For decades, the risk of exposure to infectious diseases in recreational beaches has been evaluated through the quantification of fecal indicator bacteria in water samples using culture methods. The analyses of sand samples have recently been developed as a complement to the monitoring of recreational waters in beach quality assessments. The growing use of molecular techniques for environmental monitoring allows for the rapid detection of pathogenic genes, thus providing more accurate information regarding the health risk of exposure to contaminated sand. The aim of this work was to determine the relationship between the fecal indicators abundance in water and sand and the presence of Shiga toxin-producer Escherichia coli (STEC) in sand by analyzing samples from touristic beaches using culture-dependent (fecal coliforms assay) and culture-independent (real-time PCR of stx1, stx2, and eae genes) techniques. We found a high concentration of coliform bacteria in water and sand in several beaches in eastern Uruguay, with different levels of sanitation networks and levels of urbanization. The presence of STEC virulence genes (mainly stx1) was confirmed in 8 out of 20 sand samples. The recreational use of sandy beaches may imply a risk to the health of its users, especially near streams and creek outflows, thus highlighting the need of monitoring sand bacteriological quality and pathogens using molecular tools. Polychaetes that inhabit the sediments of estuaries are important prey to many species all around the world. Laeonereis acuta is a deposit feeder living in estuaries along the Atlantic coast of South America. Ragworms accumulate metals from the sediment, and represent a means of entry of sediment contaminants into the trophic network. The concentrations of Zn, Pb, Cu, Cr and Cd were determined in polychaetes and sediments (total and extractable) from six estuarine beaches of Río de la Plata. The associations between total and extractable concentrations in sediment and accumulated concentrations in L. acuta were analyzed. Sediments extracted by weak acid digestion appear to be a good proxy model of bioavailable metal fractions in the sediment. The association between metals in sediment and polychaetes denotes the role of L. acuta as an important link in metal trophic transfer from sediments to potential fish and bird predators of the worm. Purpose: Tomato products are a dietary source of natural antioxidants, especially lycopene, which accumulates in the liver, where it exerts biological effects. Taking into consideration this fact, the aim of the present study was to ascertain the effect of tomato consumption on biomarkers and gene expression related to lipid metabolism in rats with induced steatosis. Methods: Adult male Sprague-Dawley rats (8 weeks old) were randomly grouped ( n = 6 rats/group) in four experimental groups: NA (normal diet and water), NL (normal diet and tomato juice), HA (high fat diet and water) and HL (high fat diet and tomato juice). After 7 weeks, rats were euthanized, and plasma, urine, feces and liver were sampled to analyze the biomarkers related to lipid metabolism, inflammation and oxidative stress. Results: The H diet induced steatosis (grade II) in the HA and HL groups, which was confirmed by the levels of alanine aminotransferase and aspartate aminotransferase, histological examination and the presence of dyslipidemia. The intake of tomato juice led to an accumulation of all- E and Z-lycopene and its metabolites in the livers of these animals; levels were higher in HL than in NL, apparently due to higher absorption (63.07 vs. 44.45 %). A significant improvement in the plasma level of high-density lipoprotein was observed in the HL group compared with HA animals, as was an alleviation of oxidative stress through reduction of isoprostanes in the urine. In relation to fatty acid gene expression, an overexpression of several genes related to fatty acid transport, lipid hydrolysis and mitochondrial and peroxisomal β-fatty acid oxidation was observed in the HL group. Conclusions: The consumption of tomato juice and tomato products reduced hallmarks of steatosis, plasmatic triglycerides and very low-density lipoproteins, and increased lipid metabolism by inducing an overexpression of genes involved in more efficient fatty acid oxidation. Four selected fresh consumption tomato varieties, harvested at different ripening stages (green, breaker, pink, red) and five industrial processing tomato varieties, harvested at red ripe stage, were analysed for lycopene and its isomers, β-carotene, total and individual phenolics, vitamin C and hydrophilic antioxidant activity. Tomato variety and ripening stage significantly affected carotenoids, total phenolic compounds and hydrophilic antioxidant activity when fresh consumption tomatoes were compared. The average of total lycopene content at the different ripening stages was 0.63, 12.20, 26.76 and 116.66 mg/kg of fresh weight (FW). Vitamin C ranged from 2.79 (breaker Cherry Pera) to 297.62 (red Cherry Pera) mg/kg FW. In green and breaker tomatoes, a positive correlation of hydrophilic antioxidant activity with vitamin C and chlorogenic acid was observed. However, in industrial tomato varieties (red ripe stage) and fresh consumption varieties harvested at pink and red stages, hydrophilic antioxidant activity was correlated with total phenolics and rutin. Tomato varieties for industrial processing did not show significant differences in total phenolic compounds and lycopene content. Lycopene ranged from 83.17 to 97.60 mg/kg FW, while total phenolic compounds varied between 257.91 and 284.13 mg/kg FW. Chlorogenic acid and rutin were the most abundant individual phenolics found in all the samples studied. Moreover, the content of total phenolics was always significantly correlated with the content of rutin. The amounts of all individual phenolics were affected by variety, with the exception of ferulic acid. Generally, these individual phenolics were more abundant in green and intermediate ripening stages, decreasing in full red tomatoes. [ABSTRACT FROM AUTHOR]

There is an increasing demand for easy and cost-effective methods to screen the toxicological impact of the growing number of chemical mixtures being generated by industry. Such a screening method has been developed using viable, genetically modified green fluorescent protein (GFP) reporter yeast that was magnetically functionalised and held within a microfluidic device. The GFP reporter yeast was used to detect genotoxicity by monitoring the exposure of the cells to a well-known genotoxic chemical (methyl methane sulfonate, MMS). The cells were magnetised using biocompatible positively charged PAH-stabilised magnetic nanoparticles with diameters around 15 nm. Gradient mixing was utilised to simultaneously expose yeast to a range of concentrations of toxins, and the effective fluorescence emitted from the produced GFP was measured. The magnetically enhanced retention of the yeast cells, with their facile subsequent removal and reloading, allowed for very convenient and rapid toxicity screening of a wide range of chemicals. This is the first report showing magnetic yeast within microfluidic devices in a simple bioassay, with potential applications to other types of fluorescent reporter yeast in toxicological and biomedical research. The microfluidic chip offers a simple and low-cost screening test that can be automated to allow multiple uses (adapted to different cell types) of the device on a wide range of chemicals and concentrations. The mean body size of limnetic cladocerans decreases from cold temperate to tropical regions, in both the northern and the southern hemisphere. This size shift has been attributed to both direct (e.g. physiological) or indirect (especially increased predation) impacts. To provide further information on the role of predation, we compiled results from several studies of subtropical Uruguayan lakes using three different approaches: (i) field observations from two lakes with contrasting fish abundance, Lakes Rivera and Rodó, (ii) fish exclusion experiments conducted in in-lake mesocosms in three lakes, and (iii) analyses of the Daphnia egg bank in the surface sediment of eighteen lakes. When fish predation pressure was low due to fish kills in Lake Rivera, large-bodied Daphnia appeared. In contrast, small-sized cladocerans were abundant in Lake Rodó, which exhibited a typical high abundance of fish. Likewise, relatively large cladocerans (e.g. Daphnia and Simocephalus) appeared in fishless mesocosms after only 2 weeks, most likely hatched from resting egg banks stored in the surface sediment, but their abundance declined again after fish stocking. Moreover, field studies showed that 9 out of 18 Uruguayan shallow lakes had resting eggs of Daphnia in their surface sediment despite that this genus was only recorded in three of the lakes in summer water samples, indicating that Daphnia might be able to build up populations at low risk of predation. Our results show that medium and large-sized zooplankton can occur in subtropical lakes when fish predation is removed. The evidence provided here collectively confirms the hypothesis that predation, rather than high-temperature induced physiological constraints, is the key factor determining the dominance of small-sized zooplankton in warm lakes.
