Until the early 1990s, cyst nematodes were abundant pathogens in fields where hosts of Heterodera spp. were frequent members of crop rotations along California’s Central Coast. To mitigate damage caused by Heterodera schachtii and H. cruciferae, the soil fumigant 1,3-dichloropropene (1,3-D) was used by more than 43% of surveyed broccoli growers. Over the last few decades, use of 1,3-D and other nematicides has dramatically diminished, suggesting a decline in nematode disease pressure. The goal of this study was to examine the hypothesis that increased population densities of nematophagous fungi contribute to the low populations of Heterodera spp. in fields frequently cropped to their hosts. In 2016, soil samples were collected from 152 Brassica fields with a broad geographical distribution, from Santa Barbara County to Santa Cruz County. The average number of Heterodera cysts per 250 cm3 of soil ranged from 0.5 to 27.5, with 62% of the soils harboring no detectable cyst nematodes and only a few samples reaching a potentially damaging threshold level. A baiting experiment with H. schachtii and cabbage was performed in a greenhouse to detect nematophagous fungi associated with nematode females as their posterior end emerged and became exposed to the soil’s rhizosphere. An Illumina-based sequence analysis of these H. schachtii females identified several known nematophagous fungi, including members of the Hyalorbilia oviparasitica clade, Pochonia chlamydosporia, certain Fusarium spp., and others. These soils clearly harbor a diverse population of hyperparasitic fungi that could be biologically suppressing cyst nematodes below a damaging threshold. The lettuce downy mildew pathogen, Bremia lactucae, is an obligate oomycete that causes extensive produce losses. Initial chlorotic symptoms that severely reduce the market value of the produce are followed by the appearance of white, downy sporulation on the abaxial side of the leaves. These spores become airborne and disseminate the pathogen. Controlling lettuce downy mildew has relied on repeated fungicide applications to prevent outbreaks. However, in addition to direct economic costs, heterogeneity and rapid adaptation of this pathogen to repeatedly applied fungicides has led to the development of fungicide-insensitivity in the pathogen. We deployed a quantitative PCR assay-based detection method using a species-specific DNA target for B. lactucae coupled with a spore trap system to measure airborne B. lactucae spore loads within three commercial fields that each contained experimental plots, designated EXP1 to EXP3. Based upon these measurements, when the spore load in the air reached a critical level (8.548 sporangia per m3 air), we advised whether or not to apply fungicides on a weekly basis within EXP1 to EXP3. This approach saved three sprays in EXP1, and one spray each in EXP2 and EXP3 without a significant increase in disease incidence. The reduction in fungicide applications to manage downy mildew can decrease lettuce production costs while slowing the development of fungicide resistance in B. lactucae by eliminating unnecessary fungicide applications.

Isolates of the Fusarium oxysporum species complex have been characterized as plant pathogens that commonly cause vascular wilt, stunting, and yellowing of the leaves in a variety of hosts. F. oxysporum species complex isolates have been grouped into formae speciales based on their ability to cause disease on a specific host. F. oxysporum f. sp. fragariae is the causal agent of Fusarium wilt of strawberry and has become a threat to production as fumigation practices have changed in California. F. oxysporum f. sp. fragariae is polyphyletic and limited genetic markers are available for its detection. In this study, next-generation sequencing and comparative genomics were used to identify a unique genetic locus that can detect all of the somatic compatibility groups of F. oxysporum f. sp. fragariae identified in California. This locus was used to develop a TaqMan quantitative polymerase chain reaction assay and an isothermal recombinase polymerase amplification (RPA) assay that have very high sensitivity and specificity for more than 180 different isolates of the pathogen tested. RPA assay results from multiple field samples were validated with pathogenicity tests of recovered isolates.

In 2014 an unidentified disease was found on lettuce and celery grown in the Salinas Valley, Monterey Co., CA. Lettuce leaf symptoms were round to oval spots, 2 to 4 mm in diameter, white to tan, with a brown border. Celery leaf spots were round to oval, 2 to 4 mm in diameter, white to tan, with a chlorotic border. Initially it was thought that the damage was caused by chemical drift; however, field personnel noted the association of the disease with nearby carrots infected with Alternaria leaf blight. Microscopic examination of lettuce and celery leaves showed obclavate, brown, multicelled conidia in the spot centers, and a fungus was consistently isolated from this tissue. Cultural, morphological, and molecular analyses confirmed the fungus to be Alternaria dauci. Isolates of A. dauci from lettuce, celery, and carrot were pathogenic on all three crops. Field surveys showed that disease severity was greatest on the side of the lettuce or celery field closest to carrots. This is the first documentation that the carrot pathogen A. dauci can cause a disease on lettuce and celery grown in the field. The disease is designated as Alternaria leaf speck. Alternaria dauci was consistently isolated from nearby spinach plants showing identical symptoms and signs. However, pathogenicity tests were inconsistent and additional studies are needed to document A. dauci on spinach.

Bremia lactucae is an obligate, oomycete pathogen of lettuce that causes leaf chlorosis and necrosis and adversely affects marketability. The disease has been managed with a combination of host resistance and fungicide applications with success over the years. Fungicide applications are routinely made under the assumption that inoculum is always present during favorable environmental conditions. This approach often leads to fungicide resistance in B. lactucae populations. Detection and quantification of airborne B. lactucae near lettuce crops provides an estimation of the inoculum load, enabling more judicious timing of fungicide applications. We developed a quantitative polymerase chain reaction (qPCR)-based assay using a target sequence in mitochondrial DNA for specific detection of B. lactucae. Validation using amplicon sequencing of DNA from 83 geographically diverse isolates, representing 14 Bremia spp., confirmed that the primers developed for the TaqMan assays are species specific and only amplify templates from B. lactucae. DNA from a single sporangium could be detected at a quantification cycle (Cq) value of 32, and Cq values >35 were considered to be nonspecific. The coefficient of determination (R2) for regression between sporangial density derived from flow cytometry and Cq values derived from the qPCR was 0.86. The assay was deployed using spore traps in the Salinas Valley, where nearly half of U.S. lettuce is produced. The deployment of this sensitive B. lactucae-specific assay resulted in the detection of the pathogen during the 2-week lettuce-free period as well as during the cropping season. These results demonstrate that this assay will be useful for quantifying inoculum load in and around the lettuce fields for the purpose of timing fungicide applications based on inoculum load.

Angular leaf spot of cucurbits is generally considered to be caused by Pseudomonas syringae pv. lachrymans. It has a worldwide distribution and has been observed to emerge sporadically under humid and wet conditions. Reports of multiple P. syringae pathovars associated with the disease and lack of molecular analysis has left the true diversity of populations in the United States unclear. In this study, we collected 27 P. syringae strains causing foliar lesions and blighting on watermelon, cantaloupe, and squash in Florida, Georgia, and California over several years. Strains were fluorescent on King’s medium B agar and displayed the typical phenotypic and biochemical characteristics of P. syringae. P. syringae pv. lachrymans is a member of genomospecies 2. However, the genetic profiles obtained through both MLSA (gyrB, rpoD, gapA, and gltA) and BOX-PCR (BOXA1R) identified 26 of the P. syringae strains to be distributed among three clades within genomospecies 1, and phylogenetically distinct from genomospecies 2 member P. syringae pv. lachrymans. A novel MLSA haplotype of the pathogen common to all states and cucurbit hosts was identified. Considerable genetic diversity among P. syringae strains infecting cucurbits is associated with the same disease, and reflects the larger ecological diversity of P. syringae populations from genomospecies 1.

Verticillium wilt caused by V. dahliae is a devastating disease of lettuce in California (CA). The disease is currently restricted to a small geographic area in central coastal CA, even though cropping patterns in other coastal lettuce production regions in the state are similar. Infested spinach seed has been implicated in the introduction of V. dahliae into lettuce fields but direct evidence linking this inoculum to wilt epidemics in lettuce is lacking. In this study, 100 commercial spinach fields in four coastal CA counties were surveyed to evaluate the frequency of Verticillium species recovered from spinach seedlings and the area under spinach production in each county was assessed. Regardless of the county, V. isaacii was the most frequently isolated species from spinach followed by V. dahliae and, less frequently, V. klebahnii. The frequency of recovery of Verticillium species was unrelated to the occurrence of Verticillium wilt on lettuce in the four counties but was related to the area under spinach production in individual counties. The transmission of V. dahliae from infested spinach seeds to lettuce was investigated in microplots. Verticillium wilt developed on lettuce following two or three plantings of Verticillium-infested spinach, in independent experiments. The pathogen recovered from the infected lettuce from microplots was confirmed as V. dahliae by polymerase chain reaction assays. In a greenhouse study, transmission of a green fluorescence protein-tagged mutant strain of V. dahliae from spinach to lettuce roots was demonstrated, after two cycles of incorporation of infected spinach residue into the soil. This study presents conclusive evidence that V. dahliae introduced via spinach seed can cause Verticillium wilt in lettuce.

Verticillium dahliae is a soilborne fungus that causes Verticillium wilt on multiple crops in central coastal California. Although spinach crops grown in this region for fresh and processing commercial production do not display Verticillium wilt symptoms, spinach seeds produced in the United States or Europe are commonly infected with V. dahliae. Planting of the infected seed increases the soil inoculum density and may introduce exotic strains that contribute to Verticillium wilt epidemics on lettuce and other crops grown in rotation with spinach. A sensitive, rapid, and reliable method for quantification of V. dahliae in spinach seed may help identify highly infected lots, curtail their planting, and minimize the spread of exotic strains via spinach seed. In this study, a quantitative real-time polymerase chain reaction (qPCR) assay was optimized and employed for detection and quantification of V. dahliae in spinach germplasm and 15 commercial spinach seed lots. The assay used a previously reported V. dahliae-specific primer pair (VertBt-F and VertBt-R) and an analytical mill for grinding tough spinach seed for DNA extraction. The assay enabled reliable quantification of V. dahliae in spinach seed, with a sensitivity limit of ≈1 infected seed per 100 (1.3% infection in a seed lot). The quantification was highly reproducible between replicate samples of a seed lot and in different real-time PCR instruments. When tested on commercial seed lots, a pathogen DNA content corresponding to a quantification cycle value of ≥31 corresponded with a percent seed infection of ≤1.3%. The assay is useful in qualitatively assessing seed lots for V. dahliae infection levels, and the results of the assay can be helpful to guide decisions on whether to apply seed treatments.

Verticillium dahliae is responsible for Verticillium wilt on a wide range of hosts, including strawberry, on which low soil inoculum densities can cause significant crop loss. Determination of inoculum density is currently done by soil plating but this can take 6 to 8 weeks to complete and delay the grower's ability to make planting decisions. To provide a faster means for estimating pathogen populations in the soil, a multiplexed TaqMan real-time polymerase chain reaction (PCR) assay based on the ribosomal DNA (rDNA) intergenic spacer (IGS) was developed for V. dahliae. The assay was specific for V. dahliae and included an internal control for evaluation of inhibition due to the presence of PCR inhibitors in DNA extracted from soil samples. An excellent correlation was observed in regression analysis (R2 = 0.96) between real-time PCR results and inoculum densities determined by soil plating in a range of field soils with pathogen densities as low as 1 to 2 microsclerotia/g of soil. Variation in copy number of the rDNA was also evaluated among isolates by SYBR Green real-time PCR amplification of the V. dahliae-specific amplicon compared with amplification of several single-copy genes and was estimated to range from ≈24 to 73 copies per haploid genome, which translated into possible differences in results among isolates of ≈1.8 cycle thresholds. Analysis of the variation in results of V. dahliae quantification among extractions of the same soil sample indicated that assaying four replicate DNA extractions for each field sample would provide accurate results. A TaqMan assay also was developed to help identify colonies of V. tricorpus on soil plates.

Since 2002, severe leaf spotting on parsley (Petroselinum crispum) has occurred in Monterey County, CA. Either of two different pathovars of Pseudomonas syringae sensu lato were isolated from diseased leaves from eight distinct outbreaks and once from the same outbreak. Fragment analysis of DNA amplified between repetitive sequence polymerase chain reaction; 16S rDNA sequence analysis; and biochemical, physiological, and host range tests identified the pathogens as Pseudomonas syringae pv. apii and P. syringae pv. coriandricola. Koch's postulates were completed for the isolates from parsley, and host range tests with parsley isolates and pathotype strains demonstrated that P. syringae pv. apii and P. syringae pv. coriandricola cause leaf spot diseases on parsley, celery, and coriander or cilantro. In a multilocus sequence typing (MLST) approach, four housekeeping gene fragments were sequenced from 10 strains isolated from parsley and 56 pathotype strains of P. syringae. Allele sequences were uploaded to the Plant-Associated Microbes Database and a phylogenetic tree was built based on concatenated sequences. Tree topology directly corresponded to P. syringae genomospecies and P. syringae pv. apii was allocated appropriately to genomospecies 3. This is the first demonstration that MLST can accurately allocate new pathogens directly to P. syringae sensu lato genomospecies. According to MLST, P. syringae pv. coriandricola is a member of genomospecies 9, P. cannabina. In a blind test, both P. syringae pv. coriandricola and P. syringae pv. apii isolates from parsley were correctly identified to pathovar. In both cases, MLST described diversity within each pathovar that was previously unknown.

Cauliflower (Brassica oleracea var. botrytis subvar. cauliflora) is susceptible to wilt caused by Verticillium dahliae but broccoli (B. oleracea var. italica subvar. cyamosa) is not. Infection of broccoli and cauliflower by a green fluorescent protein-expressing isolate of V. dahliae was examined using epifluorescence and confocal laser-scanning microscopy to follow infection and colonization in relation to plant phenology. Plant glucosinolate, phenolic, and lignin contents were also assayed at 0, 4, 14, and 28 days postinoculation. V. dahliae consistently infected and colonized the vascular tissues of all cauliflower plants regardless of age at inoculation, with the pathogen ultimately appearing in the developing seed; however, colonization decreased with plant age. In broccoli, V. dahliae infected and colonized root and stem xylem tissues of plants inoculated at 1, 2, or 3 weeks postemergence. However, V. dahliae colonized only the root xylem and the epidermal and cortical tissues of broccoli plants inoculated at 4, 5, and 6 weeks postemergence. The frequency of reisolation of V. dahliae from the stems (4 to 22%) and roots (10 to 40%) of mature broccoli plants was lower than for cauliflower stems (25 to 64%) and roots (31 to 71%). The mean level of aliphatic glucosinolates in broccoli roots was 6.18 times higher than in the shoots and did not vary with age, whereas it was 3.65 times higher in cauliflower shoots than in the roots and there was a proportional increase with age. Indole glucosinolate content was identical in both cauliflower and broccoli, and both indole and aromatic glucosinolates did not vary with plant age in either crop. Qualitative differences in characterized glucosinolates were observed between broccoli and cauliflower but no differences were observed between inoculated and noninoculated plants for either broccoli or cauliflower. However, the phenolic and lignin contents were significantly higher in broccoli following inoculation than in noninoculated broccoli or inoculated cauliflower plants. The increased resistance of broccoli to V. dahliae infection was related to the increase in phenolic and lignin contents. Significant differential accumulation of glucosinolates associated with plant phenology may also contribute to the resistant and susceptible reactions of broccoli and cauliflower, respectively, against V. dahliae.

Increasing demands for value-added salad products have triggered revolutionary changes in the production practices of vegetable salad crops in recent years. One of the pivotal changes is the adaptation of 2-m-wide beds for increased vegetable biomass per unit area. The move away from the traditional 1-m-wide raised beds in cool-season vegetable production and the associated irrigation practices potentially can have a major influence on diseases affecting cool-season vegetables. To assess the potential impacts of this shift on lettuce drop caused by Sclerotinia minor and S. sclerotiorum, the two bed widths and different irrigation frequencies within each were compared in two separate field experiments over four lettuce crops in 2 years. Treatments included 1- and 2-m bed widths with twice-weekly, weekly and biweekly drip irrigation serving as subplot treatments that were begun immediately following thinning. Incidence of lettuce drop was evaluated weekly thereafter until maturity. For S. sclerotiorum, 36 half-liter soil samples were also collected once each season and assayed for the number of sclerotia, and apothecia were counted weekly in a 10-m2 area for each plot. Regardless of the species, the effects of bed width and irrigation frequency were both significant. Twice-weekly irrigation and 2-m bed width resulted in higher lettuce drop incidence than other treatments. For S. sclerotiorum, twice-weekly irrigation and 2-m bed width also significantly increased the number of apothecia per unit area and the accumulation of soilborne sclerotia over multiple cropping seasons. Results demonstrated that the 2-m bed width combined with the practiced frequency of irrigations can result in higher lettuce drop caused by S. minor and increased incidence of airborne infection by S. sclerotiorum in the Salinas Valley where, historically, it has not been a serious threat. Increased incidence of S. sclerotiorum infection in commercial lettuce fields in the Salinas Valley between 2001 and 2006 validates these experimental results. These relatively new crop production practices can alter the balance of the two Sclerotinia spp. that has long existed in California.

In California, parsley (Petroselinum crispum [Mill.] Fuss) is a versatile leafy green commodity that is grown for fresh, frozen, dried, and processed products. In May 2017, parsley grown in a commercial field in Santa Barbara County, CA, showed symptoms of an unknown disease. Plants were stunted, wilted, and had chlorotic foliage. The large taproots were discolored, and small feeder roots were sparse and necrotic. Affected plants appeared in clusters, and disease incidence was approximately 5%. To recover possible pathogens, roots were surface sterilized in 0.01% bleach for 30 s and plated on corn meal agar amended with pimaricin, ampicillin, rifampicin, and pentachloronitrobenzene (PARP) (Kannwischer et al. 1978). After 3 days, rapidly growing, coenocytic mycelia typical of a Pythium species grew out from all roots. Older cultures showed globose sporangia and ornamented oogonia. Oogonia had an average diameter of 23 µm, and projections were conical, measuring an average length of 3 µm. Five isolates were tested using a recombinase polymerase amplification genus-specific Phytophthora assay (targeting the trnM-trnP-trnM region in the mitochondria) and were found to be negative (Miles et al. 2015). To determine the species, DNA was extracted from mycelial plugs using a QIAamp DNA Mini Kit (Qiagen, Hilden, Germany), and the internal transcribed spacer (ITS) regions 1 and 2 were amplified and sequenced using ITS1 (5′-TCCGTAGGTGAACCTGCGG-3′) and ITS4 primers (5′-TCCTCCGCTTATTGATATGC-3′) (GenBank accession no. MG593942). The sequences were 99% homologous to Pythium mastophorum strain CBS 375.72 (GenBank accession no. AY598661). To test for pathogenicity, 4-week-old plants of flat leaf (cv. Italian Dark Green Flat) and curly leaf (cv. Moss Curled) parsley were inoculated using a root-dip method for five isolates. Inoculum was made by blending five fully colonized V8 plates with 250 ml of sterile water. Parsley root balls were soaked for 10 to 15 min in the inoculum and then repotted in a 1:2 sand/soil mixture. In 2 to 3 weeks, inoculated plants became stunted and chlorotic and had symptoms that matched those seen in the field. Roots were severely decayed. Roots plated onto PARP resulted in colonies that morphologically and molecularly were identical to the original isolates. Control plants of both cultivars were inoculated with a mixture of sterile V8 media and water using the above protocol. These plants developed no foliar or root symptoms. This test was completed three times with consistent results. We believe this is the first report of P. mastophorum infecting parsley in California. P. mastophorum has been reported on parsley only in southeastern Australia (Petkowski et al. 2013). In California, this species has been reported on celery (Vazquez et al. 1996). Because affected parsley plants were completely unharvestable, this disease could be a significant concern for growers if inoculum spreads throughout this and other parsley growing regions in the state.

Arugula (Eruca sativa is a leafy crucifer that is grown in coastal California as a fresh market commodity in bagged salad mixes. In January 2016, a foliar disease developed on of commercial produced arugula in the Salinas Valley (Monterey County). Initial leaf symptoms consisted of chlorotic lesions with diffuse margins and black veins; lesions tended to originate from leaf margins and have an angular V-shape. As disease progressed, lesions became tan and dry, resulting in a papery leaf texture. Disease incidence in the affected field reached approximately 5%; however, due to quality standards, sections of the crop that exhibited any symptoms were not harvested, resulting in greater than 5% loss. Bright yellow, mucoid bacterial colonies were recovered when surface sterilized, macerated leaf tissue was streaked onto sucrose peptone agar. Five representative isolates originally from five different plants produced yellow mucoid colonies on yeast extract dextrose calcium carbonate agar and were gram-negative,...

Mustard cover crops have been suggested as a potential biofumigant for managing soilborne agricultural pests and weeds. We conducted several experiments in commercial lettuce fields in the Salinas Valley, CA, to evaluate the effects of mustard cover crops on lettuce drop caused by Sclerotinia minor and on weed density and seed viability. In a long-term study, we measured the effects of white and Indian mustard cover crops on the density of S. minor sclerotia in soil, lettuce drop incidence, weed densities, weed seed viability, and crop yield in head lettuce. We also tested broccoli and rye cover crop treatments and a fallow control. Across several short-term studies, we evaluated the density of S. minor sclerotia in soil, lettuce drop incidence, weed densities, and weed seed viability following cover cropping with a mustard species blend. Numbers of sclerotia in soil were low in most experimental locations and were not affected by cover cropping. Mustard cover crops did not reduce disease incidence in the long-term experiment but the incidence of lettuce drop was lower in mustard-cover-cropped plots across the short-term experiments. With the exception of common purslane and hairy nightshade, weed densities and weed seed viability were not significantly reduced by cover cropping with mustard. Head lettuce yield was significantly higher in mustard-cover-cropped plots compared with a fallow control. Glucosinolate content in the two mustard species was similar to those measured in other studies but, when converted to an equivalent of a commercial fumigant, the concentrations were much lower than the labeled rate for lettuce production. Although mustard cover cropping resulted in yield benefits in this study, there was little to no disease or weed suppression.

The effects of broccoli and lettuce rotations on population densities of Verticillium dahliae and Pythium spp. in soil and on strawberry (Fragaria × ananassa) growth, yield, and Verticillium wilt were evaluated in conventional and organic production systems in California for 2 years. Under both management systems, strawberry was planted after two successive crops of broccoli or lettuce. The control treatment in the conventional field was strawberry planted in soils fumigated with methyl bromide + chloropicrin. Preplant densities of V. dahliae and Pythium sp. did not differ in these fields. At the end of the second broccoli crop, V. dahliae densities in conventional plots had declined by 44% in both years. In contrast, after the second broccoli crop, densities in organic fields decreased 47% in 2000 and 25% in 2001. In general, there were no differences in V. dahliae inoculum densities in organic and conventional plots following lettuce rotations. After the second vegetable production cycle, population densities of V. dahliae in broccoli rotated organic (24 CFU/g of soil in 2000 and 27 CFU/g of soil in 2001) or conventional (23 CFU/g of soil in 2000 and 19 CFU/g of soil in 2001) fields were significantly lower than those in lettuce rotated organic (40 CFU/g of soil in 2000 and 42 CFU/g of soil in 2001) or conventional (39 CFU/g of soil in 2000 and 35 CFU/g of soil in 2001) fields. However, crop rotation treatments had no consistent effect on the inoculum densities of Pythium spp. Canopy diameters of strawberry plants grown in rotation with broccoli were not different from those in fumigated control plots, whereas those from lettuce plots were 10% smaller. Strawberry plant nutrient analysis showed that fertilizer inputs into organic or conventional production were not responsible for the observed differences in plant size. Increases in strawberry yields were not consistent between years. Verticillium wilt incidence on strawberry was 12 to 24% lower in fields rotated with broccoli compared with fields rotated with lettuce. Wilt severity on strawberry was 22 to 36% lower in fields rotated with broccoli compared with those rotated with lettuce. The strategy of using broccoli rotation coupled with postharvest incorporation of broccoli residue continues to show promise as a tool in the management of Verticillium wilt in both conventional and organic strawberry production systems.

Phytophthora ramorum has been found in potting media of containerized plants; however, the role of infested media on disease development under nursery conditions is unknown. This study assesses pathogen survival, sporulation, and infectivity to rhododendron plants in nursery pots with infected leaf litter that were maintained under greenhouse and field conditions. The influence of environmental conditions and irrigation method on disease incidence was also assessed. Infected leaf disks were buried below the soil surface of potted rhododendrons and retrieved at approximately 10-week intervals for up to 66 weeks. Pathogen survival was assessed by either isolation or induction of sporulation in water over three experimental periods. P. ramorum was recovered from infected leaf disks incubated in planted pots for longer than 1 year. Chlamydospores and sporangia formed on hydrated leaf disks but relative production of each spore type varied with the duration of incubation in soil. Root infections were detected after 40 weeks in infested soil. Foliar infections developed on lower leaves but only after spring rain events. Sprinkler irrigation promoted the development of foliar infections; no disease incidence was observed in drip-irrigated plants unless foliage was in direct contact with infested soil. Management implications are discussed.

A pear bait monitoring system was used to detect and quantify Phytophthora ramorum propagules in streams that flow through woodland areas with sudden oak death in Santa Cruz County, CA from 2001 to 2007. Stream propagules were detected most frequently or occurred in highest concentrations in winter and spring. The stream propagule concentration was characterized with statistical models using temperature and rainfall variables from 2004 to 2007. The highest concentrations of propagules occurred when stream sampling was preceded by about 2 months with low maximum daily temperatures and by 4 days with high rainfall. The occurrence of propagules in streams in the summer was mostly associated with infected leaves from the native host Umbellaria californica that prematurely abscised and fell into the water. When the stream water was used for irrigating rhododendron nursery stock from 2004 to 2007, disease occurred only three times in the two wettest springs (2005 and 2006) on plants sprinkler irrigated with stream water with relatively high concentrations of propagules. Disease incidence was described with a statistical model using the concentration of infective propagules as measured by pear baiting and consecutive hours of leaf wetness measured by electronic sensors at rhododendron height. The concentration of infective propagules was significantly reduced after water was pumped from the stream and applied through sprinklers.

Spinach downy mildew, caused by Peronospora farinosa f. sp. spinaciae, is the most economically important disease of spinach worldwide. During the past few years, spinach cultivars resistant to the seven previously described races of P. farinosa f. sp. spinaciae were observed to be severely affected by downy mildew in both the United States and the European Union. Four new isolates of P. farinosa f. sp. spinaciae were collected from California and The Netherlands and characterized based on disease reactions on two modified sets of spinach differentials. The results led to the description of three new races of the downy mildew pathogen, designated races 8, 9, and 10. Four differential cultivars with resistance to races 1 to 7 were used to distinguish the three new races. Dolphin was susceptible to races 8 and 10 but resistant to race 9; Lion was susceptible to race 10 but resistant to races 8 and 9; Lazio was resistant to races 1 to 7 as well as races 8, 9, and 10; and Tarpy was susceptible to all three new races. The three new races also were used to evaluate the disease reactions on 43 contemporary commercial spinach cultivars in greenhouse trials. A survey of 58 isolates of P. farinosa f. sp. spinaciae collected in California and Arizona between 2004 and 2006 revealed that race 10 predominated in the areas sampled.

Beginning in 2000 and continuing into 2004, a previously undescribed disease caused significant damage to romaine lettuce (Lactuca sativa) in the coastal Salinas Valley of California. Symptoms were brownish black, sunken cavities on the crown and upper taproot. Cavities were firm, lacked signs of fungal growth, and resulted in cracking and weakening of the crown. Affected plants were stunted, uneven in growth, and unmarketable. Crisphead lettuce also developed these symptoms, although disease incidence was always lower than that for romaine. A fungus was consistently isolated from cavities. Using morphological features and molecular methods, the fungus was identified as Phoma exigua. Pathogenicity of isolates from romaine and crisphead lettuce was demonstrated on both of these lettuce types. Replicated field studies showed that azoxystrobin, boscalid, and cyprodinil + fludioxonil fungicides effectively controlled the disease. This is the first report of a crown disease caused by P. exigua in the United States. A similar disease, Phoma basal rot on greenhouse grown lettuce in the United Kingdom, had been previously observed, but no report has been published.

Morphology, life cycle biology, and DNA sequence analysis of rust fungi on garlic and chives from California. Phytopathology 94:569-577. In the late 1990s, commercial garlic fields in California (CA) were devastated by an outbreak of rust caused by Puccinia allii. We compared collections of the pathogen from garlic (Allium sativum) and chives (A. schoenoprasum) in central CA and Oregon (OR) to collections from garlic and leek (A. porrum and A. ampeloprasum) in the Middle East. Teliospores from the CA and OR collections were smaller in length, width, and projected cross-sectional area compared with collections from the Middle East. CA and OR collections had a shortened life cycle, in which pycnia and aecia were not formed. Germinating teliospores produced a two-celled promycelium, resulting in two basidiospores, each initially with two nuclei, indicating that this rust was homothallic. In addition, the morphology of the substomatal vesicles was different between the CA-OR (fusiform) and the Middle Eastern (bulbous) collections. DNA sequence analysis of the nuclear ribosomal internal transcribed spacer region showed that the CA and OR rust collections formed a well-supported cluster distinct from the Middle Eastern and European samples. These results suggest that the rust on garlic and chives in CA and OR is a different species than the rust fungus on garlic and leek in the Middle East. Since 1996, commercial spinach cultivars with resistance to four previously described races of Peronospora farinosa f. sp. spinaciae (races 1, 2, 3, and 4) were observed with high incidences of downy mildew both in California and Europe. Isolates of P. farinosa f. sp. spinaciae collected in California between 1997 and 2001, Arizona in 1999, and a single isolate collected in the Netherlands in 1996 were examined for their disease reaction on differential spinach cultivars and a set of commercial spinach cultivars. Disease reactions on the differential cultivars indicated the occurrence of three new races of P. farinosa f. sp. spinaciae. Two newly identified races, designated race 5 (isolate CA1) and race 6 (isolate SP1), were detected in the United States. The isolate from the Netherlands also was distinct and designated race 7 (isolate JVN7). Some cultivars with resistance to races 1, 2, 3, and 4 were susceptible to race 5, whereas others were resistant, indicating that resistance to a given race may be governed by different genes (or alleles) depending on the source of resistance. A survey of races in California indicated that races 5 and 6 predominated. Although the majority of the cultivars examined were susceptible to race 6 based on the traditional qualitative cotyledon inoculation assay, significant quantitative differences in resistance to race 6 were observed using a true-leaf greenhouse screening procedure. Although more work is needed to confirm the results of the true-leaf assays, the quantitative resistance observed using this procedure appears to be race specific.

Field experiments were conducted at Spence Road site and at the Hartnell College East Campus site in Salinas, CA, to determine the effects of crop rotation with broccoli or a fallow period on lettuce drop caused by Sclerotinia minor and the density of pathogen sclerotia in the soil. Treatments at the Spence Road site with low inoculum density (3 of soil) distributed randomly included: successive crops of lettuce (LLL), lettuce rotated with broccoli (LBL), and lettuce followed by a fallow period (LFL). Treatments at the Hartnell site with high inoculum density (>7 sclerotia per 100 cm3 of soil) distributed uniformly included: continuous lettuce (LLLL), broccoli-lettuce-broccoli-lettuce (BLBL), broccoli-broccoli-lettuce-lettuce (BBLL), and fallow-lettuce-fallow-lettuce (FLFL). At the Spence Road site, continuous lettuce did not increase lettuce drop incidence for at least 2 years, although an increase in soilborne sclerotia was observed annually but was below the threshold at which a correlation between inoculum density and disease incidence is observed. Rotation with broccoli resulted in small reductions in disease incidence only in the first year. The density of sclerotia was lowest in the LFL treatment, and the highest in the LLL. At the Hartnell site, rotation with broccoli significantly reduced both sclerotia and lettuce drop incidence. The number of broccoli crops rather than the sequence of lettuce rotations with broccoli was critical for reducing the numbers of S. minor sclerotia in soil. Fallowing after a lettuce crop resulted in marginal reductions in sclerotia and lettuce drop incidence. Viability of recovered sclerotia was not significantly different between treatments, although differences between seasons were detected. Results suggest that rotations with broccoli can be a practical lettuce drop management strategy.

A new canker disease, commonly known as sudden oak death, of Lithocarpus densiflorus, Quercus agrifolia, Q. kelloggii, and Q. parvula var. shrevei in California is shown to be caused by Phytophthora ramorum. The pathogen is a recently described species that previously was known only from Germany and the Netherlands on Rhododendron spp. and a Viburnum sp. This disease has reached epidemic proportions in forests along approximately 300 km of the central coast of California. The most consistent and diagnostic symptoms on trees are cankers that develop before foliage symptoms become evident. Cankers have brown or black discolored outer bark and seep dark red sap. Cankers occur on the trunk at the root crown up to 20 m above the ground, but do not enlarge below the soil line into the roots. Individual cankers are delimited by thin black lines in the inner bark and can be over 2 m in length. In L. densiflorus saplings, P. ramorum was isolated from branches as small as 5 mm in diameter. L. densiflorus and Q. agrifolia were inoculated with P. ramorum in the field and greenhouse, and symptoms similar to those of naturally infected trees developed. The pathogen was reisolated from the inoculated plants, which confirmed pathogenicity.

Schoch, C. L., Crous, P. W., Polizzi, G., and Koike, S. T. 2001. Female fertility and single nucleotide polymorphism comparisons in Cylindrocladium pauciramosum. Plant Dis. 85:941-946. Cylindrocladium pauciramosum is well established in South America, and has recently been collected from nurseries in South Africa, Italy, and the United States. Isolates were compared with respect to the percentages of hermaphrodites and the respective mating types in the different samples. Based on these data, the effective population size could be determined for the different areas studied. All nurseries had mating type ratios significantly different from an idealized 1:1 ratio. In the South African nursery, the MAT-1 mating type was dominant, while the MAT-2 mating type dominated in other samplings. This is consistent with an introduction of a small starter population. High percentages of hermaphrodites also agreed with recent introductions into nurseries in Italy and the United States. Variability of DNA sequences of the 5′ end of the β-tubulin gene from a set of C. pauciramosum isolates from different geographic regions was low to high. Isolates from South Africa, the United States, and Australia had identical βtubulin DNA sequences; this sequence was also found in the Italian sample, along with another unique group. Finally, a group of isolates obtained from South and Central America had the highest variation of all isolates investigated, and also included isolates that shared single nucleotide variations with another species, C. candelabrum. These findings suggest that C. pauciramosum most likely has a Central or South American center of origin.

Lettuce growers in coastal California have relied mainly on protective fungicide sprays to control downy mildew. Thus, timing of sprays before infection is critical for optimal results. A leaf-wetness-driven, infection-based advisory system, previously developed, did not always perform satisfactorily. In this study, the advisory system was modified by incorporating a pathogen survival component (system 1) or both survival and sporulation components (system 2). These systems were then evaluated in commercial lettuce fields in coastal California during 1996-1998. Three or four treatments were carried out in each field: (i) no spray; (ii) sprays as scheduled by the growers; (iii) sprays following modified system 1; and (iv) sprays following the original advisory system (1996) or modified system 2 (1998). Downy mildew incidence was evaluated every 2 to 9 days. In fields with drip irrigation, the number of fungicide applications was reduced by one or two regardless of the advisory system used compared to the grower’s calendar-based schedule, although one unnecessary spray was recommended in 1996 at Soledad and 1997 at Salinas. In 1998, a devastating outbreak of rust disease severely damaged the garlic crop in California, resulting in yield losses of 51% and an economic loss of 27% to the industry. The disease also occurred in 1999 and 2000, indicating that rust may have become an annual problem in some parts of the state. The presence of urediniospores, two-celled teliospores, and telial paraphyses indicated that the pathogen was Puccinia allii. Isolates from garlic infected onion and chives, but not leek, elephant garlic, or shallot in inoculation experiments. Garlic cloves obtained from diseased plants were planted under controlled conditions, but the resulting plants did not develop rust. Fungicide trials were conducted for 3 years and showed that none of the currently registered materials gave satisfactory control. However, tebuconazole and azoxystrobin provided good protection against rust if sprayed at 10-day intervals. A variety trial of 34 garlic cultivars and selections was planted, inoculated, and evaluated for resistance to rust. Although there was variability in rust severity among the selections, acceptable levels of resistance were not observed in any cultivar.

Wilt incited by Verticillium dahliae has recently become an important disease on cauliflower in the Salinas Valley. Although broccoli is closely related to cauliflower, wilt has not occurred on this host and V. dahliae isolates from cauliflower were weakly pathogenic on broccoli in greenhouse inoculations. In this study, the effectiveness of broccoli residues on propagule attrition in soil and wilt incidence on cauliflower was determined in a commercial field infested with V. dahliae microsclerotia. The treatments were broccoli residue with tarp, broccoli residue without tarp, chloropicrin, metham sodium, control with tarp, control without tarp, cauliflower followed by cauliflower, and cauliflower followed by fallow. Approximately 200 kg of chopped broccoli was uniformly disk incorporated into the corresponding plots (36 m2). Densities of V. dahliae microsclerotia were determined at 0, 30, 90, and 145 days after treatment in 1993 and 0, 30, 74, 109, and 140 days after treatment in 1994 using the modified Anderson sampler technique. Cauliflower cultivar White Rock was planted in all plots. Plant height, number of marketable heads, head weight, and wilt severity were determined at maturity. Even though the pre-treatment number of V. dahliae propagules in broccoli-treated and control plots were similar, by the end of the cauliflower cropping season numbers either were the same or increased in control plots compared with a reduction in numbers in broccoli residue-treated plots during both seasons. There were no differences between tarped and non-tarped plots either in broccoli residue-treated or control plots. In fumigated plots, propagules declined initially but later returned to pre-treatment levels by the end of the cropping season. Continuous cauliflower or fallowing after one cauliflower crop resulted in stable or increased levels of microsclerotia. Verticillium wilt severity was lowest in metham sodium-treated plots, followed by broccoli residue-treated plots and chloropicrin in both years. Plant height, marketable heads, and head weight were higher in broccoli-treated than in control plots. These results suggest that broccoli residues reduce V. dahliae microsclerotia in soil and wilt of cauliflower as much as or more than chloropicrin and metham sodium, and that rotation with broccoli may be a feasible approach to manage Verticillium wilt in cauliflower and other susceptible crops.

During 1996 and 1997, a new and damaging disease of leek (Allium porrum) was observed on greenhouse-produced transplants and field-grown plants in California. Symptoms were water-soaked lesions at leaf tips, which eventually expanded down the length of the leaf and resulted in brown, elongated, stripe-like lesions with yellow margins. Diseased leaves eventually wilted. A blue fluorescent pseudomonad was consistently recovered from lesions, and biochemical and physiological tests indicated that it was Pseudomonas syringae. Pathogenicity tests established that representative strains of this P. syringae induced disease symptoms in leek that were similar to those observed on leek plants in the greenhouse and field, and that this bacterium caused similar symptoms in onion, chives, and garlic plants. Representative strains were further characterized by fatty acid analysis, repetitive bacterial sequence-polymerase chain reaction (rep-PCR), and rDNA sequencing. Fatty acid analysis confirmed that these isolates were P. syringae, but did not provide a clear pathovar designation. Rep-PCR analysis revealed that all the California leek P. syringae strains had identical DNA fingerprints and that these strains were indistinguishable from those of known strains of P. syringae pv. porri. In addition, the rDNA sequence of the spacer region between 16S and 23S rDNA genes was identical among the California leek P. syringae strains and P. syringae pv. porri. Together, these results established that the new leek disease in California is caused by P. syringae pv. porri. P. syringae pv. porri was recovered from a commercial leek seed lot imported into California, which suggests that the pathogen was introduced in association with seed. Commercial leek production in California is favorable for development of this disease because transplants are produced in greenhouses with high plant densities, overhead irrigation, and mowing of plants.

Experiments were conducted in field plots to evaluate the effects of broccoli residue on population dynamics of Verticillium dahliae in soil and on Verticillium wilt development on cauliflower under furrow and subsurface-drip irrigation and three irrigation regimes in 1994 and 1995. Treatments were a factorial combination of three main plots (broccoli crop grown, harvested, and residue incorporated in V.dahliae-infested plots; no broccoli crop or residue in infested plots; and fumigated control plots), two subplots (furrow and subsurface-drip irrigation), and three sub-subplots (deficit, moderate, and excessive irrigation regimes) arranged in a split-split-plot design with three replications. Soil samples collected at various times were assayed for V. dahliae propagules using the modified Anderson sampler technique. Incidence and severity of Verticillium wilt on cauliflower were assessed at 7- to 10-day intervals beginning a month after cauliflower transplanting and continuing until harvest. Number of propagules in all broccoli plots declined significantly (P < 0.05) after residue incorporation and continued to decline throughout the cauliflower season. The overall reduction in the number of propagules after two broccoli crops was approximately 94%, in contrast to the fivefold increase in the number of propagules in infested main plots without broccoli after two cauliflower crops. Disease incidence and severity were both reduced approximately 50% (P < 0.05) in broccoli treatments compared with no broccoli treatments. Differences between furrow and subsurface-drip irrigation were not significant, but incidence and severity were significantly (P < 0.05) lower in the deficit irrigation regime compared with the other two regimes. Abundance of microsclerotia of V. dahliae on cauliflower roots about 8 weeks after cauliflower harvest was significantly (P < 0.05) lower in treatments with broccoli compared with treatments without broccoli. Rotating broccoli with cauliflower and incorporating broccoli residues into the soils is a novel means of managing Verticillium wilt on cauliflower and perhaps on other susceptible crops. This practice would be successful regardless of the irrigation methods or regimes followed on the susceptible crops.

Lettuce mosaic potyvirus (LMV) outbreaks in the Salinas Valley of California have increased in recent years despite a comprehensive lettuce mosaic management program. In a previous study, it was established that new serologically and/or genetically distinct LMV strains were not responsible for these outbreaks. Therefore, the role of alternate hosts was investigated. Here we provide evidence that the ornamental plant, Gazania spp., is an alternate host of LMV. A number of lines of evidence were used to establish that gazanias were infected by LMV, including indirect enzyme-linked immunosorbent assay, dot blot hybridization with an LMV coat protein (CP) probe, and sap- and aphid-transmission of LMV from gazania to lettuce and Nicotiana benthamiana. Additionally, an LMV CP fragment was amplified from an infected gazania plant by reverse transcription-polymerase chain reaction, and the nucleotide sequence of this fragment was 99% identical to the CP sequence of an LMV isolate from the Salinas Valley. We further show that LMV-infected gazanias are widely distributed in the Salinas Valley, and are present near areas where LMV outbreaks have occurred. Field experiments demonstrated that LMV could be transmitted from infected gazania to lettuce. LMV-infected gazanias also were found in local nurseries, suggesting that infection of gazania with LMV had occurred prior to planting in the Salinas Valley (i.e., lettuce was not serving as the source of inoculum for infection of gazania in the Salinas Valley). These results establish that gazania is an alternate host of LMV, and suggest that gazanias may be a primary inoculum source for recent lettuce mosaic outbreaks in the Salinas Valley.

Cucurbit downy mildew (CDM), caused by Pseudoperonospora cubensis, is one of the most important diseases affecting cucurbits worldwide. In the USA, host resistance in cucumber had adequately controlled the disease with very minimal application of fungicides from the late 1960s to 2004. In 2004, there was a resurgence of the disease that devastated the cucumber crop in several states in the eastern USA. Since then, host plant resistance alone has not been sufficient to adequately control the disease and now control relies heavily on application of fungicides. To effectively apply fungicides in a timely manner, cucurbit growers, extension personnel, and crop consultants and advisors can now utilize information on disease occurrence and predicted spread disseminated through the United States Department of Agriculture's CDM ipmPIPE decision support system developed by scientists at North Carolina State University. Based on a survey conducted in Georgia, North Carolina, and Michigan, the CDM ipmPIPE resulted in an average reduction of 2 to 3 fungicide applications in 2009 compared to calendar-based fungicide sprays. With approximately 122,000 acres of cucurbits in these three states, this translates to more than $6 million in savings to the producers in these three states. Economic savings and positive environmental implications of reduced fungicide applications demonstrate the value of a coordinated national monitoring network for management of a plant disease that is disseminated aerially over long distances. Xanthomonas leaf spot is a new disease that has occurred on catnip (Nepeta cataria). Catnip is grown commercially in California for use as herbs, seasonings, and tea. This disease has developed recently on catnip transplants that are produced in enclosed greenhouses. Symptoms consist of small brown flecks that are visible from both sides of a leaf. The flecks later develop into larger, dark brown, angular leaf spots. Severe infection reduced the quality and marketability of the transplants. Xanthomonas campestris, as identified by biochemical, physiological, and molecular tests, was consistently isolated from symptomatic plants, and selected strains caused similar symptoms when inoculated onto catnip test plants. However, catnip strains failed to cause any symptoms when inoculated onto nine other plants in the Lamiaceae family and five other hosts of known X. campestris pathovars. Catnip plants showed no symptoms when inoculated with X. campestris pvs. campestris, carotae, and vesicatoria. Catnip also was not susceptible to the X. campestris pathogen isolated from lavender. This is the first report of a bacterial disease of catnip caused by a Xanthomonas pathogen, and the catnip strains may be a new and distinct pathovar of X. campestris.

In the summer of 2009, a leaf spot disease occurred on 100% of Gaillardia × grandiflora cv. Goblin in a commercial nursery in coastal Monterey Co., CA. Nearly all of the affected plants were unsalable. The causal agent was determined to be Entyloma gaillardianum based on morphological features, host, and ITS region. This species has not been reported previously from this host in North America. Accepted for publication 16 March 2010. Published 28 April 2010.

The causal agent was identified as Entyloma australe Speg. based on host and on pathogen morphology and ITS region sequence. The size, shape, and color of teliospores and the production of fascicles of conidiophores fit the descriptions for E. australe. This appears to be the only Entyloma species occurring on Physalis. Poison-hemlock is a widely distributed biennial especially known for its toxic piperidine alkaloids that can poison livestock that eat this plant. In 2008, a powdery mildew was observed on poison-hemlock growing in coastal California. The causal agent was determined to be Erysiphe heraclei DC., previously unreported on this host in North America. Accepted for publication 18 December 2008. Published 12 March 2009.

Fungicides were evaluated for pre- and post-infection control of ramorum blight, caused by Phytophthora ramorum, on Rhododendron cvs. Cunningham's White and Irish Lace, Camellia japonica, Pieris japonica, and Viburnum tinus. Cyazofamid, dimethomorph, mefenoxam, pyraclostrobin, and fenamidone applied as foliar sprays consistently provided preventative control as indicated by reduced lesion size compared to water controls. These fungicides provided preventative activity for at least 28 days in the tested species except in Rhododendron where fungicides were active for at least 14 days following application. With preventative fungicide applications, the pathogen was recovered from most fungicide-treated leaves by isolation onto selective media. Dimethomorph consistently reduced the percent recovery from diseased Rhododendron leaves. With post-infection treatments, the fungicides did not significantly reduce lesion growth and percent recovery of the pathogen. The pathogen was recovered from lesions consistently at least 6 weeks after fungicide application in Rhododendron regardless of treatment on intact and fallen diseased leaves. However, the cultures resulting from isolations of diseased tissue treated with cyazofamid and dimethomorph were significantly slower growing than those cultures from other fungicide treatments. During the summer of 2007 the authors encountered a previously unrecorded powdery mildew disease on several plants of the perennial ornamental plant Stachys byzantina K. Koch ex Scheele (common names: woolly hedgenettle, woolly betony, and lamb's ear) growing in a plant bed near a commercial building in Salinas (Monterey Co.), California. This report documents the occurrence of Neoerysiphe galeopsidis on this host in North America. Accepted for publication 11 October 2007. Published 17 December 2007.

Description and first report of powdery mildew caused by Podosphaera epilobii on E. ciliatum in North America are presented in this brief. The weed is commonly found in California's Salinas Valley (Monterey Co.). Confirmation of the powdery mildew disease indicates that this weed will not be a source of the powdery mildew diseases that affect nearby vegetable and ornamental crops. Lettuce plants were artificially inoculated with three isolates of Xanthomonas campestris pv. vitians in field and greenhouse evaluations for genetic variation in resistance to bacterial leaf spot. The cultivar Little Gem had the least amount of disease, whether evaluated for disease severity or disease incidence. Disease severity levels for cultivar Batavia Reine des Glaces were not significantly different from those of Little Gem. Several cultivars including Vista Verde, Pybas 251, Pacific, Clemente, Salinas 88, and Sniper were highly susceptible. There was a strong correlation between results obtained in the greenhouse and those obtained in the field (r = 0.77, P = 0.009) indicating that greenhouse assays may be used to increase the number of cultivars evaluated and to speed breeding efforts. The efficacy of Serenade, Maneb, Cuprofix, and mixtures of these products were evaluated for efficacy in reducing disease incidence and severity of bacterial leaf spot of lettuce caused by Xanthomonas campestris pv. vitians in six trials at two locations between 2001 and 2003. Additionally, Messenger, elemental copper, Quadris, and Actigard were evaluated in at least two of these trials. Mixtures of Serenade and Cuprofix reduced disease incidence in three of six experiments and reduced disease severity in a fourth experiment. Treatment with the Maneb and Cuprofix mixture was less effective but resulted in significantly less disease and lower severity in single experiments. These results indicate that a mixture of Serenade and Cuprofix may be a useful addition to an integrated bacterial leaf spot disease management strategy but this treatment may be too variable to justify it as a stand-alone treatment.
