Eucheumatoid cultivation is an important alternative livelihood for small-scale farmers in coastal areas in east Malaysia. Over the last 50 years, the same clones of the red eucheumatoid Kappaphycus alvarezii and Kappaphycus striatus have been vegetatively propagated for cultivation resulting in low genetic diversity. This has made the cultivars more vulnerable to pests and diseases, which has been further exacerbated by climate change. There is an urgent need, therefore, to develop new cultivars that are resilient to pests and diseases, as well as environmental change. In this study, wild eucheumatoids were collected from four regions in Semporna, East Malaysia: Kerindingan, Sibuhun, Sebangkat and Omadal. These eucheumatoids were then grown at a commercial seaweed farm in Silungun, Semporna using two methods: i) tie-tie and ii) basket net for 60 days and their performance was evaluated based on growth and survival rates. A higher mean specific growth rate (SGR) was recorded by wild-collected Kappaphycus spp. (2.15 ± 1.90% day−1) and Eucheuma denticulatum (2.83 ± 3.27% day−1) when tie-tie was used. However, survival of the wild-collected eucheumatoids was better with the basket net, which led to a reduction in grazing and prevented detachment from culture lines. Of 212 original individuals, 72 survived throughout the study, despite the prevalence of pests and ice-ice disease. Of these, 46.15% Kappaphycus spp. and 50.75% E. denticulatum survived when cultivated using the basket net and 17.20% Kappaphycus spp. and 30.77% E. denticulatum survived when cultivated using tie-tie. The mean SGR and survival rate were significantly correlated with temperature, salinity, pH, current velocity and rainfall (p < 0.05). In an integrated multitrophic aquaculture (IMTA) system, seaweeds serve as extractive species that utilize excess nutrients, thereby reducing the risk of eutrophication and promoting sustainable aquaculture. However, the use of excessive fish feeds and the resultant faecal waste as nutrient streams can contribute to variations in nitrogen and phosphorus levels (e.g., primarily NH4+ and PO4−3) in the surrounding area and this may impact the physiology of the integrated seaweeds, particularly on how these species take up inorganic nutrients. In this study, the effect of different PO4−3 levels on NH4+ uptake of the three commercially important eucheumatoids Kappaphycus alvarezii, Kappaphycus striatus and Eucheuma denticulatum was examined under laboratory conditions. Seaweed thalli (n = 4) were incubated in seawater media containing 30 µM NH4+, and 0, 0.5, 1.0, 1.5, 3.0 or 5.0 µM PO4−3 for 1 h under a saturating light level of 116 ± 7.13 µmol photons m−2 s−1 inside a temperature-controlled laboratory. Species-specific responses to PO4−3 levels were observed. For K. alvarezii, maximum NH4+ uptake (17.8 ± 1.6 µmol gDW−1 h−1) was observed at 0.5 µM PO4−3 and the uptake rate declined at higher PO4−3 levels. For K. striatus, NH4+ uptake increased with increasing PO4−3 levels, with maximum N uptake (6.35 ± 0.9 µmol gDW−1 h−1) observed at 5.0 µM PO4−3. For E. denticulatum, maximum NH4+ uptake (14.6 ± 1.4 µmol gDW−1 h−1) was observed at 1.0 µM PO4−3. Our results suggest that among the three eucheumatoid species, the NH4+ uptake of K. striatus persists even at high levels of PO4−3. However, our results also showed that K. striatus had the lowest range of NH4+ uptake rates. Studies have shown that cultivars of the carrageenophytes Kappaphycus and Eucheuma are clones of a limited number of strains originally domesticated from wild populations. For the development and selection of new cultivars, it is important that a comprehensive record of available variants exists. This study was conducted to provide up-to-date analysis and compilation of the current state of cultivars as the last list of cultivars was compiled nearly a decade ago. The present study analyzed the cox2–3 spacer and cox1 (1356 bp) genetic diversity of cultivars collected from 2019 to 2020 from the east coast of Sabah where the seaweed farms are concentrated. These data were compared with cultivars reported from 2010 to 2012 to assess changes, if any, to the gene pool of farmed eucheumatoid in Malaysia. Kappaphycus alvarezii, K. striatus, and K. malesianus are currently cultivated while Eucheuma denticulatum is no longer an important cultivar compared to a decade ago, probably due to its lower price. Analysis of the cox2–3 spacer revealed a new haplotype, LBT10, and, by including published GenBank data, a further four previously unnamed haplotypes were recognized from Sabah. This study confirms that there is a limited gene pool within cultivars in Malaysia and suggests the need for new or genetically diverse cultivars which can adapt to a changing environment, to ensure a more sustainable carrageenan industry. The impact of cottonii (COT, Kappaphycus alvarezii) and spinosum (SPI, Eucheuma denticulatum) flour (1–10% w/w) on rheological properties of wheat (WHE) and quality of bread was investigated. Wheat-COT and WHE-SPI had higher water absorption capacities, dough development times, dough stabilities and farinograph quality numbers than WHE dough. Extensograms of composite dough were largely similar to those of WHE at the same incubation times. Dough energy decreased whereas elasticity ratio (ratio of maximum resistance to extensibility) increased when incubation time was increased from 45 to 135 min. Wheat-COT and WHE-SPI had lower pasting temperatures but higher peak, final, breakdown and setback viscosities than WHE flour. Specific volume of bread containing COT (5% w/w) or SPI (7.5% w/w) was not different (p > 0.05) from that of WHE bread. Crumb texture properties of these composite breads were also largely similar to those of WHE bread. The physical properties of WHE dough and bread were influenced by gluten, whereas those of WHE-COT and WHE-SPI were influenced by gluten in addition to κ-carrageenan in COT and ι-carrageenan in SPI. Protein, ash and dietary fibre contents increased (p < 0.05) whereas carbohydrate content of bread decreased (p < 0.05) when WHE was replaced with COT or SPI (5% w/w). [ABSTRACT FROM AUTHOR]

Farming of eucheumatoid seaweeds is a widespread, promising activity and an important livelihood option in many tropical coastal areas as for example in East Africa, Western Indian Ocean (WIO). Compared to other types of aquaculture, seaweed farming has generally low impact on the environment. Nonetheless, there are potential direct or indirect negative effects of seaweed farming, such as introduction of alien species and changes in local environmental conditions. Although farming has been practiced in this region during several decades, the knowledge concerning the actual environmental impacts from faming non-native eucheumatoid haplotypes and consequently how to manage farming activities to mitigate those is highly limited. In this review, we provide a summary of the current scientific knowledge of potential direct and indirect negative environmental effects linked to eucheumatoid seaweed farming such as alterations of benthic macrophyte habitats and loss of native biodiversity. Furthermore, we highlight knowledge gaps that are of importance to address in the near future, e.g., large-scale ecosystem effects and farms as potential vectors of pathogens. We also provide a number of feasible management recommendations to be implemented for a continued development of environmentally sustainable seaweed farming practices in the WIO region, which includes spatial planning of farms to avoid sensitive areas and farming of native haplotypes of eucheumatoids instead of introduced specimens. Indonesia is the world largest producer of the red seaweeds Kappaphycus and Eucheuma; however, this country is facing significant challenges such as disease outbreaks, epiphyte infestations and a loss in seedling quality. Biosecurity practices have been widely adopted in other aquaculture sectors and when enforced can help to limit the introduction and spread of diseases and pests. To assess current capacity for biosecurity in seaweed aquaculture in Indonesia, a systematic analysis of policy frameworks including legislation, regulatory tools, and national standards was conducted. Biosecurity themes and risks were used to evaluate current national biosecurity content. The results identified major challenges faced by the industry in order to implement biosecurity policies in practice. Barriers to implementation included unspecific reference to the seaweed aquaculture sector, limited variety of approaches to biosecurity, limited use of up-to-date scientific evidence, insufficient guidance for the use of precaution and insufficient inclusion of specific biosecurity hazards. In general, although national regulations are currently under revision, current policies indicate a lack of clarity where biosecurity is included. Six recommendations are suggested to incorporate proactive biosecurity actions into current frameworks, with the aim of improving the health and sustainability of the seaweed aquaculture sector in Indonesia. Eucheumatopsis isiformis Núñez-Resendiz, Dreckmann & Sentíes is a red alga with great economic potential due to its iota-carrageenan content that could serve as a substitute for traditional carrageenan sources (Freile-Pelegrín and Robledo, J Appl Phycol 20:537–541, 2008) such as the red alga Kappaphycus alvarezii (Loureiro et al., New Phytol 206:489–492, 2015), if it can be reliably and sustainably cultivated in open water. Due to prevailing and changing climatic conditions, it is necessary to retain germplasm and to cultivate this alga in tank systems. To develop the necessary, large volumes of seed stock required for stocking open water cultivation farms, three brown seaweed-derived biostimulants were tested. One of these was manufactured from the kelp Ecklonia maxima, the other two were manufactured from the intertidal fucoid Ascophyllum nodosum. Apical segments of E. isiformis were treated either by dipping or by continuous exposure to the biostimulants, at different concentrations. Growth was assessed every seven days by measuring the maximum length of each segment, number of lateral shoots produced by each shoot, fresh weight, and the maximum diameter. The constant exposure treatment produced a greater number of shoots per propagule with no significant effects driven by any of the biostimulants. Increasing or decreasing the concentration of the biostimulants to improve the performance of the dipping protocol was not effective, as compared with a constant exposure protocol. This study supports the increasing number of research articles related to the positive benefits of using macroalgal-derived extracts as biostimulants to support seaweed cultivation. Pests and diseases of eucheumatoid farming were studied in Lian Bay, Hainan Province, China, from March 2009 to December 2013. Neosiphonia savatieri, ice-ice disease, and the herbivorous fish Siganus fuscescens were found to be the dominant pests and disease. Infections of N. savatieri increased severely in May 2009 and especially so in August 2009. Then, they remained severe the whole year round, until all the Kappaphycus spp. in the bay was wiped out by October 2010. The outbreak pattern of ice-ice disease was the same as that for N. savatieri. Based on our results, the period mostly from May to August and also into October was the period of frequent outbreak of the epiphyte and ice-ice disease at this Kappaphycus spp. farm. Different from the Kappaphycus spp., Eucheuma denticulatum showed more resistance to N. savatieri and ice-ice disease at the same time and location. A reduced occurrence of N. savatieri and ice-ice disease infections in Kappaphycus spp. was observed when Kappaphycus spp. were co-cultured with E. denticulatum during July-August but not during September-October. The fish S. fuscescens swarmed into the bay from the outside sea from April to May, when the seawater temperature increased to above 26 °C. Interestingly, the S. fuscescens usually prefer N. savatieri to Kappaphycus alvarezii, and they grazed on K. alvarezii only after the amount of N. savatieri became insufficient to meet their needs. Acyclovir is a commonly-used drug for treating herpes simplex virus (HSV) infections, but with its wide clinical application, more and more resistant strains have been found. Therefore, seeking a drug that can act against acyclovir-resistant virus has become an important goal of drug screening and development. In this study, plaque reduction assay, real-time PCR, Western blot, and immunofluorescence technique were used to investigate the antiviral effect of Eucheuma gelatinae polysaccharide (EGP) on HSV and to preliminarily clarify the in vitro anti-HSV mechanism of EGP. EGP was found to significantly inhibit HSV infection in vitro and displayed a good inhibitory effect on acyclovir-resistant strains. More detailed experiments have shown that EGP prevented early HSV-1 infection through directly inactivating HSV-1 particles and impairing virus attachment, but without effect on viral penetration. EGP also inhibited the RNA synthesis of HSV-1 early gene and late gene as well as viral DNA replication; no effect on immediate-early gene synthesis was observed. Besides, through immunofluorescence and western blot, we found that EGP significantly affected the protein synthesis of HSV-1. Taken together, these results demonstrate that EGP exerts its anti-HSV activity mainly through impeding early HSV-1 infection and inhibiting viral RNA and DNA syntheses. The weak cytotoxicity, strong viral inactivation as well as attachment inhibition activity enable EGP to be a virucide candidate for HSV therapy, especially for drug-resistant strains. The commercial importance of carrageenophytes Kappaphycus and Eucheuma is well known, with much interest in terms of cultivation, marketing, and research. Considering the many lucrative prospects, these red seaweeds were introduced into various parts of the world for farming, where merely a few were comprehensively documented. Despite being extensively cultivated throughout Southeast Asia, the genetic diversity of Kappaphycus and Eucheuma is poorly studied, where heavy reliance is placed on the use of local or commercial names for identifications. This study used the mitochondrial-encoded cox1 and cox2-3 spacer genetic markers to investigate the Kappaphycus and Eucheuma haplotypes, cultivated and wild, available throughout Southeast Asia. Concatenated cox1- cox2-3 spacer datasets were also analyzed. The near full-length cox1 gene is preferred at revealing the genetic diversity of Kappaphycus and Eucheuma, provided a larger reference database is available. Both molecular markers were capable of delineating common members of the genus Kappaphycus (i.e., Kappaphycus alvarezii, Kappaphycus striatus, and Kappaphycus cottonii) and Eucheuma denticulatum, and revealed interesting genotypes and new species which may be potential alternatives to the common cultivars as well as materials for research. The Brunei-Indonesia-Malaysia-Philippines East Asia Growth Area (BIMP-EAGA) is located within the Coral Triangle, known to have the world's richest biodiversity in marine flora and fauna. This region lies within the 10° N and 10° S of the Equator where natural populations of both Kappaphycus and Eucheuma grow luxuriantly and abundantly. It is in this same region where commercial cultivation of Kappaphycus and Eucheuma began in the Philippines around the mid-1960s. Commercial farming of Kappaphycus (which was originally called Eucheuma) was successful in the Philippines from the early 1970s, after which the technology was transferred to Indonesia and Malaysia in the late 1970s. No seaweed cultivation has been reported in Brunei. At present, carrageenophytes are cultivated in sub-tropical to tropical countries circumferentially around the globe within the 10° N and S of the Equator. However, their combined production is still low as compared to Indonesia, the Philippines, and Malaysia. Notably, few improvements in farming techniques have been made since its first introduction. Some of the major improvements were the introduction of deep-water farming using hanging long lines, multiple rafts, and spider webs in the Philippines; the use of short and long 'loops', instead of plastic 'tie-tie' in Indonesia; and mechanization in harvesting and use of solar 'greenhouse' drying in Malaysia. Commercial cultivation of tropical red seaweeds in the BIMP-EAGA region is dominated by Kappaphycus and Eucheuma (carrageenophytes) and Gracilaria (agarophytes) and the area became the major region for the production of carageenophytes and agarophytes globally. In particular, Indonesia is a major center for the production of Gracilaria. There is an increasing demand for other agarophytes/carrageenophytes in the international market such as Gelidium spp., Pterocladia spp., Porphyroglossum sp., and Ptilophora sp. for paper and ethanol production in Indonesia and Malaysia, and Halymenia for phycoerythrin pigments in the Philippines currently pursued in an experimental stage. A summary of the present status, problems, sustainability, and challenges for the cultivation of tropical red seaweeds in the BIMP-EAGA region are discussed in this paper. [ABSTRACT FROM AUTHOR]

Seaweed farming in the Western Indian Ocean (WIO) Region is carried out in a number of countries, most of them farming Eucheuma denticulatum, Kappaphycus alvarezii and Kappaphycus striatum. These species are farmed mostly in Tanzania with limited production in Madagascar, Mozambique and Kenya; current production (2012) stands at 15,966 t (dry weight) year of Eucheuma and Kappaphycus, valued at US$ 4.2 million with 95 % of this tonnage coming from Tanzania. Other countries in the region have limited or no seaweed production owing to problems of epiphytes, ice ice and markets. The problem of epiphytes coupled with ice ice that WIO countries are facing causes die-off of Kappaphycus which is the preferred species in foreign markets for its thicker gel, kappa carrageenan (vs. the weaker iota carrageenan from Eucheuma). New efforts are put to curb these problems including moving seaweed farms to deeper waters and cultivation trials of other carrageenophytes as well as agar-producing species, agarophytes. Research work has been initiated to evaluate Gracilaria and Hypnea farming and processing in Tanzania, the Republic of Mauritius and Mayotte. Gracilaria farming is at experimental stages as a biofilter of fishpond effluents and as potential species for the production of agar with growth rates of 1.5-1.9 % day. Hypnea farming is only being initiated in Mauritius and Mayotte at present. Other innovations including value addition by making various seaweed products and encouraging the consumption of seaweed as food at least in Tanzania and Mauritius are increasing further the importance of the seaweed farming and processing industry in the WIO Region. The genera Kappaphycus Doty and Eucheuma J. Agardh are important sources of carrageenan in Malaysia, offering lucrative revenues to the carrageenan industry, economy, and the local community. The extensive range of morphotypes and the lack of distinct morphological characteristics led to the application of molecular systematics in elucidating this taxonomic confusion. Local varieties of Kappaphycus and Eucheuma, identified using putative external morphology, were analyzed using the mitochondrial cox2-3 spacer and plastid RuBisCO spacer molecular markers. Phylogenetic analysis of these and non-local specimens indicate that Kappaphycus and Eucheuma are genetically distinct. Three main genotypes of Kappaphycus alvarezii were identified, of which two are extant in Hawaii. Morphological and color variations are not supported by molecular data, indicating that most of the local names are not genetically based. Both the cox2-3 spacer and RuBisCO spacer generated phylogenetic trees with similar topology except in variation of nodal supports. The two markers showed clear separation between Kappaphycus and Eucheuma and the existence of three Malaysian Kappaphycus cultivars. Cox2-3 spacer data is more variable and provides better resolution than the RuBisCO spacer, showing that Kappaphycus is more diversified with a larger number of genotypes, strains, and species which are unique to Southeast Asia. Kappaphycus sp. ' Aring-aring' appeared to be phenotypically and genotypically different from other Kappaphycus congeners, whereas Kappaphycus striatum exhibited two different genotypes. Our data indicate that Eucheuma denticulatum is the dominant species in Malaysian waters and also suggested paraphyly in Eucheuma which will require further studies. The application of molecular taxonomy on Malaysian Kappaphycus and Eucheuma proves useful, offering valuable insights into the taxonomy and distribution of these commercially important Rhodophytes. Seaweed farming has a crucial role in the development of future sustainable mariculture. In the same time, spreading of introduced species or genotypes from farms may threaten local ecosystems. We analyzed a molecular marker (mitochondrial cox2-3 spacers) from cultivated and wild specimen of the widely farmed seaweeds Eucheuma and Kappaphycus, collected in Zanzibar on the African east coast where commercial farming was introduced in 1989. Genotypes of presumed Asian origin were found growing on coral reefs and drifting in seagrass meadows, indicating that genotypes introduced for farming have established successfully in the wild in Zanzibar. Only a very low number of genotypes, all of Asian origin, were found in the farms. This indicates a low accessible gene pool, which can limit the capacity for adaptation to changed conditions and disease resistance in the farming system. African genotypes were found in a few sites, showing the potential for future farming of native strains. The ecological effects of the Asian genotypes introduced to coral reefs should also be further investigated in order to evaluate the risk connected with further introductions of new foreign strains. The variety described in this paper is rather a rare and interesting one. Locally called 'endong', this was discovered first in a farm in Tawi-Tawi, Sulu Archipelago, Philippines. During harvest, seaweed farmers used to mix this with 'tambalang' ( Kappaphycus alvarezii (Doty) Doty ex Silva var. tambalang Doty, the 'cottonii' of commerce) because of the presumption of it producing the same carrageenan type as that of 'tambalang', viz., kappa-carrageenan. The two farmed varieties are similar in appearance. Complaints arose from seaweed processing plants when the resulting gelling properties (hard and brittle gel) expected of the kappa-carrageenan-producing 'tambalang' were not met. Because of its lax habit and long, smooth, and naked basal axes, 'endong' was initially thought to be related to 'tambalang' and differ from the farmed 'spinosum' ( Eucheuma denticulatum (Burman) Collins & Hervey) whose typical or common form has shorter axes covered from base to apex with spinous determinate (ramuli) or indeterminate branchlets arranged irregularly and in whorls at predictable interval. However, a closer look at 'endong' shows that it also has spinous branchlets arranged in whorls, but confined to the apical portion of the branches. Moreover, its internal axial anatomy shows rhizoidal filaments forming a core. Both characters fit well the distinguishing taxonomic criteria established for E. denticulatum. Chemical characterization of its carrageenan and the determination of its phylogenetic affinities support our morphological findings. Hence, we report and describe 'endong' as a rare variety, and we name it E. denticulatum (Burman) Collins & Hervey var. endong Trono & Ganzon-Fortes var. nov. Seaweed is a macroscopic marine algae which has been used as human food since ancient times. Food values of seaweed are mainly due to significant amounts of protein, vitamin, mineral, trace elements and dietary fibre apart from substances of antibiotic nature. In this study edible red seaweed Eucheuma ( Kappaphycus alvarezzi), was used as an ingredient in the preparation of spice adjunct. Functional properties such as soluble solids, water-holding capacity, oil holding capacity and swelling capacity of Eucheuma were studied. Steamed Eucheuma powder was added at levels of 15, 20 and 25% in the preparation of spice adjunct. Sensory analysis and consumer acceptability study of spice adjunct showed that the incorporation of Eucheuma upto 20% had high acceptability. Spice adjunct containing 20% Eucheuma had 6.2% moisture, 22.2% ash, 29.2% fat, 10.0% protein, 9.4% crude fibre and 23% carbohydrates. Based on equilibrium relative humidity studies metallized polyester was selected as the packaging material. Storage studies indicated that metallized polyester provided a shelf life of 120 days at both the ambient and accelerated conditions. [ABSTRACT FROM AUTHOR]

The systematic classification of the Eucheumatoideae is difficult because of their variable morphology and interpretation of reproductive structures. Kappaphycus and Eucheuma specimens cultivated on the Hainan and Fujian coast of China were introduced from Vietnam, the Philippines and Indonesia. Combined with morphological characteristics, all Kappaphycus and Eucheuma cultivated strains were identified by internal transcribed spacer (ITS) sequences. The phylogenetic tree was constructed using neighbor-joining and maximum likelihood methods. The results indicate that different ITS sequence lengths occurred in the different genera and species. An obvious difference in morphology could be found in the protuberance shape between Kappaphycus and Eucheuma. The protuberance in Eucheuma was thorn-like and in Kappaphycus was wartlike or papillate. Their ITS sequence lengths differed significantly in nucleotide variation rates up to 58.55%-63.90%. All nucleotide variations occurred in the ITS1 and ITS2 regions except for five nucleotide transversions in the 5.8S rDNA region. In addition, the difference was at the branches among congeneric species. Kappaphycus sp. had branches with small buds, while K. alvarezii did not have such a feature. The nucleotide variation rates varied from 7.02% to 7.48% among species; within the same species of the clades it was <1.20%. Eucheumatoideae algae cultivated in China consisted of three clades, K. alvarezii, Kappaphycus sp., and E. denticulatum. The results indicate that ITS sequence analysis was an effective way for identification of interspecies and intraspecies phylogenetic relationships and might provide a clue for molecular identification of algal Eucheumatoideae. Carrageenan, a hydrocolloid used in the food and pharmaceutical industry mainly as a thickener and stabilizer, has an annual global market of US $450 million. In Mexico, carrageenan imports doubled during the last years, reaching 4,400 t in 2008. Any carrageenan industrial project demands a regular supply of raw material from reliable sources capable to meet and maintain required volumes and qualities. Some carrageenophytes have been exploited traditionally in Baja California Peninsula (i.e., Chondracanthus canaliculatus). The recent demand, however, for carrageenan has increased the development of cultivation of economically important seaweeds mainly in the tropics. In the Yucatan peninsula, experimental cultivation of native and exotic species has been carried out during the last 10 years. Eucheuma isiforme, Solieria filiformis, Halymenia floresii, and the introduced Kappaphycus alvarezii represent potential raw material sources for the production of carrageenan. Demand for iota carrageenan-producing species which could be sourced from native species is not a market priority. On the contrary, the most in demand are kappa and lambda carrageenan-producing species. Abstract  The yield and physicochemical properties of native and alkali treated carrageenan from Eucheuma isiforme harvested from the Nicaraguan coast were investigated. The native carrageenan yield was 57.2% of dry weight and decreased to 43.5% when the alga was alkali treated. Native carrageenan viscosities showed significant differences between native (144.6 ± 3.3 cPs) and treated carrageenan (113.9 ± 2.6 cPs) (p −2). Chemical analysis and FTIR spectra revealed that Eucheuma isiforme from Nicaragua is a good source of relatively pure iota-carrageenan with sufficient quality to serve as a substitute for traditional iota-carrageenan sources. The tensile and tear strength of carrageenan film from Philippines Eucheuma species were investigated using NEC tensilon universal-testing machine according to American Society for Testing Materials methods. These properties are important for assessing carrageenan film as packaging material. The κ and ι types were used in the study. The effect of glycerine on the tensile and tear strength including elongation was also evaluated. Addition of glycerine tended to lower the tensile strength of the film and increase its elongation properties including the tear strength. Carrageenan film without glycerine was much stronger. Glycerine made the film more flexible and easy to deform. The composite film of carrageenan and konjac gum did not exhibit elongation. It also showed higher tensile strength than did the composite film of carrageenan and xanthan gum. Compared with ι-type carrageenan film, κ-type carrageenan film without glycerine was more comparable to low-density polyethylene (LDPE) film in terms of tensile strength as was the composite film of carrageenan–konjac gum. The κ-type carrageenan film with glycerine was more comparable to LDPE film in terms of tear strength. The elongation reading for carrageenan film was lower than that for LDPE film. Morphologic studies showed that the carrageenan film had sets of pores distributed randomly at different places as compared to LDPE film. The mechanism of infection by Vibrio sp. P11 promoting the ice-ice disease in Kappaphycus alvarezii was investigated in vitro. Its intensity of infection differs from that of another ice-ice promoter (Cytophaga sp. P25) by promoting the disease much faster. However, when secondary infection by other bacteria starts, its ability to compete with these bacteria gradually diminishes, whereas, infection by P25, although not displaying such drastic effects as P11, shows consistent competitive ability against other bacteria. Time-series infection experiments with application of polyclonal antibodies to specifically detect Vibrio sp. P11 revealed that this bacterium has a high affinity for the seaweed especially when the latter is stressed. It promotes the disease after a rapid increase in cell density of up to 107 g−1 (wet wt.) in the first 24 h. This bacterial cell build-up may take only 1–2 h on stressed thalli, but takes about 24 h on non-stressed thalli. Build-up is not sustainable in non-stressed thalli as high density is usually followed by a sudden decline in cell number believed to result from an algal defence against potential pathogens. Inoculation of the bacterium on thalli incubated in continuous culture system extends the time of bacterial attachment due to laminar flow and, possibly, competition by existing bacteria on the seaweed surface and in ambient seawater medium.
