Exposing honey bees to isopentylacetate (IPA) can cause stress-related changes in learning performance. In bees of foraging age, IPA's effects on learning are mimicked by C-type allatostatins (AstCC, AstCCC) injected into the brain. Here we ask whether allatostatins induce a similar response in young (6-day-old) bees and if so, whether their effects on learning performance are modulated by queen mandibular pheromone (QMP). We found that young bees exposed to IPA responded less to the conditioned stimulus during training than controls (Type 1-like stress response). AstCC treatment induced a similar response, but only in bees maintained without QMP. Bees exposed to QMP responded to AstCC with increased odour responsiveness and odour generalisation in the 1-h memory test (Type 2-like response). Type 2-like responses could be induced also by the A-type allatostatin, AstA. However, in bees exposed to QMP, AstA-induced odour generalisation was absent. Effects of AstCCC treatment in young bees were weak, indicating that responsiveness to this peptide changes with age. Our findings are consistent with the hypothesis that honey bee allatostatins play a role in stress reactivity, but suggest in addition that allatostatin signalling is age dependent and susceptible to modulation by pheromone released by the queen bee.

Olfactory learning and memory processes in Drosophila have been well investigated with aversive conditioning, but appetitive conditioning has rarely been documented. Here, we report for the first time individual olfactory conditioning of proboscis activity in restrained Drosophila melanogaster. The protocol was adapted from those developed for proboscis extension conditioning in the honeybee Apis mellifera. After establishing a scale of small proboscis movements necessary to characterize responses to olfactory stimulation, we applied Pavlovian conditioning, with five trials consisting of paired presentation of a banana odour and a sucrose reward. Drosophila showed conditioned proboscis activity to the odour, with a twofold increase of percentage of responses after the first trial. No change occurred in flies experiencing unpaired presentations of the stimuli, confirming an associative basis for this form of olfactory learning. The adenylyl cyclase mutant rutabaga did not exhibit learning in this paradigm. This protocol generated at least a short-term memory of 15 min, but no significant associative memory was detected at 1 h. We also showed that learning performance was dependent on food motivation, by comparing flies subjected to different starvation regimes.

The processes whereby developing neurones acquire morphological features that are common to entire populations (thereby allowing the definition of neuronal types) are still poorly understood. A mathematical model of neuronal arborizations may be useful to extract basic parameters or organization rules, hence helping to achieve a better understanding of the underlying growth processes. We present a parsimonious statistical model, intended to describe the topological organization of neuritic arborizations with a minimal number of parameters. It is based on a probability of splitting which depends only on the centrifugal order of segments. We compare the predictions made by the model of several topological properties of neurones with the corresponding actual values measured on a sample of honeybee (olfactory) antennal lobe neurones grown in primary culture, described in a previous study. The comparison is performed for three populations of segments corresponding to three neuronal morphological types previously identified and described in this sample. We show that simple assumptions together with the knowledge of a very small number of parameters allow the topological reconstruction of representative (bi-dimensional) biological neurones. We discuss the biological significance (in terms of possible factors involved in the determinism of neuronal types) of both common properties and cell-type specific features, observed on the neurones and predicted by the model.
