Osteosarcoma is an aggressive form of bone cancer and affects the health in children and adolescents. Although conventional treatment improves the osteosarcoma survival, some patients have metastasis and drug resistance, leading to a worse prognosis. Therefore, it is necessary to explore the molecular mechanism of osteosarcoma occurrence and progression, which could discover the novel treatment for osteosarcoma. Long noncoding RNAs (lncRNAs) have been reported to regulate osteosarcoma occurrence and malignant progression. LncRNA HOXA-AS3 facilitates the tumorigenesis and progression in a variety of human cancers. However, the underlying mechanism of lncRNA HOXA-AS3-induced oncogenesis is poorly determined in osteosarcoma. To address this point, we utilized several cellular biological strategies and molecular approaches to explore the biological functions and mechanisms of lncRNA HOXA-AS3 in osteosarcoma cells. We found that lncRNA HOXA-AS3 facilitates cell proliferation and invasion via targeting miR-218-5p/FOXP1 axis in osteosarcoma. A novel construction strategy is introduced for an ultrasensitive dynamic light scattering (DLS) immunosensor targeting alpha fetoprotein (AFP). This approach relies on a self-assembled heptamer fusion protein (A1-C4bpα), incorporating the dual functions of multivalent recognition and crosslinking aggregation amplification due to the presence of seven AFP-specific A1 nanobodies on the A1-C4bpα heptamer. Leveraging antibody-functionalized magnetic nanoparticles for target AFP capture and DLS signal output, the proposed heptamer-assisted DLS immunosensor offers high sensitivity, strong specificity, and ease of operation. Under the optimized conditions, the designed DLS immunosensor demonstrates excellent linear detection of AFP in the concentration range 0.06 ng mL−1 to 512 ng mL−1, with a detection limit of 15 pg mL−1. The selectivity, accuracy, precision, practicability, and reliability of this newly developed method were further validated through an assay of AFP levels in spiked and actual human serum samples. This work introduces a novel approach for constructing ultrasensitive DLS immunosensors, easily extendable to the sensitive determination of other targets via simply replacing the nanobody sequence, holding great promise in various applications, particularly in disease diagnosis. Background: Rapid cycling bipolar disorder (RCBD), characterized by four or more episodes per year, is a complex subtype of bipolar disorder (BD) with poorly understood characteristics. Method: This multicenter, observational, longitudinal cohort study enrolled 520 BD patients across seven psychiatric institutions in China from January 2013 to January 2014. Participants were divided into RCBD and non-RCBD (NRCBD) groups based on the frequency of mood episodes in the preceding year. Data collection utilized a standardized form, supplemented by a medical record review, focusing on sociodemographic, clinical, and treatment characteristics. Statistical analysis involved independent samples t-tests, Kruskal–Wallis H tests, Chi-square or Fisher's exact tests, with Bonferroni correction applied to account for multiple comparisons, and multivariable logistic regression to identify characteristics associated with RCBD. Results: Among the BD cohort, 9.4% were identified as current RCBD. Compared to NRCBD, RCBD patients had a shorter duration from the first psychiatric consultation to the diagnosis of BD, a reduced duration of their longest period of euthymia, a lower proportion of lifetime hospitalization history due to BD, and less use of electroconvulsive therapy (ECT) within the last 12 months. Additionally, they presented higher baseline scores on the Mood Disorder Questionnaire (MDQ) and the Brief 16-item Quick Inventory of Depressive Symptomatology Self-Report (QIDS-SR16). However, after applying the Bonferroni correction, these differences were not statistically significant. Multivariable logistic regression analysis identified three factors that were independently associated with RCBD: time from first psychiatric consultation to BD diagnosis (Odds Ratio [OR] = 0.512, P = 0.0416), lifetime hospitalization history due to BD (OR = 0.516, P = 0.0476), and ECT treatment within the past 12 months (OR = 0.293, P = 0.0472). Conclusion: This study revealed that the duration from first psychiatric consultation to BD diagnosis, lifetime hospitalization history due to BD, and ECT treatment in the past year were associated with RCBD. Spinal cord injury (SCI) is one of the most devastating traumas, and the aberrant proliferation of astrocytes usually causes neurological deficits. However, the mechanism underlying astrocyte over-proliferation after SCI is unclear. Grin2c (glutamate ionotropic receptor type 2c) plays an essential role in cell proliferation. Our bioinformatic analysis indicated that Grin2c and Ca2+ transport functions were inhibited in astrocytes after SCI. Suppression of Grin2c stimulated astrocyte proliferation by inhibiting the Ca2+/calmodulin-dependent protein kinase 2b (CaMK2b) pathway in vitro. By screening different inflammatory factors, interleukin 1α (IL1α) was further found to inhibit Grin2c/Ca2+/CaMK2b and enhance astrocyte proliferation in an oxidative damage model. Blockade of IL1α using neutralizing antibody resulted in increased Grin2c expression and the inhibition of astrocyte proliferation post-SCI. Overall, this study suggests that IL1α promotes astrocyte proliferation by suppressing the Grin2c/Ca2+/CaMK2b pathway after SCI, revealing a novel pathological mechanism of astrocyte proliferation, and may provide potential targets for SCI repair. The article presents the results of experimental studies on the gasification of mixtures of brown coal and polyethylene (up to 20 wt% fraction) in a laboratory reactor. The work aims to study the agglomeration process during the heating and oxidation of the mixtures. The measurement results (gas composition, pressure drop) provide indirect information on the dynamics of thermal decomposition and structural changes in the fuel bed. We have shown that the interaction between polyethylene and a coal surface leads to the formation of dense agglomerates, in which the molten polymer acts as a binder. Clinkers form as a result of interfacial interactions between components and filtration flow rearranging. The hydrogen/carbon ratio in the solid residue of coal-polyethylene co-gasification increases from 0.07–0.2 to 1.11, indicating the formation of stable hydrocarbon compounds on the carbon surface. The conducted research makes it possible to identify possible interactions between chemical reactions and transfer processes that lead to agglomeration in mixtures of coal with polyethylene. The latest development of Generative Artificial Intelligence (GenAI), particularly ChatGPT, has drawn the attention of educational researchers and practitioners. We have witnessed many innovative uses of ChatGPT in STEM classrooms. However, studies regarding students' perceptions of ChatGPT as a virtual tutoring tool in STEM education are rare. The current study investigated undergraduate students' perceptions of using ChatGPT in a physics class as an assistant tool for addressing physics questions. Specifically, the study examined the accuracy of ChatGPT in answering physics questions, the relationship between students' ChatGPT trust levels and answer accuracy, and the influence of trust on students' perceptions of ChatGPT. Our finding indicates that despite the inaccuracy of GenAI in question answering, most students trust its ability to provide correct answers. Trust in GenAI is also associated with students' perceptions of GenAI. Soot generation is a major challenge in industries. The elimination of soot is particularly crucial to reduce pollutant emissions and boost carbon conversion. The mechanisms for soot oxidation are complex, with quantified models obtained under in situ conditions still missing. We prepare soot samples via noncatalytic partial oxidation of methane. Various oxidation models are established based on the results of in situ transmission electron microscopy experiments. A quantified maturity parameter is proposed and used to categorize the soot particles according to the nanostructure at various maturity levels, which in turn lead to different oxidation mechanisms. To tackle the challenges in the kinetic analysis of soot aggregates, a simplification model is proposed and soot oxidation rates are quantified. In addition, a special core-shell separation model is revealed through in situ analysis and kinetic studies. In this study, we obtain important quantified models for soot oxidation under in situ conditions. Using a newly developed experimental setup, the features and advantages of an autothermal single-casing atmospheric subbituminous coal fluidized bed air-blown gasifier, combining a combustion and gasification section, and mixing the dispersed phase (inert material, char) and heat exchange between them through an annular transfer device, have been revealed. To increase the efficiency of the gasifier, an experimental-computational method was developed find the conditions for optimal operation, combining changing the annular flow's geometry and regulating the primary air for gasification. A simple and reliable multizone thermodynamic calculation model makes it possible to predict the composition of char and syngas in the gasification section with acceptable accuracy. This method confirmed that a two-section fluidized bed gasifier can provide efficient gasification of solid fuels and is suitable for use in small-scale cogeneration plants. Syngas with a heating value of 3.6–4.5 MJ/m3 and CGE of 38.2%–42.3% was obtained in the experimental setup without optimizing the primary air flow rate. With optimization, the indicators increased to the heating value of syngas of 5.20–5.34 MJ/m3 and CGE of 42.5%–50.0%. With heat regeneration of 0.8, CGE increases to 70%. [ABSTRACT FROM AUTHOR]

With the increase in extremely low birth weight (ELBW) infants, their outcome attracted worldwide attention. However, in China, the related studies are rare. The hospitalized records of ELBW infants discharged from twenty-six neonatal intensive care units in Guangdong Province of China during 2008–2017 were analyzed. A total of 2575 ELBW infants were enrolled and the overall survival rate was 55.11%. From 2008 to 2017, the number of ELBW infants increased rapidly from 91 to 466, and the survival rate improved steadily from 41.76% to 62.02%. Increased survival is closely related to birth weight (BW), regional economic development, and specialized hospital. The incidence of complications was neonatal respiratory distress syndrome (85.2%), oxygen dependency at 28 days (63.7%), retinopathy of prematurity (39.3%), intraventricular hemorrhage (29.4%), necrotizing enterocolitis (12.0%), and periventricular leukomalacia (8.0%). Among the 1156 nonsurvivors, 90.0% of infants died during the neonatal period (≤ 28 days). A total of 768 ELBW infants died after treatment withdrawal, for reasons of economic and/or poor outcome. Corticotropin-releasing factor (CRF) neurons are one of the most densely distributed cell types in the central amygdala (CeA), and are involved in a wide range of behaviors including anxiety and learning. However, the fundamental input circuits and patterns of CeA-CRF neurons are still unclear. Here, we generate a monosynaptic-input map onto CeA-CRF neurons at single-cell resolution via a retrograde rabies-virus system. We find all inputs are located in 44 nested subregions that directly innervate CeA-CRF neurons; most of them are top-down convergent inputs expressing Ca2+/calmodulin-dependent protein kinase II, and are centralized in cortex, especially in the layer 4 of the somatosensory cortex, which may directly relay information from the thalamus. While the bottom-up divergent inputs have the highest proportion of glutamate decarboxylase expression. Finally, en passant structures of single input neuron are revealed by in-situ reconstruction in a modified 3D-reference atlas, represented by a Periaqueductal gray-Subparafascicular nucleus-Subthalamic nucleus-Globus pallidus-Caudoputamen-CeA pathway. Taken together, our findings provide morphological and connectivity properties of inputs onto CeA-CRF neurons, which may provide insights for future studies interrogating circuit mechanisms of CeA-CRF neurons in mediating various functions. Purpose: Autophagy and EMT (epithelial–mesenchymal transition) are the two principal biological processes and ideal therapeutic targets during cancer development. Autophagy, a highly conserved process for degrading dysfunctional cellular components, plays a dual role in tumors depending on the tumor stage and tissue types. The EMT process is the transition differentiation from an epithelial cell to a mesenchymal-like cell and acquiring metastatic potential. There is evidence that the crosstalk between autophagy and EMT is complex in cancer. In recent years, more studies have shown that long non-coding RNAs (lncRNAs) and microRNAs (miRNAs) are involved in autophagy, EMT, and their crosstalk. Therefore, accurate understanding of the regulatory mechanisms of lncRNAs and miRNAs in autophagy, EMT and their interactions is crucial for the clinical management of cancers. Methods: An extensive literature search was conducted on the Google Scholar and PubMed databases. The keywords used for the search included: autophagy, EMT, crosstalk, lncRNAs, miRNAs, cancers, diagnostic biomarkers, and therapeutic targets. This search provided relevant articles published in peer-reviewed journals until 2021. Data from these various studies were extracted and used in this review. Results: The results showed that lncRNAs/miRNAs as tumor inhibitors or tumor inducers could regulate autophagy, EMT, and their interaction by regulating several molecular signaling pathways. The lncRNAs/miRNAs involved in autophagy and EMT processes could have potential uses in cancer diagnosis, prognosis, and therapy. Brachial plexus avulsion (BPA) is a devastating traumatic peripheral nerve injury complicated with paralysis of the upper extremity. We previously reported that leucine-rich repeat and immunoglobulin-like domain-containing NOGO receptor-interacting protein 1 (LINGO-1) has a potent role in inhibiting neuron survival and axonal regeneration after the central nervous system (CNS) damage and miR-615 is a potential microRNA (miRNA) negatively regulated LINGO-1. However, the effect of miR-615 in BPA remains to be elucidated. Accumulating evidence indicates that pluronic F-127 (PF-127) hydrogel could serve as a promising vehicle for miRNA encapsulation. Thus, to further explore the potential role of hydrogel-miR-615 in BPA-reimplantation, the present study established the BPA rat model and injected miR-615 agomir encapsulated by PF-127 hydrogel into the reimplantation site using a microsyringe. In this study, results indicated that hydrogel-miR-615 agomir effectively alleviated motoneuron loss by LINGO-1 inhibition, promoted musculocutaneous nerve regeneration and myelination, reduced astrocytes activation, promoted angiogenesis and attenuated peripheral amyotrophy, leading to improved motor functional rehabilitation of the upper extremity. Pedestrian detection has a pivotal role in the field of computer vision. Recently, deep convolutional neural networks (CNNs) have been demonstrated to achieve appealing performance in object detection compared to hand-crafted methods, with single shot multiBox detector (SSD) being one of state-of-the-art methods in terms of both speed and accuracy. In this paper, we propose a novel framework which is able to perform pedestrian detection by not only considering local features but also by incorporating global information into features to make them more discriminative for this task. Specifically, we first integrate feature pyramid network into the SSD detection framework. Next, a Squeeze-and-Excitation network is proposed to encode global information. Hence, the features become more focused on pedestrians, in particular those of small scale and with occlusion. We further introduce a network in network fusion module, which enhances the features by incorporating local details. In this way our framework is able to suppress background information and highlights pedestrian elements. Experimental results show that the proposed framework can achieve comparable detection results to state-of-the-art methods and run an average of 17 frames per second (fps) on NVidia TITAN X GPU with image size of 600 × 600 . The two-component Cry48Aa/Cry49Aa toxin produced by Lysinibacillus sphaericus shows specifically toxic to Culex quinquefasciatus mosquito larvae. Cry49Aa C-terminal domain is responsible for specific binding to the larval gut cell membrane, while its N-terminal domain is required for interaction with Cry48Aa. To investigate functional role of cysteine in Cry49Aa, four cysteine residues at positions 70, 91, 183, and 258 were substituted by alanine. All mutants showed similar crystalline morphology and comparable yield to that of the wild type except that the yield of the C91A mutant was low. Four cysteine residues did not involve in disulfide bond formation within or between Cry49Aa molecules. Cys91, Cys183, and Cys258 are essential for larvicidal activity against C. quinquefasciatus larvae, while Cys70 is not. Substitution at C91, C183, and C258 caused weaker Cry48Aa- Cry49Aa interaction, while mutations at C183 and C258 reduced the binding capacities to the larval gut cell membrane. Thus, Cysteine residues at position 91, 183, and 258 in Cry49Aa are required for full toxicity of Cry48Aa/Cry49Aa toxin. [ABSTRACT FROM AUTHOR]

Autophagy has been proved to play a vital role in cardiac hypertrophy. The present study was designed to investigate the relationship between miR-100-5p and autophagy in the development of cardiac hypertrophy. Here, miR-100-5p expression was detected in abdominal aortic coarctation (AAC)-induced cardiac hypertrophy rats and Angiotensin II (Ang II)-stimulated cardiomyocytes. In vitro and in vivo experiments were performed to explore the function of miR-100-5p on autophagy and cardiac hypertrophy. We also investigated the mechanism of miR-100-5p on autophagy with dual-luciferase reporter assays, RNA immunoprecipitation (RIP), quantitative real-time PCR (qRT-PCR), western blot, immunofluorescence, and transmission electron microscopy (TEM). The results showed that miR-100-5p was highly expressed in hypertrophic hearts and Ang II-induced cardiomyocytes. Overexpression of miR-100-5p promoted the expression of cardiac hypertrophy markers ANP, BNP and β-MHC and cell surface area, while those were suppressed by miR-100-5p inhibitor. Knockdown of miR-100-5p by antagomiR significantly improves cardiac function and attenuate cardiac hypertrophy in vivo. Mechanistic investigation has found that miR-100-5p promote autophagy by targeting mTOR. Inhibition of autophagy by 3-methyladenine (3-MA) or mTOR overexpression could reverse the function of miR-100-5p in cardiac hypertrophy. The environmental behavior and ultimate fate of the engineered nanoparticles could inevitably be affected by the widespread dissolved organic matter and sunlight irradiation in the natural environment. In this study, an analysis combined with experimental investigations was used to explore the influence of humic acid (HA) and UV irradiation on the changes in surface charge and aggregation behavior of TiO2 nanoparticles. A prolonged UV irradiation caused the point of zero charge (PZC) of TiO2 nanoparticles shifted to lower values and the aggregate size reached to a maximum value at the PZC. TEM image and FT-IR spectroscopy analysis confirmed the formation of TiO2 cluster aggregates and successive growth of the hydroxyl groups on different sites on TiO2 surface, respectively, although no effect of irradiation on the phase transformation of TiO2 nanoparticles was observed through the use of Raman spectroscopy. The fluorescence spectroscopy analysis illustrated the interactions between the surface hydroxyls formed during UV irradiation on TiO2 surface and functional groups on HA molecules, which accelerated the aggregation process of TiO2 nanoparticles. Widespread HA and UV irradiation in the natural environment significantly altered the surface properties and stability of the nanoparticles in aqueous environments. [ABSTRACT FROM AUTHOR]

In this paper, we propose a novel visual object tracking method using a part based appearance model. First, a local kernel feature is developed to encode edge information of patches. Next, bounding box of the target is divided into multiple parts. Then, each part uses correlation filter based tracking to predict position in the next frame. The matrix cosine similarity is utilized to measure reliabilities of the patches. Finally, optimal target location is predicted via maximizing the likelihood, which is obtained by adaptively fusing the reliable patches locations. In this study, the effect of ash removal on Shenfu bituminous coal was investigated. The coal was pretreated by hydrofluoric acid (HF) pickling, and the raw/pretreated coal chars were prepared at 900 °C in a fixed bed reactor. The structure of coal and char were detected by Fourier transform infrared (FTIR) and Raman spectroscopy. The reactivity was tested in a thermogravimetric analyzer, including coal pyrolysis and char gasification. The reaction kinetics was analyzed through the Coats–Redfern method, master plots, the model-free and model-fitting method. The results show that the HF pickling can remove silicon from coal efficiently, and the macromolecular framework of coal is quite stable according to FTIR. The Raman parameters imply some carbonaceous structure on coal surface changed. For slow pyrolysis of coal, the effect of heating rate is considered. The changes of pyrolysis characteristics and kinetics are insignificant. For char gasification, the reactivity under isothermal and non-isothermal condition are discussed with an emphasis in different residence time of devolatilization process. In kinetic control region (low temperature), the activation energy (Ea) is very close (about 240 kJ/mol) for all chars. With the temperature increases, the reactivity of raw coal char is more easily suffered by diffusion. The random pore model is more suitable for the ash-free coal char, and the char with long residence time has a larger value of structural parameter ψ and smaller value of pre-exponential factor A. The greenhouse problem has a significant effect on our communities such as, health and climate. Carbon dioxide is one of the main gases that cause global warming. Therefore, CO2 capture techniques have been the focus of attention these days. The chemical looping combustion technique adopted the air reactor and fuel reactor to recycle heat energy. This study presents a numerical and experimental investigation on a fuel reactor in chemical looping combustor (CLC) system. The present numerical model is introduced by the kinetic theory of granular flow and coupled with gas–solid flow with chemical reactions to simulate the combustion of solids in the CLC. The k–ε turbulent model was used to model the gas phase and the particle phase. The developed model simplify the prediction of flow patterns, particle velocities, gas velocities, and composition profiles of gas products and the distribution of heterogeneous reaction rates under the same operating conditions. The predicted and experimental results were compared according to the basis of determination coefficient (R2). In addition the results showed that there is a good agreement between the predicted and experimental data. The value of (R2) for CO, CO2 and CH4 was 0.959, 0.925 and 0.969 respectively. LINGO-1(LRR and Ig domain–containing NOGO receptor interacting protein 1) is a viable target for spinal cord injury (SCI) repair due to its potent negative regulation in neuron survival and axonal regeneration. Although promising, the intracellular mechanism underlying LINGO-1 regulation is unclear. Here, we identified miR-615 as a potential microRNA (miRNA) that directly targets LINGO-1 by binding its 3′-untranslated region (3′-UTR) and caused the translation inhibition of LINGO-1. MiR-615 negatively regulated LINGO-1 during neural stem cell (NSC) differentiation and facilitated its neuronal differentiation in vitro. Interestingly, compared to the control, neurons differentiated from miR-615-treated NSCs were immature with short processes. Further results showed LINGO-1/epidermal growth factor receptor (EGFR) signaling may be involved in this process, as blockade of EGFR using specific antagonist resulted in mature neurons with long processes. Furthermore, intrathecal administration of miR-615 agomir in SCI rats effectively knocked down LINGO-1, increased neuronal survival, enhanced axonal extension and myelination, and improved recovery of hindlimbs motor functions. This work thus uncovers miR-615 as an effective miRNA that regulates LINGO-1 in NSC and SCI animals, and suggests miR-615 as a potential therapeutic target for traumatic central nervous system (CNS) injury. The Cry48Aa/Cry49Aa binary toxin from Lysinibacillus sphaericus is composed of a three-domain Cry-like toxin (Cry48Aa) and a binary-like protein (Cry49Aa) that work together to kill Culex quinquefasciatus mosquito larvae through a novel interaction between its two components. The aim of this study was to identify the functional regions of Cry48Aa that were involved in the interaction with Cry49Aa. Eight Cry48Aa truncated fragments were constructed from both N- and C-termini and expressed in Escherichia coli. Only the individual or combined N69K truncated fragment, a Cry48Aa N-terminal derivative consisting of three domains, showed larvicidal activity against C. quinquefasciatus larvae, while the other fragments exhibited significant loss of biological activity. Visual object tracking is a fundamental problem in computer vision. It heavily relies on feature description for the appearance of object. In this paper, we present a robust algorithm which exploits the locally adaptive regression kernel (LARK) feature for visual tracking. The proposed approach formulates the LARK feature in a tracking by detection framework. In addition, we compute a target-specific saliency map as LARK feature with the guidance of the tracking framework. The tracking problem is solved by maximizing an object location likelihood function. We adopt Fast Fourier Transform for fast learning and detection in this work. This paper proposes an online object tracking algorithm in which the object tracking is achieved by using multi-task sparse learning and non-negative matrix factorization under the particle filtering framework. The object appearance is first modeled by subspace learning to reflect the target variations across frames. Combination of non-negative components is learned from examples observed in previous frames. In order to robust tracking an object, group sparsity constraints are included to the non-negativity one. Furthermore, the alternating direction method of multipliers algorithm is employed to compute the model efficiently. This exploratory study examined the motivational effects of an instructional method that involved students in the rubric development on animation instruction. Drawing upon Keller's ARCS motivation model, this study compared student motivation regarding attention (A), relevance (R), confidence (C), and satisfaction (S) towards animation instruction before and after their involvement in the rubric development. This study recruited seventeen undergraduates participating in a technology integration course and administered motivation surveys before and after implementing the instructional method. The survey results revealed that student motivation was significantly increased in terms of relevance. Motivated students developed a self-developed rubric by drawing upon their experience and perspectives of creating animated research presentation videos. In this paper, we present a multi-scale predictions fusion region-based Fully Convolutional Networks (MSPF-RFCN) to robustly detect and classify human hands under various challenging conditions. In our approach, the input image is passed through the proposed network to generate score maps, based on multi-scale predictions fusion. The network has been specifically designed to deal with small objects. It uses an architecture based on region proposals generated at multiple scales. Our method is evaluated on challenging hand datasets, namely the Vision for Intelligent Vehicles and Applications (VIVA) Challenge and the Oxford hand dataset. It is compared against recent hand detection algorithms. The experimental results demonstrate that our proposed method achieves state-of-the-art detection for hands of various sizes. In this paper, we propose a hybrid correlation filter based tracking method which depends on coupled interactions between a global filter and two local filters. Specifically, a local kernel feature with Gaussian curvature is developed to encode object appearance. Then the global filter and the two local filters independently track the target. The peak-to-sidelobe ratio (PSR) is employed to measure the reliability of the tracking results. Next, the global filter and the two local filters jointly determine the target position. In this way, the proposed hybrid model deals well with challenging situations, e.g., partial occlusion and scale changes. In this paper, we present a low-rank sparse tracking method which builds upon the particle filtering framework. The proposed method learns the local dense scale-invariant feature transform features corresponding to candidate samples jointly by exploiting the underlying sparse and low-rank constraints. Furthermore, the alternating direction method of multipliers method guarantees the optimization equation can be solved accurately and robustly. We evaluate our proposed tracking method against 9 state-of-the-art trackers on a set of 64 challenging sequences. Unmanned aerial vehicle (UAV)-based target tracking is a long-standing problem in UAV applications. In this paper, we develop a local kernel feature to encode properties of UAV tracking object. Meanwhile, object proposals can provide a reliable prior knowledge to identify tracking target being an object or not. Therefore, we propose to integrate detection proposal method into a tracking by detection framework. More specifically, we adopt edge box proposals and random samplings as training examples and then train these examples for tracking task. The structured support vector machine is employed to implement training and detecting procedure. To reveal the effectiveness of our method, experiment is performed on the UAV123 benchmark dataset. Multiple feature object representation has been proved as a robust approach for visual tracking. Different types of situations such as occlusion, rotation and illumination may occur during tracking, especially long sequences. Robust tracking could be obtained as multiple features could complement each other. In this paper, we cast visual tracking as a novel multi-task sparse learning problem and exploit various types of visual features, such as intensity, color, texture and edge, where each feature can be sparsely represented by a linear combination of atoms from an adaptive feature template. We use an on-line feature selection mechanism based on the two-class variance ratio measure, applied to log likelihood distributions computed with respect to a given feature from samples of object and background pixels. The proposed method is integrated in a particle filtering framework. We jointly consider the underlying relationship across different particles, and tackle it in a unified robust multi-task formulation. In addition, to capture the frequently emerging outlier tasks, we make fully use of a decomposition model which enables a more robust and accurate approximation. We show that the proposed model can be efficiently solved using the Alternating direction method of multipliers (ADMM) with a small number of closed-form updates. Four types of features are implemented and tested on numerous benchmark video sequences. Osteoporosis (OP) is an increasing public health problem worldwide. Genetic factors are considered to be major contributors to the pathogenesis of OP. The aim of this study was to investigate the association of the purinergic P2X7 receptor (P2X7R) and estrogen receptor-α (ER-α) genes with OP risk, and the effect of the possible interaction between the two genes on predisposition to OP in Chinese postmenopausal women. A total of 596 subjects, including 350 OP patients and 246 controls, were recruited in this case-control study. Five functional single-nucleotide polymorphisms (SNPs) in the P2X7R gene (rs2393799, rs7958311, rs1718119, rs2230911, rs3751143) and two ER-α PvuII and XbaI polymorphisms were genotyped and analyzed. Single-gene variant analysis showed that the carriers of the CC genotype of P2X7R rs3751143 revealed an increased OP risk. Haplotype rs1718119G-rs2230911G-rs3751143C also appeared to be a significant 'risk' haplotype with OP. For the ER-α gene, no evidence of significant association of PvuII or XbaI polymorphism with OP risk was found. Moreover, there was a significant gene-gene interaction between P2X7R rs3751143 and ER-α PvuII; the cross-validation consistency was 10/10 and the testing accuracy was 0.5818 (P = 0.0107). A 1.67-fold-increased risk for OP was detected in individuals carrying the genotypes of AC or CC of rs3751143 and Pp or PP of PvuII compared to subjects with AA of rs3751143 and pp of PvuII. Our findings suggest an important association of the P2X7R rs3751143CC genotype and the rs1718119G-rs2230911G-rs3751143C haplotype with an increased OP risk. Background: The aim was to explore the factors contributing to a high degree of satisfaction following total hip arthroplasty in patients with ankylosing spondylitis and hip joint fusion.Methods: From January 2001 to June 2015, 43 patients with ankylosing spondylitis were treated with total hip arthroplasty. The patients were divided into two groups, including 20 patients in non-fusion groups, with 25 cases of hip non-fusion, and 23 patients in fusion groups, with 40 cases of hip fusion. Patients were followed for an average of 27 months. Harris hip scores (HHSs) and Barthel scores were recorded pre-operatively and post-operatively. The improvement rates of both groups were calculated, and the data were compared and analyzed. The Self-Administered Patient Satisfaction Scale (SAPS) was used to assess the degree of satisfaction in each group.Results: The post-operative HHS and Barthel scores of two groups both significantly improved. In non-fusion groups compared with fusion groups, there were significant differences in the pre-operative HHSs, the improvement of the HHSs, the pre-operative Barthel score, the improvement of the Barthel score, and the satisfaction score. However, there were no significant differences in the post-operative HHSs and Barthel scores.Conclusion: Patients in the hip fusion group had higher satisfaction scores. The improvements in joint function and self-care ability after total hip arthroplasty are the decisive factors in determining patient satisfaction for patients with ankylosing spondylitis combined with hip fusion. Fluorescent light-up probes based on aggregation-induced emission (AIE)-active molecules have recently attracted great research interest due to the intelligent fluorescence activation mechanism and high sensitivity. In this work, an AIE-silica nanosphere (SiO NP)-based label-free fluorescent aptasensor for the sensitive 'turn-on' detection of prostate-specific antigen (PSA) is reported for the first time. The positively charged amino-functionalized SiO NPs were used as efficient nanocapturer to electrostatically adsorb single-stranded PSA aptamer (PA) to form SiO NP-PA nanocomposite as well as adsorb negatively charged tetraphenylethylene derivative 3 (TPE3) to form AIE-SiO NP nanocomposite. The binding of the aptamer to the target PSA could induce a rigid aptamer conformation, resulting in the release of the PA away from the surface of SiO NPs. This made the AIE molecules TPE3 aggregate on the SiO NP surface and emit high fluorescence. With the advantages of simple design and rapid responses, the proposed aptasensor showed high sensitivity and selectivity for PSA with a detection limit of 0.5 ng/mL. The aptasensor was further applied in human serum samples with satisfactory results. Osteoporosis (OP) is a major public health problem worldwide. Genetic factors are considered to be major contributors to the pathogenesis of OP. The purinergic P2X7 receptor (P2X7R) has been shown to play a role in the regulation of osteoblast and osteoclast activity and has been considered as an important candidate gene for OP. A case-control study was performed to investigate the associations of functional single nucleotide polymorphisms (SNPs) in the P2X7R gene (rs2393799, rs7958311, rs1718119, rs2230911, and rs3751143) with susceptibility to OP in 400 Chinese OP patients and 400 controls. Results showed that rs3751143 was associated with OP; in particular, carriers of the C allele and CC/(AC + CC) genotypes were at a higher risk of OP, but no significant association of rs2230911, rs7958311, rs1718119, and rs2393799 with OP risk was observed. Analysis of the haplotypes revealed one haplotype (rs1718119G-rs2230911G-rs3751143C) that appeared to be a significant 'risk' haplotype with OP. The rs3751143 polymorphism was associated with osteoclast apoptosis; ATP-induced caspase-1 activity of osteoclasts with AC and CC genotypes is lower than that of osteoclasts with AA genotype in vitro. The authors have investigated (a) the self-assembly of single-stranded DNA (ssDNA) on glass surfaces, and (b) the interaction of DNA with liquid crystals (LCs) on solid surfaces. The results suggest that ssDNA (compared to dsDNA) on the solid interface causes particularly different orientations in LCs. The LC molecules assume a uniform homeotropic orientation on the surface with a typical surface ssDNA coverage of ~2.4 × 10 molecules per square cm. Once complementary DNA is hybridized on the surface, the homotropic orientation of the LCs becomes disrupted. This orientation transition can be visually observed by using a crossed polarizer. The findings were exploiting to design an assay for target DNA (= analyte DNA) that has an ~0.1 nM detection limit. The assay is highly selective and can easily differentiate target DNA from single-base mismatch and non-complementary DNA. Diabetic nephropathy (DN) is one of the major complications of diabetes. A tremendous amount of genetic variations have been identified to be associated with DN. However, most of them only generate from statistical associations at the DNA level, generally without direct functional evidence regarding their association mechanisms underlying DN. Based on the publicly available datasets and resources, this study performed integrative analyses (expression quantitative trait loci analysis, differential gene expression analysis and functional prediction analysis) to detect the molecular functional mechanisms underlying the associations for DN. Among 150 selected (P < E-4) genetic associations that were archived in the public databases, two single nucleotide polymorphisms (SNPs) (rs3135377 and rs9469220) have been found to act as cis-effect regulators of the 'identified' gene (HLA-DRA and HLA-DRB1). These eQTL genes have differential expression signals in the DN-associated cell groups. These SNPs were predicted as regulatory sites by utilizing online prediction tools. Our data suggest potential mechanistic links underlying the association between DN and two identified SNPs. These results could help us to have a deeper understanding of the functional relevance of genetic variants with susceptibility to DN, which is useful for pursuit of in-depth validation studies to dissect their involvements and molecular functional mechanisms in DN. Diabetic neuropathy (DNP) is a frequent chronic complication of diabetes mellitus with potentially life-threatening outcomes. High glucose and elevated free fatty acids (FFAs) have been recently recognized as major causes of nervous system damage in diabetes. Our previous study has indicated extracellular stimuli, such as high glucose and/or FFA stress, may activate the p38 mitogen-activated protein kinase (MAPK) signaling pathway and induce a p38 MAPK-dependent sensitization of the P2X7 receptor and release of inflammatory factors in PC12 cells, while the mechanisms underlying remain to be elucidated. Long noncoding RNAs (lncRNAs) play important roles in diverse biological processes, including activation of a series of pathway signalings. Here, we showed combined high d-glucose and FFAs (HGHF) induced an increment of lncRNA-NONRATT021972 (NONCODE ID, nc021972) in PC12 cells. Nc021972 small interference RNA (siRNA) alleviated HGHF-induced activation of p38 MAPK, expression of the P2X7 receptor, and [Ca] increment upon P2X7 receptor activation. Further experiments showed that there existed a crosstalk between nc021972 and the p38 MAPK signaling pathway. Inhibition of p38 MAPK signaling decreased nc021972-induced expression of the P2X7 receptor and [Ca] increment upon P2X7 receptor activation. Also, nc021972 siRNA inhibited HGHF-induced PC12 release of TNF-α and IL-6 and rescued decreased cell viability mediated by the P2X7 receptor. Alpine treeline ecotones are harsh environment for tree establishment due to low temperature. Tree establishment at treelines requires favorable climate, suitable microsites, and viable seeds. But most researches have been addressed treeline microclimate and its effects on tree regeneration, the knowledge of seed quantity and quality and its controls on seedling recruitment were limited. We measured seed rain, soil seed bank, seed germination rate and seedling recruitment in natural forests in combination with seed transplanting manipulation to evaluate the controls of seed quantity and quality on seedling recruitment of Abies georgei var. smithii (smith fir) along altitudinal gradient, with focus on treeline ecotone in the Sygera Mountains, southeastern Tibetan Plateau. Both seed quantity and seed quality of smith fir decreased with increasing altitude and was thereby associated with decline in seed germination rate. Seed quantity and seedling recruitment were better in north-facing slope than in south slope. The treeline ecotone above 4200 m appeared as the threshold altitude to sharply decrease seed quality and seedling recruitment. The emergence and overwintering rates of transplanting seeds from 3600−3800 m also went down remarkably above 4200 m at north-facing slope. It also underpins the fact that treeline ecotone is the bottleneck of seedling recruitment. Our results suggest that seed quantity and quality are the principal limitation of treeline upward advance. The purpose of this paper is to present the course design and evaluative data associated with the learning experiences of practicing teachers engaged in a gamified approach to a graduate level course on technology integration. Twenty-two teachers across three offerings of the course completed a survey examining their experience with the gamified course and course elements. Survey mean scores were positive overall. Participants reported they were motivated by the gaming principles incorporated into the course, including the use of badges and awards and the opportunity to tailor course experience to their own interests. Participant responses to open-ended items similarly revealed that recognition and autonomy were important aspects of their learning experience. The objective of the study is to evaluate Homer1 expression in rats with Type 2 diabetes mellitus (T2DM) and investigate the mechanism by which Homer1 influences the pathogenesis of diabetes through study on rat model with decreased Homer1 expression. Rat model of T2DM was constructed and blood insulin concentration was measured. Homer1 mRNA and protein expressions in rat pancreatic tissue were determined using RT-PCR as well as Western blotting. Homer1 expression in human monocytic THP-1 cells was interfered using short hairpin RNA, and its effect on phosphorylation of extracellular signal-regulated kinase (ERK) was assessed. Fasting glucose concentration in rat model of T2DM was significantly higher than that of normal rats (13.1 ± 2.4 vs 5.1 ± 1.1 mmol/L), and fasting blood insulin concentration of diabetic group was significantly lower than that of normal group (13.6 ± 1.9 18.3 ± 2.2 mIU/L) ( P < 0.05). Homer1 mRNA and protein expressions in pancreatic tissue of rats with T2DM were significantly higher than those of normal rats ( P < 0.05). Level of ERK phosphorylation in pancreatic tissue of rats with T2DM was significantly higher than that of normal rats. Homer1 mRNA level in rat pancreatic tissue of T2DM was positively correlated with the area of pancreatic islets ( r = 0.526, P = 0.014). Homer1 mRNA level was significantly inhibited in high-glucose and high-fat stimulated human monotypic THP-1 cells with interfered Homer1. Compared with controls, P-ERK phosphorylation was significantly decreased in THP-1 cells with interfered Homer1 ( P < 0.05). As K-12 schools increase the presence of and access to technology, the role of school librarians is changing. Many librarians have not been prepared by their colleges and universities for the changes demanded by their jobs. In this article, we discuss the skills that 47 school librarians reported having learned on the job as opposed to in librarian preparation programs. More than half indicated that they had to develop technological skills while on the job. This can be explained by technology's influence on society, librarians being early adopters and leaders of technology and technology's ability to enable learning. Because of these technological skills indicated by school librarians, we provide resources and share suggestions for how school librarians can build a Professional Learning Network (PLN) to learn about and with technology.
