L-Proline (l-Pro) is a non-essential amino acid which, in high concentrations, can cause neurological problems including seizures, although the causative mechanism for this is unclear. Here, we studied the impact of physiological levels of proline on brain energy metabolism and investigated the metabolism of l-Pro itself, using the cortical brain tissue slice and stable isotope labelling from [1-13 C]glucose and [1,2-13 C]acetate detected by NMR spectroscopy and LCMS. l-Pro was actively taken up by the slices and significantly reduced the total metabolic pools of all measured metabolites with glutamine the least affected, while reducing net flux of 13C into glycolytic byproducts (lactate and alanine). Conversely, net flux into Krebs cycle intermediates was increased, suggesting that L-Pro at lower concentrations was driving increased mitochondrial activity in both neurons and glia at the expense of glycolysis and metabolic pool sizes. As there was no evidence of metabolism of [1-13 C] l-Pro in slices under normo-glycemic conditions, the effect of proline on metabolism was not due to displacement of metabolites by added l-Pro. Comparison of the metabolic fingerprint generated by l-Pro in slices metabolizing [3-13 C]pyruvate with that generated by ligands active in the GABAergic system suggested that l-Pro may engender effects similar to that of the inhibitory neurotransmitter and metabolite γ-aminobutyric acid (GABA), in line with previous suggestions that l-Pro may be a GABA mimetic in addition to its role as a modulator of mitochondrial metabolism. Cytokine storm is a hallmark for acute systemic inflammatory disease like sepsis. Intrinsic microbiome-derived short-chain fatty acid (SCFAs) like acetate modulates immune cell function and metabolism has been well studied. However, it remains poorly investigated about the effects and the underlying mechanism of exogenous acetate in acute inflammation like sepsis. Here, we observed that serum acetate accumulates in patients undergoing abdominal gastrointestinal surgery and in septic mice. Short exposure to high-dose exogenous acetate protects mice from sepsis by inhibiting glycolysis in macrophages, both in vivo and in vitro. Hypoxia-inducible factor 1 subunit alpha (HIF-1α) stabilization or overexpression reverses the decreased glycolysis and pro-inflammatory cytokine production in macrophages and abrogates acetate's protective effect in septic mice. Meanwhile, we also found acetyl-CoA synthetase-2, but not GPR41 or GPR43, plays a key role in acetate's immunosuppressive effect. Acetate transiently increases acetyl-coenzyme A production, promoting histone acetylation and decreasing acetyl-transfer to NF-κB p65. These findings suggest that short exposure to mM-level acetate inhibits macrophage immune response linked to HIF-1α-dependent glycolysis. Mixotrophic cultivation of Haematococcus lacustris is one of the most promising strategies to produce natural astaxanthin. During mixotrophic growth, microalgae assimilate and metabolize organic carbon in addition to photosynthetic growth, resulting in increased biomass productivity. Several studies have evaluated the effect of different organic carbon sources on mixotrophic growth in various microalgae species. However, knowledge of detailed growth kinetics as a function of substrate concentration and light intensity is lacking. In this study, the growth kinetics of H. lacustris using four different carbon sources and the effect of light under mixotrophic and photoautotrophic conditions are described. Mixotrophic cultivation showed significant differences in respect to applied substrate and achieved maximum specific growth rates of 0.91 ± 0.13, 0.19 ± 0.05, 0.36 ± 0.05, and 0.23 ± 0.05 day−1, for acetate, methanol, glucose, and glycerol, respectively. Optimal growth at mixotrophic conditions using acetate was 1.8 times higher than the sum of hetero- and photoautotrophic growth. Furthermore, the optimum light intensity was 1.3 times higher for mixotrophic than for autotrophic growth. Thus, mixotrophy increases light intensity tolerance. These results indicate a strong interconnection between carbon metabolism and photosynthetic activity and lay the foundation for more detailed mathematical models describing the mixotrophic growth of H. lacustris. Waste glycerol is produced in excess by several industries, such as during biodiesel production. In this work, the metabolic versatility of anaerobic sludge was explored towards waste glycerol valorization. By applying different environmental (methanogenic and sulfate-reducing) conditions, three distinct microbial cultures were obtained from the same inoculum (anaerobic granular sludge), with high microbial specialization, within three different phyla (Thermodesulfobacteriota, Euryarchaeota and Pseudomonadota). The cultures are capable of glycerol conversion through different pathways: (i) glycerol conversion to methane by a bacterium closely related to Solidesulfovibrio alcoholivorans (99.8% 16S rRNA gene identity), in syntrophic relationship with Methanofollis liminatans (98.8% identity), (ii) fermentation to propionate by Propionivibrio pelophilus strain asp66 (98.6% identity), with a propionate yield of 0.88 mmol mmol−1 (0.71 mg mg−1) and a propionate purity of 80–97% and (iii) acetate production coupled to sulfate reduction by Desulfolutivibrio sulfoxidireducens (98.3% identity). In conclusion, starting from the same inoculum, we could drive the metabolic and functional potential of the microbiota towards the formation of several valuable products that can be used in industrial applications or as energy carriers. The factors limiting micropollutant biodegradation in the environment and how to stimulate this process have often been investigated. However, little information is available on the capacity of microbial communities to retain micropollutant biodegradation capacity in the absence of micropollutants or to reactivate micropollutant biodegradation in systems with fluctuating micropollutant concentrations. This study investigated how a period of 2 months without the addition of micropollutants and other organic carbon affected micropollutant biodegradation by a micropollutant-degrading microbial community. Stimulation of micropollutant biodegradation was performed by adding different types of dissolved organic carbon (DOC)—extracted from natural sources and acetate—increasing 10 × the micropollutant concentration, and inoculating with activated sludge. The results show that the capacity to biodegrade 3 micropollutants was permanently lost. However, the biodegradation activity of 2,4-D, antipyrine, chloridazon, and its metabolites restarted when these micropollutants were re-added to the community. Threshold concentrations similar to those obtained before the period of no substrate addition were achieved, but biodegradation rates were lower for some compounds. Through the addition of high acetate concentrations (108 mg-C/L), gabapentin biodegradation activity was regained, but 2,4-D biodegradation capacity was lost. An increase of bentazon concentration from 50 to 500 µg/L was necessary for biodegradation to be reactivated. These results provide initial insights into the longevity of micropollutant biodegradation capacity in the absence of the substance and strategies for reactivating micropollutant biodegrading communities. Production of medium chain length polyhydroxyalkanoate (mcl-PHA) was attempted using Pseudomonas gessardii NIBRBAC000509957, which was isolated from Sunchang, Jeollabuk-do, Republic of Korea (35°24′27.7"N, 127°09′13.0"E) and effectively utilized acetate and formate as carbon sources. We first evaluated the utilization of acetate as a carbon source, revealing optimal growth at 5 g/L acetate. Then, formate was supplied to the acetate minimal medium as a carbon source to enhance cell growth. After overexpressing the acetate and formate assimilation pathway enzymes, this strain grew at a significantly higher rate in the medium. As this strain naturally produces PHA, it was further engineered metabolically to enhance mcl-PHA production. The engineered strain produced 0.40 g/L of mcl-PHA with a biomass content of 30.43% in fed-batch fermentation. Escherichia coli MLB (MG1655 ΔpflB ΔldhA), which can hardly grow on glucose with little succinate accumulation under anaerobic conditions. Two-stage fermentation is a fermentation in which the first stage is used for cell growth and the second stage is used for product production. The ability of glucose consumption and succinate production of MLB under anaerobic conditions can be improved significantly by using acetate as the solo carbon source under aerobic condition during the two-stage fermentation. Then, the adaptive laboratory evolution (ALE) of growing on acetate was applied here. We assumed that the activities of succinate production related enzymes might be further improved in this study. E. coli MLB46-05 evolved from MLB and it had an improved growth phenotype on acetate. Interestingly, in MLB46-05, the yield and tolerance of succinic acid in the anaerobic condition of two-stage fermentation were improved significantly. According to transcriptome analysis, upregulation of the glyoxylate cycle and the activity of stress regulatory factors are the possible reasons for the elevated yield. And the increased tolerance to acetate made it more tolerant to high concentrations of glucose and succinate. Polycystic ovarian syndrome (PCOS) is well documented as the leading cause of infertility globally, and its long-term complications include non-alcoholic fatty liver disease (NAFLD) which results in a cascade of events leading to liver cirrhosis as well as hepatic carcinoma. Paraoxonase-1 (PON-1) is an antiatherosclerotic component, which protects against hepatic injury. Short-chain fatty acids (SCFAs), particularly acetate, are synthesized by the gut microbiota and play a central role in metabolic regulation. The present study investigated the impact of acetate on PCOS-associated hepatic dysregulation and probable involvement of PON-1. Female Wistar rats (8-week-old) were assigned into groups: control (CTL), acetate (ACT), letrozole (LET), and LET + ACT. Induction of PCOS was by administration of letrozole (1 mg/kg) for 21 days. After PCOS was confirmed in the rats, acetate (200 mg/kg) was administered for a period of 6 weeks. PCOS animals were characterized with multiple ovarian cysts; excessive androgen; reduced insulin sensitivity; metabolic hormone (leptin and adiponectin) dysregulation; dyslipidemia; hepatic lipid peroxidation; lipotoxicity; increased hepatic caspase-6, as well as proinflammatory markers (NF-kB/SDF-1); and decreased antioxidant defense (NrF2) and HIF-1α. These perturbations were accompanied by suppression of PON-1 level. Nevertheless, administration of acetate mitigated these hepatic metabolic/cellular alterations. The present results provide evidence for the therapeutic potential of acetate in ameliorating hepatic dysmetabolism, inflammation, oxidative stress, and cellular apoptosis in PCOS through modulation of PON-1. The anodic electrooxidation of ethanol to value-added acetate is an excellent example of replacing the oxygen evolution reaction to promote the cathodic hydrogen evolution reaction and save energy. Herein, we present a colloidal strategy to produce Ni-Fe bimetallic alloy nanoparticles (NPs) as efficient electrocatalysts for the electrooxidation of ethanol in alkaline media. Ni-Fe alloy NPs deliver a current density of 100 mA·cm−2 in a 1.0 M KOH solution containing 1.0 M ethanol merely at 1.5 V vs. reversible hydrogen electrode (RHE), well above the performance of other electrocatalysts in a similar system. Within continuous 10 h testing at this external potential, this electrode is able to produce an average of 0.49 mmol·cm−2·h−1 of acetate with an ethanol-to-acetate Faradaic efficiency of 80%. A series of spectroscopy techniques are used to probe the electrocatalytic process and analyze the electrolyte. Additionally, density functional theory (DFT) calculations demonstrate that the iron in the alloy NPs significantly enhances the electroconductivity and electron transfer, shifts the rate-limiting step, and lowers the energy barrier during the ethanol-to-acetate reaction pathway. Drosophila melanogaster is unique among animal models because it has a fully defined synthetic diet available to study nutrient-gene interactions. However, use of this diet is limited to adult studies due to impaired larval development and survival. Here, we provide an adjusted formula that reduces the developmental period, restores fat levels, enhances body mass, and fully rescues survivorship without compromise to adult lifespan. To demonstrate an application of this formula, we explored pre-adult diet compositions of therapeutic potential in a model of an inherited metabolic disorder affecting the metabolism of branched-chain amino acids. We reveal rapid, specific, and predictable nutrient effects on the disease state consistent with observations from mouse and patient studies. Environmental concerns about residues and the traditional disposal methods are driving the search for more environmentally conscious processes, such as pyrolysis and gasification. Their main final product is synthesis gas (syngas) composed of CO, CO2, H2, and methane. Syngas can be converted into various products using CO-tolerant microorganisms. Among them, Rhodospirillum rubrum is highlighted for its biotechnological potential. However, the extent to which high doses of CO affect its physiology is still opaque. For this reason, we have studied R. rubrum behavior under high levels of this gas (up to 2.5 bar), revealing a profound dependence on the presence or absence of light. In darkness, the key variable affected was the lag phase, where the highest levels of CO retarded growth to more than 20 days. Under light, R. rubrum ability to convert CO into CO2 and H2 depended on the presence of an additional carbon source, such as acetate. In those conditions where CO was completely exhausted, CO2 fixation was unblocked, leading to a diauxic growth. To enhance R. rubrum tolerance to CO in darkness, a UV-accelerated adaptive laboratory evolution (UVa-ALE) trial was conducted to isolate clones with shorter lag phases, resulting in the isolation of clones 1.4-2B and 1.7-2A. The adaptation of 1.4-2B was mainly based on mutated enzymes with a metabolic function, while 1.7-3A was mostly affected at regulatory genes, including the anti-repressor PpaA/AerR. Despite these mutations having slight effects on biomass and pigment levels, they successfully provoked a significant reduction in the lag phase (−50%). Bacterial infections are a significant concern in eel aquaculture. Edwardsiella causes severe systemic infections and high mortality rates. This study aimed to demonstrate that synbiotic oral administration could improve eels' gut environment, health, and aquaculture productivity. After 1 month of administration in Japanese eel Anguilla japonica using synbiotics consisting of prebiotic 1-kestose, a fructooligosaccharide with a degree of polymerization of 3, and the probiotic Lactiplantibacillus plantarum FM8, isolated from fermented food, we discovered that feed efficiency was improved by more than 20%. This may partly be because the synbiotics significantly increased intestinal acetate concentrations (P = 0.0144). 16S rRNA gene amplicon sequencing and linear discriminant analysis effect size revealed that the relative abundance of the Romboutsia was significantly higher, and that of the Edwardsiella was significantly lower in the eel intestine of the synbiotic group (P = 0.0166 and 0.0497, respectively). The BLAST search revealed that all amplicon sequence variants classified as genus Edwardsiella had > 98% identity with the 16S rRNA gene of Edwardsiella tarda, Edwardsiella piscicida, or Edwardsiella anguillarum. Our synbiotic administration in eels exhibited a breakthrough effect in significantly reducing Edwardsiella and increasing acetate concentrations in the intestine, suggesting that our synbiotics could reduce the incidence of eel diseases and improve aquaculture productivity. Despite the effectiveness of doxorubicin (DOX) in the management of a wide range of cancers, a major challenge is its cardio-toxic effect. Oxidative stress, inflammation, and apoptosis are major pathways for the cardiotoxic effect of DOX. On the other hand, acetate reportedly exerts antioxidant, anti-inflammatory, and anti-apoptotic activities. This particular research assessed the impact of acetate on cardiotoxicity induced by DOX. Mechanistically, acetate dramatically inhibited DOX-induced upregulation of xanthine oxidase and uric acid pathway as well as downregulation of Nrf2/HO-1 signaling and its upstream proteins (reduced glutathione peroxidase, superoxide dismutase, glutathione-S-transferase, glutathione, and catalase, glutathione reductase). In addition, acetate markedly attenuated DOX-driven rise inTNF-α, NFkB IL-6 and IL-1β expression, and myeloperoxidase activity. Furthermore, acetate significantly ameliorated DOX-led suppression of Bcl-2 and Ca2+-ATPase activity and upregulation of Bax, caspase 3, and caspase 9 actions. Improved body weight, heart structural integrity, and cardiac function as depicted by cardiac injury markers convoyed these cascades of events. Summarily, the present study demonstrated that acetate protects against DOX-induced cardiotoxicity by upregulating Nrf2/HO-1 signaling and downregulating NFkB-mediated activation of Bax/Bcl-2 and caspase signaling. Renewable energy sources draw attention day by day and studies in this field are increasing. However, in addition to producing energy, the efficient use of the energy produced is of greater importance. Thermoelectric materials come into play at this stage, but the production of materials at the desired level in terms of efficiency is still a problem. In this study, CaMnO3 structure with perovskite structure, which allows the production of many high-tech materials with its variable structure, was produced with acetate starting materials. Continuously prolonged cardiac hypertrophy results in maladaptive myocardial remodeling, which affects cardiac function and can eventually lead to heart failure. Short-chain fatty acids (SCFAs), including acetate, propionate, and butyrate, have been reported to be associated with cardiovascular diseases (CVD). Gut microbiota may mediate between dietary fiber and SCFA effects on cardiac hypertrophy. The mice model of isoproterenol (ISO)-induced cardiac hypertrophy was constructed and verified for physiological, functional, and fibrotic alterations in this study. Both high-fiber and acetate diet improved physiological indexes, ameliorated cardiac functions, and relieved fibrotic alterations in model mice hearts; collectively, cardiac hypertrophy in mice receiving both high-fiber and acetate diet improved. Following 16s rDNA sequencing and integrative bioinformatics, analyses indicated that both high-fiber and acetate diet caused alterations in mice gut microbiota compared with the ISO group, including OTU composition and abundance. In conclusion, high-fiber and acetate diet improve the physiological status, cardiac functions, and fibrotic alterations in ISO-induced hypertrophic mice. Besides, considering the alterations in mice gut microbiota in response to single ISO, both high-fiber and acetate diet treatment, gut microbiota might mediate the favorable benefits of both high-fiber and acetate diet on cardiac hypertrophy. Background Staphylococcus aureus spreads its infections through bioflms. This usually happens in the stationary phase of S. aureus growth where it utilizes accumulated acetate as a carbon source via the phosphotrans-acetylase-acetate kinase (Pta-Ack) pathway. In which acetate kinase (ackA) catalyzes the substrate-level phosphorylation, a vital secondary energy-yielding pathway that promotes bioflms formation aids bacterium survival in hostile environments. In this study, we describe the cloning, sequencing, and expression of S. aureus ackA gene. The expression analysis of ackA gene in methicillin-resistant strains of S. aureus (MRSA) correlates with ackA activity and bioflm units. The uniqueness of ackA was analyzed by using in silico methods. Results Elevated ackA gene expression was observed in MRSA strains, which correlates with increased ackA activity and bioflm units, explaining ackA role in MRSA growth and pathogenicity. The pure recombinant acetate kinase showed a molecular weight of 44 kDa, with enzyme activity of 3.35 ± 0.05 μM/ml/min. The presence of ACKA-1, ACKA-2 sites, one ATP, and fve serine/threonine-protein kinase sites in the ackA gene (KC954623.1) indicated that acetyl phosphate production is strongly controlled. The comparative structural analysis of S. aureus ackA with ackA structures of Mycobacterium avium (3P4I) and Salmonella typhimurium (3SLC) exhibited variations as indicated by the RMSD values 1.877 Å and 2.141 Å respectively, explaining why ackA functions are diferently placed in bacteria, concurring its involvement in S. aureus pathogenesis. Conclusions Overall fndings of this study highlight the correlation of ackA expression profoundly increases survival capacity through bioflm formation, which is a pathogenic factor in MRSA and plays a pivotal role in infection spreading. Acetate, a major by‐product of glycolytic metabolism in Escherichia coli and many other microorganisms, has long been considered a toxic waste compound that inhibits microbial growth. This counterproductive auto‐inhibition represents a major problem in biotechnology and has puzzled the scientific community for decades. Recent studies have however revealed that acetate is also a co‐substrate of glycolytic nutrients and a global regulator of E. coli metabolism and physiology. Here, we used a systems biology strategy to investigate the mutual regulation of glycolytic and acetate metabolism in E. coli. Computational and experimental analyses demonstrate that decreasing the glycolytic flux enhances co‐utilization of acetate with glucose. Acetate metabolism thus compensates for the reduction in glycolytic flux and eventually buffers carbon uptake so that acetate, rather than being toxic, actually enhances E. coli growth under these conditions. We validated this mechanism using three orthogonal strategies: chemical inhibition of glucose uptake, glycolytic mutant strains, and alternative substrates with a natively low glycolytic flux. In summary, acetate makes E. coli more robust to glycolytic perturbations and is a valuable nutrient, with a beneficial effect on microbial growth. Abstract Mutations in AT‐rich interactive domain‐containing protein 1A (ARID1A) cause Coffin‐Siris syndrome (CSS), a rare genetic disorder that results in mild to severe intellectual disabilities. However, the biological role of ARID1A in the brain remains unclear. In this study, we report that the haploinsufficiency of ARID1A in excitatory neurons causes cognitive impairment and defects in hippocampal synaptic transmission and dendritic morphology in mice. Similarly, human embryonic stem cell‐derived excitatory neurons with deleted ARID1A exhibit fewer dendritic branches and spines, and abnormal electrophysiological activity. Importantly, supplementation of acetate, an epigenetic metabolite, can ameliorate the morphological and electrophysiological deficits observed in mice with Arid1a haploinsufficiency, as well as in ARID1A‐null human excitatory neurons. Mechanistically, transcriptomic and ChIP‐seq analyses demonstrate that acetate supplementation can increase the levels of H3K27 acetylation at the promoters of key regulatory genes associated with neural development and synaptic transmission. Collectively, these findings support the essential roles of ARID1A in the excitatory neurons and cognition and suggest that acetate supplementation could be a potential therapeutic intervention for CSS.

This work presents the investigation of ultrasonic speed propagation waves and aqueous solutions densities of acetates and sulfates of single and double- charged cations in a wide range of salt concentration. The variation of the relative adiabatic compressibility of the solution on the salt concentration has two or several straight lines. For each salt at a certain concentration (Cp), the transition from one straight line to another is occurred. Each of these lines corresponds to a certain value of the adiabatic compressibility of water. It is shown that presence of ions decreases the adiabatic compressibility of water. The hydrate numbers and sizes of nanoparticles (hydrated ions) are estimated as a function of the salt concentration. The obtained results show that the number of hydration decreases stepwise with increasing salt concentration, i.e., in certain intervals the salt concentration remains constant. The evaluated hydrate numbers and radii of hydrated ions (nanoparticle sizes) by the acoustic method are in a good agreement with the literature data. [ABSTRACT FROM AUTHOR]

Introduction/objectives: Persistent hyperuricemia is a key factor in gout; however, only 13.5% of hyperuricemic individuals manifest the disease. The gut microbiota could be one of the many factors underlying this phenomenon. We aimed to assess the difference in taxonomic and predicted functional profiles of the gut microbiota between asymptomatic hyperuricemia (AH) individuals and gout patients. Methods: The V3–V4 region of the 16S rRNA gene of the gut microbiota of AH individuals, gout patients, and controls was sequenced. Bioinformatic analyses were carried out with QIIME2 and phyloseq to determine the difference in the relative abundance of bacterial genera among the study groups. Tax4fun2 was used to predict the functional profile of the gut microbiota. Results: AH individuals presented a higher abundance of butyrate- and propionate-producing bacteria than gout patients; however, the latter had more bacteria capable of producing acetate. The abundance of Prevotella genus bacteria was not significantly different between the patients but was higher than that in controls. This result was corroborated by the functional profile, in which AH individuals had less pyruvate oxidase abundance than gout patients and less abundance of an enzyme that regulates glutamate synthetase activation than controls. Conclusion: We observed a distinctive taxonomic profile in AH individuals characterized by a higher abundance of short-chain fatty acid-producing bacteria in comparison to those observed in gout patients. Furthermore, we provide scientific evidence that indicates that the gut microbiota of AH individuals could provide anti-inflammatory mediators, which prevent the appearance of gout flares. Purpose of Review: Epidemiologic studies and clinical trials have demonstrated the benefits of dietary fiber. This occurs through a combination of the physiochemical properties of fiber and through microbial fermentation that occurs in the colon which result in the production of short-chain fatty acids (SCFA). The purpose of this review is to highlight the physiochemical properties of fiber that result in the range of physiologic effects and to review the literature on the health benefits of acetate, propionate, and butyrate. Recent Findings: Of the variety of properties and functions exerted by dietary fibers, the fermentability and production of SCFA's are emphasized in this review. Studies done in both animal and humans reveal the anti-obesity, anti-inflammatory, and possible anti-neoplastic roles SCFAs exert at the mucosal level. Summary: Many clinical questions remain regarding the optimal dose, type, and method of delivery of fiber to exert the desired beneficial effects. It has the potential to be used in the management of clinical symptoms, prevention of disease, and improvement in human health. Acetyl-CoA synthetase (ACS) is the enzyme that irreversibly catalyzes the synthesis of acetyl-CoA from acetate, CoA-SH, and ATP via acetyl-AMP as an intermediate. In this study, we demonstrated that AcsA1 (MSMEG_6179) is the predominantly expressed ACS among four ACSs (MSMEG_6179, MSMEG_0718, MSMEG_3986, and MSMEG_5650) found in Mycobacterium smegmatis and that a deletion mutation of acsA1 in M. smegmatis led to its compromised growth on acetate as the sole carbon source. Expression of acsA1 was demonstrated to be induced during growth on acetate as the sole carbon source. The acsA1 gene was shown to be negatively regulated by Crp1 (MSMEG_6189) that is the major cAMP receptor protein (CRP) in M. smegmatis. Using DNase I footprinting analysis and site-directed mutagenesis, a CRP-binding site (GGTGA-N6-TCACA) was identified in the upstream regulatory region of acsA1, which is important for repression of acsA1 expression. We also demonstrated that inhibition of the respiratory electron transport chain by inactivation of the major terminal oxidase, aa3 cytochrome c oxidase, led to a decrease in acsA1 expression probably through the activation of CRP. In conclusion, AcsA1 is the major ACS in M. smegmatis and its gene is under the negative regulation of Crp1, which contributes to some extent to the induction of acsA1 expression under acetate conditions. Purpose: Polycystic ovary syndrome (PCOS) is a complex reproductive event that is delineated by endocrine/metabolic disorders. Alteration of kisspeptin status in the hypothalamus and adipose tissue is critical to increased endocrine/metabolic derangements in PCOS individuals, aggravating the clinical manifestation of PCOS and its complications. Short chain fatty acids (SCFAs) are crucial modulators of metabolic homeostasis. However, the role of SCFAs, particularly acetate on hypothalamic-adipose kisspeptin status (HAKS) in PCOS model is unknown. The present study hypothesized acetate as a key player in restoration of deranged HAKS, associated with experimental PCOS model. Methods: Three groups (n = 6/group) of female Wistar rats (120–150 g) were used. The groups were treated (po) for 21 days with vehicle, letrozole (1 mg/kg) with/without acetate (200 mg/kg) respectively. Results: Letrozole-treated animals had impaired glucose homeostasis, elevated testosterone, leptin and LH/FSH ratio and decreased GnRH and adiponectin with ovarian tissues revealing degenerated follicles and disrupted morphology. These animals also showed increased concentration of hypothalamic triglyceride (TG)/total cholesterol (TC), free fatty acid (FFA), and decreased concentration of TG/TC/FFA in visceral adipose tissue (VAT) with an increase in hypothalamic and VAT malondialdehyde, NF-κB/TNF-α and decreased glutathione/G6PD and hypothalamic but not VAT kisspeptin. Immunohistochemical analysis revealed the expression of NLRP3 inflammasome in the hypothalamus and VAT and all these changes were attenuated by acetate. Conclusions: Altogether, the present results demonstrate that PCOS is characterized with hypothalamic-adipose inflammation, associated with immunohistochemical expression of NLRP3 with significant alteration of hypothalamic but not adipose kisspeptin. Salinity is an important abiotic stress which limits normal plant growth and productivity, particularly in arid and semi-arid areas. The foliar spray of acetic acid has recently been considered to enhance tolerance of abiotic stress, especially drought stress in plants but its effect on increased salinity tolerance in horticultural crops remains elusive. Hydroponically grown strawberry plants that were subjected to 40 mM sodium chloride for 90 days, were sprayed with 1- or 2-mM acetic acid (AA) three times including a week before, simultaneously and a week after application of salinity treatment. According to the results the electrolyte leakage, lipid peroxidation, and hydrogen peroxide, were increased by 45%, 46%, and 54%, respectively, under saline condition. While, the maximum quantum yield of PSII (Fv/Fm) decreased to 14% under salinity treatment. The maximum alleviation effect of salt stress was recorded in 1 mM foliar application of acetic acid, especially when applied 1 week before and simultaneous with salinity stress, resulting in significant decreases in electrolyte leakage to 13% and 15%, hydrogen peroxide to 30% and 32%, respectively, while the decline in lipid peroxidation was only observed in 1 week before salt stress to 39% compared to the control. The fruit yield reduced about 71% under 40 mM NaCl compared to the control, however, yield reduction was quite lower under 1- and 2-mM acetic acid treatments (31% and 45%, respectively). Acetic acid considerably increased antioxidant enzymes' activity including peroxidase (POD), superoxide dismutase (SOD), and catalase (CAT) as well as the concentration of non-enzymatic antioxidants such as phenolic compounds under salinity stress conditions, especially when applied 1 mM AA a week before and simultaneously with salinity (25% and 24%, respectively). These results confirmed that acetic acid treatment especially with 1 mM concentration a week before and simultaneously with salinity treatment can be considered to improve the salt tolerance in strawberry under saline conditions. Purpose: Urolithiasis is a common urologic disease. Higher consumption of vinegar was associated with a lower risk of urolithiasis. Recent studies reported that disorder of gut microbiota and injury of the tight junction of renal tubular epithelial cells were associated with the formation of renal calcium oxalate (CaOx) stones. We aimed to explore the mechanism of vinegar reduced renal CaOx stone formation by regulating gut microbiota and the tight junction of renal tubular epithelial cells. Methods: Thirty Sprague–Dawley rats were randomly allocated to control group, model group and vinegar group. Rats in control group got 2 ml/kg of sterile water by gavage. Model group rats were additionally supplied with drinking water with 1% (v/v) ethylene glycol (EG) every day. Rats in vinegar group had 1% (v/v) EG in drinking water and were gavaged with 2 ml/kg of vinegar (5% acetate) every day. Results: Vinegar reduced renal CaOx crystals and urinary oxalate. Vinegar increased the relative abundances of Ruminococcus gauvreauii, Ruminococcus torques, Ruminococcus-2, Moryella, Enterococcus, Alistipes, and Parabacteroides in the gut microbiota. Blood acetate increased in vinegar group. The renal tight junction occludin protein decreased in the model group and increased in the vinegar group. Studies in vitro verified that acetate could reverse the decline in occludin expression induced by CaOx crystals and inhibit CaOx crystal adhesion to cells. Purpose: NO3−-N reduction is the main way of soil nitrogen (N) loss, especially in saline-alkaline soil, and NO3−-N-reducing Fe(II) oxidation (NRFO) is an important but less known process for NO3−-N reduction. An anoxic cultivation experiment was carried out on coastal saline-alkaline paddy soil to determine the effect of the external environment on NRFO process and microbial community composition. Methods: A laboratory cultivation experiment was studied to elucidate the effect of pH and acetate on NRFO in saline-alkaline soil. Nine different treatments were designed with two factors: (1) three pH levels: 7.5, 8.0, and 8.5; (2) three acetate concentrations: 15 (C1), 20 (C2), and 25 mM (C3). Concentration of Fe(II), Fe(III), NO3−-N, NO2−-N, and NH4+ and dynamic changes in microorganisms were assessed within 36 days. Results: The reduction of NO3−-N showed a positive correlationship with Fe(II) oxidation and Fe(III) formation. In addition to Fe(II), acetate also participated in NO3−-N reduction as an electron donor. The reduction rate of NO3−-N was the highest in pH 8.5 + C3 treatment, in which the contribution of Fe(II) was 46.10%. The loss of NO3−-N was the least in pH 7.5 + C2 treatment, and the contribution of Fe(II) was 37.03%. Illumina high-throughput sequencing showed that the phyla of Proteobacteria and Firmicutes were enriched in all treatments. The classes of Alphaproteobacteria, Bacilli, and Gammaproteobacteria were found to be the dominant in all treatments. Pseudomonas, a NO3−-N reduction bacteria, was dominant in pH 8.5 + C3 treatment, in which the relative abundance was 24%. Meanwhile, a Fe(II) oxidizing bacteria, Acidovorax, was dominant in the pH 8.5 + C1 treatment, and the relative abundance was 15%. Conclusion: In coastal saline-alkaline paddy field, high C input and high pH (pH 8.5 + C3 treatment) could increase NO3−-N reduction, which might be the important reasons for N loss. Our study focused on a mouse model of obesity induced by a high-fat diet (HFD). We administered Semaglutide intraperitoneally (Ozempic ®—0.05 mg/Kg—translational dose) every seven days for six weeks. HFD-fed mice had higher blood glucose, lipid profile, and insulin resistance. Moreover, mice fed HFD showed high gut levels of TLR4, NF-kB, TNF-α, IL-1β, and nitrotyrosine and low levels of occludin, indicating intestinal inflammation and permeability, culminating in higher serum levels of IL-1β and LPS. Treatment with semaglutide counteracted the dyslipidemia and insulin resistance, reducing gut and serum inflammatory markers. Structural changes in gut microbiome were determined by 16S rRNA sequencing. Semaglutide reduced the relative abundance of Firmicutes and augmented that of Bacteroidetes. Meanwhile, semaglutide dramatically changed the overall composition and promoted the growth of acetate-producing bacteria (Bacteroides acidifaciens and Blautia coccoides), increasing hypothalamic acetate levels. Semaglutide intervention increased the number of hypothalamic GLP-1R+ neurons that mediate endogenous action on feeding and energy. In addition, semaglutide treatment reversed the hypothalamic neuroinflammation HDF-induced decreasing TLR4/MyD88/NF-κB signaling and JNK and AMPK levels, improving the hypothalamic insulin resistance. Also, semaglutide modulated the intestinal microbiota, promoting the growth of acetate-producing bacteria, inducing high levels of hypothalamic acetate, and increasing GPR43+ /POMC+ neurons. In the ARC, acetate activated the GPR43 and its downstream PI3K-Akt pathway, which activates POMC neurons by repressing the FoxO-1. Thus, among the multifactorial effectors of hypothalamic energy homeostasis, possibly higher levels of acetate derived from the intestinal microbiota contribute to reducing food intake. Background: Tumor equivalent uniform dose (EUD) is proposed as a predictor of patient outcome after liver radioembolization (RE) of hepatocellular carcinoma (HCC) and can be evaluated with 90Y-TOF-PET. The aim is to evaluate the correlation between PET-based tumors EUD and the clinical response evaluated with dual molecular tracer (11C-acetate and 18F-FDG) PET/CT post-RE. Methods: 34 HCC tumors in 22 patients were prospectively evaluated. The metabolic response was characterized by the total lesion metabolism variation (ΔTLM) between baseline and follow-up. This response allowed to compute a tumor control probability (TCP) as a function of the tumor EUD. Results: The absorbed dose response correlation was highly significant (R = 0.72, P < 0.001). With an absorbed dose threshold of 40 Gy, the metabolic response was strongly different in both groups (median response 35% versus 100%, P < 0.001). Post-RE TCP as a function of the EUD was very similar to that observed in external beam radiation therapy (EBRT), with TCP values equal to 0.5 and 0.95 for a EUD of 51 Gy and 100 Gy, respectively. The TCP was perfectly predicted by the Poisson model assuming an inter tumor radiosensitivity variation of 30% around the HCC cell in vitro value. Conclusions: EUD-based 90Y TOF-PET/CT predicts the metabolic response post-RE in HCC assessed using dual molecular PET tracers and provides a similar TCP curve to that observed in EBRT. In vivo and in vitro HCC radiosensitivities are similar. Both TCPs show that a EUD of 100 Gy is needed to control HCC for the three devices (resin spheres, glass spheres, EBRT). The Schiff-base probe H2VL [6,6'-((1E,1'E)-hydrazine-1,2 diylidenebis(methanylylidene))bis(2-methoxyphenol)] is synthesized and structurally characterized by single crystal X-ray diffraction (SCXRD). H2VL is able to detect selectively acetate ion (OAc-) colorimetrically over other anions with 1:1 co-ordination. The detection limit is found to be 4.93 µM. On the other hand, fluorescence intensity of the receptor is drastically enhanced with Zn2+ and Cd2+ in the presence of acetate as counter anion. N, N-Dimethyl formamide (DMF) or Dimethylsulphoxide (DMSO) and acetate (OAc-) was the best solvent and counter anion for Zn2+/Cd2+ -sensing compared with other solvents and anions, respectively. Detection limit for Zn2+ and Cd2+ are calculated to be 1.94 µM and 1.99 µM, respectively. The strong selective emissive behavior could be attributed to the CHEF (chelation-enhanced fluorescence) process. According to the changes in output emission intensity in DMSO in response to the set of ions (Zn2+, Cd2+ and OAc¯) as input variables, the function of 3-input multifunctional molecular logic circuits has been demonstrated. The electrochemical process of microbial electrosynthesis (MES) is used to drive the metabolism of electroactive microorganisms for the production of valuable chemicals and fuels. MES combines the advantages of electrochemistry, engineering, and microbiology and offers alternative production processes based on renewable raw materials and regenerative energies. In addition to the reactor concept and electrode design, the biocatalysts used have a significant influence on the performance of MES. Thus, pure and mixed cultures can be used as biocatalysts. By using mixed cultures, interactions between organisms, such as the direct interspecies electron transfer (DIET) or syntrophic interactions, influence the performance in terms of productivity and the product range of MES. This review focuses on the comparison of pure and mixed cultures in microbial electrosynthesis. The performance indicators, such as productivities and coulombic efficiencies (CEs), for both procedural methods are discussed. Typical products in MES are methane and acetate, therefore these processes are the focus of this review. In general, most studies used mixed cultures as biocatalyst, as more advanced performance of mixed cultures has been seen for both products. When comparing pure and mixed cultures in equivalent experimental setups a 3-fold higher methane and a nearly 2-fold higher acetate production rate can be achieved in mixed cultures. However, studies of pure culture MES for methane production have shown some improvement through reactor optimization and operational mode reaching similar performance indicators as mixed culture MES. Overall, the review gives an overview of the advantages and disadvantages of using pure or mixed cultures in MES. Short-chain fatty acids (SCFAs) are volatile fatty acids produced by gut microbial fermentation of dietary nondigestible carbohydrates. Acetate, propionate, and butyrate SCFA measures are important to clinical and nutritional studies for their established roles in promoting healthy immune and gut function. Additionally, circulating SCFAs may influence the metabolism and allied function of additional tissues and organs. The accurate quantification of SCFAs in plasma/serum is critical to understanding the biological role of SCFAs. The low concentrations of circulating SCFAs and their volatile nature present challenges for quantitative analysis. Herein, we report a sensitive method for SCFA quantification via extraction with methyl tert-butyl ether after plasma/serum acidification. The organic extract of SCFAs is injected directly with separation and detection using a polar GC column coupled to mass spectrometry. The solvent-to-sample ratio, plasma volume, and amount of HCl needed for SCFA protonation were optimized. Method validation shows good within-day and inter-day repeatability. The limit of detection was 0.3–0.6 µg/mL for acetate and 0.03–0.12 µg/mL for propionate and butyrate. Successful application of this method on clinical plasma and serum samples was demonstrated in six datasets. By simplifying the sample preparation procedure, the present method reduces the risk of contamination, lowers the cost of analysis, increases throughput, and offers the potential for automated sample preparation. This study aimed to optimize the growth conditions of the microalgae Auxenochlorella protothecoides to maximize the biomass and lipid content. The pH values and acetate concentrations of a tris-acetate-phosphate medium were varied for a photoheterotrophically grown culture of A. protothecoides. Approximately 23% of the microalgal biomass consisted of saturated fatty acids (SFAs). The highest lipid content (47.8%) was achieved with a medium containing 17 mM acetate at pH 6.2, and a medium containing 50 mM acetate at pH 5.8 resulted in the highest SFA content (43.7%). Surface response methodology was used to analyze the effects of two independent variables, pH (5.8–7.2) and acetate (0–35 mM), on lipid and biomass content. Four quadratic models, using the Box-Wilson central composite design, were proposed to correlate the optimal pH and acetate concentrations that would achieve the maximum biomass and lipid content. The study demonstrated that medium containing 24.4 mM acetate at pH 5.96 resulted in the highest lipid content (47.8%), and resulted in the highest biomass (1537 mg/L). In this study, the photoheterotrophic consortium C4 was used to produce the copolymer [P(3HB-co-3HV)]. PHA production was enhanced by using response surface methodology (RSM) to determine the effects of different concentrations of acetate and butyrate in mixtures (0.5–3 g L−1), ammonium sulfate and their combinations. This is relevant because PHA accumulation is stimulated by nitrogen limitation. The type and concentration of the substrate determines the monomeric composition and the PHA content (% per cell dry mass (CDM)). The RSM, carbon balance and metabolic behavior analysis results showed that at the lowest ammonium concentration, 0.1 g L−1, and when acetate was in a higher proportion than butyrate, biomass production was favored. In contrast, when the butyrate proportion was high, PHA production increased, reaching a highest production of 58% per CDM. The better conditions were evaluated in a 3-L reactor, and a maximum P(3HB-co-3HV) of 67% was determined. The predominant microbial population consisted of four major species, Macelibacteroides fermentans (37%), Rhodopseudomonas palustris (22%), Acinetobacter sp. (35%), and Clostridium propionicum (2%). Insights into the understanding of copolymer production by photoheterotrophic mixed cultures constitute the basis for developing coupled processes from organic residues. Purpose: Several studies have established impaired testicular function in obese male population, including the young males with childhood obesity, contributing to increased male infertility, which is a universal trend in the last few decades. Short chain fatty acids (SCFAs) have been recently demonstrated to inhibit progression to metabolic comorbidities. The present study therefore hypothesized that SCFAs, acetate attenuates testicular dysfunction in high fat diet (HFD)-induced obese rat model, possibly by modulating Nrf2/PPAR-γ. Methods: Adult male Wistar rats weighing 160–190 g were randomly allotted into three groups (n = 6/group): The groups received vehicle (distilled water), 40% HFD and sodium acetate (200 mg/kg) plus 40% HFD respectively. The administration lasted for 12 weeks. Results: HFD caused obesity, which is characterized with increased body weight and visceral adiposity and insulin resistance/hyperinsulinemia. In addition, it increased testicular lipid deposition, malondialdehyde, pro-inflammatory mediators, lactate/pyruvate ratio, γ-Glutamyl transferase, and circulating leptin as well as decreased testicular glutathione, nitric oxide, Nrf2, PPAR-γ and circulating follicle stimulating hormone and testosterone without a significant change in testicular lactate dehydrogenase, blood glucose and luteinizing hormone when compared to the control group. Nevertheless, administration of acetate reversed the HFD-induced alterations. Conclusion: The present results demonstrates that HFD causes obesity-driven testicular dysfunction, associated with testicular lipid deposition, oxidative stress, and inflammation. Microbial electrosynthesis (MES) is an effective approach to driving the CO2 reduction to multi-carbon organic products using renewable energy. In this work, the MES of acetate from CO2 was realized by mixed bacterial consortia, in which Acetobacterium sp. acted as the dominant acetate synthesis microbial flora. To improve synthesis efficiency of MES process, hydrogen evolution reaction (HER) electrocatalyst of Pt nanoparticles on reduced graphene oxide (PtNPs/rGO) was embedded on the biocathode of carbon felt. Results showed that loading the HER catalyst of PtNPs/rGO can significantly improve the MES performance. When 0.04 mg/cm2 Pt nanoparticles was loaded on the cathode, the highest acetate synthesis rate can reach 26.2 g/m2/day, which was twofold higher than that of bare carbon felt. Moreover, PtNPs/rGO incorporated carbon felt cathode showed much lower overpotential than bare carbon felt for hydrogen evolution reaction. Hence, the increased local H2 concentration around cathode enhanced the MES performance. These findings suggested that the artificial composite system composed by HER electrocatalysts will be a promising approach to enhance the electron utilization and CO2 reduction reaction, which acted as a prospective move to meet the needs of carbon cycling and sustainable energy in the future. The high temperature in which sugarcane vinasse (SV) is generated (~ 90 °C) and the positive effect of higher temperatures in biochemical reactions have motivated the evaluation of SV anaerobic digestion (AD) under extreme temperature conditions. Two-stage (acidogenic/methanogenic) and single-stage (methanogenic) AD of SV were evaluated under 70 °C in structured-bed reactors. The extreme temperature was beneficial to the acidogenic step of the two-stage AD process. The methane production, however, was hindered at 70 °C. The VMP of the single and two-stage reactors accounted, respectively, for only 13% and 7% of the production rate reported in sugarcane vinasse AD at 55 °C. At 70 °C, the main genera responsible for methane production was Methanothermobacter and the acetoclastic methanogenesis did not occur, resulting in acetic acid build up (15,800 mg L−1). These results brought a new perspective for sugarcane vinasse management, with acetic acid production alternatively to methanization. In this perspective, two-stage process would be composed of acidogenic and acetogenic reactors, and beyond acetate, hydrogen and other soluble compounds could be recovered in a complete biorefinery process. Objectives: To determine furfural biotransformation capabilities of Acinetobacter baylyi ADP1 and Acinetobacter schindleri ACE. Results: Acinetobacter baylyi ADP1 and A. schindleri ACE could not use furfural as sole carbon source but when acetate was used as substrate, ADP1 and ACE biotransformed 1 g furfural/l in 5 and 9 h, respectively. In both cases, the product of this biotransformation was difurfuryl-ether as shown by FT-IR and 1H and 13C NMR spectroscopy. The presence of furfural decreased the specific growth rate in acetate by 27% in ADP1 and 53% in ACE. For both strains, the MIC of furfural was 1.25 g/l. Nonetheless, ADP1 biotransformed 2 g furfural/l at a rate of 1 g/l/h in the stationary phase of growth. A transcriptional analysis of possible dehydrogenases involved in this biotransformation, identified that the areB and frmA genes were highly overexpressed after the exposure of ADP1 to furfural. The products of these genes are a benzyl-alcohol dehydrogenase and an alcohol dehydrogenase. Conclusions: Acinetobacter baylyi ADP1 is a candidate for the biological detoxification of furfural, a fermentation inhibitor present in lignocellulosic hydrolysates, with the possible direct involvement of the AreB and FrmA enzymes in the process. The accumulation of acetate in Escherichia coli inhibits cell growth and desired protein synthesis, and cell density and protein expression are increased by reduction of acetate excretion. Dissolved oxygen (DO) is an important parameter for acetate synthesis, and the accumulation of acetate is inversely correlated to DO level. In this study, the effect of DO levels on glutamate dehydrogenase (GDH) expression was investigated, and then different DO control strategies were tested for effects on GDH expression. DO control strategy IV (50% 0–9 h, 30% 9–18 h) provided the highest cell density (15.43 g/L) and GDH concentration (3.42 g/L), values 1.59- and 1.99-times higher than those achieved at 10% DO. The accumulation of acetate was 2.24 g/L with DO control strategy IV, a decrease of 40.74% relative to that achieved for growth at 10% DO. Additionally, under DO control strategy IV, there was lower expression of PoxB, a key enzyme for acetate synthesis, at both the transcriptional and translational level. At the same time, higher transcription and protein expression levels were observed for a glyoxylate shunt gene (aceA), an acetate uptake gene (acs), gluconeogensis and anaplerotic pathways genes (pckA, ppsA, ppc, and sfcA), and a TCA cycle gene (gltA). The flux of acetate with DO strategy IV was 8.4%, a decrease of 62.33% compared with the flux at 10% DO. In contrast to what is observed in aerobic uplands, microbial biomass and the presence of electron acceptors, such as iron oxides, play a crucial role in regulating soil organic C (SOC) mineralisation in paddy soils. However, the related underlying mechanisms are still poorly explored. We conducted an anaerobic incubation study to investigate changes in CO2 emissions from SOC and acetate (13C-labeleld) in response to iron oxide (ferrihydrite and goethite) addition in chloroform-fumigated and unfumigated paddy soils. The iron oxides, as electron acceptors, increased CO2 emissions from SOC with stronger impact under ferrihydrite than goethite addition. However, the acetate addition, as a preferable C source for reducing microbes, decreased SOC mineralisation and caused a negative priming effect. CO2 emission from both acetate and SOC was affected by microbial biomass change. In the acetate-treated soil, goethite in the fumigated soil (i.e. high microbial biomass) increased CO2 emissions from acetate, providing electron acceptors, and decreased CO2 emissions from SOC. Ferrihydrite accepted electrons and adsorbed acetate, resulting in a slight decline in CO2 emission from acetate. However, in the fumigated soil (i.e. low microbial biomass), both iron oxide additions reduced CO2 emissions from acetate and SOC and likely the dominant role of both iron oxides shifted from being electron acceptors to being adsorbents, thus limiting acetate accessibility to microorganisms. Acetaldehyde and acetic acid/acetate, the active metabolites of alcohol (ethanol, EtOH), generate actions of their own ranging from behavioral, physiological, to pathological/cancerogenic effects. EtOH and acetaldehyde have been studied to some depth, whereas the effects of acetic acid have been less well explored. In this study, we investigated the effect of acetic acid on big conductance calcium-activated potassium (BK) channels present in GH3 rat pituitary tumor cells in more detail. In whole cell voltage clamp recordings, extracellular application of acetic acid increased total outward currents in a dose-dependent manner. This effect was prevented after the application of the specific BK channel blocker paxilline. Acetic acid action was pH-dependent—in whole cell current and single BK channel recordings, open probability (Po) was significantly increased by extracellular pH reduction and decreased by neutral or base pH. Acetic acid hyperpolarized the membrane potential, whereas acidic physiological solution had a depolarizing effect. Moreover, acetic acid reduced calcium (Ca2+) oscillations and exocytosis of growth hormone contained secretory granules from GH3 cells. These effects were partially prevented by BK inhibitors—tetraethylammonium or paxillin. In conclusion, our experiments indicate that acetic acid activates BK channels in GH3 cells which eventually contribute to acetic acid-induced membrane hyperpolarization, cessation of Ca2+ oscillations, and decrease of growth hormone release. In this study, simultaneous removal of ammonium plus phenol could be achieved by Alcaligenes faecalis strain WY-01 with the addition of acetate, although acetate delayed the phenol degradation, probably due to the delayed expression of phenol hydroxylase gene under the presence of acetate. Moreover, the successful expression of key enzyme genes in strain WY-01 provided some evidence to illustrate its metabolic pathways of ammonium and phenol under aerobic conditions. Furthermore, SEM was used to clarify the role of acetate in resisting phenol toxicity, and these results demonstrated that strain WY-01 has the ability to form cell flocs when sodium acetate is used as co-substrate for a high concentration of phenol, and these flocs could protect cells against the toxicity of phenol, further enhancing phenol degradation in a high concentration of phenol. The purpose of this study was to assess the effects of acetate and β-hydroxybutyrate alone or in combination on lipogenic genes and their associated regulatory proteins in dairy cow mammary epithelial cells (DCMEC) using quantitative reverse transcription polymerase chain reaction (qRT-PCR), western blotting, lipid droplet staining and a triglyceride content detection kit, to determine whether SCFA are related to milk fat synthesis regulation in DCMEC. Our experiment shows that addition of different concentrations of acetate, β-hydroxybutyrate and their combinations to DCMEC increase in relative mRNA abundance of lipogenic genes and key transcription factors suggest an increase in lipogenic capacity, which is supported by an increased in cytosolic triglyceride content. Similarly, the protein expression level of acetyl-coenzyme A carboxylase (ACACA), fatty acid synthase (FASN) and sterol-coenzyme desaturase-1 (SCD1) genes and the transcription factor sterol regulatory element-binding protein-1 (SREBP1) were found to be increased by addition of acetate, β-hydroxybutyrate and their combinations. The expression pattern of fat-related genes and proteins showed similar trends in almost all treatments, suggesting that common transcription factor are regulating these genes. These results show that acetate and β-hydroxybutyrate regulate fat synthesis, further confirming that SCFAs work by targeting genes to activate the SREBP1 and insulin-induced gene 1 protein (INSIG1) signalling pathways in DCMEC. The non-heterocystous cyanobacterium Arthrospira platensis was investigated for the conditions under which high accumulation of polyhydroxybutyrate (PHB) occurred. Cells grown photoautotrophically under nitrogen-deprived conditions had high accumulation of PHB whose granules were clearly visualized using Nile-Red staining. A further increase of PHB was observed in cells grown photoheterotrophically for 3 days under nitrogen-deprived condition with 0.50%(w/v) acetate supplementation (−N +Act) showing the maximum PHB contents of 19.2 ± 0.5%(w/w DW). This was significantly higher than that in cells grown under the same condition but with either 0.1%(w/v) butyrate, or 0.75%(w/v) glucose, or 0.1%(w/v) propionate supplementation (P < 0.05). RT-PCR analysis revealed that photoheterotrophically grown cells under −N+ Act condition had a significant increase of phaB and phaC transcript levels (P < 0.05) compared with those supplemented with other organic carbons. The representative FTIR spectra of the extracted polymer from A. platensis showed prominent peaks at 1721 cm−1 and 1279 cm−1 for the carbonyl (C=O) stretching of the ester group and asymmetric C–O–C stretching vibration, respectively. This spectral pattern is comparable with that of the standard PHB spectra, thus confirming the chemical nature of the extract. The overall results demonstrated that the enhanced PHB accumulation in A. platensis was attributed to the increased expression of phaB and phaC responsible for increased PHB biosynthesis when cells were grown photoheterotrophically in nitrogen-deprived medium containing organic carbon compounds. Advancing the understanding of stability behavior and aggregation mechanisms of quantum dot (QD) nanoparticles in natural systems is fundamental to elucidate their fate and transport, bioavailability, environmental toxicity, and subsequent risks to environmental and public health. This study investigates the aggregation kinetics and colloidal stability of QDs as a function of pH and organic ligands—acetate, oxalate, and citrate. Results indicated an influence of solution chemistry upon both the aggregation kinetics and colloidal stability of QDs. The zeta potential of QDs, with a point of zero charge (pHPZC) between pH 1.5 and 3.5, decreased (from positive to negative) with increasing solution pH. The diameter of QD aggregates was ~500 nm in the region of pHPZC and decreased with pH when pH > pHPZC to 40–50 nm. Organic ligands enhanced the negative zeta potentials of QDs at pH = 1.5 and pH = 3.5. The impact of ligands on the levels and rates of aggregation was pH dependent; furthermore, the presence of ligands increased the diameters of all QD nanoaggregates at pH 3.5 (e.g., 817 nm for 0.001 M citrate). QDs and organic ligand-QD nanoparticle complexes remained stable across pH values 5–9. In terms of environmental and toxicological risk assessments, results revealed that QDs and organic ligand-QD nanoparticle complexes remain stable across a significant range of pH values (5–9), indicating that this stability behavior could enhance the mobility, transport, and residence time of QDs in terrestrial and aqueous environments, and facilitate the bioavailability of QDs, therefore augmenting the adverse effects of QDs in the environment. The stages of formation and development of activated sludge biofilm carrying out the anammox process in a laboratory sequencing batch reactor (SBR) with complete biomass retention on the carrier were investigated using light, epifluorescence, and electron microscopy. Light microscopy revealed biofilm formation on the carrier to occur within one week. Rod-shaped and filamentous microorganisms were the first to attach on the carrier and acted as a skeleton for biofilm formation, thus playing an important role in colonization of the carrier. Epifluorescence microscopy revealed physiologically active anammox bacteria of the genera Candidatus "Brocadia" and Ca. "Jettenia" in the biofilms. Selective autofluorescence of the colonies of anammox bacteria Ca. "Jettenia" was observed. This autofluorescence was probably caused by specific proteins in the extracellular polymeric substances (EPS), their synthesis and/or amount depending on the colony age and the physiological state of the cells. Structural organization of the colonies of anammox bacteria was investigated by electron microscopy. High-throughput sequencing of the 16S rRNA gene fragments revealed the presence of sequences affiliated with members of the phyla Chloroflexi, Bacteroidetes, Planctomycetes, and Proteobacteria in the biofilms. Apart from anammox bacteria, anaerobic and facultatively anaerobic organotrophs, stage I nitrifiers, denitrifiers, and sulfate reducers were detected. Members of the genus Ca. "Brocadia" were predominant among the anammox bacteria, probably due to better adhesion of their cells to the carrier or to their competitive advantage over Ca. Purpose: Earthworm mucus is a ubiquitous source of C and N for soil biochemical processes and likely fuels direct interactions with other soil organisms; however, its chemical composition is little known and was the research question addressed by this research. A method was developed for the collection of sufficient amounts of cutaneous mucus from highly isotopically labelled earthworms with the objective to analyse mucus chemically by state-of-the-art nuclear magnetic resonance (NMR) techniques. Materials and methods: A clean sand method was developed to collect cutaneous mucus from highly 13C, 15N-labelled endogeic earthworms (Allolobophora chlorotica). Mucus was analysed by 1D and 2D NMR. Results and discussion: NMR spectra showed a complex mixture dominated by carbohydrates with a high aliphatic/lipid content and relatively few resonances for protein/peptide material. A very large contribution from acetate was detected, likely in the form of acetylated carbohydrates. Conclusions: The ample presence of acetate is noteworthy because acetate is an important component in soil carbon cycling and methanogenic pathways. Earthworm mucus may be involved in novel direct biochemical interactions with soil (and possibly also sediment) microorganisms and plant roots that have not been recognised before. Phloroglucinol is a three-hydroxyl phenolic compound and has diverse physiological and pharmacological activities such as antivirus and anti-inflammatory activities. Chemical synthesis of phloroglucinol suffered from many drawbacks such as high cost and environmental pollution. To avoid the above issues, microbial phloroglucinol biosynthesis was successfully accomplished in this study, while the abundant and low-cost acetate was used as the main carbon source. Firstly, the toxicity of phloroglucinol was tested, and E. coli BL21(DE3) could tolerate 5 g/L phloroglucinol. The ability of phloroglucinol synthase (PhlD) for catalyzing malonyl-CoA to phloroglucinol was confirmed, and E. coli BL21(DE3) expressing PhlD and acetyl-CoA carboxylase (ACCase) could produce 1107 ± 12 mg/L phloroglucinol from glucose. Then, E. coli BL21(DE3) was engineered to utilize acetate to produce 228 ± 15 mg/L phloroglucinol. Then, the endogenous citrate synthase (GltA) which could catalyze oxaloacetate and acetyl-CoA generated from acetate to citrate was knocked down by CRISPRi system in order to enhance the carbon flux for phloroglucinol production, and the titer was improved to 284 ± 8 mg/L. This work demonstrated that acetate could be used as low-cost substrate to achieve the biosynthesis of phloroglucinol and provided an example of effective utilization of acetate. Purpose: Increased circulating concentrations of short-chain fatty acids (SCFA) achieved by ingestion of high-fibre diets is associated with anti-inflammatory effects through promotion of FoxP3+ regulatory T(reg) cells in mouse models. This study aimed to determine whether similar increments in blood SCFA levels can be achieved in humans and whether these are associated with similar immune modulatory effects. Methods: In a pilot single-blinded, randomised, controlled cross-over study in ten healthy subjects, the effects were determined of high- (39 g/day) and low-fibre (18 g/day) intake (all food provided) on SCFA (gas chromatography), proportions of Treg cells (flow cytometry) and a panel of cytokines (multiplex methodology) measured in peripheral blood at day 5 of each diet. Results: Actual fibre intake differed between the diets by 19 [16–21] g/day (P< 0.001). Median [range] total plasma SCFA levels with high-fibre intake were 174.5 [104.8–249.5] µmol/L, which were greater than those associated with low-fibre intake at 59.0 [26.5–79.9] (P < 0.001). Differences were significantly different for both acetate and propionate. The frequencies of total CD4 T cells and T-regulatory cells, and concentrations of inflammatory and anti-inflammatory cytokines were not significantly different between the dietary interventions. Conclusions: Plasma SCFA levels can be modulated by altering dietary fibre consumption in healthy individuals with increments similar to those achieved in murine studies. Five days of diet intervention did not result in changes in regulatory T-cell proportions and cytokine concentrations in peripheral blood, and may require longer duration of dietary change. Chlamydomonas reinhardtii P.A. Dangeard is a unicellular green alga capable to assimilate acetate. C. reinhardtii growth and metabolism distinctly depend on trophic conditions. Its influence on batch culture interferes with changes in the medium composition and metabolism of microalgae occurring during culture growth. The aim of this work was to estimate the effect of acetate on changes in the expression of 32 genes encoding enzymes of central metabolism and plastid transporters during growth of batch cultures of C. reinhardtii cc-124. In autotrophic conditions, transcription profiles considerably differed from the profiles in the presence of acetate. The strongest influence of trophic conditions was observed in the log phase of growth. In the presence of acetate, a more intense expression of gene ACS2 encoding plastid acetyl-CoA synthase and of the genes encoding subunits of enzymes directing acetyl groups to the synthesis of fatty acids was recorded. Elevated expression of genes PCK1 and PPT1 under mixotrophic conditions may be corresponded to the entry of acetate carbon to gluconeogenesis. In the presence of acetate, a high expression level of the starch metabolism genes was observed. Autotrophic conditions were notable for an elevated accumulation of transcripts of the genes encoding subunits of citrate lyase, which may be responsible for an outflow of acetyl groups from the Krebs cycle. Moreover, a higher level of expression was shown for genes of plastid transporters participating in the export of sugars from plastids, which is associated with the redistribution of reducing power in the cell. Methane production usually increases from the acidic sphagnum-dominated ombrotrophic peatlands to minerotrophic ones with more neutral pH and higher coverage of vascular plants. Along this ombrotrophic–minerotrophic gradient, pH, microbial communities, and properties of dissolved organic matter in porewater all vary greatly. The hydrographic change resulted from permafrost thaw and projected global warming can potentially connect the minerotrophic and ombrotrophic sites via porewater and turn acidic bogs to minerotrophic fens. It is thus very important to investigate how the anaerobic carbon degradation processes respond to changes in fundamental factors like pH, temperature, properties of dissolved organic matter, and microbial communities resulted from such hydrographic change. In this study, one ombrotrophic (pH = 3.9) and one minerotrophic peatland site were sampled in Fairbanks, Alaska in Sep 2017 and a 42-day-period anaerobic laboratory incubation was conducted to study the changes in anaerobic carbon degradation processes including primary and secondary fermentation, methanogenesis, and acetogenesis when pH, temperature, and porewater were manipulated individually and a combination of two or three of these factors. The results suggested lowering pH would inhibit many anaerobic carbon degradation processes in the minerotrophic peatland except primary fermentation. Elevating pH in the ombrotrophic site did not stimulate its methanogen community, but primary fermentation responded better with increasing pH than with increasing temperature alone. Replacing the porewater in the minerotrophic site with that from the ombrotrophic site with high aromaticity did not inhibit methanogenesis but potentially brought in highly efficient primary fermenters. Acetoclastic methanogenesis, acetogenesis, and syntrophy only exist in the minerotrophic site but not at the ombrotrophic one. Porewater from the minerotrophic site could potentially introduce acetoclastic methanogens and syntrophs to the ombrotrophic site but would not make them active unless both pH and temperature were increased. When ground water connects ombrotrophic and minerotrophic peatlands due to thawing of permafrost, secondary fermenters and acetoclastic methanogens could be introduced to acidic bogs and cooperate efficiently to degrade the stored carbon in ombrotrophic peatlands especially under elevated temperature conditions.
