Circulating tumor cells (CTCs) survive in the bloodstream and then seed and invade to foster tumor metastasis. The arrest of cancer cells is favored by permissive flow forces and geometrical constraints. Through the use of high-throughput microfluidic devices designed to mimic capillary-sized vessels, we applied pressure differences to cancer cells (MCF-7 cell line) and recorded the cell traverse-vessel behaviors. Our results showed that cancer cells transform from a Newtonian droplet state to an adhesion/migration state when cancer cells traverse artificial vessels. To explain these phenomena, a modified Newtonian droplet model was also proposed. These phenomena and the modified model may reveal how CTCs in the blood seed and invade vessels under suitable conditions. [ABSTRACT FROM AUTHOR]

The causes and consequences of aging have always been a concern. In recent studies, changes in the stress response capacity of cells during aging were quantitatively analyzed. It was found that aging was accompanied by a decline in response capacity. When the response capacity decreased to a critical value, which we assumed was the internal noise level, the cell soon died. To survive, the response capacity should be, at minimum, sufficiently strong to resist intracellular noise. Here, we discuss the role of stress response capacity in aging and conjecture that lifespan might be extended by enhancing stress response capacity. [ABSTRACT FROM AUTHOR]

Density functional theory calculation is used to investigate the oxidation of cyclo-olefin (cyclobutene, cyclopentene, cyclohexene, cycloheptene, and cyclo-octene) by the complex [FeIV(O)(TQA)(NCMe)]2+, which has S = 2 ground state, and the effect of electronic factors and steric hindrance on reaction barriers. Our results suggest that the oxo–iron(IV) complex can oxidise C–H and C = C bonds via a single-state mechanism, and two different ways of electron transport exist. The energy barriers initially decrease with increasing substrate size, and the trend then reverses. Comparison of the energy barrier in different systems reveals that except for the reaction between [FeIV(O)(TQA)(NCMe)]2+ and cycloheptene, oxo–iron(IV) complexes prefer epoxidation to hydroxylation. However, the hydroxylated product is more stable than the corresponding epoxidated product. This result indicates that the products of epoxidation tend to decompose first. The energy barrier of hydroxylation and epoxidation originates from the balance of orbital interaction and Pauli repulsion from the equatorial ligand and protons on the approaching substrate. In this regard, we calculate the weak interaction between two fragments (oxo–iron complex and substrates) using the independent gradient model and drawn the corresponding 3D isosurface representations of reactants. The widespread use of caffeine in food and drug industries has caused great environmental pollution. Herein, an efficient caffeine-degrading strain Paraburkholderia caffeinilytica CF1 isolated from a tea garden in China can utilize caffeine as its sole carbon and nitrogen source. Combination of chromatographic and spectrophotometric techniques confirmed that strain CF1 adopts N-demethylation pathway for caffeine degradation. Whole genome sequencing of strain CF1 reveals that it has two chromosomes with sizes 3.62 Mb and 4.53 Mb, and a 174-kb mega-plasmid. The plasmid P1 specifically harbors the genes essential for caffeine metabolism. By analyzing the sequence alignment and quantitative real-time PCR data, the redundant gene cluster of caffeine degradation was elucidated. Genes related to catalyzing the N1-demethylation of caffeine to theobromine, the first step of caffeine degradation were heterologously expressed, and methylxanthine N1-demethylase was purified and characterized. Above all, this study systematically unravels the molecular mechanism of caffeine degradation by Paraburkholderia. L-arabinose isomerase (L-AI) (EC 5. 3. 1. 4. L-AI) that mediates the isomerization of D-galactose to D-tagatose was isolated from Lactobacillus brevis (MF 465792), and was further purified and characterized. Pure enzyme with molecular weight of 60.1 kDa was successfully obtained after the purification using Native-PAGE gel extraction method, which was a monomer in solution. The L-AI was found to be stable at 45-75 °C, and at pH 7.0-9.0. Its optimum temperature and pH was determined as 65 °C and 7.0, respectively. Besides, we found that Ca2+, Cu2+, and Ba2+ ions inhibited the enzyme activity, whereas the enzyme activity was significantly enhanced in the presence of Mg2+, Mn2+, or Co2+ ions. The optimum concentration of Mn2+ and Co2+ was determined to be 1 mM. Furthermore, we characterized the kinetic parameters for L-AI and determined the Km (129 mM) and the Vmax (0.045 mM min− 1) values. Notably, L. brevisL-AI exhibited a high bioconversion yield of 43% from D-galactose to D-tagatose under the optimal condition, and appeared to be a more efficient catalyst compared with other L-AIs from various organisms. Resveratrol is now gaining much attention because of its pharmacological properties. Polygonum cuspidatum has the highest content of resveratrol in plants and is the best material offering resveratrol. However, the content of resveratrol in P. cuspidatum is much lower compared with its glycoside polydatin. In this study, enzymatic transformation of polydatin to resveratrol by piceid-β- d-glucosidase from Aspergillus oryzae sp. 100 was investigated. The biotransformation conditions were optimized. Under the optimized conditions of 60 °C, pH 5.0, substrate concentration of 40 g/L and piceid-β- d-glucosidase activity of 5 U/mL, enzymatic transformation of polydatin from P. cuspidatum was successfully performed, during which 22.5 g/L of resveratrol was produced after reacting for 4 h, with the substrate conversion rate of 2 g/h/U of piceid-β- d-glucosidase. A feasible and environment friendly process of enzymatic transformation of polydatin to resveratrol was developed, which provides a promising and competitive alternative for the production of resveratrol.
