Objective: To investigate the relationship between systemic inflammatory response index (SII), serum thymidine kinase 1 (TK1), C-reactive protein/albumin (CRP/ALB) ratio and postoperative recurrence and metastasis in patients with triple negative breast cancer (TNBC). Methods: 268 TNBC patients admitted to the Affiliated Hospital of Xuzhou Medical University from January 2018 to May 2020 were selected and divided into recurrent metastasis group and non recurrent metastasis group according to whether they had recurrent metastasis 3 years after surgery. Calculated preoperative SII, CRP/ALB ratio and detected serum TK1 level. The factors of postoperative recurrence and metastasis in TNBC patients were analyzed by multivariate Logistic regression, the predictive value of SII, serum TKI and CRP/ALB ratio for postoperative recurrence and metastasis in TNBC patients was analyzed by receiver operating characteristic (ROC) curve. Results: After 3 years of follow-up, the recurrence and metastasis rate of 268 TNBC patients was 25.00% (67/268). Compared with non recurrence and metastasis group, the SII, serum TKI and CRP/ALB ratio in recurrence and metastasis group increased (P<0.05). Independent risk factors for postoperative recurrent metastasis in TNBC patients were TNM stage III, lymph node metastasis and SII, serum TKI, and an elevated CRP/ALB ratio P<0.05). The area under the curve of SII, serum TKI and CRP/ALB ratio in predicting postoperative recurrence and metastasis of TNBC patients was 0.917, which was greater than 0.789, 0.779 and 0.781 predicted by SII, serum TKI and CRP/ALB ratio alone. Conclusions: The increase of SII, serum TKI and CRP/ALB ratio are closely relate to the recurrence and metastasis of TNBC patients after operation, the combination of SII, serum TKI and CRP/ALB ratio has a high predictive value for the recurrence and metastasis of TNBC patients after operation. Objective: To investigate the relationship between the expression of NUAK family SNF1-like kinase 1 (NUAKI) and NUAK family SNF1-like kinase 2 (NUAK2) protein and clinicopathological features and prognosis in non-specific invasive breast cancer (IBC-NST). Methods: 132 patients with IBC-NST admitted to the Affiliated Hospital of Xuzhou Medical University from June 2018 to June 2020 were selected. Detected the expression of NUAK2 and NUAKI proteins in cancer tissue and adjacent tissues, the relationship between the expression of NUAK2, NUAK1 protein in cancer tissues and clinicopathological features were analyzed. IBC-NST patients were followed up for 3 years after discharge, the survival difference between patients with negative and positive expression of NUAK1 and NUAK2 were analyzed by Kaplan-Meier survival curve, the factors affecting the prognosis of IBC-NST patients were analyzed by multivariate COX regression analysis. Results: Compared with adjacent tissues, IBC-NST had a higher positive expression rate of NUAK2 and NUAKI proteins in cancer tissues (P<0.05). The positive expression rates of NUAK2 and NUAKI proteins in IBC-NST cancer tissue were related to the degree of differentiation, TNM staging, and lymph node metastasis (P<0.05). The 3-year progression-free survival (PFS) rate in IBC-NST patients with NUAKI positive expression was lower than that in IBC-NST patients with NUAKI negative expression (P<0.05), and the 3-year PFS rate in IBC-NST patients with NUAK2 positive expression was lower than that in IBC-NST patients with NUAK2 negative expression (P<0.05). Risk factors for poor prognosis in IBC-NST patients included NUAKI positive expression, lymph node metastasis, and NUAK2 positive expression (P<0.05). Conclusion: The positive expression rates of NUAKI and NUAK2 proteins in IBC-NST cancer tissue are significantly increased. To explore the distribution of pathogenic bacteria in sputum of patients with ICU in hospitals and analyze their drug resistance in order to provide the necessary bacteriological basis for clinical empirical antibiotic selection. Eighty-four patients who were admitted to an ICU in a certain hospital from January 2017 to December 2019 were selected as the research subjects, sputum samples were collected, and bacteriological culture and drug sensitivity tests were performed. (1) A total of 91 strains were extracted from 84 patients, of which 45 were G-bacteria, accounting for 49.45 %, 36 were G+ bacteria, accounting for 39.56 %, and 10 were fungi, accounting for 10.99 %. (2) Analysis showed that Imipenem has better sensitivity to G-bacteria, and vancomycin has better sensitivity to G+ bacteria. Vancomycin-resistant enterococci and staphylococci have not been found. There are many types of pathogenic bacteria in sputum of ICU patients. Among them, G-bacteria are the main pathogens. The pathogens show multiple drug resistance. Objective: To investigate the effect of additional acid etching on the shear bond strength of Self-etching adhesive applied to primary teeth dentin. Methods: 28 healthy deciduous teeth were randomly selected, the lip and cheek enamel were removed, the dentin bonding surface was exposed, and 56 samples along the proximal and distal directions were formed. They were randomly divided into 7 groups (n=8), nonetching groups(Al,A2,A3 group)was coated with AdperTM Easy One (AEO), Xeno-V (XV) and OptiBond All In One (AIO) self-etching adhesive, and additional etching groups(Bl, B2, B3 group )were etched with 35% phosphoric acid for 15 s before application of the Self-etching adhesive. Control group(group C)was primed with Bond & Bond NTTM (NT), and then use Z350 composite resin to build a resin column with a diameter of 3 mm. Shear test was conducted to test the shear bond strength and the morphology of the fracture surface was observed by scanning electron microscopy. Results: The shear bond strength values of group B1 and group B2 were significantly higher than those of group Al and group A2 (P<0.001). There was no significant difference in the shear bond strength between group B3 and group A3(P=0.94). The shear bond strength value of group A2 was lower than that of group C (P < 0.05). The shear bond strength of group B 1 was significantly higher than that of group C (P < 00 1 ). The result of TEM observation indicates that the fracture surface morphology of each group was mostly dentin and composite resin interface damage. Nonetching groups (group Al, A2, and A3) mostly occurred at the bottom of the mixed layer, and the resin protrusions were less and lower than the small nozzles. In the fracture surface of the specimens of group B1 and group B2, most of the dentinal tubules were filled with resin protrusions, and the fracture occurred mostly in the upper middle part of the mixed layer. In the fracture surface of the B3 group, the dentinal tubules were emptied and the resin was less. Conclusions: Additional acid etching improve the shear bond strength of AEO and XV self-etching adhesives applied to primary teeth dentin .The self-etching adhesive agent of the deciduous dentin can achieve the bond strength of the total-etching adhesive agent, but the use of a one-step self-etch adhesive with prior acid etching can result in a greater bond strength in primary tooth dentin. Objective： To compare the effects of simple radiotherapy and cisplatin combined with fluorouracil in the concurrent chemoradiotherapy for patients with advanced cervical cancer. Methods： 87 cases of patients with advanced cervical cancer who were treated in our hospital from March 2012 to March 2014were selected and randomly divided into the control group which was treated with simple radiotherapy and the observation group which was treated with concurrent chemoradiotherapy, with intravenous infusion of 600 mg/m2of 5- fluorouracil, and 50 mg/m2of cisplatin once a week, for a total of 6 cycles. The extracorporeal radiotherapy was performed within half an hour alter each chemotherapy ended. The clinical effects, incidence of adverse reactions such as thrombocytopenia, decreased WBC, impaired renal function and nausea and vomiting were compared between the two groups. Both groups of patients were followed up for 3 years, the recurrence rate, metastasis rate and long-term survival rate were observed and compared between the two groups. Results： Alter treatment, the effective rate of observation group was 90.63%（39/43）, which was significantly higher than that of the control group79.04%（32/44）（P〈0.05）; after 3 years of follow-up, the recurrence rate and metastasis rate of the observation group were significantly lower than those of the control group （P〈0.05）, the 3 year survival rate of observation group was significantly higher than that of the control group （P〈0.05）; the incidence of adverse reactions such as thrombocytopenia, decreased WBC, impaired renal function and nausea and vomiting in the observation group was significantly higher than that of the control group （P〈0.05）. Objective: To analyze the quality of pre-operation sleep and anxiety condition in patients with coronary artery disease and investigate its influencing factors. Methods: One hundred cases of cardiac surgical inpatients were investigated by using Pittsburgh Sleep Quality Index(PSQI) scale, Hospital Anxiety and Depression Scale(HADS) and self-made scale. The data were analyzed by t-test and X2 test. Results: 1The average total PSQI sore was 9.42±4.90 on 10 days pre-operation and 12.39±3.93 1 day pre-operation. 2The HADS score was significantly higher on 1 day pre-operation. The higher score was, the worse the sleep quality was. 3 The influencing factors of sleep quality of patients before operation were anxiety, preoperative, preparation, medical staff behavior, physical discomfort, environmental noise and so on. Conclusions: The pre-operation cardiac surgical inpatients had bad sleep quality and had a positive correlation with anxiety level. Objective: To determine the expression change of micro RNA in endothelial cells under oxidative stress. Methods:Human Umbilical Vein Endothelial Cells cultured by ECM, were stimulated by different concentrations of hydrogen peroxide(0 umol/L,200 μmol/L, 500 μmol/L, 800 μmol/L) for 24 hours. Cells apoptosis assessed by Annexin V/PI staining and flow cytometry. Total RNA was isolated from cells, and micro RNA expression levels were detected by using quantitive real-time PCR. Bioinformatics prediction software were applied to predict potential target genes of these mi RNAs. Results: The apoptosis rate of endothelial cells increased significantly after hydrogen peroxide stimulation. Compared with control cells, the apoptosis rate of 200 μmol/L, 500 μmol/L and 800μmol/L hydrogen peroxide treatment group were significantly increased(13.31 % vs 4.75 %, 35.9 % vs 4.75 %, 89.75 % vs 4.75 %,respectively, P<0.01). Quantitive real-time PCR results showed that the expression of mi RNA appeared obviously disorder, of which mi R-92 a, mi R-126 were decreased, while mi R-181 a, mi R-217, mi R-34 a and mi R-320 were increased notably(P<0.05). Mi R-92 a and mi R-320 might regulate the expression of multiple genes related to the apoptosis of endothelial cells. Conclusion: The abnormal expression mi RNAs in HUVECs after oxidative stress induced apoptosis suggested that these mi RNAs maybe involved in regulation of endothelial cells function. Objective: To study the protective effect of autophagy on endothelial cells apoptosis induced by high glucose.Methods: Human coronary artery endothelial cells cultured by conventional media(Control group), conventional media contraining 30mmol/L D-glucose(High-glucose group), high-glucose media plus 100 nmol/L Rapamycin(RAPA group), or high-glucose media plus5mmol/L 3-Methyladenine(3-MA group). Cells vitality and apoptosis were measured by CCK-8 assay and flow cytometry. The autophagy marking protein(Beclin1) expression was determined by western blot. Results:(1) Endothelial cell viability decreased significantly after high-glucose(30 mmol/L) stimulation, with 55.0% to the control group(P<0.01). Beclin1 expressed increased, and the level of cell apoptosis was significanly increased, with 2 times to the control group.(2) Compared with high-glucose group, the cell viability and the beclin1 expression of RAPA group were both increased(P<0.01). The level of cell apoptosis was significanly decreased,with 70.1% to the high-glucose group.(3) Compared with high-glucose group, the cell viability and the beclin1 expression of RAPA group were both decreased(P<0.01). The level of cell apoptosis was significanly increased, with 1.42 times to the high-glucose group.Conclusion: Cell autophagy might have an apoptosis-protective effect on the human coronary artery endothelial cells induced by high-glucose. Objective: To determine the expression change of microRNA(miRNA) in endothelial cell injury induced by high-glucose. Methods: Human coronary artery endothelial cells cultured by conventional methods, were stimulated by using different concentrations of D-glucose solution (0 mmol/L, 5 mmol/L, 15 mmol/L and 25 mmol/L). Cells vitality and apoptosis were measured by CCK-8 assay and flow cytometry. Total RNA was isolated from cells in 24 h after high-glucose induced, and miRNA expression levels were detected by using quantitative real-time PCR. Bioinformatics prediction software including Targetscan and PicTar etc, were applied to predict potential target genes of these miRNAs. Results: Endothelial cell viability decreased significantly after high-glucose(25 mmol/L) stimulation, with 67.5% to the control cells(P<0.01), and the apoptosis level were 4.5 times compared with control cells(P<0.01). Quantitative real-time PCR results showed that the expression of miRNA appeared obvious disorder, which were miR-451, miR-504, miR-302 d,miR-18b*, miR-198, miR-328 and miR-517 c were increased, while miR-29 c, miR-100*, miR-137, miR-660 and miR-217 were decreased notably (P<0.05). MiR-217 and miR-451 might regulate the expression of multiple genes related to endothelial cell function by Bioinformatics prediction. Conclusion: The abnormal expression miRNAs in endothelial cells after high glucose induced injury suggested that these miRNAs maybe be involved in regulation of endothelial cells function.
