Abstract

Solid-state fermentation has a special advantage of preventing the foaming problem that obstructs submerged fermentation processes for rhamnolipid production. In the present work, a 50:50 mixture of sugarcane bagasse and sunflower seed meal was selected as the optimum substrate for rhamnolipid production using a Pseudomonas aeruginosa mutant 15GR and an impregnating solution including 5% v/v glycerol. Using Box–Behnken design, the optimum fermentation conditions were found to be an inoculum size 1% v/v, temperature 30 °C and unlike other studies, pH 8. These optimized conditions yielded a 67% enhancement of rhamnolipid levels reaching 46.85 g rhamnolipids per liter of impregnating solution, after 10 days, which was about 5.5 folds higher than that obtained by submerged liquid fermentation. Although maximum rhamnolipids concentration was obtained after 10 days of incubation, rhamnolipids concentration already reached high levels (41.87 g/l) after only 6 days. This rhamnolipid level was obtained in a shorter time and using lower carbon source concentrations than most studies reported so far. The findings obtained indicate an enormous potential for employing solid-state fermentation for rhamnolipid production by the studied isolate.

Abstract

Purpose Wheat straw is an agricultural waste which can be used as a cost effective animal feed. However, high hemicellulose and phytic acid content in wheat straw prevents it as a primary feed choice. Utilization of wheat straw in solid-state fermentation may result in wheat straw valorization and enzyme production. In this study, phytase production in solid-state fermentation of wheat straw using Aspergillus ficuum and valorization of wheat straw were evaluated. Methods A two-step experimental design procedure was employed for screening and optimization of influencing factors on phytase production. Effects of different nutritional and environmental factors were investigated by one factor at the time method (OFATM). To reach higher amounts of phytase, response surface methodology (RSM) was employed to optimize phytase production as a function of three of the most effective factors. Results Optimization of the significant parameters resulted in an increase in the phytase activity from 0.74 ± 0.12 to a maximum of 16.46 ± 0.56 Units per gram dry substrate (U gds−1). The high degree of the fungal phytase activity on wheat straw resulted in the decrease in phytic acid content by 57.4%, as compared to the untreated sample. Scanning electron microscopy (SEM) and FTIR structural analysis showed intensive fungal growth on wheat straw, and partial removal of hemicelluloses, lignin and phytic acid. Conclusion The study demonstrated the feasibility of wheat straw utilization in solid-state fermentation using Aspergillus ficuum toward the production of phytase and valorization of wheat straw as an animal feed.

Abstract

The process parameters governing the production of fibrinolytic enzyme in solid state fermentation employing Bacillus cereus IND5 and using cuttle fish waste and cow dung substrate were optimized. The pH value of the medium, moisture content, sucrose, casein and magnesium sulfate were considered for two-level full factorial design and pH, casein and magnesium sulfate were identified as the important factors for fibrinolytic enzyme production. Central composite design was applied to investigate the interactive effect among variables (pH, casein and magnesium sulfate) and response surface plots were created to find the pinnacle of process response. The optimized levels of factors were pH 7.8, 1.1% casein and 0.1% magnesium sulfate. Enzyme production was increased 2.5-fold after statistical approach. The enzyme was purified up to a specific activity of 364.5 U/g proteins and its molecular weight was 47 kDa. It was stable at pH 8.0 and was highly active at 50 °C. The mixture of cuttle fish waste and cow dung could find great application as solid substrate for the production of fibrinolytic enzyme.
