The development of detailed national pathways towards sustainable food and land systems aims to provide stakeholders with clarity on how long-term goals could be achieved and to reduce roadblocks in the way to making commitments. However, the inability to perfectly capture the relationships between all variables in a system and the unknown probability of future values (deep uncertainty) makes it very difficult to design scenarios that account for the full breadth of system uncertainty. Here we use scenario discovery to systematically explore the effect of different parameter ranges on model outputs, and design resilient pathways to sustainability in which multiple target achievement requires a broad portfolio of solutions. We use a model of the Australian food and land system, the FABLE (Food, Agriculture, Biodiversity, Land-use, Energy) Calculator, to investigate conditions for achieving a sustainable Australian food and land system under scenarios based on the Shared Socioeconomic Pathways (SSP) 1, 2, and 3 narratives. Here we link the FABLE Calculator with a Monte Carlo simulation tool to explore hundreds of thousands of scenarios. This allows us to identify the ranges of systemic drivers that achieve multiple sustainability targets around diets, net forest growth, agricultural water consumption, greenhouse gas emissions, biodiversity conservation, and exports by 2050. Our results show that livestock productivity and density, afforestation, and dietary change are powerful influencers for sustainability target achievement. Around 10% of the SSP1 scenarios could achieve all modelled sustainability targets. However, practically none of the scenarios based on SSP2 and SSP3 narratives could achieve such targets. The results suggest that there are options to achieve a more sustainable and resilient Australian food and land-use system with better socio-economic and environmental outcomes than under current trends. However, its achievement requires significant structural changes and coordinated interventions in several components of the domestic food and land system to increase its resilience and environmental and socio-economic performance. The Sustainable Development Goals (SDGs) recognise the importance of action across all scales to achieve a sustainable future. To contribute to overall national- and global-scale SDG achievement, local communities need to focus on a locally-relevant subset of goals and understand potential future pathways for key drivers which influence local sustainability. We developed a participatory method to co-create local socioeconomic pathways by downscaling the SDGs and driving forces of the shared socioeconomic pathways (SSPs) via a local case study in southern Australia through contextual analysis and community engagement. We linked the SSPs and SDGs by identifying driving forces and describing how they affect the achievement of local SDGs. We co-created six local socioeconomic pathways with the local community which track towards futures with different levels of fulfilment of the SDGs and each encompasses a narrative storyline incorporating locally-specific ideas from the community. We tested and validated the local pathways with the community. This method extends the SSPs in two dimensions-into the broader field of sustainability via the SDGs, and by recontextualizing them at the local scale. Introduction: Patients with congenital agammaglobulinemia, characterized by a defect in B lymphocyte differentiation causing B alymphocytosis, require life-long IgG replacement. There is scant literature regarding the effectiveness of IgG treatment at preventing mucosal (particularly sinopulmonary tract) infection and whether current management adequately restores "normal" health and quality of life (QoL). We aimed to document infective episodes pre- and post-commencing IgG replacement, determine any change in lung function and structure and assess respiratory status and QoL in a cohort of patients treated in Newcastle., Methods: Clinical data were extracted from medical records of 15 patients identified from the immunology database, focusing on infective episodes, serial chest CT and spirometry results. Thirteen patients completed a selection of standardized and validated questionnaires assessing physical health, respiratory health and QoL., Results: Pediatric patients on IgG therapy suffered fewer infections per patient year (0.74) than adults (2.13). 6/14 patients showed deteriorating respiratory status despite adequate therapy. Health questionnaires revealed a significant burden of respiratory disease on a patient's life., Conclusion: Clinical data showed patients with congenital agammaglobulinemia receiving immunoglobulin therapy retained a higher than average infection rate, most of which affected mucosal barriers. Most patients self-reported worse respiratory symptoms, a lower respiratory-related QoL and a lower general health QoL relative to a healthy population. Most participants had progressive structural lung damage and decreased lung function. These results suggest that current management is not entirely effective at preventing deterioration of respiratory health or restoring QoL.

Changing unsustainable natural resource use in agricultural landscapes is a complex social-ecological challenge that cannot be addressed through traditional reductionist science. More holistic and inclusive (or transdisciplinary) processes are needed. This paper describes a transdisciplinary project for natural resource management planning in two regions (Eyre Peninsula and South Australian Murray-Darling Basin) of southern Australia. With regional staff, we reviewed previous planning to gain an understanding of the processes used and to identify possible improvement in plan development and its operation. We then used an envisioning process to develop a value-rich narrative of regional aspirations to assist stakeholder engagement and inform the development of a land use management option assessment tool called the landscape futures analysis tool (LFAT). Finally, we undertook an assessment of the effectiveness of the process through semi-structured stakeholder interviews. The planning process review highlighted the opinion that the regional plans were not well informed by available science, that they lacked flexibility, and were only intermittently used after publication. The envisioning process identified shared values-generally described as a trust, language that is easily understood, wise use of resources, collaboration and inclusiveness. LFAT was designed to bring the best available science together in a form that would have use in planning, during community consultation and in assessing regional management operations. The LFAT provided spatially detailed but simple models of agricultural yields and incomes, plant biodiversity, weed distribution, and carbon sequestration associated with future combinations of climate, commodity and carbon prices, and costs of production. Stakeholders were impressed by the presentation and demonstration results of the software. While there was anecdotal evidence that the project provided learning opportunities and increased understanding of potential land use change associated with management options under global change, the direct evidence of influence in the updated regional plan was limited. This project had elements required for success in transdisciplinary research, but penetration seems limited. Contributing factors appear to be a complexity of climate effects with economic uncertainty, lack of having the project embedded in the plan revision process, limited continuity and capacity of end users and limited after project support and promotion. Strategies are required to minimise the controlling influence that these limitations can have.

The members of the Rho family of small GTPases are involved in an array of cellular processes, including regulation of the actin cytoskeleton, cell polarity, microtubule dynamics, membrane transport, and transcription factor activity. Recent findings have implicated the Rho-proteins as key regulators of angiogenesis, modulating a diversity of cellular processes, including vascular permeability, extracellular matrix remodeling, migration, proliferation, morphogenesis, and survival.

The Rho family of small GTPases is involved in a diverse array of cellular processes, including regulation of the actin cytoskeleton, cell polarity, microtubule dynamics, membrane transport pathways and transcription factor activity. Recent findings have implicated the Rho proteins as key regulators of the skeletal myogenic program; however, much controversy presently exists as to the precise role of these proteins in this process. This review examines the present controversial findings pertaining to the Rho family's regulation of skeletal myogenesis and extrapolates from both other differentiation systems and recently published data the possible mechanisms by which these proteins function in the myogenic cascade.

The 15 N abundance of tissues of five Prosopis specimens at our primary study site (a Prosopis woodland at Harper's Well in the Sonoran desert of Southern California) was determined over two growing seasons 1980 and 1981. The 15 N abundance of soil and of tissues of presumed non-N 2 -fixing (control) plants was also measured. Prosopis tissues were significantly lower in 15 N than either soil N or corresponding tissues of presumed non-N 2 -fixing plants which derive their N entirely from soil. Soil N was also significantly higher in 15 N than atmospheric N 2 . We conclude that it is feasible to use variations in the natural abundance of 15 N as an index of N 2 -fixation in this kind of ecosystem, and that N 2 -fixation is of considerable importance to Prosopis growing at this site.We also determined the 15 N abundance of leaf tissue of presumed N 2 -fixing and control plants growing at the same site at six additional sites (five in the Sonoran desert of southern California and one in Baja California, Mexico near the town of Catavina). Four of these additional sites were dominated by Prosopis and two were mixed communities. There were statistically significant differences between the 15 N abundances of the pooled legume population and control plants at all sites, although not every legume specimen exhibited this difference. From 15 N abundance data we estimated the fractional contribution of biologically fixed N to the N economy of desert legumes. We concluded that N 2 -fixation is very important to Prosopis at six of seven sites in the Sonoran Desert. At the site where Prosopis did not appear to be fixing N 2 , N 2 -fixation was important only for legumes of the sub-family Papilionoideae, Lupinus, Dalea, Astragalus and Lotus.
