High-carbohydrate diets (HCD) can induce the occurrence of nonalcoholic fatty liver disease (NAFLD), characterized by dramatic accumulation of hepatic lipid droplets (LDs). However, the potential molecular mechanisms are still largely unknown. In this study, we investigated the role of autophagy in the process of HCD-induced changes of hepatic lipid metabolism, and to examine the process of underlying mechanisms during these molecular contexts. We found that HCD significantly increased hepatic lipid accumulation and activated autophagy. Using primary hepatocytes, we found that HG increased lipid accumulation and stimulated the release of NEFA by autophagy-mediated lipophagy, and that lipophagy significantly alleviated high glucose (HG)-induced lipid accumulation. Oxidative and endoplasmic reticulum (ER) stress pathways played crucial regulatory roles in HG-induced lipophagy activation and HG-induced changes of lipid metabolism. Further investigation found that HG-activated lipophagy and HG-induced changes of lipid metabolism were via enhancing carbohydrate response element-binding protein (ChREBP) DNA binding capacity at PPARγ promoter region, which in turn induced transcriptional activation of the key genes related to lipogenesis and autophagy. The present study, for the first time, revealed the novel mechanism for lipophagy mediating HCD-induced changes of lipid metabolism by oxidative stress and ER stress, and ChREBP/PPARγ pathways. Purpose: An association between dietary carbohydrate intake and prostate cancer (PCa) prognosis is biologically plausible, but data are scarce. This prospective cohort study examined the relation between pre-diagnostic carbohydrate intake and treatment failure following radical prostatectomy for clinically early-stage PCa.Methods: We identified 205 men awaiting radical prostatectomy and assessed their usual dietary intake of carbohydrates using the 110-item Block food frequency questionnaire. We also evaluated carbohydrate intake quality using a score based on the consumption of sugars relative to fiber, fat, and protein. Logistic regression analyzed their associations with the odds of treatment failure, defined as a detectable and rising serum prostate-specific antigen (PSA) or receiving androgen deprivation therapy (ADT) within 2 years.Results: Sucrose consumption was associated with a higher odds and fiber consumption with a lower odds of ADT after accounting for age, race/ethnicity, body mass index, and tumor characteristics (odds ratio [OR] (95% confidence interval [CI]) 5.68 (1.71, 18.9) for 3rd vs. 1st sucrose tertile and 0.88 (0.81, 0.96) per gram of fiber/day, respectively). Increasing carbohydrate intake quality also associated with a lower odds of ADT (OR (95% CI) 0.78 (0.66, 0.92) per unit increase in score, range 0-12).Conclusions: Pre-diagnostic dietary carbohydrate intake composition and quality influence the risk of primary treatment failure for early-stage PCa. In the present study, three cDNAs of AdipoRs (MpAdipoR1a, MpAdipoR1b, and MpAdipoR2) were identified from juvenile black carp Mylopharyngodon piceus. There were 375, 378, and 356 amino acids in the MpAdipoR1a, MpAdipoR1b, and MpAdipoR2, respectively. BLAST analysis reveals that MpAdipoRs share high identities with other known AdipoRs from zebrafish, rainbow trout, human, etc. And there were all seven transmembrane regions in the amino acid sequences of MpAdipoR1s and MpAdipoR2, respectively. The relative expression levels of MpAdipoR1s were higher in the liver, blood, brain, and eyes in black carp (p < 0.05). Relatively higher expression of MpAdipoR2 was detected in the liver (p < 0.05) and then in the adipose tissues and blood by real-time PCR assays. The relative expression levels of AdipoR mRNA in the liver, muscle, brain, blood, and adipose tissues were detected by real-time PCR in black carp fed with four levels of dietary carbohydrate (CHO) (10.65, 19.43, 28.84, and 37.91%) for 9 weeks, respectively. The expression levels of MpAdipoR1s in the liver, muscle, brain, and blood were induced and reached to the maximum at optimal dietary CHO (19.43 or 28.84%) and then were decreased at 37.91% dietary CHO, although there were no significant differences on the expression levels of MpAdipoR1a in the liver between 19.43, 28.84, and 37.91% dietary CHO. Similarly, MpAdipoR2 were also induced to the maximal levels in the liver, muscle, brain, and blood at optimal dietary CHO (19.43 or 28.84%) compared with that at 10.65% dietary CHO, although no significant differences were observed on the expression levels of MpAdipoR2 in the liver and muscle between groups fed 19.43 and 37.91% dietary CHO (p > 0.05). However, the expression levels of MpAdipoRs in the adipose tissues were significantly downregulated at excessive dietary CHO (37.91%) compared with other groups (p < 0.05). These results indicated that MpAdipoRs were inducible proteins and might enhance the understanding of their vital roles in the regulation of glucose metabolic homeostasis in black carp. Most research on nutritional effects on aging has focussed on the impact of manipulating single dietary factors such as total calorie intake or each of the macronutrients individually. More recent studies using a nutritional geometric approach called the Geometric Framework have facilitated an understanding of how aging is influenced across a landscape of diets that vary orthogonally in macronutrient and total energy content. Such studies have been performed using ad libitum feeding regimes, thus taking into account compensatory feeding responses that are inevitable in a non-constrained environment. Geometric Framework studies on insects and mice have revealed that diets low in protein and high in carbohydrates generate longest lifespans in ad libitum-fed animals while low total energy intake (caloric restriction by dietary dilution) has minimal effect. These conclusions are supported indirectly by observational studies in humans and a heterogeneous group of other types of interventional studies in insects and rodents. Due to compensatory feeding for protein dilution, low-protein, high-carbohydrate diets are often associated with increased food intake and body fat, a phenomenon called protein leverage. Purpose: We compared the effects of a eucaloric moderate-fat diet (18 % protein, 36 % fat, and 46 % carbohydrate), a eucaloric low-fat high-carbohydrate diet (18 % protein, 18 % fat, and 64 % carbohydrate), and a low-calorie (33 % reduced) low-fat high-carbohydrate diet on biomarkers of systemic inflammation. Methods: We randomly assigned 102 participants (age 21-76 years and BMI 19.2-35.5 kg/m) to the three different diets for 6 weeks in a parallel design intervention trial. All foods were provided. Ninety-three participants completed all study procedures; 92 were included in the analyses. Endpoints included plasma C-reactive protein (CRP), interleukin-6 (IL-6), soluble tumor necrosis factor receptors I and II (sTNFRI and II), and adiponectin. Results: In the unadjusted primary analyses, none of the endpoints were differentially affected by the dietary interventions despite the significantly greater reductions in body weight and fat mass in participants consuming the low-calorie low-fat diet compared to the eucaloric diets ( p < 0.001). When including weight change in the model in secondary analysis, adiponectin tended to be increased with weight loss (time × weight change interaction, p = 0.051). Adjusted for weight change, adiponectin was reduced in the groups consuming the low-fat diets relative to the moderate-fat diet ( p = 0.008). No effect of the intervention diets or weight loss on CRP, IL-6, or sTNFRI and II was seen in these secondary analyses. Conclusions: In relatively healthy adults, moderate weight loss had minimal effects on systemic inflammation, and raised plasma adiponectin only modestly. A lower dietary fat and higher carbohydrate content had little impact on measures of systemic inflammation, but reduced adiponectin concentrations compared to a moderate-fat diet. Teleost fishes represent a highly diverse group consisting of more than 20,000 species living across all aquatic environments. This group has significant economical, societal and environmental impacts, yet research efforts have concentrated primarily on salmonid and cyprinid species. This review examines carbohydrate/glucose metabolism and its regulation in these model species including the role of hormones and diet. Over the past decade, molecular tools have been used to address some of the downstream components of these processes and these are incorporated to better understand the roles played by carbohydrates and their regulatory paths. Glucose metabolism remains a contentious area as many fish species are traditionally considered glucose intolerant and, therefore, one might expect that the use and storage of glucose would be considered of minor importance. However, the actual picture is not so clear since the apparent intolerance of fish to carbohydrates is not evident in herbivorous and omnivorous species and even in carnivorous species, glucose is important for specific tissues and/or for specific activities. Thus, our aim is to up-date carbohydrate metabolism in fish, placing it to the context of these new experimental tools and its relationship to dietary intake. Silver sea bream, Sparus sarba, were fed two diets of different carbohydrate levels (2 and 20% dextrin) for 4 weeks, and the effects on organ indices, liver composition, serum metabolite and hormone levels and gene expression profile of key enzymes of carbohydrate metabolism in the liver were investigated. By using real-time PCR, mRNA expression levels of carbohydrate metabolic enzymes including glucokinase (GK, glycolysis), glucose-6-phosphatase (G6Pase, gluconeogenesis), glycogen synthase (GS, glycogenesis), glycogen phosphorylase (GP, glycogenolysis) and glucose-6-phosphate dehydrogenase (G6PDH, pentose phosphate pathway) in liver of sea bream have been examined, and it was found that high dietary carbohydrate level increased mRNA level of GK but decreased mRNA levels of G6Pase and GP. However, mRNA levels of GS and G6PDH were not significantly influenced by dietary carbohydrate. Silver sea bream fed high dietary carbohydrate had higher hepatosomatic index (HSI), liver glycogen and protein, but there were no significant changes in gonadosomatic index (GSI), serum glucose and protein level, as well as liver lipid and moisture level. Pituitary growth hormone (GH) and hepatic insulin-like growth factor I (IGF-I) transcript abundance were assayed by real-time PCR, and it was found that both parameters remained unchanged in fish fed different dietary carbohydrate levels. Serum triiodothyronine (T) and thyroxine (T) were not significantly affected by dietary carbohydrate levels, but lower serum cortisol level was found in fish fed high dietary carbohydrate level. These results suggest that silver sea bream is able to adapt to a diet with high carbohydrate content (up to 20% dextrin), the consumption of which would lead to fundamental re-organization of carbohydrate metabolism resulting in hepatic glycogen deposition. To investigate the effects of different carbohydrate (CHO) levels in the diet of top-mouth culter Erythroculter ilishaeformis Bleeker, fish were randomly divided into five treatment groups. A control group was fed a diet with no CHO, and four treatment groups were fed with 14, 21, 27, and 34 % CHO, respectively, for 70 days. Serum cortisol level, glucose concentration, alanine aminotransferase activity, and malondialdehyde content of fish offered the diet containing 27 % CHO or 34 % CHO were significantly greater than those of fish offered the diet containing 14 % CHO or 21 % CHO. Serum lysozyme activity, total antioxidation capacity, and superoxide dismutase activity of fish offered the diet containing 27 % CHO or 34 % CHO also were significantly lower than those of fish offered the diet containing 14 % CHO. The relative levels of hepatic heat shock cognate protein 70 messenger RNA (mRNA) of fish offered the diet containing 27 % CHO after being deprived of food for 48 h or 34 % CHO at 48 h after feeding were significantly higher than those of fish offered the diet containing 21 % CHO or 14 % CHO. The results of this study suggest that ingestion of 27 or 34 % dietary CHO can impact the nonspecific immune ability of E. ilishaeformis and also cause metabolic stress. [ABSTRACT FROM AUTHOR]

It is recommended that patients with diabetes reduce their intake of saturated fat and increase their intake of monounsaturated fat or carbohydrate. However, high-carbohydrate diets may result in higher saturated fatty acids in VLDL-triacylglycerol. This is attributed to de novo lipogenesis, although synthesis of specific fatty acids is rarely measured. The objective of this study was to examine the contribution of de novo fatty acid synthesis to VLDL-triacylglycerol composition. It was hypothesised that levels of total and de novo synthesised fatty acids would increase with increased carbohydrate intake in diabetic participants. Seven individuals with type 2 diabetes mellitus and seven matched non-diabetic controls consumed two diets differing in fat energy (lower fat <25%, higher fat >35%) for 3 days in a randomised crossover design. Blood samples were drawn before and 24 h after the ingestion of 2H-labelled water. In the control participants, the higher-fat diet resulted in a 40% reduction in VLDL-triacylglycerol fatty acids because of decreases in myristic, palmitic, palmitoleic and linoleic acids, but the opposite trend occurred in participants with diabetes. The lower-fat diet increased the fractional synthesis rate by 35% and 25% in the control and diabetes participants, respectively (range: 0–33%). Palmitate accounted for 71% of fatty acids synthesised (range: 44–84% total de novo synthesised fatty acids). 2H incorporation was used for the first time in humans showing variability in the synthesis rate of specific fatty acids, even palmitic acid. A lower-fat diet stimulated saturated fatty acid synthesis at high rates, but no net stimulation of synthesis of any fatty acid occurred in the diabetes group. Background:It has been proposed that the development of obesity in humans is influenced by the relative proportions of the two major phyla of bacteria (Bacteroidetes and Firmicutes) present in the large intestine.Objective:To examine the relationships between body mass index, weight loss and the major bacterial groups detected in fecal samples.Design:Major groups of fecal bacteria were monitored using fluorescent in situ hybridization (FISH) in obese and non-obese subjects under conditions of weight maintenance, and in obese male volunteers undergoing weight loss on two different reduced carbohydrate weight-loss diets given successively for 4 weeks each.Results:We detected no difference between obese and non-obese individuals in the proportion of Bacteroidetes measured in fecal samples, and no significant change in the percentage of Bacteroidetes in feces from obese subjects on weight loss diets. Two experiments were conducted to determine the capability of the giant gouramy Osphronemus gouramyto assimilate dietary carbohydrate. In experiment 1, fingerlings with an initial body weight of 29-32 g were fed diets containing 20.8, 35.6, 49.8 and 57.0% carbohydrate three times daily, to satiation, for 40 days. In experiment 2, subadults with an initial body weight of 78.7-79.5 g were fed diets containing 21.2, 30.1, 38.6 and 47.5% carbohydrate three times daily, to satiation, for 60 days. The diets had the same amount of protein and the same energy content. The results of experiment 1 showed that the blood glucose levels of fish fed high carbohydrate diets (49.8 and 57.0%) in the 18 h postprandial were lower than those of fish fed low carbohydrate diets (20.8 and 35.6%). The peak of the blood glucose levels in all treatments was found 5 h postprandial. Experiment 2 showed that the blood glucose level of fish was the same among treatments; and the peak of the blood glucose levels was found 9 h postprandially. It was also found that the protein retention, daily growth rate and feed efficiency of the dietary 20.8% carbohydrate levels in experiment 1 were all significantly higher than in other treatments; whereas those in experiment 2 showed no significant difference among the treatments (P > 0.05). In both experiments 1 and 2, lipid retention increased significantly as the dietary carbohydrate level was elevated (P < 0.05). It was concluded that fingerlings of giant gouramy have a lower capability for utilizing dietary carbohydrate, and of the treatments, the 20.8% diet was optimal, whereas subadults could utilize diets with higher carbohydrate levels, as high as 47.5%. The activity and abundance of the Na+/glucose co-transporter (SGLT1) was assessed in brush-border-membrane vesicles (BBMV) isolated from the intestine of grass- and roughage- (GR) consuming ruminants (sheep and dairy cattle), during the transition from the pre-ruminant to the mature ruminant state. The abundance of SGLT1 messenger ribonucleic acid (mRNA) was also compared in the intestinal tissue of the same animals. The dramatic developmental decline in the activity and expression of SGLT1 appears to be typical of GR-consuming ruminants and is coincident with the significant decline in the levels of lumenal monosaccharides. Expression of the ovine SGLT1 complementary deoxyribonucleic acid (cDNA) in Xenopus laevis oocytes confirmed that the isolated cDNA encodes for a functional Na+/glucose co-transporter. Determination of a bovine intestinal SGLT1 protein sequence (amino acids 347–658) indicated 99% similarity to the ovine SGLT1 protein with differences in the carboxyl terminus. In contrast to GR-consuming ruminants, the abundance of SGLT1 protein and SGLT1 mRNA remained significantly high in the intestine of ruminants in both the intermediate-mixed (IM) feeding goat and fallow deer and the concentrate-selecting (CS) moose and roe deer, dietary groups correlating with the availability of monosaccharides in the intestinal lumen. Dietary manipulations involving high carbohydrate feeding increase VI cardiac myosin isoform expression in hormonally deficient rats. The purpose of this study was to determine if extremes in dietary carbohydrate availability could alter cardiac myosin isoform patterns in normal weanling and adult rats. Three and six weeks of dietary manipulations (either high or low carbohydrate diets) failed to change calcium-activated myofibril ATPase activity, calcium regulated myofibril ATPase activity, or the myosin isoform distribution in the adult. In contrast, a four week, high carbohydrate diet reduced calcium activated myosin ATPase activity by 33%, calcium regulated myofibril ATPase activity by 10%, and Vl isoform expression by 66% in weanling rats. Although the low carbohydrate diet caused no change in the myosin ATPase properties, it decreased VI isoform expression by 17%. These results show that carbohydrate availability can alter cardiac myosin isoform expression in normal rats, but only at weanling age. The reason for this age-related contrast in response to dietary manipulations is unknown at this stage. The dietary manipulations may have acted directly on the heart by creating a state of malnutrition, or indirectly, by altering some developmental process which links maturation of the sympathetic nervous system with myosin isoform expression. Weanling male rats were fed a copper-deficient diet devoid of cholesterol. The effects of varying the source of carbohydrate and supplements of copper and zinc on cardiovascular pathology and some biochemical and physiological parameters were investigated. It was found that cardiomyopathy developed in copper-deficient groups. Sucrose, in contrast to starch or starch:lactose (1:1), caused significant exacerbation of this situation. Increasing dietary Cu to 8 ppm prevented or minimized the development of cardiomyopathy. Angiopathy occurred only when dietary zinc was at the lower level (20 ppm). Dietary copper supplements to 8.0 ppm did not alter this situation, but 120 ppm Zn in the drinking water did reduce the angiopathy almost to the control level, except in the groups in which sucrose was fed. Serum cholesterol was only elevated significantly over the control value when dietary copper was deficient and sucrose was the carbohydrate source. The data point to independent action of dietary copper or zinc on the myocardium or vessels, respectively, with sucrose interacting to make copper and zinc supplements less active than when starch or starch/lactose was fed. We have studied levels of glycated haemoglobin in a sample of 223 people aged over 40 years without known diabetes mellitus screened in a community study. Each had a glucose tolerance test and glycated haemoglobin measured by four methods - agar gel electrophoresis with and without removal of Schiff base, affinity chromatography and isoelectric focusing. The correlation coefficients between 2 h blood glucose and levels of glycated haemoglobin were between 0.43 and 0.64. This poor correlation was not explained on the basis of assay or biological variability of either 2 h blood glucose or glycated haemoglobin. Multiple regression analysis showed that other assays of glycated haemoglobin contributed to the variance of any single glycated haemoglobin value by 0.1%-52.9% (median 12.8%) compared to the variance of 18.6%-41.4% (median 30.8%) explained by 2 h blood glucose alone, suggesting that in a non-diabetic population, the degree of glucose intolerance may explain only one third of the variance of glycated haemoglobin levels, but other factors operate to produce consistent changes in levels of glycated haemoglobin. Investigation of 42 subjects with consistently high (20 subjects) or low (22 subjects) levels of glycated haemoglobin relative to their 2 h blood glucose level showed no difference in age, gender, body mass index, haemoglobin levels or smoking, although 50% of low glycators had impaired glucose tolerance. Neither ambient bloodglucose levels, as estimated on two five-point blood-glucose profiles, nor dietary intake of carbohydrate, starch, sugars, fibre or alcohol, explained the difference between high and low glycators. To clarify the separate influences of digestible carbohydrate and of dietary fibre on blood glucose control and serum lipoproteins, 14 diabetic patients (six Type 1 and eight Type 2) were submitted to three weight-maintaining diets for 10 days each: (1) low carbohydrate/low fibre diet with 42% carbohydrate and 20g fibre; (2) high carbohydrate/low fibre diet (carbohydrate 53%, fibre 16g); (3) high carbohydrate/ high fibre diet (carbohydrate 53%, fibre 54 g). In comparison with the low carbohydrate/low fibre diet, the 2-h post-prandial blood glucose and the daily blood glucose profile decreased significantly on the high carbohydrate/high fibre diet, without significant changes during the high carbohydrate/low fibre diet. The diet-induced modifications of blood glucose control were similar in both types of diabetic patients (twoway analysis of variance: F=5.86, p<0.02 for dietary treatment and F=2.09, NS for type of diabetes). Total and lowdensity lipoprotein cholesterol were also decreased after the high carbohydrate/high fibre diet in comparison with the low carbohydrate/low fibre diet ( p<0.001 for both), while they were not significantly modified after the high carbohydrate/ low fibre diet. Again the modifications of low density lipoprotein cholesterol induced by diet were similar in both types of diabetic patients (F=10.02, p<0.005 for dietary treatment and F=0.14 for type of diabetes, NS). High-density lipoprotein cholesterol was lower after the two test diets than after the low carbohydrate/low fibre diet. However statistical significance was found only for the high carbohydrate/high fibre diet ( p<0.005). In conclusion: (1) a simple increase of digestible carbohydrate without a parallel increase of dietary fibre does not help in improving the metabolic control of diabetic patients; (2) the hypoglycaemic and hypolipidaemic effects of high carbohydrate/high-fibre diets are due principally to dietary fibre. The amount of carbohydrate released at 1 and 5 h by digestion in vitro of 2 g carbohydrate portions of 14 foods by human digestive juices was compared with the area under the 2-h blood glucose response curve when 50 g carbohydrate portions were fed to groups of five to ten healthy volunteers. A significant relationship was found between the amounts of sugars and oligosaccharides liberated at 1 and 5 h and the food blood glucose area expressed as a percentage of the blood glucose area for 50 g glucose (r=0.8627 and 0.8618, p<0.001). A significant relationship was also found between the glycaemic index and the food fibre content (p<0.02) and between the glycaemic index and the glucose trapping capacity of the foods (p<0.05). Legumes as a group liberated 56% less sugars and oligosaccharides (p<0.01) than the eight cereal foods over 5 h. It is suggested that such studies in vitro may help to identify food of use for diabetic patients, and at the same time throw further light on factors which affect post-prandial glycaemia.
