The frequent use of electronic devices in daily lives, predominantly reliant on non-ionizing radiation, has increased the prevalence of electromagnetic radiation (EMR) in natural environment. In light of this, effects of EMR at frequencies of 1800 MHz and 2400 MHz characterized by a power of 10.0 dBm (0.01 W), across varying exposure durations of 1 h/day, 2 h/day, 4 h/day, 6 h/day, and 8 h/day for 7 days, in Allium cepa L. were studied. The effects of the treatment on the morphological features (root length, fresh weight, and dry weight of roots) and biochemical characteristics (protein content and antioxidative enzymes, namely, ascorbate peroxidase (APX), glutathione reductase (GR), superoxide dismutase (SOD), glutathione-S-transferase (GST), guaiacol peroxidase (POD), catalase (CAT), and dehydroascorbate (DHAR)) were studied in roots and bulbs of Allium cepa L. Further genotoxicity for different exposure periods at both frequencies was also conducted. Prolonged exposure to electromagnetic radiation (EMR) at both frequencies was found to reduce root length, fresh weight, and dry weight of plant. Furthermore, significant effects were observed on protein content, indicating a reduction with prolonged exposure duration. Investigation into the activities of antioxidative enzymes such as APX, GR, GST, DHAR, CAT, SOD, and POD at a frequency of 1800 MHz and 2400 MHz in roots and bulbs demonstrated a significant enhancement in enzyme activity during 6 h/day and 8 h/day exposure periods. Additional investigation during genotoxicity studies demonstrated the induction of chromosomal aberrations in the root tip cells of the Allium cepa L. plant test system. The current study revealed the initiation of oxidative stress and genotoxicity resulting from long-term exposure to electromagnetic radiation in the studied plant test systems. Benign tumors, but rarely cancer, are common in patients with tuberous sclerosis complex (TSC). Blood samples from patients undergoing treatment for TSC at our institution were analyzed for their individual sensitivity to ionizing radiation. Blood samples were collected from 13 adult patients with TSC. The samples were irradiated ex vivo and analyzed by 3-color fluorescence in situ hybridization. In each patient, aberrations were analyzed in 200 metaphases of chromosomes 1, 2, and 4 and scored as breaks. Radiosensitivity was determined by mean breaks per metaphase (B/M) and compared to both healthy donors and oncologic patients. The radiosensitivity (B/M) of the TSC patient cohort (n = 13; female: 46.2%, B/M: 0.48 ± 0.11) was clearly increased compared to healthy individuals of similar age (n = 90; female: 54.4%; B/M: 0.40 ± 0.09; p = 0.001). There was no difference compared to age-matched oncological patients (n = 78; female: 67.9%; B/M 0.49 ± 0.14; p = 0.246). Similarly, the proportion of radiosensitive (B/M > 0.5) and distinctly radiosensitive individuals (B/M > 0.6) was increased in the TSC and oncological patient cohorts (TSC: 30.8% and 7.7%, oncological patients: 46.2% and 14.1%) compared to the healthy individuals (11.1% and 2.2%). Purpose: Ionizing radiation-based technologies are extensively used in the diagnosis and treatment of diseases. While utilizing the technologies, exposure to a certain amount of radiation is unavoidable. Data can be obtained from participants who received radiation during medical imaging and therapeutic purposes to predict the effects of low-dose radiation. Methods: To understand the effects of low-dose radiation, participants (n = 22) who received radioactive I-131 for scan/therapy were used as a model in this study. Blood samples were drawn pre- and post-administration of I-131. Biological effects were measured using markers of DNA damage (γ-H2AX, micronucleus (MN), and chromosomal aberrations (CA)) and response to damage through gene expression changes (ATM, CDKN1A, DDB2, FDXR, and PCNA) in blood samples. Results: Mean frequency of γ-H2AX foci in pre-samples was 0.28 ± 0.16, and post-samples were 1.03 ± 0.60. γ-H2AX foci frequency obtained from post-samples showed significant (p < 0.0001) and a heterogeneous increase in all the participants (received I-131 for scan/therapy) when compared to pre-samples. A significant increase (p < 0.0001) in MN and CA frequency was also observed in participants who received the I-131 therapy. Gene expression analysis indicates that all genes (ATM, CDKN1A, DDB2, FDXR, and PCNA) were altered in post-samples, although with varying degrees, suggesting that the cellular responses to DNA damage, such as damage repair, cell cycle regulation to aid in repair and apoptosis are increased, which priority is given to repair, followed by apoptosis. Space rockets are regularly launched from the Baikonur Cosmodrome in Kazakhstan. The ecological impact of territories affected by the consequences of space rocket activities on the environment is a significant concern. On July 27, 2006, the Dnepr rocket was launched from the Baikonur Cosmodrome, resulting in an abnormal situation and its emergency fall. The rocket fuel components, heptyl, and amyl, led to decontamination and reclamation activities. However, the self-restoration of soil and vegetation cover is currently in the earliest stages. Consequently, an assessment of the remote consequences of the accidental fall of the Dnepr launch vehicle for environmental objects was conducted. The study involved eco-toxicological characterization of habitat conditions, assessment of species diversity, determination of key morphophenetic indicators, and the genetic status of indicator species as the main parameters. The results revealed an increased level of genotoxicity in soils at the accident site. While there were deviations from the norm in some morphophenetic and cytogenetic parameters in indicator animals, they were not critical for their existence in this territory. Glyphosate and 2,4-dichlorophenoxyacetic acid (2,4-D) are the most widely used pesticides in the world. In this work, the cytogenotoxic potential of these herbicides and their binary mixtures was investigated using A. cepa test. 2,4-D was assayed at 0.4 – 400 μg L−1 and glyphosate at 0.65 – 650 μg L−1 concentration range. The concentrations evaluated cover those permitted by Brazilian legislation for the protection of aquatic life and human health. Sixteen binary mixtures were examined to replicate different contamination scenarios. When analyzed separately, 2,4-D at 400 μg L−1 inhibited the mitotic index, while glyphosate at 0.65 and 65 μg L−1 increased it. In addition, the herbicides increased the frequency of chromosomal aberrations (2,4-D at 0.4, 4, and 40 μg L−1 and glyphosate at 0.65, 65, and 650 μg L−1) and micronuclei (2,4-D at 400 μg L−1 and glyphosate at 650 μg L−1). Regarding the mixtures, six of them were cytotoxic while eight were genotoxic. Additive, synergistic, or potentiated effects were observed. However, none of the mixtures tested increased the frequency of micronuclei. The results showed that even concentrations allowed by Brazilian legislation pose a potential risk to exposed organisms. There is little research on the cytogenotoxic effects of 2,4-D + glyphosate mixtures, so this is the first study to analyze a large number of combinations. The detection of additive, synergistic, or potentiated toxicities highlights the need for better pesticide legislation and discussions among regulators and environmental managers regarding pesticide mixtures in water. Circulating T-lymphocytes are used as "natural biodosimeters" for estimating radiation doses, since the frequency of chromosomal aberrations induced in them is proportional to the accumulated dose. Moreover, stable chromosomal aberrations (translocations) are detected years and decades after exposure. Internal incorporation of radionuclides often leads to non-uniform exposure, which resulted in difficulties in the application of retrospective biodosimetry using T-lymphocytes. Some properties of T-lymphocytes complicate retrospective biodosimetry in this case: (1) the thymic production of T-cells depends significantly on age, the maximum is observed in early childhood; (2) the "lymphocyte-dosimeter" accumulates changes (translocations) while circulating through the body. The objective of this paper is to describe the technical characteristics of the model of age dynamics and T-cell biokinetics and approaches to assessing the dose to circulating lymphocytes under various exposure scenarios. The model allows to quantify the fractions of T-lymphocytes that were formed before and after exposure. The model takes into account the time fractions that circulating lymphocytes spend in various lymphoid organs. Age-related thymic involution was also considered. The model predicts that after internal exposure to 90Sr, the doses to T-lymphocytes can differ significantly from the doses to the bone marrow and other tissues. For uniform external γ-exposure, and for internal exposure due to non-bone -seeking radionuclides (for example, 144Ce), predicted doses to T-lymphocytes are very close to bone marrow doses. This study aimed to investigation the effect of environmental changing associated with the elevation gradient on cytogenetic traits of Taraxacum officinale during its invasion in high altitude areas. We chose common dandelion as the object of research, because representatives of this species are able to inhabit a large range of elevations. The cytogenetic characteristics of the plants growing at altitudes of 200, 600, 1300, 2050, 2700 and 3050 m a.s.l in the Central Caucasus region have been studied. We determined the frequency of micronuclei, chromosomal aberrations, mitotic index and chromosome number in the cells of the apical root meristem using seed progeny of plants growing at different elevations. In the populations of natural habitats studied by us, all individuals were triploid with the exception of single ones. The level of ploidy increased in accordance with the altitude gradient when T. officinale invade in the disturbed highland areas. In the seed progeny of highland plants cell proliferation and the ratio of mitosis phases do not change along the elevation gradient. The frequency of chromosome aberrations increased by 1.3–4.6 times in the highland populations. The main types of mutations were fragments, lost and lagging chromosomes. The reasons for this are the influence of unfavorable conditions of high-altitude areas and high level of ploidy in plants of these populations. Astronauts travelling in space will be exposed to mixed beams of particle radiation and photons. Exposure limits that correspond to defined cancer risk are calculated by multiplying absorbed doses by a radiation-type specific quality factor that reflects the biological effectiveness of the particle without considering possible interaction with photons. We have shown previously that alpha radiation and X-rays may interact resulting in synergistic DNA damage responses in human peripheral blood lymphocytes but the level of intra-individual variability was high. In order to assess the variability and validate the synergism, blood from two male donors was drawn at 9 time points during 3 seasons of the year and exposed to 0–2 Gy of X-rays, alpha particles or 1:1 mixture of both (half the dose each). DNA damage response was quantified by chromosomal aberrations and by mRNA levels of 3 radiation-responsive genes FDXR, CDKN1A and MDM2 measured 24 h post exposure. The quality of response in terms of differential expression of alternative transcripts was assessed by using two primer pairs per gene. A consistently higher than expected effect of mixed beams was found in both donors for chromosomal aberrations and gene expression with some seasonal variability for the latter. Topotecan administered intraperitoneally at single doses of 0.25, 0.5, and 1 mg/kg induced chromosomal aberrations in bone marrow cells of F1(CBA×C57BL/6) hybrid mice in a dose-dependent manner. A tyrosyl-DNA phosphodiesterase 1 (TDP1) inhibitor, an usnic acid derivative OL9-116 was inactive in a dose range of 20-240 mg/kg, but enhanced the cytogenetic effect of topotecan (0.25 mg/kg) at a dose of 40 mg/kg (per os). The TDP1 inhibitor, a coumarin derivative TX-2552 (at doses of 20, 40, 80, and 160 mg/kg per os), increased the level of aberrant metaphases induced by topotecan (0.25 mg/kg) by 2.1-2.6 times, but was inactive at a dose of 10 mg/kg. The results indicate that TDP1 inhibitors enhance the clastogenic activity of topotecan in mouse bone marrow cells in vivo and are characterized by different dose profiles of the co-mutagenic effects. Among the compounds present in necro-leachate, a liquid released during the process of decomposition of the human body, are the biogenic amines cadaverine and putrescine. Although some studies on necro-leachate have indicated a potential ecotoxicological and public health risk associated with it, the research on this type of contamination is still rather limited. This study presents information about the phytotoxic and cytogenotoxic potential of cadaverine and putrescine, evaluated separately and within a mixture. Phytotoxicity was evaluated through a germination test, the initial growth of seedlings with Lactuca sativa, and cytogenotoxicity through chromosomal aberration and micronucleus tests with Allium cepa. The L. sativa results showed a phytotoxic effect for the evaluated amines, by reducing root (> 90%) and hypocotyl (> 80%) elongation. The co-exposure of cadaverine and putrescine potentiated cytogenotoxic activity by aneugenic action in the meristematic cells of A. cepa. From this result, it is possible to infer the eco-toxicogenic potential of cadaverine and putrescine. This study not only highlights the importance of the phytotoxic and cytogenotoxic effects of these amines but also emphasizes the urgent need for further investigation into contamination originating from cemetery environments. The effect of a pulsed magnetic field (PMF) on meristem cells of onion seedlings was compared with the effects of acute gamma irradiation using the allium test. It was found that a PMF with a carrier frequency of 1.8 MHz, a pulse repetition rate of 28 kHz, and a magnetic field induction of 75 mT per pulse leads to an increase in the mitotic index, mainly due to an increase in the proportion of cells in the prophase and an increase in the frequency of cells with chromosome aberrations in the ana-telophase and does not affect the frequency of cells with micronuclei. It has been suggested that a PMF causes nonspecific oxidative stress in plant cells, accompanied by a delay in the cell cycle at the check point (G2/M) and induction of DNA damage. Chromosome aberrations are an example of the mutation process that happens at the chromosome level of DNA packaging. The occurrence of inversions and complex translocations has not been studied well enough due to the complexity of their detection using the conventional methods of differential staining. It is well-known that ionizing radiation is one of the factors that increases the frequency of chromosomal rearrangements. The objective of this pilot research was to study the frequency of inversions involving telomere regions of the chromosomes and complex translocations in T-cells of 25 chronically exposed residents of the Southern Urals. Inversions were studied in the group of 12 exposed individuals (one man, 11 women, exposure doses ranging from 0.001 to 2.9 Gy) using the Q-FISH method. Complex translocations were analyzed in the group of 13 people (nine women, four men, exposure dose ranging from 0.5 to 3.1 Gy) with the help of M-FISH method. The groups did not overlap. As a result of this analysis, it has been revealed that complex translocations occur six times less frequently than simple ones (р < 0.001). Chromatid variants of inversions predominated as compared to chromosomal ones (9 : 0.17 per 100 cells, p < 0.001). This paper presents a synthesis of silver nanoparticles by the novel method with grape pomace extract obtained by eco-friendly "green" solvent, namely, deep eutectic solvent (DES). The electrical conductivity of deep eutectic solvents was measured, and the width of electrochemical stability window was evaluated depending on water content in the solvent (0–25%). The DES was used to obtain the extract, and the latter was used in synthesis nanoparticles. The bands found in the FTIR spectrum were found to be associated with phenolic compounds. Grape pomace extract (GPE-DES) were analyzed by HPLC-DAD-MS to identify extracted compounds. The phytochemical profile of GPE-DES exhibits a high amount of the hydroxycinnamic acids (~23%), followed by anthocyanins (~19%). Furthermore, caffeic acid (164.5 μg/g), gallic acid (792.4 μg/g), and (+)-catechin (511.3 μg/g) were quantified with HPLC. A sharp absorbance peak at 436 nm was found using a UV-visible spectrophotometer, confirming the presence of AgNPs. The extensive spectroscopic study enabled the identification of synthesized AgNPs. The silver nanoparticles with round shape were noticed on the scanning electron microscopy micrographs with the particle size in the range of 10–20 nm. High antibacterial potential has been proven against gram-positive spore-forming B. subtilis and gram-negative bacteria E. coli with 9 mm and 16 mm zone inhibition, respectively, observed by agar well diffusion method. Mitotic index (MI) and chromosomal aberrations (CAs) were assessed by Allium cepa assay. The synthesized silver nanoparticles do not induce cytogenotoxic and genotoxic changes in Allium cepa L. with nanoparticles at concentration up to 10%. In the current study, two plants, viz., Pisum sativum L. and Hordeum vulgare L., were exposed to nano- and macro-dispersed ZnO at 1, 10, and 30 times of maximal permissible concentration (MPC). The main objective of the study is to depict and compare the genotoxicity in terms of chromosomal anomalies, cytotoxicity (i.e., mitotic index), and phytotoxicity (viz., germination, morphometry, maximal quantum yield, and chlorophyll fluorescence imaging) of macro- and nano-forms of ZnO along with their accumulation and translocation. In the case of genotoxic and cytotoxic responses, the maximal effect was observed at 30 MPC, regardless of the macro- or nano-forms of ZnO. The phytotoxic observations revealed that the treatment with macro- and nano-forms of ZnO significantly affected the germination rate, germination energy, and length of roots and shoots of H. vulgare in a dose-dependent manner. The factor toxicity index of treated soil demonstrated that toxicity soared as concentrations increased and that at 30 MPC, toxicity was average and high in macro- and nano-dispersed ZnO, respectively. Furthermore, the photosynthetic parameters were observed to be negatively affected in both treatments, but the maximal effect was observed in the case of nano-dispersed form. It was noted that the mobility of nano-dispersed ZnO in the soil was higher than macro-dispersed. The increased mobility of nano-dispersed ZnO might have boosted their accumulation and translocation that subsequently led to the oxidative stress due to the accelerated production of reactive oxygen species, thus strengthen toxicity implications in plants. In this study, we investigate the toxicity of commercial formulations based on glyphosate, 2,4-D, imidacloprid, and iprodione, in isolation and mixed, on Allium cepa. The mixtures consisted of combinations in the lowest (M1), intermediate (M2), and highest concentrations (M3) of each pesticide. We measured physiological (germination rate, germination speed, and radicular length) and cyto-genotoxic (mitotic index and frequency of aberrant cells) parameters. In addition, we analyzed the cell cycle progression and cell death induction by flow cytometry. When applied in isolation, the pesticides changed the parameters evaluated. M1 and M2 inhibited root length and increased the frequency of aberrant cells. Their genotoxic effect was equivalent to that of pesticides applied in isolation. Furthermore, M1 and M2 caused cell death and M2 changed the cell cycle progression. M3 had the greatest deleterious effect on A. cepa. This mixture inhibited root length and promoted an additive or synergistic effect on the mitotic index. In addition, M3 changed all parameters analyzed by flow cytometry. The most sensitive biomolecule under radiation exposure is DNA, whose damage manifests itself in the form of chromosomal aberrations (CA). The processes of DNA methylation, which are involved in the regulation of gene expression, replication, DNA repair, etc., are also affected by gamma radiation. The aim of the study was to evaluate the relationship between the degree of DNA methylation and the frequency of CA after acute in vitro irradiation of human blood lymphocytes with gamma radiation. The study involved ten conditionally healthy workers of the Siberian Chemical Plant in whose blood lymphocytes the degree of methylation of CpG-dinucleotides (wide-genome bisulfite sequencing, XmaI-reduced representation bisulfite sequencing—XmaI-RRBS) and the frequency of CA (cytogenetic study) after acute in vitro irradiation of blood with doses of 0 and 1.5 Gy were evaluated. After acute exposure to gamma radiation in lymphocytes, the frequency of aberrant cells, dicentric chromosomes, and chromatid and chromosomal fragments increased. Correlation analysis of the status of CpG-dinucleotide methylation and the frequency of CA revealed changes in the degree of methylation of 97 genes, which strongly correlated positively (56 genes) or negatively (41 genes) with an increased frequency of CA. A primary genome-wide screening of genes whose methylation is correlated with a high frequency of CA was carried out. Many of the identified genes are promising as potential markers of radiation exposure and to study the mechanisms of formation of radiosensitivity of the body and radioresistance of tumors during radiation therapy. The physical and chemical characteristics of the soil can influence plant growth. When sewage sludge (SS) is applied as a soil fertilizer, the accumulation of non-essential elements contained in it can be toxic for plants. The aim of this study was to understand the effect of SS dosage on the cell cycle of Lactuca sativa L. meristematic cells and on the initial growth of L. sativa and Passiflora alata Curtis. Nine concentrations of SS + distilled water (mg dm−3) corresponding to 0, 20, 40, 60, 80, 120, 160, 320, and 520 t ha−1 were tested in four replicates of 25 seeds. Chemical analysis showed an increase in pH of the sludge from 0 to 80 t ha−1 SS followed by its stabilization thereafter. The highest electrical conductivity was observed at 520 t ha−1 SS. SS negatively affected the germination and initial growth of seedlings from P. alata and L. sativa. Cytogenetic analysis on 6000 L. sativa meristematic cells for each treatment revealed that SS could adversely affect the genetic stability of this species. SS concentrations above 120 t ha−1 adversely affected the germination and early seedling growth of L. sativa and P. alata. Data from studies of the genotoxic activity of psychotropic drugs published over the past 25 years were analyzed. Information describing the genotoxicity of psychotropic drugs was found to be fragmentary and inconsistent, and the experimental conditions in which they were produced often fail to meet accepted requirements. Among the 94 drugs considered, sufficient data for concluding that genotoxic properties are present or absent were found for only 9.6%. There is a need for a large-scale systematic reassessment of the genotoxicity of psychotropic drugs, especially first-generation drugs, based on modern methodology, including studies of mutagen-modifying activity. The desirability of monitoring the genotoxic status of patients treated with psychotropic drugs is emphasized, and this should contribute to an adequate assessment of the genotoxic risk of their use and determination of objective approaches to selecting drugs for safe therapy. Background: The treatment with chemotherapy may develop secondary tumors as a result of chemo genotoxicity. Sperm defects is another complication associated with chemo treatment. In this study the genotoxicity of vinblastine (VB) was estimated in both somatic and germ cells. Materials: 85 mice were taken. Four single doses of VB at 3, 4.5, 6 and 10 mg/kg and three successive doses at 3, 4.5 and 6 mg/kg were taken for estimation of chromosomal aberrations (CAs). Four single doses of VB were involved in estimating the DNA fragmentation, and comet assay. For sperm abnormalities mice were injected with three successive doses of VB at 3, 4.5, and 6 mg/kg. Results: The results demonstrated a significant frequency of DNA fragmentation in spleen cells and in the percentage of CAs in bone marrow. Numerical and structural aberrations were recorded with a pronounced number of polyploidy metaphases which reached (11.60%) after treatment with 6 mg/kg for three successive days vs zero for control. VB also induced a significant percentage of CAs in spermatocytes in the form of univalent. Sperm defects in the form of coiled tail, absence of acrosome and shapeless head and a significant DNA damage in the testes were recorded. The frequency of sperm abnormalities reached 11.06 ± 0.14 after treatment with highest tested dose (6 mg/kg) vs 3.04 ± 0.19 for control. Conclusion: VB is genotoxic in somatic and germ cells. Repeated heating of edible oils at high temperatures and its use in cooking food generates polycyclic aromatic hydrocarbons (PAHs) that have carcinogenic potential. The use of repeatedly heated cooking oils (RHCO) is a common practice in India. The present investigation in Wistar rats was done to determine the genotoxic potential of consumption of food cooked in sunflower oil that has been repeatedly heated to boiling. The rats were fed a diet cooked-fried in such oil. The biomarkers of genotoxicity, comet assay, micronucleus test, and chromosomal aberrations in peripheral blood lymphocytes (PBL) of Wistar rats were used. Results of the present investigation reveal that rats fed on food cooked in oil that was 5 times repeatedly boiled induced significant Deoxy ribonucleic acid (DNA) damage in PBL and liver homogenate. Increased frequency of micronuclei and chromosomal aberrations in blood and bone marrow of rats were also observed. A similar observation was found in rats that were fed food cooked in oil that was boiled 3 times. However, the results of genotoxicity in rats that ate food cooked in oil heated only once were not statistically significant in comparison to the control rats that fed on food made in heated oil (not boiled). Intake of food cooked in repeatedly heated oil of different heating grades induced significant genotoxicity in rats evident by increased DNA damage and frequency of micronuclei and chromosomal aberrations. The presence of PAHs in heated oils triggers the generation of free radicals which could be the possible causative factor for the induced genetic damage. Unprecedented growth in the communication sector and expanded usage of the number of wireless devices in the past few decades have resulted in a tremendous increase in emissions of non-ionizing electromagnetic radiations (EMRs) in the environment. The widespread EMRs have induced many significant changes in biological systems leading to oxidative stress as well as DNA damage. Considering this, the present study was planned to study the effects of EMRs at 900 MHz frequency with the power density of 10.0 dBm (0.01 W) at variable exposure periods (0.5 h, 1 h, 2 h, 4 h, and 8 h per day for 7 days) on percentage germination, morphological characteristics, protein content, lipid peroxidation in terms of malondialdehyde content (MDA), and antioxidant defense system of Trigonella foenum-graecum test system. The genotoxicity was also evaluated using similar conditions. It was observed that EMRs significantly decreased the germination percentage at an exposure time of 4 h and 8 h. Fresh weight and dry weight of root and shoot did not show significant variations, while the root and shoot length have shown significant variations for 4 h and 8 h exposure period. Further, EMRs enhanced MDA indicating lipid peroxidation. In response to exposure of EMRs, there was a significant up-regulation in the activities of enzymes such as ascorbate peroxidase (APX), superoxide dismutase (SOD), glutathione-S-transferase (GST), guaiacol peroxidase (POD), and glutathione reductase (GR) in the roots and shoots of Trigonella-foenum graecum. The genotoxicity study showed the induction of chromosomal aberrations in root tip cells of the Trigonella foenum-graecum test system. Bisphenol S (BPS) is an analog of bisphenol A, which is used as substitute of BPA in many products like airport luggage tags, baby bottles, plastics, and epoxy resins etc. Bisphenol S can cause toxic effects in different organisms, i.e., mice, rat, zebrafish, and C.elegans, etc. Bisphenol S is also known as "endocrine disruptor" due to its ability to mimic the endocrine receptors. So, the aim of this study was to evaluate the cytotoxic and genotoxic effects of bisphenol S on meristematic cells present in onion root tips through Allium cepa (A.cepa) and comet tests. Root growth inhibition was evaluated by root growth inhibition assay. Mitotic index (MI) and chromosomal aberrations (CAs) were assessed by A.cepa assay. DNA damage was evaluated by comet assay. Root growth of A.cepa was inhibited due to bisphenol S. LC50 value calculated by root growth inhibition assay for bisphenol S was (2.6±0.63, 50 μg/ml). Mitotic index was reduced, and chromosomal aberrations were observed, i.e., stickiness, polyploidy, and disturbed ana-telophase in anaphase and telophase stages of mitosis. In case of comet assay, DNA damage was increased in statistically significant manner (p ≤ 0.05). It was concluded that bisphenol S constitutes cytotoxic and genotoxic effects on A. cepa root meristematic cells. Eprinomectin, a veterinary drug within the family of avermectins, is widely used in the agricultural sector to combat a variety of parasites, mainly nematodes. However, only 10% of the drug is metabolized in the organism, so large quantities of the drug are released into the environment through urine and/or feces. Soil is the first and main environmental compartment to be contaminated by it, and nontargeted organisms can be affected. Thus, the present study aims to evaluate the phytotoxicity (through the evaluation of germination, root development, and germination speed) and genotoxicity (through an assessment of the induction of micronuclei and chromosomal aberrations) of eprinomectin. For the analyses, Allium cepa seeds were germinated in soil contaminated with a range of concentrations of eprinomectin: from 0.5 to 62.5 μg/g for the genotoxicity test and from 0.5 to 128.0 μg/g for the phytotoxicity test. The results showed that seed germination was not affected, but root development was affected at concentrations of 0.5 μg/g, 1.0 μg/g, 4.0 μg/g, 8.0 μg/g, 64.0 μg/g, and 128.0 μg/g, and germination speed was significantly changed at concentrations of 1.0 μg/g, 4.0 μg/g, 16.0 μg/g, 32.0 μg/g, and 64.0 μg/g. Significant differences in the mitotic index and genotoxicity index were observed only at concentrations of 2.5 μg/g and 12.5 μg/g, respectively. Only the 0.5 μg/g concentration did not show significant induction of micronuclei in the meristematic cells, but the damage observed at other concentrations did not persist in F1 cells. Studies suggest that chronic lead (Pb) exposure may induce deoxyribonucleic acid (DNA) damage. However, there is no synthesised evidence in this regard. We systematically reviewed existing literature and synthesised evidence on the association between chronic Pb exposure and markers of genotoxicity. Observational studies reporting biomarkers of DNA damage among occupationally Pb-exposed and unexposed controls were systematically searched from PubMed, Scopus and Embase databases from inception to January 2022. The markers included were micronucleus frequency (MN), chromosomal aberrations, comet assay, and 8-hydroxy-deoxyguanosine. During the execution of this review, we followed the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines. Mean differences in the biological markers of DNA damage between Pb-exposed and control groups were pooled using the random-effects model. The heterogeneity was assessed using the Cochran-Q test and I2 statistic. The review included forty-five studies comparing markers of DNA damage between Pb-exposed and unexposed. The primary studies utilised buccal and/or peripheral leukocytes for evaluating the DNA damage. The pooled quantitative results revealed significantly higher DNA damage characterised by increased levels of MN and SCE frequency, chromosomal aberrations, and oxidative DNA damage (comet assay and 8-OHdG) among Pb-exposed than the unexposed. However, studies included in the review exhibited high levels of heterogeneity among the studies. Chronic Pb exposure is associated with DNA damage. However, high-quality, multicentred studies are required to strengthen present observations and further understand the Pb's role in inducing DNA damage. CRD42022286810. Pethoxamid is chloroacetamide herbicide. Pethoxamid is commonly used to kill different weeds in various crops. Pethoxamid can leach in the water and soil and can cause toxic effects to other non-target species. Current study is therefore aimed to perform the investigation of the cytotoxic and genotoxic effects of pethoxamid on Allium cepa cells.The root growth, mitotic index (MI), chromosomal aberrations (CAs), and DNA damage were assessed through root growth inhibition, A. cepa ana-telophase, and alkaline comet assays, respectively. Furthermore, molecular docking was performed to evaluate binding affinity of pethoxamid on DNA and very-long-chain fatty acid (VLCFA) synthases. In root growth inhibition test, onion root length was statistically significantly decreased in a concentration dependent manner. Concentration- and time-dependent decreases in MI were observed, whereas increase in CAs such as disturbed ana-telophase, chromosome laggards, stickiness, anaphase bridges, and DNA damage was caused by the pethoxamid on A. cepa root cells. Molecular docking revealed that pethoxamid binds selectively to GC-rich regions in the minor groove of the DNA structure and showed remarkable binding affinity against all synthases taking part in the sequential biosynthesis of VLCFAs. Purpose: Kinase inhibitors (KI) are known to increase radiosensitivity, which can lead to increased risk of side effects. Data about interactions of commonly used KI with ionizing radiation on healthy tissue are rare. Patients and methods: Freshly drawn blood samples were analyzed using three-color FISH (fluorescence in situ hybridization) to measure individual radiosensitivity via chromosomal aberrations after irradiation (2 Gy). Thresholds of 0.5 and 0.6 breaks/metaphase (B/M) indicate moderate or clearly increased radiosensitivity. Results: The cohorts consisted of healthy individuals (NEG, n = 219), radiosensitive patients (POS, n = 24), cancer patients (n = 452) and cancer patients during KI therapy (n = 49). In healthy individuals radiosensitivity (≥ 0.6 B/M) was clearly increased in 5% of all cases, while in the radiosensitive cohort 79% were elevated. KI therapy increased the rate of sensitive patients (≥ 0.6 B/M) to 35% significantly compared to 19% in cancer patients without KI (p = 0.014). Increased radiosensitivity of peripheral blood mononuclear cells (PBMCs) among patients occurred in six of seven KI subgroups. The mean B/M values significantly increased during KI therapy (0.47 ± 0.20 B/M without compared to 0.50 ± 0.19 B/M with KI, p = 0.047). Plants are continuously exposed to high concentrations of engineered nickel oxide nanoparticles (NiO-NP) from increasingly contaminated soil and water. As they are the primary producers in any ecosystem, their behaviour to NiO-NP excess merit closer scrutiny. Ni is an essential micronutrient. Existing reports confirm phytotoxicity of NiO-NP, primarily through ROS paroxysm induced when they enter and accumulate in plant tissues. In the present study, the authors have endeavoured to deduce the link between chromosomal aberrations occurrence and cell cycle perturbations in Allium cepa in response to NiO-NP. A wide range of NiO-NP concentrations (10–500 mg L−1) were taken to understand dosage effect on cell cycle discrepancies. Detailed assessments of cytotoxic and genotoxic markers, like changes in the frequency of MI, MN and aberration indices, showed that with increase in NiO-NP concentration induced genotoxicity were noted. Instances of clastogenic and physiological aberrations, coincided with a dose dependent decrease in MI. This was concomitant with a dip in G1 population and sharp increase at the G2/M peak as seen using Flow Cytometry in samples undergoing similar treatment regimes. Maximum perturbation in the cell cycle dynamics coincided with maximum MI loss and increased frequency of aberrations. Instances of extreme low MI, heavy chromosomal condensation and disarray at the highest concentrations of NiO-NP used (250–500 mg L−1) were also reported. This led to initiation of cell death cascades, as evident from minimizing of the G1 population. For a reliable characterization of environmental quality, it is recommended to supplement the data obtained in natural populations with laboratory experiments using a test object. Biocides, which are found in nature as persistent pollutants, pose a great danger to the ecosystem. Methylisothiazolinone (MIT), a widely used biocide, reaches plants by mixing with water and soil. Vermicompost tea (VCT), which strengthens the plant defence mechanism and increases its growth and development, is a liquid fertiliser consisting of the cooperation of worms with microbes. In the present study, after applying 0.4 g/L (EC50/2), 0.8 g/L (EC50), and 1.6 g/L (EC50 × 2) MIT concentrations without and with VCT on forage pea (Pisum sativum), root lengths, mitotic index data, chromosome and nuclei abnormalities, and DNA damage level were determined. When VCT applied and non-applied groups were compared, it was found that, especially in the VCT applied group, they cope with the stress conditions created by MIT. In addition, positive effects were observed in root lengths, mitotic index data, and amount of cell nuclei abnormalities. On the territory of South Kazakhstan (Almaty region) there are old abandoned warehouses with prohibited and unused pesticides, the active substances and metabolites of which pollute natural water sources and pastures of farm animals located nearby. The analysis of blood samples of 50 heads of cattle from five monitoring points of the Almaty region was carried out for the presence of micronuclei in erythrocytes, genomic mutations and chromosomal aberrations in lymphocytes. Cell cultivation and preparation of preparations were carried out by standard cytogenetic methods. The frequency of erythrocytes with micronuclei in cattle kept in experimental plots was 3.3%, which is 4.7 times higher than in the control group. The incidence of genomic mutations in the blood system of experimental groups of animals exceeded that in the control group by an average of 1.6 times, and chromosomal aberrations—by 9.1 times. The proportion of hypodiploid cells was 46% of the total number of genomic mutations. Polyploidy accounted for up to 17%, and hyperdiploidy was found in 5% of cells. The analysis of individual indicators of general cytogenetic instability in the blood system of animals showed that hyperdiploidy and chromosomal aberrations are the main components of this indicator, which, on average for five monitoring sites, exceeded the control data by 7.9 times. Statistical data processing allows us to make a conclusion about the genotoxic effect of prohibited and unused pesticides on the body of cattle, which have clastogenic, aneogenous and mutagenic effects even after 30 years. The present study assessed the utility of Allium cepa based cyto-genotoxicity bioassays in evaluating the arsenic toxicity and remediation potential of Pteris vittata on contaminated soil of Lakhimpur-Kheri district. Untreated and P. vittata treated soil extracts were used for cyto-genotoxicity tests in A. cepa. Results showed that P. vittata extracted high concentration of arsenic, which ranged from 220 to 1420 mgkg−1 in different soils. Cyto-genotoxic assessment of A. cepa showed that extract of P. vittata treated soil had lower cyto-genotoxic effects as compared to untreated soil. A higher mitotic index (10%) while lower mitotic depression (29%), relative abnormality rate (10%), chromosomal aberrations (1%) and micronuclei (2%) were detected in root meristematic cells of A. cepa exposed to remediated soil extract in comparison to untreated soil. The studies provide a simple, rapid and economic cyto-genotoxicity bioassay tool for evaluating toxicity of contaminated soils of contaminated soils as well as revealed the phytoremdiation property of P. vittata against arsenic toxicity. Spontaneous abortion occurs in 8–20% of recognized pregnancies and usually takes place in the first trimester (7–11 weeks). There are many causes of pregnancy loss, but the most important (about 75%) is the presence of chromosomal aberrations. We present the results of oligonucleotide array application in a cohort of 62 miscarriage cases. The inclusion criteria for the study were the loss after 8th week of pregnancy and the appearance of recurrent miscarriages. DNA was extracted from trophoblast or fetal skin fibroblasts. In the 62 tested materials from recurrent miscarriages, the detection rate was 56.5% (35/62). The most commonly found were aneuploidies (65%) (chromosomal trisomy 14, 16, 18, 21, and 22), Turner syndrome, and triploidy (17.1%). Other chromosomal abnormalities included pathogenic and likely pathogenic structural aberrations: 1) pathogenic: deletion 7p22.3p12.3 and duplication 9p24.3p13.2 inherited from the normal father, deletion 3q13.31q22.2 and deletion 3q22.3q23 of unknown inheritance and duplication of 17p12 inherited from father with foot malformation; 2) likely pathogenic variants: deletion 17p13.1 inherited from normal mother, deletion 5q14.3 of unknown inheritance and de novo deletion 1q21.1q21.2. Among these aberrations, six CNVs (copy number variants) were responsible for the miscarriage: deletion 7p22.3p12.3 and duplication 9p24.3p13.2, deletion 3q13.31q22.2 and deletion 3q22.3q23, and deletion 17p13.1 and deletion 1q21.1q21.2. Other two findings were classified as incidental findings (deletion 5q14.3 and 17p12 duplication). Our research shows that 17% of the aberrations (6/35 abnormal results) that cannot be identified by the routine kariotype analysis are structural aberrations containing genes important for fetal development, the mutations of which may cause spontaneous abortion. Despite the great efforts of basic and clinical research on lung cancer (LC), the prevention and treatment of this type of cancer show still far from satisfactory indicators. The complex mechanisms of the LC biology are already recognized, but there is a significant gap in the knowledge about the interaction of key biomarkers in the etiology and progression of this disease. The present work is aimed at studying the relationship between the telomere length (TL) and frequencies of chromosomal aberrations (CA) and micronuclei (MN) in leukocytes of primary lung cancer patients. In 67 men with lung cancer and 77 control donors (men), the baseline frequencies of CA and MN were determined in blood leukocytes, and TL was also studied. As a result, a significant increase in TL was found in lung cancer patients compared with control donors (25.86 ± 2.13 versus 15.29 ± 1.22, respectively, p = 0.00001). In leukocytes of LC patients, a significant increase in the baseline level of structural aberrations of chromosomes, as well as micronuclei containing centromeres, was noted. Only in the sample of lung cancer patients were direct correlations of TL with the frequency of chromosomal type aberrations and with the frequency of paired fragments revealed. In the same sample, significant inverse correlations of TL with the frequency of binucleated leukocytes containing nuclear protrusions and with the frequency of cells entering apoptosis were recorded. Saccharin has been a controversial dietary component because of its potential to aggravate a number of human health disorders. The genotoxic properties of saccharin affect the normal karyokinesis process and results in chromosomal aberrations. In this backdrop, the current study has been formulated to evaluate the genotoxic potential of aqueous saccharin solution using Allium cepa L. assay. Three different concentrations of saccharin solution (0.25%, 0.5% and 1%) and a negative control were used for the root growth inhibition assay. The root length of Allium cepa significantly reduced under saccharin treatments and the effective concentration (EC50) value of the aqueous saccharin solution was determined as 0.587%. The genotoxic effects were determined for the root tip meristematic cells through mitotic index (MI) and mitotic depression (MD) at 3, 6 and 12 h of study. The MI registered highest value without saccharin at low exposure period (17.0 ± 1.5%) while a significant reduction with saccharin application culminated in the lowest MI for 1% saccharin solution at 12 h (3.4 ± 0.4%). MD was found to have a concomitant increase regarding the exposure of the chemical. Higher dose of saccharin for longer exposure time had a gradual decrease in the prophase and simultaneous increase in metaphase and anaphase. Further microscopic observations revealed eight different types of chromosomal aberrations in the A. cepa root tip cells with Anaphase Bridge assuming the highest score. A cytogenetic study of cell lines of mesenchymal multipotent stromal cells (MMSCs) isolated from human bone marrow, adipose tissue, placenta, and gingival mucosa was carried out. Karyological analysis using two chromosome-staining methods (GTG and mFISH) showed a normal diploid karyotype of most MMSC lines up to the fifth passage of cultivation. Clonal chromosomal translocations were detected in 3 of the 22 investigated MMSCs, which is 14%. Analysis of 22 MMSC lines revealed a rather low frequency of cells with chromosomal aberrations (CAs) (within 5%). Among cells with chromosomal damage (3–5%) identified by mFISH analysis of 1440 metaphase plates, unstable chromosomal aberrations (fragments, dicentric chromosomes) account for about 65% of all damage, and stable (translocations) account for 35%. These results confirm the importance of cytogenetic studies of MMSC cell lines intended for medical purposes. Gathering data on chromosomal and karyotypic variability at different periods of cultivation will make it possible to determine the acceptable limits for the selection of genetically stable diploid MMSC cell lines in order to prevent undesirable consequences of their use for therapeutic purposes. In this study, the protective effects of trans-resveratrol (t-resv) against 1,4-dioxane-induced toxicity in meristematic cells were investigated. For this purpose, Allium test was used and the alterations in all experimental groups were examined by using physiological, cytogenetic, biochemical, and anatomical parameters. In order to elucidate the toxicity mechanism, interactions of 1,4-dioxane and intracellular antioxidant molecules were investigated by molecular docking. As a result of the analysis, it was determined that 1,4-dioxane causes serious abnormalities in Allium cepa meristematic cells. In 1,4-dioxane-treated group, germination percentage was regressed 1.6 times, root length was reduced 12.7 times, and weight gain was decreased 7.7 times compared to control group. T-resv administration with 1,4-dioxane resulted in an improvement in physiological parameters and reduced the relative injury rate from 0.4 to 0.16. Mitotic index (MI), micronucleus (MN), and chromosomal abnormality (CAs) frequencies were investigated as cytogenetic parameters. 1,4-Dioxane decreased MI index, and increased CAs and MN frequency. In addition, it was determined by the comet test that 1,4-dioxane caused deterioration in DNA integrity. T-resv treatment was found to cause a dose-dependent improvement in genotoxic effects. Changes in the antioxidant system in all experimental groups were determined by measuring malondialdehyde (MDA) and glutathione (GSH) levels, superoxide dismutase (SOD), and catalase (CAT) enzyme activities. 1,4-Dioxane caused alterations in all tested parameters, causing deterioration in the oxidant/antioxidant balance in the cell. A 200-mg/L t-resv+1,4-dioxane treatment caused a 1.9-fold decrease in MDA level which is indicator of lipid peroxidation compared to only 1,4-dioxane-treated group. The mechanism of the disruption in antioxidant/oxidant dynamics and genetic integrity was elucidated by molecular docking analysis of 1,4-dioxane with antioxidant molecules and DNA. In 1,4-dioxane treatment group, anatomical changes such as cell deformation, flattened cell nucleus, and thickening of cortex cell wall were observed. The frequency of these changes decreased with t-resv administration. As a result, it was determined that 1,4-dioxane caused a versatile toxicity in A. cepa meristematic cells, while t-resv was found to have a dose-dependent protective feature against 1,4-dioxane-induced toxicity. Copper oxychloride gained great importance due to its broad-spectrum antifungal action to combat various fungal diseases of plants. However, excess quantity of cupric fungicides on plants causes enzymatic changes and toxic effects. Thus, the current study was aimed to investigate the cytotoxicity and genotoxicity of copper oxychloride on Allium cepa root cells. The root growth, mitotic index (MI), chromosomal aberrations (CAs), and DNA damage were assessed through root growth inhibition, A. cepa ana-telophase, and alkaline comet assays. Furthermore, molecular docking was performed to evaluate binding affinities of two copper oxychloride polymorphs (atacamite and paratacamite) on DNA. In root growth inhibition test, onion root length was statistically significantly decreased by changing the copper oxychloride concentration from lower (2.64±0.11 cm) to higher (0.92±0.12 cm). Concentration- and time-dependent decrease in MI was observed whereas increase in CAs such as disturbed ana-telophase, chromosome laggards, stickiness, anaphase bridges, and DNA damage were caused by the copper oxychloride on A. cepa root cells. Molecular docking results revealed that the two main polymorphs of copper oxychloride (atacamite and paratacamite) bind selectively to G and C nucleotides on the B-DNA structure. It is concluded that the atacamite- and paratacamite-induced DNA damage may be through minor groove recognition and intercalation. Findings of the current study revealed the cytotoxic and genotoxic effects of copper oxychloride on A. cepa root cells. Objective: It includes the investigation of the toxic effect of the food preservative benzoic acid during the development of Drosophila melanogaster. Method: Male and female flies of Drosophila melanogaster were exposed to various concentrations of benzoic acid (50–3000 mg/l) in culture medium. Total numbers of pupae and young adults generated in each culture tube were counted for 100 h. Ten randomly selected lacto-acetic-orcein stained polytene chromosomes of the third instar larvae of each group were made to assess chromosomal aberrations. Numbers of hemocytes or neuroblasts in third instar larvae with altered nuclear morphologies were scored in 20 randomly selected DAPI stained cells of each group in a single slide. Results: Benzoic acid acted in a dose dependent manner decreasing the population of pupae and young adults but their significant reductions (*p < 0.05, **p < 0.0001) were noticed under the exposure of the flies to ≥ 250 mg/l of this food additive. Developmental inhibition of the flies was accompanied with significant (*p < 0.05, **p < 0.0001) increase in the number of structural abnormalities (ectopic pairing, inversion loops, chromosomal shrinkage, breakage, fusion and asynapsis) of the larval polytene chromosomes due to benzoic acid ≥ 250 mg/l. DAPI staining of the nuclei of larval hemocytes and neuroblasts revealed their significant (*p < 0.05, **p < 0.0001) structural alterations (fragmented, triangular, diffused, spearhead, small globules, etc.) under the dietary exposure of the flies to benzoic acid ≥ 250 mg/l and ≥ 500 mg/l, respectively. Conclusion: Larval Chromosomal abnormalities and damages in the nuclei of the hemocytes and neuroblasts might reflect inhibition of gene expressions that reduced their defensive functions and growth promoting activities. As Drosophila melanogaster possesses substantial similarity with human genome, results of our study emphasize the vulnerability of developing human to this food additive due to maternal dietary intake during pregnancies. Industrial effluents contain hazardous substances that can be a serious threat to the agriculture and human health. In the present study, the cytotoxic and genotoxic impacts of agricultural soil from the industrial area of Dera Bassi (Punjab, India) have been evaluated. Assays such as defects in DNA repair in K-12 mutants of Escherichia coli and chromosomal aberrations in Allium cepa were used to estimate the acute toxicity and chromosomal mutagenesis, respectively. Atomic absorption spectrometry and GC-MS analysis revealed contamination of the soil with high concentrations of heavy metals and organic compounds, respectively. Dichloromethane extract of site I soil sample caused maximum damage to 40 μL mL−1 DNA repair defective mutants and showed 38 and 49% survival in lexA and recA mutants, respectively, which was least among all the sites. In A. cepa test, an inverse relationship between soil extract concentration and the mitotic index was observed. Exposure of growing roots of A. cepa to soil extracts induced chromosomal abnormalities and alterations in mitotic phases in root tip cells. The study concludes that agricultural sites near the industrial area were contaminated with genotoxic and mutagenic compounds. We studied the effect of technogenic radiation on the degree of promoter methylation in genes involved in apoptosis in blood lymphocytes of workers exposed to long-term γ-radiation during their professional activities. Blood samples for the analysis were obtained from 11 conventionally healthy men aged from 54 to 71 years (mean 66 years), workers of the Siberian Group of Chemical Enterprises working experience from 27 to 40 years (mean 30 years); the external exposure dose was 175.88 mSv (158.20-207.81 mSv). In all examined subjects, the degree of methylation of the promoters of apoptosis-related genes ranged from 0.22 to 50.00%. A correlation was found between the degree of methylation of BCLAF1 promoters (p=0.035) with the age of workers, BAX promoters (p=0.0289) with high content of aberrant cells, and APAF1 promoters (p=0.0152) with increased number of dicentric chromosomes. A relationship was found between the dose of external irradiation and the degree of methylation of gene promoters of BAD (p=0.0388), BID (р=0.0426), and HRK (р=0.0101) genes. [ABSTRACT FROM AUTHOR]

By increasing the undesirable side effects of synthetic food pigments on human health, using safe natural food pigment become an urgent issue. Incorporate corn starch with oils conducted a high impact on red pigment production by Monascus purpureus. Fortification the medium with sesame oil raised the pigment production by 80% and the dry mass by 63% compared with free oil medium. Response surface methodology maximizes the production with 114.6% (12.8 A500) using medium constituents (g/l); Sesame oil 5; Corn starch 30; Yeast extract 1.5; KH2PO4 2.5 and MgSO4.7H2O 0.1. After evaluating red pigment stability in three common food components, citric acid showed a great effect on residual stability percentage compared with ascorbic and salicylic acid which decrease slightly the residual stability percentage at light and dark conditions. The mitotic index of red pigment was lower than the negative control at all tested concentrations. Different types of mitotic chromosomal abnormalities e.g. lagging chromosome, chromosomal bridge, chromosome and chromatin fragments, outside chromosome, chromosomal stickiness and micro nuclei were recorded. Insignificant increase in total mitotic aberrations percentage in all tested root tips treated with all concentrations of red pigment (1.23, 1.58, 1.63, 2.32 and 2.40%) compared with negative control (0.91%). There was a significant increase in total aberrations percentage after treatment with all concentrations (10, 15, 20 and 25%) of positive control (2.93, 3.00, 3.55 and 6.53 respectively) except (5%) which was insignificant (2.71%). In order to increase genetic variability for improvement and better adaptation in the present days' changing climate, present investigation has been done in Andrographis paniculata (Burm. f.) Nees (also known as Kalmegh in Ayurveda), an ethno-medicinal wild plant with highly immune-boosting properties. For the purpose, healthy, dry and homogeneous seeds were treated with four doses of gamma rays from Co-60 sources at the dose rate of 1.55 Gy per second; selected on the basis of LD50 i.e. 25 Gy (gray), 50, 100 and 200 Gy along with one set of non-irradiated seeds and sown in triplicates in a completely randomized block design (CRBD). Higher doses were detrimental hence not suggested for further mutation breeding experiments. However, lower doses (viz. 25 and 50 Gy) of gamma rays were stimulatory; had induced some significant (p > 0.5) variations in several lucrative traits of Andrographis paniculata (Kalmegh) viz. Plant height, leaf area, number of branches per plant, length of internodes, fruit length, and seeds per fruit. Some induced novel mutants (viz. bushy, dark green leaves mutant and tricotyledonous leaf mutant in M2 generation) have also been procured. Background: Chronic lymphocytic leukemia (CLL) is the most common form of adult leukemia. This disease is genetically heterogeneous, and approximately 85% of patients with CLL harbor chromosomal aberrations that are considered effective prognostic biomarkers. The most frequent aberrations include deletions in 13q14, followed by trisomy 12, and deletions in 11q22.3 and 17p13 (TP53). Currently, fluorescence in situ hybridization (FISH) is the most widely used molecular cytogenetic technique to detect these aberrations. However, FISH is laborious, time-consuming, expensive, and has a low throughput. In contrast, multiplex ligation-dependent probe amplification (MLPA) is a reliable, cost-effective, and relatively rapid technique that can be used as a first-line screening tool and complement with FISH analysis. This study aimed to evaluate the contributions of MLPA as a routine standalone screening platform for recurrent chromosomal aberrations in CLL in comparison to other procedures. Thirty patients with CLL were screened for the most common genomic aberrations using MLPA with SALSA MLPA probemix P038-B1 CLL and FISH. Results: In 24 of the 30 cases (80%), the MLPA and FISH results were concordant. Discordant results were attributed to a low percentage of mosaicism. Moreover, the MLPA probemix contains probes that target other genomic areas known to be linked to CLL in addition to those targeting common recurrent CLL aberrations. Conclusions: The usage of MLPA as the first screening platform followed by FISH technique for only the negative cases is the most appropriate approach for CLL diagnosis and prognosis. The current study investigated the protective and antioxidant potential of Aegle marmelos leaf extract against Cadmium-induced alterations in hepatic and renal tissues of BALB/c mice. Cadmium (Cd) is known for its toxicity and induction of oxidative stress even at a low level of exposure with prolonged persistence in the environment. Its metabolism and excretion depend on the endogenous antioxidant defense system, which gets compromised. For the present study thirty BALB/c mice were equally divided into six groups, namely Control (normal saline via gavage for 10 days), Cadmium (intraperitoneal dose of 5 mg/kg b.wt. for 5 days), Aegle alone (500 mg/kg b.wt. for 10 days via gavage), Pre-treatment of Aegle (500 mg/kg b.wt. for 10 days+5 mg/kg b.wt. of Cd for 5 days), Post-treatment of Aegle (5 mg/kg b.wt. of Cd for 5 days+500 mg/kg b.wt. for 10 days) and Vehicle group (0.5 mL oil/kg b.wt. through gavage for 10 days). The liver and kidney tissues of different groups were screened for oxidative stress indices, antioxidant enzyme systems, hepatic-renal biomarkers, histopathological alterations, chromosomal aberrations, and micronucleus formation. Sub-acute Cadmium exposure caused marked oxidative damage and tissue dysfunction evident from increased lipid peroxidation, liver-kidney serum function markers, and a subsequent decrease in reduced glutathione, catalase, superoxide dismutase, glutathione-S-transferase, glutathione reductase (p < 0.001). Histopathological analysis exhibited both reversible and non-reversible alterations.Additionally, a marked increase in chromosomal aberrations and micronucleation was observed. Both pre-and post-treatments with Aegle significantly restored altered antioxidant enzyme status, serum function markers, histomorphological alterations and reduced the percentage of chromosomal aberrations and micronucleation in the treated groups. Aegle exerts its protective efficacy due to its rich bio-active phytoconstituents, which regulated Cadmium evoked reactive oxygen species (ROS) and enhanced endogenous defense system to combat the cyto-genotoxic alterations. Eutrophication is one of the environmental problems arising from the increase of essential nutrient concentrations, mainly phosphorus and nitrogen. In contrast to excess phosphorus, the depletion of phosphate rock deposits used for the production of fertilizers compromises the food supply. Therefore, the development of technologies that propose the recovery of the phosphorus contained in eutrophic environments for its later use for agricultural fertilization purposes is very important to ensure global food security. This work aimed to evaluate the toxic potential of the sawdust (biosorbent previously used for phosphorus adsorption) in order to enable its application in agriculture. For this, toxicity experiments with Lactuca sativa (lettuce) and Allium cepa (onion) seeds were performed. The phytotoxic potential was assessed by means of the seed germination index and physiological parameters such as radicle and hypocotyl growth. Cytotoxicity, genotoxicity, and mutagenicity tests were also performed on onion seeds. From statistical tests, it was possible to affirm that the sawdust did not promote inhibition of seed germination and radicle and hypocotyl growth. No genotoxicity, cytotoxicity and, mutagenicity were observed, which allowed to state that the sawdust is not toxic to the onion species, which reinforces the possibility of application of the biosorbent for soil fertilization purposes. Therefore, the use of sawdust for phosphorus biosorption with the subsequent agricultural application is promising and quite important from a global food security point of view. TP53 (p53) is a pivotal player in tumor suppression with fifty percent of all invasive tumors displaying mutations in the TP53 gene. In the present study, we characterized colon cancer cells (HCT116 p53 −/−) with TP53 deletion, a sub-line derived from HCT116-p53 +/+ cells. RNA sequencing and network analyses were performed to identify novel drug resistance mechanisms. Chromosomal aberrations were identified by multicolor fluorescence in situ hybridization (mFISH) and array comparative genomic hybridization (aCGH). Numerous genes were overexpressed in HCT116 p53 −/− cells: RND3/RhoE (235.6-fold up-regulated), DCLK1 (60.2-fold up-regulated), LBH (31.9-fold up-regulated), MYB (28.9-fold up-regulated), TACSTD2 (110.1-fold down-regulated), NRIP1 (81.5-fold down-regulated) and HLA-DMB (69.7-fold down-regulated) are among the identified genes with potential influence on multidrug resistance (MDR) and they are associated with cancer progression and tumorigenesis, according to previously published studies. Probably due to TP53 deletion, disturbances in DNA repair and apoptosis are leading to aberrancies in cellular and organismal organization, ultimately increasing tumorigenesis and cancer progression potential. With NFκB, PI3K and HSP70, being at the center of merged protein network, and TH1-2 pathways, being among the influenced pathways, it can be speculated that the inflammatory pathway contributes to a resistance phenotype together with cell cycle regulation and heat-shock response. HCT116-p53 −/− cells have more chromosomal aberrations, gains and losses in copy numbers than HCT116-p53 +/+ cells. In conclusion, numerous genomic aberrations, which might be associated with yet unknown drug resistance mechanisms, were identified. The application of pesticides to get more agricultural products is increasing day by day. The use of a huge amount of pesticides raises public concerns about safety. Tetraconazole is a widely used and successful fungicide. Possible toxic, cytotoxic, and genotoxic effects of different doses of tetraconazole (1.00 mg/L, 5.00 mg/L, and 10.00 mg/L) were evaluated on the meristematic cells of Allium cepa L. root tips by means of physiological, cytogenetic, biochemical, and anatomical parameters. EC50 value for tetraconazole in terms of growth inhibition was calculated as 6.7 mg/L. Increasing doses of tetraconazole resulted in reduced germination ratio, root length, and weight gain. Total activities of superoxide dismutase (SOD) and catalase (CAT) enzymes as well as malondialdehyde (MDA) content were increased as a result of oxidative stress. As an evidence of genotoxicity, mitotic index (MI) level decreased, while scores for micronucleus (MN) and chromosomal aberrations (CAs) rose. In addition, various meristematic cell damages were detected in root tips of tetraconazole applied bulbs. As a result, the multiple toxic, cytotoxic, and genotoxic effects of tetraconazole fungicide were demonstrated through a wide range of parameters on A. cepa, which was found to be a versatile tool for testing hazardous pesticides. The Itapemirim River is considered one of the most important water resources in the state of Espírito Santo, Brazil. However, environmental problems due to continuous anthropogenic contamination are threatening its potential use. This study assessed water quality by analyzing abiotic and toxicogenetic aspects of the water from four stations along the river. Samples were collected in both dry and rainy seasons. Most of the abiotic variables were below the threshold established by CONAMA Resolution No. 357/2005, and so were most of the metals. However, Al and Cu contents were above those allowed by legislation, ranging from 0.2 to 0.9 mg/L. Regarding toxicogenetic aspects, genotoxic effects were observed in meristematic cells of Allium cepa, in micronucleus test and comet assay of Oreochromis niloticus, and CHO-K1 cells. Mutagenic effects were significant at RI 02 (0.34), RI 03 (0.46), and RI 04 (0.12) stations on the first campaign in A. cepa F1 cells, compared to the negative control (0.0). The second campaign revealed the same results, but with the addition of samples from RI 01 (0.17) and RI 03 (0.18) showing mutagenicity in the micronucleus test with fish erythrocytes when compared to the negative control (0.3). Essentially, all the samples evaluated in both campaigns showed damage in A. cepa, O. niloticus, and CHO-K1 cells, thus demonstrating that the water quality of the Itapemirim River is compromised and requires action plans for its recovery.
