This edited volume focuses on the interplay between sleep and circadian rhythms with health, aging and longevity. Sleep is absolutely important for human health and survival, as insufficient sleep is associated with a plethora of conditions, including the poor quality of life, onset of several diseases, and premature death. The sleep–wake cycle is an evolutionary conserved neurobiological phenomenon, and is a prominent manifestation of the biological clocks localised in the suprachiasmatic nucleus (SCN). Understanding bidirectional relationship between sleep and circadian rhythms is of utmost importance and urgency, especially in the context of modern lifestyle where sleep is often out of phase with the internal body clocks, social jetlag, artificial lights and so on. The 25 chapters by leading researchers and experts from 11 countries are arranged into seven sections: understanding sleep and clock interlink in health and longevity; sleep, aging and longevity; clock, aging and longevity; melatonin, sleep and clock; genetic regulation of sleep and clock; therapeutic interventions in sleep disorders and clock misalignment; and experimental models to study sleep and clocks in aging and longevity. This book is useful for advanced undergraduate and graduate students, and researchers, educators, and other biomedical professionals.

This open access book provides insight on how to interpret capability in ageing – one's individual ability to perform actions in order to reach goals one has reason to value – from a multidisciplinary approach. With for the first time in history there being more people in the world aged 60 years and over than there are children below the age of 5, the book describes this demographic trends as well as the large global challenges and important societal implications this will have such as a worldwide increase in the number of persons affected with dementia, and in the ratio of retired persons to those still in the labor market. Through contributions from many different research areas, it discussed how capability depends on interactions between the individual (e.g. health, genetics, personality, intellectual capacity), environment (e.g. family, friends, home, work place), and society (e.g. political decisions, ageism, historical period). The final chapter summarizes the differencesand similarities in these contributions. As such this book provides an interesting read for students, teachers and researchers at different levels and from different fields interested in capability and multidisciplinary research.

This book describes the recent advancement of basic research on the biology of aging and longevity studies in various organisms, as well as the neurobiology of aging and neurodegeneration mechanisms. This new volume in the Subcellular Biochemistry series will focus on the biochemistry and cellular biology of aging processes in human cells. The chapters will be written by experts in their respective fields and will focus on a number of the current key areas of research in subcellular aging research. Main topics for discussion are mitochondrial aging, protein homeostasis and aging and the genetic processes that are involved in aging. There will also be chapters that are dedicated to the study of the roles of a variety of vitamins and minerals on aging and a number of other external factors (microbiological, ROS, inflammation, nutrition). This book will provide the reader with a state of the art overview of the subcellular aging field. This book will be published in cooperation with a second volume that will discuss the translation of the cell biology of aging to a more clinical setting and it is hoped that the combination of these two volumes will bring a deeper understanding of the links between the cell and the body during aging.

This edited work presents the most current evidence on osteosarcopenia from bench to bedside, which is expected to facilitate the understanding of this syndrome and to develop preventive and therapeutic strategies. With our aging population, chronic diseases such as osteoporosis and sarcopenia are becoming highly prevalent. Fortunately, our understanding of the bone and muscle interactions has increased in recent years. This has allowed to the coining of the term osteosarcopenia to describe a syndrome in which these two diseases overlap. This overlap between osteoporosis and sarcopenia has major negative effects not only on our older adults but also on health systems worldwide. Readers will find a highly translational approach that starts with a summary of recent discoveries on stem cells biology, muscle and bone interactions – including the role of local bone and muscle fat – followed by comprehensive reviews on myokines (i.e. myostatin), osteokines (i.e.osteocalcin) and adipokines (i.e. interleukins) as major players and determinants of bone and muscle loss with aging. In addition, the role of sex steroids (i.e. estrogens, androgens), and calciotropic hormones (i.e. parathyroid hormone, vitamin D) in the pathogenesis of this syndrome is also reviewed. This volume of the subcellular Biochemistry series will attempt to bridge the gap between the subcellular events that are related to aging as they were described in the first volume of this set of two books and the reality of aging as this is seen in clinical practice. All chapters will start from the biochemistry or cell biology, where the data is available and work up towards the understanding that we have of aging in the various areas that are related to the subject. Key focus points for this volume are nutrition, external factors and genetics on aging. There will also be chapters that will focus on various organs or tissues in which aging has been well studied, like the eyes, the muscles, the immune system and the bones. The aim of the book project and the book project that is published in concert with this volume is to bring the subcellular and clinical areas into closer contact.

Biological aging as the time-depending general decline of biological systems associated with a progressively increasing mortality risk is a general phenomenom of great significance. The underlying processes are very complex and depending on genetic and environment factors. These factors encode or affect a network of interconnected cellular pathways. In no system this network has been deciphered in greater detail. However, the strategy of studying various biological systems has let to the identification of pathways and specific modules and makes it obvious that aging is the result of different overlapping mechanisms and pathways. Some of these appear to be conserved ('public') among species, others are specific or'private'and only of significance in one or a few organisms. The proportion of the population over 65 years of age is increasing steadily in most industrialized countries. In the United States the proportion of elderly people has risen from four percent in 1900 to 11% in 1978, and is projected to be 14% by the year 2000. The occurrence of debilitating chronic diseases in the elderly increases with each additional year. These diseases, along with the natural loss of tissue function that occurs throughout adult life, impose a heavy burden on the health care system. Nutri­ tion plays an important etiologic role in many of these degenerative changes. Conse­ quently, the aging segment of the population presents a challenge to the nutrition scientist, who should be able to recommend optimal intakes of nutrients to minimize the functional losses associated with aging and to optimize the health of those already elderly. This sixth volume in the series Human Nutrition: A Comprehensive Treatise provides a conspectus of the various interactions of nutrition with the aging process and a comprehensive survey of current knowledge of the amounts of individual nutrients needed by the elderly. The volume begins with a general survey of the multifaceted relationship of nutrition to aging, followed by four chapters on how nutrition can affect age-related changes in selected body functions. The next six chapters cover the avail­ able evidence regarding the needs of the elderly for dietary energy, protein, calcium, trace elements, vitamins, and fiber.

The objective of this book is to provide information that will be useful to people in a variety of disciplines who wish to learn more about normal aging processes in the human body. Although gerontologists in the biological sciences are making great strides in research on human aging and documenting this work in mono­ graphs, texts, and review chapters, this information is generally not easily acces­ sible nor is it comprehensible to nonprofessionals in these fields. This book is intended to provide a summary of this work, along with its implications for psy­ chological functioning of the aging individual. The majority of the book is devoted to describing the results of research on the physiological changes in the human body with aging and to seeking explana­ tions for these age effects. This description has been approached in such a way as to make it readable for the nonspecialist, but also to focus on research issues that will be useful reading for those who are currently working in these parti­ cular areas. In addition, throughout the book, I have tried to develop some themes regarding physiological and psychological adaptation during adulthood.

It is a truism that as we age there are a number of underlying physiological changes conspiring to alter our level of behavioral and cognitive function­ ing. Despite the inherent interrelatedness of these behavioral and cognitive changes, all too often the papers we read confine themselves to specific, isolated components of the developing process. Although exceptions nat­ urally exist, we believe that these exceptions should become rule. Although an integrated approach is important in all areas of adult devel­ opment, it is perhaps particularly germane in the study of atypical aging. Here, changes in overall functioning can occur in rapid succession, with the synchrony of decline between different subprocesses making it difficult to factor changes in one process from changes in another. For example, because changes in cognitive functioning co-occur with other dramatic changes in (motoric) response capacities, it is unclear how one can effec­ tively study changes in the ability to cognize independent of changes in the very mechanisms (ability to execute motor sequences) so often used to index cognitive performance.

Aging is loosely defined as the accumulation of changes in an organism over time. At the cellular level such changes are distinct and multidimensional: DNA replication ceases, cells stop dividing, they become senescent and eventually die. DNA metabolism and chromosomal maintenance, together with protein metabolism are critical in the aging process. The focus of this book is on the role of protein metabolism and homeostasis in aging. An overview is provided of the current knowledge in the area, including protein synthesis, accuracy and repair, post-translational modifications, degradation and turnover, and how they define and influence aging. The chapters mainly focus on well-characterised factors and pathways, but new areas are also presented, where associations with aging are just being elucidated by current experimental data. Protein turnover, the balance between protein synthesis and protein degradation are carefully maintained in healthy cells. Chapters 1 and 2 illustrate that aging cells are characterised by alterations in the rate, level and accuracy of protein synthesis compared to young ones, and that mRNA translation, essential for cell growth and survival, is controlled at multiple levels. The theory that growth and somatic maintenance are believed to be antagonistic processes is described in Chapter 3: inhibition of protein synthesis results in decreased rates of growth and development, but also confers an extension of lifespan, as shown for example by the effects of dietary restriction in various models organisms.

Just as the health costs of aging threaten to bankrupt developed countries, this book makes the scientific case that a biological'bailout'could be on the way, and that human aging can be different in the future than it is today. Here 40 authors argue how our improving understanding of the biology of aging and selected technologies should enable the successful use of many different and complementary methods for ameliorating aging, and why such interventions are appropriate based on our current historical, anthropological, philosophical, ethical, evolutionary, and biological context. Challenging concepts are presented together with in-depth reviews and paradigm-breaking proposals that collectively illustrate the potential for changing aging as never before. The proposals extend from today to a future many decades from now in which the control of aging may become effectively complete. Aging is inevitable: this is gerontological dogma. And humans do inevitably grow old, which is probably why it seems so unlikely to us that other forms of life could escape aging. Escaping aging is not escaping death. Death is an inherent part of life, and it can strike any time. But the question is whether death necessarily becomes more likely as life proceeds. And it does not. The theoretical results in this monograph indicate that life provides alternative strategies. While some organisms will deteriorate over adult ages, for others mortality appears to fall or remain constant, at least over an extended period of life after reproductive maturity. This is empirically observed especially for species that keep on growing during adult ages. Perhaps the diversity of aging matches the diversity of life. My thesis, the central insight of this monograph, is: to deeply understand why some species age it is necessary to understand why other species do not.

Stress is a double edged sword. Whereas chronic stress has well known harmful effects, recent research shows that intermittent exposure of cells and organisms to mild stress can improve various biological parameters, including severe stress tolerance, health span and longevity. Mild stress-induced hormesis is potentially a powerful tool in gerontological research and interventions. This book brings together some of the world leaders in this area of research who present a critical analysis of the ongoing research with respect to the effects and mechanisms of action of various types of mild stress, such as irradiation, heat, physical exercise, hypergravity, pro-oxidants and nutritional components, on a variety of biological systems ranging from fruitflies to human cells and bodies. Their separate chapters and a final collective chapter open up a whole new world of possibilities for future research in aging, successful aging, and health span extension.
