Despite research has identified several social resources to promote employees' knowledge sharing behavior, studies on the effect of personal resources are limited. Thus, our research intends to contribute by examining the role of employee mindfulness as an essential personal resource in influencing their knowledge sharing behavior. Based on the conservation of resources (COR) theory, we propose that employee mindfulness can promote employees' knowledge sharing behavior through enhancing their thriving at work, and this effect is moderated by abusive supervision. Using a field study with multi-wave data, we found that employee mindfulness was positively related to employees' knowledge sharing behavior through their thriving at work, and this positive effect was stronger when abusive supervision was high than low. Currently, the precise and detailed anatomical data of the normal uterus, especially the myometrium thickness in various parts of the uterus, are lacking. This study aims to provide normal references for uterine size in healthy reproductive-aged Chinese women to facilitate the application of hysteroscopic surgery. A total of 298 women of reproductive age with normal uterine were included. Parity was significantly correlated with uterine measurements (P < 0.05), and age impacted several measurements (P < 0.05). At each uterine site examined, the myometrium was thinner in nulliparous women than in parous or primiparous women (P < 0.001). Similarly, the extrauterine measurements for parous or primiparous women were larger than those for nulliparous women. Weight affected some external measurements but not myometrial thicknesses, while height did not affect uterine measurements (P > 0.05). There was a positive correlation between body mass index (BMI) and extrauterine measurements as well as myometrial thickness (P < 0.05). The mathematical model of the uterine size for women of reproductive age was constructed stratified by parity. The study is the first to provide a detailed statistical description of the accurate anatomical parameters of the uterus in Chinese reproductive-aged women and has great significance for improving the safety and effectiveness of hysteroscopic surgery for patients. Purpose: A barrier to widespread adoption of chimeric antigen receptor (CAR) T-cell therapy is toxicity. To address this, we recently developed a novel antibody-T-cell receptor (AbTCR) platform (trademarked as ARTEMIS®) which was designed to leverage natural immune receptor signaling and regulation. The AbTCR platform includes a gamma/delta (γδ) TCR-based AbTCR construct and a separate co-stimulatory molecule, both engineered to be tumor-specific. Here, we aim to assess the safety and preliminary efficacy of a CD19-directed AbTCR T-cell therapy. Methods: We generated ET019003 T cells, which are autologous CD19-directed AbTCR T cells. We then conducted an early phase I study to evaluate the safety and preliminary efficacy of ET019003 T cells for the treatment of CD19-positive relapsed/refractory (r/r) B-cell lymphoma. Results: Sixteen patients enrolled in this study and 12 patients were treated. Of the 12 patients treated, 6 patients (50%) achieved a complete response (CR), and 4 (33%) achieved a partial response (PR) (best objective response rate [ORR] of 83%). CRs were durable, including 2 patients with ongoing CRs for 22.7 months and 23.2 months. ET019003 was well-tolerated with an attractive safety profile. No patients experienced severe (grade ≥ 3) cytokine release syndrome (CRS) and only 1 patient experienced immune effector cell-associated neurotoxicity syndrome (ICANS) of any grade. Significant elevations of cytokine levels were not seen, even in patients with marked expansion of ET019003 T cells. Conclusion: This study provides initial clinical validation of the AbTCR platform as a novel cancer treatment with the potential to provide durable clinical benefit with low toxicity. Key message: A new strategy that integrated multiple public data resources was established to construct root gene co-expression network and mine genes regulating root system architecture in maize. A root gene co-expression network, containing 13,874 genes, was constructed. A total of 53 root hub genes and 16 priority root candidate genes were identified. One priority root candidate was further functionally verified using overexpression transgenic maize lines. Root system architecture (RSA) is crucial for crops productivity and stress tolerance. In maize, few RSA genes are functionally cloned, and effective discovery of RSA genes remains a great of challenge. In this work, we established a strategy to mine maize RSA genes by integrating functionally characterized root genes, root transcriptome, weighted gene co-expression network analysis (WGCNA) and genome-wide association analysis (GWAS) of RSA traits based on public data resources. A total of 589 maize root genes were collected by searching well-characterized root genes in maize or homologous genes of other species. We performed WGCNA to construct a maize root gene co-expression network containing 13874 genes based on public available root transcriptome data, and further discovered the 53 hub genes related to root traits. In addition, by the prediction function of obtained root gene co-expression network, a total of 1082 new root candidate genes were explored. By further overlapping the obtained new root candidate gene with the root-related GWAS of RSA candidate genes, 16 priority root candidate genes were identified. Finally, a priority root candidate gene, Zm00001d023379 (encodes pyruvate kinase 2), was validated to modulate root open angle and shoot-borne roots number using its overexpression transgenic lines. Our results develop an integration analysis method for effectively exploring regulatory genes of RSA in maize and open a new avenue to mine the candidate genes underlying complex traits. Polybrominated diphenyl ethers (PBDEs) have been used as brominated flame retardants worldwide and are correlated with extensive environmental pollution and human health concerns. This study is aimed at analyzing the concentrations of PBDEs and at evaluating their temporal trends among a population of blood donors (n = 33) over a 4-year period. A total of 132 serum samples were used for PBDE detection. Nine PBDE congeners were quantified in serum samples by gas chromatography with mass spectrometry (GC–MS). The median concentrations of Σ9PBDEs in each year were 33.46, 29.75, 30.85, and 35.02 ng/g lipid, respectively. Most of the PBDE congeners showed a downward trend from 2013 to 2014 and then increased after 2014. No correlations between age and PBDE congener concentrations were observed, while concentrations of each congener and Σ9PBDE were nearly always lower in females than in males, especially in BDE-66, BDE-153, BDE-183, BDE-190, and Σ9PBDE. We also found that the intake of fish, fruit, and eggs in the daily diet was related to the exposure level of PBDEs. Our results suggest that, as deca-BDE is still produced and used in China, diet is an important exposure pathway for PBDEs, and follow-up studies will be required to improve our understanding of the behaviors of PBDE isomers in humans and the exposure levels. While it is known that exposure to disinfection by-products (DBPs), including trihalomethanes (THMs), impairs liver function, few epidemiological studies have explored this association. Here, we determined the concentrations of four urinary trihalomethanes (chloroform [TCM], and three Br-THMs, bromodichloromethane [BDCM], dibromochloromethane [DBCM], and bromoform [TBM]), and nine serum liver function indicators in 182 adults ≥ 18 years of age, examined at a medical examination center in Wuxi, China, in 2020 and 2021. Generalized linear model analysis revealed positive associations between urinary DBCM and alanine aminotransferase (ALT), aspartate aminotransferase (AST), total bilirubin (TBIL), total protein (TP), and albumin (ALB). Urinary Br-THMs and total THMs (TTHMs) were positively associated with ALT, AST, TBIL, indirect bilirubin (IBIL), TP, and ALB (all P < 0.05). Urinary THMs were not associated with alkaline phosphatase (ALP) or glutamine transaminase (GGT) (all P > 0.05). Generalized additive model–based penalized regression splines were used to confirm these associations. In conclusion, THM exposure was associated with altered serum biomarkers of liver function. Background: Lidocaine administered through the working channel of a flexible bronchoscope can provide effective local anesthesia but cannot achieve good distribution in the airway. This study was undertaken to determine whether lidocaine delivered via a multi-orifice epidural catheter (three orifices/openings) is superior to conventional method and if a better distribution and decreased the cough reflex can be achieved. Methods: The patients (N = 100; 50 in each group) were randomized to receive either topical airway anesthesia by the "spray-as-you-go" technique via conventional application (group C) through the working channel of the bronchoscope or via a triple-orifice epidural catheter (group E). The primary outcome measurement was the cough severity, which was documented using a 4-point scale. Bronchoscopists and nurses assessed the coughing. The visual analogue scale (VAS) score for cough, total consumption of propofol and lidocaine, requirement frequency of propofol and topical anesthesia, PACU retention time, and adverse events were also compared. Results: There was a significant difference in the median cough severity scores between the two groups (group C: 3 vs. group E: 2, P = 0.004). The median visual analogue scale (VAS) scores for the cough, were significantly higher in group C than those in group E (bronchoscopist: 3 vs. 2 P = 0.002; nurse: 3 vs. 2, P < 0.001). The incidence of cough was significantly higher in group C in the trachea, left and right bronchi. The highest respiratory rate was higher in group C than in group E (P < 0.01). Eight patients in group C and two patients in group E had an oxygen saturation below 90% during flexible bronchoscopy(FB) (P = 0.046). More patients in group C required extra topical anesthesia than in group E (P < 0.001). The total lidocaine consumption was also higher in group C than that in group E (P < 0.001). Conclusions: Endotracheal topical anesthesia via the multi-orifice epidural catheter (three holes/openings) during flexible bronchoscopy using the "spray-as-you-go" technique was appeared to be superior to the conventional method. Characterising and understanding the plasma properties of a rf-coupled electric propulsion device is crucial during testing, qualification and development. Therefore, the optimization of existing diagnostic systems as well as the development of new ones is an important area of electric propulsion research. Here, we present an approach to non-invasively determine the plasma parameters of an operating radio-frequency ion-thruster. For this purpose, a correlation between non-invasive optical emission (OE) spectroscopy and intrusive Langmuir probe diagnostics measurements is established for a reference system. Both types of measurements are performed simultaneously for a wide range of operation points yielding a large reference data set. Based on a principal component analysis (PCA), a correlation between plasma parameters and corresponding OE spectra at different operational points is established. This correlation can then be applied to OE spectra of the plasma of an operating thruster to obtain non-invasively the corresponding plasma parameters, i.e., without having to employ intrusive Langmuir probes. This approach for evaluating optical spectroscopic data in terms of plasma parameters has no need for a theoretical microscopic modeling of the plasma. This makes this approach very versatile and easily transferable to cases where other propellants are used, since no knowledge of excitation cross sections or transition matrix elements and other microscopic parameters of the species of the plasma is required. Excess amount of nitrogen in wastewater has caused serious concerns, such as water eutrophication. Paracoccus pantotrophus MA3, a novel isolated strain of heterotrophic nitrification–anaerobic denitrification bacteria, was evaluated for nitrogen removal using formic acid as the sole carbon source. The results showed that the maximum ammonium removal efficiency was observed under the optimum conditions of 26.25 carbon to nitrogen ratio, 3.39% (v/v) inoculation amount, 34.64 °C temperature, and at 180 rpm shaking speed, respectively. In addition, quantitative real-time PCR technique analysis assured that the gene expression level of formate dehydrogenase, formate tetrahydrofolate ligase, 5,10-methylenetetrahydrofolate dehydrogenase, serine hydroxymethyltransferase, respiratory nitrate reductase beta subunit, L-glutamine synthetase, glutamate dehydrogenase, and glutamate synthase were up-regulated compared to the control group, and combined with nitrogen mass balance analysis to conclude that most of the ammonium was removed by assimilation. A small amount of nitrate and nearly no nitrite were accumulated during heterotrophic nitrification. MA3 exhibited significant denitrification potential under anaerobic conditions with a maximum nitrate removal rate of 4.39 mg/L/h, and the only gas produced was N2. Since late 2019, the outbreak of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and the resultant spread of COVID-19 have given rise to a worldwide health crisis that is posing great challenges to public health and clinical treatment, in addition to serving as a formidable threat to the global economy. To obtain an effective tool to prevent and diagnose viral infections, we attempted to obtain human antibody fragments that can effectively neutralize viral infection and be utilized for rapid virus detection. To this end, several human monoclonal antibodies were isolated by bio-panning a phage-displayed human antibody library, Tomlinson I. The selected clones were demonstrated to bind to the S1 domain of the spike glycoprotein of SARS-CoV-2. Moreover, clone A7 in Fab and IgG formats were found to effectively neutralize the binding of S protein to angiotensin-converting enzyme 2 in the low nM range. In addition, this clone was successfully converted to quench-based fluorescent immunosensors (Quenchbodies) that allowed antigen detection within a few minutes, with the help of a handy fluorometer. This study uses organizational support theory and broaden-and-build theory to examine the antecedents and consequences of workplace gratitude. Specifically, we explored the moderating role of job satisfaction and the mediating role of gratitude at work, including team error management climate as a predictor and OCBs and creativity as outcome variables. Using multiphase, multisource, and multilevel data of 229 employees from 33 teams of two architectural companies located in the east of China, we found that team error management climate is conducive to enhancing employees' workplace gratitude, and that increased workplace gratitude then stimulates more organizational citizenship behaviors (OCBs) and creativity among employees. The positive effect of team error management climate on workplace gratitude and the indirect effect of team error management climate on OCBs and creativity are stronger for employees with lower job satisfaction. With the increase in population growth and environmental pollution, the daily protein supply is facing great challenges. Single-cell protein (SCP) produced by microorganism fermentation is a good alternative for substituting plant- and animal-derived proteins. In this study, Paracoccus communis MA5 isolated from soil previously demonstrated an excellent ability to synthesize SCP directly from sodium formate. To investigate the central metabolic network of formic acid assimilation and protein synthesis, genome-scale analyses were performed. Genomic analysis showed that complete tetrahydrofolate cycle-, serine cycle-, glycolytic pathway-, tricarboxylic acid (TCA) cycle- and nitrogen metabolism-relevant genes were annotated in the genome. These pathways play key roles in the conversion of formic acid into proteins. Transcriptional analysis showed that sodium formate stress could stimulate the metabolic pathway in response to environmental stress, but weaken the sulfur metabolic pathway to inhibit amino acid synthesis, resulting in a decrease in protein content (30% vs 44%). However, under culture conditions with ammonium sulfate, metabolic pathways associated with protein synthesis were accelerated, causing an increase in protein content (53% vs 44%); while the tetrahydrofolate cycle associated with formic acid assimilation was inhibited, causing a 62.5% decrease in growth rate (OD600: 0.21 vs 0.56). These results provide evidence of protein synthesis from sodium formate in strain MA5 at the gene level and lay a theoretical foundation for the optimization of fermentation systems using formic acid as a carbon source. We present a non-invasive approach for determining plasma parameters such as electron temperature and density inside a radio-frequency ion thruster (RIT) using optical emission spectroscopy (OES) in conjunction with principal component analysis (PCA). Instead of relying on a theoretical microscopic model of the plasma emission to extract plasma parameters from the OES, an empirical correlation is established on the basis of conducting simultaneous OES and Langmuir diagnostics. The measured reference spectra are simplified and a PCA is performed. The PCA results are correlated with the plasma parameters of the Langmuir measurements yielding a one-to-one correspondence. This correlation allows us to derive the plasma parameters by analysis of a non-invasively determined emission spectrum without additional Langmuir measurements. We show how the plasma parameters can be calculated from OES measurements using this correlation. Under the assumption that the electronic system thermalizes on much shorter time scales than the period of the RF signal driving the thruster, we can also use time-resolved spectral data to determine the time evolution of plasma parameters. Key message: Sunlight enhanced peel color and significantly up-regulated the expression of PyMYB10 and PybHLH genes. MYB-bHLH-WD40 transcriptional complex forms in the light and is involved in regulating anthocyanin accumulation in the peel. Anthocyanin is the major pigment in the peel of Yunnan red pear (Pyrus pyrifolia (Burm.) Nak.). A transcriptional activation protein complex, involving members of the transcription factor classes of MYB, bHLH and WD40, regulates anthocyanin biosynthesis. This complex was examined in the peel of red pear. In order to clarify the interaction of PyMYB10, PybHLH and PyWD40, fruit were bagged then peel samples collected 0, 3, 5, and 7 days after bag removal. Samples were used for Western blotting and protein interaction analysis. The results showed that sunlight enhanced peel color and significantly up-regulated the expression of both PyMYB10 and PybHLH genes. Co-immunoprecipitation (Co-IP) analysis showed that PybHLH interacted with PyMYB10 or PyWD40, and PyMYB10 interacted with PyWD40. Using onion cells as a model system, bimolecular fluorescence complementation (BiFC) confirmed these interactions and showed that the interaction localized to the nuclei. GST Pull down and Far-Western blotting assays demonstrated that PybHLH interacted with PyMYB10 or PyWD40, respectively, and PyMYB10 interacted with PyWD40 in vitro. In addition, EMSA assay showed that PyMYB10 can directly bind to the promoter of the gene encoding the anthocyanin biosynthesis enzyme anthocyanidin synthase (PyANS). Taken together, these results showed that the ternary complex of PyMYB10, PybHLH and PyWD40 transcription factors forms to regulate anthocyanin biosynthesis and accumulation in Yunnan red pear. High-power fiber laser has been proven to be feasible for cutting carbon fiber reinforced polymers with several advantages including noncontact force, high efficiency and flexibility, while the characteristics of thermal damage and heat conduction in materials are not yet fully understood. Continuous-wave fiber laser was applied in this work to cut 2.0-mm-thick carbon fiber reinforced polymer laminates with different layup configurations. The influence of processing parameters including laser power and cutting speed on thermal damage was investigated. The characteristics of various thermal defects on different positions of machined surface were analyzed using high-resolution SEM and mathematical models. Interestingly, swollen fibers were observed and they connected together to form irregular swollen masses. According to further analysis on the initial heat distribution, it showed that cutting speed was the main factor affecting heat accumulation. In addition, modified heat conduction model was developed to analyze heat transfer within unidirectional carbon fiber reinforced polymer laminates in comparison with experimental results, which can be applied to predict heat affect zone during high-power fiber laser cutting composite materials. In the present study, the root zone of Canna flaccida Salisb. was selected as the biological component. We found that the adsorption rate of formaldehyde (FA) by C. flaccida was initially low, before increasing substantially. 13CNMR analysis indicated that C. flaccida can metabolize H13CHO and the major metabolic products were organic acid and amino acids. On this basis, we had designed a suspended growth bioreactor (SGB) to remove FA. An SGB was tested under a series of different conditions and the results showed that FA removal efficiency was strongly influenced by water volume, root zone weight, and gas flow rate. More than 52% of the FA was removed in 1 h and the maximum elimination capacity reached 35.112 mg m−3 h−1. After 24 h, no FA remained in the SGB. Running for 15 days, the concentration of FA in the lab space decreased from 1.12 to 0.82 mg m−3, and the concentration of FA in the SGB reached a low level. We explored whether polybrominated diphenyl ethers (PBDEs) exposed in cord blood could have any potential relationship with perinatal outcomes. Participants were pregnant females (n = 222) who were recruited from a prospective birth cohort (Laizhou Wan Birth Cohort, LWBC) between September 2010 and February 2012. We measured eight PBDE congeners (BDE-28, BDE-47, BDE-85, BDE-99, BDE-100, BDE-153, BDE-154, and BDE-183) in cord serum and examined their relationship with perinatal outcomes. The median levels of BDE-28, BDE-47, BDE-85, BDE-99, BDE-100, BDE-153, BDE-154, and BDE-183 were 2.92, 3.93, 2.29, 7.03, 3.03, 3.14, 1.46, and 2.55 ng/g lipids, respectively. For each log unit increase in BDE-47, BDE-100, and ∑4PBDEs, gestational age increased by 0.70 weeks (95% confidence interval [CI] 0.25, 1.15), 0.48 weeks (95% CI 0.03, 0.94), and 0.73 weeks (95% CI 0.12, 1.34), respectively. We also found that BDE-47 was positively associated with head circumference (β = 0.42, 95% CI 0.00, 0.84). Given that our study area is one of the major brominated flame retardant production areas in China, and the cord PBDEs levels were relatively higher than those reported in most other Asian areas, more studies on the effects of in utero PBDE exposure on fetal growth and child development are warranted. Main conclusion: Over-expression of AtFDH controlled by the promoter of Rubisco small subunit in chloroplasts increases formaldehyde uptake and metabolism in tobacco leaves. Our previous study showed that formaldehyde (HCHO) uptake and resistance in tobacco are weaker than in Arabidopsis. Formate dehydrogenase in Arabidopsis (AtFDH) is a key enzyme in HCHO metabolism by oxidation of HCOOH to CO, which enters the Calvin cycle to be assimilated into glucose. HCHO metabolic mechanism in tobacco differs from that in Arabidopsis. In this study, AtFDH was over-expressed in the chloroplasts of transgenic tobacco using a light inducible promoter. C-NMR analysis showed that the carbon flux from HCHO metabolism was not introduced into the Calvin cycle to produce glucose in transgenic tobacco leaves. However, the over-expression of AtFDH significantly enhanced the HCHO metabolism in transgenic leaves. Consequently, the productions of [4-C]Asn, [3-C]Gln, [U-C]oxalate, and HCOOH were notably greater in transgenic leaves than in non-transformed leaves after treatment with HCHO. The increased stomatal conductance and aperture in transgenic leaves might be ascribed to the increased yield of oxalate in the guard cells with over-expressed AtFDH in chloroplasts. Accordingly, the transgenic plants exhibited a stronger capacity to absorb gaseous HCHO. Furthermore, the higher proline content in transgenic leaves compared with non-transformed leaves under HCHO stress might be attributable to the excess formate accumulation and Gln production. This study evaluated the clinical and cost-effectiveness of prophylactic use of fluconazole versus mould-active triazoles (voriconazole and posaconazole) in adult patients with acute lymphoblastic leukemia (ALL). A decision analytical model was developed with inputs from a 7-year retrospective study (2009-2016) of 103 consecutive adult patients with ALL who received antifungal prophylaxis. Information on the administration of antifungal agents, clinical outcomes, and costs were collected. One-way sensitivity analyses and probabilistic sensitivity analysis were performed. The mould-active triazoles group was associated with higher life-years (3.71 vs 3.59) and lower total costs (US$4886 vs US$5722) per patient compared with fluconazole. One-way sensitivity analyses revealed that varying all of the key variables in the model did not affect the robustness of the results. Probabilistic sensitivity analysis demonstrated that mould-active triazoles had a probability of 77.1 and 90.1% of providing a dominant and cost-effective option relative to fluconazole, respectively. Mould-active triazoles should be regarded as preferable to fluconazole as the first-line prophylactic for adult patients with ALL accompanied by uncommon severe vinca alkaloid-induced neurotoxicity. Key message: AtPrx64 is one of the peroxidases gene up-regulated in Al stress and has some functions in the formation of plant second cell wall. Its overexpression may improve plant tolerance to Al by some ways. Studies on its function under Al stress may help us to understand the mechanism of plant tolerance to Al stress. Abstract: In Arabidopsis thaliana, the expressions of some genes ( AtPrxs) encoding class III plant peroxidases have been found to be either up-regulated or down-regulated under aluminum (Al) stress. Among 73 genes that encode AtPrxs in Arabidopsis, AtPrx64 is always up-regulated by Al stress, suggesting this gene plays protective roles in response to such stress. In this study, transgenic tobacco plants were generated to examine the effects of overexpressing of AtPrx64 gene on the tolerance to Al stress. The results showed that overexpression of AtPrx64 gene increased the root growth and reduced the accumulation of Al and ROS in the roots. Compared with wild type controls, transgenic tobaccos had much less soluble proteins and malondialdehyde in roots and much more root citrate exudation. Phosphatidylcholine enriched with docosahexaenoic acid (DHA) was successfully produced by phospholipase A1-catalysed acidolysis. Reverse micelles were firstly selected as the reaction system to avoid the process of immobilization and to ensure the efficient catalysis of phospholipase A1. Parameters were optimized for a high incorporation of phosphatidylcholine enriched with DHA (DHA-PC). A response-surface design with four factors, including the water content, enzyme loading, pH and substrate-mass ratio were used to evaluate the influence of the major factors and to predict the optimal reaction conditions. The results indicated that the optimal reaction conditions for the production of DHA-PC were a water content of 0.4%, an enzyme loading of 40%, pH 6.92 and a substrate-mass ratio of 2.13. As a major antioxidant in plants, ascorbic acid (AsA) plays a very important role in the response to aluminum (Al) stress. However, the effect of AsA on the mitigation of Al toxicity and the mechanism of nitrate nitrogen (NO -N) uptake by plants under Al stress are unclear. In this study, a hydroponic experiment was conducted using peak 1 A rice (sterile line, Indica) with weaker resistance to Al and peak 1 superior 5 rice (F1 hybrid, Indica) with stronger resistance to Al to study the effects of exogenous AsA on the physiological and biochemical responses to NO -N uptake by rice roots exposed to 50 μmol L Al. Al stress induced increases in the concentrations of HO and malondialdehyde (MDA) and in the activities of antioxidant enzymes [such as superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), and ascorbate peroxidase (APX)]. Plasma membrane (PM) H-ATPase and H-pump activities, endogenous AsA content and NO -N uptake in rice roots decreased under Al stress. After treatment with 2 mmol L exogenous AsA combined with Al, concentrations of HO and MDA in roots notably decreased, and endogenous AsA content and activities of SOD, POD, CAT, and APX in rice roots increased significantly; furthermore, the interaction of PM H-ATPase and the 14-3-3 protein was also enhanced significantly compared with that in control plants without AsA treatment, which clearly increased NO -N uptake. Based on all of these data, the application of AsA significantly reduced the accumulation of HO and MDA and increased the activities of PM H-ATPase and the H-pump by increasing the endogenous AsA content, the antioxidant enzyme activities, and the interaction of PM H-ATPase and the 14-3-3 protein in the roots of the two rice cultivars under Al stress, thereby improving the uptake of NO -N in rice. Although MeHg is a confirmed neurotoxin at sufficiently high dosages, the effect of prenatal low-level Hg exposure on infant neurodevelopment is still unknown. Four hundred ten mother-infant pairs were recruited in rural northern China from September 2010 to September 2012. We investigated maternal diet during pregnancy, collected maternal and umbilical blood samples, and tested infant neurodevelopment using Gesell developmental schedules (GDS) at the age of 1 year old. The geometric means (GM) of Hg exposure in maternal and umbilical blood were 0.72 and 1.2 μg/L, respectively. Positive associations between Hg levels and the frequency of total fish consumption were found (Spearman's correlation, r = 0.09 for maternal Hg, r = 0.14 for umbilical Hg, p < 0.01). After adjusting for confounding factors, we found a log-unit increase in umbilical blood Hg levels was associated with a 4.22-point (95 % confidence interval (95 %CI) 0.77 to 7.67) increase in the adaptive domain and a 4.06-point (95 %CI 0.51 to 7.62) increase in the social domain. No significant association has been found between other domains and Hg levels in our study. In further analysis, we noticed the positive correlation between the developmental quotient (DQ) scores of the adaptive domain and frequencies of total fish consumption ( p = 0.04). Our results failed to indicate the adverse effects between prenatal Hg exposure and infant neurodevelopment. However, given the low-level Hg exposure and its predominant source, the potential positive associations in our study may be attributed to the beneficial nutrients of fish consumption. Although pyrethroid insecticides are widely used, little is known about potential adverse effects on fetal growth. Participating 454 mother-infant pairs were recruited from a prospective birth cohort in rural northern China between September 2010 and 2012. We measured five non-specific pyrethroid metabolites in maternal urine at delivery and examined their association with birth outcomes including birth weight, length, head circumference, and gestational duration. The creatinine-adjusted medians of pyrethroid metabolites in urine were 0.51 μg/g for cis-DCCA, 0.65 μg/g for trans-DCCA, and 0.68 μg/g for 3-PBA. The pregnant women had substantially higher levels of urinary pyrethroid metabolites compared with those reported in developed countries. A increase in total (the sum of cis-DCCA, trans-DCCA, and 3-PBA) but not individual urinary metabolite levels was associated with a decrease in birth weight (adjusted β=-96.76 g per log10 unit increase, 95% confidence interval=-173.15 to -20.37). No associations were found between individual or total metabolite levels and birth length, head circumference, or gestational duration. We report an adverse association of prenatal exposure to pyrethroids as measured by urinary metabolites with birth weight. Arsenic trioxide (ATO) combined with dexamethasone, melphalan or other cytostatic agents had been used to treat refractory or relapsed multiple myeloma (MM) patients. We assessed the safety and efficacy of ATO combined with vindesine/cyclophosphamide/melphalan/prednisone (VCMP) or vindesine/doxorubicin/dexamethsone (VAD) chemotherapy for MM patients who failed more than two different prior regimens. All patients received ATO (0.25 mg/kg day) for 10 days/cycle combined with VCMP or VAD in 30-day cycles. Vindesine (1.4 mg/m) was given intravenously on day 1, cyclophosphamide (400 mg/m day) was given intravenously on days 2, 4, 6, 8, 10, melphalan (6 mg/day) and prednisone (1 mg/kg day) were given orally day 1 to day 10 for VCMP regimen. VAD regimen consisted of vindesine 1 mg/day and doxorubicin 10 mg/day intravenously drip for 4 days with oral dexamethasone 40 mg/day for days 1-4, 9-12, 17-20. Patients who completed at least one cycle were evaluated for response to treatment. Objective responses occurred in 35 of 63 (56 %) patients, including seven complete, 14 partial and 14 minor responses. Median progression-free survival and overall survival were 6 and 23 months respectively. 12 patients had elevated serum creatinine levels (SCr) at baseline, and 9 of 12 (75 %) showed decreased SCr levels during treatment. Frequent Grade 3/4 non-hematological adverse events included arrhythmia, hypertension, fatigue and neuropathy. Manganese (Mn) is an essential mineral nutrient in mammals. The physiological role of Mn in animal models is well documented, but little is known about the adverse effects of Mn deficiency or overexposure in humans, including pregnancy outcomes such as birth weight. We examined the relationship of the maternal and cord blood Mn levels with birth weight in a cohort of 172 mother-infant pairs born in Shanghai, China. Non-linear spline and quadratic regression models were used to test the hypothesis of an inverted U-shaped association between the Mn levels and birth weight. The median (range) levels of Mn in the maternal and cord blood were 5.38 (2.34-30.37) μg/dL and 7.66 (2.57-34.23) μg/dL, respectively. An inverted U-shaped relationship was observed between maternal Mn and birth weight after adjusting for potential confounders. The birth weight increased with Mn levels up to 4.18 μg/dL, and a slight reduction in weight was observed at higher levels. The cord blood Mn levels were not found to be associated with birth weight. Both lower and higher Mn exposures are associated with lower birth weight, which may influence important developmental parameters; the association of higher Mn levels with lower weight was weak and imprecise. A foliar spray of 5 % (v/v) methanol had better effects on the growth of the Tamba black soybean (TBS) in an acidic soil (pH 4.6) than in a neutral soil (pH 7.1). To better understand the molecular mechanisms behind the methanol-enhanced growth of the TBS under acidic soil conditions, the Affymetrix GeneChip Soybean Genome was used to identify methanol-responsive genes in TBS leaves sprayed with 5 % methanol for 24 h. The results showed that a total of 516 transcripts (including 190 upregulated genes and 326 downregulated genes) were significantly regulated (at least two fold) by methanol. RT-PCR analysis verified that the microarray data were reliable and reproducible. The expression of six POD and other antioxidant-related genes was upregulated in methanol-treated TBS leaves. This might be an important molecular mechanism through which methanol application ameliorated the effects of acidic soil stress on TBS growth. Methanol also induced the expression of three CaMs, which might be involved in the regulation of TBS growth and development through interactions with various clients. The expression of two gibberellin-related genes and two auxin-related genes was also upregulated by methanol. Changes in the expression of these genes might alter the distribution and content of gibberellin and auxin in TBS plants, thereby enhancing the growth of stem and leaves. RT-PCR analysis confirmed that the expressions of the selected photosynthesis-associated genes were all upregulated during the methanol treatment period. Consistent with these results, the photosynthesis rate was also obviously elevated in methanol-treated TBS leaves. Nitrogen metabolism is essential for plant growth and development. The glutamine synthetase (GS) enzyme and the Dof transcription factor play important roles in the regulation of nitrogen metabolism. Using expression vectors, transgenic tobacco lines overexpressing each of Dof1 ( Dof1.7), GS1 ( GLN1;4) and GS2 were developed. cDNA clones of each of these transgenes were derived from Arabidopsis thaliana, and each transgene was under the control of the Rubisco small subunit promoter, PrbcS-a leaf-specific light-inducible promoter. Under low-nitrogen conditions, plant length, leaf surface, total protein, chlorophyll, as well as sucrose and glucose contents in leaves of transgenic tobacco lines were higher than those of wild-type plants. In addition, transgenic plants exhibited increased activities of enzymes involved in carbon and nitrogen metabolism, such as GS, nitrate reductase (NR), phosphoenolpyruvate carboxylase (PEPC) and pyruvate kinase (PK). The contents of most amino acids were significantly higher in transgenic plants than those of wild-type plants, whereas levels of nitrate, malic acid, and citric acid were significantly lower than those of wild-type plants. These findings suggest that Dof1 and GS work together to regulate the nitrogen metabolic pathway in plants, and this enhances nitrogen assimilation in transgenic tobacco plants grown under low-nitrogen conditions. In this study, the effects of Al stress on the activity and transcription of antioxidant enzymes were investigated in an acid-resistant black soybean (RB) and an acid-sensitive black soybean (SB) under hydroponic conditions to further clarify the role of antioxidant enzymes in the plant's response to Al stress. The results indicated that oxidative stress was induced in the roots and leaves of RB and SB and that the stress level was higher in SB than in RB. Changes in the catalase (CAT) activity in response to Al stress occurred faster in RB roots and leaves than in SB. As the duration of Al stress increased, the peroxidase (POD) activity was enhanced more pronouncedly in RB roots and leaves than in SB. The activity of superoxide dismutase (SOD) in the roots and leaves of RB and SB was not responsive to Al stress. A high transcription level of a selected POD gene was detected in RB leaves, but no transcription of this POD gene was observed in SB leaves under Al stress. Moreover, the transcription level of this POD gene was higher in RB roots than in SB roots. Under Al stress, the transcription of two selected SOD genes showed an increasing trend in RB but decreased in SB. Furthermore, the transcription levels of these two selected SOD genes were always higher in RB than in SB. The above results suggest that not only does RB have a higher level of antioxidant enzyme activities but also that antioxidant enzyme genes can be upregulated by Al stress. Phosphoenolpyruvate carboxylase (PEPC) and citrate synthase (CS) are two key enzymes in organic acid synthesis metabolism. In the present study, a cytoplasmic form of CS from tobacco and a mutant (with reduced sensitivity to organic acid inhibition) PEPC from Synechococcus vulcanus were overexpressed simultaneously using a light-inducible promoter in tobacco leaves. The analysis for enzyme activity showed that CS and PEPC enzyme activities were increased by 235% to 257% and 218% to 236% in the selected cs and pepc (double-gene) overexpression lines, respectively, compared with those in the wild-type plants (WT). The measurement for the relative root elongation rate of the tobacco plants exposed to 30 μM aluminum (Al) indicated that Al tolerance in the double-gene overexpression lines was stronger than that of the transgenic cs or pepc lines and WT plants. The C-NMR analysis with NaHCO showed that overexpression of CS and PEPC in the transgenic tobacco successfully constructed a new citrate synthesis pathway. Under the conditions with Al stress, the amount of citrate secreted from the double-transgenic tobacco roots was the largest among the tested plants. When grown on sandy soil supplied with a nutritional solution containing 500 μM Al, the growth of the double-transgenic tobacco was better than that of the transgenic cs or pepc tobacco and WT, and their root biomass was the highest among the tested plants. These results demonstrated that construction of a new citrate synthesis pathway by simultaneous overexpression of CS and PEPC in the cytoplasm of transgenic plant leaves could enhance Al resistance in plants. To better understand the molecular basis of plant responses to nitrate stress, suppression subtractive hybridization (SSH) was used to identify the potential important or novel genes involved in the early stage of spinach responses to nitrate stress. Complementary DNAs (cDNAs) of 15 min, 1 h, 2 h, 6 h and 24 h of 160 mM nitrate stress treatment seedlings were used as tester, and cDNAs of normal nutrient solution seedlings of the same time were used as driver. The SSH analysis identified 189 non-redundant putative nitrate stress responsive cDNAs out of 798 clones. These ESTs were categorized into 12 functional groups, with the largest group of genes involved in metabolism, followed by the group of genes related to cell rescue, defense and virulence. Reverse transcriptase (RT)-PCR was conducted for several genes, confirming the induction by nitrate stress. The results indicated that osmolyte biosynthesis genes, reactive oxygen scavengers, transporters, signaling components and transcription factor played important roles in fighting against nitrate stress. Aluminum (Al) toxicity is a major factor that limits crop productivity in acid soils. The toxic effects of Al exposure on Al-tolerant Cryptococcus humicola were analyzed at the physiological level. The exposure to 20 mM Al led to a clear increase in malondialdehyde content and a significant decrease in the levels of protein carbonyls, suggesting that Al stress caused oxidative damage to membrane lipids but not to proteins. Suppression subtractive hybridization (SSH) results showed that when C. humicola was exposed to 20 mM Al, a total of 141 ESTs were differentially expressed. Of them, 27 had unknown functions and 48 were newly identified in this study. The genes with known functions were categorized into seven groups. The largest group was related to metabolism and energy, followed by protein synthesis and processing, cell structure, signal transduction and transcription, transporters, stress and defense. Reverse transcription (RT)-PCR analysis of certain genes was performed to confirm the reliability of the SSH data. Nine selected genes were upregulated by Al in a time-dependent manner. The potential functions of some genes involved in Al-tolerance were predicted and are discussed. The color of fruit skin is an important quality parameter, and in many plants, it is the result of coordinative regulation of the anthocyanin pathway. To characterize the mechanism involved in fruit peel coloration of Yunnan red pear ( Pyrus pyrifolia), we constructed a subtractive cDNA library using the suppression subtractive hybridization (SSH) technology. cDNA of red peel exposed to sunlight (for 2, 4, 6, and 8 days) was subtracted from that of white skin unexposed to sunlight. Over 100 differentially expressed ESTs were obtained, putatively involved in primary and secondary metabolism, stress, and defense response. Expression analysis using semiquantitative reverse transcription polymerase chain reaction (RT-PCR) for 13 genes was performed with two pear cultivars, light-skinned 'Zaobaimi' and red-skinned 'Yunhong-1', which had been bagged and then exposed to sunlight for 0, 1, 2, 3, 5, and 7 days before harvest. This analysis showed that genes encoding for a metallothionein-like protein and a NADP-malic acid enzyme were constitutively expressed, whereas other selected genes were either down- or up-regulated. Semiquantitative RT-PCR analysis for 7 anthocyanin biosynthetic pathway genes and 3 putative regulatory genes was also performed. Results showed that an R2R3 MYB transcription factor PyMYB10 was up-regulated in both the less-colored pear 'Zaobaimi' and well-colored red pear Yunhong-1 after the bag was removed, but that kinetics differed between cultivars. Other anthocyanin-related genes appeared to be coordinately regulated by the MYB-bHLH-WD40 complex. DFR and ANS genes seemed to be limiting factors for the peel coloration of less-colored pear 'Zaobaimi', while all biosynthetic steps are up-regulated by 7 days after bag removal in red fruit. Increasing evidences have revealed a close interaction between the intestinal microbes and host growth performance. The shrimp (Litopenaeus vannamei) gut harbors a diverse microbial community, yet its associations with dietary, body weight and weaning age remain a matter of debate. In this study, we analyzed the effects of different dietary (fishmeal group (NC), krill meal group (KM)) and different growth stages (age from 42 day-old to 98 day-old) of the shrimp on the intestinal microbiota. High throughput sequencing of the 16S rRNA genes of shrimp intestinal microbes determined the novelty of bacteria in the shrimp gut microbiota and a core of 58 Operation Taxonomic Units (OTUs) was present among the shrimp gut samples. Analysis results indicated that the development of the shrimp gut microbiota is a dynamic process with three stages across the age according to the gut microbiota compositions. Furthermore, the dietary of KM group did not significantly change the intestinal microbiota of the shrimps compared with NC group. Intriguingly, compared to NC group, we observed in KM group that a fluctuation of the shrimp gut microbiota coincided with the shrimp body weight gain between weeks 6-7. Six OTUs associated with the microbiota change in KM group were identified. This finding strongly suggests that the shrimp gut microbiota may be correlated with the shrimp body weight likely by influencing nutrient uptake in the gut. Nanoparticles are a unique structural scaffold for the fabrication of molecular dyad-like structures, in particular, when the functional moieties are bound onto the nanoparticle surface by conjugated interfacial bonds. In the present study, stable ruthenium nanoparticles (RuHC12) were prepared by the self-assembly of 1-dodecyne onto the nanoparticle surface forming ruthenium-vinylidene interfacial bonds and further functionalized with acetylene derivatives of coumarin and perylene by olefin metathesis reactions. Steady-state photoluminescence measurements of ethynylcoumarin-functionalized ruthenium (RuEC) nanoparticles showed that whereas the excitation and emission maxima remained almost unchanged, the normalized emission intensity was significantly enhanced, as compared to that of RuHC12, due to the much higher quantum yield of the coumarin moieties. By contrast, for the nanoparticles cofunctionalized with ethynylcoumarin and ethynylperylene (RuECEP), the excitation and emission maxima were close to those of ethynylperylene-functionalized ruthenium (RuEP) nanoparticles, but the normalized emissions of RuECEP were markedly quenched as compared to those of RuEP, due to effective photoinduced intraparticle charge transfer from the electron-donating coumarin moieties to the electro-accepting perylene moieties via the conjugated metal-ligand interfacial bonds in RuECEP. This is in sharp contrast to conventional coumarin-perylene dyads where energy transfer plays a dominant role and the perylene emissions were actually markedly enhanced.
