Epidermal stem cells (ESCs) were specific stem cell, which have the potential of proliferation and multi-directional differentiation. Epidermal stem cell located in epidermal basal layer and the eminence of hair follicle. At present, researches of epidermal stem cells in isolation, purifying, culture have made great progress. It was mainly applied in skin wound healing, building of tissue-engineered skin and gene therapy. Objective: To investigate the differential expression of Integrin β1 in epidermal stem cells among different age groups. Methods: The Epidermal stem cells were obtained by digesting human foreskin with Dispase II and BSA and Trypsin. The expression of Integrin β1 of epidermal stem cells was detected by Flow cytometry (FCM). Results: The epidermal stem cells marked with β1-integrin and sorted by flow cytometry were similar in shape and size among different age groups. The cell growth state of the group aged 7were the best among the three groups. The rate of positive expression of β1-integrin in the group aged 7, aged 16 and aged 30 were 93%, 56%, 3.5%, respectively. Conclusions: The expression of integrin β1 in three epidermal stem cells was decreased with the age, suggesting that the stem cells in skin tissues are decreased with age, and the lower age will be the best choice for tissue engineering.
