This study developed a new method for the analysis of 20 phenolic compounds in human urine. The urine samples were prepared by hydrochloric acid (HCl) hydrolysis, liquid–liquid extraction (LLE), and solid-phase extraction (SPE) cleanup. We found that HCl hydrolysis is of similar effectiveness to, and much cheaper than, the traditional enzymatic method. Vanillic acid was co-eluted with butyl paraben and interfered with the determination of butyl paraben in urine. K2CO3-treated-silica-gel SPE was designed to efficiently eliminate interference from the endogenous organic acids (especially vanillic acid) in urine. After derivatization, the samples were analyzed by large-volume-injection gas chromatography–tandem mass spectrometry (LVI-GC–MS–MS). Good linearity (R 2 ≥ 0.996) was established in the range 0.1–100 ng mL−1 for all analytes. Method detection limits (MDLs) were 0.7–9.8 pg mL−1. Intraday (n = 5) and interday (n = 5 days) validation was performed, with satisfactory accuracy (recovery: 70–126 % and 73–107 %, respectively) and precision (RSD ≤ 19 %) at two levels (low: 0.1 and 0.5 ng mL−1; high: 5 and 10 ng mL−1). The method was used in a population study and achieved more than 85 % detection for most analytes; mean analyte concentrations were in the range 0.01–185 ng mL−1. Interleukin 1β (IL-1β) is a key orchestrator of inflammation and host defense that exerts its effects through IL-1 receptor type I (IL-1RI) and IL-1 receptor accessory protein (IL-1RAcP). How IL-1RAcP is recruited by IL-1β-IL-1RI to form the signaling-competent complex remains elusive. Here we present the crystal structure of IL-1β bound to IL-1 receptor type II (IL-1RII) and IL-1RAcP. IL-1β-IL-1RII generated a composite binding surface to recruit IL-1RAcP. Biochemical analysis demonstrated that IL-1β-IL-1RI and IL-1β-IL-1RII interacted similarly with IL-1RAcP. It also showed the importance of two loops of IL-1 receptor antagonist (IL-1Ra) in determining its antagonism. Our results provide a structural basis for assembly and activation of the IL-1 receptor and offer a general cytokine-receptor architecture that governs the IL-1 family of cytokines. In the present study, we developed a gas chromatography–quadrupole mass spectrometry method for analyzing nine pyrethroid insecticides in sediment after accelerated solvent extraction and solid phase extraction cleanup. The operation was optimized, and negative chemical ionization was selected to improve analytical selectivity. The sediment matrix effects on qualification were evaluated, and matrix-matched standard solutions, along with the internal standard calibration, were used to reduce the matrix-induced chromatographic response enhancement. The method detection limits were 0.68 to 1.43 μg/kg dry weight (dw), and recoveries were 70.3 to 143.3%, 61.1 to 169.7%, and 65.7 to 118.8%, with relative SDs of 4.2% to 32.1%, 4.9 to 23.6%, and 1.5 to 23.3% at the spiked levels of 1.0, 5.0 and 20 μg/kg dw, respectively. Pyrethroid insecticides have been implicated as the cause of sediment toxicity to Hyalella azteca in both agricultural and urban areas of California; however, for a subset of these toxic sediments (~30%), the cause of toxicity remains unidentified. This article describes the analytical method development for seven additional pesticides that are being examined to determine if they might play a role in the unexplained toxicity. A pressurized liquid extraction method was optimized to simultaneously extract diazinon, methyl parathion, oxyfluorfen, dicofol, fenpropathrin, pyraclostrobin, and indoxacarb from sediment, and the extracts were cleaned using a two-step solid-phase extraction procedure. The final extract was analyzed for the target pesticides by gas chromatography/nitrogen–phosphorus detector (GC/NPD), and gas chromatography/electron capture detector (GC/ECD), after sulfur was removed by shaking with copper and cold crystallization. Three sediments were used as reference matrices to assess method accuracy and precision. Method detection limits were 0.23–1.8 ng/g dry sediment using seven replicates of sediment spiked at 1.0 ng/g dry sediment. Recoveries ranged from 61.6 to 118% with relative standard deviations of 2.1–17% when spiked at 5.0 and 50 ng/g dry sediment. The three reference sediments, spiked with 50 ng/g dry weight of the pesticide mixture, were aged for 0.25, 1, 4, 7, and 14 days. Recoveries of the pesticides in the sediments generally decreased with increased aging time, but the magnitude of the decline was pesticide and sediment dependent. The fur seal ( Callorhinus ursinus) population has decreased in their primary breeding grounds in the Bering Sea; contamination is among suspected causes. Our goal was to better understand the extent of contamination of seal tissues with certain organochlorine compounds by measuring the concentrations of polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs) in fur seal tissues from St. Paul Island, to gain a better perspective of tissue congener distribution and to evaluate the observed PCB levels against toxicologically significant levels for modes of action. Concentrations of 145 PCB congeners (∑145PCBs) and 12 OCPs were measured with gas chromatography–ion trap mass spectrometry in 8 different tissues of 10 male northern fur seals. The mean concentrations of ∑OCPs [in ng/g lipid weight (lw)] were 1180 in blubber, 985 in the heart, 1007 in the liver, 817 in the kidney, 941 in muscle, 660 in reproductive tissues, 204 in the brain, and 322 in the lung. The mean concentrations of ∑145PCBs (in ng/g lw) were 823 in blubber, 777 in the liver, 732 in the heart, 646 in reproductive tissues, 638 in muscle, 587 in the kidney, 128 in the lung, and 74.3 in brain tissues. Concentrations of PCBs affecting the aryl hydrocarbon receptor expressed as total PCB toxic equivalents (∑PCB-TEQs) ranged from 0.3 to 545 pg/g lw for the various tissues. The major contributors to ∑PCB-TEQs are CB-118 in muscle, brain, lung, kidney, and liver, CB-126 in blubber, and CB-118 and CB-126 equally in the heart and reproductive tissues. Concentrations of PCBs affecting Ca2+ homeostatsis expressed as the neurotoxic equivalent (NEQ) showed ∑PCB-NEQs ranged from 17.7 to 215 ng/g lw in all tissues. Although no composite measure of perturbation of thyroid function is available, sufficient amounts of congeners with high binding to the thyroxine transport system were present to warrant consideration of this mode of action in future studies. Analyses of 145 PCBs and mode of action evaluation suggest that PCB contamination could potentially exert an effect on the Alaskan northern fur seal population although the PCB concentrations have been decreasing in the fur seals over the last decade. The expression cassette I10 containing the new-found flocculation gene, FLONS, was transformed into an industrial strain Saccharomyces cerevisiae YSF5. Upstream activating sequences of the S. cerevisiae alcohol dehydrogenase II ( ADH2) gene promoter (PU-ADH2) were used to regulate the expression of FLONS; α-acetolactate synthase gene ILV2 was chosen for homologous recombination of I10 to the YSF5 chromosome; copper binding metallothionein (encoded by CUP1) was used for selection of transformants. Ten randomly selected transformants exhibited increased flocculation ability of 1.5 to 2.3 fold more than the original strain. Based on their sensitivity to glucose, maltose and sucrose, flocculation property of the transformants was supported to be NewFlo-type. After successive subculture, the introduced CUP1 remained in the transformants. At the end of simulated fermentation test, diacetyl content of the culture media of 5I-1 was 0.45 g l−1, lower than YSF5 (0.48 g l−1). [ABSTRACT FROM AUTHOR]

To actively express an outer membrane protein, protein I (PI), from different strains of Neisseria gonorrhoeae in E.␣coli, PI gene fragments from two reference strains and four clinical isolates of Neisseria gonorrhoeae were obtained with PCR amplification. They were cloned into the PCR cloning vector pBS-T to form pBS-T-PI and sequenced. Subsequently, they were cloned into an expression vector pET-30b (+) to generate pET-PI recombinants. After inducing with isopropyl-β- d -thiogalactopyranoside (IPTG), the expressed PI proteins were analysed by SDS-PAGE, Western blotting and ELISA. The results implied that we had successfully constructed the PI gene recombinants from both reference strains and clinical isolates and obtained the recombinant proteins expressed in E. coli at relatively high levels, and the expressed proteins had the immunological activity with the corresponding antibodies. This research will be very helpful for the further study of these proteins in generating preventive vaccines on Neisseria gonorrhoeae infection and clinical diagnosis. [ABSTRACT FROM AUTHOR]
