Textile effluent discharge can negatively impact the environment and living organisms due to its potential toxicity, higher percentages of total organic carbon (TOC) contents, and so on. The study investigates the extraordinary performance of the electrocoagulation process (ECP) combined with powdered activated carbon (PAC) as a highly effective and environmental friendly method of treating textile effluents. This scientific work mainly includes the focus on removing toxic components in textile effluents, such as high concentrations of colour and TOC using synthesized PAC derived from coconut shells coupled with the ECP (ECP-PAC). Initially, PAC was characterized by using XRD, Raman, BET, FTIR, and TGA studies. Subsequently, the pilot-scale ECP-PAC batch reactor was constructed with iron (Fe) as an anode and copper (Cu) as a cathode. The pilot-scale ECP-PAC batch reactor has achieved higher treatment efficiency in a shorter reaction time with low energy consumption compared to a stand-alone ECP. Further, the optimum conditions for effective ECP-PAC have been optimized, such as pH 7.5, applied current density (0-50 mA/cm 2 ), reaction time (0-30 min), electrode combinations (Fe-Cu) with electrode distances of 5 cm apart, and an optimum dose of 5 g/L of PAC. Specifically, 98% of the colour and 96% of the TOC contents present in the industrial textile effluent were treated in 15 and 30 min, respectively. In quantitative perspectives, the developed batch reactor has sharply decreased TOC (324.1 mg/L), IC (1410 mg/L) and TC (1019 mg/L) to 13.55 mg/L (96%), 31.49 mg/L (97%), and 48.05 mg/L (95%), respectively, in 30 min demonstrating its sensitivity and selectivity with the utmost care. Moreover, the physicochemical properties of the treated water were convincingly assessed. That is, it remains suitable for the seed germination of mung bean and chlorophyll content study. Thus, the developed methodology could effectively reduce freshwater consumption in the agricultural sector, increase freshwater availability in water-scarce regions, and facilitate the increase of the recharging capacity of groundwater tables., (© 2024. The materials for water treatment have been evolving in multitude of dimensions, indicating the importance of water reuse and increasing level of water pollution around the globe. Among the various materials that are utilized in wastewater treatment, the material that has attracted the research community for the past decades is the metal organic framework (MOF). In this work one of the water stable and microporous MOF, UiO-66, and its aminated version has been employed to adsorb an anionic azo dye, direct blue-6 (DB-6), from the aqueous matrix. Performance of both the MOFs was compared to know the efficiency under varying solution conditions. The optimized parameters for DB-6 adsorption by UiO-66 was performed using response surface methodology. This numerical optimization was further extended with canonical and ridge analysis. Under optimal conditions, the materials were exhibiting a good adsorption capacity of 754.4 mg/g. The materials were analyzed in terms of morphology, crystallinity, thermal stability, and surface area using instruments like X-ray diffraction, electron microscopy, thermogravimetric analysis, and BET surface area analysis. The mechanism of interaction between UiO-66 and DB-6 molecule was elucidated with the help of XPS analysis which helps to know the main interacting group of UiO-66. Purpose: Spermatogonial stem cells (SSCs) are the source for the mature male gamete. SSC technology in humans is mainly focusing on preserving fertility in cancer patients. Whereas in livestock, it is used for mining the factors associated with male fertility. The review discusses the present status of SSC biology, methodologies developed for in vitro culture, and challenges ahead in establishing SSC technology for the propagation of superior germplasm with special reference to livestock., Method: Published literatures from PubMed and Google Scholar on topics of SSCs isolation, purification, characterization, short and long-term culture of SSCs, stemness maintenance, epigenetic modifications of SSCs, growth factors, and SSC cryopreservation and transplantation were used for the study., Result: The fine-tuning of SSC isolation and culture conditions with special reference to feeder cells, growth factors, and additives need to be refined for livestock. An insight into the molecular mechanisms involved in maintaining stemness and proliferation of SSCs could facilitate the dissemination of superior germplasm through transplantation and transgenesis. The epigenetic influence on the composition and expression of the biomolecules during in vitro differentiation of cultured cells is essential for sustaining fertility. The development of surrogate males through gene-editing will be historic achievement for the foothold of the SSCs technology., Conclusion: Detailed studies on the species-specific factors regulating the stemness and differentiation of the SSCs are required for the development of a long-term culture system and in vitro spermatogenesis in livestock. Minerals are required in small amounts but play significant roles in many physiological functions related with growth, reproduction, and health of goats such as biochemical, molecular systems, and optimized enzymatic activities due to their roles as co-factors to metalloenzymes. Among them, zinc (Zn) and copper (Cu) are leading essential elements in goat nutrition, because of their role across several biological functions. The proportion of these minerals availability and absorption from the ingested feed is usually less, because of their complexities with un-degradable parts of feed resources. Hence, their exogenous supplementation is required for normal animal functions. On this background, this review presents findings associated with supplementation of these minerals in organic form as a way for improving the fertility of male goats with special focus on physico-chemical-kinetics of the semen for improving the application of reproductive technologies. This review emphasizes the organic sources of these minerals to replace the inorganic sources, based on their significance in improving semen qualities, antioxidant protection, and mediation of molecular activities. This review also discusses salient routes of Zn and Cu absorption and identifies the need for molecular exploration for positive outcomes with supplementation of these minerals as an area of the future goat nutrition-reproduction improvement strategy., (© 2021. Photoionization (PI) and photoinduced electron transfer (PET) dynamics of coumarin 450 (C450) in micelles were investigated in the time domains of micro to femtoseconds using steady-state and time-resolved absorption and fluorescence spectroscopy. The PI of C450 occurs inside the micelles leads to the formation of C450 cation radical (CR) and hydrated electron, which is characterized by the respective transient absorption. The PI of C450 is monophotonic in nature and the yield is dependent on the charge of the micelles. The observation of amine CR in the transient absorption confirms the PET from amine to the excited state of C450 in micelles, which results in the quenching of both fluorescence intensity and lifetime. The decrease in femtosecond fluorescent decay of C450 and the absence of transient C450 radical anion in the presence of amine implies that the concerted ultrafast PET promoted PI and PET to the C450 CR-electron pair. The decrease in the time constant for the formation of relaxed state in the presence of amines is due to the ultrafast PET to the C450 CR-electron pair, which prevents the formation of a relaxed state through recombination at a longer time scale. In the present investigation, the recombination dynamics of the CR-electron pair is justified as one of the origins of the slow solvation in micelles. The influence of amine concentration on the decay of C450 CR indicates ET reaction between C450 CR and amine, which is further confirmed by the bleach recovery of C450 ground state in the presence of amine., (© 2021. Purpose: The objective of the present study was to purify sheep spermatogonial stem cells (SSCs) from testicular isolate using combined enrichment methods and to study the effect of growth factors on SSC stemness during culture., Methods: The testicular cells from prepubertal male sheep were isolated, and SSCs were purified using Ficoll gradients (10 and 12%) followed by differential plating (laminin with BSA). SSCs were cultured with StemPro®-34 SFM, additives, and FBS for 7 days. The various doses (ng/ml) of growth factors, EGF at 10, 15, and 20, GDNF at 40, 70, and 100 and IGF-1 at 50, 100, and 150 were tested for the proliferation and stemness of SSCs in vitro. The stemness in cultured cells was assessed using SSC markers PLZF, ITGA6, and GFRα1., Results: Ficoll density gradient separation significantly (p < 0.05) increased the percentage of SSCs in 12% fraction (35.1 ± 3.8 vs 11.2 ± 3.7). Subsequently, purification using laminin with BSA plating further enriched SSCs to 61.7 ± 4.7%. GDNF at 40 ng/ml, EGF at 15 and 20 ng/ml and IGF1 at 100 and 150 ng/ml significantly (p < 0.05) improved proliferation and stemness of SSCs up to 7 days in culture. GDNF at 40 ng/ml outperformed other growth factors tested and could maintain the ovine SSCs proliferation and stemness for 36 days., Conclusions: The combined enrichment method employing density gradient centrifugation and laminin with BSA plating improves the purification efficiency of ovine SSCs. GDNF at 40 ng/ml is essential for optimal proliferation and sustenance of stemness of ovine SSCs in vitro.

The current study deals with the adsorption of hexavalent chromium using acid (H 3 PO 4 )-activated water caltrop (Trapa natans) shell (PWCS) using an up-flow packed bed column. Characteristics of breakthrough curve was obtained by investigating the effect of several operating parameters viz. inlet flow rate (2-6 mL/min), initial metal ion concentration (50-150 mg/L), and adsorbent's column bed height (1-3 cm). Elevated time of breakthrough curve was reported with elevated adsorbent bed height and vice versa with enhanced initial metal concentration and inlet flow rate of sorbate solution. Process design and breakthrough curves under varying conditions were predicted by applying column models like Thomas model, Adams-Bohart model, Yoon Nelson model, and bed depth service Time (BDST). Column behavior was better described by the BDST and Thomas model and simultaneously gave a good fit with the experimental data of breakthrough curves. The percentage removal for Cr(VI) from aqueous solution having pH 2 and temperature 303.15 K was observed to be 52.46%. The maximum adsorption capacity was found to be 87.31 mg/g according to the Thomas model. Conclusively, phosphoric acid-modified T. natans shell (PWCS) showed better sorption potential for of Cr(VI) species from simulated wastewater.

Current study deals with the comparative assessment for efficient adsorption of Cr(VI) from simulated wastewater using raw (NPP), phosphoric acid-activated (PPP) and sulphuric acid-activated (SPP) Pongamia pinnata shells. Physico-chemical alterations of the adsorbent were characterised by scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FT-IR), zeta-potential analysis, energy-dispersive X-ray spectroscopy (EDS) and total pore analysis using Brunauer-Emmett-Teller (BET). Parameters influencing the efficient biosorption of Cr(VI) species viz. initial pH of Cr(VI) solution, dosage of biosorbent, biosorbent-Cr(VI) contact period, initial concentration of Cr(VI) ions and reaction temperature were optimised. Various two-parameter and three-parameter isotherm models, kinetic models and thermodynamic studies were performed using equilibrium data. Langmuir adsorption capacity for NPP (raw biomass), PPP (phosphoric acid-activated biomass) and SPP (sulphuric acid-activated biomass) was found to be 96.2, 152 and 192 mg/g, respectively. All the biosorbents gave best fit for pseudo-second-order model. Thermodynamic studies suggest spontaneous and endothermic interaction with increased degree of randomness. Effect of co-existing cations and anions on Cr(VI) biosorption onto the biosorbents implied that minimal competition and the biosorption capacity of the biosorbents for Cr(VI) species remained unaffected. Regeneration studies suggest that activated biosorbents can be used up to three times with continuous desorption.

Environmental contaminants are gaining more attention in the livestock sector lately due to their harmful effects on productivity and fertility of livestock. Recent research indicates that many domestic ruminants are becoming subfertile/infertile due to confounding reasons associated with management. Contaminants like metals, metalloids, herbicides, pesticides, insecticides, chemicals, or natural contaminants are present everywhere in day to day life and are becoming a threat to the livestock. Studies on a broad-spectrum of animals suggest that high doses of acute or low doses of chronic exposure to the contaminants lead to disruption of multi-organs/systems including reproductive function. The lowered reproductive efficiency in animals is attributed to the endocrine disruptor activities of the environmental contaminants on the gonads, affecting gametogenesis and steroidogenesis. In vitro studies on testicular cells and the semen suggest that spermatozoa are more susceptible to damage by environmental contaminants. The quality of the semen happens to be a critical factor in the livestock industry. Contaminants affecting gametogenesis and steroidogenesis may lead to devastating consequences to the livestock reproduction, and thus the production. However, there is a lack of collective data on the effect of such environmental contaminants on the fertility of male domestic ruminants. This review discusses the studies related to the impact of environmental contaminants on male fertility in large (bull and buffalo) and small (sheep and goat) ruminants by focusing on the underlying molecular interactions between the contaminants and gonads.

A novel biosorbent Phanera vahlii fruit biomass (PVF) and its biochar and chemically modified forms were studied for the elimination of Cr(VI) from synthetic solutions. Biosorbents were characterized through BET, FTIR, FESEM, EDX, and TGA technique. The parameters influencing biosorption were optimized and found as pH 2.0, temperature 303 K, initial metal concentration 500 mg/L, and biosorbent dosage 0.5 g/L. The ideal contact time was 180 min for all biosorbents. Freundlich isotherm was found to have good correlation with investigational data, which indicated that biosorption takes place in multiple layer style. Langmuir adsorption isotherm yielded the highest biosorption capacity (Q o ) to be 159.1, 225.1, 244.1, and 278.5 mg/g for Phanera vahlii fruit biomass, Phanera vahlii biochar, Phanera vahlii phosphoric acid activated carbon, and Phanera vahlii zinc chloride activated carbon, respectively. Experimental data had good correlation with pseudo-second-order kinetic model fitted. Thermodynamic studies indicated the biosorption process to be spontaneous, stable, and endothermic. Thus, it was concluded that Phanera vahlii fruit biomass and the derived activated carbons are promising biosorbents for adsorption of chromium from aqueous solutions. Graphical abstract.

This study reveals the efficient bio-sorption of Cr(VI) species from simulated wastewater using raw (RSV), acid-activated (PSV) and chelated-activated (PSV-C) Sterculia villosa Roxb. shells. Batch optimization experiments were carried out for determining the optimal pH, dosage of bio-sorbent, initial Cr(VI) concentration and incubation temperature. Physico-chemical alterations in the bio-sorbents prior and following bio-sorption of Cr(VI) species were characterized. Equilibrated experimental data at different temperatures was evaluated as a function of time for understanding the isotherm, kinetics and thermodynamics of the bio-sorption process. Sorption rates for all the bio-sorbents (RSV, PSV and PSV-C) alternately fitted to the Langmuir isotherm model and Langmuir maximum adsorption capacity for RSV, PSV and PSV-C were found to be 57.78, 163.51 and 188.68 mg/g, respectively. Cr(VI) sorption by these bio-sorbents followed pseudo-second-order kinetics. Thermodynamic studies suggest endothermic interaction and increased degree of randomness between the bio-sorbents and Cr(VI) species. Regeneration of the bio-sorbents following sorption-desorption revealed that it can be reused and recycled for several times for Cr(VI) removal from wastewater. Graphical abstract.

The photoreceptor UV RESISTANCE LOCUS 8 (UVR8) activates photomorphogenic responses when plants are exposed to ultraviolet-B (UV-B) light. However, whereas the absorption spectrum of UVR8 peaks at 280 nm, action spectra for several photomorphogenic UV-B responses show maximal photon effectiveness at 290-300 nm. To investigate this apparent discrepancy we measured the effectiveness of UV wavelengths in initiating two responses in Arabidopsis: photoconversion of homodimeric UVR8 into the monomeric form, which is active in signaling, and accumulation of transcripts of the ELONGATED HYPOCOTYL 5 (HY5) transcription factor, which has a key role in UVR8-mediated responses. When purified UVR8 or Arabidopsis leaf extracts were exposed to UV light monomerisation was maximal at approximately 280 nm, which correlates with the UVR8 absorption spectrum. When intact plants were exposed to UV, monomerisation was most strongly initiated at approximately 290 nm, and this shift in maximal effectiveness could be explained by strong absorption or reflectance at 280 nm by leaf tissue. Notably, the action spectrum for accumulation of HY5 transcripts in the same leaf tissue samples used to assay UVR8 dimer/monomer status peaked at approximately 300 nm. Possible reasons for the difference in maximal photon effectiveness of UVR8 monomerisation and HY5 transcript accumulation in leaf tissue are discussed.

Coral reefs are degraded by the synergistic action of climate and anthropogenic stressors. Coral cover in the Palk Bay reef at the northern Indian Ocean largely declined in the past decade due to frequent bleaching events, tsunami and increased fishing activities. In this study, we carried out a comparative assessment to assess the differences in the recovery and resilience of three spatially distant reefs viz. Vedhalai, Mandapam and Pamban along Palk Bay affected by moderate, severe and low fishing pressure respectively. The assessment was based on the juvenile coral recruitment pattern and its survivability combined with availability of hard substratum, live coral cover and herbivore reef fish stock. The Vedhalai reef has the highest coral cover (14.6 ± 6.3%), and ≥90% of the live corals in Vedhalai and Mandapam were affected by turf algal overgrowth. The density of herbivore reef fish was low in Vedhalai and Mandapam reefs compared to the Pamban reef with relatively few grazing species. The juvenile coral diversity and density were high in the Pamban reef and low in Vedhalai and Mandapam reefs despite high hard substratum cover. In total, 22 species of juvenile corals of 10 genera were recorded in Palk Bay. Comparison of the species diversity of juvenile corals with adult ones suggested that the Pamban reef is connected with other distant reefs whereas Vedhalai and Mandapam reefs were self-seeded. There was no statistically significant difference in the survivability of juvenile corals between the study sites, and in total, ≥90% of the juvenile corals survived the high sedimentation stress triggered by the northeast monsoon and bleaching stress that occurred recurrently. Our results indicated that the human activities indirectly affected the juvenile coral recruitment by degrading the live coral cover and contributed to the spatial variation in the recovery and resilience of the Palk Bay reef. Low species diversity of the juvenile corals will increase the vulnerability of the Palk Bay reef to species-specific endemic threats.

The arsenic (As) is a multi system effector including reproduction. The present study examined the association of graded doses of As(V) on testicular microenvironment and sperm function in mice. Thirty-six adult male mice were randomly assigned to six groups (n = 6). Group A served as control without test chemical. The groups B, C, D, E, and F were administered graded doses of 10, 25, 50, 100, and 200 ppm As(V), respectively, through drinking water for 40 days. A dose-dependant significant (P < 0.05) decrements were observed in epididymal sperm kinematic attributes (progressive motility, rapid, fast progressive, VCL, VSL, VAP, LIN, STR, WOB and TYPE A (STR >80 %, ALH 2.5 μm) by CASA), viability, plasma membrane functional integrity, and mitochondrial membrane potential which were associated with insignificant decrease in serum testosterone levels. The histoarchitectural studies of testes showed progressive loss of spermatozoa concentration in the seminiferous tubules as the As(V) dose increased. The mice exposed to As(V) had an increase in the As accumulation, protein carbonylation, and lipid peroxidation levels associated with alterations in SOD, CAT, and GST activities in the testes. In conclusion, higher doses of As(V) (more than 50 ppm) were found to be testicular toxicants which impaired semen quality by inducing oxidative stress in the testicular microenvironment.

The objective of this was to establish the effects of red spectrum of light (650 nm, treated n = 12) and normal spectrum of light (450 nm control = 12) on GnRH-I mRNA expression, amplitude and frequency of luteinizing hormone (LH), and egg production from 72-82 weeks of age in white leghorn hens. Birds exposed to red spectrum of wavelength significantly improved (P < 0.01) steroid hormone, and egg production improved over old laying 72 to 82 weeks. Weekly interval profiles followed the same pattern. At 77th weeks of age blood, samples from both the groups were collected at every 3 h for 36 h to study the pulsatile secretion of LH surges. Plasma LH concentration was higher (P < 0.01) in treated birds with more number of frequencies and amplitude LH surges in plasma of treated birds. LH frequencies were more pronounced and advanced during 36 h of sampling at 3 h interval in treated birds. Weekly interval of plasma LH, E2β, and P(4) concentrations increased (P < 0.01) in treated birds from 72 to 82 weeks of age. GnRH-I mRNA concentration was significantly (P < 0.01) higher in birds exposed to red spectrum of light compared to controls. It is hypothesized that exposure of birds to red spectrum of light-enhanced (P < 0.01) GnRH-I mRNA with more number of yellow yolky follicles was found in birds exposed to red spectrum of light during 77 days (72-82 weeks of age) of experimental period. It is concluded that higher levels of GnRH-I mRNA, LH, E2β, and P(4) concentration with lower incidence of pause days enabled the birds to lay more eggs even later in the productive period by modulating the wavelengths of light under normal husbandry conditions.

Unlabelled: Artemisia annua L. produces a number of sesquiterpene synthases, which catalyze the conversion of farnesyl diphosphate to various sesquiterpenes. The cDNAs encoding amorpha-4,11-diene synthase (ADS), a key enzyme in the artemisinin biosynthesis, and epi-cedrol synthase (ECS), a complex sesquiterpene cyclization synthase, were cloned into Cowpea mosaic virus-based viral vector (pEAQ-HT) with Kozak consensus motif and C-terminal histidine tag. The plasmids were transformed into Agrobacterium LBA4404 and, agroinfiltrated into Nicotiana benthamiana leaves along with vector (pJL3:p19) containing Tomato bushy stunt virus post-transcriptional gene silencing suppressor. Quantitative PCR was carried out to measure the transcript levels at 0, 3, 6, 9, 12 and 15 days post-infiltration (dpi). The highest relative expression was observed at 9 dpi for both genes. Transiently expressed recombinant proteins of ADS and ECS were confirmed by SDS-PAGE and western blot. Recombinant proteins were extracted from 9 dpi leaves and purified by immobilized metal ion affinity chromatography using histidine tag, which produced yields of 90 and 96 mg kg⁻¹ fresh weight of leaves for ADS and ECS, respectively. Activities of the purified enzymes were assayed using gas chromatography-mass spectrometry for product identification and quantification using valencene as internal standard. The recombinant ADS and ECS converted farnesyl diphosphate into amorpha-4,11-diene (97 %) and epi-cedrol (96 %) as the major products, respectively. The purified enzymes exhibited the specific activity of 0.002 and 0.01 μmol min⁻¹ mg⁻¹ protein for ADS and ECS, respectively. The apparent k(cat) values were 2.1 × 10⁻³ s⁻¹ and 11 × 10⁻³ s⁻¹ for ADS and ECS, respectively., Key Message: Agroinfiltration of leaves of Nicotiana bentamiana can be used to produce recombinant biosynthetic enzymes as exemplified by two sesquiterpene synthases from Artemisia annua in relatively high yields.

Plant flavonoids are emerging as potent therapeutic drugs effective against a wide range of free radical-mediated diseases. Morin (3,5,7,2',4'-pentahydroxyflavone), a member of flavonols, is an important bioactive compound by interacting with nucleic acids, enzymes and protein. In this study, we found that morin (30 mg/kg body weight) by oral administration offers protection against hyperammonemia by means of reducing blood ammonia, oxidative stress and enhancing antioxidant status in ammonium chloride-induced (100 mg/kg body weight; i.p) hyperammonemic rats. Enhanced blood ammonia, plasma urea, lipid peroxidation in circulation and tissues (liver and brain) of ammonium chloride-treated rats was accompanied by a significant decrease in the tissues levels of superoxide dismutase (SOD), catalase, reduced glutathione (GSH) and glutathione peroxidase (GPx). Morin administered rats showed a significant reduction in ammonia, urea, lipid peroxidation with a simultaneous elevation in antioxidant levels. Cotreatment with morin prevented the elevation of liver marker enzymes induced by ammonium chloride. The body weight of the animals decreased significantly on ammonium chloride administration when compared with control group. However, cotreatment with morin significantly prevented the decrease of the body weight caused by ammonium chloride. Hyperammonemic rats show liver fibrosis, steatosis, sinusoidal dilatation, etc., along with necrosis, microcystic degeneration in brain. All these changes were reduced in hyperammonemic rats treated with Morin, which too correlated with the biochemical observations. In conclusion, these findings indicate that morin exert antioxidant potential and offer protection against ammonium chloride-induced hyperammonemia. But the exact underlying mechanism needs to be elucidated.
