Knowledge on the interaction between non-vascular plants and pathogens is very scarce. This study is the first to report on the transmission of a satellite-associated cucumber mosaic virus (CMVsat) into the moss Physcomitrium patens, and only the second report on the successful mechanical inoculation of a virus into P. patens. Inoculation was successful on agar and on soil, the latter possibly constituting a more natural environment for the interaction. Additionally, this growth condition allowed the monitoring of the infection for an extended period of time without contamination. The concentration of the CMVsat coat protein was higher in plants incubated for 10 days than those incubated for 5 days. The CMVsat infection did not activate the biosynthesis of salicylic acid in moss. Flavonoids were predominantly located near the outer part of the P. patens cell and in the nucleus, the major compound was quercetin and its concentration was higher after infection. Moreover, the concentration of indole-3-acetic acid (IAA) also increased. The level of CMVsat was lower in GH3-double knockouts which, under control conditions, showed higher IAA levels than wild type, and IAA did not change after infection in GH3-double knockouts. The high IAA concentrations in the knockouts could have been a factor that reduced viral infection. An interaction between SA, quercetin and IAA during the response of P. patens towards CMVsat is possible and therefore a challenging subject for future research. [ABSTRACT FROM AUTHOR]

Phytoplasmas are phytopathogenic bacteria that cause serious damage to agriculture. A quarantine pathogen, the flavescence dorée phytoplasma (FDp), often associated with grapevine yellows disease, affects viticultural production across Europe. However, the mechanisms of FDp pathogenicity are not still elucidated. In this study, symptomatic and asymptomatic grapevine (Vitis vinifera L. var. ‘Pinot gris’) leaves were sampled. Two different FDp genotypes (M38 and M54) were identified, and genotype-dependent changes to grapevine physiological responses through the development of FDp infection were analysed. Correlation analyses established a potential link between measured physiological parameters and relative FDp DNA abundance. Increased malondialdehyde levels pointed to the oxidative stress in infected leaves, and highly correlated with the activation of L-ascorbic acid synthesis. Levels of hydrogen peroxide were reduced in infected leaves, possibly as an FDp mechanism to avoid plant-derived oxidative damage. Genotype M54 was associated with a lower accumulation of soluble sugars and lower damage to photosynthetic pigments while retaining a higher titre than M38. Therefore, pronounced phytoplasma genotype-dependent changes in grapevine physiology, potentially caused by the differences between M54 and M38 on the level of the efficiency of their effectors should be further investigated. Altogether, results provide data on certain targets of FDp in grapevine and could assist in the identification of potential specific effectors of this phytoplasma to aid the efforts of FDp management in European vineyards. The purpose of this study was to compare the polyphenol, antioxidant and antidiabetic potential of powdered Matcha and bagged Sencha tea during in vitro digestion. Total phenols (TP), flavonoids (TF), flavanols (TFLA), antioxidant and antidiabetic (α-glucosidase inhibition) activity were higher in Matcha tea before and in most in vitro digestion phases. Upon gastric digestion, in Matcha tea TP, TF, TFLA were 2.6, 1.4 and 1.2 times significantly higher (p ≤ 0.05), respectively; gallic acid, gallocatechin, epigallocatechin, quercetin and kaempferol 1.5, 1.6, 1.8, 1.7, 1.2 times, respectively; whereas antioxidant activity was significantly (p ≤ 0.05) higher 3.2 and 1.1 times with ABTS and FRAP and α-glucosidase inhibition 1.8 times. After the intestinal phase, TP and TFLA were 3.4 and 1.7 times significantly (p ≤ 0.05) higher, respectively, antioxidant activity was significantly (p ≤ 0.05) higher 2.4 and 2.0 times with ABTS and FRAP, respectively, while inhibition of α-glucosidase was 1.7 time significantly (p ≤ 0.05) higher in Matcha tea, but the differences in TF, TP and identified phenolics (with the exception of gallic acid) between Matcha and Sencha tea were neutralized. Our results are the first to demonstrate that, during digestion of Matcha powder together with its water extract, Matcha polyphenols are more bioavailable and possess higher antioxidant and antidiabetic activity compared to Sencha. Due to extreme climate changes, plants are exposed to severe soil water content, i.e. flooding and drought. The aim of this study was to assess the effect of different soil water content on the nutritional potential of young Chinese cabbage (Brassica rapa ssp. pekinensis) on the level of phenolics, sugars, photosynthetic pigments and antioxidant capacity. Total phenolic acids were induced in plants grown under drought conditions. Both types of stress increased soluble sugars in Chinese cabbage but reduced total tannins. Plants grown under drought had more L-ascorbic acid than the control group; however, the concentration of ferulic acid and quercetin was reduced. On the other hand, excess of water increased the amount of sinapic and ferulic acid, main hydroxycinnamic acids in Chinese cabbage. Photosynthetic pigments were more susceptible to flooding (reduced amount) than drought, the only exception was chlorophyll b whose concentration was significantly higher in plants grown under drought. In addition, among all the measured parameters, chlorophyll b reacted most specifically against flooding (decreased) and drought (increased). Chinese cabbage grown under flooding had reduced amount of porphyrins compared to the one grown under normal and drought conditions, but showed higher antioxidant capacity (ABTS). Based on the results, the use of excess water could be considered as a possibility when growing young Chinese cabbage since it increased the concentration of antioxidants sinapic and ferulic acid and resulted in higher antioxidant capacity, recorded by ABTS assay, compared to control plants. The phytochemical and antioxidant properties of mature (head stage) Chinese cabbage (Brassica rapa ssp. pekinensis) are known; however, data on the phenolic profile, vitamin C (L-ascorbic acid) content and antioxidant capacity of its fresh sprouts are lacking. Since the human consumption of fresh cruciferous sprouts has significantly increased in recent years, their nutritional characterization has become a somewhat urgent matter. Therefore, in this study the contents of total phenolics, flavonols and hydroxycinnamic acids were measured spectrophotometrically, whereas individual flavonoids, phenolic acids and vitamin C were identified and quantified using a newly-developed high performance liquid chromatography method. Also, the antioxidant capacity of five Chinese cabbage sprout growth stages was determined. These stages contained either cotyledons only (seedlings), cotyledons and two leaves, four leaves, six leaves, or ten leaves. Principal component analysis (PCA) and hierarchical clustering (HC) were implemented in order to visualize the classification trend between the stages. Seedlings contained more sinapic acid and vitamin C than older plants. Plants containing six or ten leaves had more ferulic acid and isorhamnetin than younger ones. Total phenolics, flavonols, hydroxycinnamic acids, quercetin and antioxidant capacity did not statistically differ between seedlings and stages with six or ten leaves and their concentrations were significantly higher than in stages with two or four leaves. PCA and HC confirmed the higher phytochemical similarity between seedlings and plants with six or ten leaves than plants with two or four leaves. Therefore, Chinese cabbage seedlings and plants with six or ten leaves should be preferred over plants with two or four leaves, which were ultimately shown to be of lesser nutritional quality. Native Crocus taxa are abundant in regions with moderate and dry climate; however, their potential in the food industry has thus far been neglected. The objective of this study was to compare the quantitative and qualitative flavonoid content and antioxidant and cytotoxic activity in organs of several populations of Croatian native taxa and assess their potential for application in the food industry. The dominant flavonoids in the native Crocus taxa were kaempferol and quercetin. Tepals contained similar or higher concentrations of flavonoid-glycosides than other organs. Tepals from Cvv1 and Cvv2 populations contained more flavonoid-glycosides than the commonly used spice saffron (C. sativus stigmas). The FRAP antioxidant activity of Cvv1 and Cvv5 tepals was similar to that of standard Trolox. DPPH inhibition of Cvv1 and Cvv3 tepals was within the range of that recorded for saffron. Cvv1 tepals significantly reduced reactive oxygen species in the broadest concentration range (50-1000 μg/ml), and showed considerable antioxidant activity in the ABTS assay, equal to 82% of standard Trolox antioxidant activity. A significantly higher concentration of kaempferol-rutinoside was recorded in this than in other taxa. The flavonoid showed a very strong or strong correlation with antioxidant assays results, and a negative correlation with cellular reactive oxygen species concentration. We therefore presumed that kaempferol-rutinoside is one of the main antioxidant phenolics in Crocus tepals. None of the tested extracts showed cytotoxicity toward Caco-2 cells. Spectrophotometric and chromatographic analysis of phenolics in water and ethanolic extracts of wild asparagus, butcher's broom and black bryony from Croatia was conducted. Their antioxidant capacity (ABTS, DPPH and FRAP assay) and energy content were determined. The gastrointestinal stability of detected phenolics was determined using a two-phase in vitro digestion method with human enzymes. The highest phenolics yield, radical scavenging activity and ferric reducing antioxidant potential were recorded in 40 % ethanolic extract of black bryony, with glycosylated forms of kaempferol as dominant components. Quercetin-3- O-rutinoside and isorhamnetin-3- O-rutinoside were dominant phenolics in all wild asparagus extracts, and salicylic acid was predominant in butcher's broom 40 % ethanolic extract. Phenolic acids of the three species were not stable during gastric and duodenal phases of simulated digestion. Two main black bryony kaempferol glycosides were best preserved after digestion (50 % of each). Black bryony contains more energy than wild asparagus and butcher's broom. Accordingly, we propose black bryony as a valuable source of antioxidant kaempferol glycosides with relevant gastrointestinal stability and higher energy content than so far more conventional vegetable wild asparagus. The roles of several phenolic compounds in plant defence response have been extensively studied, yet little is known about the role of flavonoids in plant-virus interaction. Quantitative and qualitative changes of selected phenolics in Arabidopsis thaliana induced by Cucumber mosaic virus containing satellite RNA (CMVsat) infection were analysed accompanied by plant hormone, chalcone synthase and pathogenesis-related gene expression analysis. Lower leaves of infected plants had a lower concentration of total phenolics compared to control plants. The concentration of kaempferol in upper leaves of all infected plants was significantly lower compared to control plants, while the expression of the chalcone synthase gene in those leaves was in most cases upregulated. All infected plants had a higher concentration of indole-3-acetic acid in lower leaves, which was accompanied with a lower concentration of kaempferol in upper leaves. Our research demonstrates a correlation between kaempferol and indole-3-acetic acid in response to CMVsat infection in Arabidopsis. We demonstrated two different metabolic patterns in infected plants suggesting the activation of two different defence responses. We also propose kaempferol to be an important part of the auxin-dependent defence response which limits systemic movement of CMVsat and that this defence response is activated prior to the well-known salicylic acid dependent defence response. Further research on kaempferol and its role in Arabidopsis-CMVsat interaction will improve our understanding on the role of flavonoids in plant defence. [ABSTRACT FROM AUTHOR]
