目的探讨他克莫司(FK506)对肝移植患者外周静脉血IL-2、IL-10基因表达的影响及意义。方法选择接受原位肝移植患者60例,均采用FK506+甲基强的松龙+吗替麦考酚酯三联用药治疗。肝移植术后第1、3周取外周血,检测血清ALT、AST及外周静脉血IL-2、IL-10基因表达。结果与肝移植术后1周比较,肝移植术后3周患者血清ALT水平明显降低(P<0.05),而血清AST水平变化不明显(P>0.05);肝移植术后3周外周静脉血IL-2基因表达明显降低(P<0.05),IL-10基因表达明显升高(P<0.01)。血清ALT水平与IL-2基因表达呈正相关,与IL-10基因表达呈负相关(P均<0.05);血清AST水平与IL-2、IL-10基因表达均无相关性(P均>0.05)。结论FK506可能通过影响外周静脉血IL-2、IL-10基因表达,从而抑制免疫排斥,诱导免疫耐受。 [ABSTRACT FROM AUTHOR]
