The PSYCHE method for pure shift NMR is exploited to generate 2D J spectra with full decoupling in one dimension and multiplet structure in the other, allowing spin–spin coupling constants to be measured even in very crowded spectra. A global and accurate mapping of the environment could be achieved if sensors and indicators are mass-produced at low cost. Printed electronics using polymeric (semi)conductors offer a platform for such sensor/indicator based circuits. Herein, we present the material concept for an electrochemical pH-threshold indicator based on a printable hybrid electrode which comprises a liquid metal alloy (GaInSn) embedded in a conducting polymer matrix (PEDOT). This hybrid electrode displays a large variation in open circuit potential versus pH in an electrochemical cell, which when connected to the gate of an electrochemical transistor leads to a dramatic change in the drain current in a narrow range of pH. Fully homodecoupled HSQC spectra can be obtained through the use of a new pulse sequence element, "perfectBIRD". By way of illustration, we show that perfectBIRD decoupling allows one-bond residual dipolar couplings (RDCs), which provide important NMR restraints for structure elucidation, to be measured with outstanding precision, even in methylene groups. The water soluble tetradentate Schiff base, N,N -bis(5-sulfonato-salicylidene)-diaminoethane (H2salen-SO3), will readily coordinate to the uranyl(VI) cation, but not to the same extent to trivalent anthanide cations. Molecular transport is an important aspect in metal-organic frameworks (MOFs) as it affects many of their applications, such as adsorption/separation, drug delivery and catalysis. Yet probing the fundamental diffusion mechanisms in MOFs is challenging, and the interplay between the MOF's features (such as the pore structure and linker dynamics) and molecular transport remains mostly unexplored. Here, the pulsed-field gradient nuclear magnetic resonance (PFG NMR) technique is used to probe the diffusion of several probe molecules, i.e. , water, xylenes and 1,3,5-triisopropylbenzene (TIPB), within the UiO-66 MOF and its derivatives (UiO-66NH 2 and UiO-66Br). Exploiting differences in the size of probe molecules we were able to probe the diffusion rate selectively in the different pore environments of the MOFs. In particular, when relatively small molecules, such as water and small hydrocarbons, were used as probes, the PFG NMR log attenuation plots were non-linear with two distinctive diffusion regions, suggesting faster diffusion in the inter-crystalline space and slower diffusion within crystal aggregates, the latter occurring mostly inside the framework of the MOFs. Conversely, experiments with a larger probe molecule, i.e. , TIPB, with a kinetic diameter of 0.95 nm, which makes it unable to access the framework windows of the MOF crystals, showed linear PFG NMR log attenuation plots, which indicates diffusion occurring in a single environment, most likely in the inter-crystalline space. Analysis of the apparent tortuosity values of the systems under investigation highlights the role of linker functionalisation in influencing the molecular diffusion of the probe molecules, which affects both intra-molecular interactions and pore accessibility within the MOF crystals. The findings of this work demonstrate that the diffusion behaviour of probe molecules within MOFs is influenced by the pore size, structure, functionalisation of the MOF linker and molecular interactions. Our study contributes to further advance the understanding of mass transport in MOFs by PFG NMR and provides insights that can inform the design and optimisation of MOF-based materials for various applications.

Practical pure shift NMR experiments, especially on instruments equipped with cryoprobes, can sometimes give very disappointing results. Here we show for the first time that this is a consequence of signal loss due to sample convection, and demonstrate a simple adjustment to common pure shift NMR experiments that restores the lost signal.

Fluorine is becoming increasingly prevalent in medicinal chemistry, both in drug molecules and in molecular probes. The presence of fluorine allows convenient monitoring of such molecules in complex environments by NMR spectroscopy. However, sensitivity is a persistent limitation of NMR, especially when molecules are present at low concentrations. Here, sensitivity issues with 1 H NMR are mitigated by sharing 19 F photochemically-induced dynamic nuclear polarisation with 1 H nuclei. Unlike direct 1 H enhancement, this method enhances 1 H signals without significantly distorting multiplet intensities, and has the potential to enable the use of suitable molecules as low-concentration probes.

Selective 1D COSY can unambiguously identify coupled spins but is often limited both by lack of selectivity, and by unfavourable multiplet lineshapes. Here, ultra-selective GEMSTONE excitation is employed with CLIP-COSY to provide through-bond correlations for nuclei whose NMR signals overlap. The new method is illustrated using the coccidiostat lasalocid and the immunosuppressant cyclosporin.

Human milk oligosaccharides belong to an important class of bioactive molecules with diverse effects on the development of infants. NMR is capable of providing vital structural information about oligosaccharides which can aid in determining structure-function relationships. However, this information is often concealed by signal overlap in 1 H spectra, due to the narrow chemical shift range and signal multiplicity. Signal overlap in oligosaccharide spectra can be greatly reduced, and resolution improved, by utilising pure shift methods. Here the benefits of combining pure shift methods with the CASPER computational approach to resonance assignment in oligosaccharides are demonstrated.

An ultra-selective 1D NMR experiment - GEMSTONE-ROESY - enables clear, unambiguous assignment of ROE signals in the not uncommon situation that traditional selective methods fail. Its usefulness is demonstrated in the analysis of the natural products cyclosporin and lacto- N -difucohexaose I, providing detailed insight into the structures and conformations of these molecules.

By screening of a collection of 50 000 small-molecule compounds, we recently identified 4-arylazo-3,5-diamino-1 H -pyrazoles as a novel group of anti-biofilm agents. Here, we report a SAR study based on 60 analogues by examining ways in which the pharmacophore can be further optimized, for example, via substitutions in the aryl ring. Accurate measurement of transverse relaxation rates in coupled spin systems is important in the study of molecular dynamics, but is severely complicated by the signal modulations caused by scalar couplings in spin echo experiments. The most widely used experiments for measuring transverse relaxation in coupled systems, CPMG and PROJECT, can suppress such modulations, but they also both suppress some relaxation contributions, and average relaxation rates between coupled spins. Here we introduce a new experiment which for the first time allows accurate broadband measurement of transverse relaxation rates of coupled protons, and hence the determination of exchange rate constants in slow exchange from relaxation measurements. The problems encountered with existing methods are illustrated, and the use of the new method is demonstrated for the classic case of hindered amide rotation and for the more challenging problem of exchange between helical enantiomers of a gold(i) complex., Competing Interests: There are no conflicts to declare., (This journal is © The Royal Society of Chemistry.)

Selective 1D TOCSY is a powerful tool in the assignment of NMR spectra of organic molecules. Here an order of magnitude improvement in selectivity, allowing overlapping multiplets to be excited separately, is achieved in a single scan using the very recent GEMSTONE method. The new experiment is illustrated using an antibiotic and a mixture of diastereomers.

Liquid biopsy holds promise towards practical implementation of personalized theranostics of cancer. In particular, circulating tumour cells (CTCs) can provide clinically actionable information that can be directly linked to prognosis or therapy decisions. In this study, gene expression patterns and genetic mutations in single CTCs are simultaneously analysed by strategically combining microfluidic technology and in situ molecular profiling technique. Towards this, the development and demonstration of the OPENchip (On-chip Post-processing ENabling chip) platform for single CTC analysis by epithelial CTC enrichment and subsequent in situ molecular profiling is reported. For in situ molecular profiling, padlock probes that identify specific desired targets to examine biomarkers of clinical relevance in cancer diagnostics were designed and used to create libraries of rolling circle amplification products. We characterize the OPENchip in terms of its capture efficiency and capture purity, and validate the probe design using different cell lines. By integrating the obtained results, molecular analyses of CTCs from metastatic breast cancer (HER2 (ERBB2) gene expression and PIK3CA mutations) and metastatic pancreatic cancer (KRAS gene mutations) patients were demonstrated without any off-chip processes. The results substantiate the potential implementation of early molecular detection of cancer through sequencing-free liquid biopsy.

The extractability and coordination chemistry for lanthanides in a binary extraction system containing aerosol OT (AOT) and a phosphine oxide (CMPO) have been investigated by means of UV-Vis and X-ray absorption fine structure (XAFS) techniques over a range of extractant concentrations. In addition to the Fourier transform EXAFS analysis, the application of wavelet transform (WT) to the Fourier transform was adapted to the Ho L-III edge to provide a better contrast between the heavier and lighter backscattering atoms at higher coordination shells in the mixed extractant system. Through the variation of the ratio of AOT and CMPO and the acid concentration of the aqueous phase, a synergistic effect appeared at a 1 : 1 ratio of AOT and CMPO using a moderate acid concentration (0.5 M HNO 3 ). It was also found that CMPO itself is directly coordinated with the metal ions by forming the bond with P[double bond, length as m-dash]O and C[double bond, length as m-dash]O, while AOT extracts the lanthanide as a hydrated form through reverse micelle formation. However, the complex of CMPO (C[double bond, length as m-dash]O-M and P[double bond, length as m-dash]O-M) to metal becomes weaker as it is mixed with AOT. Our results indicate that complexation from the CMPO is aiding the control of water extraction through reverse micelles from AOT, which results in the cooperative effect of certain mixtures of AOT-CMPO. The information from this coordination environment offers insight into the role of reverse micelle formation in a mixed-extractant solvent extraction system.

"Pure shift" methods can greatly improve the resolution of proton NMR spectra. However, current pure shift spectra show small periodic artefacts that prevent their use for studying dilute mixture components. A new technique, compatible with all current pure shift methods, is presented that suppresses such sidebands to arbitrary order, allowing ultraclean spectra to be obtained.

The coordination of the trivalent 4f ions, Ln = La 3+ , Dy 3+ , and Lu 3+ , with neutral and acidic organophosphorus reagents, both individually and combined, was studied by use of X-ray absorption spectroscopy. These studies provide metrical information about the interatomic interactions between these cations and the ligands tri-n-butyl phosphate (TBP) and di-n-butyl phosphoric acid (HDBP), whose behavior are of practical importance to chemical separation processes that are currently used on an industrial scale. Previous studies have suggested the existence of complexes involving a mixture of ligands, accounting for extraction synergy. Through systematic variation of the aqueous phase acidity and extractant concentration and combination, we have found that complexes with Ln and TBP : HDBP at any mixture and HDBP alone involve direct Ln-O interactions involving 6 oxygen atoms and distant Ln-P interactions involving on average 3-5 phosphorus atoms per Ln ion. It was also found that Ln complexes formed by TBP alone seem to favor eight oxygen coordination, though we were unable to obtain metrical results regarding the distant Ln-P interactions due to the low signal attributed to a lower concentration of Ln ions in the organic phases. Our study does not support the existence of mixed Ln-TBP-HDBP complexes but, rather, indicates that the lanthanides are extracted as either Ln-HDBP complexes or Ln-TBP complexes and that these complexes exist in different ratios depending on the conditions of the extraction system. This fundamental structural information offers insight into the solvent extraction processes that are taking place and are of particular importance to issues arising from the separation and disposal of radioactive materials from used nuclear fuel.

A new method for the analysis of low level impurities in sparsely fluorinated species allows measurement of clean high dynamic range 19 F spectra, fully decoupled and free of interfering signals from 13 C isotopomers.

Conventional solvent extraction of selected f-element cations by bis(2-ethylhexyl)phosphoric acid (HDEHP) yields increased extraction from aqueous to organic solution along the series Np(v) < Cm(iii) < Eu(iii) < U(vi), with distribution ratios all within two orders of magnitude. However, in the presence of the water-soluble tetradentate Schiff base (N,N'-bis(5-sulfonatosalicylidene)-ethylenediamine or H 2 salenSO 3 ), selective complexation of the two actinyl cations (Np(v) and U(vi)) resulted in an extraction order of Np(v) < U(vi) ≪ Eu(iii) < Cm(iii). The extraction of neither Cm(iii) or Eu(iii) by HDEHP are significantly impacted by the presence of the aqueous phase Schiff base. Despite observed hydrolytic decomposition of H 2 salenSO 3 in aqueous solutions, the calculated high conditional stability constant (β 11 = 26) for the complex [UO 2 (salenSO 3 )] 2- demonstrates its capacity for aqueous hold-back of U(vi). UV-visible-NIR spectroscopy of solutions prepared with a Np(vi) stock and H 2 salenSO 3 suggest that reduction of Np(vi) to Np(v) by the ligand was rapid, resulting in a pentavalent Np complex that was substantially retained in the aqueous phase. Results from 1 H NMR of aqueous solutions of H 2 salenSO 3 with U(vi) and La(iii), Eu(iii), and Lu(iii) provides additional evidence that the ligand readily chelates U(vi), but has only weak interactions with trivalent lanthanide ions.

A new pulse sequence, CHORUS Oneshot, allows measurements of diffusion-ordered spectroscopy (DOSY) spectra over the full chemical shift range of (19)F for the first time. Swept-frequency pulses are used to give very broadband excitation; the sequence is a prototype for a large family of very broadband liquid state NMR methods.

We demonstrate a novel digital microfluidic nucleic acid amplification concept which is based on padlock probe mediated DNA detection and isothermal circle-to-circle amplification (C2CA). This assay platform combines two digital approaches. First, digital microfluidic manipulation of droplets which serve as micro-reaction chambers and shuttling magnetic particles between these droplets facilitates the integration of complex solid phase multistep assays. We demonstrate an optimized novel particle extraction and transfer protocol for superparamagnetic particles on a digital microfluidic chip that allows for nearly 100% extraction efficiencies securing high assay performance. Second, the compartmentalization required for digital single molecule detection is solved by simple molecular biological means, circumventing the need for complex microfabrication procedures necessary for most, if not all, other digital nucleic acid detection methods. For that purpose, padlock probes are circularized in a strictly target dependent ligation reaction and amplified through two rounds of rolling circle amplification, including an intermediate digestion step. The reaction results in hundreds of 500 nm sized individually countable DNA nanospheres per detected target molecule. We demonstrate that integrated miniaturized digital microfluidic C2CA results in equally high numbers of C2CA products μL(-1) as off-chip tube control experiments indicating high assay performance without signal loss. As low as 1 aM synthetic Pseudomonas aeruginosa DNA was detected with a linear dynamic range over 4 orders of magnitude up to 10 fM proving excellent suitability for infectious disease diagnostics.

An NMR method is reported that allows diastereomeric ratios to be determined even in crowded spectra or where chemical shift differences are small compared to multiplet widths. Band-selective pure shift NMR collapses multiplets to singlets, greatly improving spectral resolution while largely retaining, or even enhancing, signal-to-noise ratio.

Diffusion-ordered spectroscopy (DOSY) is one of the most powerful methods for intact mixture analysis by NMR. However, the separation of overlapped spectra by current DOSY methods typically requires a minimum of 30% difference in diffusion coefficient. Here we present a new algorithm (OUTSCORE) that can improve the situation by almost an order of magnitude, allowing the unmixing of severely overlapped species of similar size, by combining least squares fitting with cross-talk minimisation, maximising spectral difference.

A simple modification of the WATERGATE solvent suppression method greatly improves the quality of (1)H NMR spectra obtainable from samples in H(2)O. The new method allows (1)H signals to be measured even when close in chemical shift to the signal of water, as for example in the NMR spectra of carbohydrates.

The spin echo is the single most important building block in modern NMR spectroscopy, but echo modulation by scalar couplings J can severely complicate its use. High resolution diffusion-ordered NMR spectroscopy allows the separation of signals from different species based on their diffusion coefficients. In general this requires that the NMR spectra of the components do not have overlapping signals, and that the diffusion coefficients are significantly different. Modifying the solvent matrix in which a sample is dissolved can change the diffusion coefficients observed, allowing resolution ("matrix-assisted DOSY"). We show here that dissolving the two naturally-occurring epimers of naringin in an aqueous solution of β-cyclodextrin causes both shift and diffusion changes, allowing the signals of the epimers to be distinguished. Chiral matrix-assisted DOSY has the potential to allow simple resolution and assignment of the spectra of epimers and enantiomers, without the need for derivatisation or for titration with a shift reagent.

A donor-functionalized, silyl-substituted pentadienyllithium has been synthesized and characterized by crystallography and NMR spectroscopy. The pentadienyl negative charge was found to be partially localized, which DFT revealed to be due to the polarizing power of the lithium cation.

The sensitive detection and quantification of DNA targets in the food industry and in environmental and clinical settings are issues of utmost importance in ensuring contamination-free food, monitoring the environment, and battling disease. Selective probes coupled with powerful amplification techniques are therefore of major interest. In this study, we set out to create an integrated microchemical chip that benefits from microfluidic chip technology in terms of sensitivity and a strong detection methodology provided jointly by padlock probes and rolling circle amplification (RCA). Here, we have integrated padlock probes and RCA into a microchip. The chip uses solid phase capture in a microchannel to enable washing cycles and decrease analytical area, and employs on-bead RCA for single-molecule amplification and detection. We investigated the effects of reagent concentration and amount of padlock probes, and demonstrated the feasibility of detecting Salmonella.

Measurement of diffusion-weighted NMR spectra as a function of time allows the time-dependence of concentration and the isolated spectrum to be found for each component in a reaction, without prior assumptions about spectra, kinetics or diffusion behaviour, by data decomposition using the PARAFAC algorithm.

The problem of NMR signal overlap in DOSY is significantly diminished by suppressing multiplet structure in the spectral dimension.

Novel propranolol analogues have been designed and synthesised and their enantioselective binding to the cellulose degrading enzyme, Cel7A, has been evaluated. Affinity and enantioselectivity have been determined by capillary electrophoresis experiments. Ligands with significantly improved affinity and selectivity have been obtained and an analysis of the results has led to insights concerning the relation between the changes in ligand structure and selectivity as well as affinity to the protein.

An improved pulse sequence for measuring diffusion-ordered COSY spectra is achieved by incorporating the diffusion encoding directly into the evolution and detection periods of a gradient-enhanced COSY sequence, giving improved sensitivity and a 32-fold reduction in minimum experiment time.
