Syphilis, caused by Treponema pallidum, is resurging globally. Molecular typing allows for the investigation of its epidemiology. In Pakistan and other nations, T. pallidum subsp. pallidum has developed widespread macrolide resistance in the past decade. A study at the Peshawar Regional Blood Centre from June 2020–June 2021 analyzed serum samples from 32,812 blood donors in Khyber Pakhtunkhwa, Pakistan, to assess circulating T. pallidum strains and antibiotic resistance. Blood samples were initially screened for T. pallidum antibodies using a chemiluminescent microparticle immunoassay (CMIA). CMIA-reactive samples underwent polymerase chain reaction (PCR) targeted the polA, tpp47, bmp, and tp0319 genes. PCR-positive samples were further analyzed for molecular subtyping using a CDC-developed procedure and tp0548 gene examination. All PCR-positive samples were analyzed for the presence of point mutations A2058G and A2059G in 23S rRNA, as well as the G1058C mutation in 16S rRNA. These mutations are known to impart antimicrobial resistance to macrolides and doxycycline, respectively. Out of 32,812 serum samples, 272 (0.83%) were CMIA-reactive, with 46 being PCR-positive. Nine T. pallidum subtypes were identified, predominantly 14d/f. The A2058G mutation in 23S rRNA was found in 78% of cases, while G1058C in 16S rRNA and A2059G in 23S rRNA were absent. The research found donor blood useful for assessing T. pallidum molecular subtypes and antibiotic resistance, especially when chancres are not present. The prevalent subtype was 14d/f (51.85%), and the high macrolide resistance of 36 (78%) indicates caution in using macrolides for syphilis treatment in Khyber Pakhtunkhwa, Pakistan. Objective: The Difficulties in Emotion Regulation Scale-18 (DERS-18) is an instrument used to measure deficits in emotion regulation. However, the instrument has not been adapted to Malaysians and has never been validated in the Malay language. This study aimed to examine the psychometric properties of the Malay version of DERS-18. Method: The DERS-18 underwent forward-backward translation and assessment of face and content validity. Both Malay version of the DERS-18 and DASS-21 were completed by 701 adolescents (44.4% boys) aged 13 and 14 years old. To assess its dependability, a floor and ceiling effect evaluation and Cronbach's analysis were both performed. A series of confirmatory factor analyses (CFA), bivariate correlation, and regression were performed to evaluate the construct and criterion validity, respectively. Results: The Malay version of DERS-18, after excluding "Awareness", indicated excellent reliability (Cronbach's α = 0.93), and acceptable internal consistency for each subscale (range of α from 0.63 to 0.82). Floor or ceiling effects were observed at item level and subscale level, but not at total level. CFA results revealed that the Malay version of the DERS-18 bifactor model (excluding "Awareness") portrayed the best construct validity (χ2/df = 2.673, RMSEA = 0.049, CFI = 0.977, TLI = 0.968) compared to a single factor, a correlated factor, and a higher-order factor model. The DERS-18 subscales (except "Awareness") and DERS-18 total scores were significantly correlated with stress, anxiety, and depression in a positive direction (r ranged from 0.62 to 0.64, p < 0.01). The general factor of the DERS-18 and its specific factors ("Clarity", "Goals", and "Non-Acceptance") significantly predicted the symptoms of stress, anxiety, and depression (R2 ranged from 0.44 to 0.46, p < 0.001). Conclusion: The Malay version of the DERS-18, excluding "Awareness", possessed good reliability, construct validity, and criterion validity to assess emotion dysregulation among Malaysian adolescents. The Zigzag ladybird beetle, Cheilomenes sexmaculata (Fabricius) (Coleoptera: Coccinellidae), is a biological control agent that feeds on a variety of aphid species. Life table and predation data of C. sexmaculata were collected under laboratory conditions at 25±2⁰C, 60±5% RH and L14: D10 h in connection with feeding on four different aphid species; Lipaphis erysimi (Kaltenbach), Myzus persicae (Sulzer), Aphis nerii (Boyer de Fonscolombe) and Diuraphis noxia (Mordvilko). Larval development of C. sexmaculata was long when fed on M. persicae (12.18 days) and shorter on D. noxia (10.64 days). The male's lifespan was longer on M. persicae (26.70 days) and shorter on L. erysimi (23.67 days). Fecundity was maximum when the beetle was fed D. noxia (316.8 eggs/female) and minimum on M. persicae (199.1 eggs/female). Net reproductive rate, intrinsic rate of increase and finite rate of increase were highest on D. noxia with values of 158.4 (offspring individual-1), 0.22 d-1, and 1.24 d-1, respectively whereas the respective parameters were lowest on L. erysimi (99.5 offspring individual-1, 0.19 d-1, and 1.20 d-1, respectively). However, the mean of the generation (T) was shorter on A. nerii (22.48 d-1) and longer on M. persicae (24.68 d-1). Based on life table parameters obtained under laboratory conditions, the most appropriate host of C. sexmaculata was D. noxia. Extremely low temperatures present various challenges to life that include ice formation and effects on metabolic capacity. Psyhcrophilic microorganisms typically have an array of mechanisms to enable survival in cold temperatures. In this study, we sequenced and analysed the genome of a psychrophilic yeast isolated in the Antarctic region, Glaciozyma antarctica. The genome annotation identified 7857 protein coding sequences. From the genome sequence analysis we were able to identify genes that encoded for proteins known to be associated with cold survival, in addition to annotating genes that are unique to G. antarctica. For genes that are known to be involved in cold adaptation such as anti-freeze proteins (AFPs), our gene expression analysis revealed that they were differentially transcribed over time and in response to different temperatures. This indicated the presence of an array of adaptation systems that can respond to a changing but persistent cold environment. We were also able to validate the activity of all the AFPs annotated where the recombinant AFPs demonstrated anti-freeze capacity. This work is an important foundation for further collective exploration into psychrophilic microbiology where among other potential, the genes unique to this species may represent a pool of novel mechanisms for cold survival. Theoretical analysis has been carried out to establish the heat transport phenomenon of six different ferromagnetic MnZnFe2O4—C2H6O2 (manganese zinc ferrite-ethylene glycol), NiZnFe2O4—C2H6O2 (Nickel zinc ferrite-ethylene glycol), Fe2O4—C2H6O2 (magnetite ferrite-ethylene glycol), NiZnFe2O4—H2O (Nickel zinc ferrite-water), MnZnFe2O4—H2O (manganese zinc ferrite-water), and Fe2O4—H2O (magnetite ferrite-water) nanofluids containing manganese zinc ferrite, Nickel zinc ferrite, and magnetite ferrite nanoparticles dispersed in a base fluid of ethylene glycol and water mixture. The performance of convective heat transfer is elevated in boundary layer flow region via nanoparticles. Magnetic dipole in presence of ferrites nanoparticles plays a vital role in controlling the thermal and momentum boundary layers. In perspective of this, the impacts of magnetic dipole on the nano boundary layer, steady, and laminar flow of incompressible ferromagnetic nanofluids are analyzed in the present study. Flow is caused by linear stretching of the surface. Fourier’s law of heat conduction is used in the evaluation of heat flux. Impacts of emerging parameters on the magneto—thermomechanical coupling are analyzed numerically. Further, it is evident that Newtonian heating has increasing behavior on the rate of heat transfer in the boundary layer. Background: Vitamin D is postulated to decrease the risk of breast cancer by inhibiting cell proliferation via the vitamin D receptor (VDR). Two common single nucleotide polymorphisms (SNPs) in the VDR gene, rs1544410 (BsmI) and rs2228570 (FokI), are inconsistently associated with breast cancer risk in Caucasian populations, while data for Asians are scarce. Here, we investigated the possible contribution of these SNPs to breast cancer risk in Pakistani breast cancer patients and in controls participating in a hospital-based breast cancer case-control study (PAK-BCCC). Methods: Genotyping of the BsmI and FokI SNPs was performed by PCR-based restriction fragment length polymorphism (RFLP) analysis of 463 genetically enriched female breast cancer cases with known BRCA1/2 status and in 1,012 controls from Pakistan. The association between SNP genotypes and breast cancer risk was investigated by logistic regression adjusted for potential breast cancer risk factors and stratified by BRCA1/2 status and family history. Odds ratios (ORs) and 95% confidence intervals (CIs) were reported. Results: The b allele of the BsmI was associated with an increased breast cancer risk (per b allele OR 1.28, 95% CI 1.09–1.49, P = 0.003). Subgroup analysis revealed that this effect was restricted to BRCA1/2 non-carriers (per b allele OR 1.33, 95% CI 1.11–1.59, P = 0.002) and was stronger in those who reported a positive family history of breast and/or ovarian cancer (per b allele OR 1.64, 95% CI 1.20–2.22, P = 0.002). No association with breast cancer risk was detected for the FokI SNP. Conclusions: The BsmI polymorphism in the VDR gene may be associated with an increased breast cancer risk in Pakistani women negative for BRCA1/2 germline mutations. [ABSTRACT FROM AUTHOR]
