In May and June of 2021, marine microbial samples were collected for DNA sequencing in East Sound, WA, USA every 4 hours for 22 days. This high temporal resolution sampling effort captured the last 3 days of a Rhizosolenia sp. bloom, the initiation and complete bloom cycle of Chaetoceros socialis (8 days), and the following bacterial bloom (2 days). Metagenomes were completed on the time series, and the dataset includes 128 size-fractionated microbial samples (0.22–1.2 µm), providing gene abundances for the dominant members of bacteria, archaea, and viruses. This dataset also has time-matched nutrient analyses, flow cytometry data, and physical parameters of the environment at a single point of sampling within a coastal ecosystem that experiences regular bloom events, facilitating a range of modeling efforts that can be leveraged to understand microbial community structure and their influences on the growth, maintenance, and senescence of phytoplankton blooms. The North Temperate Lakes Long-Term Ecological Research (NTL-LTER) program has been extensively used to improve understanding of how aquatic ecosystems respond to environmental stressors, climate fluctuations, and human activities. Here, we report on the metagenomes of samples collected between 2000 and 2019 from Lake Mendota, a freshwater eutrophic lake within the NTL-LTER site. We utilized the distributed metagenome assembler MetaHipMer to coassemble over 10 terabases (Tbp) of data from 471 individual Illumina-sequenced metagenomes. A total of 95,523,664 contigs were assembled and binned to generate 1,894 non-redundant metagenome-assembled genomes (MAGs) with ≥50% completeness and ≤10% contamination. Phylogenomic analysis revealed that the MAGs were nearly exclusively bacterial, dominated by Pseudomonadota (Proteobacteria, N = 623) and Bacteroidota (N = 321). Nine eukaryotic MAGs were identified by eukCC with six assigned to the phylum Chlorophyta. Additionally, 6,350 high-quality viral sequences were identified by geNomad with the majority classified in the phylum Uroviricota. This expansive coassembled metagenomic dataset provides an unprecedented foundation to advance understanding of microbial communities in freshwater ecosystems and explore temporal ecosystem dynamics. Purpose: Trial data shows modest reductions in leg pain, disability and surgery avoidance following epidural steroid injections (ESI) for severe sciatica. Despite their common use, there is no clear evidence about which patients are more likely to benefit from ESI. The aim of this study was to generate consensus on potential predictors of outcome following ESI for disc-related sciatica. Methods: A list of potential predictors of outcome was generated during a consensus meeting of seven experts. The items were subsequently presented in a two round on-line Delphi study to generate consensus among experts on which items are potential predictors of outcome. Consensus was defined as 70% agreement among participants. Results: Sixty-one items were generated during the consensus meeting. Of ninety experts invited to participate in the on-line Delphi study, 44 (48%) and 33 (73%) took part in rounds one and two respectively. Twenty-eight additional items suggested by participants in round one were included in round two. Overall, 14 items reached consensus reflecting domains of health, medication use, pain intensity, psychosocial factors, imaging findings and type of injection. Context: Freshwater biodiversity is declining at unparalleled rates, but fundamental questions remain over how it is distributed at the spatial scales most relevant for conservation management. Objectives: Here, we test the hypothesis that freshwater biodiversity is distributed across standing waterbody types in a pattern that is reproducible across disparate biota and contrasting landscapes, such that conservation efforts can be aligned across landscapes and taxa. Methods: We analysed the richness, composition and distribution of macrophytes, molluscs, beetles and odonates from 199 standing waterbodies (lakes, ponds, ditches and canals) nested within UK landscapes with contrasting dominant land use (agricultural, upland and suburban). Results: We found a common pattern in the distribution of our biodiversity indicators across waterbody types in all landscapes that was largely repeated across biota; lakes consistently had the highest or equal alpha diversity and supported a greater proportion of the sampled species pool in each landscape (mean = 86%) in comparison to ponds (74%). Landscape-specific waterbody types (ditches and canals) also contributed significantly to the regional species pool (69 and 33% respectively). Each waterbody type contributed uniquely to landscape biodiversity and usually species of conservation concern, rather than simply supporting a subset of ubiquitous species found in lakes. Conclusions: Landscape-wide management strategies that encompass multiple habitats and biota should prove advantageous and generalisable. However, our study landscapes suggest that long-term biodiversity conservation should also recognise lakes as a priority for nature recovery, both to minimise further losses and to maintain the largest reservoir of biodiversity. Female Weddell seals (Leptonychotes weddellii) display a mixed capital-income breeding strategy, losing up to 40% of their body mass between birthing and weaning their pups. How and when they regain energy stores, however, remains to be fully explored. To better understand the foraging by lactating Weddell seals, we fitted time-depth recorders and head-mounted cameras on 26 seals in Erebus Bay, Ross Sea, for ~ 5 days in November and December 2018 and 2019. We aimed to (1) identify prey species and foraging depth and (2) investigate relationships between seal physiology and demographics and probability of foraging. We recorded 2782 dives, 903 of which were > 50 m, maximum depth was 449.3 m and maximum duration was 31.1 min. Pup age likely contributes to the probability of a lactating Weddell seal foraging (Est. = 1.21 (SD = 0.61), z = 1.97, p = 0.0484). Among 846 prey encounters, the most frequent prey items were crustaceans (46.2%) and Antarctic silverfish (Pleuragramma antarcticum, 19.0%); two encounters were observed with juvenile Antarctic toothfish (Dissostichus mawsoni, 0.2%). We identified substantial variability in foraging behaviour, individually and between locations, and found that lactating seals target many species and some may specialise on certain prey groups. Wood is a sustainable, benign, and high-performing green structural material readily available in nature that can be used to replace structural materials. However, insufficient mechanical performance (compared to metals and plastic), moisture sensitivity, and susceptibility to microorganism attack make it challenging to use wood as it is for advanced engineering applications. We here present an efficient approach to fabricating densified wood with minimal time and waste generation, demonstrating high mechanical strength, and decreased water penetration on the surface. Wood slabs were treated with deep eutectic solvents (DESs) to solubilize the lignin, followed by in-situ regeneration of dissolved lignin in the wood. Then, the slabs were densified with heat and pressure, turning the wood into a functionalized densified material. Lignin regeneration and morphological changes were observed via two-photon microscopy and Scanning Electron Microscopy (SEM), respectively. The final product is less susceptible to water absorption on the surface and has enhanced flexural strength (> 50% higher), surface hardness (100% increased), and minimal set recovery compared to natural wood. The improved mechanical performance is due to regenerated lignin which acts as a glue and fills spaces present within the interconnected cellulose network inside the wood, forming a highly dense composite during densification. Structural diversity (SD) characterizes the volume and physical arrangement of biotic components in an ecosystem which control critical ecosystem functions and processes. LiDAR data provides detailed 3-D spatial position information of components and has been widely used to calculate SD. However, the intensive computation of SD metrics from extensive LiDAR datasets is time-consuming and challenging for researchers who lack access to high-performance computing resources. Moreover, a lack of understanding of LiDAR data and algorithms could lead to inconsistent SD metrics. Here, we developed a SD product using the Discrete-Return LiDAR Point Cloud from the NEON Aerial Observation Platform. This product provides SD metrics detailing height, density, openness, and complexity at a spatial resolution of 30 m, aligned to the Landsat grids, for 211 site-years for 45 Terrestrial NEON sites from 2013 to 2022. To accommodate various ecosystems with different understory heights, it includes three different cut-off heights (0.5 m, 2 m, and 5 m). Most climate proxies of sea surface temperatures suffer from severe limitations when applied to cold temperatures that characterize Arctic environments. These limitations prevent us from constraining uncertainties for some of the most sensitive climate tipping points that can trigger rapid and dramatic global climate change such as Arctic/Polar Amplification, the disruption of the Atlantic Meridional Overturning Circulation, sea ice loss, and permafrost melting. Here, we present an approach to reconstructing sea surface temperatures globally using paired Mg/Ca - δ18Oc recorded in tests of the polar to subpolar planktonic foraminifera Neogloboquadrina pachyderma. We show that the fidelity of Mg/Ca-based paleoclimate reconstructions is compromised by variations in seawater carbonate chemistry which can be successfully quantified and isolated from paleotemperature reconstructions using a multiproxy approach. By applying the calibration to the last glacial maximum, we show that marine polar amplification has been underestimated by up to 3.0 ± 1.0 °C in model-based estimates. Standard climate proxies cannot quantify sea surface temperatures below 4 °C. Calcium phosphate-based materials (CaP) are introduced as potential dental pulp capping materials for deciduous teeth. The present study investigated the influence of inorganic phosphate (Pi) on regulating stem cells isolated from human exfoliated deciduous teeth (SHED). SHEDs were treated with Pi. Cell cycle progression and apoptosis were examined using flow cytometry analysis. Osteo/odontogenic and adipogenic differentiation were analyzed using alizarin red S and oil red O staining, respectively. The mRNA expression profile was investigated using a high-throughput RNA sequencing technique. Pi increased the late apoptotic cell population while cell cycle progression was not altered. Pi upregulated osteo/odontoblastic gene expression and enhanced calcium deposition. Pi-induced mineralization was reversed by pretreatment of cells with Foscarnet, or p38 inhibitor. Pi treatment inhibited adipogenic differentiation as determined by decreased PPARγ expression and reduced intracellular lipid accumulation. Bioinformatic analysis of gene expression profiles demonstrated several involved pathways, including PI3K/AKT, MAPK, EGFR, and VEGF signaling. Background: Invasive lobular carcinoma (ILC) of the breast grows in a diffuse pattern, resulting in a high risk of positive margins at surgical resection. Oncoplastic approaches have been shown to reduce this risk, but concerns persist around the safety of immediate oncoplastic surgery for those with ILC. This study evaluated the short- and long-term oncologic outcomes of immediate oncoplastic surgery for patients with ILC. Methods: This study retrospectively analyzed an institutional database of stages I to III ILC patients who underwent breast-conserving surgery (BCS) with or without immediate oncoplastic surgery (oncoplastic closure or oncoplastic reduction mammoplasty [ORM]). The study compared positive margin rates, rates of successful BCS, and recurrence-free survival (RFS) by type of surgery. Results: For 494 patients the findings showed that the use of immediate ORM was associated with significantly lower odds of positive margins (odds ratio [OR], 0.34; 95 % confidence interval [CI], 0.17–0.66; p = 0.002). Both lumpectomy with oncoplastic closure and ORM were significantly associated with higher rates of successful BCS than standard lumpectomy (94.2 %, 87.8 %, and 73.9 %, respectively; p < 0.001). No difference in RFS was observed between those undergoing immediate oncoplastic surgery and those undergoing standard lumpectomy alone. Conclusions: The patients with stages I to III ILC who underwent immediate oncoplastic surgery had significant benefits including lower odds of positive margins and higher rates of successful BCS, with both types of immediate oncoplastic surgery showing similar RFS compared with lumpectomy alone. This supports the oncologic safety of immediate oncoplastic surgery for diffusely growing tumors such as ILC, providing it an ideal option for patients desiring BCS. [ABSTRACT FROM AUTHOR]

Intentionally walking to the beat of an auditory stimulus seems effortless for most humans. However, studies have revealed significant individual differences in the spontaneous tendency to synchronize. Some individuals tend to adapt their walking pace to the beat, while others show little or no adjustment. To fill this gap we introduce the Ramp protocol, which measures spontaneous adaptation to a change in an auditory rhythmic stimulus in a gait task. First, participants walk at their preferred cadence without stimulation. After several steps, a metronome is presented, timed to match the participant's heel-strike. Then, the metronome tempo progressively departs from the participant's cadence by either accelerating or decelerating. The implementation of the Ramp protocol required real-time detection of heel-strike and auditory stimuli aligned with participants' preferred cadence. To achieve this, we developed the TeensyStep device, which we validated compared to a gold standard for step detection. We also demonstrated the sensitivity of the Ramp protocol to individual differences in the spontaneous response to a tempo-changing rhythmic stimulus by introducing a new measure: the Response Score. This new method and quantification of spontaneous response to rhythmic stimuli holds promise for highlighting and distinguishing different profiles of adaptation in a gait task. The South African blueberry industry has grown consistently over the past few years due to an increase in local and international demand. Like many other crops, blueberries are susceptible to grey mould caused by Botrytis cinerea, resulting in significant reductions in yield. In this study, Botrytis isolates were recovered from symptomatic blueberry fruit, flowers, leaves and undifferentiated buds, but were not found as endophytes in healthy tissues. Phylogenetic analysis of the concatenated glyceraldehyde-3-phosphate dehydrogenase (G3PDH), heat-shock protein 60 (HSP60) and DNA-dependent RNA polymerase subunit II (RPB2) genes identified 88 isolates as B. cinerea strains as well as a single B. pelargonii strain. Many early childhood teachers seek to promote diversity in their classrooms through the use of multicultural children's literature. While these efforts are well-intentioned, teachers may not be fully aware of the issues of culture potentially hidden within such books, nor may they have support in considering the authenticity of the texts they use. While these issues are pervasive within books representing all cultural groups, recent research by Indigenous scholars has highlighted the concerns and implications within Indigenous children's literature. This article is grounded within Indigenous ways of knowing to provide a helpful tool for supporting teachers as they seek to curate authentic Indigenous children's literature for classrooms. Mosquitoes visit flowers to obtain sugar or other nutrients and therefore possibly serve as major or minor pollinators of some plant species. They also often derive plant nutrients from other sources, such as extrafloral nectaries and honeydew. In a few cases, the plant-mosquito relationship is close, and mosquito pollination has been confirmed. Most plant species visited by mosquitoes, however, appear to depend on multiple means of pollination, particularly other flower-feeding insects. In addition, most mosquito species visit the flowers of many kinds of plants, possibly dispersing pollen in both biologically meaningful and irrelevant ways. This apparent lack of selectivity by both plants and mosquitoes liberates each of them from dependence on an unreliable pollen vehicle or nutrient source. A hypothetical pollinating role for the two top vectors of devastating human-disease pathogens, Anopheles gambiae or Aedes aegypti, relies on indirect evidence. So far, this evidence suggests that their participation in pollen transfer of native, introduced, or beneficial plants is negligible. The few plant species likely to be pollinated by these vectors are mostly invasive, harmful weeds associated with humans. That conclusion draws support from four characteristics of these vectors: (1) the numerous alternative potential pollinators of the flowers they visit; (2) their common use of diverse non-floral sources of nutrients; (3) the females' infrequent sugar feeding and heavy reliance on human blood for energy; and (4) their relatively low population densities. From these traits it follows that focused suppression or elimination of these two vectors, by whatever means, is highly unlikely to have adverse effects on pollination in endemic biotic communities or on ornamental plants or food crops. Purpose of Review: The development and progression of heart failure is characterized by metabolic and physiologic adaptations allowing patients to cope with cardiac insufficiency. This review explores the changes in metabolism in heart failure and the potential role of biomarkers, particularly ketone bodies, in staging and prognosticating heart failure progression. Recent Findings: Recent insights into myocardial metabolism shed light on the heart's response to stress, highlighting the shift towards reliance on ketone bodies as an alternative fuel source. Elevated blood ketone levels have been shown to correlate with the severity of cardiac dysfunction, emphasizing their potential as prognostic indicators. Furthermore, studies exploring therapeutic interventions targeting specific metabolic pathways offer promise for improving outcomes in heart failure. Summary: Ketones have prognostic utility in heart failure, and potentially, an avenue for therapeutic intervention. Challenges remain in deciphering the optimal balance between metabolic support and exacerbating cardiac remodeling. Recycling end-of-life tires (EOLT) presents a sustainable solution for addressing a major waste issue in many countries. This study focuses on the reuse of EOLT as construction materials, particularly in the form of EOLT-based rubberised concrete. However, several research gaps hinder the understanding of this construction material for implementation into practice. This study delves into key parameters, including admixture composition, aging, thickness, rubber distribution, and surface roughness, which play pivotal roles in designing and implementing rubberised concrete noise barriers. The paper presents the results of investigations into the performance of fibre-reinforced rubberised concrete when entrained with air, shedding light on flexural toughness and post-crack behaviour. The effects of incorporating fly ash and ground granulated blast furnace slag (GGBFS) as cement replacements are also examined. The acoustic performance of fibre-reinforced rubber concrete is studied, including the impact of sample conditioning (surface saturated dry or dry). The results indicate that air-entraining admixtures, the replacement of coarse sand with tire-derived rubber shreds, and the inclusion of recycled polypropylene fibre significantly enhance the mechanical and acoustic properties of the concrete. For instance, compressive strength improves by 43%, flexural strength by 120% and acoustic performance nearly twice, while water absorption and volume of permeable voids remain relatively unaffected. This study suggests an optimized sustainable mix design with rubber replacing more than 75% of the aggregate volume. It underscores the potential of EOLT-based rubberised concrete as an environmentally responsible construction material, offering enhanced performance across multiple domains, including noise attenuation barriers. Research on Mesophotic Coral Ecosystems (MCEs; 30–150 m) has grown exponentially in the last few decades, highlighting their rich diversity and extensive distribution. However, they are still largely underexplored compared to shallow-water coral reefs and frequently remain under-protected and under-represented in marine spatial planning. One reason for the imbalance between the high ecological value of MCEs and the limited levels of protection may be that baseline data on MCEs are largely missing to date, yet are crucial to provide evidence-based information for management actions. Here, we present data on the alpha and beta diversity of the benthic communities within MCEs in the Chagos Archipelago, Indian Ocean. Using imagery collected from Remotely Operated Vehicle surveys, benthic invertebrate megafauna were surveyed along the entire depth gradient from shallow to lower mesophotic depths (15–160 m). The diversity of the benthic communities decreased with increasing depth, from shallow water to the lower mesophotic zone. Nevertheless, the deepest parts of MCEs in the Archipelago displayed higher species richness values than several other shallow subtropical regions. In addition, the benthic communities showed high dissimilarity along the depth gradient, indicating that the key driver of community composition change with depth is species turnover (species replacement), revealing the uniqueness of MCEs. This study presents novel findings on MCEs in the central Indian Ocean, demonstrating that they host a high and unique benthic diversity, and highlighting the need to protect these ecosystems to preserve the overall biodiversity of coral reefs. In this paper, we study the asymmetric effects of different types of capital-embodied technological change, as proxied by tangible and intangible assets, on relative wages (high- to medium-skilled, high- to low-skilled and medium- to low-skilled workers), relying upon the technology-skill complementarity and polarization of the labor force frameworks. We also consider two additional major channels that contribute to shaping wage differentials: globalization (in terms of trade openness and global value chains participation) and labor market institutions. The empirical analysis is carried out using a panel dataset comprising 17 mostly advanced European economies and 5 industries, with annual observations spanning the period 2008–2017. Our findings suggest that software and databases—as a proxy for intangible technologies—exert downward pressure on low-skilled wages, while robotics is associated with a polarization of the wage distribution at the expense of middle-skilled labor. Additionally, less-skilled workers' relative wages are negatively affected by trade openness and global value chain participation, but positively influenced by sector-specific labor market regulations. Impairments of object recognition are core features of neurodegenerative syndromes, in particular posterior cortical atrophy (PCA; the 'visual-variant Alzheimer's disease'). These impairments arise from damage to higher-level cortical visual regions and are often missed or misattributed to common ophthalmological conditions. Consequently, diagnosis can be delayed for years with considerable implications for patients. We report a new test for the rapid measurement of cortical visual loss – the Graded Incomplete Letters Test (GILT). The GILT is an optimised psychophysical variation of a test used to diagnose cortical visual impairment, which measures thresholds for recognising letters under levels of increasing visual degradation (decreasing "completeness") in a similar fashion to ophthalmic tests. The GILT was administered to UK Biobank participants (total n=2,359) and participants with neurodegenerative conditions characterised by initial cortical visual (PCA, n=18) or memory loss (typical Alzheimer's disease, n=9). UK Biobank participants, including both typical adults and those with ophthalmological conditions, were able to recognise letters under low levels of completeness. In contrast, participants with PCA consistently made errors with only modest decreases in completeness. GILT sensitivity to PCA was 83.3% for participants reaching the 80% accuracy cut-off, increasing to 88.9% using alternative cut-offs (60% or 100% accuracy). Specificity values were consistently over 94% when compared to UK Biobank participants without or with documented visual conditions, regardless of accuracy cut-off. These first-release UK Biobank and clinical verification data suggest the GILT has utility in both rapidly detecting visual perceptual losses following posterior cortical damage and differentiating perceptual losses from common eye-related conditions. Pyroprocessing holds the key to unlocking a more sustainable future for nuclear energy by handling various types of spent nuclear fuel (SNFs) and reducing radioactive waste volumes. This review examines the role of electrochemical monitoring in molten salt pyroprocessing of SNFs, emphasizing its importance in enhancing process efficiency and nonproliferation. Challenges associated with the monitoring of multi-element environments, flow environments, and sensor stability are discussed. We prove L p (R d) × L q (R d) → L r (R d) bounds for certain lacunary bilinear maximal averaging operators with parameters satisfying the Hölder relation 1 / p + 1 / q = 1 / r . The boundedness region that we get contains at least the interior of the Hölder boundedness region of the associated single scale bilinear averaging operator. In the case of the lacunary bilinear spherical maximal function in d ≥ 2 , we prove boundedness for any p , q ∈ (1 , ∞ ] 2 , which is sharp up to boundary; we then show how to extend this result to a more degenerate family of surfaces where some curvatures are allowed to vanish. For the lacunary triangle averaging maximal operator, we have results in d ≥ 7 , and the description of the sharp region will depend on a sharp description of the Hölder bounds for the single scale triangle averaging operator, which is still open. Rural regions have higher incidence rates of colorectal cancer (CRC) than urban regions, while screening is lower in rural residents than in urban residents. Rural residents experience barriers to CRC screening due to a lack of access to healthcare, perceived lack of privacy, and other cultural factors. The fecal immunochemical tests (FITs) may provide an inexpensive and convenient alternative for those lacking access to a colonoscopy or sigmoidoscopy. In a clinical trial, we investigated factors that influenced the return of a FIT kit for CRC screening among 1,230 rural residents in a midwestern U.S. state. Participants were selected from two cancer screening databases maintained by the state health department. Participants returning and not returning the FIT kit were compared for differences by age category (45–54, 55–64, 65–74), gender (female vs. male), race (non-white vs. white), and ethnicity (Hispanic vs. not Hispanic) using chi-square tests and logistic regression. Cox proportional hazard models and Kaplan–Meier curves were used to assess differences in FIT kit return time by age and gender. The youngest age group was significantly less likely to return the FIT kit and were slower at returning it when they did. In models adjusted for age category, females were significantly more likely to return the FIT kit than males and returned the FIT kit sooner than males. The results suggest that efforts are needed to reach those 45 to 55, especially males, who are not likely to see the need for CRC screening. [ABSTRACT FROM AUTHOR]

While the nuclear contribution to stopping power, i.e., elastic scattering between atoms, is well understood in the literature, the route for gathering data on the electronic contribution has for decades remained costly and reliant on many simplifying assumptions, including that materials are isotropic. We establish a method that combines time-dependent density functional theory (TDDFT) and machine learning to reduce the time to assess new materials to hours on a supercomputer and provide valuable data on how atomic details influence electronic stopping. Our approach uses TDDFT to compute the electronic stopping from first principles in several directions and then machine learning to interpolate to other directions at a cost of 10 million times fewer core-hours. We demonstrate the combined approach in a study of proton irradiation in aluminum and employ it to predict how the depth of maximum energy deposition, the "Bragg Peak," varies depending on the incident angle—a quantity otherwise inaccessible to modelers and far outside the scales of quantum mechanical simulations. The lack of any experimental information requirement makes our method applicable to most materials, and its speed makes it a prime candidate for enabling quantum-to-continuum models of radiation damage. The use of antimicrobial drugs in food-producing animals contributes to the selection pressure on pathogenic and commensal bacteria to become resistant. This study aims to evaluate the existence of trade-offs between treatment effectiveness, cost, and the dynamics of resistance in gut commensal bacteria. We developed a within-host ordinary differential equation model to track the dynamics of antimicrobial drug concentrations and bacterial populations in the site of infection (lung) and the gut. The model was parameterized to represent enrofloxacin treatment for bovine respiratory disease (BRD) caused by Pastereulla multocida in cattle. Three approved enrofloxacin dosing regimens were compared for their effects on resistance on P. multocida and commensal E. coli: 12.5 mg/kg and 7.5 mg/kg as a single dose, and 5 mg/kg as three doses. Additionally, we explored non-FDA-approved regimes. Our results indicated that both 12.5 mg/kg and 7.5 mg/kg as a single dose scenario increased the most the treatment costs and prevalence of P. multocida resistance in the lungs, while 5 mg/kg as three doses increased resistance in commensal E. coli bacteria in the gut the most out of the approved scenarios. A proposed non-FDA-approved scenario (7.5 mg/kg, two doses 24 h apart) showed low economic costs, minimal P. multocida, and moderate effects on resistant E. coli. Overall, the scenarios that decrease P. multocida, including resistant P. multocida did not coincide with those that decrease resistant E. coli the most, suggesting a trade-off between both outcomes. The sensitivity analysis suggests that bacterial populations were the most sensitive to drug conversion factors into plasma (β ), elimination of the drug from the colon (ϑ ), fifty percent sensitive bacteria (P. multocida) killing effect ( L s50 ), fifty percent of bacteria (E. coli) above ECOFF killing effect ( C r50 ), and net drug transfer rate in the lung (γ ) parameters. Identifying mechanisms that drive population divergence under varying geographic and ecological scenarios can inform our understanding of evolution and speciation. In particular, analysis of genetic data from island populations with known colonisation timelines allows us to identify potential source populations of diverging island subspecies and current relationships among populations. Silvereyes (Zosterops lateralis) are a small passerine that have served as a valuable study system to investigate evolutionary patterns on both large and small geographic scales. We examined genetic relatedness and diversity of two silvereye subspecies, the mainland Z. l. cornwalli and island Z. l. chlorocephalus, and used 18 077 single nucleotide polymorphisms (SNPs), to compare locations across southeast Queensland, Australia. Although silvereyes are prolific island colonisers our findings revealed population divergence over relatively small spatial scales was strongly influenced by geographic isolation mediated by water barriers. Strong genetic connectivity was displayed between mainland sites, but minimal inter-island connectivity was shown despite comparable sampling distances. Genetic diversity analysis showed little difference in heterozygosity between island and mainland populations, but lower inbreeding scores among the island populations. Our study confirmed the range of the Z. l. chlorocephalus subspecies throughout the southern Great Barrier Reef. Our results show that water barriers and not geographic distance per se are important in driving incipient divergence in island populations. This helps to explain the relatively high number of phenotypically differentiated, but often geographically proximate, island silvereye subspecies compared to a lower number of phenotypically less well-defined Australian continental subspecies. Histiocytoid cardiomyopathy (HC) is an arrhythmogenic disorder, usually involving children under two years of age with a strong Caucasian and female predominance. The disease is fatal in the vast majority and diagnosis is nearly always established at autopsy, but this is only possible with adequate myocardial sampling. Meticulous gross and histological examination of the heart in collaboration with a cardiovascular-trained pathologist maximises the opportunity to make specific diagnoses (and therefore rule out the differentials of SIDS, SUDC and child abuse), guide genetic testing, and inform potentially life-saving medical interventions for blood relations. We present a typical HC case presenting as sudden death, without prodrome, in a previously healthy 18-month-old boy. The disease is characterised histologically by discrete groups of enlarged, polygonal histiocyte-like cells with distinct margins and abundant faintly eosinophilic foamy cytoplasm. Cells often contain coarse granules, microvacuoles and irregular, round nuclei. In our case, dysplastic fascicles were predominantly located immediately deep to the endocardium of the left ventricle. We report our own autopsy findings with histological images, and discuss the expected clinical, morphological and ultrastructural features of the disease. [ABSTRACT FROM AUTHOR]

Recent approaches have investigated assisting users in making early trade-off decisions when the future evolution of project elements is uncertain. These approaches have demonstrated promise in their analytical capabilities; yet, stakeholders have expressed concerns about the readability of the models and resulting analysis, which builds upon Tropos. Tropos is based on formal semantics enabling automated analysis; however, this creates a problem of interpreting evidence pairs. The aim of our broader research project is to improve the process of model comprehension and decision-making by improving how analysts interpret and make decisions. We extend and evaluate a prior approach, called EVO, which uses color to visualize evidence pairs. In this article, we explore the effectiveness of EVO with and without the impacts of tooling through a two-phased empirical study. All subjects in both phases were untrained modelers, given training at study time. First, we conduct an experiment to measure any effect of using colors to represent evidence pairs. Second, we explore how subjects engage in decision-making activities (with or without color) through a user study. Adoptive cellular therapy (ACT) using memory-like (ML) natural killer (NK) cells, generated through overnight ex vivo activation with IL-12, IL-15, and IL-18, has shown promise for treating hematologic malignancies. We recently reported that a multifunctional fusion molecule, HCW9201, comprising IL-12, IL-15, and IL-18 domains could replace individual cytokines for priming human ML NK cell programming ("Prime" step). However, this approach does not include ex vivo expansion, thereby limiting the ability to test different doses and schedules. Here, we report the design and generation of a multifunctional fusion molecule, HCW9206, consisting of human IL-7, IL-15, and IL-21 cytokines. We observed > 300-fold expansion for HCW9201-primed human NK cells cultured for 14 days with HCW9206 and HCW9101, an IgG1 antibody, recognizing the scaffold domain of HCW9206 ("Expand" step). This expansion was dependent on both HCW9206 cytokines and interactions of the IgG1 mAb with CD16 receptors on NK cells. The resulting "Prime and Expand" ML NK cells exhibited elevated metabolic capacity, stable epigenetic IFNG promoter demethylation, enhanced antitumor activity in vitro and in vivo, and superior persistence in NSG mice. Thus, the "Prime and Expand" strategy represents a simple feeder cell-free approach to streamline manufacturing of clinical-grade ML NK cells to support multidose and off-the-shelf ACT. Biosurfactants (BSFs) are molecules produced by microorganisms from various carbon sources, with applications in bioremediation and petroleum recovery. However, the production cost limits large-scale applications. This study optimized BSFs production by Bacillus velezensis (strain MO13) using residual glycerin as a substrate. The spherical quadratic central composite design (CCD) model was used to standardize carbon source concentration (30 g/L), temperature (34 °C), pH (7.2), stirring (239 rpm), and aeration (0.775 vvm) in a 5‐L bioreactor. Maximum BSFs production reached 1527.6 mg/L of surfactins and 176.88 mg/L of iturins, a threefold increase through optimization. Microbial development, substrate consumption, concentration of BSFs, and surface tension were also evaluated on the bioprocess dynamics. Mass spectrometry Q-TOF-MS identified five surfactin and two iturin isoforms produced by B. velezensis MO13. Active and passive continental margins exhibit significant differences in sediment transport. However, opportunities to quantify these specific sedimentary processes remain limited. The northeastern South China Sea hosts both active and passive continental margins, offering a distinct opportunity to compare the relative contributions from a large passive continental margin with those from a smaller but more dynamic active margin. We present detrital zircon U-Pb ages from marine sediments in the northeastern South China Sea to decipher their provenance and reveal the control of transport processes in the margins. Our results show varied provenance of sediments from the shelf to the trench. In the continental shelf, sediments were primarily derived from the Jiulong and Gaoping rivers, controlled by the topography and ocean currents in the passive margin. In contrast, the sediments in the submarine canyons predominantly originated from the Gaoping and Zhuoshui rivers in western Taiwan Island. Quantitative results indicate that the Gaoping submarine canyon provided approximately 86.2% of sediments to the Manila Trench by directly linking to the Gaoping River in the active margin. Based on the provenance results, we propose a distinct sedimentary process in the active and passive margins. In passive margins, sediments from drainage systems enter the shelf and are transported by ocean currents, but topographic highs often block these sediments, preventing them from reaching the deep sea. In contrast, active margins feature narrow shelves, which facilitate direct connections between submarine canyons and river mouths, allowing gravity flows to carry sediments directly to the trenches. Our study provides an understanding of the sediments transported from rivers to trenches in the northeastern South China Sea and highlights the controls (ocean currents, dramatic topography, and gravity flows) during the sediment transport in the continental margins. Invasive species are an increasing source of economic loss, costing nations billions of dollars annually. Significant financial resources are spent to manage invasive species, but few comprehensive syntheses of the economic expenditures associated with this management effort exist. As a relatively affluent developed country, the United States should serve as a model of how to both manage invasive species and, more critically, understand the economic costs of doing so. To begin understanding the scale of expenditures on invasive species in the U.S., our goal was to quantify spending on invasive species management at the state level. We contacted natural resource management officials from all 50 states following a standardized protocol. While 47 of the 50 states provided expenditures for at least one of the five years requested (2017–2021), the distribution of expenditures by state varied dramatically, suggesting that actual expenditures might be much higher than those reported. While most states shared annual expenditures, they varied by an order of magnitude from $28,370 for Connecticut to $118,695,389 for Washington. Specifically, a widespread lack of careful and consistent expense tracking and coordination within and between states made clear and correct evaluation difficult. While the expenditures we obtained are almost certainly a significant underestimate, they also represent a serious lack of accounting at a state level. Invasive fruit-eating animals (hereafter frugivores) can form novel mutualisms with fruiting plants and alter seed dispersal. Plant-frugivore interactions are often linked to frugivore preference for certain fruit traits, but for invasive frugivores, it is uncertain if novelty—whether or not a fruit is familiar—impacts foraging decisions. We experimentally tested fruit preferences of the four most abundant and frugivorous bird species on O'ahu, a Hawaiian Island, all of which are globally invasive songbirds. With captive wild birds, we tested for preference in relation to fruit size, color, nutrients, and bird sex. We also tested how novelty of fruits affected trait-based preferences, with fruits from plant species either established in the wild or novel. Using arrays that offered multiple fruit species, we conducted 252 trials with 111 plant species, 59.5% of which were considered novel. From all fruits tested, three bird species preferred smaller fruits, and color preferences varied among bird species. Novelty of fruits influenced preference in three ways. First, birds preferred fruits from plants that were established in the wild over novel fruits. Second, repeated exposure to novel fruits increased the probability of interactions. Third, preferred novel fruits were smaller and redder than preferred fruits from established plants. Finally, bird sex and fruit nutrient content did not affect preference. Together, these results suggest that preference for fruit traits varies among invasive bird species and can be flexible when fruits are novel to birds, increasing the likelihood of novel mutualisms and making it increasingly difficult to predict invasion impacts. Urban heat island effects (UHIEs) and spatial growth remain global existential challenges in the 21st Century. Nevertheless, research on UHIEs in Ghana remains limited, particularly in the northern part. Based on the interrelationship between land surface temperatures (LST) and UHIs, the present study, using spatial data and statistical analytical techniques, comprehensively analyses the nexus between Tamale’s spatial growth and LST and their implication for UHIEs. The results indicate a notable increase in developed areas from 1.89% in 2003 to 3.7% in 2013 and 9.2% in 2023. Notably, the city’s vegetation cover decreased from 3.48% to 2.1% between 2003–2013 and 2013–2023. Factors contributing to the decline in Tamale’s vegetation cover include increasing housing supply, grey infrastructural development, and intensified economic activities. Additionally, Tamale’s surface temperature rose from 45.72 °C to 51.24 °C between 2003 and 2013 and from 51.24 ℃ to 62.9 ℃ between 2013 and 2023. We observed from further analysis that a positive relationship exists between land cover and vegetation, and a weak relationship between land cover, vegetation, and LST. Specifically, there is no statistical evidence to explain temperature variability. Despite this, the study confirms the interplay between spatial growth, LST, and the potential implications for urban planning and sustainable cities in northern Ghana. Given the high rate of spatial expansion, Tamale should anticipate a heightened prevalence and temperature surges, contributing to UHIEs. We conclude the study with recommendations for remedial measures to alleviate the adverse impacts of rising LST and emphasise the importance of initiative-taking urban planning strategies to foster sustainable development amidst rapid urbanisation and climate change challenges. Oxacillin (bactericidal) and tetracycline (bacteriostatic) are clinically relevant antibiotics that are routinely prescribed to treat Staphylococcus aureus infections but not conventionally used in combination. There is an urgent need for treatment regimens that can act upon biofilms during infection, associated with chronic infections on indwelling devices, as well as acute planktonic (systemic) infection. Here we show that in an in vitro model oxacillin and tetracycline act synergistically against S. aureus UAMS-1 biofilms, reducing the concentration of both antibiotics necessary to eradicate an established biofilm. Using an in vivo zebrafish larval infection model with S.aureus NewHG, they display improved bacterial clearance compared to each drug alone and can counteract a loss of host phagocytes, an important innate defence against S. aureus. In these cases, the bacteriostatic nature of tetracycline enhances rather than dampens the bactericidal action of oxacillin, although an exact mechanism remains to be elucidated. We suggest a dual therapy could be of clinical use against biofilm-forming S. aureus and has a potential use in patients with a compromised immune system. [ABSTRACT FROM AUTHOR]

Brucellosis infects humans and animals worldwide but is particularly prevalent in Asia. In many Asian countries, molecular diagnostic tools for accurate molecular diagnostics and molecular epidemiology are lacking. Nonetheless, some countries have conducted in-depth molecular epidemiological studies. The objective of this study was to reveal the genetic relationships, geographic origins, and distributions of Brucella strains across Asia for two primary species, B. abortus and B. melitensis. For this, we systematically searched genotyping data from published studies on the molecular epidemiology of Brucella species for both humans and livestock in Asia. We used data from multilocus sequence typing (MLST), multiple-locus variable-number tandem repeat analysis (MLVA), and whole genome sequencing analysis of Brucella strains. We also analyzed the MLVA genotypes of 129 B. abortus isolates and 242 B. melitensis isolates with known origins in Asia from an online MLVA database using MLVA-11 data in minimum spanning trees and MLVA-16 data in neighbor-joining trees. We found that the B. melitensis East Mediterranean lineage is predominant across the continent, with only a small number of samples from the Africa and Americas lineages, and none from the West Mediterranean lineage. The "abortus C" genotype was the most common group of B. abortus in Asia, with limited genetic variation for this species. Several studies also reported that Near Eastern countries frequently encounter human brucellosis cases of B. abortus from genotypes 42 and 43. Our study highlights the inconsistent collection of genetic data for Brucella species across Asia and a need for more extensive sampling in most countries. Finally, a consistent nomenclature is necessary to define various groupings of strains within a lineage (i.e., clade) so uniform terminology should denote particular genetic groups that are understood by all researchers. ProXp-ala is a key component of the translational machinery in all three Domains of life. This enzyme helps to maintain the fidelity of proline codon translation through aminoacyl-tRNAPro proofreading. In the first step of tRNA aminoacylation, the cognate aminoacyl-tRNA synthetase (aaRS) binds and activates an amino acid in the enzyme's synthetic active site. If a non-cognate amino acid passes this first selection step and is charged onto the tRNA, a distinct aaRS editing active site may recognize the mischarged tRNA and deacylate it. Alternatively, this editing reaction may be carried out by a separate enzyme that deacylates the mischarged tRNA in trans. ProXp-ala is responsible for editing Ala mischarged onto tRNAPro. Since trans-editing domains such as ProXp-ala bind their substrates after release from the synthetase, they must recognize not only the mischarged amino acid, but also the specific tRNA. Previous studies showed that Caulobacter crescentus (Cc) ProXp-ala distinguishes tRNAPro from tRNAAla, in part, based on the unique tRNAPro acceptor stem base pair C1:G72. Previous crystallographic and NMR data also revealed a role for conformational selection by the ProXp-ala α2 helix in Ala- versus Pro-tRNAPro substrate discrimination. The α2 helix makes lattice contacts in the crystal, which left some uncertainty as to its position in solution. We report resonance assignments for the substrate-free Cc ProXp-ala and the NMR-derived three-dimensional structure of the protein. We formally introduce IsaVODEs (Isabelle verification with Ordinary Differential Equations), an open, compositional and extensible framework for the verification of cyber-physical systems. We extend a previous semantic approach with methods and techniques that increase its expressivity, proof automation, and scalability to the level of state-of-the-art deductive verification tools. Our contributions include a user-friendly specification language, a flexible hybrid store model, including vectors and matrices, and separation-logic-style rules for local reasoning with hybrid stores using a novel form of differentiation called framed Fréchet derivatives. The formalisation of correctness specifications with forward predicate transformers, the certification of flows as unique solutions to systems of ordinary differential equations, and invariant reasoning for such systems also contribute to the scalability and usability of our framework. In combination, these features make our framework flexible and adaptable to several verification workflows. Purpose: This paper describes a conceptual framework of maintenance of physical activity (PA) and its application to future intervention design. Methods: Evidence from systematic literature reviews and in-depth (N = 27) qualitative interviews with individuals with cancer were used to develop a conceptual framework of long-term physical activity behaviour. Determinants of long-term PA were listed and linked with domains of the Theoretical Domains Framework which in turn were linked to associated behaviour change techniques (BCTs) and finally to proposed mechanisms of action (MoA). Results: The conceptual framework is presented within the context of non-modifiable contextual factors (such as demographic and material resources) and in the presence of learnt and adapted behavioural determinants of skills, competence and autonomous motivation that must be established as part of the initiation of physical activity behaviour. An inventory of 8 determinants of engagement in long-term PA after cancer was developed. Clusters of BCTs are presented along with proposed MoA which can be tested using mediation analysis in future trials. Conclusion: Understanding the processes of PA maintenance after cancer and presentation of implementable and testable intervention components and mechanisms of action to promote continued PA can inform future intervention development. Implications for Cancer Survivors: This resource can act as a starting point for selection of intervention components for those developing future interventions. The Large Fiber Array Spectroscopic Telescope (LFAST) concept involves the utilization of 2640 individual, 0.76 m "unit telescopes" to collect light from a target object. Through optical fibers, the lights from all telescope foci are directed to a high-resolution spectrograph, with fibers subtending a 1.47 arcsec diameter on the sky. The total light-collecting area of all telescopes combined is 1200 m2. Given the telescope's application for spectroscopy of seeing-limited images, the need for coherence is eliminated. The primary goal of the LFAST prototype is to demonstrate a telescope design that can be rapidly and cost-effectively replicated to achieve a 1200 m2 collecting area and match the capabilities of extremely large telescopes (ELTs) presently under construction. Engineering, manufacturing, and design challenges unique to LFAST differ significantly from those encountered in traditional ELTs. Considering that the unit telescope is to be replicated thousands of times, reducing the manufacturing cost per telescope is essential. The aim of the described design is to achieve this goal, with strong considerations for manufacturability, alignment, few moving parts, and high yield. Experience from the first prototype unit telescope built in 2022 will be leveraged to build the first 20-unit tracking module in 2024, with a collecting area of a 3.4-m-diameter mirror. The design is minimalistic and features commercial tolerances, thereby ensuring cost-effectiveness in manufacturing, assembly, and alignment processes. Moreover, the telescope is designed to operate at angles as low as 70° from the zenith by laterally translating a single lens to correct for atmospheric dispersion. We present a combined average of over 80% encircled energy across 2640 Monte Carlo trials. Simulations were conducted using commercially specified tolerances and capabilities to model the overall performance of the array. Highlights: LFAST is an "array telescope" designed to combine light from multiple telescopes. We design a telescope that is highly performing, even at elevations as low as 70° from the zenith. The durability of copper (Cu) canisters against corrosion is critical for the licensing of deep geological repositories. Assessing oxic corrosion, a primary degradation mechanism, is essential for ensuring the reliability of such repositories. Due to the complex interactions influencing oxic corrosion, a comprehensive model is necessary for evaluating Cu canister corrosion. This study develops a model for the KAERI Reference Disposal System (KRS), incorporating mixed-potential theory with key O2 consumption reactions, including Cu corrosion, Cu(I) oxidation, FeS2 oxidation, aerobic microbial activity, and O2 dissolution and consumption. Simulation of 11 scenarios revealed that the representative KRS case would experience a maximum corrosion depth of 9.3 μm on the Cu canister after 2.3 years due to oxic corrosion, under conditions that are unfavorable for the initiation of pitting corrosion. These results suggest that oxic corrosion is not a threat to the isolation of spent nuclear fuels in KRS. [ABSTRACT FROM AUTHOR]

Quantum dots are promising candidates for telecom single photon sources due to their tunable emission across the different low-loss telecommunications bands, making them compatible with existing fiber networks. Their suitability for integration into photonic structures allows for enhanced brightness through the Purcell effect, supporting efficient quantum communication technologies. Our work focuses on InAs/InP QDs created via droplet epitaxy MOVPE to operate within the telecoms C-band. We observe a short radiative lifetime of 340 ps, arising from a Purcell factor of 5, owing to integration of the QD within a low-mode-volume photonic crystal cavity. Through in-situ control of the sample temperature, we show both temperature tuning of the QD's emission wavelength and a preserved single photon emission purity at temperatures up to 25K. These findings suggest the viability of QD-based, cryogen-free C-band single photon sources, supporting applicability in quantum communication technologies. [ABSTRACT FROM AUTHOR]

Background: CBASP is a learning acquisition model of psychotherapy and until this present study, has not been systematically dismantled to identify the active ingredient. Methods: The present investigation is the first formal dismantling pilot-study conducted to date on the Cognitive Behavioral Analysis System of Psychotherapy (CBASP). The two major treatment components of CBASP, Situational Analysis (SA) and the Interpersonal Discrimination Exercise (IDE), were administered alone and with CBASP treatment-as-usual (the combination of both) to determine if there were active ingredients operative in the model. Secondly, we questioned if the dismantling of CBASP should continue in a larger study in an effort to answer the greater dismantling question of whether to administer both SA and IDE separately based on the active ingredient outcomes. Results: The study did achieve visually inspected differential data patterns across the three groups of the dependent measures. Based on these outcomes, a larger dismantling study seems warranted and is recommended. Purpose: The epidemiology of paediatric fractures has been previously described, however there is limited data available on open fractures in this population. The purpose of this study was to investigate trends, mechanism of injury (MOI) and severity of paediatric open fractures and undertake an epidemiological study. Methods: All children ≤ 16.0 years presenting with open fractures were identified between 01/04/2013 and 01/04/2023. Those with craniofacial, thoracic and distal phalangeal fractures were excluded. Incidence was calculated based on those presenting within the local geographical region. Social deprivation was measured using the Index of Multiple Deprivation (IMD). Results: There were 208 open fractures with a median age of 11.0(q1 7.4-q3 13.4) years, and 153(74.6%) were in males. The MOIs were road traffic collisions 73(35.1%), sports/play 45(21.6%), fall > 2m 29(13.9%), simple fall 25(12.0%), crush 16(7.7%), bites 8(3.8%), assault 6(2.9%), and other 6(2.9%). Nineteen children (9.1%) presented with polytrauma. Gustilo-Anderson grade for long bone fractures were I-61(29.3%), II-24(11.5%), IIIa-36(17.3%), IIIb-30(14.4%) and IIIc-7(3.4%). There were 129 children presenting within the local geographical region providing an annual incidence of 8.0/100,000. Radius and ulna were the most frequently injured 49(38.0%) followed by tibia and fibula 44(34.1%). There were 69(53.5%) children presenting from an IMD quintile 1 with open fractures. Conclusion: Paediatric open fractures are commonly seen in the adolescent male and affect those who are from a more socially deprived background. These injuries account for 3.2% of fractures admitted to a MTC. Data suggests children principally sustain open fractures through two distinct injury patterns and ten-year trends suggests that there is a gradual decline in the annual incidence.
