worst-case scenario. In Europe, up to four generations per year were predicted. The predicted yearly average number of moths per trap per week (± standard deviation) was 5 (± 4), 17 (± 5), and 139 (± 22) in the best, median-, and worst-case assessment scenarios, respectively. Model results showed that Southern and Central Europe up to the 48th parallel north might be exposed to the risk of transient populations. Depending on the latitude and on the period of arrival of the propagule, 1–2 transient generations per year might be expected. The model can be used to define strategies for reducing the risks of establishment of the pest at the country level. Acute myeloid leukemia may be characterized by a fraction of leukemia stem cells (LSCs) that sustain disease propagation eventually leading to relapse. Yet, the contribution of LSCs to early therapy resistance and AML regeneration remains controversial. We prospectively identify LSCs in AML patients and xenografts by single-cell RNA sequencing coupled with functional validation by a microRNA-126 reporter enriching for LSCs. Through nucleophosmin 1 (NPM1) mutation calling or chromosomal monosomy detection in single-cell transcriptomes, we discriminate LSCs from regenerating hematopoiesis, and assess their longitudinal response to chemotherapy. Chemotherapy induced a generalized inflammatory and senescence-associated response. Moreover, we observe heterogeneity within progenitor AML cells, some of which proliferate and differentiate with expression of oxidative-phosphorylation (OxPhos) signatures, while others are OxPhos (low) miR-126 (high) and display enforced stemness and quiescence features. miR-126 (high) LSCs are enriched at diagnosis in chemotherapy-refractory AML and at relapse, and their transcriptional signature robustly stratifies patients for survival in large AML cohorts. Relapse within acute myeloid leukaemia may be driven by the presence of leukaemia stem cells. The pilocarpine-induced (PILO) model has helped elucidate the electrophysiological and molecular aspects related to mesial temporal lobe epilepsy. It has been suggested that the extensive cell death and edema observed in the brains of these animals could be induced by increased inflammatory responses, such as the rapid release of the inflammatory cytokine interleukin 1 beta (Il1b). In this study, we investigate the role of endogenous Il1b in the acute phase of the PILO model. Our aim is twofold. First, we want to determine whether it is feasible to silence Il1b in the central nervous system using a non-invasive procedure. Second, we aim to investigate the effect of silencing endogenous Il1b and its antagonist, Il1rn.We used RNA interference applied non-invasively to knockdown Il1b and its endogenous antagonist Il1rn. We found that knocking down Il1b prior to pilocarpine injection increased the mortality rate of treated animals. Furthermore, we observed that, when exposing the animals to more Il1b by silencing its endogenous antagonist Il1rn, there was a better response to status epilepticus with decreased animal mortality in the acute phase of the PILO model. Thus, we show the feasibility of using a novel, less invasive approach to study genes involved in the inflammatory response in the central nervous system. Furthermore, our results provide suggestive evidence that modulating endogenous Il1b improves animal survival in the acute phase of the PILO model and may have effects that extend into the chronic phase. Multiferroics, showing the coexistence of two or more ferroic orderings at room temperature, could harness a revolution in multifunctional devices. However, most of the multiferroic compounds known to date are not magnetically and electrically ordered at ambient conditions, so the discovery of new materials is pivotal to allow the development of the field. In this work, we show that BaFe2O4 is a previously unrecognized room temperature multiferroic. X-ray and neutron diffraction allowed to reveal the polar crystal structure of the compound as well as its antiferromagnetic behavior, confirmed by bulk magnetometry characterizations. Piezo force microscopy and electrical measurements show the polarization to be switchable by the application of an external field, while symmetry analysis and calculations based on density functional theory reveal the improper nature of the ferroelectric component. Considering the present findings, we propose BaFe2O4 as a Bi- and Pb-free model for the search of new advanced multiferroic materials. The application of multiferroics is often limited by low ordering temperatures. CuSbSe2 is a promising absorber nano-material for thin-film solar cells, for its attractive photovoltaic properties, low cost, and environmentally friendly constituent elements. In this work, the interest was focused on the investigation of the effect of three types of substrates (FTO, ITO, and Mo) and deposition time on the characteristics of the electrodeposited films and on the efficiency of the CuSbSe2-based PV cells. The adopted architecture for this is the typical of substrate/CuSbSe2/CdS/i-ZnO/Al-ZnO. XRD reveals that CuSbSe2 thin films deposited on different substrates are grown in orthorhombic CuSbSe2 chalcostibite form (JCPDS: 98-041-8754), all with 65 nm crystallite size. However, the deposition time affects their preferential directions. SEM-EDX analysis shows that the sample deposited on ITO (45 min) has an almost stoichiometric composition with the lowest impurity content. Despite this, the FTO-based solar cell (60 min) shows the best efficiency (0.51%), compared to the ITO-based cell (0.12%), due to higher Jsc values. [ABSTRACT FROM AUTHOR]

We developed a mechanistic, stage-structured model simulating the phenology of Popillia japonica. The model simulates the influence of soil temperature on the larval diapause termination and on the development rate function of post-overwintering larvae and pupae. Model parameters are estimated based on literature evidence for pupae development and on a parameterisation process that allows estimating parameters for larval diapause termination and for the development rate function (and the related uncertainty) of post-overwintering larvae. Data used for model parameterisation and validation refer to time-series adult trap catches collected during the P. japonica monitoring programme performed by the Phytosanitary Service of Lombardy Region within the infested area in Lombardy (Italy) from 2015 to 2019. A total of 12 randomly selected locations are used to estimate biologically realistic model parameters (parameterisation dataset). We applied a Jackknife nonparametric resampling procedure on the parameterisation dataset to quantify uncertainty associated with parameters' estimates. Parameterised model is then validated on time-series adult trap catches data referring to a different set of 12 randomly selected locations (validation dataset) surveyed in Lombardy. The model successfully predicted the beginning of adult emergence and the overall curve of adult emergence in the validation dataset. The model presented can support the definition of the best timing for the implementation of monitoring and control activities for the local and the area-wide management of P. japonica. The Japanese beetle Popillia japonica is an invasive alien species recently introduced and established in Northern Italy. Adult beetles are very polyphagous and feed on vines, fruit trees, forest trees, crops, vegetables, ornamental and wild plant species. Eggs are usually laid by females in moist grassland in the summer, singly or in small clusters. Larvae feed on roots and may be transported in soil of plants for planting grown in containers. Restrictions on movement of plants grown in containers from infested to non-infested areas imposed by phytosanitary regulations have a significant economic impact on the nursery industry. An innovative approach was used to exclude beetle oviposition by weed mulching available for container-grown nursery stocks, and by testing larval survival to the application of chemical (cypermethrin) and organic (Heterorhabditis bacteriophora and Metarhizium brunneum) commercial pesticides registered for European nurseries. The high effectiveness of the method makes it a suitable component of a systems approach strategy for pest risk management, in order to achieve a safe production and trade of nursery plant material in areas infested by the Japanese beetle. Understanding how climate change might influence the distribution and abundance of crop pests is fundamental for the development and the implementation of pest management strategies. Here we present and apply a modelling framework assessing the non-linear physiological responses of the life-history strategies of the Mediterranean fruit fly (Ceratitis capitata, Wiedemann) to temperature. The model is used to explore how climate change might influence the distribution and abundance of this pest in Europe. We estimated the change in the distribution, abundance and activity of this species under current (year 2020) and future (years 2030 and 2050) climatic scenarios. The effects of climate change on the distribution, abundance and activity of C. capitata are heterogeneous both in time and in space. A northward expansion of the species, an increase in the altitudinal limit marking the presence of the species, and an overall increase in population abundance is expected in areas that might become more suitable under a changing climate. On the contrary, stable or reduced population abundances can be expected in areas where climate change leads to equally suitable or less suitable conditions. This heterogeneity reflects the contribution of both spatial variability in the predicted climatic patterns and non-linearity in the responses of the species' life-history strategies to temperature. During 2020, a total of 64 wild boar carcasses were tested for Enterobacteriaceae count (EBC), Salmonella and Yersinia enterocolitica in the abdominal region (i) within 5 h after hunting in the game collection point and (ii) before dressing and processing in the game-handling establishment (GHE) (49 carcasses—average time interval between (i) and (ii): 4.3 days). Because of COVID-19 restrictions, 15 carcasses were transported to a near slaughterhouse (average time interval between (i) and (ii): 2.3 days). Mesenteric lymph nodes (MLNs) were collected and tested for Salmonella and Y. enterocolitica. Results are shown in relation to sampling A (49 carcasses—GHE) and sampling B (15 carcasses—slaughterhouse). Sampling A: EBC median values were (i) 2.51 log10 CFU/cm2 and (ii) 2.79 log10 CFU/cm2. EBC increase between (i) and (ii) was statistically significant (p = 0.001). Salmonella prevalence on carcasses varied from (i) 2.0 to (ii) 6.1%. Sampling B: EBC median values were (i) 3.1 log10 CFU/cm2 and (ii) 3.32 log10 CFU/cm2. EBC increase between (i) and (ii) was not statistically significant (p = 0.191). Salmonella prevalence on carcasses varied from (i) 6.7 to (ii) 0.0%. The prevalence (sampling A + B) of lymphatic Salmonella carriers was 7.8% (5/64). From carcasses and/or MNLs, the serovars Enteritidis, Typhimurium, Agama, Zaiman and Diarizonae O:50 (z) were detected. Y. enterocolitica was never isolated. Long chilling periods prior to wild game processing should be avoided, and carcasses should be tested at GHE rather than after shooting to proper reflect the microbial load of wild boar meat entering the food chain. Activating mutations in the BRAF-MAPK pathway have been reported in histiocytoses, hematological inflammatory neoplasms characterized by multi-organ dissemination of pro-inflammatory myeloid cells. Here, we generate a humanized mouse model of transplantation of human hematopoietic stem and progenitor cells (HSPCs) expressing the activated form of BRAF (BRAFV600E). All mice transplanted with BRAFV600E-expressing HSPCs succumb to bone marrow failure, displaying myeloid-restricted hematopoiesis and multi-organ dissemination of aberrant mononuclear phagocytes. At the basis of this aggressive phenotype, we uncover the engagement of a senescence program, characterized by DNA damage response activation and a senescence-associated secretory phenotype, which affects also non-mutated bystander cells. Mechanistically, we identify TNFα as a key determinant of paracrine senescence and myeloid-restricted hematopoiesis and show that its inhibition dampens inflammation, delays disease onset and rescues hematopoietic defects in bystander cells. Our work establishes that senescence in the human hematopoietic system links oncogene-activation to the systemic inflammation observed in histiocytic neoplasms. BRAF-MAPK activating mutations are reported in histiocytoses—hematological neoplasms with widespread pro-inflammatory myeloid cells. We developed a reaction–diffusion model to describe the spatio-temporal dynamics of the Japanese beetle (Popillia japonica Newman), based on adult abundance data collected by the Regional Phytosanitary Service during the monitoring activities in the infested area in Lombardy (northern Italy), from 2015 to 2021. The model simulated the abundance of the pest, with a discrete time step of one year along linear trajectories departing from an initial point of establishment. The model allowed the determination of the rate of expansion (i.e. the speed at which the leading edge of a population wave moves over time) of the pest along 13 different trajectories, ranging from 4.5 to 13.8 km/y, with a mean value of 8.2 km/y. Finally, we developed a land suitability index that summarises the effect of land use on the trajectory-specific rate of expansion of P. japonica. Specifically, the model revealed an increase in the rate of expansion of 260 m per year for each additional percentage point in the land suitability index. The model presented and the knowledge acquired in this work represent an important step forward in the comprehension of P. japonica population dynamics, and they represent important elements for the development of a decision support tool for pest risk managers to design and implement scientifically driven management actions. Background: The implementation of hybrid Bt cotton unique to India has been heralded as a grand success by government agencies, seed companies and other proponents, and yet yields have stagnated at low levels and production costs have risen 2.5–3-fold. The low-yield hybrid cotton system of India contributes thousands of farmer suicides to the annual national toll. Conceptual and methodological barriers have hindered bioeconomic analysis of the ecological and social sustainability of such cross-scale agro-ecological problems in time and geographic space, under global technology and climate change. As a paradigm shift, we use conceptually simple, parameter-sparse, theoretically based, mechanistic, weather-driven physiologically based demographic models (PBDMs) to deconstruct the bio-economics of the Indian cotton system. Results: Our analysis of Indian hybrid cotton system explains some extant ecological and economic problems, and suggests a viable solution. Specifically, the model accurately captured the age-stage mass dynamics of rainfed and irrigated cotton growth/development and the interactions with the key pest pink bollworm across five south-central Indian states, and enabled identification of proximate bioeconomic factors responsible for low yield and their relationship to farmer suicides. The results are reinforced by analysis of Ministry of Agriculture annual state-level data. We explain why short-season, high-density non-GM cotton is a highly viable solution for Indian cotton farmers in rainfed and irrigated cotton areas of the five states, and possibly nationally. The transition from a theoretical bioeconomic construct to a real-world regional bioeconomic analysis proved seamless. Conclusions: The hybrid long-season Bt technology for rainfed and irrigated cotton is unique to India, and is a value capture mechanism. This technology is suboptimal leading to stagnant yields, high input costs, increased insecticide use, and low farmer incomes that increase economic distress that is a proximate cause of cotton farmer suicides. The current GM Bt technology adds costs in rainfed cotton without commensurate increases in yield. Non-GM pure-line high-density short-season varieties could double rainfed cotton yield, reduce costs, decrease insecticide use, and help ameliorate suicides. Yersiniosis was the fourth reported zoonosis in the European Union in 2018. As well-known, pigs are recognized important reservoirs of Yersinia enterocolitica. The study was focused on Y. enterocolitica detection in mesenteric lymph nodes and faeces of 305 wild boars, but Yersinia bercovieri was more common, being isolated from 108 animals (35.4%). Cold season (p = 1.17 × 10–5) and young age (p = 0.004) significantly increased Y. bercovieri detection. Y. enterocolitica 1A belonging to six serotypes (O:4.32–4.33; O:5; O:6.30–6.31; O:7.8–8–8.19; O:10–34; O:12.25–12.26) was isolated from 8.2% (25/305) of the animals. Spittlebugs (Hemiptera: Aphrophoridae) are the dominant xylem-sap feeders in the Mediterranean area and the only proven vectors of Xylella fastidiosa ST53, the causal agent of the olive dieback epidemic in Apulia, Italy. We have investigated the structured population phenology, abundance and seasonal movement between crops and wild plant species of both the nymphal and adult stages of different spittlebug species in olive groves. Field surveys were conducted during the 2016–2018 period in four olive orchards located in coastal and inland areas in the Apulia and Liguria regions in Italy. The nymphal population in the herbaceous cover was estimated using quadrat samplings. Adults were collected through sweep nets on three different vegetational components: herbaceous cover, olive canopy and wild woody plants. Philaenus spumarius was the most abundant species; its nymphs were collected from early March and reached a peak around mid-April, when the 4th instar was prevalent. Spittlebug adults were collected from late April until late autumn. P. spumarius adults were abundant on the herbaceous cover and olive trees in late spring, and they then dispersed to wild woody hosts during the summer and returned to the olive groves in autumn when searching for oviposition sites in the herbaceous cover. A relatively high abundance of P. spumarius was observed on olive trees during summer in the Liguria Region. The present work provides a large amount of data on the life cycle of spittlebugs within an olive agroecosystem that can be used to design effective control programmes against these vectors in infected areas and to assess the risk of the establishment and spread of X. fastidiosa to Xylella-free areas. The study assessed Salmonella carriage in wild boars (Sus scrofa) and compared their isolates with those recovered from the domestic swine population of the same area of northern Italy (Emilia-Romagna), characterized by intensive pig farming and rather high density of wild boars. A total of 189 wild boars hunted during twelve months (2017–2018) were tested for Salmonella in mesenteric lymph nodes (MLN) and faecal samples. Antimicrobial resistance of recovered strains was tested against 14 antimicrobials. Salmonella was detected in 33/189 wild boars (17.5%), specifically from 30/189 MLN (15.9%) and 6/189 faecal samples (3.2%). Three animals were positive in both samples. Thirteen Salmonella serovars were identified, i.e. Typhimurium (the most common), Bovismorbificans, Coeln, Derby, Enteritidis, Gaminara, Hessarek, Houtenae IV, Kottbus, Napoli, Stanleyville, Thompson and Veneziana. Salmonella carriage was higher in warm than in cold months (P = 0.0013). Pregnancy status was never associated with Salmonella carriage, with significant difference in the recovery of the pathogen between non-pregnant and pregnant females (P = 0.003). Only one resistance pattern to streptomycin and tetracycline was found in 15 isolates (41.7%) belonging to Typhimurium (14/14; 100%) and Kottbus (1/3; 33.3%) serovars. Overlap with isolates from farmed pigs was limited at serotype level (Typhimurium, Derby, Enteritis, Bovismorbificans, Kottbus) and absent at PFGE level, and also antimicrobial resistance patterns were substantially different. This evidence indicates a substantial segregation of the two animal populations with regard to infectious contacts, possibly suggesting that biosecurity measures in place at industrial farm level limit the exchange of Salmonella. The production of pasture in Ethiopia was simulated by means of a dynamic model. Most of the country is characterized by a tropical monsoon climate with mild temperatures and precipitation mainly concentrated in the June-September period (main rainy season). The production model is driven by solar radiation and takes into account limitations due to relocation, maintenance respiration, conversion to final dry matter, temperature, water stress, and nutrients availability. The model also considers the senescence of grassland which strongly limits the nutritional value of grasses for livestock. The simulation for the 1982-2009 period, performed on gridded daily time series of rainfall and maximum and minimum temperature with a resolution of 0.5°, provided results comparable with values reported in literature. Yearly mean yield in Ethiopia ranged between 1.8 metric ton per hectare (t ha-1) (2002) and 2.6 t ha−1 (1989) of dry matter with values above 2.5 t ha-1 attained in 1983, 1985, 1989, and 2008. The Ethiopian territory has been subdivided in 1494 cells and a frequency distribution of the per-cell yearly mean pasture production has been obtained. This distribution ranges from 0 to 7 t ha-1 and it shows a right skewed distribution and a modal class between 1.5-2 t ha-1. Simulation carried out on long time series for this peculiar tropical environment give rise to as lot of results relevant by the agroecological point of view on space variability of pasture production, main limiting factors (solar radiation, precipitation, temperature), and relevant meteo-climatic cycles affecting pasture production (seasonal and inter yearly variability, ENSO). In order to set priorities in management of costly and ecosystem-damaging species, policymakers and managers need accurate predictions not only about where a specific invader may establish but also about its potential abundance at different geographical scales. This is because density or biomass per unit area of an invasive species is a key predictor of the magnitude of environmental and economic impact in the invaded habitat. Here, we present a physiologically based demographic model describing and explaining the population dynamics of a widespread freshwater invader, the golden apple snail Pomacea canaliculata, which is causing severe environmental and economic impacts in invaded wetlands and rice fields in Southeastern Asia and has also been introduced to North America and Europe . The model is based on bio-demographic functions for mortality, development and fecundity rates that are driven by water temperature for the aquatic stages (juveniles and adults) and by air temperature for the aerial egg masses. Our model has been validated against data on the current distribution in South America and Japan, and produced consistent and realistic patterns of reproduction, growth, maturation and mortality under different scenarios in accordance to what is known from real P. canaliculata populations in different regions and climates. The model further shows that P. canaliculata will use two different reproductive strategies (semelparity and iteroparity) within the potential area of establishment, a plasticity that may explain the high invasiveness of this species across a wide range of habitats with different climates. Our results also suggest that densities, and thus the magnitude of environmental and agricultural damage, will be largely different in locations with distinct climatic regimes within the potential area of establishment. Control interventions in sustainable pest management schemes are set according to the phenology and the population abundance of the pests. This information can be obtained using suitable mathematical models that describe the population dynamics based on individual life history responses to environmental conditions and resource availability. These responses are described by development, fecundity and survival rate functions, which can be estimated from laboratory experiments. If experimental data are not available, data on field population dynamics can be used for their estimation. This is the case of the extrinsic mortality term that appears in the mortality rate function due to biotic factors. We propose a Bayesian approach to estimate the probability density functions of the parameters in the extrinsic mortality rate function, starting from data on population abundance. The method investigates the time variability in the mortality parameters by comparing simulated and observed trajectories. The grape berry moth, a pest of great importance in European vineyards, has been considered as a case study. Simulated data have been considered to evaluate the convergence of the algorithm, while field data have been used to obtain estimates of the mortality for the grape berry moth. Multimodal human brain mapping has been proposed as an integrated approach capable of improving the recognition of the cortical correlates of specific neurological functions. We used simultaneous EEG-fMRI (functional magnetic resonance imaging) and EEG-TD-fNIRS (time domain functional near-infrared spectroscopy) recordings to compare different hemodynamic methods with changes in EEG in ten patients with progressive myoclonic epilepsy and 12 healthy controls. We evaluated OHb, HHb and Blood oxygen level-dependent (BOLD) changes and event-related desynchronization/synchronization (ERD/ERS) in the α and β bands of all of the subjects while they performed a simple motor task. The general linear model was used to obtain comparable fMRI and TD-fNIRS activation maps. We also analyzed cortical thickness in order to evaluate any structural changes. In the patients, the TD-NIRS and fMRI data significantly correlated and showed a significant lessening of the increase in OHb and the decrease in BOLD. The post-movement β rebound was minimal or absent in patients. Cortical thickness was moderately reduced in the motor area of the patients and correlated with the reduction in the hemodynamic signals. The fMRI and TD-NIRS results were consistent, significantly correlated and showed smaller hemodynamic changes in the patients. This finding may be partially attributable to mild cortical thickening. However, cortical hyperexcitability, which is known to generate myoclonic jerks and probably accounts for the lack of EEG β-ERS, did not reflect any increased energy requirement. Electrophysiological studies indicate that Unverricht-Lundborg's disease (ULD), the most common form of progressive myoclonus epilepsy in Europe, is characterized by the involvement of multiple cortical regions in degenerative changes that lead to enhanced excitation and deficient inhibition. We searched for the haemodynamic correlates of these effects using functional MRI (fMRI) of self-paced index extensions, a well-accepted task highlighting significant differences. EEG and fMRI were simultaneously acquired in 11 ULD patients and 16 controls, performing the index extensions individually (event-related task) as well as repetitively (block task). ERD/ERS analysis was performed for the EEG data in the alpha and beta bands. fMRI time-series were analyzed using the traditional general linear model, as well as with an assumption-free approach, and by means of cross-region correlations representing functional connectivity. In line with the existing literature, ULD patients had enhanced desynchronization in the alpha band and reduced post-movement synchronization in the beta band. By contrast, fMRI did not reveal any difference between the two groups; there were no activation intensity, latency or extent effects, no significant engagement of additional regions, and no changes to functional connectivity. We conclude that, so long as the patients are executing a task which does not induce obvious action myoclonus, the hypothesized abnormalities in pyramidal neuron and interneuron dynamics are relatively subtle, embodied in processes which are not metabolically-demanding and take place at a time-scale invisible to fMRI. One of the putative causative genes for juvenile myoclonic epilepsy (JME) is EFHC1. We report here the expression profile and distribution of Efhc1 messenger RNA (mRNA) during mouse and rat brain development. Real-time polymerase chain reaction revealed that there is no difference in the expression of Efhc1 mRNA between right and left hemispheres in both species. In addition, the highest levels of Efhc1 mRNA were found at intra-uterine stages in mouse and in adulthood in rat. In common, there was a progressive decrease in Efhc1 expression from 1-day-old neonates to 14-day-old animals in both species. In situ hybridization studies showed that rat and mouse Efhc1 mRNAs are expressed in ependymal cells of ventricle walls. Our findings suggest that Efhc1 expression is more important during initial phases of brain development and that at this stage it could be involved in key developmental mechanisms underlying JME. Mutations in LGI1 were described in patients with autosomal dominant partial epilepsy with auditory features (ADPEAF), and recent clinical findings have implicated LGI1 in human brain development. However, the precise role of LGI1 in epileptogenesis remains largely unknown. The objective of this study was to determine the expression pattern of Lgi1 in mice brain during development and in adult animals. Real-time polymerase chain reaction (PCR) quantification and Western blot experiments showed a relative low expression during intrauterine stages, increasing until adulthood. In addition, we did not find significant differences between left and right hemispheres. The hippocampus presented higher levels of Lgi1 expression when compared to the neocortex and the cerebellum of adult animals; however, these results did not reach statistical significance. This study was the first to determine a specific profile of Lgi1 gene expression during central nervous system development, which suggests a possible inhibitory function in latter stages of development. A conceptual framework is developed and used for improving the livelihood of Sub-Saharan communities faced with multiple stresses resulting from adverse environments, vector-transmitted diseases, and limited food. Ecosocial systems are the units for management. The accumulation of ecological, economic, and social capital is the objective of management, the reduction of maintenance costs is the key strategy, and technologies must satisfy ecological, economic, and social criteria. Interacting social system components are communities, facilitators, scientists, and political as well as administrative organizations. An adaptive approach to management allows flexibility in human and financial resource allocation and results in changes in knowledge and efficacy of management operations. The implementation strategy at two sites in Ethiopia relies on a two-stage procedure. In Stage 1, human and cattle health management set the conditions for development. In Stage 2, the ecosocial system is kept on a trajectory to enhanced ecological, economic, and social capital. In each stage, the assignment of responsibilities leads to four implementation phases. In Stage 1, successful precision targeting of tsetse and drug-based Trypanosoma spp. control increased the economic and social capital at the Luke site in southwestern Ethiopia but threatened the ecological capital because of overstocking and land degradation. In Stage 2, a women’s group living in the outskirts of Addis Ababa (Ethiopia) transformed barren land into a horticultural enterprise to enhance ecosystem service provision, including production of horticultural goods, and increase soil fertility expressed by nutrients and water availability. The spatial distribution of motile life stages of the two-spotted spider mite Tetranychus urticae Koch in Kenyan small-scale tomato fields was described by Taylor's power law and an enumerative sampling plan was designed for research purposes. The exponential increase of T. urticae populations during three growing seasons permits the development and use of a simple exponential model for the design of a threshold-based chemical control system. For this purpose, a critical threshold of 440 motile mites per sample unit at the end of the growing season (12 weeks after transplanting) was translated into a proportion of 0.83 infested units in sample 3, i.e. five weeks after transplanting. Asequential binomial sampling plan with respect to the proportion of 0.83, five weeks after transplanting, was designed. The exponential model was extended to account for the influence of weather and host plant species. Model development and parameter estimation were based on three data sets (Kenyan tomato fields, Italian and Californian strawberry fields). The model satisfactorily predicted a positive influence on growth rates by (i) changing the host plant from tomato to strawberry, and (ii) temperature, while a negative effect resulted from (iii) rainfall; both (ii) and (iii) are controlled by temperature-rainfall interactions. In contrast, the estimated parameter values did not satisfactorily describe the expected responses at specific temperature and rainfall values. Nevertheless, the model allowed the rating of host plant species in the field. A fourth data set from Swiss apple orchards was used to test the model, and population build-up on the apple host plant appeared to be higher than on tomatoes but lower than on strawberry. Similarly to the half-doped manganese oxides with perovskite structure like La0.5Ca0.5MnO3, the double perovskite compound NaMn7O12 contains an equal number of Mn3+ and Mn4+ ions in the corner-sharing network of MnO6 octahedra and exhibits a CE order of the charge and spins of these ions at low temperature. Though, in NaMn7O12 the order is complete, the charge and spin ordering transitions are sharp and the system is free of disorder, phase coexistence or structural inhomogeneities thanks to the absence of chemical substitutions. Here we discuss two unusual features of the CE structure of NaMn7O12: (1) the e g 3 d x 2− y 2 orbital ordering expected from the direct crystallographic observation of compressed Mn3+O6 octahedra below the charge ordering transition; (2) the existence of a large amount of low-energy excitations evidenced by the low temperature behavior of the specific heat. We present a comparative Mössbauer study of the 151Eu motions in the nonsuperconducting and superconducting cuprates EuSr2Cu3O7−δ (ESCO) and EuBa2Cu3O7−δ (EBCO), respectively, with the aim of investigating atomic-motion anharmonicities in these materials. In this study, a comparison with the results of a similar analysis of powdered samples of EBCO having oriented grains is also performed. We show that the Eu ion in ESCO, as well as in the common EBCO cuprates, does not move in a parabolic potential, but in a potential with a flat bottom. Imatinib is the tyrosine kinase inhibitor used as the gold standard for the treatment of Chronic Myeloid Leukemia. However, about 30% of patients do not respond well to this therapy. Variants in drug administration, distribution, metabolism and excretion (ADME) genes play an important role in drug resistance especially in admixed populations. We investigated 129 patients diagnosed with Chronic Myeloid Leukemia treated with imatinib as first choice therapy. The participants of the study are highly admixed, populations that exhibit genetic diversity and complexity due to the contributions of multiple ancestral groups. Thus, the aim of this work was to investigate the association of 30 SNVs in genes related to response to treatment with Imatinibe in CML. Our results indicated that for the rs2290573 of the ULK3 gene, patients with the recessive AA genotype are three times more likely to develop resistance over time (secondary resistance) (p = 0.019, OR = 3.19, IC 95%= 1.21–8.36). Finally, we performed interaction analysis between the investigated variants and found several associations between SNVs and secondary resistance. We concluded that the variant rs2290573 of the ULK3 gene may be relevant for predicting treatment response of CML with imatinib, as well as possible treatment resistance. In this work we study from the mathematical and numerical point of view a problem arising in vector-borne plant diseases. The model is written as a nonlinear system composed of a parabolic partial differential equation for the vector abundance function and a first-order ordinary differential equation for the plant health function. An existence and uniqueness result is proved using backward finite differences, uniform estimates and passing to the limit. The regularity of the solution is also obtained. Then, using the finite element method and the implicit Euler scheme, fully discrete approximations are introduced. A discrete stability property and a main a priori error estimates result are proved using a discrete version of Gronwall’s lemma and some estimates on the different approaches. Finally, some numerical results, in one and two dimensions, are presented to demonstrate the accuracy of the approximation and the behaviour of the solution. [ABSTRACT FROM AUTHOR]
