Nicotinamide adenine dinucleotide (NAD +) plays a pivotal role in numerous cellular functions. Reduced NAD + levels are postulated to be associated with cancer. As interest in understanding NAD + dynamics in cancer patients with therapeutic applications in mind grows, there remains a shortage of comprehensive data. This study delves into NAD + dynamics in patients undergoing surgery for different digestive system cancers. This prospective study enrolled 99 patients with eight different cancers. Fasting blood samples were obtained during the perioperative period. The concentrations of NAD + , nicotinamide mononucleotide (NMN), and nicotinamide riboside were analyzed using tandem mass spectrometry. After erythrocyte volume adjustment, NAD + remained relatively stable after surgery. Meanwhile, NMN decreased the day after surgery and displayed a recovery trend. Interestingly, liver and pancreatic cancer patients exhibited poor postoperative NMN recovery, suggesting a potential cancer type-specific influence on NAD + metabolism. This study illuminated the behavior of NAD + in surgically treated cancer patients. We identified which cancer types have particularly low levels and at what point depletion occurs during the perioperative period. These insights suggest the need for personalized NAD + supplementation strategies, calibrated to individual patient needs and treatment timelines. Clinical trial registration jRCT1020210066. Recently, an astatine-labeled prostate-specific membrane antigen (PSMA) ligand ([211At]PSMA-5) has been developed for the targeted alpha therapy of patients with prostate cancer. This manual delineates its physicochemical characteristics to assist healthcare professionals in understanding the α-ray-emitting drug of [211At]PSMA-5 when administered to patients. The safety considerations regarding the handling and use of this drug in clinical trials are outlined, based on the proper usage manual of previous studies. The dose limits, as defined by the guidelines of the International Commission on Radiological Protection (ICRP) and the International Atomic Energy Agency (IAEA), are assessed for patients' caregivers and the general public. According to the calculations provided in this manual, clinical trials involving [211At]PSMA-5 can be safely conducted for these populations even if patients are released after its administration. Moreover, this manual provides comprehensive guidance on the handling of [211At]PSMA-5 for healthcare facilities, and compiles a list of precautionary measures to be distributed among patients and their caregivers. While this manual was created by a research team supported by Ministry of Health, Labour, and Welfare in Japan and approved by Japanese Society of Nuclear Medicine, its applicability extends to healthcare providers in other countries. In this article, a pseudo-stress for local residual and an algebraically derived consistent tangent are applied to elastic–plastic boundary value problems of shells. The authors define a coupled problem of the weak form of the equilibrium equation for the overall structure and the constrained equations for stress state, which include the yield condition and the plane stress condition, at every material point. Since the pseudo-stress for the residuals of yield criterion and plane stress state is incorporated into the linearized weak form of the equilibrium equation, the proposed scheme does not have any local iterative calculations and enables us to decrease the residuals in the coupled boundary value problems simultaneously. In the reverse transition in pipe flow, turbulent flow changes to less disturbed laminar flow. The entropy of the flow appears to decrease. This study examined the reverse transition experimentally and theoretically using entropy change and momentum balance models, not in terms of disturbance in the flow. The reverse transition was accomplished by decreasing the Reynolds number. The transitions approximately correlated with local Reynolds numbers. The initial Reynolds number of the transition became larger, and the pressure at low Reynolds numbers was greater than in ordinary pipe flow. These behaviours were caused by turbulent flow in the pipe undergoing a reverse transition. In this article, an introduction of pseudo-stress for local residual and an algebraic derivation of consistent tangent are presented. The authors define a coupled problem of the equilibrium equation for the overall structure and the constrained equations for stress state at every material point, and the pseudo-stress and the derived consistent tangent can be implemented easily to finite element analysis. In the proposed block Newton method, the internal variables are also updated algebraically without any local iterative calculations. In genetic counseling, information must be provided in ways that the client and general public can understand to ensure that decisions are made autonomously. To realize this, we must assess the extent of knowledge held by the general public regarding genetics. To identify the client's original knowledge before genetic counseling, we explored the fundamental knowledge related to genetic counseling that is taught in Japanese compulsory education. A qualitative study was conducted. We selected 50 textbooks for compulsory education (Japanese, social studies, science, health and physical education, technology and home economics, morality, and life) that had been used in more than half of the districts in Japan. The text data were analyzed using qualitative content analysis, and quantitative data were analyzed for methodological triangulation. Codes, subcategories, and categories were generated from the contexts that met the following criteria: the contents included in the official textbook for clinical geneticists, contents derived from such descriptions that were related to genetic counseling, and contents clearly related to genetics. Among the 50 textbooks, 33 textbooks contained fundamental knowledge regarding genetic counseling. A qualitative content analysis identified four major categories: (1) basics of genetics, (2) understanding and control of diseases, (3) efforts and barriers to the realization of a harmonious society, and (4) technology and humans. We found that fundamental knowledge related to genetic counseling is directly or indirectly taught in compulsory education. Background: This study evaluated the physical performance of a positron emission tomography (PET) system dedicated to the head and breast according to the National Electrical Manufacturers Association (NEMA) NU2-2012 standard. Methods: The spatial resolution, sensitivity, scatter fraction, count rate characteristics, corrections for count losses and randoms, and image quality of the system were determined. All measurements were performed according to the NEMA NU2-2012 acquisition protocols, but image quality was assessed using a brain-sized phantom. Furthermore, scans of the three-dimensional (3D) Hoffmann brain phantom and mini-Derenzo phantom were acquired to allow visual evaluation of the imaging performance for small structures. Results: The tangential, radial, and axial full width at half maximum (FWHM) at a 10-mm offset in half the axial field of view were measured as 2.3, 2.5, and 2.9 mm, respectively. The average system sensitivity at the center of the field of view and at a 10-cm radial offset was 7.18 and 8.65 cps/kBq, respectively. The peak noise-equivalent counting rate was 35.2 kcps at 4.8 kBq/ml. The corresponding scatter fraction at the peak noise-equivalent counting rate was 46.8%. The peak true rate and scatter fraction at 8.6 kBq/ml were 127.8 kcps and 54.3%, respectively. The percent contrast value for a 10-mm sphere was approximately 50%. On the 3D Hoffman brain phantom image, the structures of the thin layers composing the phantom were visualized on the sagittal and coronal images. On the mini-Derenzo phantom, each of the 1.6-mm rods was clearly visualized. Objectives: This study evaluates the phantom attenuation correction (PAC) method as an alternative to maximum-likelihood attenuation correction factor (ML-ACF) correction in time-of-flight (TOF) brain positron emission tomography (PET) studies. Methods: In the PAC algorithm, a template emission image λ Ref and a template attenuation coefficient image μ Ref are prepared as a data set based on phantom geometry. Position-aligned attenuation coefficient image μ Acq is derived by aligning μ Ref using parameters that match the template emission image λ Ref to measured emission image λ Acq . Then, attenuation coefficient image μ Acq combined with a headrest image is used for scatter and attenuation correction in the image reconstruction. To evaluate the PAC algorithm as an alternative to ML-ACF, Hoffman 3D brain and cylindrical phantoms were measured to obtain the image quality indexes of contrast and uniformity. These phantoms were also wrapped with a radioactive sheet to obtain attenuation coefficient images using ML-ACF. Emission images were reconstructed with attenuation correction by PAC and ML-ACF, and the results were compared using contrast and uniformity as well as visual assessment. CT attenuation correction (CT-AC) was also applied as a reference. Results: The contrast obtained by ML-ACF was slightly overestimated due to its unique experimental condition for applying ML-ACF in Hoffman 3D brain phantom but the uniformity was almost equivalent among ML-ACF, CT-AC, and PAC. PAC showed reasonable result without overestimation compared to ML-ACF and CT-AC. Conclusions: PAC is an attenuation correction method that can ensure the performance in phantom test, and is considered to be a reasonable alternative to clinically used ML-ACF-based attenuation correction. In this manuscript, we present the guideline for use of meta-[211At] astatobenzylguanidine ([211At] MABG), a newly introduced alpha emitting radiopharmaceutical to the up-coming World's first clinical trial for targeted alpha therapy (TAT) at Fukushima Medical University in Japan, focusing on radiation safety issues in Japan. This guideline was prepared based on a study supported by the Ministry of Health, Labour, and Welfare, and approved by the Japanese Society of Nuclear Medicine on Oct. 5th, 2021. The study showed that patients receiving [211At] MABG do not need to be admitted to a radiotherapy room and that TAT using [211At] MABG is possible on an outpatient basis. The radiation exposure from the patient is within the safety standards of the ICRP and IAEA recommendations for the general public and caregivers or patient's family members. In this guideline, the following contents are also included: precautions for patients and their families, safety management associated with the use of [211At] MABG, education and training, and disposal of medical radioactive contaminants. TAT using [211At] MABG in Japan should be carried out according to this guideline. Although this guideline is based on the medical environment and laws and regulations in Japan, the issues for radiation protection and evaluation methodology presented in this guideline are useful and internationally acceptable as well. Aim: The aim of this study was to evaluate an image reconstruction algorithm, including a new maximum-likelihood attenuation correction factor (ML-ACF) for time of flight (TOF) brain positron emission tomography (PET). Methods: The implemented algorithm combines an ML-ACF method that simultaneously estimates both the emission image and attenuation sinogram from TOF emission data, and a scaling method based on anatomical features. To evaluate the algorithm's quantitative accuracy, three-dimensional brain phantom images were acquired and soft-tissue attenuation coefficients and emission values were analyzed. Results: The heterogeneous distributions of attenuation coefficients in soft tissue, skull, and nasal cavity were sufficiently visualized. The attenuation coefficient of soft tissue remained within 5% of theoretical value. Attenuation-corrected emission showed no lateral differences, and significant differences among soft tissue were within the error range. Objective: This study aimed at investigating the correlation between recurrent visual hallucinations (VHs) and regional cerebral blood flow (rCBF) in patients with dementia with Lewy bodies (DLB). Methods: In 147 DLB patients, the correlation between noise pareidolia scores and rCBF in brain perfusion single photon emission computed tomography (SPECT) was evaluated. The 147 subjects comprised 52 probable and 95 possible DLB patients, of whom 107 did not have visual hallucinations and 40 had visual hallucinations. Brain perfusion SPECT was then performed, and memory impairment was assessed using the Mini-Mental State Examination (MMSE), while the optical illusion "pareidolia" (the tendency to see a specific image in a random visual pattern) was evaluated using noise pareidolia test. The correlations between rCBF and MMSE or noise pareidolia scores were then analyzed. Results: The rCBF and MMSE scores were positively correlated, and rCBF was correlated with MMSE scores in a region that was consistent with a previously reported memory-related site. There was no correlation between noise pareidolia scores and occipital CBF, but there were weak correlations between noise pareidolia scores and rCBF in the bilateral frontal lobes (Brodmann area [BA]8 and BA9), left cingulate cortex (BA31), and left angular and supramarginal gyri (BA39 and BA40) in DLB patients. Conclusion: Weak correlation was found between noise pareidolia scores and rCBF in several sites (BA8, BA9, BA31, BA39 and BA40) other than in occipital lobe. These findings suggest that DLB hallucinations may be manifested by more complex brain network disorders, rather than by primary visual cortex disorders alone. [ABSTRACT FROM AUTHOR]

Pv11 is an insect cell line established from the midge Polypedilum vanderplanki, whose larval form exhibits an extreme desiccation tolerance known as anhydrobiosis. Pv11 itself is also capable of anhydrobiosis, which is induced by trehalose treatment. Here we report the successful construction of a genome editing system for Pv11 cells and its application to the identification of signaling pathways involved in anhydrobiosis. Using the Cas9-mediated gene knock-in system, we established Pv11 cells that stably expressed GCaMP3 to monitor intracellular Ca2+ mobilization. Intriguingly, trehalose treatment evoked a transient increase in cytosolic Ca2+ concentration, and further experiments revealed that the calmodulin–calcineurin–NFAT pathway contributes to tolerance of trehalose treatment as well as desiccation tolerance, while the calmodulin–calmodulin kinase–CREB pathway conferred only desiccation tolerance on Pv11 cells. Thus, our results show a critical contribution of the trehalose-induced Ca2+ surge to anhydrobiosis and demonstrate temporally different roles for each signaling pathway. [ABSTRACT FROM AUTHOR]

Background: Appendicitis is the most common cause of acute abdomen. Although emergency surgery used to be the standard treatment for both simple and complex appendicitis, there are now options for interval surgery, laparoscopic surgery, and even non-surgical treatment. In this study, we aimed to establish better treatment strategies for pediatric acute appendicitis and to find out whether minimally invasive treatment is superior to the traditional open approach. We retrospectively reviewed the cases of acute appendicitis treated in our hospital during the period from 2013 to 2018. The patients who underwent appendectomy were divided into four groups. Group 1 underwent early appendectomy with open approach, group2 underwent interval appendectomy with open approach, group 3 underwent early appendectomy with laparoscopic approach, and group 4 underwent interval appendectomy with laparoscopic approach. In addition to the above groups, the non-surgical treatment group was also studied. Clinical presentation, laboratory findings, imaging results, operative time, morbidity, and length of hospital stay were reviewed. Results: One hundred six children's records were reviewed. Thirty-five of them were selected for non-surgery as they were cases with no fecal stone and first onset appendicitis, and 15 of these 35 patients (42.9%) relapsed after antibiotic treatment. As for the appendix diameter, the relapse group was significantly larger than the non-relapse group (p=0.007). In cases of surgery, group 4 had significantly less intraoperative blood loss than group 1 (p<0.001). Group 4 had significantly fewer postoperative complications than groups 1 and 2 [group 4 vs. group 1 (p=0.009), group 4 vs. group 2 (p=0.034)]. The postoperative hospital stay in groups 2 and 4 were significantly shorter than group 1 [group 1 vs. group 2 (p=0.015), group 1 vs. group 4 (p<0.001)]. On the contrary, group 1 had significantly shorter total hospital stay than groups 2 and 4 [group 1 vs. group 2 (p=0.029), group 1 vs. group 4 (p<0.001)]. Conclusion: Interval laparoscopic appendectomy and non-surgical treatment were safe and effective in children. From the viewpoint of avoiding unnecessary emergency surgery and prolonged hospitalization, we believe that interval laparoscopic appendectomy or non-surgical treatment should be performed after identifying patients who do not require surgery, paying attention to the risk factors for relapse. Liver cancer is the fourth leading cause of cancer-related death. Hepatocellular carcinoma (HCC) is a primary liver cancer that results from chronic hepatitis caused by multiple predisposing factors such as viral infection, alcohol consumption, and non-alcoholic fatty liver disease. Accumulating studies have indicated that dysfunction of the gut epithelial barrier and hepatic translocation of gut microbes may be implicated in the pathogenesis of HCC. However, the translocated bacteria in HCC patients remains unclear. Here, we characterised tumour-associated microbiota in patients with liver cancer and focused on HCC. We observed that the number of amplicon sequence variants in tumour-associated microbiota was significantly higher compared with that in non-tumour regions of the liver. The tumour-associated microbiota consisted of Bacteroidetes, Firmicutes, and Proteobacteria as the dominant phyla. We identified an unclassified genus that belonged to the Bacteroides, Romboutsia, uncultured bacterium of Lachnospiraceae as a signature taxon for primary liver cancer. Additionally, we identified Ruminococcus gnavus as a signature taxon for HCC patients infected with hepatitis B and/or hepatitis C viruses. Purpose: It is usually easy to judge whether amyloid PET images should be interpreted as positive or negative for amyloid deposits by visual inspection or quantitative measurement standard uptake value ratio (SUVR), but the findings are equivocal in some cases. As conventional mean cortical SUVR (mcSUVR) measures accumulation in both gray matter (GM) and white matter, it may mis-estimate amyloid deposits. The purpose of the study was to develop a regional GM-dedicated SUVR measuring (GMSUVR) system for amyloid PET images with 3D-MRI, and evaluate its utility for detecting amyloid deposits in equivocal cases. Methods: Of 126 subjects who underwent amyloid PET with 11C-PiB and 3D-MRI, the area of amyloid-positive regions and the critical regional GMSUVR thresholds were first determined in 15 amyloid-positive and 15 amyloid-negative patients, using the automatic volumetric measurement of segmented brain images system. We then tested 36 amyloid-negative, 60 amyloid-positive, and 13 equivocal subjects with this GMSUVR system and with conventional mcSUVR. Results: Sensitivity, specificity, accuracy, positive predictive value (PPV) and negative predictive value (NPV) were 100%, 92%, 97%, 95%, and 100% for the GMSUVR system; and 97%, 86%, 93%, 92% and 94%, respectively, for mcSUVR. In 24 cases in which the findings were equivocal or discordant, the sensitivity, specificity, accuracy, PPV, and NPV were all 100% for the GMSUVR system; and were 90%, 33%, 83%, 90%, and 33%, respectively, for mcSUVR. Conclusion: The regional GMSUVR measurement method was well able to discriminate between amyloid-positive and -negative subjects, even in cases where amyloid deposition was equivocal. Objective: An artificial intelligence (AI)-based algorithm typically requires a considerable amount of training data; however, few training images are available for dementia with Lewy bodies and frontotemporal lobar degeneration. Therefore, this study aims to present the potential of cycle-consistent generative adversarial networks (CycleGAN) to obtain enough number of training images for AI-based computer-aided diagnosis (CAD) algorithms for diagnosing dementia. Methods: We trained CycleGAN using 43 amyloid-negative and 45 positive images in slice-by-slice. Results: The CycleGAN can be used to synthesize reasonable amyloid-positive images, and the continuity of slices was preserved. Objective: This study aims to develop an algorithm named AutoRef to delineate a reference region for quantitative PET amyloid imaging.Methods: AutoRef sets the reference region automatically using a distinguishing feature in the kinetics of reference region. This is reflected in the shapes of the tissue time activity curve. A statistical shape recognition algorithm of the gaussian mixture model is applied with considering spatial and temporal information on a reference region. We evaluate the BPND with manually set reference region and AutoRef using 86 cases (43 positive cases, 10 equivocal cases, and 33 negative cases) of dynamically scanned 11C-Pittsburgh Compound-B.Results: From the Bland-Altman plot, the difference between two BPND is 0.099 ± 0.21 as standard deviation, and no significant systematic error is observed between the BPND with AutoRef and with manual definition of a reference region. Although a proportional error is detected, it is smaller than the 95% limits of agreement. Therefore, the proportional error is negligibly small.Conclusions: AutoRef presents the same performance as the manual definition of the reference region. Further, since AutoRef is more algorithmic than the ordinary manual definition of the reference region, there are few operator-oriented uncertainties in AutoRef. This paper presents a numerical procedure to couple shell to solid elements by using the Nitsche's method. The continuity of displacements can be satisfied approximately with the penalty method, which is effective in setting the penalty parameter to a sufficiently large value. When the continuity of only displacements on the interface is applied between shell and solid elements, an unreasonable deformation may be observed near the interface. In this work, the continuity of the stress vector on the interface is considered by employing the Nitsche's method, and hence a reasonable deformation can be obtained on the interface. The authors propose two types of shell elements coupled with solid elements in this paper. One of them is the conventional MITC4 shell element, which is one of the most popular elements in engineering applications. This approach shows the capability of discretizing the domain of the structure with the different types of elements. The other is the shell element with additional degrees of freedom to represent thickness-stretch developed by the authors. In this approach, the continuity of displacements including the deformation in the thickness direction on the interface can be considered. Several numerical examples are presented to examine the fundamental performance of the proposed procedure. This paper presents a quadrilateral shell element incorporating thickness-stretch, and demonstrates its performance in small and large deformation analyses for hyperelastic material and elastoplastic models. In terms of geometry, the proposed shell element is based on the formulation of the MITC4 shell element, with additional degrees of freedom to represent thickness-stretch. To consider the change in thickness, we introduce a displacement variation to the MITC4 shell element, in the thickness direction. After the thickness direction is expressed in terms of the director vectors that are defined at each midsurface node, additional nodes are placed along the thickness direction from the bottom surface to the top surface. The thickness-stretch is described by the movement of these additional nodes. The additional degrees of freedom are used to compute the transverse normal strain without assuming the plane stress condition. Hence, the three dimensional constitutive equation can be employed in the proposed formulation without any modification. By virtue of not imposing the plane stress condition, the surface traction is evaluated at the surface where the traction is applied, whereas it is assessed at the midsurface for conventional shell elements. Several numerical examples are presented to examine the fundamental performance of the proposed shell element. In particular, the proposed approach is capable of evaluating the change in thickness and the stress distribution when the effect of the surface traction is included. Objective: To improve the reliability and convenience of the calibration procedure of positron emission tomography (PET) scanners, we have been developing a novel calibration path based on traceable point-like sources. When using Na sources, special care should be taken to avoid the effects of 1.275-MeV γ rays accompanying β decays. The purpose of this study is to validate this new calibration scheme with traceable point-like Na sources on various types of PET scanners. Method: Traceable point-like Na sources with a spherical absorber design that assures uniform angular distribution of the emitted annihilation photons were used. The tested PET scanners included a clinical whole-body PET scanner, four types of clinical PET/CT scanners from different manufacturers, and a small-animal PET scanner. The region of interest (ROI) diameter dependence of ROI values was represented with a fitting function, which was assumed to consist of a recovery part due to spatial resolution and a quadratic background part originating from the scattered γ rays. Results: The observed ROI radius dependence was well represented with the assumed fitting function ( R > 0.994). The calibration factors determined using the point-like sources were consistent with those by the standard cross-calibration method within an uncertainty of ±4 %, which was reasonable considering the uncertainty in the standard cross-calibration method. Purpose: Thus far, cylindrical phantoms with F or Ge/Ga have been used in the standard techniques for determining the cross-calibration factors (CCFs) of PET scanners. This paper describes a new practical method that uses a point-like Na radioactive source for determining CCFs. Methods: A point-like Na radioactive source (about 700 kBq) was equipped with a spherical aluminium absorber capsule to ensure the symmetry of the emitted photons. During measurements, the source was moved in the axial direction to cover the whole axial field of view (FOV) of a clinical PET scanner, SET-2400 W. The region-of-interest (ROI) values obtained in reconstructed images without scatter and attenuation corrections were used to calculate the CCFs of the PET scanner. Results: The CCFs obtained by the proposed method agreed with those obtained by the standard cross-calibration (CC) method within a precision of 1.5% (σ). The temporal variations in the CCFs by the standard CC method were reproduced by the proposed method with a precision better than 1%. Conclusion: The CC method with a moving Na point-like radioactive source is practically useful for determining the CCFs of PET scanners and monitoring their variations. [ABSTRACT FROM AUTHOR]

Radioactive sources for evaluating sensitivity and uncertainty in the radioactivity measurements performed using PET scanners must be equipped with absorber materials that ensure the annihilation of positrons. Attenuation and scattering owing to the absorber materials produce uncertainty in the performance evaluation. The aim of this study is to propose a point-like radioactive source with multiple absorber capsules, for which evaluation can be independent of scatter and attenuation owing to the source absorbers. The point-like source consists of a small spherical radioactive part and a set of successively sized cylindrical aluminum absorber capsules. Data were collected for different total absorber thicknesses. By an extrapolation technique, the effects of the source absorbers were eliminated. Sensitivity and uncertainty in the radioactivity measurements of PET scanners were evaluated with this technique. Sensitivity and uncertainty of radioactivity measurement to the point-like radioactive source were evaluated successfully with this method. The proposed point-like radioactive source is useful for evaluating performance characteristics of PET scanners in a way that is independent of the effects of the source absorbers. The article reports on the X-ray diffraction, morphology, and thermoelectric properties of magnesium silicide bulk sample prepared by using a compact body of sodium silicon powder and magnesium melt. Details of the preparation of sodium silicon are provided. The thickness of the wall is cited. The sample of magnesium silicide exhibited p-type thermoelectric and semiconducting behavior, which was attributed to sodium doping of less than 0.5 wt% in magnesium silicide.

By a search for novel human imprinted genes in the vicinity of the imprinted gene MEST, at chromosome 7q32, we identified the carboxypeptidase A4 gene (CPA4) in a gene cluster of the carboxypeptidase family, 200 kb centromeric to MEST. Because CPA4 was originally identified as a protein induced in a prostate cancer cell line (PC-3) by histone deacetylase inhibitors, and was located at the putative prostate cancer-aggressiveness locus at 7q32, we investigated its imprinting status in fetal tissues and in adult benign hypertrophic prostate (BPH). RT-PCR using four intragenic polymorphisms as markers showed that CPA4 was expressed preferentially from the maternal allele in the fetal heart, lung, liver, intestine, kidney, adrenal gland, and spleen, but not in the fetal brain. It was also preferentially expressed in the BPH. These findings support that CPA4 is imprinted and may become a strong candidate gene for prostate cancer-aggressiveness. As a Silver-Russell syndrome (SRS) locus has been proposed to be located to a region near MEST and to be involved in imprinting, CPA4 would have been a candidate gene for SRS. Cleft of the soft palate (CSP) and the hard palate are subtypes of cleft palate. Patients with either condition often have difficulty with speech and swallowing. Nonsyndromic, cleft palate isolated has been reported to be associated with several genes, but to our knowledge, there have been no detailed genetic investigations of CSP. We performed a genome-wide linkage analysis using a single-nucleotide polymorphism-based microarray platform and successively using microsatellite markers in a family in which six members, across three successive generations, had CSP. A maximum LOD score of 2.408 was obtained at 2p24.2–24.1 and 2p21–p12, assuming autosomal dominant inheritance. Our results suggest that either of these regions is responsible for this type of CSP. During embryogenesis, cells repeatedly divide and dynamically change their positions in three-dimensional (3D) space. A robust and accurate algorithm to acquire the 3D positions of the cells would help to reveal the mechanisms of embryogenesis. To acquire quantitative criteria of embryogenesis from time-series 3D microscopic images, image processing algorithms such as segmentation have been applied. Because the cells in embryos are considerably crowded, an algorithm to segment individual cells in detail and accurately is needed. To quantify the nuclear region of every cell from a time-series 3D fluorescence microscopic image of living cells, we developed QCANet, a convolutional neural network-based segmentation algorithm for 3D fluorescence bioimages. We demonstrated that QCANet outperformed 3D Mask R-CNN, which is currently considered as the best algorithm of instance segmentation. We showed that QCANet can be applied not only to developing mouse embryos but also to developing embryos of two other model species. Using QCANet, we were able to extract several quantitative criteria of embryogenesis from 11 early mouse embryos. We showed that the extracted criteria could be used to evaluate the differences between individual embryos. The larvae of the African midge, Polypedilum vanderplanki, can enter an ametabolic state called anhydrobiosis to overcome fatal desiccation stress. The Pv11 cell line, derived from P. vanderplanki embryo, shows desiccation tolerance when treated with trehalose before desiccation and resumes proliferation after rehydration. However, the molecular mechanisms of this desiccation tolerance remain unknown. Here, we performed high-throughput CAGE-seq of mRNA and a differentially expressed gene analysis in trehalose-treated, desiccated, and rehydrated Pv11 cells, followed by gene ontology analysis of the identified differentially expressed genes. We detected differentially expressed genes after trehalose treatment involved in various stress responses, detoxification of harmful chemicals, and regulation of oxidoreduction that were upregulated. In the desiccation phase, L-isoaspartyl methyltransferase and heat shock proteins were upregulated and ribosomal proteins were downregulated. Analysis of differentially expressed genes during rehydration supported the notion that homologous recombination, nucleotide excision repair, and non-homologous recombination were involved in the recovery process. This prospective observational study compare urine nephrin:creatinine ratio (NCR, ng/mg) with serum soluble fms-like tyrosine kinase-1:placental growth factor ratio (FPR, pg/pg) for preeclampsia (PE) prediction among unselected asymptomatic pregnant women in 2nd trimester. NCR and FPR were determined in 254 paired urine/blood samples collected simultaneously from 254 women at median gestational week (GW) 24 (range, 22-27) without hypertension or significant proteinuria in pregnancy (SPIP). Fifteen (5.9%) developed SPIP and hypertension at GW 34.0 (26.0-38.6) and 35.3 (27.6-38.6), respectively, and were diagnosed with PE at GW 35.7 (27.6-38.6). The 90th percentile level determined in 239 women normotensive throughout pregnancy gave NCR (139) sensitivity and positive predictive values (PPV) of 60% (9/15) and 27% (9/33), while those for serum FPR (4.85) were 40% (6/15) and 20% (6/30), respectively. Relative risks (95%CI) of later PE were 10.0 (3.82-26.4; 27% [9/33] vs. 2.7% [6/221]) and 4.98 (1.91-13.0; 20% [6/30] vs. 4.0% [9/224]) for NCR-positive and FPR-positive women, respectively. Cut-offs suggested by ROC gave NCR (86.6) sensitivity and PPV of 87% (13/15) and 17% (13/79), and FPR (8.8) values of 40% (6/15) and 40% (6/15), respectively. Epigenetic modifications are thought to serve as a memory of exposure to in utero environments. However, few human studies have investigated the associations between maternal nutritional conditions during pregnancy and epigenetic alterations in offspring. In this study, we report genome-wide methylation profiles for 33 postpartum placentas from pregnancies of normal and foetal growth restriction with various extents of maternal gestational weight gain. Epigenetic alterations accumulate in the placenta under adverse in utero environments, as shown by application of Smirnov-Grubbs' outlier test. Moreover, hypermethylation occurs frequently at the promoter regions of transcriptional regulator genes, including polycomb targets and zinc-finger genes, as shown by annotations of the genomic and functional features of loci with altered DNA methylation. Aberrant epigenetic modifications at such developmental regulator loci, if occurring in foetuses as well, will elevate the risk of developing various diseases, including metabolic and mental disorders, later in life.
